Description
本開示は、糖タンパク質CD8に特異的に結合する抗体および抗体の抗原結合フラグメント、これらの抗体、放射性標識された抗CD8抗体、蛍光標識された抗CD8抗体を使用する治療および診断方法、ならびにそれらの撮像における使用に関する。

特定の抗腫瘍療法の対象の適合性または対応性を予測および監視するための診断ツールに対するニーズがある。
CD8に結合するモノクローナル抗体およびその抗原結合フラグメントが、本明細書に提供される。抗体は、とりわけ、CD8を発現する免疫細胞を標的化するために、およびCD8陽性T細胞活性を調節するために有用であり得る。ある特定の実施形態では、抗体は、CD8陽性T細胞活性を阻害または中和するために有用であり、例えば、CD8陽性T細胞におけるIFNγ産生の阻害および/または活性化T細胞における転写因子アクチベータータンパク質(AP-1)の阻害に有用である。いくつかの実施形態では、抗体および抗原結合フラグメントは、インビボでのCD8結合に有用である。抗体は、CD8陽性T細胞活性化に関連する疾患または状態の処置に有用である。

精製された抗CD8 mAbに結合するhCD8α.mmhの平衡解離定数(KD値)を、リアルタイム表面プラズモン共鳴バイオセンサを用いて決定し、Sierra Sensor MASS-1を使用して、高容量アミンセンサ表面を、ポリクローナルヤギ抗マウスFc抗体(GE、#BR-1008-38)とのアミンカップリングにより誘導体化して、精製された抗CD8 mAbを捕捉した。SPR結合試験を、0.01MのHEPES(pH7.4)、0.15MのNaCl、3mMのEDTA、0.05%v/vの界面活性剤P20(HBS-ET泳動緩衝液)から構成される緩衝液中で実施した。HBS-ET泳動緩衝液中で調製されたC末端myc-myc-ポリヒスチジンタグ(hCD8α.mmh、REGN3940)を有するhCD8αの異なる濃度(300nM~3.7nM、3倍希釈物)を、50μL/分の流量で、抗CD8 mAbを捕捉した表面上に注入した。hCD8α.mmhの捕捉されたモノクローナル抗体への結合を4分間監視し、HBS-ET泳動緩衝液中のhCD8α.mmhの解離を10分間監視した。結合動力学的実験を、25℃で実施した。Scrubber 2.0c曲線適合ソフトウェアを用いてリアルタイムセンサーグラムを1:1結合モデルへフィッティングすることによって、動力学的会合(ka)および解離(kd)速度定数を決定した。結合解離平衡定数(KD)および解離半減期(t1/2)を、動力学的速度定数から以下のように計算した:
CD8抗体またはアイソタイプ対照抗体の一次ヒトCD8陽性T細胞およびカニクイザルT細胞への結合を評価するために、フローサイトメトリーを実施した。

mAb1およびアイソタイプ対照抗体を、200nMの濃度から始まる、ヒトCD8陽性T細胞には8点滴定で、またはカニクイザルT細胞には11点滴定でのいずれかにおいて、染色緩衝液(2%のFBSを含有するPBS)中で4倍連続希釈した。一次抗体を含まない、染色緩衝液のみの試料も対照として含まれた。抗体滴定物を、V底マイクロプレートに50ulウェルで播種した。一次ヒトおよびカ二ガニカルサルT細胞を、PBS中で1:1000に希釈されたLIVE/DEAD(商標)Fixable Violet Dead Cell染色(Invitrogen)を用いて15分間染色した。細胞を2回洗浄し、2%のFBSを含有するPBS中で再懸濁した。CD4+サルT細胞を取り除くために、カニクイザルCD4+T細胞と反応するBD BiosciencesからのCD4細胞を、サルT細胞と氷上で30分間インキュベートし、続いて染色緩衝液で1回洗浄した。染色緩衝液中のヒトCD8陽性およびサルT細胞は、約150,000個のT細胞を含有する細胞懸濁液50ulが、滴定抗体を含有する96ウェルV底マイクロプレートのウェルに添加されるように播種した。従って、抗体を2倍希釈し、それにより、最終濃度は、ヒトCD8陽性T細胞とインキュベートした抗体では100nM~24pMの範囲であって、またはサルT細胞とインキュベートした抗体では100nM~0.10pMの範囲であった。細胞を、氷上で30分間一次抗体とインキュベートし、染色緩衝液(2%のFBSで補充されたPBS)で二回洗浄し、二次アロフィコシアニン(APC)ヤギ抗マウスIgG抗体を、2μg/mLの濃度ですべてのウェルに添加し、氷上で30分間インキュベートした。次いで、試料を染色緩衝液で1回洗浄した、その後、染色緩衝液で1:1で希釈されたBD Cytofixに固定させた。固定緩衝液を除去した後、細胞を染色緩衝液中に再懸濁し、濾過し、その後、ベックマン・コールターCytoflexフローサイトメトリー測定器上で分析した。試料をFlowJo10ソフトウェアで分析し、その結果、抗体結合に対して生存可能なCD8陽性の単一細胞のみを評価した。APCの幾何学的MFIを決定し、抗体濃度に対してプロットし、GraphPad Prism(商標)において4パラメータロジスティック方程式を用いて、100nMの濃度から始まる、ヒトCD8陽性T細胞には8データ点に基づいて、またはカニクイザルT細胞には12データ点に基づいて、EC50値を決定した。
