Description
細胞媒介性免疫系の一部を形成するTリンパ球(又はT細胞)は、病原体の根絶において主要な役割を果たす。T細胞は胸腺において発達し、T細胞の表面上にT細胞受容体分子を発現し、これは有核細胞上に発現された主要組織適合性複合体(MHC)分子上に提示されたペプチド(抗原提示)の認識を可能とする。病原体の抗原、すなわち、MHC分子によって提示された外来抗原は強力なT細胞応答を惹起し、一方、このようなT細胞の発達中の胸腺における自己抗原特異的T細胞のネガティブ選択のために、自己抗原は通常はT細胞応答を生じない。したがって、免疫系は、外来抗原又は自己抗原を提示している有核細胞を区別することができ、強力なサイトカイン遊離及びT細胞の細胞障害性機序を介して、感染した細胞を特異的に標的化及び根絶することができる。
本発明は、抗原特異的T細胞受容体、並びに、それを含む核酸、ベクター、宿主細胞;並びに、その様々な使用及び適用を提供する。
本発明者らは、腫瘍関連抗原(TAA)PRAMEを発現している細胞を特異的に認識することができ;そして、サイトカインの産生及び標的細胞の細胞溶解などの有利なエフェクター機能を示す、T細胞クローンを同定した。それ故、該T細胞クローン及びそれらのT細胞受容体は、非常に特異的かつ効果的ながんの処置のための有望なツールである。したがって、同定されたPRAME特異的TCRは、がんの養子T細胞療法に適している。高度に特異的にPRAMEに結合することのできるTCRの同定は、エクスビボにおいてT細胞を「武装化」しドナーにそれらを再導入することを可能とし、そこでそれらはPRAMEを発現しているがん細胞を効果的に認識し特異的に排除することができる。さらに、本明細書において提供される新規なTCRの抗原結合領域を使用して、直接投与のために容易に利用可能であるさらなる機能的な部分(例えば、薬物、標識、又は他の免疫細胞を誘引するさらなる結合ドメイン)を含む可溶性構築物を設計することができる。
CDR3ドメイン
第一の態様では、したがって、本発明は、(i)CAVHSTAQAGTALIF(配列番号1)の配列を含むか若しくはからなるT細胞受容体α鎖CDR3、及び/又は(ii)CASSTHRGQTNYGYTF(配列番号2)のアミノ酸配列を含むか若しくはからなるT細胞受容体β鎖CDR3を含む、T細胞受容体(TCR)に関する。
以前に示されているように、本発明のTCRは、MHC分子、特にMHC−I分子、特にHLA−A、好ましくはHLA−A*02、特に対立遺伝子HLA−A*02:01によってコードされるHLA−A2分子上に提示されると、それらの抗原標的PRAME100〜108を認識すると考えられる(T細胞又はTCRは、特定のMHC分子に「拘束」されると言われる)。また、本発明のTCRは、他のHLA−A*02対立遺伝子上に提示されたそれらの抗原標的を認識すると考えられ得る。以前に記載されているように、TCRα鎖及びβ鎖のCDR1及びCDR2は主にMHCの認識に関与していると考えられる。HLA−A*02拘束性の抗原認識に関与していることが知られているCDR1配列及びCDR2配列の「プール」は限定され、本発明のCDR3ドメインは原則的に配列番号34〜224に示されるCDR1ドメイン及びCDR2ドメインのいずれとも組み合わせることができると考えられ、ただし、TCRは、その抗原標的、好ましくはそのHLA−A*02結合形のその抗原標的を認識するその能力を、添付の実施例において評価されたTCRと類似しているか、同じであるか、又はさらにはより高い程度で保持している。CDR1ドメイン及びCDR2ドメインの有用な例としては、配列番号5に示されるようなVSGLRG配列を含むか又はからなるCDR1α、配列番号3に示されるようなLYSAGEE配列を含むか又はからなるCDR2α、配列番号6に示されるようなSGDLS配列を含むか又はからなるCDR1β、及び配列番号4に示されるようなYYNGEE配列を含むか又はからなるCDR2βが挙げられる。該CDR配列は図9にも示される。
本発明はさらに、配列番号15に示されるようなアミノ酸配列を含むか若しくはからなるTCRα鎖可変領域、及び/又は、配列番号16に示されるようなアミノ酸配列を含むか若しくはからなるTCRβ鎖可変領域を含む、TCRを提供する。該α鎖配列及びβ鎖配列は図9にも示されている(通常のフォント)。
TCRはさらに、そのα鎖及び/又はβ鎖に定常(C)領域を含んでいてもよい。定常領域は、ヒト定常領域であり得るか又は別の種に由来し得、これにより「キメラ」TCRが生じる。例えば、ヒトα鎖及び/又はβ鎖は、TCRα鎖及びβ鎖の選択的な対形成及びCD3共受容体とのより安定な会合を支持することによって、ヒトTCRの表面発現を増強させることが判明しているそれらのマウス対応物によって置換(「マウス化」)されていてもよい。α鎖の適切な定常領域は、例えば、配列番号17(ヒト)、19(最小限にマウス化されている)及び21(マウス)から選択され得る。β鎖の適切な定常領域は、配列番号18(ヒト)、20(最小限にマウス化されている)及び22(マウス)から選択され得る。本明細書の「TCR配列変異体」の章にさらに説明されているように、完全ヒト定常領域をそれらのマウス対応物によって置き換える代わりに、ヒト定常領域内のいくつかのアミノ酸のみを、マウス定常領域の対応するアミノ酸と交換することも可能である(「最小限のマウス化」)。
本発明のTCRの有用な例としては、配列番号7、9、11又は13に示されているようなアミノ酸配列を含むか又はからなるα鎖と、配列番号8、10、12又は14に示されているようなアミノ酸配列を含むか又はからなるβ鎖とを含むTCRが挙げられる。したがって、本発明はとりわけ、配列番号7に示されるようなアミノ酸配列を含むか又はからなるα鎖と、配列番号8に示されるようなアミノ酸配列を含むか又はからなるβ鎖とを含むか又はからなるTCR;配列番号9に示されるようなアミノ酸配列を含むか又はからなるα鎖と、配列番号10に示されるようなアミノ酸配列を含むか又はからなるβ鎖とを含むか又はからなるTCR;配列番号11に示されるようなアミノ酸配列を含むか又はからなるα鎖と、配列番号12に示されるようなアミノ酸配列を含むか又はからなるβ鎖とを含むか又はからなるTCR(両方共が図9にも示されている);及び、配列番号13に示されるようなアミノ酸配列を含むか又はからなるα鎖と、配列番号14に示されるようなアミノ酸配列を含むか又はからなるβ鎖とを含むか又はからなるTCRを提供する。
