Description

プールしたヒト血漿寒冷沈降物を開始材料(すなわち、フィブリノーゲン含有原材料)として使用した。簡単に説明すると、プールした血漿寒冷沈降物は、20mMクエン酸3
ナトリウム、200mMイプシロン−アミノカプロン酸(ε−ACA)、60IU/mLヘパリンおよび500mM NaCl(pH7.2±2)を含有する抽出緩衝液中において31±2℃で30分間可溶化した(緩衝液4g当たり寒冷沈降物1g)。次に、水酸化アルミニウム2%(w/w)を可溶化した寒冷沈降物に25%(w/w)の濃度で添加した。その後、水酸化アルミニウムゲルを遠心または深層濾過のいずれかによって除去し、フィブリノーゲン含有上清をHCICクロマトグラフィー樹脂によるクロマトグラフィーでさらに精製するために回収した。
HEA Hypercelによって低下したフィブリノーゲン溶液における不純物レベル
実施例1によって調製したフィブリノーゲンを含有する可溶化寒冷沈降物約48.5mLを、pH6.5、7.0、7.5、8.0または8.5のいずれかの25mMトリスで予め平衡化した5mL HEA Hypercel(商標)カラムに添加した。HCIC精製は、フィブリノーゲンに関してネガティブモードで実施され、フィブリノーゲンを未結合フロースルー画分に素通りさせ、一方、t−PA、プラスミノーゲンおよび第II因子は樹脂に結合して維持した。図4aは、HEA Hypercel(商標)を使用したポストクロマトグラフィー精製後のフィブリノーゲン、プラスミノーゲン、t−PAおよび第II因子の工程回収率を示す。結果は、pHはHCIC樹脂へのプラスミノーゲンおよび第II因子の結合にはほとんどまたは全く影響を及ぼさず、一方、樹脂へのt−PAの結合は、6.5〜7.0の低いpH範囲で最も効果的に見えることを示す。フィブリノーゲンの回収率は、カラム洗浄を実施しなかったにもかかわらず、様々なpH条件下で、素通り画分において90%を上回った。図4aに示したように、これらの結果は、HCIC樹脂によって可溶化寒冷沈降物などの粗フィブリノーゲン含有原材料中におけるプロテアーゼが効果的に除去されることを示している。例えば、pH7.0で実施した実験条件は、可溶化寒冷沈降物溶液からの第II因子は≧99.9%、t−PAは≧88.3%およびプラスミノーゲンは≧%98.2%低下することを示した。
HEA Hypercelによって精製されたフィブリノーゲン溶液中における不純物のレベル
実施例1によって生成したアルハイドロゲル(商標)吸着工程後に得られたフィブリノーゲン含有上清約500mLを、25mMトリスpH7.0で予め平衡化したHEA Hypercel(商標)の樹脂36mLを充填したXK16/30カラムに添加した。素通り画分は、フィブリノーゲン、プラスミノーゲン、t−PAおよび第II因子を試験するために収集した。結果の概要を以下の表1に示す。
HEA Hypercelによって精製したフィブリノーゲン溶液中における不純物のレベルに対するグリシン沈殿の効果
実施例1によって生成したアルハイドロゲル吸着工程後のフィブリノーゲン含有上清に、グリシン2.4M、2.7M NaCl、2.1mM CaCl2および23mMクエン酸3ナトリウムを含む生理食塩水(pH6.6〜7.3)を添加することによって、さらに沈殿工程を行った。可溶化した沈殿物を30℃まで温めてから、同様に30℃でインキュベートしたグリシン緩衝液を、生成物の緩衝液に対する比1:2で添加した。混合物を10分間撹拌し、得られた沈殿物を遠心によって液相から回収した。主にフィブロネクチンおよびIgGを含有する液相を廃棄して、フィブリノーゲン含有沈殿物を収集し、100mM NaCl、1.1mM CaCl2、10mMクエン酸3ナトリウム、10mMトリス−(ヒドロキシメチルメチルアミン)および4.5mMスクロースを含有する可溶化緩衝液(pH7.0)に再懸濁した。可溶化したフィブリノーゲン中間体(250mL)は、1μmフィルターを使用して清澄にしてから、HEA Hypercel(商標)樹脂36mLを充填し、25mMトリスpH7.0で予め平衡化したXK16/30カラムに通過させた。素通り画分は、フィブリノーゲン、プラスミノーゲン、t−PAおよび第II因子を試験するために収集し、結果の概要を以下の表2に示した。