ヒトCD8陽性T細胞およびカニクイザルT細胞に結合するmAb1抗体のフローサイトメトリー分析。
mAb1のヒトおよびサルCD8に結合する能力を、フローサイトメトリーにより評価した(図1)。これらの実験では、無関係なアイソタイプと一致した抗体を、陰性対照として使用した。mAb1の用量依存的結合を、ヒトおよびサルCD8陽性T細胞の両方で観察した。mAb1は、25nMにおいて、アイソタイプ対照抗体と比較して、MFIのおおよそ2,778倍の増加を伴いヒトCD8陽性T細胞に対して0.37nMのEC50値を示した。mAb1は、0.33nMのEC50値でカニクイザルT細胞と結合し、25nMにおいて、アイソタイプ対照と比較してMFIのおよそ1,475倍の増加を示した。表3を参照されたい。アイソタイプ対照は、ヒトまたはサルT細胞のいずれかに対しても用量依存的結合を示さなかった。これらの結果は、mAb1が、ヒトおよびサルのCD8と交差反応し、両方の種のCD8に類似のEC50で結合することを示している。
T細胞は、T細胞受容体(TCR)が標的細胞上のMHC分子によって提示される外来抗原を特異的に認識するときに活性化される。この相互作用は、相互作用する標的細胞上の、MHCIIまたはMHCIの非可変領域にそれぞれ結合するT細胞上の共受容体(CD4およびCD8など)の存在によって強化され得る。さらに、これらの共受容体は、チロシンタンパク質キナーゼLckとのそれらの細胞質ドメインとの会合を通してT細胞活性を調節するのに直接的な役割を有する。共受容体とMHC分子との間の相互作用を妨害することは、T細胞活性に影響を与える可能性がある。CD8特異的抗体がT細胞活性を変化させるかどうかを識別するために、混合リンパ球反応(MLR)アッセイを採用した。MLRアッセイは、インビトロのT細胞を活性化する生理学的に妥当な手段である。一方向性MLRでは、1つの個体からの白血球を、別の遺伝子学的に異なる個体の増殖阻止白血球と共培養する。同種の決定因子の不適合性は、T細胞活性化につながり、これはサイトカインの産生および/または増殖によって評価され得る。サイトカインIFNγおよびIL-2、ならびに増殖は、一般に、CD4+T細胞活性のための読み出しとして使用される。しかしながら、CD8陽性エフェクターT細胞活性は、IFNγのそれらの産生によって最良に反映されるが、IL-2および増殖はバイスタンダー効果の結果であり得、活性化CD8陽性T細胞の割合に直接関連していないことが観察されている(Anthony et al.2012-Dissecting the T Cell Response:Proliferation Assays vs.Cytokine Signatures by ELISPOT-Cells,1,127-140)。
PBMCをヒト白血球パックから単離し、その後、陰性単離によって処理し、によって加工して、非付着のCD8陽性T細胞を得た。CD8陽性T細胞を使用して一方向性MLRアッセイを実施し、mAb1が、IFNγ産生によって示されるT細胞活性に影響を及ぼすかどうかを評価した。
ヒトPBMCを、NY血液センターから得られた健康なドナーからの末梢血の4つの白血球パックから単離した。PBMCの単離を、50mlのSepMate(商標)管を使用して、製造業者の推奨されるプロトコルに従って、密度勾配遠心分離によって達成した。簡潔に述べると、15mlのFicoll-Paque PLUSを50mlのSepMate(商標)管に層状に置き、その後、PBSで1:2で希釈した30mlの白血球を添加した。次のステップを、SepMate(商標)の製造業者のプロトコルに従って行った。単離されたPBMCの一部(>300×10^6)を、10%のDMSOを含有するFBS中で、バイアル当たり5000万個の細胞の濃度で凍結させた。PBMC単離後、Miltenyi BiotecからのヒトCD8 T細胞単離キットを、製造業者のプロトコルに従ってを使用して、CD8陽性T細胞を濃縮した。続いて、単離されたCD8陽性T細胞を、10%のDMSOを含有するFBS中で、バイアル当たり5000万個の細胞の濃度で凍結させた。PBMCおよびCD8陽性T細胞を、ベンゾナーゼヌクレアーゼを含有する、1次培養培地(10%のウシ胎児血清および0.01mMのベータ-メルカプトエタノールを補充したX-Vivo15培地)において、500Uのベンゾナーゼヌクレアーゼを含有する一次培養培地10ml当たり5000万個の濃度で、MLRアッセイセットアップの当日に解凍した。
