Description

細胞を、プロテアーゼ阻害剤(COMPLETE、Mini、EDTAフリーの錠剤;ROCHE、Basel,Switzerland)およびホスファターゼ阻害剤(PHOSSTOP Phosphatase Inhibitor Cocktail、ROCHE、Basel,Switzerland)を補充したレポーター溶解緩衝液(1x、PROMEGA)で溶解した。タンパク質を10%SDS-PAGEで分離し、PVDF膜に転写し、室温で2時間または4℃で一晩、一次抗体でプローブした。二次抗体とインキュベーションして展開した後、フィルム(BIOMAX MRフィルム;CARESTREAM HEALTH、Rochester,NY)上で、またはCHEMIDOC IMAGING SYSTEM(BIO-RAD、Hercules.CA)を使用してバンドを検出し、IMAGE STUDIO(商標)LITEソフトウェア(LI-COR、Lincoln,NE)またはIMAGE LABソフトウェア(BIO-RAD、Hercules,CA)を使用して定量化した。
RNeasy Mini Kit(QIAGEN、Hilden,Germany)を使用して全RNAを単離した。分光光度法によりRNA濃度を決定した後、100~1000ngの全RNAをcDNA合成に使用した。REVERSE TRANSCRIPTION SYSTEM(PROMEGA、Madison,WI)を使用して逆転写を行った。10マイクロリットルの反応液を100μlに希釈し、ITAQ(商標)UNIVERSAL SYBR(登録商標)GREEN SUPERMIX(BIORAD、Hercules,CA)を使用する定量的リアルタイムPCRのために1マイクロリットルを取り出した。プライマーの特異性は、融解曲線分析およびゲル電気泳動により確認した。qPCRは、CFX CONNECT REAL TIME PCR DETECTION SYSTEM(BIORAD、Hercules,CA)で行った。試料を3通りアッセイした。相対的なRNAレベルは、ΔΔCt法を使用して定量化し(75)、特に指定がない限り、内因性対照GAPDHに対して正規化した。
細胞を3回洗浄した後、室温で15分間、PBS中の4%パラホルムアルデヒドで固定した。固定した細胞をPBSで洗浄し、PBS中の0.2% Triton X-100で10分間透過処理した。PBSで洗浄した後、トランスウェルインサートをブロッキング試薬(DAKO PROTEIN BLOCK SERUM FREE、AGILENT)で1時間ブロックし、一次抗体とともに4℃で一晩インキュベートした。核を2.5μg/mlのDAPIで5分間染色し、次いでPBSで2回洗浄した。次いで、清浄なカミソリの刃を使用してトランスウェル膜を注意深く切り取り、ProLong褪色防止用封入剤(FISHER、Waltham,MA)を使用してスライドガラスに封入した。スライドをLeica DMI6000顕微鏡下で検査し、オープンソースソフトウェアMANAGERまたはVOLOCITYソフトウェア(VELOCITY INC.)を使用して写真をキャプチャした。ARDS患者からのヒト肺の組織切片は、Institutional Review Board承認のプロトコルを使用して、University of Rochesterの病理学部門によって提供された。全ての試料は剖検で採取された。合計で、6人のARDS患者から16の切片と、3人のARDSのない対照患者から7つの切片を得た。対応する各セクションのH&E染色は、様々な程度の肺損傷および浮腫の内容を示す。免疫蛍光染色のために、組織切片を脱パラフィンして再水和した。次いで、抗原検索ステップを実行して、後続の抗体結合および免疫蛍光のためにエピトープを露出させた。
アッセイの前に、12ウェルトランスウェルプレート(孔径0.4μmのポリエステル膜インサートを備えた12mmトランスウェル;CORNING、Corning,NY)上で培養した細胞を組織培養フードに15分間移し、培地を室温に平衡化させた。TEERは、上皮電圧計(EVOM2;WORLD PRECISION INSTRUMENTS、Sarasota,FL)を使用して測定した。3つの読み取り値を記録し、各ウェルについて平均した。TEERを計算するために、細胞を含まないフィブロネクチンでコーティングしたインサートの抵抗(ブランク抵抗)を、測定された抵抗から差し引き、次いで1.12cm2を乗じてインサートの表面積を求めた。
