タンパク質の大規模かつ経済的な精製は、バイオ医薬品業界のますます重要な問題である。治療用タンパク質は通常、対象のタンパク質をコードする遺伝子を含む組み替えプラスミドから対象のタンパク質を発現するように設計された、原核生物または真核生物の細胞株を用いて生成する。細胞に与えられる成分と細胞の副産物との混合物からの所望のタンパク質の十分な(例えばヒトの治療に十分使用できる)純度での分離は、生物製剤の製造者に対して恐るべき課題として提起されている。
本発明は、アフィニティークロマトグラフィーを用いるタンパク質の精製中に不純物を除去する、効果の高い、独特な手段を提供する。具体的には、本手法は非常に高いpH(例えば、9〜11)のアルカリ洗浄溶液を利用する。アルカリ洗浄溶液の別の特徴は、それらはアルギニンまたはアルギニン誘導体の存在を必要としないことである。アルカリ洗浄溶液は、アフィニティークロマトグラフィーマトリックスに適用されるフィード物質からの、宿主細胞のタンパク質(HCP)の除去に極めて効果的である。下記の研究例に示すように、この手法は頑強で、異なるmAbサブクラスに効果的に利用できる。アフィニティークロマトグラフィーにおけるこのような洗浄溶液の使用は、高い処理収率および生成物の完全性を維持する。加えてこの手法は、処理の効率を上げ、展開時間を短くする。
本開示はより簡単に理解されるように、特定の用語を最初に定義する。本出願において、本明細書で他に明確に与えられる場合を除き、下記の各用語は下記で説明する意味を持つものとする。さらなる定義は、本出願のあらゆる場所で、説明される。
ある態様において、本発明の方法は、商業、実験、または他の応用において有益である、製薬、診断、農業、および/またはあらゆる多様な他の性質を持つタンパク質を含むが、これらに限定されない、いかなる対象のタンパク質の精製に使用してもよい。加えて、対象のタンパク質はタンパク質治療薬であってもよい。ある実施態様において、本発明の方法で精製されたタンパク質を、プロセシングまたは修飾しても良い。例えば、本発明に従って得られる対象のタンパク質をグリコシル化しても良い。
本発明の方法は、対象のタンパク質を含むあらゆる混合物に適用できる。一つの実施態様において、混合物は、精製しようとするタンパク質(例えば、天然または遺伝子的に設計される)を発現する生細胞から得られるか、または、それによって生成される。場合により、培養細胞中の細胞は、対象のタンパク質をコードする核酸を含む発現コンストラクトを遺伝子導入した細胞を含む。タンパク質を生成するために、細胞を遺伝子操作する方法は当分野でよく知られている。例えば、Ausubel et al., eds., Current Protocols in Molecular Biology, Wiley, New York (1990)ならびにU.S. Patent Nos. 5,534,615および4,816,567参照、これらはそれぞれ、引用により本明細書に明確に組み入れられる。このような方法は、タンパク質をコードし、発現を可能にする核酸を、生きた宿主細胞に導入することを含む。これらの宿主細胞は、培養液中で成長する、細菌性細胞、真菌細胞、昆虫細胞または、好ましくは動物細胞であり得る。細菌性宿主細胞は、限定されないが、大腸菌細胞を含む。適切な大腸菌株の例は:HB101、DH5α、GM2929、JM109、KW251、NM538、NM539、および外来のDNAを切断できないいかなる大腸菌株をも含む。用いられる真菌宿主細胞は、限定ではないが、サッカロマイセス・セレビシエ(Saccharomyces cerevisiae)、ピキア・パストリス(Pichia pastoris)およびアスペルギルス(Aspergillus)細胞を含んでもよい。昆虫細胞は、限定ではないが、カイコ(Bombyx mori)、ヨトウガ(Mamestra drassicae)、ツマジロクサヨトウ(Spodoptera frugiperda)、イラクサギンウワバ(Trichoplusia ni)、キイロショウジョウバエ(Drosophilia melanogaster)を含んでもよい。