一次細胞培養培地(125ul/ウェル)を、丸底マイクロタイタープレートの各ウェルに播種した。mAb1およびアイソタイプ対照の、400nMの濃度で始まる、3点の10倍連続希釈を、一次培養培地中で行った。これから、25ul抗体を、丸底マイクロプレートのウェルに三通りで播種した。抗体は、各ウェルの全容量の8分の1であり、最終抗体濃度50nM、5nM、および0.5nMにした。抗体を含まず、一次培養培地のみのウェルも、対照として含めた。同じ3人のドナーからの陰性単離されたCD8陽性T細胞およびこれらの同じ3人のドナーからのPBMC、並びについあのドナーをMLRアッセイで使用した。PBMCを、12×10^6細胞/mlの濃度で一次刺激培地中で50ug/mLまで希釈したマイトマイシンCで処理した。37℃/5%のCO2で1時間のインキュベーション後に、PBMCを50mlの円錐形管に集めて、一次細胞培養培地で合計3回洗浄した。これらの細胞を一次培養培地中の12×10^6細胞/mlの最終濃度に再懸濁し、25ulを丸底マイクロタイタープレートのウェルに添加し、ウェル当たり300,000個のPBMCの最終濃度を得た。さらに、PBMCを含まない、培地およびT細胞のみのウェルもまた、対照して含まれ、T細胞単独でIFNγを産生することができるかどうかを決定した。T細胞を、一次培養培地中で7×10^6細胞/mlの濃度に調製し、丸底マイクロタイタープレートのウェルに播種して、これにより、各ウェルにおけるT細胞の最終濃度は、175,000であった。PBMCの単独では、IFNγ産生に寄与しないことを確認するために、T細胞を含まない、培地のみのウェルもまた、対照としての役割を果たすために含まれた。1人のドナーのみのT細胞および1人のドナーのマイトマイシンC処置されたPBMCが、各ウェルに含まれた。3人のドナーのT細胞の各々は、それ自体のまたは異なるドナーのPBMCとペアリングした。37℃/5%のCO2での72時間のインキュベーション後に、マイクロタイタープレートを遠心分離して細胞をペレット化して、20ulの培地上清を収集した。収集した上清からの5ulを、製造業者のプロトコルに従ってヒトIFNγAlphaLISAアッセイで試験した。測定値を、マルチラベルプレートリーダーEnvision(PerkinElmer)上で取得した。生のRLU値をGraphPad Prism(商標)の棒グラフにプロットし、抗体を含有するウェルにおけるIFNγ産生量を、PBMCおよびT細胞のみを含有するウェルと比較し、IFNγ産生の阻害率として計算した。
mAB1がCD8 T細胞活性に影響を与える能力を、一方向性MLRにおけるIFNγ産生により測定した(図2)。これらの実験では、無関係なアイソタイプと一致した抗体を、対照として使用した。2つの異なるT細胞/PBMCペアについての結果および代表的画像は、mAb1が、用量依存的にIFNγ産生を減少させることができることを示している。この阻害の程度は、1人のドナー/PBMCペア(MLR反応1)が、IFNγの、10%の阻害を、5nMのmAb1処置において示し、一方別のドナー/PBMCペア(MLR反応2)が、IFNγの>50%の阻害を示すために、ドナー依存性であるように見える。両方の反応では、アイソタイプ対照は、5nMで、IFNγ産生に及ぼす最小の影響を有した。表4を参照されたい。
IFNγ阻害率の計算:
T細胞は、T細胞受容体(TCR)が、抗原提示細胞(APC)上で、主要組織適合性複合体(MHC)分子(ヒト白血球抗原(HLA)としても知られる)によって提示された外来性抗原を特異的に認識するときに活性化される。この相互作用は、相互作用するAPC上の、MHCIIまたはMHCIの非可変領域にそれぞれ結合するT細胞上の共受容体(CD4およびCD8など)の存在によって強化され得る。さらに、これらの共受容体は、チロシンタンパク質キナーゼLckとのそれらの細胞質ドメインとの会合を通してT細胞活性を調節するのに直接的な役割を有する(Goldrath et al.,Selecting and maintaining a diverse T cell repertoire,Nature 402:255-262,1999;Denkberg et al.Critical Role for CD8 in Binding of MHC Tetramers to TCR:CD8 Antibodies Block Specific Binding of Human Tumor-Specific MHC-Peptide Tetramers to TCR,The Journal of Immunology,2001,167:270-276;Cantrell et al.,T cell Antigen Receptor Signal Transduction,Immunology,2002,105.4:369-374;およびWang et al.2009)。
