本発明により使用される希土類イオンをドープした結晶性マトリックスを有するフォトルミネッセンスナノ粒子は、当業者に知られている任意の従来の方法によって調製することができる。
ナノ粒子の結合試薬への結合
ラテラルフローアッセイデバイスで従来使用されているプローブと同様に、本発明により使用されるルミネッセンスナノ粒子は、分析される物質に特異的な少なくとも1つの結合試薬に結合される。
1つまたは複数の結合試薬に結合された粒子を適切に調製するために、結合/グラフトの適切な方法を使用することは、当業者次第である。ルミネッセンスナノ粒子の表面官能化に使用される結合試薬の量は、粒子の量を考慮して調整される。
また、本発明のルミネッセンスナノ粒子は、その表面の電荷および性質、形状およびサイズに応じて、毛細管現象試験デバイス内、例えばニトロセルロース膜内でのルミネッセンスナノ粒子の移動を容易にする、以下「移動剤」と呼ばれる薬剤でコーティングすることができる。
上記のように、上記で定義されたフォトルミネッセンスナノ粒子は、本発明によれば、毛細管現象試験、例えば「ラテラルフロー」アッセイにおけるプローブとして使用される。
-液体サンプル、および場合により希釈剤を沈着させるためのゾーン(1)、
-沈着ゾーンの下流に配置され、「標識ゾーン」と呼ばれ、分析される物質に特異的に結合する少なくとも1つの試薬に結合された本発明によるフォトルミネッセンス無機ナノ粒子(プローブ)が担持されたゾーン(2)、
-標識ゾーン(2)の下流に配置され、「検出ゾーン」とも呼ばれ、分析される物質に特異的な少なくとも1つの捕捉試薬が固定化される、反応ゾーン(3)、
-検出ゾーン(3)の下流に位置し、分析物に特異的に結合する試薬に特異的な少なくとも1つの第2の捕捉試薬が固定化される、対照ゾーン(4)を含む。
(i)毛細管現象試験デバイスの沈着ゾーン(1)のレベルで、分析される液体サンプル、および場合により希釈剤を適用すること、
(ii)フォトルミネッセンスナノ粒子によって生成されたルミネッセンスが反応ゾーン(3)で検出されるまで、および/またはルミネッセンスが移動対照ゾーン(4)で検出されるまで、デバイスをインキュベートすること、
(iii)結果を読んで解釈することを含む。
従来の毛細管現象試験と同様に、本発明の方法により実施される試験(検出および/または定量化)の評価は、検出ゾーン、および場合により対照ゾーンを観察することによって行われる。
本発明による毛細管現象試験デバイスの検出ゾーンおよび対照ゾーンの観察は、より具体的には、(i)フォトルミネッセンスナノ粒子の励起のステップおよび(ii)ルミネッセンス発光の検出のステップを使用する。
上記で定義された本発明による毛細管現象試験デバイス、および
デバイスの検出ゾーン、および場合により対照ゾーンのレベルで固定化されたプローブにより生成されたルミネッセンスを検出するためのデバイスを含む。
次に、ルミネッセンス結果を解釈することができる。
・2つのバンドが存在する場合、試験は陽性である;
・単一の対照バンドが存在する場合、試験は陰性である;
・単一の試験バンドが存在する場合、試験は無効である;
・バンドが存在しない場合、試験は無効である。
1.フォトルミネッセンスプローブの調製
1.1.フォトルミネッセンス無機ナノ粒子の合成
1.1.a Y0.6Eu0.4VO4ナノ粒子の合成
メタバナジン酸アンモニウムNH4VO3をメタバナジン酸イオンVO3 -の供給源として使用し、オルトバナジン酸VO4 3-は、塩基、この場合は水酸化テトラメチルアンモニウム、N(CH3)4OHとの反応後にその場で得られる。Y3+およびEu3+イオンの供給源として、硝酸イットリウムおよび硝酸ユウロピウムを使用した。
ボトル内で16時間放置した後のナノ粒子の溶液の目視観察は、均一に拡散する溶液を示している。