本明細書において提供されるTCRは有益には、(ヒト)PRAMEに結合することができる。PRAME(腫瘍において優先的に発現されている悪性黒色腫抗原、Uniprotアクセッション番号P78395)は、MAPE(腫瘍において優先的に発現されている悪性黒色腫抗原)及びOIP4(OPA相互作用タンパク質4)とも称されるが、これは機能が不明な精巣癌抗原(CTA)であると報告されている。
以前に記載されているように、「TCR」という用語は、TCRの配列変異体、断片、及び構築物をはじめとする、TCRの変異体を包含する。全てのTCR変異体は、本発明のTCRの機能的変異体であると考えられる。本明細書において使用する「機能的変異体」という用語は、親のTCR、その可変領域、又はその抗原結合領域に対してかなりの又は有意な配列同一率又は類似率を有する、TCRのポリペプチド又はタンパク質を指し、これはその生物学的活性、すなわち、本発明の親TCRが抗原特異性を示す抗原標的に対して、本明細書に開示されかつ添付の実施例で評価されたTCRと類似しているか、同じであるか、又はさらにはより高い程度で特異的に結合する能力を共有する。
「TCR変異体」という用語は、本明細書に開示されたTCRの「配列変異体」、すなわち、上記のような本発明のTCR(「親」TCRとも称される)のアミノ酸配列を実質的に含んでいるが、「親」TCRアミノ酸配列と比較して少なくとも1つのアミノ酸の改変(すなわち、置換、欠失、又は挿入)を含有している変異体を含み、ただし、該変異体は好ましくは、本発明の「親」TCRの抗原特異性を保持している。本発明のTCR配列変異体は典型的には、適切なヌクレオチド変化を「親」TCRをコードしている核酸に導入することによって、又はペプチド合成によって調製される。一般的には、前記のアミノ酸改変は、TCRの可変領域又は定常領域に導入され得るか又は存在し得、結合強度及び結合特異性、翻訳後プロセシング(例えばグリコシル化)、熱力学的安定性、溶解度、表面発現、又はTCRの構築のような特性を調節する役目を果たし得る。
定常領域へのジスルフィド結合の付加は、TCRα鎖及びβ鎖の正しい対形成を促進すると報告されている(Kuball J et al. Blood. 2007 Mar 15; 109(6):2331-8)。したがって、定常領域への1つ以上のシステイン結合の付加も本明細書において考えられる。
以前に記載されているように、TCRのマウス化(すなわち、α鎖及びβ鎖におけるヒト定常領域を、そのマウス対応物と交換すること)は、宿主細胞におけるTCRの細胞表面発現を向上させるために一般的に適用される技術である。特定の理論に拘りたくはないが、マウス化TCRは、CD3共受容体とより効果的に会合し;及び/又は、互いに優先的に対を形成し、所望の抗原特異性を有するTCRを発現するようにエクスビボにおいて工学操作されたヒトT細胞上で混合型TCRを形成する傾向がより少ないが、依然としてそれらの「元来の」TCRを保持及び発現していると考えられる。
本明細書において使用する「TCR」という用語はTCR構築物をさらに含む。「構築物」という用語は、本発明のTCRの少なくとも1つの抗原結合ドメインを含むタンパク質又はポリペプチドを含むが、必ずしも天然TCRの基本的構造(すなわち、TCRα鎖及びTCRβ鎖に取り込まれた可変ドメインが、ヘテロ二量体を形成している)を共有していない。TCR構築物及び断片は典型的には、慣用的な遺伝子工学操作法によって得られ、追加の機能的タンパク質又はポリペプチドドメインを含むようにしばしば人工的に構築される。前記によると、本発明のTCR構築物及び断片は、本明細書の何処か他の場所で開示されているような少なくとも1つのCDR3α及び/又は少なくとも1つのCDR3βを含むと考えられる。本明細書においてさらに考えられるのは、本明細書において例示されているようなさらに他のタンパク質ドメイン又は部分と場合により組み合わせた、少なくとも1つのCDR1α、CDR2α、CDR1β、CDR2β、α鎖可変領域、β鎖可変領域、α鎖及び/又はβ鎖、あるいはその組合せを含んでいる、構築物及び断片である。本明細書に提供されたTCR構築物及び断片は、上記されそして添付の実施例において評価されている本発明のTCRと同じ抗原標的に特異的に結合することができると考えられる。
「TCR構築物」という用語は、少なくとも1つのTCRα鎖可変領域又はTCRα鎖と少なくとも1つのTCRβ鎖可変領域が互いに共有結合で連結されている、ヘテロ二量体及び多量体を包含する。本発明による多価TCR構築物は、その最も単純な形態では、好ましくはリンカー分子を介して、互いに会合した(例えば共有結合により又は別の方法で連結された)2つ又は3つ又は4つ又はそれ以上のTCRの多量体を含む。
「TCR構築物」という用語はまた、少なくとも1つのTCRα鎖、TCRα鎖可変領域若しくはCDR3α、及び/又は少なくとも1つのTCRβ鎖、TCRβ鎖可変領域若しくはCDR3β;並びにさらなる1つ以上の融合成分(群)を含む融合タンパク質又はポリペプチドに関する。有用な成分としては、Fc受容体;Fcドメイン(IgA、IgD、IgG、IgE、及びIgMに由来);サイトカイン(例えばIL−2又はIL−15);毒素;抗体若しくはその抗原結合断片(例えば抗CD3抗体、抗CD28抗体、抗CD5抗体、抗CD16抗体、若しくは抗CD56抗体、又はその抗原結合断片);CD247(CD3ζ)、CD28、CD137、CD134ドメイン;又はその任意の組合せが挙げられる。
本発明のTCRは、「単離された」又は「実質的に純粋な」形態で提供され得る。本明細書において使用される場合の「単離された」又は「実質的に純粋な」は、TCRが、その産生環境の成分から同定、分離、及び/又は回収され、これにより「単離された」TCRは、その治療使用又は診断使用を妨害する可能性のあるその産生環境からの他の混入成分を含まないか又は実質的に含まないことを意味する。混入成分としては、酵素、ホルモン、及び他のタンパク質性又は非タンパク質性溶質が挙げられ得る。したがって、「単離された」TCRは、これらの混入成分を除去するか又は実質的に除去する少なくとも1回の精製工程によって調製されるだろう。前記の定義は、必要な変更を加えて、「単離された」ポリヌクレオチド/核酸に対して同等に適用可能である。
