Description
したがって、黄斑変性などの眼疾患のより根本的かつ持続的な治療技術の開発が要求される。
一例は、VEGF−A遺伝子を不活性化させる製剤を含む、眼疾患の予防および/または治療用組成物を提供する。
他の例は、VEGFA遺伝子の特定標的部位(target site or target region)を標的化するためのガイドRNAを提供する。
他の例は、前記ガイドRNAまたはVEGFA遺伝子特異的リボ核酸タンパク質(RNP)を含む薬学組成物を提供する。
前記不活性化の標的となるVEGF−A遺伝子は、眼、例えば、新生血管性眼疾患が発生した眼、または新生血管性眼疾患の病変部位に位置するものであってもよい。
(1)VEGF−A遺伝子の全体または1−50bpまたは1−40bpの長さの連続的または不連続的一部分の欠失;
(2)VEGF−A遺伝子中、1−20個、1−15個、または1−10個の連続的または不連続的なヌクレオチドの野生型VEGF−A遺伝子と相異なるヌクレオチドへの置換;
(3)VEGF−A遺伝子内の1−20個、1−15個、または1−10個のヌクレオチドの挿入(付加)、この時、前記挿入されるヌクレオチドは、それぞれ独立してA、T、C、およびGの中から選択される;および
(4)これらの組合せ。
(1)VEGF−A遺伝子内のCas9タンパク質のPAM(proto−spacer−adjacent Motif)配列に隣接した1−50bpまたは1−40bpの長さの連続的または不連続的部位に位置する1つ以上のヌクレオチドの欠失;
(2)VEGF−A遺伝子内のCas9タンパク質のPAM配列に隣接した1−50bpまたは1−40bpの長さの連続的または不連続的部位に位置する1−20個、1−15個、または1−10個の連続的または不連続的なヌクレオチドの野生型VEGF−A遺伝子と相異なるヌクレオチドへの置換;
(3)VEGF−A遺伝子内のCas9タンパク質のPAM配列に隣接した1−50bpまたは1−40bpの長さの連続的または不連続的部位に1−20個、1−15個、または1−10個のヌクレオチドの挿入(付加)、この時、前記挿入されるヌクレオチドは、それぞれ独立してA、T、C、およびGの中から選択される;および
(4)これらの組合せ。
(a)生体(または病変部位)または細胞などに投与される前にCas9タンパク質とVEGF−A遺伝子特異的ガイドRNAとが結合して形成された(つまり、投与前に予め組込まれた)複合体、つまり、リボ核酸タンパク質(ribonucleoprotein;RNP)形態であるか(この場合、リボ核酸タンパク質形態で細胞膜を通過して細胞内または生体内に伝達される)、
(b)Cas9タンパク質を暗号化するDNAおよびVEGF−A遺伝子特異的ガイドRNAを暗号化するDNAがそれぞれ別個のベクターを通して生体内または細胞内に投与(または伝達)されるか、1つのベクターを通して共に生体内または細胞内に投与(または伝達)されて、生体または細胞内で複合体をなすもの、
(c)Cas9タンパク質を暗号化するRNA(mRNA)およびVEGF−A遺伝子特異的ガイドRNAを含むRNA混合物形態、または
(d)Cas9タンパク質を暗号化する遺伝子(DNA)を含む組換えベクターおよびVEGF−A遺伝子特異的ガイドRNA(例えば、in vitro転写によって得られる)の混合物であってもよい。
(a)生体(または病変部位)または細胞などに投与される前にCas9タンパク質とVEGF−A遺伝子特異的ガイドRNAが結合して形成された(つまり、投与前に予め組込まれた)複合体、つまり、リボ核酸タンパク質(ribonucleoprotein;RNP)(この場合、リボ核酸タンパク質形態で細胞膜を通過して細胞内または生体内に伝達される);

1.Cas9 RNPの製造
精製されたCas9タンパク質は、ToolGen Inc.、South Koreaから購入して準備した。sgRNAは、T7ポリメラーゼ(New England Biolabs)を用いて、製造業者のプロトコルに従ってin vitro転写によって生成した。簡略に、2つの相補的なオリゴヌクレオチド(表1参照)をannealingおよびextensionしてsgRNAのテンプレートを生成した。
