Description
本出願は、参照によりその全体が本明細書に組み込まれる2016年3月18日に出願された米国特許出願番号第62/310,640号への35U.S.C.§l19(e)の下での優先権を主張するものである。
Claims (16)
- マイクロ流体デバイス内に配置されたキャピラリフローチャネルであって、近位開口部及び遠位開口部を備えるキャピラリフローチャネルと、
フィルタを備え、前記近位開口部に配置されたフィルタポケットと、
前記フィルタポケットに対して遠位に配置された混合ウェルと、
前記混合ウェルに対して遠位に配置された、試薬を収容する乾燥試薬ゾーンと、
前記乾燥試薬ゾーンに対して遠位に配置されたピンチ領域と、
前記ピンチ領域に対して遠位に配置された検出ゾーンと、
を備え、
前記遠位開口部は、流量を制御するべく先細りしていて、
前記フィルタポケット、前記混合ウェル、前記乾燥試薬ゾーン及び前記ピンチ領域は、流体連通しており、
前記ピンチ領域は、当該マイクロ流体デバイスの長さに実質的に垂直な方向にローブしている、前記キャピラリフローチャネルの部分を有する、マイクロ流体デバイス。 - 前記フィルタポケットが、液体サンプルを受け入れるように構成された凹部を有するサンプル入口、フィルタランディング、及び前記液体サンプルの受け入れ時に空気が排気されることを可能にするように構成されたベントを備える、請求項1に記載のマイクロ流体デバイス。
- 前記フィルタランディングが、前記フィルタポケットの遠位縁から延びる***したプラトーを含む、請求項2に記載のマイクロ流体デバイス。
- 前記乾燥試薬ゾーンが、前記サンプルを前記試薬ゾーンの親水性領域に維持するように構成された、キャピラリフローに垂直な高さを有する疎水性インク壁を含む、請求項1に記載のマイクロ流体デバイス。
- 前記フィルタポケットが、血漿を前記サンプル入口から前記フィルタランディングへ誘導するために配置されたキャットウォークストリップをさらに備える、請求項2に記載のマイクロ流体デバイス。
- 前記検出ゾーンの洗浄中に過剰液体サンプルを保持するように構成された廃液チャネルをさらに備える、請求項1に記載のマイクロ流体デバイス。
- 前記基板の一部が前記廃液チャネルを覆い、該一部は、流量を増加させ且つ洗浄時間を減少させるべく疎水性インクでプリントされる、請求項6に記載のマイクロ流体デバイス。
- 前記キャピラリフローチャネルが、上側基板と下側基板との間に配置される、請求項1に記載のマイクロ流体デバイス。
- 前記下側基板が、第1の深さを有する第1の部分と、前記第1の深さよりも小さい第2の深さを有する第2の部分を備える、請求項8に記載のマイクロ流体デバイス。
- 前記第1の深さを有する部分が凸形であり、前記第2の深さを有する部分が平坦である、請求項9に記載のマイクロ流体デバイス。
- 前記フィルタポケットが、前記濾過された液体を毛管作用により前記キャピラリチャンバに沿って前記キャピラリチャネルの混合ウェルへ移動させるように構成される、請求項1に記載のマイクロ流体デバイス。
- サンプル中のターゲットを検出するための方法であって、
近位開口部及び流量を制御するべく先細りしている遠位開口部を有するキャピラリフローチャネルを備えたマイクロ流体デバイスに関する読取り装置と共に動作可能な関係性に当該マイクロ流体デバイスを位置決めすることと、
前記キャピラリフローチャネルの近位部にあるフィルタポケットであって、液体サンプルを生成するべく前記サンプルの液体部分を分離するフィルタポケットにサンプルを導入することと、
前記液体サンプルを、デバイス間の濾過成分の変動を最小にするべく混合ウェルに通すことと、
前記液体サンプル中で乾燥試薬を再構成するように構成された乾燥試薬ゾーンに、前記混合ウェルからの前記液体サンプルを通すことと、
当該マイクロ流体デバイスの長さに実質的に垂直な方向にローブしている、前記キャピラリフローチャネルの部分を有するピンチ領域であって、減少した拡散距離でインキュベートするための試薬の付加的な時間をもたらすことにより前記アッセイの感度を高めるように構成されたピンチ領域に、前記乾燥試薬ゾーンからの前記液体サンプルを通すことと、
前記ピンチ領域からの液体サンプルを、ターゲットの存在を検出するように構成された検出ゾーンに通すことと、
前記検出ゾーンにおいて固相取り込みスポットと接触させることと、
前記固相取り込みスポットからの信号を読み取ることと、
前記検出ゾーンからの液体サンプルを、前記検出ゾーンの洗浄からの過剰液体サンプルを保持するように構成された廃液チャネルに通すことと、
を含む、方法。 - 患者サンプル中の心筋トロポニンの存在又は欠如を判定するための、請求項12に記載の方法であって、
a)存在する場合、トロポニンの結合パートナー及び検出可能な分子を含む標識でトロポニンを標識することと、
b)ハンドヘルドアッセイにおける標識の存在又は欠如を判定することによりサンプル中のトロポニンを検出することと、
を含み、
前記標識の存在の検出は、前記サンプル中のトロポニンの存在を示し、前記アッセイは、約20%未満の変動係数と共に約3pg/mLの定量限界を有する、方法。 - 前記心筋トロポニンが心筋トロポニンI(cTnI)である、請求項13に記載の方法。
- 前記心筋トロポニンが心筋トロポニンT(cTnT)である、請求項13に記載の方法。
- 前記心筋トロポニンがcTnIとcTnTの複合体である、請求項13に記載の方法。