本発明の方法は第1のクロマトグラフィーマトリックス(例えば、アフィニティークロマトグラフィーマトリックス)をpHが少なくとも9.0であるアルカリ洗浄溶液と接触させることを含む。このようなアルカリ洗浄溶液はアルギニンまたはアルギニン誘導体の存在を必要としない。場合により、このようなアルカリ洗浄溶液は、緩衝作用のない塩を含んでも含まなくてもよい。
臨床の要求のため、多数のモノクローナル抗体が現在、治療に用いるための開発下にある。市場の競合において、バイオ医薬品会社は継続して、前臨床および臨床上の製造における、開発のタイムライン(timeline)の加速およびより効率的な方法の作成に目を向けている。ひとつの手法は、必要な生成物の仕様を満たすように精製のスキームを改良し、合理化することである。この理由のため、各ユニット操作における不純物の排除能の最大化が望まれている[1,4]。しかしながら、精製のフィードストリームは、宿主細胞タンパク質、DNA、外来性および内在性ウイルス、内毒素、および凝集体を含む、除去が重要であり、かつ、困難であり得る様々な不純物を含む。産業上の精製の開発は、これらの生成物の品質特性に追随しており、生成物の承認基準のための一般的なガイドラインを定めてきた(表1)[2,3,4]。不必要な精製の工程はよくタンパク質の回収率を下げ、操作時間および費用を増やし得るため、理想的には、多段階精製スキームにおける各ユニットの操作は全体の生成物の質に対して、指定された役割を持つべきである。にもかかわらず、大規模な精製スキームおよび無関係な手法はよく、方法の頑強性を保証するため、不純物の排除に必要とされる。

本研究で用いる重要な品質特性および認容された承認基準
培養細胞
完全なヒトモノクローナル抗体を含むチャイニーズハムスター卵巣(CHO)細胞の培養細胞上清を用いた。mAb1はpI7.6の4型IgG分子であり、mAb2はpI8.4のIgG1分子であり、mAb3はpI9.2のIgG1分子である。
全てのキャプチャークロマトグラフィー実験には、GE HealthcareのMabSelect樹脂を用いた。調整用の規模でのカラムベースのクロマトグラフィー実験は、Unicorn 6.3ソフトウェアで制御するAKTA Avant装置(GE Lifesciences)を用いて実行した。全てのカラムは樹脂の製造者の推奨に従って、研究室内で充填した。
ロード処理後、カラムを最初にPBS、pH7.4、二番目に5mMコハク酸、pH5.8で洗浄する。10mMリン酸、pH3.0緩衝液を用いてカラムからmAbを溶出する。洗浄の添加物の添加による、クロマトグラフィーの性能を評価するため、追加の種々の洗浄を洗浄1と洗浄2の間に挿入した。テストした種々の洗浄は、20mMコハク酸500mM NaCl、500mML−アルギニンpH5.8、20mMコハク酸0.1%トリトンX100pH5.8、20mMコハク酸5%PEG400pH5.8、および17mMリン酸ナトリウム4%カプリル酸pH7.4を含んでいた。
ロード処理後、カラムを最初にPBS、pH7.4緩衝液で洗浄し、次に200mM炭酸ナトリウム、1M塩化ナトリウム緩衝液、pH9.6または10.4を用いて、第2の洗浄工程を行った。第2の洗浄工程の次に、100mM炭酸ナトリウム緩衝液、pH9.6または10.4を用いて第3の洗浄工程に進み、次に35mMリン酸ナトリウム、pH6.0緩衝液を用いて第4の洗浄工程を行う。10mMリン酸、pH3.0緩衝液を用いてカラムからmAbを溶出する。
溶出後、アフィニティークロマトグラフィーからの生成物のプールは、0.1N塩酸を用いて生成物を低いpH(3.4から3.6)に調製し、その後1時間保つことによって、低いpHでのウイルス不活化を受けた。生成物を次に2Mトリスで中和(7.0から7.4)した。中和後の生成物のプールを0.2μMのフィルターで濾過した。