pET28−NLS−Cas9ベクター(図8;Cas9:Streptococcus pyogenes由来である(配列番号358))を大腸菌菌株BL21(DE3)に形質転換させた後、0.5mM isopropyl β−D−1−thiogalactopyranoside(IPTG)を処理して、18℃で12時間、Cas9タンパク質の発現を誘導した。前記大腸菌細胞を超音波処理して溶解させ、20,000gで30分間遠心分離した後、溶解性溶解液(soluble lysate)を取ってNi−NTAビーズ(Qiagen)と混合し、Cy3染料(GE Healthcare)を1:10の比率(Cas9タンパク質:Cy3染料分子)で添加した。前記混合物を暗条件および4℃条件で一晩(12時間以上)培養した。溶離緩衝液(elution buffer;50mM Tris−HCl[pH7.6]、150−500mM NaCl、10−25%(w/v)グリセロール、0.2Mイミダゾール)を用いてCy3−標識Cas9を溶出させ、透析用緩衝液(dialyzing buffer;20mM HEPES pH7.5、150mM KCl、1mM DTT、10%(w/v)グリセロール)を用いて透析した。精製されたCy3−標識Cas9タンパク質をUltracel 100K cellulose column(Millipore)を用いて濃縮させた。Cy3−標識Cas9タンパク質の純度はSDS−PAGEによって測定した。Cy3標識効率は、Cas9タンパク質(280nm)および接合されたCy3染料分子の吸収スペクトルを比較して測定した。
マウスNIH3T3(ATCCR CRL−1658TM)およびARPE−19(ヒト網膜色素上皮細胞;ATCCR CRL−2302TM)細胞株を、10%(v/v)BCSまたはFBSが補充されたDulbecco’s Modified Eagle Medium(DMEM)で、5%CO2、37℃、および湿潤大気条件下で培養した(NIH3T3およびARPE−19細胞はmycoplasma汚染の有無が試験されていない)。形質感染1日前に、NIH3T3およびARPE−19細胞を24−ウェルプレートに2x104細胞/ウェルの量で接種した。各ウェルには抗生剤無含有成長培地250μlが添加されている。
形質感染後1日目に、細胞を、室温で10分間、4%(w/v)PFA(paraformaldehyde)で固定させた後、室温で15分間、4’,6−diamidino−2−phenylindole(DAPI、1μg/ml、Sigma Aldrich)で染色した。前記細胞をx630倍率で共焦点顕微鏡(LSM510、Carl Zeiss)で視角化した。スキャニング媒介変数は次の通りにした:scaling(x=0.14μm/pixel、y=0.14μm/pixel、z=1μm/pixel)、dimensions(x=1024、y=1024、z=6、channels:3、12−bit)(with objective C−Apochromat 63x/1.20W Korr UV−VIS−IR)。ZEN2ソフトウェア(黒色板、Ver10.0、Carl Zeiss)を用いてCy3陽性核(Cy3 positive nuclei)を計数した。Cy3陽性核の頻度(frequency)を定量化するために、我々は倍率630倍で観察される視野範囲でCy3染色された核を有する細胞の数と全体細胞数を数え、4つの視野にわたって観察されるCy3陽性核の平均百分率を計算した(n=3)。
DNeasy Tissue Kit(Qiagen)を用いて、製造業者のプロトコルに従って細胞および組織から遺伝体DNA(genomic DNA)を分離した。PCRを用いて標的部位を増幅させた後、生成物を熱循環器を用いて変性させアニーリングさせた。この時使用されたプライマーを下記の表2にまとめた:
Phusionポリメラーゼ(Thermo Fisher Scientific)を用いて、遺伝体DNAからオン−ターゲット(on−target)および潜在的オフ−ターゲット(off−target)領域を増幅させた。Illumina MiSeq(LAS Inc.韓国)を用いてPCR増幅物の対をなした配列の分析を行った。使用されたプライマーを表3〜表5にまとめた:
easy−spinTM Total RNA抽出キット(iNtRON、韓国)を用いて、製造会社のプロトコルに従ってNIH3T3およびARPE−19細胞から全体RNAを分離した。Superscript II(Enzynomics、South Korea)を用いて250ngのRNAを逆転写させた。SYBR Green(KAPA)および次のプライマーを用いて定量的PCR(Quantitative PCR;qPCR)を行った:
