実験の手順を下記に示す。
機能性組成物を、主剤をTA、重合剤をトリス(2-アミノエチル)アミン、主剤Aと重合剤Bのモル比率をA:B=15:85として調製し、その機能性組成物2.5mLをドロップ-キャスト法により50mm×50mmのガラス基板に塗布して多孔性の機能性膜を形成した。したがって、この機能性膜の面密度は0.15mg/cm2である。
DPPH試薬(東京化成工業(株)製)は、アセトニトリルを溶媒として、0.16mmol/mg、0.33mmol/mgおよび0.67mmol/mgの三種類の濃度の溶液を用意した。それぞれの溶液のDPPHの量は、12μM、25μMおよび50μMである。
そして、それぞれのDPPH溶液50mLを別々のビーカーに入れ、上記機能性膜が形成された試料を24時間浸漬させた。その間、各DPPH溶液の波長520nmにおける光吸収を測定した。ここで、光吸収測定には、V-670(ASCO製)を用いた。

In the meantime, the light absorption at a wavelength of 520 nm of each DPPH solution was measured. Here, V-670 (manufactured by ASCO) was used for optical absorption measurement.

図11は、図8から図10のデータを基に作成した図で、波長520nmにおける吸光度の初期値比α/α0の経過時間依存性を示す。吸光度の初期値比α/α0は経過時間とともに単調に減少し、DPPH溶液の濃度が0.16mmol/mgの場合は約6時間でその減少は飽和する。DPPH溶液の濃度が0.33mmol/mgおよび0.67mmol/mgの場合は、その減少は24時間で飽和に近づく。
このことから、この機能性膜は十分高い抗酸化機能を有することが実証された。
なお、参考までに、機能性膜をDPPH溶液に24時間浸漬させた後のDPPH溶液吸光度初期値比α/α0のDPPH濃度依存性を図12に示す。DPPH溶液吸光度初期値比α/α0はDPPH濃度とリニアな関係にある。

From this, it was demonstrated that this functional film has a sufficiently high antioxidant function.
For reference, FIG. 12 shows the DPPH concentration dependence of the DPPH solution absorbance initial value ratio α/ α0 after the functional film was immersed in the DPPH solution for 24 hours. The DPPH solution absorbance initial value ratio α/ α0 has a linear relationship with the DPPH concentration.

実施例4は、機能性膜の滅菌・抗菌作用について調べた結果を示す。
フィルム密着法により、本発明の機能性膜の滅菌・抗菌効果評価を行った。
被検菌は、大腸菌、黄色ブドウ球菌およびメシチリン耐性黄色ブドウ球菌(MRSA)で、機能性膜としては、主剤をTA、重合剤をトリス(2-アミノエチル)アミン、主剤Aと重合剤Bのモル比率をA:B=15:85として作製された膜を用いた。
具体的には、ガラス基板に上記機能性膜を塗布形成した後、ピーリングして作製したフィルム試料と、コントロールとして何も塗布しないフィルムによる非処理フィルム試料とを準備し、各フィルム試料を大腸菌を培養させた培養体に接触させて、大腸菌に対する殺菌作用を評価した。その評価の方法の詳細はJIS2801のフィルム試験法に準拠させ、各フィルム試料の培養体への接触時間は24時間とした。また、菌を黄色ブドウ球菌やMRSAに換えて同様の測定を行った。その結果を下記表1に示す。ここで、初期値は、培養体の細菌数(細菌コロニー数)である。各々3サンプルにつき測定を行った。

The details of the evaluation method conformed to the film test method of JIS2801, and the contact time of each film sample with the culture medium was 24 hours. In addition, similar measurements were performed by replacing the bacteria with Staphylococcus aureus or MRSA. The results are shown in Table 1 below. Here, the initial value is the number of bacteria in the culture (the number of bacterial colonies). Three samples were measured for each.

最初に、主剤をTA、重合剤をトリス(2-アミノエチル)アミン、主剤Aと重合剤Bのモル比率をA:B=15:85としてガラス基板上に形成した機能性膜を準備した。そして、その機能性膜の一部の水酸基をPCL(東京化成工業(株)製)で置換した試料とDFU(ALDRICH製)で置換した疎水化処理試料を作製した。ここで、PCLおよびDFUの置換は、溶媒をジエチルエーテルとしたPCL溶液およびDFU溶液を準備し、それらの溶液に機能性膜を形成した試料を室温(25℃)で2時間浸漬して行った。PCL溶液の濃度は0.6,1.2,2.4および4.9μmol/mLの4種類、DFU溶液の濃度は0.6,1.2,2.4および4.8μmol/mLの4種類計8種類とした。

ここで、水の接触角測定にはDrop Master DMs-401(協和界面科学(株)製)を用い、測定は大気圧下で行った。また、コントラストの高い水滴像を得るために、水には濃度0.1mMでローダミンBを添加した。水の滴下にはマイクロシリンジを用い、1滴当たりの水の滴下量は2.0μLとした。
また、参考までに、ガラス基板上に濃度1.2μmol/mLのPCLを塗布し、その後乾燥を行った試料と、1.2μmol/mLのDFUを塗布し、その後乾燥を行った計2つの試料を別に準備し、その水の接触角も上記と同様に測定した。

For reference, a total of two samples were prepared by applying PCL at a concentration of 1.2 μmol/mL on a glass substrate and then drying, and by applying DFU at a concentration of 1.2 μmol/mL and then drying. was prepared separately, and its contact angle with water was measured in the same manner as above.

その結果、PCLで表面処理した場合もDFUで表面処理した場合も高い水の接触角が得られ、特にPCLやDFUの濃度を1.2μmol/mLとした場合には、PCLが126.8±0.8°、DFUが130.8±0.9°と共に120°を超える水の接触角となった。また、DFUを用いた場合は、0.6μmol/mLから4.8μmol/mLに至るまで水の接触角は127.7±2.4°から132.0±2.2°の範囲にあり、ほぼ濃度に依存せず130°付近の高い水の接触角が得られた。
一方、ガラス基板上に直接PCLを塗布形成した場合の水の接触角は62.7±1.6°であり、DFUのそれは74.1±2.9°と本発明の多孔を有する機能性膜を用いた場合より大幅に低い接触角であった。また、PCLやDFUで表面処理をしていない本機能性膜での水の接触角は約0°であった。
この結果、本発明の機能性膜は、PCLやDFUによる表面処理により、水の接触角で120°を超える高い撥水性を得ることができることが実証された。これは、PCLやDFUによる疎水性付与と機能性膜の多孔に伴う凹凸形状効果の相乗効果によると考えられる。

The contact angle was significantly lower than with the membrane. In addition, the contact angle of water on this functional film, which was not surface-treated with PCL or DFU, was about 0°.
As a result, it was demonstrated that the functional film of the present invention can obtain high water repellency with a water contact angle exceeding 120° by surface treatment with PCL or DFU. This is considered to be due to the synergistic effect of the hydrophobicity imparted by PCL or DFU and the uneven shape effect associated with the pores of the functional film.
