本発明は本明細書に記述された特定の方法および実験条件に限定されないが、それはこのような方法および条件は変化する場合があるからである。本発明の範囲は請求項によって定義されるので、本明細書に使用される用語は、特定の実施形態のみを記述する目的で使用されており、意図しないことも理解される。

Pdcd1(CD279とも呼ばれる)は、アポトーシスを受けているT細胞ハイブリドーマの上方制御された遺伝子として最初に発見された(Ishida, Y. et al. (1992) EMBO J. 11(11):3887−3895)。Pdcd1遺伝子は、N末端免疫グロブリンV(IgV)領域を含むタイプI膜タンパク質(PD−1と呼ばれる)であるPD−1をコードする5つのエクソン、柄(〜20アミノ酸の長さ)、膜貫通領域、および免疫受容体チロシン依存性抑制モチーフ(ITIM)および免疫受容体チロシン依存性スイッチモチーフ(ITSM)の両方を含む細胞内尾部から成る。PD−1は、例えば、B細胞、樹状細胞、活性化単球、ナチュラルキラー(NK)細胞および活性化T細胞などの、多くの細胞タイプ上で発現される(Keir, M.E., et al. (2008) Annu. Rev. Immunol. 26:677−704)。PD−1のさまざまなスプライスバリアントも報告されており、どのエクソンを欠くかに基づいて異なる(Nielsen, C. et al. (2005) Cell. Immunol. 235:109−116)。実際に、特定のスプライスバリアントは、自己免疫性疾患の原因因子として観察されている(Wan, B. et al. PD−1タンパク質をコードする非ヒト動物の内因性遺伝子座(例えば、Pdcd1遺伝子座)の遺伝子改変から生じる非ヒト動物の細胞表面上に、ヒト化PD−1タンパク質を発現する非ヒト動物が提供されている。本明細書に記述された適切な例には齧歯類、特にマウスが含まれる。
PD−1機能の調査ではさまざまなPdcd1変異体およびトランスジェニック非ヒト動物の使用を採用した(例えば、Nishimura, H. et al. (1998) Intern. Immunol. 10(10):1563−1572; Nishimura, H. et al. (1999) Immunity 11:141−151; Nishimura, H. et al. (2001) Science 291:319−322; Iwai, Y. et al. (2004) Intern. Immunol. 17(2):133−144; Keir, M. E. et al. (2005) J. Immunol. 175:7372−7379; Keir, M. E. et al. (2007) J. Immunol. 179:5064−5070; Carter, L.L. et al. (2007) J. Neuroimmunol. 182:124−134; Chen, L. et al. (2007) Europ. Soc. Organ Transplant. 21:21−29; Okazaki, T. et al. (2011) J. Exp. Med. 208(2):395−407;米国特許第7,414,171号、およびヨーロッパ特許第1 334 659 B1を参照、これらは参照により本明細書に組み込まれる)。このような変異体およびトランスジェニック動物は、PD−1発現の分子側面、さまざまな細胞プロセスの機能および調節を決定する上で有用であった。しかし、それらには限界がある。例えば、マウスPdcd1遺伝子のエクソン1への、ヒトPD−1 cDNAのノックインによって作製されたPD−1欠損マウスは、PMA(Carter, L.L.ら、上記参照)で刺激した後でもヒトPD−1を発現しなかった。さらに、異なる遺伝的背景のPD−1変異動物での表現型のかなりの差が、特に、さまざまな機能および/または調節活性をPD−1に割り当てようとする際、調査を複雑化した。それでも、PD−1を過剰発現するその他のトランスジェニック動物が作製されてきた(Chen, L.ら、上記参照)。このような動物は、導入遺伝子の異なる発現パターンを示しており、これは構築物のデザインに合理的に起因しうる。さらに、同じソース遺伝物質(すなわちマウス)を使用したために、PD−1過剰発現は、導入遺伝子の潜在的な位置効果のために、トランスジェニックPD−1ではなく、内因性PD−1に対応していた可能性がある。PD−1トランスジェニックマウスは、一部のPD−1媒介生物機能を明らかにする上で有用なことが示されているが、それらは得られた結果で変動性を示しており、これは少なくとも一部は、マウスを作製するために用いられた異なるアプローチに基づく。従って、PD−1媒介生物学を利用している現在のインビボシステムは不完全である。PD−1媒介生物機能およびシグナル伝達経路の分子側面は、トランスジェニックマウスではその潜在的能力の限界まで利用されていない。
