本発明の製剤は、polyglutamine−tract binding protein1(PQBP−1)をコードする所定のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する。以下、本発明の製剤について説明する。
本発明の神経幹細胞の増殖の促進に用いられる製剤は、PQBP−1をコードする以下の(a)から(d)のいずれかに記載のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する。

本発明のDNAは、上記(a)〜(f)のいずれかのDNAである。
本発明の予防又は治療剤で使用されるポリペプチドは、前述のDNAによりコードされ、例えば、配列番号2記載のアミノ酸配列を有し、又は配列番号2記載のアミノ酸配列において1もしくは複数のアミノ酸が置換、欠失及び/又は付加されたアミノ酸配列を有する。
発現ベクターは、適当なベクターに上述のDNAを挿入することにより、作製できる。「適当なベクター」とは、原核生物及び/又は真核生物の各種の宿主内で複製保持又は自己増殖できるものであればよく、使用の目的に応じて適宜選択できるものである。例えば、DNAを大量に取得したい場合には高コピーベクターを選択でき、ポリペプチドを取得したい場合には発現ベクターを選択できる。その具体例としては、特に限定されず、例えば、特開平11−29599号公報に記載された公知のベクターが挙げられる。
形質転換体は、前述のDNAを含むベクターを宿主に導入することにより、作製できる。このような宿主は、本発明のベクターに適合し形質転換され得るものであればよく、その具体例としては、特に限定されないが、細菌、酵母、動物細胞、昆虫細胞等の、公知の天然細胞もしくは人工的に樹立された細胞(特開平11−29599号公報参照)、あるいは、ヒト、マウス等の動物が挙げられる。
本発明に係るスクリーニング方法は、神経幹細胞の減少に関連する疾患の予防もしくは治療に用いられる製剤の候補物質のスクリーニング方法、又は、シナプス後部形成の減少に関連する疾患の予防もしくは治療に用いられる製剤の候補物質のスクリーニング方法である。
本発明の神経幹細胞の減少に関連する疾患の予防又は治療に用いられる製剤の候補物質のスクリーニング方法は、被験物質を、動物細胞又はヒトを除く動物に投与する工程と、被験物質が投与された動物細胞又はヒトを除く動物の、PQBP−1による神経幹細胞増殖促進の程度を測定する工程と、測定結果に基づいて、神経幹細胞の減少に関連する疾患の予防又は治療に用いられる製剤の候補物質として被験物質を選択する工程と、を有する。
本発明のシナプス後部形成の減少に関連する疾患の予防又は治療に用いられる製剤の候補物質のスクリーニング方法は、被験物質を、動物細胞又はヒトを除く動物に投与する工程と、被験物質が投与された動物細胞又はヒトを除く動物の、PQBP−1によるシナプス後部形成の促進の程度を測定する工程と、測定結果に基づいて、シナプス後部形成の減少に関連する疾患の予防又は治療に用いられる製剤の候補物質として被験物質を選択する工程と、を有する。
投与工程は、被験物質を、動物細胞又はヒトを除く動物に投与する工程である。
測定工程は、被験物質が投与された動物細胞又はヒトを除く動物の、内在性PQBP−1の発現量又はPQBP−1による神経幹細胞増殖促進の程度、もしくは、PQBP−1によるシナプス後部形成の促進の程度を測定する工程である。
選択工程は、上記測定結果に基づいて、神経幹細胞の減少に関連する疾患の予防もしくは治療に用いられる製剤、又は、シナプス後部形成の減少に関連する疾患の予防もしくは治療に用いられる製剤の候補物質として被験物質を選択する工程である。
個体の発生に必須な遺伝子は単純な方法ではノックアウト個体を得ることができないが、Cre−loxP系等を用いることで特定の条件下でのみ遺伝子ノックアウトすることが可能である。本実施例では、Nestinが神経幹細胞で選択的に発現すること、及び、Synapsin1が成熟神経細胞で選択的に発現することを利用して、神経幹細胞を標的とするノックアウトマウス(Nestin−Cre−cKOマウス)、及び、成熟神経細胞を標的とするノックアウトマウス(Synapsin1−Cre−cKOマウス)を作成した。以下、Nestin−Cre−cKOマウスは、神経幹細胞でPQBP−1遺伝子がノックアウトされたマウス、Synapsin1−Cre−cKOマウスは、成熟神経細胞でPQBP−1遺伝子がノックアウトされたマウスをいう。