TCRシグナル伝達事象は、アクチベータ-タンパク質1(AP-1)、活性化T細胞の核因子(NFAT)または活性化B細胞の核因子カッパ軽鎖-エンハンサー(NFκB)などの様々な転写因子によって駆動されるレポーター遺伝子によって監視され得る(Shapiro et al.,Cutting Edge:Nuclear Factor of Activated T Cells and AP-1 Are Insufficient for IL-2 Promoter Activation:Requirement for CD28 Up-Regulation of RE/AP,The Journal of Immunology,1998,161(12):6455-6458)。
マウス線維芽細胞3T3細胞株をHLA-A*02対立遺伝子(受託番号P01892-1)およびヒトβ2-ミクログロブリン(hβ2M;受託番号NP_004039.1)と共に、NY-ESO-1 157-165、癌-精巣抗原NY-ESO-1に由来するHLA-A2*02拘束性ペプチド(受託番号NP_001318.1)を安定して過剰発現するように改変した。
開発されたバイオアッセイにおいて、改変されたAPC上のHLA-A2/NYESO1(157-165)MHCI/ペプチド複合体は、1G4 TCRに結合し、かつこれを刺激し(Robbins et al.,Single and Dual Amino Acid Substitutions in TCR CDRs Can Enhance Antigen-Specific T Cell Functions,J.Immunol.2008;180(9):6116-6131)、改変されたレポーターT細胞株においてAP-1の転写活性の増加をもたらす。AP-1は、次に、アッセイの読み出しとして使用される、ルシフェラーゼレポーター遺伝子の転写を活性化する。このバイオアッセイでは、CD8モノクローナル抗体を試験して、それらの遮断活性を評価した。
10%のFBSおよびP/S/Gで補充されたRPMI1640をアッセイ培地として用いて、細胞懸濁液および抗体希釈物を調製して、実験当日に抗CD8抗体のスクリーニングを実施した。

表5および図3は、mAb1および市販のクローンRPA-T8が、それぞれ、1.2nMおよび161pMのIC50で、改変されたT細胞におけるルシフェラーゼ活性を低下させることを示している。アイソタイプ1およびアイソタイプ2は、予想される用量依存的阻害を示していない。100nMでは、mAb1は、T細胞活性を約89.7%低下させ、一方でクローンRPA-T8は、ブ97.9%遮断する。クローンRPA-T8と比べると、mAb1は、CD8/MHCIの相互作用をより弱く遮断する。両方の抗体は、BiacoreおよびELISA実験において、ヒトCD8αサブユニットに結合することが示された。
凍結組織試料(Raji/PBMC腫瘍、マウス脾臓、および黒色腫組織)を、プロテアーゼ阻害剤を含む1×RIPA溶解緩衝液(Thermo Fisher Scientific)中で溶解した。組織を小片に切断し、密着したダウンス型ホモジナイザー中で1mLの溶解緩衝液で均質化した。溶解物を、超音波処理を10分ごとに30秒間行いながら、氷上で30分間インキュベートし、完全なタンパク質抽出物を得た。溶解物を、14,000gで10分間遠心分離した。タンパク質濃度をBCAアッセイにより測定した。各試料を1mg/mLに希釈し、14,000gで10分間遠心分離し、アリコートで-80℃にて保存した。
CD8α発現は、RBMC/Raji注入したマウスからの5個の腫瘍および脾臓、Rajiのみを注入したマウスの2個の腫瘍および脾臓、10個の黒色腫臨床試料、ならびに5個の黒色腫の正常な隣接組織(NAT)で分析した。組織重量、タンパク質量、抽出収率、およびCD8発現を、表6に列挙した。Bmaxを、1g/mLでの腫瘍密度の見積りで、以下の方程式に基づいて計算した。
親抗CD8抗体、mAb1(配列番号2/10のHCVR/LCVR配列ペアを有する)、およびアイソタイプ対照抗体を放射性標識を用いた免疫PET試験に好適であるように修飾するために、キレート化剤、p-SCN-bn-デフェロキサミン(DFO;Macrocylics、カタログ番号:B-705))を、これらの抗体に結合させた。
UV/VIS分光法によって示されるように、mAb1はリジンを介してDFOと首尾よくコンジュゲートされた。計算されたキレート部分対抗体比は、1.0~2.0の予想される範囲内であった。SECトレースは、検出可能な低分子量種を有さない97.5%の単量体生成物を示している。この結果は、還元状態および非還元状態の両方の電気泳動図によって裏付けられる。
免疫PETインビボ試験での使用に関しては、DFOとコンジュゲートされた抗CD8抗体、mAb1-L2を、89Zrで放射性標識した。