本発明のTCRは、可溶化形で提供され得る。可溶性TCRは、可溶性TCRによって認識される抗原標的を発現している例えばがん細胞に対して、標的治療剤又はエフェクター細胞を特異的に標的化させる、診断ツール、及び担体又は「アダプター」として有用である。可溶性TCR(sTCR)は典型的には、TCRα鎖及び/若しくはβ鎖、又はその可変領域若しくはCDRを含み、場合によりジスルフィド結合を介して安定化されているか、又は例えば本発明のTCR構築物の内容で上記されているような適切なリンカー分子を介して共有結合で連結されている断片又は構築物であろう。それらは典型的には、例えば膜貫通領域を含まないだろう。いくつかの環境では、分子の溶解度、並びに/又はα鎖及びβ鎖の正しい折り畳み及び対形成(所望であれば)(特に前記の特色を提供しない組換え宿主において産生される場合)を増強させるために、ポリペプチド配列内にアミノ酸改変を導入してもよい。例えば、産生宿主細胞としてE.coliを使用する場合、TCRα鎖及びβ鎖の折り畳み及び対形成は典型的にはインビトロで成し遂げられる。それ故、本発明によるTCRは、例えば、本明細書の何処か他の場所で記載されているような追加のシステイン残基を含み得る。
本発明のTCRはさらに、以下に記載のような1つ以上の改変を含み得る。以下に記載の改変は典型的には共有結合による改変であり、当技術分野において公知である標準的な技術を使用して成し遂げられ得る。いくつかの環境では、TCR内のアミノ酸の改変が、該改変の導入を促進するために必要とされ得る。
本発明のTCR、特に(可溶性)TCRは標識されていてもよい。有用な標識は当技術分野において公知であり、場合により様々な長さのリンカーを介して慣用的な方法を使用してTCR又はTCR変異体に結合させることができる。「標識」又は「標識基」という用語は、任意の検出可能な標識を指す。一般的に、標識は、それらを検出しようとするアッセイに応じて、様々なクラスに分類される。以下の例には、同位体標識(これは放射性又は重同位体、例えば放射性同位体又は放射性核種(例えば3H、14C、15N、35S、89Zr、90Y、99Tc、111In、125I、131Iであり得る);磁気標識(例えば磁気粒子);酸化還元活性部分;光学色素(発色団、リン光体、及びフルオロフォアを含むがこれらに限定されない)、例えば蛍光基(例えばFITC、ローダミン、ランタニド、リン光体)、化学発光基、及びフルオロフォア(これは「低分子」フルオロフォア又はタンパク質性フルオロフォアのいずれかであり得る);酵素基(例えばセイヨウワサビペルオキシダーゼ、β−ガラクトシダーゼ、ルシフェラーゼ、アルカリホスファターゼ);ビオチニル化基;又は二次リポーターによって認識される予め決定されたポリペプチドエピトープ(例えばロイシンジッパー対配列、二次抗体の結合部位、金属結合ドメイン、エピトープタグなど)が含まれるがこれらに限定されない。TCR、TCR変異体、又は特に可溶性のTCR構築物(例えば、本明細書に記載のような少なくとも1つのTCRα鎖及び/又はTCRβ鎖を含むもの)を診断使用のために意図する場合には、標識が特に考えられる。
本発明のTCR、特に可溶性TCRは、例えば、免疫原性を低下させるために、流体力学的サイズ(溶液中のサイズ)、溶解度及び/又は安定性(例えば、タンパク質分解に対する増強された保護による)を増加させるために、並びに/あるいは、血清中半減期を延長させるために、さらに他の機能的部分を付着させることによって改変され得る。
改変されたグリコシル化パターンを有するTCRも本明細書において考えられる。当技術分野において公知であるように、グリコシル化パターンは、アミノ酸配列(例えば、以下に考察されている、特定のグリコシル化アミノ酸残基の有無)及び/又はタンパク質が産生される宿主細胞若しくは生物に依存し得る。ポリペプチドのグリコシル化は典型的には、N結合又はO結合のいずれかである。N結合は、アスパラギン残基の側鎖への糖質部分の付着である。結合分子に対するN結合グリコシル化部位の付加は簡便には、アスパラギン−X−セリン及びアスパラギン−X−トレオニン(Xはプロリンを除く任意のアミノ酸である)から選択された1つ以上のトリペプチド配列を含有するようにアミノ酸配列を改変することによって成し遂げられる。O結合グルコシル化部位は、出発配列への1つ以上のセリン残基又はトレオニン残基による付加又は置換によって導入され得る。
本発明のTCR、特に可溶性TCRに、低分子化合物などの薬物を付加することも考えられ得る。連結は、共有結合を介して、又は正電気力を通してなどの非共有結合的な相互作用を介して成し遂げられ得る。当技術分野において公知である様々なリンカーを、薬物抱合体を形成するために使用することができる。
本発明のTCR、特に可溶性TCRは、それぞれの分子(タグ)の同定、追跡、精製、及び/又は単離を補助する追加ドメインを導入するために改変されていてもよい。このようなタグの非制限的な例としては、Myc−タグ、HAT−タグ、HA−タグ、TAP−タグ、GST−タグ、キチン結合ドメイン(CBD−タグ)、マルトース結合タンパク質(MBP−タグ)、Flag−タグ、Strep−タグ、及びその変種(例えば、Strep II−タグ)、His−タグ、CD20、Her2/neuタグ、myc−タグ、FLAG−タグ、T7−タグ、HA(ヘマグルチニン)−タグ、又はGFP−タグとして知られるペプチドモチーフを含む。
本明細書においてさらに、本明細書に記載のような1つ以上のポリヌクレオチドを含むベクターも提供される。「ベクター」は、(外来)遺伝子材料を宿主細胞に導入するためのビヒクルとして使用される核酸分子であり、その宿主細胞で例えば、複製及び/又は発現させることができる。