蛍光トレーサーに対する透過性は、以前に記載した修正プロトコルを使用して測定した(76)。TEER測定後、上部および下部のトランスウェルチャンバーをP緩衝液(pH7.4の10mM HEPES、1mMピルビン酸ナトリウム、10mMグルコース、3mM CaCl2、および145mM NaCl)で2回洗浄した。100μg/mLの40kD FITC-デキストランおよび100μg/mLの3kD テキサスレッド-デキストランを含む500マイクロリットルの新たに調製した溶液を、頂端区画に加えた。1000マイクロリットルのP緩衝液を下部チャンバーに加えた。37℃で2時間インキュベートした後、100μlの基本培地を回収し、輸送されたデキストランの蛍光をSPECTRAMAX M5マルチモードマイクロプレートリーダー(MOLECULAR DEVICES、San Jose,CA)で測定した。励起波長および発光波長は、それぞれ、FITCで492nmおよび520nm、テキサスレッドで596nmおよび615nmである。標準曲線と比較することによりトレーサーの量を算出した。透過係数は、以下の式を使用して決定した(77):Pc(cm/min)=V/(AxCo)x(C/T)、式中、Vは下部区画容積(1ml)、Aは膜の表面積(ここで使用される12ウェルトランスウェルの場合は1.12cm2)、Coは時間0での上部区画のデキストラン濃度(0.1mg/ml)、Cは時間T(2時間)での下部区画のデキストラン濃度である。
1枚の100mmプレートからの細胞を1mlのIP溶解緩衝液(1% NP-40、50mM Tris HCl、pH8.0)で溶解し、25ゲージシリンジで10回ホモジナイズした。製造者(MILTENYI BIOTEC、Bergisch-Gladbach,Germany)の指示に従ってμMACS Protein Gキットを使用して免疫沈降を行った。事前に清澄化した試料を、抗MRCKα抗体(PA1-10038、1:50希釈;FISHER、Waltham,MA)、抗β1抗体(UPSTATE、05-382、1:250希釈)、または対照としてのIgGとともに4℃で一晩インキュベートした。溶出液をSDS-PAGEグラジエントゲル(4~20%)により分析した。各レーンをほぼ同じサイズの10個の小片に切断した。次いで、ゲルのバンドを脱染し、還元し、トリプシンで一晩消化した。次いで、消化されたペプチド混合物を、Orbitrapシステムを使用してLC-MS/MS分析に供した。
Thermoの原データをmgf形式に変換した。結果として得られたピークリストを、PROTEIN PROSPECTOR(v5.22.0)を使用して以下の設定で検索した:最大1つの切断部位が欠落しているプロテアーゼとしてのトリプシン、MSに対する10ppmの質量許容誤差、MS/MSについてそれぞれ0.5Da(イオントラップ)および0.05Da(ORBITRAP)、固定としてのカルバミドメチル化(C)、可変修飾としての酸化(M)およびリン酸化(S/T/Y)。PROTEIN PROSPECTORからの結果を取得し、社内のPythonスクリプトを使用してクリーンアップした。NSAF測定を使用したタンパク質の定量化については以前に記載されている(23)。データの正規化、注釈、および統計分析は、Perseusを使用して実行した(78)。NSAFの統計分析には、スチューデントのt検定が使用された(79)。
肺胞上皮バリアの細胞間結合の大部分は、隣接するATI細胞間にあり、その表面の95%を覆っている(5)。残念ながら、既存の細胞株はインビボでのATIの遺伝的特徴および表現型特徴を完全に再現しておらず、ATI細胞を直接単離することは技術的な課題をもたらす(19)。これを克服するために、ラットの初代ATII細胞が使用されたが、それは、該細胞を単離してインビトロで培養するとATI細胞に分化することができるためである(20)。プロセス中の表現型の変化を追跡するために、ATII(SPC)またはATI(T1α)に特異的な遺伝子についてqPCR分析を行った(図1A)。SPC mRNAレベルは、単離後24時間から48時間で15分の1に低下したことから、ATII表現型の喪失が示唆される。対照的に、T1αレベルは、単離後5日目まで継続的に増加し、ATI表現型への移行を示した。