合成は、0.1Mのイオン(Y3++Dy3+)でY(NO3)3およびDy(NO3)3からなる溶液2を除いて、例1.1.a.の合成と同じである。溶液2を、シリンジポンプで溶液1に1mL/分の流量で滴下する。
合成は、0.1Mのイオン(Y3++Sm3+)でY(NO3)3およびSm(NO3)3からなる溶液2を除いて、例1.1.a.の合成と同じである。溶液2を、シリンジポンプで溶液1に1mL/分の流量で滴下する。
合成は、0.1Mのイオン(Lu3++Eu3+)でLu(NO3)3およびEu(NO3)3からなる溶液2を除いて、例1.1.a.の合成と同じである。溶液2を、シリンジポンプで溶液1に1mL/分の流量で滴下する。
合成は、0.1Mのイオン(Lu3++Dy3+)でLu(NO3)3およびDy(NO3)3からなる溶液2を除いて、例1.1.a.の合成と同じである。溶液2を、シリンジポンプで溶液1に1mL/分の流量で滴下する。
合成は、0.1Mのイオン(La3++Eu3+)でLa(NO3)3およびEu(NO3)3からなる溶液2を除いて、例1.1.a.の合成と同じである。溶液2を、シリンジポンプで溶液1に1mL/分の流量で滴下する。
これらのナノ粒子の合成は、オルトバナジン酸前駆体から出発して、以下のように行われる。0.1MのNaVO4の10mL水溶液(溶液1)を新たに調製し、1MのNaOH溶液でpHを12.6~13に調整する。
異なるVO4 3-:PO4 3-比でマトリックス中にVO4 3-およびPO4 3-イオンの混合物を含むナノ粒子も合成した。
ポイント1.1.a.に記載したように得られたY0.6Eu0.4VO4ナノ粒子は、以下のプロトコルにより抗体に結合される。
ナノ粒子の合成の終了時に、ナノ粒子の溶液を17000gで3分間遠心分離して、ナノ粒子の凝集体を沈殿させ、上清を回収する。サイズによる選択を行う。この目的のために、1900gで3分間の遠心分離を数回行う。各遠心分離に続いて、超音波処理器でナノ粒子を再分散させ、次いで、ナノ粒子のサイズをDLS-ゼータ電位装置(Zetasizer Nano ZS90、Malvern社)を使用して決定する。
3つ口タイプの500mLフラスコに225mLの無水エタノールを入れ、最終濃度1.125mMに相当する265μLのAPTES(3-アミノプロピルトリエトキシシラン)(分子量221.37g/mol、Sigma Aldrich社)を添加する。この量は、導入されたバナジン酸の5当量に相当する。次いで、コンデンサをフラスコに取り付ける。全体をフラスコヒーターに配置し、フードの下に置く。混合物を90℃の還流下で加熱する。3つ口フラスコの入口の1つに、pH9で75mLの水中ナノ粒子のコロイド溶液(バナジン酸イオンの濃度[V]=3mM)を、蠕動ポンプを使用して1mL/分の流量で滴下する。全体を撹拌しながら24時間加熱する。
グラフト化を開始する前に、溶媒の移動を行う。
COOHで表面グラフト化されたナノ粒子の結合は、以下のプロトコルにより行う。
1.MES緩衝液(pH5~6)中のEDC/Sulfo-NHS(それぞれ濃度500mg/mLおよび500mg/mL)の混合溶液を新たに調製する。
2.事前に調製した3mLの溶液に、90nMのNP(この場合は参考文献Casanovaら[37]により、バナジン酸イオンの濃度から算出したナノ粒子の濃度)を添加し、室温で25分間、撹拌しながら反応させる。
3.NPを少なくとも2回、MilliQ水で遠心分離(13000gで60分間、Legend Micro 17R、Thermo Scientific社)ですばやく洗浄し、過剰な試薬を除去する。
4.pH7.3のリン酸ナトリウム緩衝液で超音波処理した後、最後のペレットを回収する。必要な量のタンパク質(抗h-FABP抗体、参照4F29、10 E1、Hytest社)を、必要な比率(タンパク質:Np)の関数として、典型的には20:1の比率のために2μM、および5mg/mLのmPEG-シラン(MW:10 kD、Laysan Bio 256-586-9004)を添加する。