標的化ベクターを使用して、J. Sambrook et al., Molecular Cloning: A Laboratory Manual (4th edition), Cold Spring Harbor Laboratory, Cold Spring Harbor Laboratory Press, New York(2012)によって記載のような当技術分野において公知の方法によって、ポリヌクレオチドを宿主細胞の染色体に組み込むことができる。簡潔に言えば、適切な手段としては、相同的組換え、すなわち組込み部位の配列に特異的に標的化するハイブリッドリコンビナーゼの使用が挙げられる。標的化ベクターは典型的には、相同的組換えに使用される前は環状及び鎖状である。代替選択肢として、外来ポリヌクレオチドは、融合PCRによって接続されたDNA断片又は合成により構築されたDNA断片であり得、これは次いで、宿主細胞に組み込まれる。また、無作為組込み又は非標的化組込みがもたらされる異種組換えを使用することも可能である。
本発明のベクターは発現ベクターであってもよい。「発現ベクター」又は「発現構築物」は、適切な宿主細胞における、異種ポリヌクレオチド配列、例えば本発明のTCRをコードしている配列の転写、及びそれらのmRNAの翻訳のために使用され得る。このプロセスはまた、本明細書において本発明のTCRの「発現」とも称される。
本発明はさらに、本明細書に記載のTCR、核酸、又はベクターを含む宿主細胞を提供する。
細胞
本発明の可溶性TCRの発現のために使用される「産生宿主細胞」は好ましくは、多量の組換えタンパク質を発現することができる。
発現ベクターを有する宿主細胞を、本明細書に提供されているTCR、特に本明細書の何処か他の場所で記載されているようなα鎖及び/又はβ鎖の生成に適した条件下で増殖させ、α鎖及び/又はβ鎖のタンパク質合成についてアッセイする。二本鎖TCRの発現のために、α鎖及びβ鎖の両方をコードしているベクターを、完全分子の発現のために宿主細胞において共発現させ得る。
本発明のTCRが一旦発現すると、それは、当技術分野において公知である任意の精製法によって、例えば、クロマトグラフィー(例えばイオン交換クロマトグラフィー(例えばヒドロキシアパタイトクロマトグラフィー)、アフィニティクロマトグラフィー、特にプロテインA、プロテインG、若しくはレクチンアフィニティクロマトグラフィー、サイズカラムクロマトグラフィー)、遠心分離、示差溶解度、疎水性相互作用クロマトグラフィーによって、又はタンパク質の精製のための任意の他の標準的な技術によって精製され得る。当業者は、回収しようとするTCRの個々の特徴に基づいて適切な精製法を容易に選択することができるだろう。
以前に記載されているように、本発明はまた、本発明のヌクレオチド配列、ベクター、又はTCRを含む「エフェクター宿主細胞」を提供する。該エフェクター宿主細胞は、慣用的な方法を使用して、本発明のTCRをコードしている核酸配列を含むように改変され、本明細書に記載のTCRを特に細胞表面上に発現すると考えられる。本発明の目的のために、「本発明のTCRを発現している改変された宿主細胞」は一般的に、例えば添付の実施例に記載のようなRNAトランスフェクションによって、本発明によるTCRを発現するように処理又は改変された(エフェクター又は産生)宿主細胞を指す。本明細書の何処か他の場所で記載されているような、他の改変法又はトランスフェクション法又は形質導入法も考えられる。したがって、「改変された宿主細胞」という用語は、本発明のTCRを好ましくは発現している「トランスフェクトされた」、「形質導入された」及び「遺伝子工学操作された」宿主細胞を含む。
本発明はさらに、1つ以上の活性薬剤として、本明細書に記載のようなTCR、核酸、ベクター、及び/又は宿主細胞、並びに場合により1つ以上の薬学的賦形剤(群)を含む医薬組成物を提供する。したがって、医薬組成物又は医薬品の製造のための該TCR、核酸、ベクター、及び宿主細胞の使用も本明細書において考えられる。
TCRポリヌクレオチド、ベクター、又は宿主細胞の正確な用量は、公知の技術を使用して当業者によって確認可能であろう。適切な用量は、十分な量の本発明の活性薬剤を提供し、好ましくは治療に有効であり、すなわち、所望の治療効果を惹起する。
「賦形剤」という用語は、充填剤、結合剤、崩壊剤、コーティング剤、吸着剤、接着防止剤、潤滑剤、保存剤、抗酸化剤、芳香剤、着色剤、甘味剤、溶媒、共溶媒、緩衝剤、キレート剤、粘度付与剤、表面活性剤、希釈剤、湿潤剤、担体、希釈剤、保存剤、乳化剤、安定化剤、及び等張化剤を含む。所望の本発明の医薬組成物を調製するために適した賦形剤を選択することは当業者の知識範囲内である。本発明の医薬組成物に使用するための例示的な担体としては、食塩水、緩衝食塩水、デキストロース、及び水が挙げられる。典型的には、適切な賦形剤の選択はとりわけ、使用される活性薬剤、処置される予定の疾患、及び医薬組成物の所望の製剤に依存するだろう。
本発明はさらに、上記に明記された1つ以上の本発明の活性薬剤(例えば、宿主細胞又はTCR構築物)と、処置される予定の疾患の治療及び/又は予防に適した1つ以上の追加の活性薬剤とを含む、医薬組成物を提供する。組合せに適した活性成分の好ましい例としては、公知の抗がん薬、例えばシスプラチン、マイタンシン誘導体、ラケルマイシン、カリケアマイシン、ドセタキセル、エトポシド、ゲムシタビン、イホスファミド、イリノテカン、メルファラン、ミトキサントロン、ポルフィマーナトリウム(sorfimer sodium)、フォトフリンII、テモゾロミド、トポテカン、グルクロン酸トリメトレキサート、アウリスタチンE、ビンクリスチン、及びドキソルビシン;並びに、ペプチド細胞毒素、例えばリシン、ジフテリア毒素、シュードモナス細菌外毒素A、DNAアーゼ及びRNAアーゼ;放射性核種、例えばヨウ素131、レニウム186、インジウム111、イットリウム90、ビスマス210及び213、アクチニウム225、及びアスタチン213;プロドラッグ、例えば抗体に指向する酵素プロドラッグ;免疫刺激剤、例えばIL−2、ケモカイン、例えばIL−8、血小板因子4、悪性黒色腫増殖刺激タンパク質など、抗体又はその断片、例えば抗CD3抗体又はその断片、補体活性化因子、異種タンパク質ドメイン、同種タンパク質ドメイン、ウイルス/細菌タンパク質ドメイン、及びウイルス/細菌ペプチドが挙げられる。
様々な経路が、本発明による医薬組成物の投与のために適用可能である。典型的には、投与は非経口的に達成されるだろう。非経口送達法としては、局所、動脈内、筋肉内、皮下、髄内、くも膜下腔内、脳室内、静脈内、腹腔内、子宮内、膣内、舌下、又は鼻腔内投与が挙げられる。