残留しているCHO宿主細胞タンパク質(CHO−HCP)の定量化のためのELISAを、TECAN(登録商標)リキッドハンドリングシステムを用いて、ハイスループット方式で行った。試料中の宿主細胞タンパク質のレベルを、CHO宿主細胞タンパク質第三世代キット(CHO Host Cell proteins 3rd generation kit)(Cat# F550, Cygnus Technologies, Southport, NC)を用いて、製造者のプロトコルに従って、定量化した。TECAN(登録商標)インフィニットM1000Proリーダーを用いて、450/650nmにおける吸光度を測定した。
試料中に残留しているCHO−DNAのレベルを決定するため、リアルタイム定量PCR(RT−PCR)を用いた。RT−PCRは、MyiQシングルカラーリアルタイムPCR解析システム(Bio-Rad Lab., Hercules, CA, USA)を用いて、製造者の指示に従い、SYBR(登録商標)グリーンPCRマスターミックス(SYBR(登録商標) green PCR master mix)(Bio-Rad Laboratories, Hercules, CA, USA)で行った。
残留しているプロテインA(rProA)の定量化のためのELISAを、TECAN(登録商標)リキッドハンドリングシステムを用いて、ハイスループット方式で行った。残留しているプロテインAを、残留プロテインAキット(residual protein A kit)(Cat # 9333-1, Repligen)を用いて、製造者のプロトコルに従って定量化した。TECAN(登録商標)インフィニットM1000Proリーダーを用いて、450/650nmにおける吸光度を測定した。
円二色性スペクトルを、恒温六連セルチェンジャーを備えるJasco J−815 CD分光光度計で収集した。mAb試料を対応する緩衝液に0.25mg/mLとなるように希釈し、スペクトルを260から195nm、1.0mmの経路長の石英セルで25℃で収集した。データをSpectraManagerソフトウェア(Jasco)で処理し、緩衝液で収集したスペクトルを、各mAbにおける実験上のペプチドの濃度、MW、およびアミノ酸数に基づいて、平均残基楕円率に規準化した。
熱安定性のプロファイルを確認するため、DSCを各ロットのmAbに対して行った。試料を対応する処理緩衝液で0.75mg/mLに標準化した。走査は、10〜100℃の温度勾配を用いて90℃/時間の勾配速度で、MICROCAL(登録商標)キャピラリーDSC上で行った。緩衝液のみにおける走査のプロファイルを試料の信号から引き、変性中間点(Tm)の値を得るため、データをMICROCAL(登録商標) Origin 7.0分析ソフトウェアを用いて非2状態モデルでフィットした。
サイズ排除クロマトグラフィーを、各捕捉戦略による精製後のmAbの純度を評価するため、Waters' Acquity H-Class Bio UPLC(登録商標)を用いて行った。この検定を行うため、Acquity UPLC(登録商標) BEH200 SEC1.7μmカラム(4.6×150mm)を用いた。ここで、移動相はpH6.8のPBSからなり、1mL/分の流速で実行された。実行中はカラムを常に25℃で維持した。
mAbを、インライン(inline)の多角度光散乱検出器と連結したサイズ排除クロマトグラフィー(SEC−MALS)で調べた。Prominence Shimadzu UFLCを連結したShodex PROTEIN KW-803カラムで、200mMリン酸カリウム150mM塩化ナトリウム、pH6.8を含み、0.02%アジ化ナトリウムを含む緩衝液中で、0.5mL/分の流速で、無勾配分離を行った。50μgの試料をSIL-20AC Prominence Shimadzuオートサンプラーを用いてカラム上に注入し、データを直列につないだ三つのオンライン(online)の検出器から得た;Prominence SPD-20ADダイオードアレイUV/Vis分光光度計に続く、Wyatt miniDAWN TREOS(登録商標)多角度光散乱検出器の後の、Wyatt OPTILAB(登録商標) T-REX示差屈折率検出器。Astra (Wyatt)およびLabsolutions (Shimadzu)を用いて、データを収集し、分析した。分子量およびサイズバリアントのパーセンテージを、分析の専門家によって得られ、同等であるまたは同等でないと要約されたものとして報告した。