20μg/mlのフィブロネクチンでコーティングされたトランスウェルプレートで培養した場合、これらの細胞は、16HBE14oまたはCalu-3細胞で測定されたものに匹敵するかまたはそれ以上の、細胞単層全体の電気抵抗の測定値であるTEERを示し(図9A)(21)、細胞膜に局在するオクルディンおよびzo-1のほぼ連続的な染色が見られた(図9B)。一方、1μg/mlのリポ多糖で処理した場合、これらの細胞はTEERの低下および4kDデキストランに対する透過性の増加を示したことから(図9Cおよび図9D)、上皮バリアの損傷が示唆される。これらのデータは、この初代ラットATI培養系が、肺胞上皮バリアを研究するための適切なモデルとしての役割を果たすことを示している。
β1サブユニットがATI細胞においてタイトジャンクションのタンパク質発現を増加させたことを考慮して、これらの細胞におけるそれらの局在を分析した。免疫蛍光染色により、細胞膜上のオクルディン、zo-1、zo-2、およびクローディン-4のレベルの上昇が確認された(図2A)。次に、肺胞上皮バリアに対するβ1サブユニットの機能的効果を調査するためにアッセイを行った。エレクトロポレーションでは細胞をトリプシン処理する必要があり、それによって細胞単層が破壊され、TEERおよび透過性アッセイが妨げられるため、ラットβ1サブユニットをTet-onプラスミドにクローニングすることにより、ドキシサイクリン誘導系を作製してβ1サブユニットの発現を制御した。これにより、エレクトロポレーション後、細胞培養培地にドキシクリンを添加するだけで、ATI細胞の遺伝子発現をオンにすることができる。このシステムを使用してβ1サブユニットの発現を制御できるかどうかを調べるために、ヒト気管支上皮細胞株である16HBE14o-細胞に、エレクトロポレーションによりpCMV-tet調節プラスミドおよびpTet3G-ヒトβ1サブユニット発現プラスミドをコトランスフェクトし、その後直ちに増加する濃度のドキシサイクリン(0、1、10、100、1000ng/ml)を加えた。イムノブロット分析は、ドキシサイクリンがトランスフェクションの24時間後にβ1サブユニットの用量依存的なアップレギュレーションを引き起こし、1000ng/mlで最大誘導が見られたことを示した(図11A)。その結果、オクルディンおよびzo-1の発現も増加し、pCMV-ラットβ1プラスミドを使用した以前の知見が裏付けられた。さらに、ルシフェラーゼレポータープラスミドを使用すると、一過性のトランスフェクションがトランスフェクション後7日まで遺伝子のアップレギュレーションをもたらすことが明らかになった(図11B)。
Na+/K+-ATPアーゼのβサブユニットは、αサブユニットの成熟および膜輸送を促進し、それによってイオン輸送活性を増加させる(22)。この活性がβ1サブユニットのバリア増強効果に必要な場合、β2またはβ3アイソフォームの過剰発現はβ1と同じ効果を有する可能性がある。これを調べるために、単離の3日後、ATI細胞がATI表現型を表示したときに、ATI細胞にマウスβ2サブユニットまたはマウスβ3サブユニットをコードするプラスミドをトランスフェクトした。次いで、24時間後、ウェスタンブロットによりタイトジャンクション構成タンパク質のレベルを評価した。β1サブユニットとは対照的に、β2アイソフォームの過剰発現はオクルディンまたはzo-1の発現を増加させず(図3A)、β3サブユニットの過剰発現は、そのレベルをそれどころか低下させた(図3B)。
上記の知見は、β1サブユニットを介した上皮バリアの緊密性がそのイオン輸送活性とは無関係であることを示唆しているため、β1相互作用タンパク質が役割を果たす可能性があるという仮説が立てられた。しかしながら、文献で報告されたこれらのタンパク質はほんのわずかであり、それらのほとんどは神経系でのみ発現される(表S1/図16)。肺上皮細胞のβ1サブユニットと相互作用しているタンパク質を体系的に特定するために、タンデム質量分析を使用した。