本発明の医薬組成物は、とりわけ、使用される活性薬剤(例えば可溶性TCR)に応じて様々な剤形で、例えば固形剤、液剤、気体剤形、若しくは凍結乾燥剤形で製剤化され得、とりわけ、軟膏、クリーム、経皮パッチ、ゲル、散剤、錠剤、液剤、エアゾール、顆粒剤、丸剤、懸濁剤、乳化剤、カプセル剤、シロップ剤、液剤、エリキシル剤、エキス剤、チンキ剤、若しくは液体エキス剤の剤形で、又は、所望の投与法に特に適した剤形で製剤化され得る。医薬品を製造するためのそれ自体公知のプロセスは、Remington's Pharmaceutical Sciences(Ed. Maack Publishing Co, Easton, Pa., 2012)第22版に示され、これは、例えば、慣用的な混合、溶解、造粒、糖衣錠作製、研和(levigating)、乳化、カプセル化、封鎖(entrapping)又は凍結乾燥プロセスを含み得る。例えば、本明細書に記載のような宿主細胞又は可溶性TCRを含む医薬組成物は典型的には、液体剤形で提供され、好ましくは薬学的に許容される緩衝液を含む。
前記を鑑みて、したがって、本発明は、医薬品としての使用のための本明細書に記載のようなTCR、核酸、ベクター、及び/又は宿主細胞を提供する。
療法のために、本発明のTCR、特に本発明の可溶性TCR、核酸、ベクター(例えばウイルスベクター)、又は宿主細胞を、それを必要とする被験者に直接投与することができる。したがって、本発明は、がんの検出、診断、予後予測、予防及び/又は治療の方法に使用するための、TCR、核酸、ベクター、又は宿主細胞を提供する。該方法は、(a)本発明の(i)TCR、(ii)核酸、(iii)ベクター、(iv)宿主細胞、及び/又は(v)医薬組成物のうち1つ以上を準備する工程;並びに(b)(i)〜(v)のうち1つ以上をそれを必要とする被験者に投与する工程を含み得る。場合により、該方法は、がん療法のさらに他の工程、例えば放射線療法、又は1つ以上の抗がん剤の投与を含み得る。

25. 改変されたリンパ球を生成するための、項目1〜12のいずれかに記載のTCR、項目13若しくは14記載の核酸、及び/又は項目15記載のベクターの使用。
本発明者らは、任意の所望のMHC拘束性及び抗原特異性を有するT細胞クローンを単離するためのインビトロにおけるプライミングアプローチを使用した。プライミングシステムは、抗原提示細胞として成熟樹状細胞(mDC)及び応答細胞として自己CD8+の豊富なT細胞を使用する。配列番号33に参照されているような完全長ヒトPRAMEアミノ酸配列をコードしているインビトロで転写されるRNA(ivtRNA)は、特異的な抗原の発生源としての役割を果たす。ivtRNAは、mDCに電気穿孔された後、完全長タンパク質に翻訳され、これは続いてプロセシングされmDCのMHC分子によってペプチドとして提示される。同じドナーに由来するivtRNAのトランスフェクトされたmDCとT細胞のインビトロにおける共培養により、対応するTCRの発生源としての役目を果たす抗原特異的T細胞が新規に誘導される。抗原特異的T細胞は様々な方法によって強化され得、これは限界希釈法又はFACSに基づいた単一細胞播種によってクローニングされる。
樹状細胞による高親和性TCRを有するT細胞のプライミングは、以下のプロトコールに従って自己MHC分子によるペプチド提示を使用して成し遂げられた(図1):
・HLA−A*02:01/PRAMEプライミング
・DC用の適切な成熟カクテルを使用して産生された8日目のmDC。
・APCの積載:ivtRNA。
・HLA−A*02:01RPAME100〜108多量体を介した強化
・FACS技術を使用した単一細胞選別
所望のPRAMEエピトープ(RPAME100〜108)を認識する候補T細胞クローン(T細胞クローンT4.8−1−29)の同定後、機能及び特異性に関する完全な特徴付けが実施された。分析は、様々なヒト腫瘍細胞株との共培養液中の単離されたT細胞クローン(T細胞クローンT4.8−1−29)のサイトカイン分泌パターン、クローンが様々な標的細胞を特異的に認識する能力、クローンの機能的結合力、並びにT2及び腫瘍細胞に対する細胞障害性を含んでいた。
実施例2.1.1:ポリサイトカイン分析
実験の設計図:ペプチドの積載されたT2細胞による刺激
・IFN−γ、IL−2、TNF−α、IL−5、IL−10、IL−6、IL12p70、IL−4、及びIL−1βを検出する、マルチプレックス(登録商標)サイトカイン分析が実施された。
・細胞の刺激:飽和量(10−5M)のPRAME100〜108ペプチド(「VLDペプチド」)又は無関係のPRAME300〜309、すなわちALYVDSLFFLペプチド(「ALYペプチド」、配列番号230)の積載されたT2細胞(HLA−A*02陽性)。
・関係のあるペプチド又は無関係のペプチドの積載されたT2細胞とT細胞の共培養液の上清を24時間後に収集し、続いて、マルチプレックス(登録商標)サイトカイン分析を使用して測定した。
・候補クローンは、バックグラウンドレベルを上回るIFN−γ、IL−2、及びTNF−α(Th1/Tc1サイトカイン)を分泌した。サイトカイン発現パターンは、良好な抗腫瘍エフェクター機能に関連したTh1表現型を反映する(図2)。
・IL−5及びIL−13(Th2/Tc2サイトカイン)分泌は検出されなかった(n.d.)。
実験の設計図:腫瘍細胞株による刺激
・IFN−γELISAを使用して、一連のヒト腫瘍細胞株を用いての刺激後のサイトカイン分泌を評価した(PRAMEの発現状態を、NanoString nCounter(登録商標)分析によって検出した)。
・T細胞クローンT4.8−1−29と、K562−A2、Mel−624.38、Colo−678、647−V及びSuDHL−6(全てHLA−A*02陽性)との共培養から最大24時間後に上清を収集した。特異的なIFN−γ分泌を、標準的なELISAを使用して評価した。
・標的細胞:
○K562−A2(HLA−A*02陽性、PRAME陽性)
○Mel−624.38(HLA−A*02陽性、PRAME陽性)
○Colo−678(HLA−A*02陽性、PRAME陰性)
○647−V(HLA−A*02陽性、PRAME陰性)
○SuDHL−6(HLA−A*02陽性、PRAME陰性)