ヒトVEGFA ELISAを行うために、Vegfa−特異的Cas9 RNP処理されたconfluent ARPE−19細胞を無血清(serum−free)培地で16時間培養した後、前記細胞培養物から無血清上澄液(supernatant)を収集し、ヒトVEGF Quantikine ELISA Kit(DVE00、R&D systems)を用いて、製造会社の指針に従って分泌されたVEGFAタンパク質水準を測定した。
DNeasy Tissue Kit(Qiagen)を用いてARPE−19細胞(ATCC)から遺伝体DNAを分離した。Digenome sequencingのために、遺伝体DNAを次の方法でin vitro切断した。簡単に説明すれば、遺伝体DNA(20μg)を、Cas9タンパク質(16.7μg)およびsgRNA(12.5μg)と共に反応緩衝液(100mM NaCl、50mM Tris−HCl、10mM MgCl2、100μg/ml BAS、pH7.9)で、37℃の条件で3時間インキュベーションして、Cas9による遺伝体DNAの切断が起こるようにした。切断された遺伝体DNAをRNase A(50μg/ml、Sigma Aldrich)で37℃で30分間処理し、DNeasy Tissue Kit(Qiagen)で精製した。Whole−genome sequencingとDigenome sequencingは、関連技術分野で知られた方法で行った(Kim,D.,Kim,S.,Kim,S.,Park,J.&Kim,J.S.Genome−wide target specificities of CRISPR−Cas9 nucleases revealed by multiplex Digenome−seq.Genome research26,406−415(2016))。
本明細書の実施例で使用されたすべての動物の管理、使用および治療は、Seoul National University Institutional Animal Care and Use Committeeで定めたガイドラインおよび「ARVO statement for the Use of Animals in Ophthalmic and Vision Research」を厳格に遵守して行った。成体(6週齢)雄SPF C57BL/6Jマウスを研究に使用した。マウスは12時間/12時間の明/暗周期下で維持させた。
tiletamineとzolazepamを1:1の重量比で混合した混合物を2.25mg/kg(体重)の量で腹腔内注射してマウスを麻酔させた。phenylephrine(0.5%(w/v))とtropicamide(0.5%(w/v))が含まれている点眼液でマウスの瞳孔を拡張させた。Indirect head set delivery system(Iridex)とレーザシステム(Ilooda)を用いてレーザ光凝固(Laser photocoagulation)を行った。レーザ波長は532nmとした。レーザ媒介変数(Laser parameters)は次の通りである:スポットサイズ:200μm、電力:1W、および露出時間:100ms。変形した視神経円盤(optic disc)周囲の12時位置(右眼)または6時位置(左眼)にレーザ火傷(laser burn)を誘導させた。硝子体出血(vitreous hemorrhage)がない泡を発生させたレーザ火傷のみを試験対象に含ませた。レーザ火傷の四分円でRNPの網膜下注入を施した。Cas9 RNP(sgRosa26(Rosa26 targeting sgRNAを含む)またはsgVegfa(Vegfa targeting sgRNAを含む))は、各マウスの左眼または右眼にランダムに割り当てた。Cas9 RNPの網膜下注射によって水ぶくれ(bleb)ができた。この水ぶくれがレーザ火傷部位と重なったことを確認した。水ぶくれがレーザ火傷部位と重なった個体を後の試験に使用した。レーザ処理7日後、眼球を室温で1時間4%(w/v)PFA(paraformaldehyde)で固定させた。RPE(retinal pigment epithelium)複合体(RPE/脈絡膜/強膜)をisolectin−B4(Thermo Fisher Scientific、cat.no.I21413、1:100)で4℃で一晩処理して免疫染色させた。染色されたRPE複合体を蛍光顕微鏡(Eclipse 90i、Nikon)に平らに載せてx40倍率で観察した。CNV面積は、blind observerがImage Jソフトウェア(1.47v、NIH)を用いて測定した。