PQBP−1遺伝子は、X染色体上の遺伝子であるから、これらcKOマウスは、X染色体が1本の雄ではPQBP−1遺伝子はノックアウトされているが、X染色体が2本の雌ではPQBP−1遺伝子は正常とノックアウトのヘテロ接合となる。
PQBP−1遺伝子が変異した患者の脳の構造を磁気共鳴画像により解析した。その結果を図1に示す。図1中、(a)は、水平面、(b)は冠状面、(c)は矢状面を示す。その結果、PQBP−1が変異した患者において、通常の皮質構造であることが確認された。
Nestin−Cre−cKOマウスの神経幹細胞(NSPC)の細胞周期の分析を行った。まず、BrdU(シグマ社製、100mg/kg体重)を、E14(胎生14日)の各妊娠マウスの腹腔内に注入した(妊娠マウスの種類は後述の表1に示す)。妊娠マウスにBruUを繰り返し注入し(3時間間隔)、累積してラベリングを行った。各マウスは、最初のBrdUの注入後、1、1.5、2、3.5、6.5、15.5又は24.5時間にそれぞれの胚脳を取り出した。取り出した胚脳を4%パラホルムアルデヒドで固定し、パラフィンに組み込んだ。胚脳の断片は、3mmの間隔で作製し、パラフィンを除き、再水和し、それから、15分間、pHが6.0の10mMクエン酸緩衝液中にいれた状態で電子レンジで加熱した。その後、抗体のインキュベーションを、マウス抗BrdU抗体(1:200、BD Biosciences社製)、ウサギ抗ホスホ−ヒストンH3(pH3)抗体、M期の細胞のマーカー(1:500、ミリポア社製)を用い、4℃、オーバーナイトで行った。二次抗体のインキュベーションは、Alexa Fluor488又はCy3複合体(1:500、Invitrogen社製)を用いて行った。脳室帯における、BrdU/pH3ダブル陽性細胞のpH3陽性細胞に対する割合を、G2/M期の長さを決定するためにBrdUの単一の注射した後、1、1.5、2時間で計算した。1、1.5、2、3.5及び6.5時間における標識化指数値(LIs)の直線グラフによって、Y軸切片(0時間でのLI)を推定し、傾きを計算した。野生型マウスの脳室帯での成長率(増殖する細胞の割合)が、1.0に近いため、0時間におけるLIと傾きは、全細胞周期(Ts/Tc)に対するS期の割合と、全細胞周期(1/Tc)の逆数をそれぞれ表す。TsとTcは、S期、全細胞周期の長さを表す。これらの値(Ts/Tcと1/Tc)から、TsとTcを算出した。それぞれの細胞周期の長さを、表1に示す。
幹細胞プールからの神経新生をBrdU及びKi67の共染色によって分析した。BrdUを妊娠マウスに腹腔内注射して24又は72時間後に、神経新生の細胞数(BrdU+/Ki67−)、幹細胞プール中に残っている細胞数(BrdU+/Ki67+)、ラベルされてない神経幹細胞(BrdU−/Ki67+)の数を計算するためにE15のそれぞれの胚脳を分析した。その結果、24時間後、72時間後において、各グループの細胞の割合に差は検出されなかった。図7は、注入して24時間後の、神経新生の細胞数、幹細胞プール中に残っている細胞数、ラベルされてない神経幹細胞(BrdU−/Ki67+)の数の相対的な割合を示すグラフである。
Nestin−Cre−cKOマウスと、Nestin−Creマウスにおける、大脳皮質の細胞死のレベルを、E10、E15、E18、P0、及びP60において、TUNEL染色によって評価した。その結果を図8に示す。図8より、Nestin−Cre−cKOマウスと、Nestin−Creマウスとの間で、細胞死した細胞の数に差がないことが確認された。