5.この溶液を撹拌しながら室温で2~4時間反応させる。
6.ブロッキング剤(1%のグリシン)を添加して、遊離COOHと反応させ、NPの表面の残留反応部位をブロックする。30分間反応させる。
7.タンパク質に結合したNPを数回、pH7.2のPBSで遠心分離フィルタ(Amicon Ultra 0.5mL、Ref UFC501096、Millipore社)を使用して遠心分離することにより、洗浄する。保存媒体であるリン酸緩衝液+Tween20(0.05%)+0.1%グリシン+10%グリセロールにNPを移す。BCAアッセイには100μLを使用する。残りの溶液をアリコートに分け、-80℃で凍結する。
例1.2の共有結合の代わりに、ナノ粒子の抗体への受動的結合も以下のように行うことができる。
・1mLのナノ粒子(5mMのバナジン酸イオンの濃度)の溶液を15000gで15分間遠心分離する。
・ペレットを800μLのMilliQ水に取り、超音波処理(Bioblock Scientific社、Ultrasonic Processor、最大出力130W、50%で40秒間動作)によって再分散させる。
・リン酸カリウム緩衝液2mM pH7.4に250μg/mLの抗体溶液100μLを添加する。
・回転させながら1時間インキュベートする。
・100μLのリン酸カリウム緩衝液20mM pH7.4/1%BSAを添加する。
・15000gで15分間遠心分離し、上清を除去する。
・ペレットを1mLのリン酸カリウム緩衝液2mM pH7.4/0.1%BSAに取る。超音波処理(Bioblock Scientific社、Ultrasonic Processor、最大出力130W、50%で40秒間動作)によって再分散させる。
・15000gで15分間遠心分離し、上清を除去する。
・ペレットを250μLのリン酸カリウム緩衝液2mM pH7.4/0.1%BSAに取る。超音波処理(Bioblock Scientific社、Ultrasonic Processor、最大出力130W、50%で40秒間動作)によって再分散させる。
タンパク質の存在を定性的または定量的に決定するための迅速なテストを開発するには、様々なパラメーターを最適化し、反応時間と試験の感度の間の妥協点を見つける必要がある。
・本質的に不活性なガラス繊維を標識ゾーン(2)(英語の用語では「コンジュゲートパッド」)(GFDX 103000、Millipore社)として使用する。
・沈着ゾーン(英語の用語では「サンプルパッド」)(1)(Ref CFSP173000、Millipore社)として表面修飾ポリエステルを使用する。それらには、タンパク質との弱い非特異的相互作用、優れた牽引力、および優れた取り扱い特性を有するという利点がある。
・ニトロセルロース膜(NC)(HF180MC100、Millipore社)を毛細管現象の手段(10)として使用する。ニトロセルロース膜は、液体の移動およびタンパク質の固定化に最適な特性を備えている。NC膜は、無孔接着プラスチック(「バッキングカード」)のサポート(6)に接着されている。
・セルロース(CFSP173000、Millipore社)を、吸収能力が高いため、吸収パッド(5)として使用する。
様々なコンポーネントを組み立てる前に、NC膜に抗体を沈着させる必要がある。
1.Abがすでに固定化されているNCの接着部分に、様々な構造(吸収パッドおよび沈着ゾーンとして機能するセルロース、Ab+NPが沈着される標識ゾーン)を組み立てる。コンポーネントは、NCのプラスチックサポートに、標識ゾーン(「コンジュゲートパッド」)、沈着ゾーン(「サンプルパッド」)、最後に吸収パッドの順序で固定させる。毛細管現象による流体の移動を改善するために、図に示すように(図1)、互いに重なり合うように様々なコンポーネントを取り付ける。
