NK-92細胞株は、非ホジキンリンパ腫と診断された50歳男性の末梢血単核球(PBMC)から樹立されたIL-2依存性のヒトNK細胞株である(Gong, et al., Leukemia. 8:652-8 (1994))。NK-92細胞株は、CD56bright、CD2、CD7、CD11a、CD28、CD45、およびCD54表面マーカーを発現するが、CD1、CD3、CD4、CD5、CD8、CD10、CD14、CD16、CD19、CD20、CD23およびCD34マーカーを提示しない。正常なNK細胞とは異なって、NK-92はほとんどのキラー細胞抑制性受容体(KIR)の発現を欠いている(Maki, et al., J Hematother Stem Cell Res. 10:369-83 (2001))。全てのNK細胞によって発現される活性化機能と抑制能力を備えたKIR受容体であるKIR2DL4のみが、NK-92細胞の表面上に検出された。
本開示は、CD96の発現を阻害するCD96を標的とした変更を含むCD96改変型NK-92細胞を提供する。いくつかの態様では、CD96改変型NK-92細胞はCD96遺伝子の破壊によって作製される。CD96遺伝子を破壊する方法としては、限定するものではないが、ジンクフィンガーヌクレアーゼ(ZFN)、Taleエフェクタードメインヌクレアーゼ(TALEN)、およびCRISPR/Casシステムを利用する方法がある。

いくつかの態様において、本明細書では、例えば実施例のセクションに記載されるような比較実験で使用するために、CD226発現が低下するように遺伝的に改変された改変型NK-92細胞を提供する。いくつかの態様では、そのような改変型NK-92細胞は、CD226発現を低下または排除するためのCD226を標的とした変更を含む。そのような細胞は、CD96またはTIGITの発現が低下するように改変されたNK-92細胞に関して上述した技術のいずれかを用いて作製することができる。例示的な方法、およびそのような方法を用いて作製された細胞は、本出願の「実施例」のセクションに記載される。
Fc受容体
いくつかの態様では、CD96を標的とした変更またはTIGITを標的とした変更を含むNK-92細胞は、細胞表面上にFc受容体を発現するようにさらに改変される。例えば、さらなる改変型NK-92細胞がモノクローナル抗体と共に投与されるいくつかの態様では、Fc受容体は、ADCCを介して標的細胞を死滅させる抗体と協調してNK細胞が機能することを可能にする。いくつかの態様では、Fc受容体はIgG Fc受容体FcγRIIIである。
いくつかの態様では、CD96改変型NK-92細胞またはTIGIT改変型NK-92細胞は、細胞表面にキメラ抗原受容体(CAR)を発現するようにさらに操作される。任意で、CARは腫瘍特異的抗原に対して特異的である。腫瘍特異的抗原は、非限定的な例として、US 2013/0189268、WO 1999024566 A1、US 7098008、およびWO 2000020460 A1に記載されており、それらの各々は、その全体が参照により本明細書に組み入れられる。腫瘍特異的抗原としては、NKG2D、CS1、GD2、CD138、EpCAM、EBNA3C、GPA7、CD244、CA-125、ETA、MAGE、CAGE、BAGE、HAGE、LAGE、PAGE、NY-SEO-1、GAGE、CEA、CD52、CD30、MUC5AC、c-Met、EGFR、FAB、WT-1、PSMA、NY-ESO1、AFP、CEA、CTAG1B、CD19およびCD33が挙げられるが、これらに限定されない。さらなる非限定的な腫瘍関連抗原およびそれらに関連する悪性腫瘍は、表2に見出すことができる。
いくつかの態様では、本発明は、少なくとも1つのサイトカインを発現するようにさらに改変されたCD96改変型NK-92細胞およびTIGIT改変型NK-92細胞を提供する。そのような細胞では、細胞内のサイトカインの発現を小胞体に向かわせることができる。いくつかの態様では、少なくとも1つのサイトカインは、IL-2、IL-12、IL-15、IL-18、IL-21またはその変異体である。一態様では、サイトカインはIL-2である。特定の態様では、IL-2は、小胞体に標的指向される変異体である。
本開示には、上記のポリヌクレオチドまたはポリペプチド、例えば、Casタンパク質、CD16、Fc受容体、CAR、および/またはIL-2と顕著な配列同一性を共有する配列も含まれる。こうした配列もまた、非改変型NK-92細胞に導入することができる。いくつかの態様では、該配列は、それらのそれぞれの天然配列に対して少なくとも70%、少なくとも80%、少なくとも85%、少なくとも88%、少なくとも95%、少なくとも98%、または少なくとも99%の配列同一性を有する。