PQBP−1によるスプライシングと関連性の強い遺伝子の分析を行うために、野生型のマウスとPQBP−1遺伝子をノックアウトしたマウスとのそれぞれのエキソンのシグナルを比較して、エキソンのレベルが著しく変化した遺伝子を見つける「エキソンエキソン分析」を行った。その差は、2つの試料の手段が同じであるという帰無仮説に基づいて、ストゥーデントのt検定により調べた。また、エキソン間の相対的な発現レベルの変化を検出する「バリアンス分析」を行った。これは、野生型及びPQBP−1をノックアウトしたマウスの各遺伝子におけるエキソンレベルのプローブセットを用いて実施した。その差は、これら2つのサンプルが同じパターン及び分散を有するという帰無仮説に基づくF−testによって調べた。なお、PQBP−1遺伝子をノックアウトしたマウスのエキソンは、E15のNestin−Cre−cKOマウスの神経幹細胞(Neural Stem Cell)、生後4週齢のNestin−Cre−cKOマウスの皮質、又は、生後4週齢のSynapsin1−Cre−cKOマウスの皮質を用いて、それぞれを野生型マウスと比較した。その結果を表2〜4に示す。なお、遺伝子が複数のエキソンプローブを保有する場合、最も低いp−値を用いた。表2は、E15のNestin−Cre−cKOマウスと野生型マウスの神経幹細胞を比較した結果を示す。表3は、生後4週齢のNestin−Cre−cKOマウスと野生型マウスの皮質を比較した結果を示す。表4は、生後4週齢のSynapsin1−Cre−cKOマウスと野生型マウスの皮質とを比較した結果を示す。
APC4遺伝子を、PQBP−1がノックアウトされたマウスに導入する試験を行った。まず、マウスのAPC4のcDNA(Genbank accession number NM_024213)を、RT−PCRにより準備し、これを、XhoI/BamHIで制限酵素処理したpIRES2−hrEGFPII(Stratagene社製)に組み込み、pApc4−IRES2−hrEGFPIIを作製した。この遺伝子を用い、野生型マウスと、Nestin−Cre−cKOマウスの胚の脳室にエレクトロポレーションにより導入した。
まず、PQBP−1遺伝子がノックアウトされたマウス(Nestin−Cre−cKOマウス)にヒトPQBP−1遺伝子を導入した。具体的には、ベクターとしてAAVベクタープラスミドを用いた。AAVベクタープラスミドは、ヒトサイトメガロウイルス前初期プロモーター(CMVプロモーター)からなる発現カセットを含有し、その後ろ側に、ヒトPQBP1又はヒトPQBP1−EGFPをコードするcDNA、AAV3ゲノムの逆方向末端反復の間のシミアンウイルス40ポリアデニル化シグナル配列(SV40ポリ(A))が続くように設計した。組換えAAVベクター(AAV−PQBP−1ベクター)は、ベクタープラスミド、AAV2 rep及びAAV1 vp発現プラスミド、及びアデノウイルスヘルパープラスミド、pHelper(Agilent Technologies社製)を用いて、HEK293細胞への一過的な導入により作製した。組換えウイルスは、2つの経時的な塩化セシウム勾配の分離形成によって精製し、ウイルス価を定量RT−PCRによって決定した。AAVベクター(AAV−PQBP−1ベクター)のインビボでの投与は、C57BL/6J妊娠マウス(E10のNestin−Cre−cKO胎児マウスを妊娠)に腹腔内投与することにより、AAV−PQBP−1ベクター(2.0×1011ゲノムコピー)をNestin−Cre−cKO胎児マウスに導入した。このAAV−PQBP−1ベクターが導入されたNestin−Cre−cKOマウス(2ヶ月(10週齢))を用いて、以下の試験を行った。
3か月の成体マウス(Nestin−Cre−cKOマウス、AAV(AAV−PQBP−1ベクター)を導入したNestin−Cre−cKOマウス)について、回転ロッドの試験を行った。まず、マウスを回転ロッド(直径3cm)に置き、回転速度を3.5回転から300秒で35rpmに直線的に増加させ、600秒経過するまで35rpmの回転数で続けた。各試験の間に10分の休憩間隔をおき、9回の試験(3日間連続して行い、1日3試験)を行った。ロッドから落下する時間を記録し、ロッドから落ちる平均時間を記録した。その結果を図19に示す。
野生型マウス、Floxマウス、Synapsin−1−Cre−cKOマウスについて、上記Nestin−Cre−cKOマウスでの行動解析と同様の方法で回転ロッドによる回転速度の試験を行ったところ、Synapsin−1−Cre−cKOマウスが、他のコントロールのマウスより、Nestin−Cre−cKOマウス同様、有意に回転ロッドに滞在した時間が短かったことが確認された(図21)。
