Description
The present invention relates to a detection apparatus and a detection method for detecting the presence or amount of a substance to be detected by utilizing surface plasmon resonance. The present invention also relates to a detection chip and a kit that can be used in the detection apparatus and the detection method.
If a minute amount of a substance to be detected such as protein or DNA in a sample can be detected with high sensitivity and quantitative value in a clinical test or the like, it becomes possible to quickly grasp the patient's condition and perform treatment. Therefore, there is a need for a method that can detect a substance to be detected in a sample with high sensitivity and quantitative.
As a detection device that can detect a substance to be detected with high sensitivity, a device using surface plasmon-field enhanced fluorescence spectroscopy (hereinafter abbreviated as “SPFS”) is known (for example, a patent). Reference 1).
In the detection device described in
In general, provision of a liquid containing a substance to be detected, a fluorescent substance, a capturing body, and the like to a container and recovery of the liquid from the container are performed using a pipette. In a detection device that automatically operates a pipette, the tip of the pipette is disposed in the vicinity of the bottom surface of the storage section, and the liquid in the storage section is collected. However, the pipette length varies from individual to individual. For this reason, when the pipette is replaced, the position of the tip of the pipette with respect to the bottom surface of the storage section changes, and as a result, the amount of remaining liquid in the storage section also changes. The amount of the remaining liquid in the container can affect the subsequent reaction. Therefore, the difference in the amount of residual liquid may affect the detection result of a very small amount of the substance to be detected.
Therefore, in the detection device described in
However, in the detection device described in
The present invention has been made in view of the above points, and is a detection apparatus and detection method using SPR, in which the position of the tip of a pipette is adjusted in advance and a substance to be detected is detected with high accuracy. It is an object of the present invention to provide a detection device and a detection method capable of performing the above. Another object of the present invention is to provide a detection chip and a kit that can be used in this detection apparatus and detection method.
In the detection apparatus and detection method using SPR, the present inventors adjust the position of the tip of the pipette using the metal film of the detection chip when detecting the presence or amount of the substance to be detected in the specimen. The present invention was completed by finding out what can be done and further studying it.

Hereinafter, an embodiment of the present invention will be described in detail with reference to the drawings. Here, as a representative example of the detection apparatus according to the present invention, a surface plasmon excitation enhanced fluorescence analyzer (SPFS apparatus) that detects the presence or amount of a substance to be detected contained in a specimen will be described.
FIG. 1 is a schematic diagram showing a configuration of a surface plasmon excitation enhanced fluorescence analyzer (SPFS apparatus) 100 according to an embodiment of the present invention. As illustrated in FIG. 1, the
As shown in FIG. 1, the
The
The shape of the
The
It should be noted that the resonance angle (and the enhancement angle in the vicinity thereof) is generally determined by the design of the
On the other hand, the
The
The material of the
Although not shown in particular, a capture body for capturing a substance to be detected is fixed on the surface of the
The
The

As shown in FIG. 1, the excitation light α enters the

The

The material of the pipette 111 is not particularly limited. Examples of the material of the pipette 111 include resin and glass. As the material of the pipette 111, polypropylene is usually used. The method for imparting conductivity to the tip of the pipette 111 is not particularly limited. For example, the
The liquid chip 115 is a container for storing a liquid such as a specimen, a labeling liquid, or a cleaning liquid. As the liquid chip 115, a plurality of containers are usually arranged according to the type of liquid, or a chip in which a plurality of containers are integrated is arranged.

The liquid
The
The liquid in the channel 51 is again sucked by the pipette 111 and discharged to the liquid chip 115 or the like. By repeating these operations, reaction with various liquids, washing, and the like can be performed, and a detection target substance labeled with a fluorescent substance can be arranged in the reaction field in the flow path 51.
The detection
The
The
The type of light source is not particularly limited, and is, for example, a laser diode (LD). Other examples of light sources include light emitting diodes, mercury lamps, and other laser light sources. When the light emitted from the light source is not a beam, the light emitted from the light source is converted into a beam by a lens, a mirror, a slit, or the like. When the light emitted from the light source is not monochromatic light, the light emitted from the light source is converted into monochromatic light by a diffraction grating or the like. Furthermore, when the light emitted from the light source is not linearly polarized light, the light emitted from the light source is converted into linearly polarized light by a polarizer or the like.
The beam shaping optical system includes, for example, a collimator, a band pass filter, a linear polarization filter, a half-wave plate, a slit, and a zoom unit. The beam shaping optical system may include all of these or a part thereof. The collimator collimates the excitation light α emitted from the light source. The band-pass filter turns the excitation light α emitted from the light source into narrowband light having only the center wavelength. This is because the excitation light α from the light source has a slight wavelength distribution width. The linear polarization filter turns the excitation light α emitted from the light source into completely linearly polarized light. The half-wave plate adjusts the polarization direction of the excitation light α so that the P-wave component is incident on the
The APC mechanism controls the light source so that the output of the light source is constant. More specifically, the APC mechanism detects the amount of light branched from the excitation light α with a photodiode (not shown) or the like. The APC mechanism controls the input energy by a regression circuit, thereby controlling the output of the light source to be constant.
The temperature adjustment mechanism is, for example, a heater or a Peltier element. The wavelength and energy of the light emitted from the light source may vary depending on the temperature. For this reason, the wavelength and energy of the light emitted from the light source are controlled to be constant by keeping the temperature of the light source constant by the temperature adjusting mechanism.
The
For example, the
As described above, among the incident angles of the excitation light α to the

The

The
The
The
The

Next, the detection operation of the SPFS device 100 (the detection method according to the embodiment of the present invention) will be described. FIG. 2 is a flowchart illustrating an example of an operation procedure of the
First, preparation for measurement is performed (step S10). Specifically, the
Next, the position of the tip of the pipette 111 is adjusted (step S20). Specifically, the
Next, the specimen is provided on the
Next, the optical blank value is measured (step S40). Here, the “optical blank value” means the amount of background light emitted above the
Next, the target substance captured on the
Next, the amount of fluorescent γ emitted from the vicinity of the surface of the
By the above procedure, the presence or amount of the substance to be detected in the liquid component of the specimen can be detected.
In the
In the
In the present embodiment, the detection device and the detection method for adjusting the position of the tip of the pipette 111 by measuring the impedance between the
FIG. 5A is a graph showing the relationship between the distance (d) between the metal film and the tip of the pipette and the capacitance (C) between the metal film and the tip of the pipette, and FIG. It is a graph which shows the relationship between the space | interval (d) between the front-end | tip part of a film | membrane and a pipette, and the resonant frequency ((omega) 0 ) between the front-end | tip part of a metal film and a pipette.
Further, in the above-described embodiment, the
In the above-described embodiment, the detection apparatus and the detection method that combine excitation light and surface plasmon by totally reflecting excitation light at the interface between the
Furthermore, although the detection apparatus and detection method using SPFS have been described in the above embodiment, the detection apparatus and detection method according to the present invention are not limited to the detection apparatus and detection method using SPFS. For example, the detection device and the detection method according to the present invention may be a detection device and a detection method using the SPR method. In this case, the
The detection apparatus and the detection method according to the present invention can detect a substance to be detected with high reliability, and are useful for, for example, examination of diseases.