いくつかの態様では、本開示のCD96改変型またはTIGIT改変型NK-92細胞は、治療用抗体および/または他の抗がん剤と組み合わせて使用される。治療用抗体を用いて、感染している細胞またはがん関連マーカーを発現している細胞を標的とすることができる。がん治療用のモノクローナル抗体の例を表3に示す。
本明細書に記載のCD96改変型またはTIGIT改変型NK-92細胞で患者を治療する方法も提供される。いくつかの態様では、患者はがんまたは感染症に罹患している。上述したように、CD96改変型またはTIGIT改変型NK-92細胞は、患者のがん細胞の表面に発現される抗原を標的とするCARを発現するようにさらに改変され得る。いくつかの態様では、上記で説明したように、CD96改変型またはTIGIT改変型NK-92細胞はまた、Fc受容体、例えばCD16、を発現させることもできる。本明細書に開示するさらなる態様では、患者はCD96改変型またはTIGIT改変型NK-92細胞および抗体で治療される。

材料および方法
細胞培養
NK-92細胞を、5%ヒト血清(Valley Biomedical社, カタログ番号HP1022)および組換えヒトIL-2(500IU/ml;Prospec社, カタログ番号Cyt-209)を補充したX-VIVO 10培養液(Lonza社, カタログ番号BE04-743Q)中で維持した。MCF-7、SKBR-3およびDaudi細胞株は、アメリカン・タイプ・カルチャー・コレクション(ATCC, Rockville, MD)から購入して、10%FBS(Gibco社, カタログ番号10438026)および1%ペニシリン/ストレプトマイシン(Gibco社, カタログ番号15070-063)を補充したRPMI-1640培養液(ThermoScientific社, カタログ番号61870-127)中で維持した。
Cas9タンパク質を安定的に発現するNK-92細胞は、NK-92親細胞にEdit-R Cas9レンチウイルスを感染させることによって作製した。簡単に説明すると、Edit-R Cas9レンチウイルスストックを作製するために、10cmペトリ皿あたり7×106個の293T細胞に、以下の量のプラスミドをトランスフェクトした:7.5μgのEdit-R-Cas9(Dharmacon社, カタログ番号CAS10138)、5μgのpCMV-ΔR8.2、および2.5μgのpCMV-VSV.G。トランスフェクションは、リポフェクタミン3000(Life Technologies社, カタログ番号L3000-008)を用いてメーカーの指示に従って行った。トランスフェクションの48時間後にウイルス上清を回収し、System Biosciences社からのPEG-it Virus Precipitation Solution(カタログ番号LV810A-1)を用いて10倍に濃縮した。5×105個のNK-92細胞に、スピノキュレーション(840g、35℃で99分)によって、TransDux(System Biosciences社, カタログ番号LV850A-1)の存在下に、24ウェルプレートで最終培養液1ml中の100μlの濃縮ウイルスを感染させた。形質導入の48時間後、Cas9を発現する細胞を、15μg/mlのブラスチシジン(InvivoGen社, カタログ番号ant-bl-1)の存在下で該細胞を増殖させることによって選択した。
ガイドRNAを、MITウェブツールhttpアドレスcrispr.mit.eduを用いて設計した。以下に各遺伝子の標的配列を示す。
MaxCyte GTエレクトロポレーターを用いたエレクトロポレーションにより、インビトロ転写されたsgRNAをCas9-NK-92細胞にトランスフェクトした。簡単に説明すると、NK-92-3-OCプロトコルを用いて、5×106個のCas9-NK-92細胞に10μgのインビトロ転写sgRNAをトランスフェクトした。各標的遺伝子について最も効率的なsgRNAを決定するために初期実験を行った。これらの実験ではsgRNA 1~4をCas9-NK-92細胞にトランスフェクトし、トランスフェクションの48時間後にフローサイトメトリーで標的遺伝子の発現を分析して、各sgRNAのノックアウト効率を決定した。
細胞表面タンパク質のサイトフルオロメトリー分析(cytofluorometric analysis)は、以下の表に示したフルオロフォアコンジュゲート化抗体を用いる直接免疫染色によって行った。簡単に説明すると、105個の細胞を100μlのフローサイトメトリー染色緩衝液(PBS、1%BSA)中の推奨量の抗体で4℃、暗所にて30分間染色した。細胞をフローサイトメトリー染色緩衝液で2回洗浄し、200μlのフローサイトメトリー染色緩衝液中に再懸濁した。サンプルをMACSQuant 10フローサイトメーター(Miltenyi社)で処理し、FlowJoソフトウェアを用いてデータを解析した。