この例は、齧歯類(例えば、マウス)などの非ヒト哺乳動物のプログラム細胞死1(PD−1)をコードする内因性Pdcd1遺伝子をヒト化する例示的方法を示している。この例に記述された方法は、任意のヒト配列、または望ましいヒト配列(または配列フラグメント)の組み合わせを使用して、非ヒト動物の内因性Pdcd1遺伝子をヒト化するために用いることができる。この例では、GenBank登録番号NM_005018.2(配列番号:23)に見られるヒトPDCD1遺伝子のエクソン2、イントロン2、およびエクソン3の最初の71 bpを含む〜883 bpのヒトDNAフラグメントが、マウスの内因性Pdcd1遺伝子のヒト化のために用いられている。内因性Pdcd1遺伝子の細胞外N末端IgV領域をコードする遺伝物質のヒト化のための標的化ベクターは、VELOCIGENE(登録商標)技術を使用して作製された(米国特許第6,586,251号およびValenzuela et al., 2003, Nature Biotech. 21(6):652−659を参照、これらは参照により本明細書に組み込まれる)。

この例は、PD−1調節因子(例えば、抗PD−1抗体)をスクリーニングし、例えば、腫瘍増殖の阻害および/または腫瘍細胞の殺傷などのさまざまな特徴を決定するために、実施例1に従ってヒト化Pdcd1遺伝子を含むように改変された非ヒト動物(例えば、齧歯類)をインビボアッセイで使用できることを実証する。この例では、腫瘍成長を阻害する最適な抗体用量および抗PD−1抗体が腫瘍細胞の殺傷を媒介する程度を決定するために、実施例1に記述された内因性Pdcd1遺伝子のヒト化に対してホモ接合性のマウスで、いくつかの抗PD−1抗体がスクリーニングされる。
この例は、実施例1によるヒト化Pdcd1遺伝子を含むように改変された非ヒト動物(例えば、齧歯類)は、PD−1調節因子(例えば、抗PD−1抗体)の最適な治療用量を決定するための腫瘍モデルに使用できることを示している。この例では、抗PD−1抗体が、定着腫瘍の治療のための最適治療用量を決定するために、実施例1に記述された内因性Pdcd1遺伝子のヒト化に対してホモ接合性のマウスに投与される。

前記齧歯類が前記ヒト化Pdcd1遺伝子からヒト化プログラム細胞死1(PD−1)ポリペプチドを発現し、
前記ヒト化PD−1ポリペプチドがヒト部分および内因性部分を備え、
前記ヒト部分がヒトPD−1ポリペプチドの細胞外部分を備え、前記内因性部分が内因性齧歯類PD−1ポリペプチドの細胞内部分を備え、
前記細胞外部分が、ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部によってコードされ、
前記齧歯類がマウスまたはラットである、齧歯類。 - 前記ヒト化PD−1ポリペプチドが齧歯類シグナルペプチドで前記齧歯類の細胞に翻訳される、請求項1に記載の齧歯類。
- 前記ヒト化PD−1ポリペプチドが内因性PD−1ポリペプチドの膜貫通部分をさらに備える、請求項1または2に記載の齧歯類。
- 前記ヒト部分がヒトPD−1ポリペプチドのアミノ酸26〜169を備える、請求項1〜3のいずれか一項に記載の齧歯類。
- 前記ヒト化Pdcd1遺伝子が内因性Pdcd1エクソン1、4および5を備える、請求項1〜4のいずれか一項に記載の齧歯類。
- 前記ヒト化Pdcd1遺伝子が内因性Pdcd1エクソン3の一部をさらに備える、請求項5に記載の齧歯類。
- 前記ヒト化Pdcd1遺伝子が齧歯類Pdcd1プロモーターと動作可能なように連結される、請求項1〜6のいずれか一項に記載の齧歯類。
- 前記齧歯類Pdcd1プロモーターが内因性齧歯類Pdcd1プロモーターである、請求項7に記載の齧歯類。
- 前記ヒト化Pdcd1遺伝子が、前記内因性Pdcd1遺伝子のゲノムフラグメントの、ヒトPdcd1遺伝子のゲノムフラグメントによる置換から生じる、請求項1〜8のいずれか一項に記載の齧歯類。