Synapsin1−Creマウス(n=3、4週齢)、Floxマウス(n=3、4週齢)、Synapsin1−Cre−cKOマウス(n=3、4週齢)の成熟神経細胞のシナプス後部形成の程度を測定した。測定は、それぞれのマウスの成熟神経細胞を、2光子顕微鏡を用いて観察することによって行った。より具体的には、あらかじめAAV−GFP(5x1011viral genome/ml)をretrosplenial dysgranular (RSD) cortex(bregmaより前後方向−2.0mm、内外方向0.6mm)内に注入しておいたそれぞれのマウスに、イソフルランによる吸入麻酔し(0.1ml/min)、観察部皮膚を切開し、ドリルを用いて頭蓋骨を薄く削り観察用のウインドウを作成した(thinned−skull法)。マルチフォトンレーザーMaiTai HP DeepSee−OL(Spectra Physics社製)、水浸型対物レンズXLPlanN25xW(オリンパス社製)を搭載した2光子レーザー顕微鏡システムFV1000MPE(オリンパス社製)を用いてスパインと樹状突起のイメージングデータを取得した。GFPはレーザー波長890nmで励起し、thinned−skull法によるウインドウよりRSD cortexを観察し、1μm間隔で1024×1024ピクセルを持つ解像度のイメージングデータを取得した。その観察結果を図23に示す。これにより、成熟神経細胞でPQBP−1遺伝子がノックアウトされたSynapsin1−Cre−cKOマウスにおいて、シナプス後部の形成が減少していることが確認された。
4週齢のSynapsin1−Cre−cKO(n=3)及び野生型のマウス(n=3)の脳をサンプルとし、GeneChip Mouse Exon 1.0 ST Array(Affymetrix社)を用いてエキソンアレイデータを取得し、以下の手順によりエキソンアレイデータ分析を行った。
高脂肪食を与えたマウスの脳におけるPQBP−1の発現を、ウエスタンブロッティングにより分析した。マウス(C57BL/6)は、6週齢から6週間の高脂肪食群(n=3、図27−30の「HFD6W」に相当)と、11週齢から1週間の高脂肪食群(n=3、図27−30の「HFD1W」に相当)を用いた。また、比較として高脂肪食を与えない通常食群(n=3、図27−30の「HFD0W」に相当)のマウスを用いた。分析は、それぞれのマウスの12週齢目に分析を行った。なお、高脂肪食としては、日本クレア社のHigh Fat Diet 32を用い、これをマウスに自由摂取させた。高脂肪食を摂取する以外の期間の高脂肪食群と、通常食群には、通常食(日本クレア社のCE−2)を自由摂取させた。水分も自由摂取とし、飼育室の明暗コントロールは12時間ごとに行い、室温25±2℃、湿度50±10%で飼育した。分析結果の写真を図27に示し、タンパク量の割合を示すグラフを図28に示す。従来より、肥満はアルツハイマー病の危険因子であると考えられていたが、この結果より、肥満によるPQBP−1の減少が、アルツハイマー病の原因となることが示唆された。
上記の高脂肪食を与えた高脂肪食群のマウスと、通常食を与えた通常食群のマウスにおける、シナプス後部の数を測定した。測定は、まず、10週齢でマウスにAAV−GFP(5x1011viral genome/ml)がretrosplenial dysgranular (RSD) cortex(bregmaより前後方向−2.0mm、内外方向0.6mm)内に注入し、12週齢でイソフルランによる吸入麻酔し(0.1ml/min)、観察部皮膚を切開し、ドリルを用いて頭蓋骨を薄く削り観察用のウインドウを作成した(thinned−skull法)。マルチフォトンレーザーMaiTai HP DeepSee−OL(Spectra Physics社製)、水浸型対物レンズXLPlanN25xW(オリンパス社製)を搭載した2光子レーザー顕微鏡システムFV1000MPE(オリンパス社製)を用いてスパインと樹状突起のイメージングデータを取得した。GFPはレーザー波長890nmで励起し、thinned−skull法によるウインドウよりRSD cortexを観察し、1μm間隔で1024x1024ピクセルを持つ解像度のイメージングデータを取得した。得られたイメージングデータをIMARISに読み込ませ、フィラメントツール(樹状突起直径:0.297−4.950μm、スパインヘッド直径:0.2−3.0μm)を用いて樹状突起とスパインを認識させ3次元モデルを構築し解析することによって、測定を行った。その結果を図29に示す。これにより、シナプス後部の数が、高脂肪食を与えることにより減少することが確認された。この結果は、肥満によるPQBP−1の減少は、シナプス後部の形成の減少を引き起こし、それがアルツハイマー病の原因となることが示唆する。