・T細胞クローンT4.8−1−29は、PRAME陽性、HLA−A*02陽性腫瘍細胞株K562−A2、及びMel−624.38(陽性対照:ペプチドでパルスされたT2細胞)との共培養液中において高いIFN−γ分泌を示した。
・PRAME陰性、HLA−A*02陽性腫瘍細胞株は全く、T細胞クローンT4.8(陰性対照;n.d.、検出されず)によって認識されなかった。
・HLA−A*02及びPRAMEを発現している腫瘍細胞株のみが、自己拘束性T細胞クローンT4.8−1−29によって認識され、これにより抗原特異性が示された(図3)。
実験の設計図:ペプチドでパルスされたT2細胞を用いての刺激
・PRAME100〜108(VLD)ペプチドの認識のための機能的なT細胞の結合力を、ペプチドの積載されたT2細胞とクローンT4.8−1−29の共培養後のIFN−γ分泌の検出によって測定した。
・標的細胞:滴定量の外因性PRAME100〜108(VLD)ペプチド(10−5M〜10−12M)の積載されたT2細胞(HLA−A*02陽性、PRAME陰性)。
・エフェクター:標的の比(E:T)は1:1。
・相対的なIFN−γ遊離は、最大遊離に対する比率で示される。機能的結合力を規定する最大半量のIFN−γ分泌は、点線によって示される。
・培養上清を、共培養から約24時間後に収集し、標準的なELISAによって評価した。
・クローンT4.8−1−29は、約1×10−9〜1×10−10モル/L[M]濃度のPRAME100〜108ペプチドで最大半量のIFN−γ分泌を示し(2回の独立した実験の平均値)、これはウイルス特異的T細胞の生理学的範囲内にあり、効率的な抗腫瘍効力にとって望ましい機能的結合力を示すと報告されている(Aleksic, M et al. Eur. J. Immunol.;42 (12):3174-3179)。
・→TCR T4.8−1−29:約1×10−9モル/L[M](図4)。
PRAME100〜108特異的T細胞クローンT4.8−1−29が同定されたので、次の工程は、対応するTCR鎖をコードしているDNA配列情報の単離、クローニングされたTCRを適切なレシピエントT細胞に導入、続いて、TCRの工学操作されたT細胞の機能的分析を含んでいた。
元来のクローンT4.8−1−29のTCRα鎖及びβ鎖のDNA配列を、社内で次世代型シークエンス(NGS−TCRseq)によって分析した。対応するTCRα及びβのDNA配列をDNA遺伝子合成(GeneArt、レーゲンスブルク)によって再構築し、ivtRNA産生のためにpGEMベクター骨格に並びに安定な形質導入のためにレトロウイルスベクターにクローニングした。
T細胞クローンT4.8−1−29のTCR配列をレシピエント細胞に導入
元来のT細胞クローンT4.8−1−29のTCR DNA配列を、電気穿孔法によるレシピエントエフェクター細胞の一過性トランスフェクションのために、完全なT4.8−1−29TCR配列をコードしているRNAにインビトロで転写したか、又は、これもまた完全なTCR T4.8−1−29配列をコードしているレトロウイルスベクター構築物を使用することによるエフェクター細胞の安定な形質導入のために使用した。
・PRAME100〜108(VLD)ペプチドでパルスされたT2細胞との共培養液中の、TCR T4.8−1−29のトランスフェクトされたレシピエントT細胞(CD8陽性レシピエントT細胞クローン+T4.8−1−29ivtRNA)の特異的なIFN−γ分泌を、標準的なELISAを使用して測定した。
・標的細胞:10−5MのVLD(関連性のある)ペプチド又は「ELAペプチド」(無関係の)ペプチド(ELAGIGILTV、MelanA、配列番号231)を用いてパルスされたT2細胞(HLA−A*02陽性、PRAME陰性)。
・TCR T4.8−1−29のトランスフェクトされたレシピエントT細胞は、関連性のあるペプチドの積載されたT2細胞に対する良好な認識を示したが、T2細胞に無関係のペプチドを積載した場合には全く認識しなかった。
・T4.8−1−29のTCRα鎖及びβ鎖のDNA配列は正しく再構築され、導入遺伝子として良好な機能を示した(図5)。

10−5MのPRAME100〜108VLDペプチド又はHLA−A*02から溶出した遍在的な自己ペプチド(131個の自己ペプチド)の積載された、T2標的細胞(HLA−A*02陽性、PRAME陰性)と共培養されたクローンT4.8−1−29のT細胞受容体を発現しているCD8+の豊富なPBMCのIFN−γ分泌を、ELISAアッセイを使用して決定した。
・クローンT4.8−1−29のT細胞受容体を発現しているCD8+の豊富なPBMCは、遍在的な自己ペプチド(陽性対照:PRAME100〜108の積載されたT2細胞)の積載されたT2細胞(HLA−A*02陽性、PRAME陰性)と共培養された場合にはIFN−γの分泌を全く示さず、これによりTCR4.8−1−29の高い特異性が反映される(図6)。
実験の設計図:ペプチドでパルスされたT2細胞の溶解
・PRAME100〜108(VLD)ペプチドでパルスされたT2細胞の溶解を、T4.8−1−29TCRを発現しているCD8の豊富なPBMCとの共培養中の蛍光標識された標的細胞の消失を決定する、TVA(商標)蛍光死滅アッセイ(CTL、セルラー・テクノロジー有限会社、米国)を使用することによって測定した。
・標的細胞:TCR T4.8−1−29を発現しているPBMCと共培養された10−5MのPRAME100〜108(VLD)(関連性のある)ペプチド又はSLL(SLLQHLIGL(配列番号229)、PRAME、無関係)ペプチドを用いて段階的なE:T比でパルスされたT2細胞(HLA−A*02陽性、PRAME陰性)。
・T4.8−1−29を発現しているPBMCは、低いE:T比でさえ、関連性のある(VLD)ペプチドの積載されたT2細胞の効率的な溶解を示す。
・関連性のないSLLペプチド(PRAME)の積載されたT2細胞は、どのE:T比でも溶解されなかった(陰性対照)(図7)。
・腫瘍細胞に対する細胞障害活性を、T細胞クローンT4.8−1−29のトランスジェニックTCRを発現しているPBMCとの共培養中の蛍光標識された標的細胞の消失を検出する、TVA(商標)蛍光死滅アッセイ(CTL、セルラー・テクノロジー有限会社、米国)を使用して分析した。