mAbの等電点(pI)のアイソフォームおよび分解物における相対的存在量の分布を知るため、全カラムイメージングのキャピラリー等電点電気泳動(iCIEF)を行った。試料のpI分布を三つの区分:酸性の群、主要なピーク、および塩基性の群、で報告し、各実験における標準試料のpI分布と比較した。pIマーカー(6.14および9.46)を含め、各注入におけるpI値の較正に用いた。試料由来のピーク下の面積を積分し、各区分におけるピークの合計を全面積の積分に対する割合として表すことによって、相対的存在量を各電気泳動図から計算した。
mAbに関連するタンパク質種(サブユニット、混入物および分解物)の相対的存在量の分子量(MW)分布を測定するために、SDSを含むゲルを充填したキャピラリー内におけるキャピラリー電気泳動を行った。サイズおよび電気泳動の移動度に基づいて、タンパク質を分離する。この方法では全長30cm(50μm ID)、有効な長さ20cmのキャピラリーおよび214nmの吸収の定点検出を用いた。注入内の相対的な泳動時間を全ての試料に存在する内部標準(10kDa)で較正する。相対的な泳動時間および補正したピークの面積を計算し、両脇の標準試料の注入と比較した。CE−SDS分布の相対的存在量を還元条件および非還元条件で別々に評価した。
マイクロプレートを0.5〜2.0μg/mLの抗体で覆った。このプレートを0.05%PBSTで洗浄し、ブロックした。このプレートを次に再度洗浄し、12の種々の濃度のmAbを添加し、インキュベートした。洗浄後、HRP結合二次抗体を添加した。インキュベーション後、プレートを洗浄し、TMB基質を添加した。プレートリーダー(Tecan Infinite M1000 Pro)で450nmおよび650nmにおける信号を測定した。結合曲線を基準試料と比較し、パーセント結合能を決定した。
それぞれのテストした洗浄緩衝液の溶解度を検査するため、製造者のプロトコルに従って、脱塩プレート(Thermo Scientific, Cat# 89808)を用いて、ハイスループット方式で、mAbを緩衝液交換した。緩衝液交換後、mAbを19〜25℃で24時間インキュベートし、上記のようにSE−UPLCで溶解度を評価した。
抗体の試料を還元し、アルキル化し、トリプシンで消化した。トリプシンペプチドをC−18カラム(Waters BEH C18, 1.7 u 2.1*100 mm 130A)で分離し、215nmおよび280nmにおいてUV検出器で検出し、次に質量分析計で検出した(LTQ-Orbitrap-Elite)。無処理のペプチドと、イオンクロマトグラムで選択した修飾したペプチドのピーク面積を比較することにより、相対定量を達成した。
アフィニティークロマトグラフィー中におけるpH9.6および10.4でのアルカリ性洗浄の有用性を評価するため、方法および生成物の品質特性を調べた。不純物の排除能を、CHO−HCP、CHO−DNA、およびrProAの分析によって調べた。純度のプロファイルを、SE−HPLC、iCIEF、CE−SDS、およびSEC−MALSを用いて調べた。機能的および構造的な分子の完全性を、結合ELISA、CD、DSC、インタクトマス(intact mass)、N末端ペプチドマッピングを用いて調べた。高いpHでの洗浄およびベースライン洗浄を伴って精製した生成物は同等の回収量であった。mAb1においては、回収量は90%から99%、mAb2では84%から90%そしてmAb3では93%から97%であった。