実施例7および8に従って調製された放射性標識された抗CD8抗体の免疫反応性(IR)を、以下のように決定した。すべての緩衝溶液/すすぎ溶液を、PBSおよび10%のウシ胎児血清(Seradigm、カタログ番号1500-500)で作製した。表13は、各IRアッセイに使用される細胞数を提供する。各アッセイについて、約107個のJRT3.T3/AP1-luc/hCD28/hCD8ΑB 1G4細胞を、0.5Mlの最終容量にさせた。20ngのそれぞれのDFO-Ab放射免疫コンジュゲートを、この溶液に添加し、チューブ回転装置上で連続的に混合しながら、インキュベータ(ThermoScientific、Forma Steri-Cycle CO2)内で、37℃、5%のCO2で45分間インキュベートした。その後、細胞を1500rpmで5分間遠心分離し、「細胞ペレットA」を作製した。上清(約0.5mL)を除去し、「細胞ペレットB」と呼ばれる、未処置細胞の別のペレットに導入し、37℃、5%のCO2で、45分間、再度インキュベートさせた。細胞ペレットBをインキュベートしている間に、細胞ペレットAを、1mLの新鮮な培地で3回すすぎ、1500rpmで5分間遠心分離した。それぞれのすすぎ物を回収し、後の分析のために保存した。45分間の細胞ペレットBのインキュベーション時間後に、これを1mLの新鮮な培地で3回すすぎ、1500rpmで5分間遠心分離した。再び、分析のためにすすぎ物を回収した。
89Zr-DFO-mAb1の投与およびPET/CT撮像
hCD8を発現する16週齢のマウスに、89Zr-DFO-mAb1を、0.5または1.5mg/kgのタンパク質用量で注射した。0.5mg/kgの用量で注射したマウスは、7μgの放射性標識されたmAb1-L2-20161115(約48μCi)および追加の8μgのDFOとコンジュゲートされていないmAb1(L1)を、最終の注入されたタンパク質用量を得るために補充として受容した。1.5mg/kgの用量で注射したマウスは、7μgの放射性標識されたmAb1-L2-20161115(約48μCi)および追加の38μgのDFOとコンジュゲートされていないmAb1(L1)を、最終の注入されたタンパク質用量を得るために補充として受容した。
生体分布試験のために、マウスを最終時点で(89Zr-DFO-mAb1投与から6日後)安楽死させて、心臓穿刺を介して血液を採取した。組織を切除し、計数管中に置き、秤量した。89Zrについての計数毎分(CPM)での計数データを、自動ガンマカウンター(Wizard 2470、Perkin Elmer)を使用して取得した。注入用量毎グラムのパーセント(%ID/g)を、注射材料から調製された標準を使用して、各試料について計算した。
この実験は、89Zr-DFO-mAb1の、hCD8を発現するマウスの脾臓およびリンパ節において内因性T細胞上で発現されるヒトCD8を標的とする能力を立証した。0.5mg/kgの投与された低タンパク質用量は、1.5mg/kg(10.32±1.54%ID/g)のより高い投与タンパク質用量と比べて、放射性トレーサー注射から6日後の血液からのより速い抗原媒介クリアランス(3.57±1.50%ID/g)を立証した。低投与タンパク質用量で注射したマウスの血液からのこのより速いクリアランスは、高い投与タンパク質用量で注射されたマウスよりも二次リンパ様器官における高い取り込みに寄与し、脾臓およびリンパ節で発現されたCD8に対する抗原特異的な標的化を立証している。hCD8を発現するマウスにおける、89Zr-DFO-mAb1の注射後6日目での生体分布からの%ID/g値を、表15に示している。hCD8を発現するマウスにおける、注射から6日後に0.5および1.5mg/kgの89Zr-DFO-mAb1の代表的iPET画像を図11に示す。
本実施例は、Raji細胞およびヒトPBMCで同時移植された雌NSGマウスにおけるジルコニウム-89標識されたDFO-抗CD8抗体コンジュゲートのインビボ撮像およびエクスビボ生体分布を記載している。
CD8標的化のための放射性標識された抗体の特異性を立証するために、2×106個のRaji細胞を単独で、または5×105個のヒトPBMC(ロット0160614、ReachBio Research Labs)と共に同時に、雌NSGマウス(8~10週齢、NOD.Cg-Prkdcscid II2rgtm1Wjl/SzJ;Jackson Labs)の右脇腹に移植した。腫瘍増殖を監視し、腫瘍移植から13~14日後に、マウスを、89Zr-DFO-mAb1投与に関して4つの群にランダム化した。RajiおよびRaji/hPBMC腫瘍は、89Zr-DFO-mAb1で投与されたとき、それぞれ、約335±68mm3および約371±40mm3であった。
皮下のRajiおよびRaji/hPBMC腫瘍を担持するマウスに、0.1mg/kg用量の89Zr-DFO-mAb1(約66μCiおよび2.8μgのタンパク質)を注射した。