- 前記ヒト化Pdcd1遺伝子が、前記内因性Pdcd1遺伝子のエクソン1、前記ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部、前記内因性Pdcd1遺伝子のエクソン3の一部、および前記内因性Pdcd1遺伝子のエクソン4〜5を備える、請求項9に記載の齧歯類。
- 単離齧歯類細胞または組織であって、そのゲノムが内因性Pdcd1遺伝子座にヒト化Pdcd1遺伝子を備え、
前記ヒト化Pdcd1遺伝子がヒト化PD−1ポリペプチドをコードし、前記ヒト化PD−1ポリペプチドがヒト部分および内因性部分を備え、
前記ヒト部分がヒトPD−1ポリペプチドの細胞外部分を備え、前記内因性部分が内因性齧歯類PD−1ポリペプチドの細胞内部分を備え、
前記細胞外部分が、ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部によってコードされ、
前記齧歯類がマウスまたはラットであり、前記齧歯類細胞が随意で齧歯類胚性幹細胞である、単離齧歯類細胞または組織。 - 請求項11に記載の齧歯類胚性幹細胞から生成される齧歯類胚。
- 齧歯類を作る方法であって、そのゲノムがヒト部分および内因性部分を持つPD−1ポリペプチドをコードするPdcd1遺伝子を備え、前記部分が齧歯類Pdcd1プロモーターと動作可能なように連結され、前記方法が、
改変されたゲノムが内因性Pdcd1遺伝子座にヒト化Pdcd1遺伝子を備えるように齧歯類のゲノムを改変し、前記改変されたゲノムを備える齧歯類を得る工程を含み、
前記齧歯類が前記ヒト化Pdcd1遺伝子からヒト化PD−1ポリペプチドを発現し、前記ヒト化PD−1ポリペプチドがヒト部分および内因性部分を備え、
前記ヒト部分がヒトPD−1ポリペプチドの細胞外部分を備え、前記内因性部分が内因性齧歯類PD−1ポリペプチドの細胞内部分を備え、
前記細胞外部分が、ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部によってコードされ、
前記齧歯類がマウスまたはラットである、方法。 - 内因性Pdcd1遺伝子からPD−1ポリペプチドを発現する齧歯類を作る方法であって、前記PD−1ポリペプチドがヒト配列を備え、前記方法が、
(a)齧歯類胚性幹細胞の内因性Pdcd1遺伝子座における内因性齧歯類Pdcd1遺伝子にヒトゲノムフラグメントを挿入して、ヒト化PD−1ポリペプチドをコードするヒト化Pdcd1遺伝子を形成させる工程であって、
前記ヒト化PD−1ポリペプチドがヒト部分および内因性部分を備え、
前記ヒト部分がヒトPD−1ポリペプチドの細胞外部分を備え、前記内因性部分が内因性齧歯類PD−1ポリペプチドの細胞内部分を含み、
前記細胞外部分が、ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部によってコードされ、
前記齧歯類がマウスまたはラットである工程と、
(b)(a)で生成された齧歯類胚性幹細胞を得る工程と、
(c)(b)の齧歯類胚性幹細胞を使用して齧歯類を作る工程とを含む方法。 - 前記ヒト化PD−1ポリペプチドが齧歯類シグナルペプチドで前記齧歯類の細胞に翻訳される、請求項13または14に記載の方法。
- 前記ヒト化PD−1ポリペプチドが内因性PD−1ポリペプチドの膜貫通部分をさらに備える、請求項13〜15のいずれか一項に記載の方法。

- 前記ヒト化Pdcd1遺伝子が、前記内因性Pdcd1遺伝子のゲノムフラグメントの、ヒトゲノムフラグメントによる置換から生じる、請求項13〜21のいずれか一項に記載の方法。
- 前記ヒト化Pdcd1遺伝子が、前記内因性Pdcd1遺伝子のエクソン1、前記ヒトPdcd1遺伝子のエクソン2およびエクソン3の一部、前記内因性Pdcd1遺伝子のエクソン3の一部、および前記内因性Pdcd1遺伝子のエクソン4〜5を備える、請求項22に記載の方法。
- ヒトPD−1を標的とする薬剤の薬物動態特性を評価する方法であって、前記方法が、
請求項1〜10のいずれか一項に記載の齧歯類に、前記薬剤を投与する工程と、
ヒトPD−1を標的とする前記薬剤の一つ以上の薬物動態特性を決定するためにアッセイを実施する工程とを含み、ここで、前記薬剤は随意で抗PD−1抗体である、方法。 - 齧歯類腫瘍モデルを生産する方法であって、
a)請求項1〜10のいずれか一項に記載の齧歯類を提供する工程と、
b)(a)の前記齧歯類に一つ以上の腫瘍細胞を移植し、
それによって前記齧歯類腫瘍モデルを得る工程と
を含む、方法。