アフィニティークロマトグラフィー中における不純物の排除能を最大化するため、出願人は最初にCHO−HCPにおける洗浄のpHの影響に目を向けた。伝統的に、プロテインAクロマトグラフィーでは、ロード処理およびチェイス(chase)に続き、溶出前にpH移行洗浄が適用される。移行洗浄はpHを下げ、溶出のために樹脂を調製することを意図する。ここで、最適なHCP排除能のpH範囲を決定するために、出願人はチェイスと同じ伝導率を用いてpH7.0ないし10.4の範囲のさらなる洗浄を加えた。図2は洗浄のpHが増加すると、溶出液中のCHO−HCPのレベルが減少することを示す。これらの結果を基に、最も良い不純物の排除能がpH9.0を超えるpHで観察され、洗浄はテストした最も高いpH(10.4)で、特に有効であった。これらの結果を確かめ、この現象がこのモノクローナル抗体に特異的でないことを確定するため、3つの異なるmAbの精製を、pH9.6および10.4の洗浄で行った。ここで本研究は、アフィニティーキャプチャー中のアルカリ性pH洗浄の戦略の利用が、mAbのフィードからのCHO宿主細胞タンパク質の除去に有効であることを示す。アフィニティーキャプチャークロマトグラフィー中のアルカリ洗浄の使用は、mAb、ProAリガンド、およびCHO−HCPタンパク質表面の静電的な性質を変えることで、生成物/不純物および/または不純物/樹脂の相互作用を攪乱し、一方、その間にmAbはカラムと結合している。アフィニティークロマトグラフィー中に、CHO−HCPは、mAbにも、ベースマトリックス(base matrix)にも、ProAリガンドにも結合できる。HCPとmAbとの相互作用が支配的な機構であるという根拠が存在する[6,9,12]。しかしながら、HCPのある一部がベースマトリックス自身[1,2,6]またはProAリガンド[12,17]と相互作用し得ることもまた明らかである。例えば、二つの異なるpHにおけるアフィニティー樹脂からの溶出は、結果として著しく異なった溶出HCPのレベルとなり得る。HCPがmAb、ベースマトリックス、またはProAリガンドのどれと相互作用するかに関わらず、非常に高いpHでのタンパク質−タンパク質相互作用の撹乱は、非常に有効であることを示す。生成物−不純物相互作用は主に静電的であるが、Mabとリガンドとの結合はより強い。プロテインAは主に、疎水性相互作用に由来する結合エネルギーの大部分による誘導適合により、四つの安定化する水素結合で、抗体と結合する[20,21]。加えて、ファンデルワールス力、疎水性相互作用および静電力のような相互作用もまた、ProA−Mab結合に寄与する[13]。ここでデータは、高いpHでの洗浄を使用して、不純物とmAb間および/または不純物と樹脂間の相互作用を選択的に攪乱する一方で、抗体は樹脂に結合させ得ることを示唆する。
SE−UPLC、iCIEF、CE−SDS、およびSEC−MALSを用いて、純度のプロファイルを調べた。表2に示すように、SE−UPLC分析は、テストした全ての三つのmAbにおいて、アルカリ性pH洗浄の使用によって生成物の純度が一パーセント以上増加したことを示す。iCIEFを用いる生成物の質の分析は、アルカリ性pH洗浄の使用によって、酸性またはアルカリ性の種のレベルが増加せず、分子のpIが変化しなかったことを示した。アルカリ性pH洗浄を用いてテストしたどのmAbにおいても脱アミドは観測されなかった。還元および非還元のCE−SDSで測定した全体の純度は、ベースライン条件と比較して有意な差を示さなかった。

mAbにおける生成物の純度のプロファイル