生体分布試験については、89Zr-DFO-mAb1投与から6日後の最終PETスキャン後に、血液を心臓穿刺を介して採取した)。マウスを安楽死させ、次いでRajiおよびRaji/hPBMC腫瘍を他の正常組織と共に切除し、計数管に入れて計量した。89Zrについての計数毎分(CPM)での計数データを、自動ガンマカウンター(Wizard 2470、Perkin Elmer)を使用して取得した。注入用量毎グラムのパーセント(%ID/g)を、注射材料から調製された標準を使用して、各試料について計算した。
本試験は、89Zr-DFO-mAb1のs.c.において腫瘍内ヒトリンパ球上で発現されたCD8に対する抗原特異的標的化を立証している。NSGマウスにおいて、Raji/hPBMC腫瘍(31.11±8.82の%ID/g)をRaiのみの腫瘍(6.39±0.93%ID/g)と比較した。Raji/hPBMCおよびRajiのみの腫瘍対血液比は、それぞれ3.32±0.11および0.43±0.07であった。さらに、Raji/hPBMC腫瘍と同時移植したマウスの脾臓における取り込みが増加している。89Zr-DFO-mAb1注射から6日後のRajiおよびRaji/hPBMC腫瘍担持マウスの代表的なiPET画像(図12)は、89Zr-DFO-mAb1のRaji腫瘍担持マウスと比べて、Raji/hPBMC腫瘍担持マウスの腫瘍および脾臓に対するより高い標的化を立証している。89Zr-DFO-mAb1の注射後6日目での生体分布からの%ID/g値(表16)は、iPET撮像データを確証している。
抗体、mAb1のおよそ10mgを、製造業者の説明書に従って、事前調整されたNap-5カラム(GE Healthcare、カタログ番号17085302)を介して、配合緩衝液(ヒスチジンベース)から50mMの炭酸塩、pH8.4に緩衝液交換した。このプロセスを四通りで実施し、各溶出物(400μL)を収集し、合計1600μLに合わせた。合わせた溶出濃度を、UV/VIS分光法(Nanodrop 2000 UV/VIS分光計、Thermo Scientific、カタログ番号ND-2000c-US-CAN)により18.1mg/mLであると決定した。

- CD8に結合する単離されたモノクローナル抗体またはその抗原結合フラグメントであって、前記抗体またはそのフラグメントが、配列番号2の重鎖可変領域(HCVR)のアミノ酸配列内に含有されている3つの重鎖CDR(HCDR1、HCDR2、およびHCDR3)と、配列番号10の軽鎖可変領域(LCVR)のアミノ酸配列内に含有されている3つの軽鎖CDR(LCDR1、LCDR2、およびLCDR3)とを含む、単離されたモノクローナル抗体またはその抗原結合フラグメント。
- 請求項1に記載の単離されたモノクローナル抗体またはその抗原結合フラグメントであって、前記抗体またはそのフラグメントが、以下の特性:
(a)完全ヒトモノクローナル抗体であること、
(b)表面プラズモン共鳴によって測定されるとき、3.5×10-8M以下のKDでCD8に結合すること、
(c)ヒトCD8αに結合すること、
(d)活性化CD8T細胞においてIFNγ産生を阻害すること、

- 請求項8に記載の発現ベクターを含有する、宿主細胞。
- 請求項1に記載の抗体またはその抗原結合フラグメントと、陽電子放出体と、を含む、放射性標識された抗体コンジュゲート。
- 請求項1に記載の抗体またはその抗原結合フラグメントと、1または複数のキレート部分と、を含み、前記1または複数のキレート部分のうちの少なくとも1つは陽電子放出体を含む、放射性標識された抗体コンジュゲート。
- 前記キレート部分が、デスフェリオキサミンを含む、請求項11または12に記載のコンジュゲート。
- 前記陽電子放出体が、89Zrである、請求項10~12のいずれか一項に記載のコンジュゲート。
- 抗体またはその抗原結合フラグメントが、式(A)の1つ、2つ、または3つの部分に共有結合している、請求項10~15のいずれか一項に記載のコンジュゲート。
- 前記抗体が、それぞれ配列番号4、6および8の3つの重鎖CDR(HCDR1、HCDR2、およびHCDR3)のアミノ酸配列と、配列番号12、14および16の3つの軽鎖CDR(LCDR1、LCDR2、およびLCDR3)のアミノ酸配列とを含む、請求項10~16のいずれか一項に記載のコンジュゲート。
- 前記抗体が、
(a)表面プラズモン共鳴によって測定されるとき、約3.5×10-8M未満の結合解離平衡定数(KD)でヒトCD8に結合すること、
(b)ヒトCD8αに結合すること、
(c)活性化CD8T細胞でのIFNγ産生を阻害すること、
(d)活性化T細胞における転写因子アクチベータータンパク質(AP-1)を阻害すること、ならびに
(e)ヒトおよびサルCD8と交差反応することからなる群から選択される1つ以上の特性を有する、請求項10~17のいずれか一項に記載のコンジュゲート。 - 前記抗体が、配列番号2/10のHCVR/LCVRアミノ酸配列ペアを含む、請求項10~18のいずれか一項に記載のコンジュゲート。