RPE複合体中のRPE細胞数を高倍率フィールド領域(high power field area)(100μmx100μm、n=8)でパラフィンに挿入された断片試料(cross section sample、4μm)内のDAPI染色された核を計数して算定した。注射後7日目に得られた断片試料(n=4)を、anti−opsin抗体(Millipore、AB5405、1:1000)とAlexa Fluor488抗体(Thermo Fisher Scientific、1:500)で免疫染色した。opsin positive領域は、blind observerがImage Jソフトウェア(1.47v、NIH)を用いて測定した。共焦点顕微鏡(LSM710、Carl Zeiss)を用いてRPE flat−mountにおけるCy3−Cas9タンパク質の細胞内分布をイメージングした。スキャニングパラメータは次の通りである:scaling(x=0.042μm/pixel、y=0.042μm/pixel、z=0.603μm/pixel)、dimensions(x=1024、y=1024、z=12、channels:2、8−bit)、and zoom(5.0)with objective C−Apochromat 40x/1.20W Korr M27.ZEN2ソフトウェアを用いてイメージを処理した。
RNPを注射して3日目にRPE複合体から遺伝体DNAを分離し、CNV面積の測定後にRNPを注射し、7日目にCNV試料から遺伝体DNAを分離し、遺伝体DNAの分離はNucleoSpin Tissue Kit(Macherey−Nagel)を用いて行った。RNP効能を評価するために、各RPE複合体を4四分円に分割し、RNPが注入された四分円を処理して遺伝体DNAを分離した。CNV領域のRPE複合体におけるindel頻度を評価するために、RPE flat−mountをイメージングした後、PBSで洗浄した。遺伝体DNAは次の2つの領域から分離した:(i)CNVのRNP注入領域を含む四分円(注入部位を代表する);および(ii)反対四分円(opposite quadrant;非注入部位を代表する)。
マウスVEGF−A ELISAの場合、眼球に計30個のレーザ火傷を誘発させた後、RNP(3μl)を網膜下空間(subretinal space)に注射した。注射後3日目、全体RPE複合体を網膜から分離し、追加分析のために凍結させた。RIPA緩衝液(50mM Tris−HCl(pH8.0)、150mM NaCl、1%lgepal CA−630、0.5%Na.deoxycholate、0.1%SDS)で細胞を溶解させ、マウスVEGF Quantikine ELISA Kit(MMV00、R&D systems)を用いて、製造業者の指示に従ってVEGF−A水準を測定した。
RNPの生体内伝達後、時間の経過によるRNP水準を分析するために、注射後1日および3日目に得られたRPE複合体に対してウェスタンブロッティングを行った。同量のタンパク質(20μg)を含有する試料を分析し;Cas9とアクチンは、それぞれ抗−HA高親和性抗体(Roche、1:1000)および抗−β−アクチン抗体(Sigma Aldrich、1:1000)を用いて検出した。ImageQuant LAS4000(GE healthcare)を用いてデジタルイメージングした。
データ分析はSPSSソフトウェアバージョン18.0(SPSS Inc.、Chicago、IL、USA)を用いて行った。P valueはunpaired、two sided Student’s t−testまたはone−way ANOVAおよびTukey post−hoc test(多数の試験群の場合)によって決定した。データはs.e.m(standard error of the mean)と共に平均値で表した。