上記のデータは、MRCKαがβ1によって誘導されるATI細胞上皮バリア増強の下流メディエーターであったことを示唆しているため、肺胞バリア機能を改善するにはMRCKαの活性増加のみで十分であると推測された。この仮説を検証するために、ATII細胞にMRCKαプラスミドをトランスフェクトし、TEERを使用してバリアの完全性を測定した。トランスフェクションの24時間後、TEERに有意差は検出されなかった。しかしながら、トランスフェクションの48時間後および72時間後の両方で、MRCKαをトランスフェクトした細胞に空のプラスミド対照と比較して有意に高い値が観察された(図6A)。この結果と一致して、オクルディンおよびzo-1は、MRCKαトランスフェクション時により高い強度および細胞間境界への局在の増加を示した(図6B)。これらの結果は、肺胞上皮バリアの完全性を促進するにはMRCKαの過剰発現のみで十分であることを示している。
これまでの結果は、β1サブユニット相互作用タンパク質MRCKαが肺胞タイトジャンクションを促進するために必要かつ必須であるという仮説を支持している。この結論をさらに実証するために、ミオシン軽鎖キナーゼ2(ミオシンホスファターゼMYPT1の阻害により直接的または間接的に)、LIMキナーゼ、およびミオシン(29-31、33、36-38)を含むMRCKα下流経路の活性化を調べるためのアッセイを行ったこれらの基質のリン酸化は、ウェスタンブロット分析を使用して評価した。β1サブユニットの過剰発現により、Ser19でミオシン軽鎖2(MLC2)のリン酸化が2倍に誘導されることがわかった(図7A)。したがって、このデータは、β1サブユニットがMRCKαと相互作用して活性化することをさらに裏付けるものである。
MRCKαが肺胞バリアの完全性を調節することを考慮して、ARDSにおいてその発現が変化するかどうかを調べた。免疫蛍光染色は、ARDS患者の肺が、対照ドナーの肺と比較してはるかに低いレベルのMRCKαを発現し(図13Aおよび図8A)、相対蛍光強度が平均30%少ないことを示した(図8B)。肺胞に加えて、小気道も、特にオクルディンが発現している繊毛および基底細胞において、高レベルのMRCKαを発現した(図13B)。重要なことに、これらの組織の染色強度もARDS患者で減少した(図13C)。総合すると、これらのデータは、肺のMRCKαのレベルがより低いことがARDSの病態に関連していることを示している。
この実施例では、インビボでのMRCKαの役割を調べるためにアッセイを行った。簡潔に述べると、肺炎感染または細菌性敗血症を模倣し、急性肺損傷/急性呼吸窮迫症候群(ALI/ARDS)を引き起こすLPSでマウスの肺を損傷させた。1日後、肺が好中球および肺水腫液で満たされたとき、図14Aおよび図14Bに示される様々なタンパク質をコードするプラスミドをエレクトロポレーションにより肺に送達した。プラスミドは、対照プラスミド(「空」、遺伝子産物を発現しないため、加えられるDNAの良好な陰性対照である)、およびNa/K-ATPアーゼ(「b1」)のβ1サブユニット、MRCKα(「MRCK」)、またはβ1サブユニットとMRCKαとの組み合わせ(「b1+MRCK」)をコードするものを含む。その2日後、肺を採取し、損傷のエンドポイントについて調べた。
この実施例では、インビボでの肺胞-毛細血管上皮-内皮バリア機能を改善するためのMRCKαの役割を調べるために、追加のアッセイを行った。
プラスミド
プラスミドpcDNA3は、PROMEGA(Madison,WI)から入手した。pCMV-MRCKαは、CMVプロモーターからヒトMRCKαを発現し、pCMV-Na+/K+-ATPアーゼβ1は、以前に記載されたように、GFPタグ付きラットNa+/K+-ATPアーゼβ1サブユニットを発現する(Gagliardi,P.A.,et al.,J Cell Biol 206:415-34、およびMachado-Aranda,D.,et al.,Am J Respir Crit Care Med 171:204-11)。QIAGEN GIGA-PREP KITS(QIAGEN、Chatsworth,CA)を使用してプラスミドを精製し、10mM Tris-HCl(pH8.0)、1mMエチレンジアミン四酢酸、および140mM NaClに懸濁した。