mAbは相対的に安定なタンパク質であり、薬剤の理想的な候補だが、mAbは、製剤原料および製剤の製造中の分解やダメージなど、化学的および物理的変化に脆弱であり得る。我々の方法の戦略はアルカリ性pH洗浄を含んでいたため、各mAbの物理化学的および構造的な完全性を評価するために研究を行った。円二色性によって、二次構造の同一性または内容には差は検出されなかった。mAbのフォールディングの程度の別の尺度である融点もまた影響がなかった。加えて、ELISA結合による活性(正常なフォールディングの尺度)は、アルカリ性pH洗浄がmAbのフォールディング、活性、および全体の機能性に影響を与えないことを示した。等電点電気泳動またはLC/MSによるトリプシンペプチドマッピングによって、高いpHで洗浄したmAbにおいて、チャージバリアント、酸化または脱アミド化に差は観察されなかった。
各組のmAb試料において、アルカリ性pH洗浄を用いて二つの試料を作成し、ベースラインの方法を用いて別の二つの試料を作成した。対照として、ベースラインの試料を緩衝液交換し、アルカリ性pH洗浄を用いて作成した二つの試料と同様の緩衝液種中の試料を作成した。結果は、全ての試料において、凝集が1%未満であることを示す。検出された凝集体のオリゴマーの状態には、ほんのわずかの差しかなかった。各試料の主要なピークにおける分子量の値は使用装置の誤差内である。

SEC−MALS分析
結合ELISAは、ある分子がその特異的なレセプターに結合する能力を調べ、正常にフォールドしたモノクローナル抗体に直接関連する。結合ELISAのデータは、洗浄の戦略に関わらず、全てのmAbが標準試料と同様に結合することを示す。これは、アルカリ性pH洗浄が分子の三次構造に有意な影響を与えなかったことを示す。

部分的に変性したmAbは正常にフォールドしている時よりも低い融解温度を示すため、示差走査熱量測定(DSC)は、mAbの構造上の性質を示すことができる生物物理学における技術である。抗体の融点に対する洗浄の影響を知るため、アルカリ性pH洗浄およびベースラインプロトコルを用いて精製した様々なmAbについて、DSCを測定した。

遠紫外の円二色性(CD)分光を用いて、両方の手順を用いて精製した三つのmAbの二次構造を特徴付けた。260nmから195nmまでの波長間における、様々な条件下での標準試料およびmAbのCDスペクトルを評価した。それぞれのmAbのアミノ酸配列に基づいて算出した分子量とアミノ酸残基の数を用いて、CD信号をミリ度から平均残基楕円率に変換した。遠紫外CDスペクトルは、mAb1では217および229nm、mAb2では217〜218nm、およびmAb3では217〜218nmに極小値を示した。mAb1の平均の平均残基楕円率は、217nmでは−3128±358deg cm2dmol−1残基−1(4条件における平均±標準偏差)および229nmでは−1738±204deg cm2dmol−1残基−1であった。mAb2の平均の平均残基楕円率は、217nmにおいて−3477±454deg cm2dmol−1残基−1であった。mAb3の平均の平均残基楕円率は、217nmにおいて−3392±83deg cm2dmol−1残基−1であった。mAbの精製に用いたプロトコルに関わらず、極小値およびスペクトルの形状はそれぞれのmAbで維持された。CDスペクトルの形状および強度に関する定性的な類似は、アルカリ性pH洗浄の戦略が研究中のmAbの二次構造に影響を与えないことを示す。mAb間に違いはあるが、それぞれ個別のmAbにおいて、テストした全ての洗浄条件下のCDスペクトルは定性的に似ていた。全ての試料のスペクトルはβシートの内容と一致した。
三つの方法によって製剤原料を還元し、アルキル化し、トリプシンで消化する。トリプシンペプチドをC−18カラムで分離し、215および280nmにおいてUV検出器で検出し、その後質量分析計で検出した(LTQ-Orbitrap-Elite)。無処理のペプチドと、イオンクロマトグラムで選択した修飾したペプチドのピーク面積を比較することにより、相対定量を達成した。重鎖のインジケータペプチドからの酸化生成物のピーク面積のパーセンテージによって、酸化をモニターし、重鎖における別のインジケータペプチドからの四つの脱アミド化生成物のピーク面積のパーセンテージによって、脱アミド化をモニターした。酸化および脱アミド化のレベルは、全ての三つのmAbにおけるベースライン試料およびアルカリ性pH試料において、同等であった。