標的細胞を、蛍光色素PKH67-GL(Sigma-Aldrich社, Saint Louis, MO)を用いて、メーカーの指示に従って染色した。標的およびエフェクターを、96ウェルプレート(Falcon BD社, Franklin Lakes, NJ)において異なるエフェクター対標的比で合わせて、短時間遠心分離し、5%ヒト血清を補充したX-VIVO 10(Lonza社, カタログ番号04-743Q)培養液中で5%CO2インキュベーター内、37℃で4時間インキュベートした。インキュベーション後、細胞を1%BSA/PBS緩衝液中10μg/mlのヨウ化プロピジウム(PI, Sigma-Aldrich社)で染色し、直ちにフローサイトメトリーにより分析した。死滅した標的細胞は、PKH67-GLおよびPIについて二重陽性として同定した。また、標的細胞およびエフェクター細胞をPIで別々に染色して、自発的細胞溶解を評価した。NKを介した細胞傷害性のパーセンテージは、エフェクターを含むサンプル中のPKH(+)/PI(+)細胞のパーセンテージから、標的細胞単独(自発的溶解)についてのPKH(+)/PI(+)細胞のパーセンテージを差し引くことによって得た。
CD96、TIGIT、CD226シングルノックアウトおよびCD96/TIGITダブルノックアウトNK-92細胞の作製
NK-92細胞株は、非ホジキンリンパ腫の50歳白人男性患者の末梢血から樹立されたナチュラルキラー様細胞株である(Gong et al., Leukemia 8:652-8 1994)。NK-92細胞はCD2、CD56およびCD57について陽性であり、CD3およびCD16について陰性である(Gong et al., 参照)。それらの増殖はIL-2依存性であり、それらは広範囲の腫瘍標的に対して強力なインビトロ細胞傷害性を発揮する。NK-92細胞は、現在知られている抑制性KIR受容体の大部分を欠いている(Maki et al., 前掲)。しかし、それらはCD226、CD96、およびTIGITを発現しており(図1)、これらはネクチンおよびネクチン様タンパク質と結合しかつNK細胞機能を調節する上で重要な役割を果たす受容体のファミリーのメンバーである。CD226は、DNAM1としても知られており、NK細胞の細胞傷害性を媒介するのに重要な活性化受容体であり(Shibuya et al., 前掲)、一方CD96およびTIGITは、NK機能活性を抑える抑制性免疫チェックポイントとして作用することが示されている(Chan et al., Nat Immunol. 15:431-8, 2014; Sarhan et al., Cancer Res. 76:5696-5706, 2016)。抗腫瘍能力を高めるために、これらの抑制性受容体の1つまたは複数を欠くNK-92変異体を作製した。
活性化CD226受容体と抑制性CD96およびTIGIT受容体は、共通のリガンドであるCD155(ポリオウイルス受容体PVRとしても知られる)を共有しており、それらは異なる親和性でリガンドCD155に結合する(Martinet et al., 前掲)。CD155発現は多くの場合腫瘍細胞においてアップレギュレートされており、その過剰発現はがんの浸潤性および転移と関連している(Hirota et al., Sloan et al., 両方とも前掲)。そのために、CD155陽性腫瘍標的に対する親細胞およびネクチン受容体ノックアウト細胞の細胞傷害能を最初に試験した。MCF-7およびSKBR-3は、CD155発現について陽性である2つの乳がん細胞株である(図3)。活性化受容体としてのCD226の役割と一致して、CD226-KO NK-92細胞によるMCF-7またはSKBR-3の死滅は、ほぼ完全に無効にされた(図4)。
Claims (39)
- CD96の発現を阻害する遺伝的変更およびTIGITの発現を阻害する遺伝的変更を含む、改変型NK-92細胞。
- CD96を標的としかつCD96の発現を阻害する干渉RNAを含む、請求項1に記載の改変型NK-92細胞。
- 前記細胞内でCD96発現をノックダウンまたはノックアウトすることによって作製される、請求項1に記載の改変型NK-92細胞。
- 前記細胞によって発現されるCD96の量が、CD96改変を有さない対応するNK-92細胞と比較して、少なくとも80%減少する、請求項2または3に記載の改変型NK-92細胞。
- TIGITを標的としかつTIGITの発現を阻害する干渉RNAを含む、請求項1に記載の改変型NK-92細胞。
- 前記細胞内でTIGIT発現をノックダウンまたはノックアウトすることによって作製される、請求項1に記載の改変型NK-92細胞。
- 前記細胞によって発現されるTIGITの量が、TIGIT改変を有さない対応するNK-92細胞と比較して、少なくとも80%減少する、請求項5または6に記載の改変型NK-92細胞。