5.5か月齢の5xFADマウス(アルツハイマー病モデルマウス)又は野生型マウスに、AAV−CMV−EGFP−PQBP−1ウイルスベクター(1x109vg/ml)又はAAV−CMV−EGFPウイルスベクター(1x109vg/ml)100μlを、osmotic pumpに充填後、マウス背部に皮下移植し、連結したガラスマイクロピペットでクモ膜下腔に72時間持続投与した。ウイルス投与開始日より14日後、thin−skull法(脳表の頭蓋骨を厚さ20−50μm程度まで薄く剥離する方法)にて、retrosplenial dysgranular(RSD) cortexの第1−2層の神経細胞のスパインを二光子顕微鏡により観察した。撮影した画像から画像解析ソフトimaris7.6.1を用いて、単位樹状長当たりのスパイン数の定量評価を行った。マウスの大脳皮質神経細胞のスパインの画像を図31に示す。図32に、マウスの大脳皮質神経細胞の樹上突起の数(number/10μm)示す。
Claims (6)
- polyglutamine−tract binding protein1(PQBP−1)をコードする以下の(a)から(d)のいずれかに記載のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する、神経幹細胞の増殖の促進に用いられる製剤。
(a)配列番号1記載の塩基配列を有するDNA
(c)配列番号2記載のアミノ酸配列において1もしくは数個のアミノ酸が置換、欠失及び/又は付加されたアミノ酸配列をコードする塩基配列を有し、かつ、神経幹細胞の増殖を促進することができるDNA
(d)配列番号1記載の塩基配列と90%以上の相同性を有する塩基配列からなり、かつ神経幹細胞の増殖を促進することができるDNA - 前記神経幹細胞の増殖の促進は、細胞周期の延長の抑制によるものである、請求項1記載の製剤。
- PQBP−1をコードする以下の(a)から(d)のいずれかに記載のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する、神経幹細胞の減少に関連する疾患の予防又は治療に用いられる製剤。
(a)配列番号1記載の塩基配列を有するDNA
(c)配列番号2記載のアミノ酸配列において1もしくは数個のアミノ酸が置換、欠失及び/又は付加されたアミノ酸配列をコードし、かつ、神経幹細胞の増殖を促進する塩基配列を有するDNA
(d)配列番号1記載の塩基配列と90%以上の相同性を有し、かつ、神経幹細胞の増殖を促進する塩基配列からなるDNA - PQBP−1をコードする以下の(a)、(e)又は(f)に記載のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する、シナプス後部形成の促進に用いられる製剤。
(a)配列番号1記載の塩基配列を有するDNA
(e)配列番号2記載のアミノ酸配列において1もしくは数個のアミノ酸が置換、欠失及び/又は付加されたアミノ酸配列をコードし、かつ、シナプス後部形成を促進する塩基配列を有するDNA
(f)配列番号1記載の塩基配列と90%以上の相同性を有し、かつ、シナプス後部形成を促進する塩基配列からなるDNA - PQBP−1をコードする以下の(a)、(e)又は(f)に記載のDNA、又はこのDNAがコードするポリペプチドを有効成分として含有する、シナプス後部形成の減少に関連する疾患の予防又は治療に用いられる製剤。
(a)配列番号1記載の塩基配列を有するDNA
(e)配列番号2記載のアミノ酸配列において1もしくは数個のアミノ酸が置換、欠失及び/又は付加されたアミノ酸配列をコードし、かつ、シナプス後部形成を促進する塩基配列を有するDNA
(f)配列番号1記載の塩基配列と90%以上の相同性を有し、かつ、シナプス後部形成を促進する塩基配列からなるDNA - 前記DNAがベクターに組み込まれている請求項1から5いずれか記載の製剤。