雄のC57BL/6マウス(9~11週)をイソフルランで麻酔し、100μgの各プラスミドを、Lin,X.,et al.,Gene Ther 23:489-99に以前に記載されたように、吸引によって、50μの10mM Tris-HCl(pH8.0)、1mM EDTA、および140mM NaCl中でマウスの肺に送達した(β1とMRCKαの両方を送達した場合、エレクトロポレーションの前にそれぞれ100μgを投与した)。ECM830エレクトロポレーター(BTX,HARVARD APPARATUS、Holliston,MA)を用いてマウスの胸部に配置された電気生理学的な皮膚電極(MEDTRONIC、Redmond,WA)を使用して、200V/cmの電界強度で8つの10ミリ秒の方形波パルスを直ちに印加した。遺伝子導入の1日前に、全てのLPSチャレンジマウスに、吸引によって、50μlのリン酸緩衝生理食塩水(PBS)中の5mg/kgのLPS(Escherichia coli O55:B5、15,000,000 エンドトキシンユニット/mg タンパク質;SIGMA-ALDRICH、St.Louis,MO)を投与した。(n=5~11マウス/群)。全ての実験手順は、American Association for the Accreditation of Laboratory Animal Care承認施設で、実験動物のケアと使用に関する施設のガイドラインに従って実施された。
この試験で使用した方法は、Lin,X.,et al.,Gene Ther 23:489-99およびMutlu,G.M.,et al.,Circ Res 94:1091-100に以前に記載されたように、生きたマウスで行われた。簡潔に述べると、37℃の体温に維持されたマウスをジアゼパム(5mg/kg、腹腔内投与)およびペントバルビタール(50mg/kg、ジアゼパムの10分後に腹腔内投与)で麻酔した。5mm、20ゲージの血管カテーテル(BECTON-DICKENSON、Sandy,UT)で気管にカニューレを挿入し、カテーテルを小動物人工呼吸器に接続した後、臭化パンクロニウム(0.04mg、腹腔内投与)で麻痺させた。マウスは、100%酸素、および1分当たり160呼吸の頻度で、10ml/kgの1回換気量で換気した。5%無酸性のエバンスブルーで標識したウシ血清アルブミン(0.15mg/ml、SIGMA、St.Louis,MO)を含む等浸透圧性の(324mOsm)0.9% NaCl溶液300mlを、10秒にわたって気管内カテーテルに注入し、その後200μlの空気を注入して液体を肺胞腔に配置した。マウスを30度に傾けて仰臥位を維持し、30分間換気した後、両側気胸を作製するために開胸し、気管カテーテルからの流体の吸引を容易にした。吸引液中のタンパク質濃度は、Bradfordアッセイ(BIO-RAD LABORATORIES、Hercules,CA)を使用して評価し、AFCは以下の式を用いて計算した:AFC=1-(C0/C30)、式中、C0は、注入前の注入液のタンパク質濃度であり、C30は、30分の機械的換気の終わりに得られた試料のタンパク質濃度である。クリアランスは、排出された総注入量/30分のパーセンテージとして表される。プロカテロール(特異的β2ARアゴニスト、10-8M)を陽性対照として注入液中で投与した。
LPS誘発急性肺損傷の総肺水分含有量への影響は、LPSの注入から72時間後に決定した。マウスを左腎動脈および左腎静脈の断裂により失血させた後、肺を切除し、表面の液体を吸い取った。湿潤肺重量を評価し、70℃のハイブリダイゼーションオーブンに72時間肺を入れた後、安定した乾燥重量を得た。
BALは、Lin,X.,et al.,Gene Ther 23:489-99およびMutlu,G.M.,et al.,Am J Respir Crit Care Med 176:582-90に以前に記載されたように行った。簡潔に述べると、滅菌PBSの2つの別々の0.5mlアリコートを、洗浄のためにマウスの肺に注入した。直ちに処理するために液体を氷上に置き、血球計数器を使用して洗浄液中の細胞の総数を数えた。BALからの細胞は、サイトスピン後にDIFF-QUIK(商標)(SIEMENS、Newark,DE)で染色した。
マウスを屠殺した直後に、20cc/kgの10%(vol/vol)緩衝化ホルマリンで肺を膨張させ、パラフィン包埋切片に使用した。切片(5μm)をヘマトキシリンおよびエオシンで染色し、盲検化し、肺の炎症反応および病理学的変化の分析について検討した。