アルカリ性pH洗浄の戦略とアルギニン洗浄の戦略の両方を用いて精製したmAbの生成物及び方法の質の比較
多くの精製の方法では、産業における品質基準に見合うように、2〜3のポリッシングクロマトグラフィーの工程を用いる。相互に関係しない不純物の排除法は、方法の頑強性を増加し得るが、製造業の経済は合理化した方法を作成するための発展を推進する。ここで、出願人は、アフィニティークロマトグラフィーにおいて、洗浄工程をさらに有効にでき、そしてそれは一以上のポリッシング工程の排除を可能にし得ることを示す。表9は、キャプチャークロマトグラフィー中の高いpHでの洗浄の使用が、CEXポリッシング工程が必要なくなる程度に不純物の排除能を改善することを示す。アフィニティークロマトグラフィー中の結合の広範な性質を探索することによって、出願人は、攻撃的なpHでの洗浄を使用して、不純物の排除能を増強できることを示す。

Claims (13)
- 対象のタンパク質および一以上の混入物を含む混合物から対象のタンパク質を精製する方法であって、対象のタンパク質が抗体であり、混合物が、哺乳類培養細胞であり、以下を含む方法:
a)混合物を第1のクロマトグラフィーマトリックスにさらす、ここで対象のタンパク質は第1のクロマトグラフィーマトリックスに結合し、第1のクロマトグラフィーが、プロテインAアフィニティークロマトグラフィーおよびプロテインGアフィニティークロマトグラフィーから選択されるアフィニティークロマトグラフィーである;
b)第1のクロマトグラフィーマトリックスを第1の洗浄溶液と接触させる、ここで第1の洗浄溶液はpHが約9.5から約10.5の間であり、第1の洗浄溶液は約0.01〜1.0Mの範囲の濃度の炭酸ナトリウムおよび約0.5〜2Mの範囲の濃度の塩化ナトリウムを含み、アルギニンまたはアルギニン誘導体を含まない;および
c)第1のクロマトグラフィーマトリックスから溶出溶液中に対象のタンパク質を溶出する。 - アフィニティークロマトグラフィーがプロテインAアフィニティークロマトグラフィーである、請求項1記載の方法。
- 抗体がモノクローナル抗体である、請求項1または2記載の方法。
- モノクローナル抗体がヒト抗体、ヒト化抗体およびキメラ抗体からなる群から選択される、請求項3記載の方法。
- 第1の洗浄溶液のpHが約9.6である、請求項1から4のいずれかに記載の方法。
- 第1の洗浄溶液のpHが約10.4である、請求項1から4のいずれかに記載の方法。
- 第1の洗浄溶液の後に、第1のクロマトグラフィーマトリックスを第2の洗浄溶液と接触させることをさらに含む、請求項1から6のいずれかに記載の方法、ここで第2の洗浄溶液はpHが少なくとも9.0であり、アルギニンまたはアルギニン誘導体を含まない。
- 第2の洗浄溶液の後に、第1のクロマトグラフィーマトリックスを第3の洗浄溶液と接触させることをさらに含む、請求項7記載の方法、ここで第3の洗浄溶液はpHが約6と約7の間であり、アルギニンまたはアルギニン誘導体を含まない。
- 第2の洗浄溶液が約0.01〜1.0Mの範囲の濃度の炭酸ナトリウムを含む、請求項1から8のいずれかに記載の方法。
- 混合物を一以上のさらなるクロマトグラフィーマトリックスにさらす、請求項1から9のいずれかに記載の方法。
- 一以上のさらなるクロマトグラフィーマトリックスが、イオン交換クロマトグラフィー(例えば、陰イオン交換クロマトグラフィーまたは陽イオン交換クロマトグラフィー)、疎水性相互作用クロマトグラフィー、およびミックスモードクロマトグラフィーから選択される、請求項10記載の方法。
- 混合物が、回収された培養細胞液、培養細胞上清、および条件培養細胞の上清、細胞溶解物、および清澄化したバルクから選択される、請求項1から11のいずれかに記載の方法。
- 哺乳類培養細胞がチャイニーズハムスター卵巣(CHO)培養細胞である、請求項1から12のいずれかに記載の方法。