マウスNIH3T3およびヒト網膜色素上皮細胞株ARPE−19で、VEGF受容体1および2に対する結合部位を暗号化するVegfa遺伝子のエクソン3とエクソン4で標的部位をターゲッティングする4個の単一鎖ガイドRNAs(sgRNAs)(Vegfa−1、2、3および4で表示)を試験した。前記4種のsgRNA(Vegfa−1、2、3および4)は参考例1を参照して作製した。
in vitroおよびin vivoでのCas9 RNPの位置をモニタリングするために、Cy3が接合されたCas9タンパク質(参考例4)を用い、Vegfa−1 sgRNAと結合したり結合しないCy3−Cas9を陽イオン性脂質と混合してNIH3T3細胞に形質感染させるか、網膜下注射によりVegfa特異的Cy3標識または非標識Cas9 RNPを成体マウスの眼球に注入して、下記の試験を行った。
Cas9 RNPがAMDマウスモデルでCNVの治療に使用できるかを調べるために、レーザで誘導された脈絡膜血管新生(laser−induced CNV)を有するマウスにVegfa特異的Cas9 RNP(Vegfa−specific Cas9 RNP;Vegfa−1 sgRNAを含む)またはRosa26特異的Cas9 RNP(Rosa26−RNP;Rosa26 sgRNAを含む)を網膜下注射して、以下の試験を行った。網膜下注射は、それ自体でCNVの大きさを増加させるため、Rosa26−RNPを対照群として使用した。
ヒト遺伝体におけるVegfa−特異的Cas9 RNPの全長遺伝体標的特異性(Genome−wide target specificity)をDigenome−seqで確認した(参考例6および9参照)。
41個のDigenome−capture site(表5参照)およびOn−target配列を含む42個の配列のSequence logoを図4Bに示した。
targeted deep sequencingによってヒトARPE−19細胞から確認されたoff−target部位およびindel頻度を図4Cに示した。図4Cで、mismatchedヌクレオチドは赤色、PAM配列は青色でそれぞれ表した。
老人性黄斑変性(AMD)または糖尿病性網膜症のような眼科疾患の治療のためにVegfa遺伝子を突然変異させる時の他の主な関心事は、眼における栄養的側面からのVegfaの役割である。Vegfa突然変異時の最も深刻な変化は円錐細胞障害(Cone dysfunction)であるが、マウスRPEでのVegfa遺伝子の条件付き欠失3日後に観察される。
Claims (8)
- Cas9タンパク質と、VEGF−A遺伝子の標的部位に特異的に結合する標的化配列を含むVEGF−A遺伝子特異的ガイドRNAとを含むリボ核酸タンパク質を含む、VEGF−Aの過発現に関連する眼疾患の予防または治療用組成物であって、前記ガイドRNAは、VEGF−A遺伝子内の配列番号1〜8の中から選択された核酸配列を含む標的部位にハイブリダイズ可能なものである、前記組成物。
- 前記ガイドRNAは、配列番号9〜16の中から選択された核酸配列を含むものである、請求項1に記載の眼疾患の予防または治療用組成物。
- 前記VEGF−Aの過発現に関連する眼疾患は、黄斑変性または網膜病症である、請求項1または2に記載の眼疾患の予防または治療用組成物。
- Cas9タンパク質およびVEGF−A遺伝子の標的部位に特異的に結合する標的化配列を含むVEGF−A遺伝子特異的ガイドRNAを含むリボ核酸タンパク質であって、前記ガイドRNAは、VEGF−A遺伝子内の配列番号1〜8の中から選択された核酸配列を含む標的部位にハイブリダイズ可能なものである、前記リボ核酸タンパク質。
- 前記ガイドRNAは、配列番号9〜16の中から選択された核酸配列を含むものである、請求項4に記載のリボ核酸タンパク質。
- VEGF−Aの過発現に関連する眼疾患の予防または治療に用いるための、Cas9タンパク質と、VEGF−A遺伝子の標的部位に特異的に結合する標的化配列を含むVEGF−A遺伝子特異的ガイドRNAとを含むリボ核酸タンパク質を含む薬学組成物であって、前記ガイドRNAは、VEGF−A遺伝子内の配列番号1〜8の中から選択された核酸配列を含む標的部位にハイブリダイズ可能なものである、前記組成物。
- 前記ガイドRNAは、配列番号9〜16の中から選択された核酸配列を含むものである、請求項6に記載の眼疾患の予防または治療に用いるための薬学組成物。
- 前記VEGF−Aの過発現に関連する眼疾患は、黄斑変性または網膜病症である、請求項6または7に記載の眼疾患の予防または治療に用いるための薬学組成物。