血漿寒冷沈降物から精製したフィブリノーゲンの調製
処理工程1−血漿寒冷沈降物の可溶化;
処理工程2−可溶化した血漿寒冷沈降物のアルハイドロゲル(商標)(水酸化アルミニウム)吸着(アルハイドロゲル(商標)濃縮物:標的15から20%w/w、10から50%w/wの範囲)および遠心または濾過助剤の存在下での深層濾過などの方法を使用したフィブリノーゲン含有上清の回収。あるいは、この工程は、フィブリノーゲンに関してネガティブモード(素通り)のHCICクロマトグラフィー工程またはフィブリノーゲンに関していずれもネガティブモードのHCICおよび陰イオン交換クロマトグラフィーの組み合わせによって置き換えることができる。HCICおよび陰イオン交換クロマトグラフィーの両方を組み合わせて使用する場合、処理工程3を選択する;
処理工程3−工程2のフィブリノーゲン含有上清からのフィブリノーゲンのグリシン沈殿。あるいは、この工程は、フィブリノーゲンに関してネガティブモードの陰イオン交換クロマトグラフィーによって置き換えることができる;
処理工程4−可溶化した工程3のグリシン沈殿物をフィブリノーゲンに関してネガティブモードのHCICクロマトグラフィー樹脂に通過させる;
処理工程5−病原体を不活性化するために、工程4で回収した精製フィブリノーゲン溶液を溶媒もしくは界面活性剤で処理するか、または低温殺菌する;
処理工程6−工程5の処理溶液をフィブリノーゲンに関してポジティブモードの陰イオン交換クロマトグラフィー樹脂に通過させ、弱く結合したタンパク質を樹脂から洗浄し、フィブリノーゲンを樹脂から溶出させる;
処理工程7−工程6の陰イオン交換樹脂から溶出したフィブリノーゲンにナノ濾過(35nmまたは20nmまたは35/20nmの組み合わせ)を行い;
処理工程8−工程7で濾過したフィブリノーゲンを限外濾過(50、100、200および300kDa膜フィルター)する。
HCICおよび陰イオン交換クロマトグラフィーの組み合わせによる精製フィブリノーゲンの調製
HEA Hypercel(商標)カラムを使用した混合型クロマトグラフィー工程で、実施例1から3で記載した工程によって、寒冷沈降物約100gを調製した3回の実験室規模の実験を完遂した。
精製したフィブリノーゲンのウイルス濾過
MacroPrep(商標)−HQ溶出工程に190mM NaCl緩衝液(21.5mS/cm)、200mM NaCl緩衝液(22.5mS/cm)、210mM NaCl緩衝液(23.5mS/cm)または1%(w/w)アルギニンを含有する200mM NaCl緩衝液(25mS/cm)のいずれかを使用した実施例6の方法に従って、20nmウイルスフィルターによる濾過性を得られたフィブリノーゲン調製物で調べた。
製物は、類似の濾過特性を生じることを示唆している。対照的に、1%(w/w)アルギニンを含有する200mM NaCl緩衝液を使用してMacroPrep(商標)HQカラムから溶出したフィブリノーゲン調製物は、フィルターに迅速に付着した(図7)。
安定性試験
