Description
本発明は、微生物(microorgansim)の検出のための方法およびシステムに関する。
生物学的試料および食物をベースとする試料の両方における細菌および他の微生物の検出に強い関心が集まっている。病原性細菌は、ヒトおよび家畜の間に相当な罹病率、ならびに莫大な経済的損失をもたらし得る。また、特定の微生物、例えば、Escherichia coliまたはSalmonella spp.で汚染された食物の摂取によってもたらされる生命を脅かすまたは致死的な疾病の集団発生を考慮すれば、微生物の検出は米国食品医薬品局(FDA)において優先度が高い。
一態様において、本発明は、試料中の微生物の検出のための、微生物の生物学を利用する。種々の微生物は、本明細書に記載の方法を使用して検出することができる。

本発明は、微生物の検出のための方法を提供する。本発明の方法およびシステムの実施形態のそれぞれは、細菌細胞を含めて、ならびに食物、水、臨床試料および市販の試料からの病原体を含めて、種々の微生物の検出および定量化に適用することができる。本発明の方法は、伝統的な生物学的増菌を必要とせずに、短時間で高い検出感度を提供する。例えば、本発明の実施形態は、試料中の単一の微生物(例えば、細菌細胞)の検出および定量化を提供することができる。
特定の実施形態において、本発明は、試料中に存在する低レベルの微生物(例えば、単一の微生物)を検出する手段として、対象の微生物に結合することができる高い特異性の作用物質を使用する。例えば、一実施形態において、本発明は、試料からの微生物の単離のために感染性因子の特異性を利用する方法およびシステムを含む。例えば、特定の実施形態において、バクテリオファージを使用して、細菌を単離し得る。

バクテリオファージタンパク質および/またはリボソームタンパク質の増幅免疫検出
特定の態様において、本発明は、試料中に存在する低レベルの分析物(例えば、対象のタンパク質)を検出する手段として、固体支持体にカップリングしている高い特異性の生体分子を利用し、シグナルをさらに増幅する。特定の実施形態において、同定されるタンパク質が直接または捕捉抗体を通して不動態化した表面に結合している免疫化学(例えば、ELISAもしくはRIA)または免疫蛍光アッセイについて、約10,000倍のシグナル増幅を達成することができる。
一実施形態において、リボソームは、ラテラルフローアッセイ(LFA)または免疫クロマトグラフィーアッセイを使用して検出し得る。このようなアッセイは、迅速でかつ行うのが容易であり得、1時間以内に視覚的な結果を生じさせ得る。一実施形態において、アッセイは、抗体−支持体および結果の読み出しとしての役割を果たすカーボンブラックナノストリング(CBNS)を使用した超高感度のラテラルフローアッセイを含み得る(Lonnbergら、J. Immunol. Methods、339巻:236〜244頁(2008年))。
感染性因子によって提供される増幅
別の実施形態において、本発明は、試料中の微生物を検出する迅速かつ高感度の方法を提供し、この方法は、試料と、遊離しているか、または固体支持体に結合している結合剤に結合している感染性因子とを接触させる工程であって、感染性因子は、マイクローブに対して特異的であり、そのため試料からのマイクローブを単離することができる工程を含む。例えば、特定の実施形態において、対象のマイクローブは、細菌であり、感染性因子は、1種または複数種のバクテリオファージである。または、他のタイプの微生物のために、他の感染性因子を使用し得る。
微生物から単離された子孫バクテリオファージおよび/またはリボソームの存在はまた、当技術分野で周知の他の方法によって決定し得る。例えば、子孫バクテリオファージは、通常のプラークアッセイ法によって、または例えば、セルソーター、例えば、蛍光活性r化細胞選別(FACS)を含めた自動化技術によって検出し得る。
本発明の方法およびシステムの実施形態のそれぞれは、試料中のマイクローブの迅速な検出および定量化を可能とすることができる。例えば、本発明による特定の方法は、最も好ましくは、約2時間以下で行うことができる。

本発明の実施形態はまた、本発明の方法を行うためのシステム(例えば、キット)を含rむ。
本発明のキットは、試料からの細菌の単離のための試薬を含み得る。
特定の実施形態において、キットは、対象のマイクローブからのリボソームおよび/またはリボソームタンパク質の検出のための試薬を含み得る。例えば、一実施形態において、キットは、例えば、E.coliまたはSalmonella spp.または哺乳動物細胞の精製したリボソームに対する、ウサギ、マウス、モルモットなどにおいて産生されたポリクローナル抗血清を含み得る。
ファージをベースとする検出技術のために、市販のファージのいずれかを使用して、本発明のキットのための試薬を作製し得る。例えば、ATCCから入手可能であるファージrタイプの一覧は、細菌およびバクテリオファージのカタログとしてこれらによって公表され、ワールドワイドウェブ上でatcc.orgにおいて、または他の公知の保管所で利用可能である。