- CD8を発現する組織の撮像方法における使用のための組成物であって、前記組成物は、請求項10~19のいずれか一項に記載の放射性標識された抗体コンジュゲートを含み、前記方法は、前記組成物を前記組織に投与することと、陽電子放出断層撮影(PET)撮像によりCD8発現を可視化することと、を含む、組成物。
- 固形腫瘍をチェックポイント阻害剤療法で処置するための方法における使用のための薬学的組成物であって、前記薬学的組成物は、請求項10~19のいずれか一項に記載の放射性標識された抗体コンジュゲートを含み、前記方法は、
(a)前記組成物を対象に投与し、固形腫瘍がCD8陽性T細胞を含むことを決定することと、
(b)前記腫瘍がCD8陽性T細胞を含む場合、前記チェックポイント阻害剤療法の1回以上の用量を前記対象に投与することと、を含む、薬学的組成物。 - 前記処置の工程(a)が、陽電子放出断層撮影(PET)撮像によって、前記腫瘍における前記放射性標識された抗体コンジュゲートの局在化の撮像を行うことと、をさらに含み、前記腫瘍における前記放射性標識された抗体コンジュゲートの存在が、前記腫瘍がCD8陽性細胞を含むことを示す、請求項21に記載の薬学的組成物。
- 前記処置が、0.1~10mg/kgの前記放射性標識された抗体コンジュゲートを投与することを含む、請求項22に記載の薬学的組成物。
- T細胞機能が、前記放射性標識された抗体コンジュゲートの投与によって損なわれない、請求項22に記載の薬学的組成物。
- 前記放射性標識された抗体コンジュゲートが、前記対象に対して皮下または静脈内に投与されることを特徴とする、請求項22または24に記載の薬学的組成物。
- PET撮像が、前記放射性標識された抗体コンジュゲートの投与の2~7日後に実施される、請求項22~25のいずれか一項に記載の薬学的組成物。
- 前記処置の工程(a)が工程(b)の前に実行される、請求項21~25のいずれか一項に記載の薬学的組成物。
- 前記処置が(c)前記対象を抗腫瘍療法の少なくとも1回の用量で処置した後に、工程(a)を繰り返すことをさらに含み、前記腫瘍における前記放射性標識された抗体コンジュゲートの局在化の面積のベースラインからの増加が、前記抗腫瘍療法の有効性を示す、請求項21~27のいずれか一項に記載の薬学的組成物。
- 前記組成物が、前記抗腫瘍療法を施した1~20週後に投与されることを特徴とする、請求項22に記載の薬学的組成物。
- 前記処置は、前記固形腫瘍がPD-1陽性であることを、放射性標識された抗PD-1コンジュゲートの投与を、それを必要とする前記対象に施すことにより決定する工程と、PET撮像によって、前記腫瘍における前記放射性標識された抗PD-1コンジュゲートの局在化を撮像する工程とを、さらに含み、前記腫瘍における前記放射性標識された抗PD-1コンジュゲートの存在が、前記腫瘍がPD-1陽性であることを示す、請求項21に記載の薬学的組成物。
- 前記抗腫瘍療法が、前記PD-1/PD-L1シグナル伝達軸の阻害剤、CTLA-4阻害剤、TIM3阻害剤、BTLA阻害剤、TIGIT阻害剤、CD47阻害剤、GITR阻害剤、別のT細胞共阻害剤もしくはリガンドの拮抗薬、インドールアミン-2,3-ジオキシゲナーゼ(IDO)阻害剤、血管内皮増殖因子(VEGF)拮抗薬、Ang2阻害剤、トランスフォーミング増殖因子ベータ(TGFβ)阻害剤、表皮成長因子受容体(EGFR)阻害剤、CD20阻害剤、腫瘍特異抗原に対する抗体、癌ワクチン、二重特異性抗体、細胞毒素、化学療法薬、シクロホスファミド、放射線療法、IL-6R阻害剤、IL-4R阻害剤、IL-10阻害剤、IL-2、IL-7、IL-21、IL-15、および抗体薬物コンジュゲート(ADC)からなる群から選択される、請求項21~30のいずれか一項に記載の薬学的組成物。
- 前記抗腫瘍療法が、REGN2810、BGB-A317、ニボルマブ、ピジリズマブ、ペンブロリズマブ、アテゾリズマブ、アベルマブ、デュルバルマブ、MDX-1105、REGN3504、イピリムマブ、抗CD-28抗体、抗2B4抗体、抗LY108抗体、抗LAIR1抗体、抗ICOS抗体、抗CD160抗体、抗VISTA抗体、アフリベルセプト、ベバシズマブ、ラニビズマブ、スニチニブ、ソラフェニブ、パゾパニブ、ネスバクマブ、エルロチニブ、セツキシマブ、リツキシマブ、抗CA9抗体、抗CA125抗体、抗黒色腫関連抗原3(MAGE3)抗体、抗癌胎児性抗原(CEA)抗体、抗ビメンチン抗体、抗腫瘍M2-PK抗体、抗前立腺特異抗原(PSA)抗体、抗ムチン-1抗体、抗MART-1抗体、抗CA19-9抗体、カルメット・ゲラン桿菌(Bacillus Calmette-Guerin)、CD3×CD20二重特異性抗体、PSMA×CD3二重特異性抗体、ダカルバジン、テモゾロミド、シクロホスファミド、ドセタキセル、ドキソルビシン、ダウノルビシン、シスプラチン、カルボプラチン、ゲムシタビン、メトトレキサート、ミトキサントロン、オキサリプラチン、パクリタキセル、ビンクリスチン、シクロホスファミド、放射線療法、サリルマブ、デュピルマブ、抗CD19-DM4 ADC、および抗DS6-DM4 ADCからなる群から選択される、請求項21~30のいずれか一項に記載の薬学的組成物。