前記の実施例5で記載した方法によって回収した精製フィブリノーゲン溶液を滅菌濾過し、9/7週にわたって2℃〜8℃または30℃で安定性試験を行った。2℃〜8℃に置いた液体フィブリノーゲン調製物は、貯蔵期間9週間後にClauss法によって測定したところ、元の活性の約90%を保持していた。30℃に置いた液体フィブリノーゲン調製物は、貯蔵期間2週間後に元の活性の約70%を保持し、少なくとも5週間はさらなる活性の喪失はなかった。60%を下回る活性のさらなる低下は、30℃で7週間インキュベートすることによって認められた。30℃でのフィブリノーゲン活性の喪失は、プロテアーゼ阻害剤(C1エステラーゼ)を添加しても貯蔵期間5週間にわたるフィブリノーゲン活性の損失を阻害しなかったので、タンパク質分解よりも熱変性によるものと考えられる。安定性データの概要を図8に示す。
HEA Hypercelを使用した第VIII因子および/またはVWFを含有する溶液中における血漿プロテアーゼレベルの低下
この実施例は、HCICクロマトグラフィー工程も、第VIII因子および/またはVWF含有調製物中におけるプロテアーゼを低下させるために使用できることを示す。この方法には、実施例4から得られたフィブリノーゲン含有溶液を、深層フィルターを使用して清澄化してから、清澄化した溶液を第2の疎水性電荷誘導クロマトグラフィー(HCIC)樹脂に通過させることが必要である。HCIC工程は、プラスミノーゲンなどのプロテアーゼが樹脂に結合し、一方、第VIII因子およびVWFが樹脂を通過することができる条件下で操作した。特に、XK50/30カラムにHEA Hypercel(商標)樹脂340mLを充填した。カラムは、50mMトリスpH6.7で予め平衡化した。次に、実施例4によって調製した清澄なフィブリノーゲン溶液をこのカラムに添加し、カラムを50mMトリスpH6.7で洗浄した。素通り画分を収集し、第VIII因子、VWF、プラスミノーゲンおよびt−PAのレベルを測定した(VWF:RCo=フォンビルブランドリストセチンコファクター)。4回の個々の実験から得られた平均結果の概要を、表5に示す。結果は、HCICクロマトグラフィー工程は第VIII因子およびVWFを含む画分からプラスミノーゲンおよびtPAなどのプロテアーゼを効果的に除去したことを示す。さらに、第VIII因子およびVWFの良好な回収率が認められた。
様々な方法から精製したフィブリノーゲンの比較研究
実施例6で記載したように、本発明の方法によって製造したフィブリノーゲン調製物を国際公開第2001048016号、国際公開第2012038410号および国際公開第2013135684号で記載された方法によって製造したフィブリノーゲン調製物と比較した。
NaCl、20mM EACA、pH8.0、10mL/分(113cm/hr)で予め平衡化した。カラム後の伝導度が調製した緩衝液の90〜110%になるまで、平衡化を継続した。次に、フィブリノーゲン溶液をこのカラムに添加し、カラムを6CVのMQ緩衝液で洗浄した。フィブリノーゲンは、ME緩衝液(500mM NaCl、1.1m
M CaCl2、10mMクエン酸Na、10mMトリスおよび45mMスクロース、pH7.0)を使用して単一ピークとして溶出した。カラムは、2CVの1M NaClを使用して再生することができた(国際公開第2001048016号、実施例2)。
Claims (1)
- フィブリノーゲンを精製する方法であって:
(i)溶液中に存在するフィブリノーゲンモノマーがイオン交換クロマトグラフィー樹脂に結合するように選択された条件下で、フィブリノーゲンを含む溶液を該樹脂に通過させる工程;
(ii)該樹脂からフィブリノーゲンモノマーを溶出緩衝液で溶出させる工程であって、該溶出緩衝液は19mS/cm〜24mS/cmの範囲の伝導率を有する、前記工程;および
(iii)工程(ii)からの溶出したフィブリノーゲンモノマーを、孔径が15nmから35nmの範囲の親水化したフッ化ポリビニリデン(PVDF)フィルターで、2%から6%(w/w)のアルギニンの存在下で濾過する工程
を含む、前記方法。