- CD96を標的としかつCD96の発現を阻害する干渉RNAと、TIGITを標的としかつTIGITの発現を阻害する干渉RNAとを含む、請求項1に記載の改変型NK-92細胞。
- 前記細胞内でCD96発現をノックダウンまたはノックアウトすることおよびTIGIT発現をノックダウンまたはノックアウトすることによって作製される、請求項1に記載の改変型NK-92細胞。
- 前記細胞によって発現されるCD96の量が、CD96改変を有さない対応するNK-92細胞と比較して、少なくとも80%減少し、かつ
前記細胞によって発現されるTIGITの量が、TIGIT改変を有さない対応するNK-92細胞と比較して、少なくとも80%減少する、
請求項8または9に記載の改変型NK-92細胞。 - CD96発現およびTIGIT発現がノックアウトされる、請求項1に記載の改変型NK-92細胞。
- 少なくとも1つのFc受容体、または少なくとも1つのキメラ抗原受容体(CAR)、または少なくとも1つのFc受容体と少なくとも1つのCARの両方を細胞表面上に発現する、請求項1~11のいずれか一項に記載の改変型NK-92細胞。
- 前記少なくとも1つのFc受容体が、CD16、またはSEQ ID NO:13を参照して番号付けされた176位にバリンを有するCD16ポリペプチドである、請求項12に記載の改変型NK-92細胞。
- 前記少なくとも1つのFc受容体が、SEQ ID NO:13のアミノ酸19~254に対して少なくとも90%の配列同一性を有するポリペプチドをコードするポリヌクレオチド配列を含み、かつSEQ ID NO:13を参照して番号付けされた176位にバリンを含む、請求項13に記載の改変型NK-92細胞。
- 前記少なくとも1つのFc受容体がFcγRIIIである、請求項12に記載の改変型NK-92細胞。
- 前記CARがFcεRIγの細胞質ドメインを含む、請求項12に記載の改変型NK-92細胞。
- 前記CARが腫瘍関連抗原を標的とする、請求項16に記載の改変型NK-92細胞。
- サイトカインをさらに発現する、請求項1~17のいずれか一項に記載の改変型NK-92細胞。
- サイトカインがインターロイキン-2またはその変異体である、請求項18に記載の改変型NK-92細胞。

- 請求項1~20のいずれか一項に記載の改変型NK-92細胞を複数含む、組成物。
- 生理学的に許容される賦形剤をさらに含む、請求項21に記載の組成物。

- 前記細胞が10回未満の集団倍加を受けている、請求項23に記載の細胞株。
- それを必要とする対象におけるがんを治療するための薬学的組成物であって、治療有効量の請求項22に記載の組成物または請求項23もしくは24に記載の細胞株を含む、薬学的組成物。
- 抗体と組み合わせて使用するための、請求項25に記載の薬学的組成物。
- 患者の体表面積1m2あたり1×108~1×1011個の細胞が患者に投与されるように使用するための、請求項25または26に記載の薬学的組成物。
- (a)請求項21もしくは22に記載の組成物または請求項23もしくは24に記載の細胞株を含む、がんを治療するためのキット。
- 対照NK-92細胞と比較して低下したレベルのCD96およびTIGITを発現するNK-92細胞を作製する方法であって、NK-92細胞におけるCD96およびTIGITの発現を遺伝的に改変する段階を含む、方法。
- CD96およびTIGITの発現を遺伝的に改変する段階が、改変されるNK-92細胞を、CD96を標的とする干渉RNAおよびTIGITを標的とする干渉RNAと接触させることを含む、請求項29に記載の方法。
- 干渉RNAが、siRNA、shRNA、マイクロRNA、または一本鎖干渉RNAである、請求項30に記載の方法。
- CD96およびTIGITの発現を遺伝的に改変する段階が、それぞれの遺伝子を、ジンクフィンガーヌクレアーゼ(ZFN)、Taleエフェクタードメインヌクレアーゼ(TALEN)、またはCRISPR/Casシステムを用いて改変することを含む、請求項30に記載の方法。
- CD96およびTIGIT標的遺伝子のそれぞれの発現を遺伝的に改変する段階が、
(i)NK-92細胞に、CRISPR(clustered regularly interspaced short palindromic repeat)関連(Cas)タンパク質を導入する段階、および
(ii)改変されるNK-92細胞にリボ核酸を導入する段階
を含み、
該リボ核酸が、該CD96およびTIGIT標的遺伝子のそれぞれの配列の標的モチーフにハイブリダイズするようにCasタンパク質を誘導し、該標的モチーフが切断される、

- CD96中の標的モチーフにハイブリダイズする前記リボ核酸がSEQ ID NO:1~4からなる群より選択され、TIGIT中の標的モチーフにハイブリダイズする前記リボ核酸がSEQ ID NO:5~8からなる群より選択される、請求項33~38のいずれか一項に記載の方法。