・標的細胞:ヒト腫瘍細胞株K562を実験に使用した。K562細胞を、ヒトHLA−A*02:01をコードしているivtRNA及び/又はヒトPRAMEをコードしているivtRNAを使用してトランスフェクトした。ヒトK562は、内因性PRAME発現を示す(ナノストリングによって決定されそして文献に報告されているような)。さらに、PRAME発現は、ヒトPRAMEをコードしているivtRNAを用いてのK562細胞のトランスフェクションによって、又はPRAME100〜108VLDペプチドの外来的積載によって増加した。
○HLA−A*02:01をコードしているivtRNAを用いてトランスフェクトされ、PRAME100〜108(VLD)ペプチドの積載されたK562:K562−(PRAME+/A2−)+A2−ivtRNA+VLDペプチド(図8A)。
○PRAMEをコードしているivtRNAを用いて形質導入されたK562:K562−(PRAME+/A2−)+PRAME−ivtRNA(図8B)。
○PRAMEをコードしているivtRNA及びHLA−A*02をコードしているivtRNAを用いてトランスフェクトされたK562:K562−(PRAME+/A2−)+A2−ivtRNA(図8C)。
○HLA−A*02をコードしているivtRNAを用いてトランスフェクトされたK562:K562−(PRAME+/A2−)+A2−ivtRNA+PRAMEivtRNA(図8D)。
・腫瘍細胞を、TCR T4.8−1−29を発現しているPBMCと段階的なE:T比で共培養した。
・PRAME ivtRNAを用いてのトランスフェクション、並びに、HLA−A*02:01を発現しているK562細胞へのVLDペプチドの積載は、トランスジェニックTCR T4.8−1−29を発現しているPBMCによる比溶解を増加させた(図8A〜D)。
実験の設計図:腫瘍細胞株による刺激
・IFN−γELISAを使用して、一連のヒト腫瘍細胞株を用いての刺激後のサイトカイン分泌を評価した(PRAMEの発現状態を、NanoString nCounter(登録商標)分析によって検出した)。
・T細胞受容体T4.8−1−29を発現しているCD8+の豊富なPBMCと、K562−B35、K562−A2、Mel−624.38、Colo−678、及びSKMEL23との共培養から最大24時間後に上清を収集した。特異的なIFN−γ分泌を、標準的なELISAを使用して評価した。
・標的細胞:
○K562−B35 (HLA−A*02陰性、PRAME陽性)
○K562−A2 (HLA−A*02陽性、PRAME陽性)
○K562−A2 VLDペプチドが積載された(HLA−A*02陽性、PRAME陽性)
○Mel−624.38 (HLA−A*02陽性、PRAME陽性)
○SkMEL23 (HLA−A*02陽性、PRAME陽性)
・T細胞受容体T4.8−1−29を発現しているCD8+の豊富なPBMCは、PRAME陽性、HLA−A*02陽性腫瘍細胞株K562−A2、さらにVLDペプチドの積載されたK562−A2との共培養液中では高いIFN−γ分泌を示し、PRAME陽性、HLA−A*02陽性、Mel−624.38、及びSkMEL23との共培養時には中程度のIFN−γ分泌を示した。
・HLA−B*35の発現を有するPRAME陽性K562はIFN−γ分泌を誘導せず、これにより、TCR T4.8−1−29のHLA−A*02拘束性が確認された。
・健康なドナーのCD8の豊富なPBMCを、TCR T4.8−1−29構築物を含有しているプラスミドを用いて形質導入した。TCRの形質導入効率を分析するために、形質導入されていない及びTCR T4.8−1−29の形質導入されたPBMCの表面染色後にFACS分析を実施した。細胞を、CD8、及びTCRβ鎖のTCR可変領域(TRBV9)に対して特異的な抗体を用いて染色した。対照エフェクター細胞個体群において、内因的にTRBV9を発現しているT細胞は8%存在するが、形質導入後にはT細胞の60%がTRBV9を発現した。このことは、50%を超える形質導入効率を示す(図11)。
・PRAME100〜108(VLD)ペプチドの認識についての機能的T細胞の結合力を、T細胞クローンT4.8−1−29(実線の曲線)又はT4.8−1−29を用いて形質導入されたエフェクターPBMC(点線の曲線)のいずれかを、ペプチドの積載されたT2細胞と共培養した後のIFN−γ分泌の検出によって測定した。T2細胞に、10−5M〜10−12Mの濃度範囲の滴定量のペプチドを積載した。共培養上清を、共培養から約24時間後に収集し、標準的なELISAによって評価し、相対的なIFN−γ遊離は最大遊離に対する比率で示される。機能的結合力を規定する最大半量のIFN−γ分泌(EC50)は点線によって示される。元来のT細胞クローン及びトランスジェニックTCRの機能的結合力は非常に類似している(図12)。
・抗原特異性を分析するために、T4.8−1−29の形質導入されたエフェクターPBMC及び形質導入されていない対照PBMCを、異なる標的細胞と共培養した。腫瘍細胞株OPM−2及びU937(HLA−A2陰性及びPRAME陰性)を、改変せずに、又はHLA−A2をコードしているivtRNAを用いてトランスフェクトさせて試験した。さらに、該細胞はまた、HLA−A2とPRAMEをコードしているivtRNA、又はHLA−A2と無関係の抗原をコードしているivtRNAの組合せを用いてトランスフェクトした後に試験した。対照として、エフェクターもまた、PRAMEVLDペプチド(10−5M)又は無関係のPRAMESLLペプチド(10−5M)の積載されたT2細胞と共に培養した。24時間共培養した後、上清を収集し、IFN−γの分泌量を、標準的なELISAによって測定した。VLDの積載されたT2細胞との共培養液中のTCRの形質導入されたPBMCについて、多量のIFN−γが測定された。また、HLA−A2及び抗原PRAMEを用いてトランスフェクトされた両方の腫瘍細胞株が、TCRの形質導入されたPBMCによるIFN−γ分泌を誘導した。抗原PRAMEと組み合わせた、必要とされるMHC拘束性成分としてHLA−A2を発現している腫瘍細胞のみが認識され、これによりT4.8−1−29を発現しているPBMCが活性化された(図13)。