肺の透過性は、血液から気道へのエバンスブルー色素(EBD)の漏出によって測定した(Baluk,P.,et al.,Br J Pharmacol 126:522-8およびMammoto,A.,et al.,Nat Commun 4:1759)。LPSの気管内投与によりマウス(n=7~11)をチャレンジし、1日後、α1-ENaCまたはβ1-Na+/K+-ATPアーゼのみを発現するプラスミドを肺にエレクトロポレーションした。遺伝子導入の47時間後、尾静脈注射によってEBD(30mg/kg、SIGMA、St.Louis,MO)を投与した。1時間後、肺を5mlの滅菌PBSで灌流して血管系内のEBDを除去し、次いで取り出し、写真を撮影し、60℃で乾燥させた。24時間後、37℃のホルムアミド(FISHER SCIENTIFIC)中でEBDを24時間抽出し、620nmおよび740nmで分光光度的に測定することにより定量化し、式E620(EBD)=E620-(1.426×E740+0.030)を用いて補正した(Standiford,TJ,et al.,J Immunol 155:1515-24)。
ウェスタンブロットは、Lin,X.,et al.Am J Respir Crit Care Med 183:1689-97に以前に記載されたように行った。簡潔に述べると、プロテアーゼ阻害剤を含む溶解緩衝液に肺組織を可溶化した。30μgの総タンパク質を10% SDS-PAGEにロードし、PVDF膜に転写し、オクルディン(THERMO FISHER SCIENTIFIC、Waltham,MA)、ZO-1(INVITROGEN、Carlsbad,CA)、またはGAPDH(SIGMA-ALDRICH、St.Louis,MO)に対する一時抗体でプローブした。二次抗体とインキュベーションして展開した後、CHEMIDOC IMAGING SYSTEM(BIO-RAD、Hercules.CA)を使用してバンドを検出し、IMAGE STUDIO(商標)LITEソフトウェア(LI-COR、Lincoln,NE)またはIMAGE LABソフトウェア(BIO-RAD、Hercules,CA)を使用して定量化した。
定量的結果は、インビボ試験では平均±SEM、インビトロ実験では平均±SDとして表される。データを一元配置ANOVAまたは二元配置ANOVAで統計的に評価し、P値<0.05は統計的に有意であると見なした。
既存のLPS誘発性肺損傷を有するマウスにMRCKαを遺伝子導入すると、肺損傷の複数の指標が減少する。
Na+/K+-ATPアーゼのβ1サブユニットをマウスの肺に遺伝子導入すると、その後のLPS誘発性肺損傷を防ぎ、既存のLPS誘発性肺損傷を治療することができる。重量分析によって測定されるこの肺水腫の減少は、Na+/K+-ATPアーゼの機能による肺からの活発な流体除去の増加と、β1サブユニットの過剰発現によって誘発されるバリア機能の増加の両方の組み合わせに起因するものであった。培養細胞では、β1サブユニットがMRCKαを介してバリア機能をアップレギュレートするシグナルを伝達することが示されたため、MRCKαの過剰発現が既存のLPS誘発性肺損傷を有する肺における肺水腫の減少にもつながり得るかどうかを調べるためにアッセイを行った。
MRCKαの遺伝子導入だけで、培養細胞で見られるように、およびβ1-Na+/K+-ATPアーゼの遺伝子導入後にマウスの肺で見られるように、タイトジャンクション構成タンパク質のレベルを増加できるかどうかを判断するために、健康な肺およびLPS損傷肺の両方でタイトジャンクション構成タンパク質ZO-1およびオクルディンの発現を測定した。LPSによる肺の損傷は、ZO-1とオクルディンの両方のレベルを低下させた。図20に示すように、生理食塩水または対照プラスミドpcDNA3の送達は、LPS損傷動物におけるZO-1またはオクルディンの発現に影響を与えなかった。対照的に、β1-Na+/K+-ATPアーゼ、MRCKα、またはこれら2つの組み合わせを発現するプラスミドの送達は、健康な動物におけるZO-1とオクルディンの両方の発現を3~4倍と大幅に増強した。β1がMRCKαを介してシグナルを伝達すると仮定すると、これらのタイトジャンクション構成タンパク質の誘導に、プラスミドの単独または組み合わせによる違いは見られなかった。これらの結果は、膜のタイトジャンクション構成タンパク質レベルおよびタイトジャンクション活性を増加させるにはMRCKαの遺伝子導入のみで十分であるという培養細胞における知見を裏付けるものである。