代わりの実施形態において、キットは、ビーズをベースとする免疫アッセイのための試薬を含み得る。
精製したリボソームの単離
E.coliはATCC(#11303)から得て、LB培地(Difco:10gのトリプトン、5gの酵母抽出物、10gのNaCl/L)中で37℃にて一晩培養した。コロニーを5mLのLB培地に入れ、37℃にて一晩インキュベートした。翌日、培養物を新鮮なLBで1:100に希釈し、細胞をKlett68の濃度に増殖させた(600nmにてOD=0.8)(Summerson光電比色計上で20Klett単位=分光光度計上で540nmにて0.1OD)が、これは約3.5時間の増殖の後の概ね4×108個の細胞/mLに対応する。次いで、細胞を、Beckman Coulter XL80Kタイプ19ローター(カタログ番号325632)において5,000rpmで15分間ペレット化した。ペレットを、1/10容量の10X Bugbuster(Novagen、PN70921−3)、0.5mg/mlのリゾチーム(Sigma L6876)、および10μg/mLの高品質デオキシリボヌクレアーゼI(リボヌクレアーゼ非含有)(USB14365)を含む、1mLのTMS(50mMのトリス−HCl、pH7.8、10mMのMgCl2、100mMのNaCl)に再懸濁した。ライセートを、Beckman Coulter Allegra 64R F2402ローターにおいて1.5mLの微量遠心管中で16,000rpmにて10分間清澄化した。上清を新鮮な微量遠心管に移し、清澄化工程を繰り返した。リボソームを、Beckman Coulter SW40Tiローターにおいて4℃にて35,000rpmで3時間遠心分離してTMS中でペレット化し、ペレットを0.5mlのTMSに再懸濁した。懸濁液を清澄化し、TMS(BioComp GradientMaster、BioComp、New Brunswick、CA)中の10〜30%(w/v)スクロース勾配へとロードし、Beckman Coulter SW40Tiローターにおいて4℃にて35,000rpmで3時間スピンした。このように得られたリボソームのバンドを、3mLシリンジの21G×1’’針の使用によって取り出した。リボソームを上記のように再びペレット化し、ペレットをPBSまたはTMSに再懸濁させた。リボソームタンパク質をSDS−PAGEによって特徴付け、粒子の純度をEMネガティブ染色によって確認した。
ウサギにおけるポリクローナル抗体の産生
実施例1において記載したように単離した精製したリボソームを、適切なアジュバントと合わせた。リボソームおよびアジュバントを、幼ウサギ(2.5〜3.0kg;10〜16週齢)の皮膚下に注射した。血液を19ゲージ針で耳中心動脈から集め、37℃にて一晩凝血および退縮させた。次いで、凝血した血液を24時間冷蔵し、その後、血清をデカントし、2500rpmで20分間の遠心分離によって清澄化した。ウサギに注射し、下記のスケジュールによって採血した。
−4日目:事前採血(pre bleed)
0日目:CFA(完全フロイントアジュバント)を使用して皮内(ID)経路によって免疫
7日目:IFA(不完全フロインドアジュバント)を使用して皮内(ID)経路によってブースタ注射
14日目:IFAを使用して皮下(SC)経路によってブースタ注射
28日目:IFAを使用して皮下(SC)経路によってブースタ注射
38日目:試験採血
40日目:免疫前の採血品および試験採血品をNGIに送る
45日目:最後の採血(承認)
47日目:最後の採血品をNGIに送る
49日目:プロジェクトの終わり(承認)。
細胞特異的抗体および磁気微粒子を使用した溶液からの細菌捕捉
溶液から損なわれていない生きた細菌細胞の捕捉を実証するために、様々な細菌種(例えば、E.coliおよびS.typhimurium)の表面エピトープに対するウサギポリクローナル抗体を作製した。
ビーズ増幅の様々なフォーマットを使用したリボソームの捕捉
実験を行って、図8、パネルA〜CおよびF(すなわち、方法1a、1b、2、3および6)において示した様々なアッセイフォーマットを比較した。本質的に、E.coliライセート(500,000個の細胞)を、6M(10,000個の細胞/μl)のチオシアン酸グアニジン中で細菌細胞を溶解することによって調製した。次いで、チオシアン酸グアニジンの濃度を、リン酸緩衝液中への希釈(例えば、450μl中への90,000個の細胞の希釈)によって0.12Mまたは0.6Mに低減させた。次いで、リボソーrムを、図8に示すリボソーム捕捉検出方法の1つを使用して捕捉した(表1)。
ラテラルフローアッセイ
LFA試験ストリップは、Medtox(A LabCorp社)と協力して開発した。ストリップは、ニトロセルロース膜、およびニトロセルロース膜と接触した吸収パッドからなる。ニトロセルロース膜は、ウサギ抗リボソーム抗体を有する試験ライン、およびrヤギ抗ウサギ抗体を有する対照ラインを含有する。ストリップの略図を、図10Aに示す。