ストリップは、例1.2で調製した抗体に結合されたY0.6Eu0.4VO4ナノ粒子を使用して調製する。
結果の定性的解釈は次の通りである:
・2つのバンドが存在する場合、試験は陽性である;
・単一の対照バンドが存在する場合、試験は陰性である;
・単一の試験バンドが存在する場合、試験は無効である;
・バンドが存在しない場合、試験は無効である。
図10は、専用アプリケーションを使用して携帯電話で撮影したデジタル写真から開始する定量分析の例を示している。携帯電話の画面の「捕捉」を押すと、白黒画像の記録が開始される。次に、「測定」を押すと、アプリケーションにより、携帯電話の画面上で検出ゾーン、次いで対照ゾーンを押すよう指示される。これにより、アプリケーションは、検出ゾーンを含む長方形内の累積ルミネッセンスレベルLD、および対照ゾーンを含む同じ大きさの長方形内の累積ルミネッセンスレベルLCを計算する。「調整」を押すと、検出ゾーンおよび対照ゾーンに対応する2つの長方形の位置が最適化され、測定シグナルが最大化される。検出ゾーンと対照ゾーンとの間のスペースの中央に位置する同じサイズの長方形内の累積発光レベルは、バックグラウンドシグナルLBを決定し、シグナルSD/C=LD/C-LBを計算するのに役立つ。アプリケーションは、比率R=SD/SCまたはR=SD/(SD+SC)を計算して表示する。Rの値は、ng/mL単位の濃度値を提供するためのキャリブレーションテーブルと比較することができる。結果は「保存」機能で保存して、後で次の結果と比較することができる。
それぞれ例1.1.dおよび1.1.eにより合成したLu0.6Eu0.4VO4およびLu0.9Dy0.1VO4ナノ粒子を、例1.3によりストレプトアビジン(SA)に結合させた(受動的結合)。「ディップスティック」ストリップは、ストレプトアビジンに結合したNPを認識するために、BSA-ビオチンをニトロセルロース膜に固定化することにより、例2により調製した。テストストリップは、抗原の非存在下で、例3によるLu0.6Eu0.4VO4-SAおよびLu0.9Dy0.1VO4-SAナノ粒子を用いて作成した。ストリップは、紫外線ランプ励起(312nm)下で視覚化した。対照ラインのレベルで2つの明確で強いバンドが形成された(図22)。
Claims (25)
- 以下の式(II)のフォトルミネッセンス無機ナノ粒子をプローブとして使用する、毛細管現象試験によって、液体サンプル中の目的の生物学的または化学的物質を検出および/または定量化するためのインビトロ方法であって、
A1-xLnxVO4(1-y)(PO4)y(II)
式中、
Aは、イットリウム(Y)、ガドリニウム(Gd)、ランタン(La)、ルテチウム(Lu)、およびそれらの混合物から選択され、
Lnは、ユウロピウム(Eu)、ジスプロシウム(Dy)、サマリウム(Sm)、ネオジム(Nd)、エルビウム(Er)、イッテルビウム(Yb)、ツリウム(Tm)、プラセオジム(Pr)、ホルミウム(Ho)、およびそれらの混合物から選択され、
0.2≦x≦0.6、および
0≦y<1、
前記方法が、一光子吸収後、ナノ粒子の発光寿命が100ms未満のルミネッセンスを、320nm以下の波長でマトリックスを励起することによって検出することを使用する、方法。 - ルミネッセンスの検出が、300nm以下で構成される波長でマトリックスを励起することによって行われる、請求項1に記載の方法。
- 液体サンプルが生物学的サンプルである、請求項1または2に記載の方法。
- サンプル中の分子、タンパク質、核酸、毒素、ウイルス、細菌または寄生虫を検出および/または定量化するための、請求項1から3のいずれか一項に記載の方法。
- 前記フォトルミネッセンスナノ粒子が、5nm以上で厳密に1μm未満の平均サイズを有する、請求項1から4のいずれか一項に記載の方法。