・抗原特異性を分析するために、T4.8−1−29の形質導入されたエフェクターPBMC及び形質導入されていない対照PBMCを、様々な標的細胞株と共培養した。腫瘍細胞株K562(HLA−A2陰性及びPRAME陽性)並びにK562_A2及びMel624.38(HLA−A陽性及びPRAME陽性)及び647−V(HLA−A2陽性及びPRAME陰性)を試験した。対照として、エフェクターも、PRAMEVLDペプチド(10−5M)又は無関係のPRAMESLLペプチド(10−5M)の積載されたT2細胞と共培養した。24時間共培養した後、上清を収集し、IFN−γの分泌量を標準的なELISAによって測定した。VLDの積載されたT2細胞との共培養中のTCRの形質導入されたPMBCにおいて多量のIFN−γが測定された。
・腫瘍細胞に対するT4.8−1−29の形質導入されたエフェクターの細胞障害活性を、細胞のライブイメージングを可能とする顕微鏡に基づいたシステムであるIncuCyte ZOOM(登録商標)(生細胞分析システム(エッセンバイオサイエンス社))を使用して分析した。
・T4.8−1−29を発現しているPBMCの安全性プロファイルを分析するために、健康なヒト組織の認識を排除しなければならない。それ故、2人の異なるドナーに由来するT4.8−1−29の形質導入されたPBMCを、HLA−A2陽性ドナーの健康な組織から得られた細胞と共培養した。一例として、形質導入されたPBMC並びに形質導入されていないPBMCを、ヒト腎臓毛細血管上皮細胞(HRCEpC)と共培養した。対照としてHRCEp細胞にさらに、VLDペプチド(10−5M)を積載した。24時間共培養した後、上清を収集し、IFN−γの分泌量を標準的なELISAによって測定した。TCRの形質導入されたPBMCは、ペプチドの積載された標的細胞との共培養時にのみ活性化されたが、改変されていないHRCEp細胞は全く認識されなかった。
Claims (16)
- TCRα鎖及びTCRβ鎖を含む、T細胞受容体であって、(i)TCRα鎖可変領域は、配列番号15に示されるアミノ酸配列を含み、そして(ii)TCRβ鎖可変領域は、配列番号16に示されるアミノ酸配列を含み、
該TCRは、VLDGLDVLL(配列番号32)のアミノ酸配列内に含まれるエピトープ又はそのMHC結合形に結合することができる、該T細胞受容体(TCR)。 - (iii)配列番号15に示されるアミノ酸配列からなるTCRα鎖可変領域、及び
(iv)配列番号16に示されるアミノ酸配列からなるTCRβ鎖可変領域
を含む、請求項1記載のTCR。 - (i)配列番号7、配列番号9、配列番号11、若しくは配列番号13から選択されたアミノ酸配列を含むか若しくはからなるTCRα鎖;及び
(ii)配列番号8、配列番号10、配列番号12、若しくは配列番号14から選択されたアミノ酸配列を含むか若しくはからなるTCRβ鎖
を含む、請求項1記載のTCR。 - 前記TCRが、天然TCR、TCR変異体、TCR断片、又はTCR構築物から選択される、請求項1〜3のいずれか一項記載のTCR。
- TCRヘテロ二量体又は多量体を形成するように互いに共有結合で連結された少なくとも1本のTCRα鎖(群)及び少なくとも1本のTCRβ鎖(群)を含む、請求項4記載のTCR構築物。
- 場合により少なくとも1つのリンカーをさらに含む、Fc受容体;IgA、IgD、IgG、IgE、及びIgMを含む、Fcドメイン;IL−2又はIL−15を含むサイトカイン;毒素;抗CD3抗体、抗CD28抗体、抗CD5抗体、抗CD16抗体、又は抗CD56抗体、又はその抗原結合断片を含む、抗体又はその抗原結合断片;CD247(CD3ζ)、CD28、CD137、CD134ドメイン、又はその組合せから場合により選択された1つ以上の融合成分(群)をさらに含む、請求項1〜5のいずれか一項記載のTCR。
- (i)配列番号15に示されるアミノ酸配列を含むか又はからなるTCRα鎖可変領域を含む、少なくとも1つのTCRα鎖;
(ii)配列番号16に示されるアミノ酸配列を含むか又はからなるTCRβ鎖可変領域を含む、少なくとも1つのTCRβ鎖;及び
(iii)リンパ球の表面上の抗原又はエピトープに対して指向された、抗体又は一本鎖抗体断片(scFv)
を含み、
TCRα鎖(群)及びTCRβ鎖(群)は互いに連結され、場合によりリンカーを介して該抗体又はscFvに融合している、請求項1〜6のいずれか一項記載のTCR。 - 請求項1〜7のいずれか一項記載のTCRをコードしている核酸。
- 配列番号23、24、25、26、27、28、29、又は30の核酸配列を含む、請求項8記載の核酸。
- 請求項8又は9のいずれか一項記載の核酸を含むベクター。
- 請求項1〜7のいずれか一項記載のTCR、請求項8若しくは9記載の核酸配列、又は請求項10記載のベクターを含む、宿主細胞。
- (i)前記TCRの発現を引き起こす条件下で請求項11記載の宿主細胞をインキュベートする工程、及び
(ii)該TCRを精製する工程
を含む、請求項1〜7のいずれか一項記載のTCRを得るための方法。 - (i)請求項1〜7のいずれか一項記載のTCR;
(ii)請求項8若しくは9のいずれか一項記載の核酸、
(iii)請求項10記載のベクター;及び/又は
(iv)請求項11記載の宿主細胞、
のうち1つ以上、及び場合により薬学的賦形剤(群)
を含む、医薬組成物又は診断用組成物。 - がんの検出、診断、予後予測、予防及び/又は治療に使用するための、請求項1〜7のいずれか一項記載のTCR、請求項8若しくは9記載の核酸、請求項10記載のベクター、及び/又は請求項11記載の宿主細胞。
- (a)被験者の試料を準備する工程、該試料は1つ以上の細胞を含む;
(b)該試料を、
(i)請求項1〜7のいずれか一項記載のTCR;
(ii)請求項11記載の宿主細胞、及び/又は
(iii)請求項13記載の医薬組成物
と接触させることにより複合体を形成する工程、並びに
(c)該複合体を検出する工程、該複合体の検出は、被験者におけるがんの存在の指標である
を含む、インビトロにおいて被験者におけるがんの存在を検出する方法。 - 改変されたリンパ球を生成するための、請求項1〜7のいずれか一項記載のTCR、請求項8若しくは9記載の核酸、及び/又は請求項10記載のベクターの使用。