ビオチン化T4バクテリオファージの調製
約2.4×1010プラーク形成単位(PFU)のT4ファージを、50μlのリン酸緩衝食塩水(phosphage buffered saline)(PBS)に希釈rした。ファージがトリス含有緩衝液、例えば、TMS中にある場合、例えば、Zebaスピンカラム(Thermo Scientific Pierce、Waltham、MA)を使用して、緩衝液の交換を行うことが必要である。
インジケーターファージまたはインジケーター細菌を使用したファージをベースとする細胞捕捉および検出
a.インジケーター細菌
子孫ファージをアッセイするためのインジケーター細菌の使用を、図11において示す。このように、このアッセイにおいて、対象の細菌試料を感染させるのに使用される親ファージを子孫ファージから分離し、次いで、子孫ファージを使用して、細菌の溶菌によって検出可能なシグナルを提供する生体分子を発現するように操作したインジケーター細菌に感染させる。例えば、細菌は、ルシフェラーゼタンパク質を発現するように操作し得る。
インジケーターファージを使用する戦略を、図13に示す。この方法において、ファージキャプシドタンパク質は、ルシフェラーゼと融合している。これにより、ビオチン化されていない(not biotinlyated)子孫ファージは、ビオチン化および直接的に定量化される親ファージから分離することができる。
可溶性ルシフェラーゼを有するインジケーターファージを使用する戦略を、図14に示す。この方法において、ファージは、Socキャプシドタンパク質の代わりにルシフェラーゼを発現する。ルシフェラーゼの発現はSocプロモーターによって駆動され、高い発現をもたらす。この場合、親ファージはルシフェラーゼ(ファージストック精製の副生成物)を含んでいないため、親ファージおよび子孫ファージを分離する必要はない。試料中の細菌の増殖性感染のみが、検出可能なルシフェラーゼを生じさせる。
Claims (10)
- 対象の細菌を検出するための方法であって、
試料中の他の成分から少なくとも1種の細菌を単離する工程と、
該少なくとも1種の細菌を複製の間可溶性組換えタンパク質を発現するように遺伝子操作された複数の溶菌性親バクテリオファージで感染させる工程であって、該可溶性組換えタンパク質の発現が、ウイルスキャプシドプロモーターにより駆動され、該親バクテリオファージが、該試料中の該少なくとも1種の細菌を見出し、結合し、感染して、溶菌性感染サイクルをもたらし、100個以上の子孫ファージ粒子を少なくとも37摂氏温度であり、そして45摂氏温度以下である温度で、約半時間から1時間で産生させる、工程と、
該少なくとも1種の感染を受けた細菌を、該細菌が溶解して該細菌中に存在する子孫バクテリオファージを放出するまでインキュベートする工程と、
該子孫バクテリオファージの複製の間に発現される該可溶性組換えタンパク質を検出する工程とを含み、
該可溶性組換えタンパク質の検出は、該細菌が該試料中に存在することを示す、方法。 - 前記親バクテリオファージが、前記子孫バクテリオファージから分離されない、請求項1に記載の方法。
- 前記可溶性組換えタンパク質が、基質を目に見える産物に変換するために使用し得る酵素、フルオロフォア、またはルミネセンス分子から選択される、請求項1に記載の方法。
- 前記可溶性組換えタンパク質が、ルシフェラーゼである、請求項3に記載の方法。
- 対象の微生物を検出するための成分を含むキットであって、
試料中の他の成分から少なくとも1種の対象の微生物を単離するための成分と;
該少なくとも1種の対象の微生物を感染させるための成分であって、該感染させるための成分が、複製の間可溶性組換えタンパク質を発現するように遺伝子操作された複数の親感染性因子であり、該可溶性組換えタンパク質の発現が、ウイルスキャプシドプロモーターにより駆動され、該親感染性因子が、該試料中の該少なくとも1種の細菌を見出し、結合し、感染して、溶菌性感染サイクルをもたらし、100個以上の子孫感染因子を少なくとも37摂氏温度であり、そして45摂氏温度以下である温度で、約半時間から1時間で産生させる、感染させるための成分と;
該子孫感染性因子の複製の間に発現される可溶性組換えタンパク質を検出するための成分とを含み、
該可溶性組換えタンパク質の検出は、該微生物が該試料中に存在することを示す、キット。 - 前記親バクテリオファージが、前記子孫バクテリオファージから分離されない、請求項1に記載の方法。
- 前記バクテリオファージがT4バクテリオファージである、請求項1に記載の方法。
- 単離する工程は、細菌学的フィルター上の試料から前記細菌を濃縮することを含む、請求項1に記載の方法。
- 前記可溶性組換えタンパク質はルシフェラーゼである、請求項1に記載の方法。
- 前記ウイルスキャプシドプロモーターが、T4 Socプロモーターである、請求項9に記載の方法。
