Claims (11)

前記第1のオリゴマー化ドメインおよび/または第2のオリゴマー化ドメイン上の前記変異の少なくとも1つは、前記少なくとも1つの第1のオリゴマー化サブユニットおよび前記少なくとも1つの第2のオリゴマー化サブユニットの自己オリゴマー化を妨げる破壊変異であり、
前記αHLモノマーおよび/またはαHLモノマーの前記コンカテマーが、1以上の配列番号3のポリペプチドを含み、
少なくとも1つの配列番号3のポリペプチドが、配列番号3の位置12または17に置換をさらに含むバリアントであり、
前記置換が1以上の正電荷を含み、T12K、T12R、N17KおよびN17Rからなる群から選択される少なくとも1つであり、
前記七量体が厳密に7つのαHLモノマーを含み、各αHLモノマーが第1のオリゴマー化ドメインおよび第2のオリゴマー化ドメインを含み、
1つのαHLモノマーの第1のオリゴマー化ドメインが他のαHLモノマーの第2のオリゴマー化ドメインに連結され、
脂質二重層中に細孔を形成する能力を保持する、
前記ヘテロオリゴマーα−溶血素(αHL)七量体。 - 前記少なくとも1つの第1のオリゴマー化サブユニット中の前記第1のオリゴマー化ドメイン上に少なくとも1つの同族および/もしくはレスキュー変異、ならびに/または前記少なくとも1つの第2のオリゴマー化サブユニット中の前記第2のオリゴマー化ドメイン中の少なくとも1つの同族および/またはレスキュー変異をさらに含み、
前記少なくとも1つの同族および/またはレスキュー変異が、前記少なくとも1つの第1のオリゴマー化サブユニットおよび前記少なくとも1つの第2のオリゴマー化サブユニット間のサブユニット間接触を決定し、
前記ヘテロオリゴマーαHL七量体中のオリゴマー化サブユニットの順序を特定する、
請求項1に記載のヘテロオリゴマーαHL七量体。 - 前記αHL七量体が、αHLモノマーのコンカテマーである少なくとも1つの第1のオリゴマー化サブユニットによって形成され、少なくとも1つの第2のオリゴマー化サブユニットは、少なくとも1つのαHLモノマーである、請求項1または2に記載のヘテロオリゴマーαHL七量体。
- 各々がαHLモノマーのコンカテマーである、少なくとも1つの第1のオリゴマー化サブユニットおよび少なくとも1つの第2のオリゴマー化サブユニットによって形成される、請求項1または2に記載のヘテロオリゴマーαHL七量体。
- 前記少なくとも1つの第1のオリゴマー化サブユニットおよび前記少なくとも1つの第2のオリゴマー化サブユニットが、αHLモノマーの各々である、請求項1または2に記載のヘテロオリゴマーαHL七量体。
- 前記αHLモノマーおよび/または前記αHLモノマーのコンカテマーが、1以上の配列番号3のポリペプチドを含む、請求項1〜5のいずれか一項に記載のヘテロオリゴマーαHL七量体。
- ポリメラーゼをさらに含み、
前記ポリメラーゼが、前記少なくとも1つの第1のオリゴマー化サブユニットまたは第2のオリゴマー化サブユニットのうち1以上に付けられている、請求項1〜6のいずれか一項に記載のヘテロオリゴマーαHL七量体。 - 請求項1〜7のいずれか一項に記載のヘテロオリゴマーαHL七量体を含む七量体細孔構築体。
- 標的分子を検出するための方法であって、
(a)検出電極に隣接して、またはその近傍に配置された膜中に、請求項8に記載の七量体細孔構築体を含むナノポアを含むチップを設置するステップと、
(b)核酸分子を前記ナノポア中に通過させるステップであって、前記核酸分子が、リポーター分子と会合され、前記核酸分子が、アドレス領域およびプローブ領域を含み、前記リポーター分子が、前記プローブ領域で前記核酸分子と会合され、前記リポーター分子が、標的分子と連結されるステップと、
(c)前記核酸分子が前記ナノポア中を通過させられる間に前記アドレス領域をシーケンシングして、前記アドレス領域の核酸配列を調べるステップと、
(d)(c)において調べられた前記アドレス領域の核酸配列に基づいて、コンピュータプロセッサーを利用して前記標的分子を同定するステップと
を含む、前記方法。 - 前記第1のオリゴマー化ドメイン中の前記少なくとも1つの同族変異が、配列番号3におけるH35Iであり、前記第2のオリゴマー化ドメイン中の前記少なくとも1つの同族変異が、配列番号3におけるY101Hである、請求項2に記載のヘテロオリゴマーαHL七量体。
- 前記1以上の変異が、配列番号3におけるH144の位置での置換を含む、請求項1〜7のいずれか1項に記載のヘテロオリゴマーαHL七量体。