タイトジャンクションのβ1-Na+/K+-ATPアーゼおよびMRCKαによる調節がLPS誘発性ALIの治療に寄与するかどうかをさらに調べるために、血液から気道へのエバンスブルー色素の漏出によって肺透過性を測定した。内皮および/または上皮バリアの破壊により、LPSに応答した肺漏出は、ナイーブマウスと比較して3倍~4倍増加した(図21)。LPS注入後の対照プラスミドpcDNA3の導入は、肺漏出に変化をもたらさず、またエレクトロポレーションの非存在下でのPBSの投与も変化をもたらさなかった。β1-Na+/K+-ATPアーゼの遺伝子導入は、MRCKα単独またはβ1サブユニットと組み合わせた遺伝子導入と同様に、LPS単独と比較して、以前にLPSによって誘発された肺漏出を著しく減少させた。まとめると、これらの結果は、MRCKαの遺伝子導入が肺漏出の阻害に重要な役割を果たし、したがって内皮および/または上皮バリア(複数可)を機能的に増強することを示唆している。
以前の研究では、エレクトロポレーションを介したβ1-Na+/K+-ATPアーゼ遺伝子導入により、肺胞液クリアランス(AFC)が単離されたラットの肺で74%、マウスの肺で43%増加したことが報告されている。培養細胞における結果は、β1Na+/K+-ATPアーゼによるMRCKαの活性化を示しているが、この活性化が双方向であるかどうか、すなわちMRCKαもまたβ1Na+/K+-ATPアーゼを活性化することができ、AFCの増加をもたらすかどうかを調べるためにアッセイが行われた。

(i)実施形態15に記載の核酸分子または核酸分子のセット、実施形態16に記載のベクター、実施形態17に記載の宿主細胞、または実施形態18に記載のウイルスもしくはウイルス様粒子、および
(ii)薬学的に許容される担体または賦形剤、を含む、医薬組成物。
(実施形態20)
実施形態15に記載の核酸分子または核酸分子のセット、実施形態16に記載のベクター、実施形態17に記載の宿主細胞、および実施形態18に記載のウイルスまたはウイルス様粒子のうちの1つ以上を含む、キット。
Claims (12)
- 上皮または内皮バリアの完全性または機能を改善するための組成物であって、筋緊張性ジストロフィーキナーゼ関連Cdc42結合キナーゼα(MRCKα)ポリペプチドまたはMRCKαポリペプチドをコードする第1の核酸を含み、
バリアの1つまたは複数の細胞におけるMRCKαのレベルが、MRCKαポリペプチドまたはMRCKαポリペプチドをコードする第1の核酸を前記1つまたは複数の細胞に導入することによって増加する、組成物。 - 前記上皮バリアが肺胞上皮バリアである、請求項1に記載の組成物。
- MRCKαのレベルが、MRCKα遺伝子の酵素レベルまたは発現レベルである、請求項1または2に記載の組成物。
- Na+/K+-ATPアーゼ(NKA)β1サブユニットポリペプチドまたはNKA β1サブユニットポリペプチドをコードする第2の核酸をさらに含み、
前記1つまたは複数の細胞におけるNKA β1サブユニットのレベルが、NKA β1サブユニットポリペプチドまたはNKA β1サブユニットポリペプチドをコードする第2の核酸を前記1つまたは複数の細胞に導入することによって増加する、請求項1~3のいずれか一項に記載の組成物。 - NKA β1サブユニットのレベルが、NKA β1遺伝子の活性レベルまたは発現レベルである、請求項4に記載の組成物。
- 前記細胞が肺胞上皮細胞である、請求項1~5のいずれか一項に記載の組成物。
- 細胞がインビトロである、請求項1~6のいずれか一項に記載の組成物。
- 前記細胞が対象のインビボである、請求項1~6のいずれか一項記載の組成物。
- 前記第1の核酸または前記第2の核酸が発現ベクター中にある、請求項1または4に記載の組成物。
- それを必要とする対象において上皮または内皮バリアの機能不全に関連する疾患または状態を治療するための医薬であって、MRCKαポリペプチドまたはMRCKαポリペプチドをコードする第1の核酸を含み、
上皮または内皮バリアの1つまたは複数の細胞におけるMRCKαのレベルが、MRCKαポリペプチドまたはMRCKαポリペプチドをコードする第1の核酸を前記1つまたは複数の細胞に導入することによって増加する、医薬。 - Na+/K+-ATPアーゼ(NKA)β1サブユニットポリペプチドまたはNKA β1サブユニットポリペプチドをコードする第2の核酸をさらに含み、