- Lnが、Eu、Dy、Sm、Yb、Er、Ndおよびそれらの混合物から選択される、請求項1から5のいずれか一項に記載の方法。
- Aが、Y、Gd、Laおよびそれらの混合物から選択される、請求項1から6のいずれか一項に記載の方法。
- 前記ナノ粒子が、その表面にテトラアルキルアンモニウムカチオンを有し、前記ナノ粒子が、pH≧5の水性媒体中でζが-28mV以下と指定されたゼータ電位を有し、厳密にイオン伝導度が100μS・cm-1未満である、請求項1から7のいずれか一項に記載の方法。
- 前記ナノ粒子が、式A1-xLnxVO4(III)のものであり、式中、A、Ln、およびxが、請求項1、6、および7のいずれか一項に定義されている通りである、請求項1から8のいずれか一項に記載の方法。
- 前記フォトルミネッセンス無機ナノ粒子が、分析される物質に特異的に結合する少なくとも1つの試薬に結合されている毛細管現象試験デバイスを使用する、請求項1から9のいずれか一項に記載の方法。
- 前記フォトルミネッセンス無機ナノ粒子が、毛細管現象試験デバイス内での移動を促進することを目的とする1つまたは複数の薬剤で表面上で官能化されている、毛細管現象試験デバイスを使用する、請求項1から10のいずれか一項に記載の方法。
- 前記液体サンプル、および場合により希釈剤を沈着させるためのゾーン(1)、
前記沈着ゾーンの下流に配置され、「標識ゾーン」と呼ばれ、分析される物質に特異的に結合する少なくとも1つの試薬に結合された前記フォトルミネッセンス無機ナノ粒子が担持された、ゾーン(2)、
前記標識ゾーン(2)の下流に配置され、「検出ゾーン」とも呼ばれ、前記分析される物質に特異的な少なくとも1つの捕捉試薬が固定化される、反応ゾーン(3)、

を含む毛細管現象試験デバイスを使用する、請求項1から11のいずれか一項に記載の方法。 - 前記方法が、少なくとも以下のステップ、
(i)前記毛細管現象試験デバイスの前記沈着ゾーン(1)のレベルで、分析される液体サンプル、および場合により希釈剤を適用すること、
(ii)前記フォトルミネッセンスナノ粒子によって生成された前記ルミネッセンスが前記反応ゾーン(3)で検出されるまで、および/または前記ルミネッセンスが移動対照ゾーン(4)で検出されるまで、前記デバイスをインキュベートすること、および
(iii)結果を読んで解釈すること、
を含む、請求項12に記載の方法。 - 前記毛細管現象試験の結果の読み取りが、アッセイの終了時に、毛細管現象試験デバイスのレベルで固定化されたプローブによって生成された前記ルミネッセンスを検出することによって行われる、請求項1から13のいずれか一項に記載の方法。
- 前記ナノ粒子が、式Y1-xEuxVO4(IV)のものであり、前記ルミネッセンスの検出が、230nm~320nmの波長でYVO4マトリックスを励起することによって行われる、請求項1から14のいずれか一項に記載の方法。
- 前記毛細管現象試験の結果の読み取りが、毛細管現象試験デバイスの直接の肉眼観察により行われる、請求項1から15のいずれか一項に記載の方法。
- 前記毛細管現象試験の結果の読み取りが、発光フィルタおよび光子検出器を含む検出装置を使用して行われる、請求項1から15のいずれか一項に記載の方法。
- 結果の解釈が、請求項12に定義された毛細管現象試験デバイスの検出ゾーン、対照ゾーンおよびバックグラウンドシグナルに対応するシグナルを決定し、前記バックグラウンドシグナルのルミネッセンスの値を差し引き、前記検出ゾーンからのシグナル対前記対照ゾーンからのシグナルの比を決定することを含む、請求項1から15および17のいずれか一項に記載の方法。
- 液体サンプル中の目的の生物学的または化学的物質を検出および/または定量化するのに有用な毛細管現象試験デバイスであって、前記デバイスは、プローブとして、以下の式(II)のフォトルミネッセンス無機ナノ粒子を含み、