- 前記PD-1/PD-L1シグナル伝達軸の前記阻害剤が、抗PD-1抗体またはその抗原結合フラグメントである、請求項31に記載の薬学的組成物。
- 前記抗PD-1抗体またはその抗原結合フラグメントが、REGN2810、ノボルマブ、またはペンブロリズマブである、請求項34に記載の薬学的組成物。
- 前記抗PD-1抗体またはその抗原結合フラグメントが、REGN2810である、請求項34に記載の薬学的組成物。
- 前記PD-1/PD-L1シグナル伝達軸の前記阻害剤が、抗PD-L1抗体またはその抗原結合フラグメントである、請求項31に記載の薬学的組成物。

前記対象に前記組成物を投与して、前記固形腫瘍におけるCD8陽性細胞の存在を決定することを含み
前記CD8陽性細胞の存在が、抗腫瘍療法に対する陽性応答を予測する、薬学的組成物。 - 抗腫瘍療法に対する対象における腫瘍の応答を監視することにおける使用のための薬学的組成物であって、前記薬学的組成物が、請求項10~19のいずれか一項に記載の放射性標識された抗体コンジュゲートを含み、前記監視することが、
(a)前記対象に対して抗腫瘍療法の1回以上の用量を投与することと、
(b)前記抗腫瘍療法を施した1~20週後に、前記対象に対して放射性標識された抗CD8抗体コンジュゲートの少なくとも1回の用量を投与して、前記固体腫瘍におけるCD8陽性細胞の存在を決定することと、を含み、
前記CD8陽性細胞の存在が、前記抗腫瘍療法に対する陽性応答を示す、薬学的組成物。 - CD8陽性T細胞を、腫瘍を有する対象における抗腫瘍療法の有効性の指標として定量化する方法における使用のための、請求項10~19のいずれか一項に記載の放射性標識された抗体コンジュゲートを含む薬学的組成物であって、前記方法が、
前記組成物を、前記腫瘍を有する対象に第1の時点で投与し、前記腫瘍におけるCD8陽性T細胞の存在を決定することと、
前記CD8陽性T細胞のレベルを、成功した抗腫瘍療法と相関させることとを含み、
ある特定の閾値を超える高レベルが、成功した抗腫瘍療法の予測または指標である、薬学的組成物。 - 経時的な腫瘍におけるT細胞の存在を監視する方法における使用のための薬学的組成物であって、前記薬学的組成物が、請求項10~19のいずれか一項に記載の放射性標識された抗体コンジュゲートを含み、前記方法が、
(a)前記組成物を、前記腫瘍を有する対象に第1の時点で投与して、前記腫瘍におけるCD8陽性T細胞の存在を決定することと、
(b)前記対象に対して抗腫瘍療法の1回以上の用量を投与することと、
(c)前記抗腫瘍療法を施した1~20週後に、前記対象に第2の時点で前記組成物を投与して、前記腫瘍におけるCD8陽性T細胞の存在を決定することと、を含み、
前記腫瘍におけるT細胞の存在が、前記抗腫瘍療法に対する陽性応答を示す、薬学的組成物。 - 前記処置の工程(c)が、前記抗腫瘍療法による処置過程にわたって繰り返される、請求項43に記載の薬学的組成物。
- 前記第1の時点が、前記処置の工程(b)の前に起こる、請求項43に記載の薬学的組成物。
- 工程(a)による前記CD8陽性T細胞が、工程(c)による前記CD8陽性T細胞に対して比較され、CD8陽性T細胞の経時的な増加が、前記抗腫瘍療法に対する陽性応答を示す、請求項43に記載の薬学的組成物。
- kが、1または2である、請求項47に記載の化合物。
- (i)CD8に結合する抗体またはその抗原結合フラグメントと、(ii)1つ以上のキレート部分と、を含む、抗体コンジュゲートであって、前記抗体またはその抗原結合フラグメントが、配列番号2の重鎖可変領域(HCVR)のアミノ酸配列内に含まれる3つの重鎖CDR(HCDR1、HCDR2およびHCDR3)、ならびに配列番号10の軽鎖可変領域(LCVR)のアミノ酸配列内に含まれる3つの軽鎖CDR(LCDR1、LCDR2およびLCDR3)を含む、抗体コンジュゲート。
- 前記コンジュゲートが、1.0~2.0のキレート部分対抗体比を有する、請求項49に記載の抗体コンジュゲート。
- 前記キレート部分対抗体比が、約1.7である、請求項49に記載の抗体コンジュゲート。
- (i)CD8に結合する抗体またはその抗原結合フラグメントと、(ii)蛍光染料と、を含む、抗体コンジュゲートであって、前記抗体またはその抗原結合フラグメントが、配列番号2の重鎖可変領域(HCVR)のアミノ酸配列内に含まれる3つの重鎖CDR(HCDR1、HCDR2およびHCDR3)、ならびに配列番号10の軽鎖可変領域(LCVR)のアミノ酸配列内に含まれる3つの軽鎖CDR(LCDR1、LCDR2およびLCDR3)を含む、抗体コンジュゲート。
- 前記蛍光染料が、近赤外染料である、請求項53に記載の抗体コンジュゲート。
