Description
a) 酸化物表面を有する担体にペプチド含有溶液を塗布し、および乾燥させ、
b) 洗浄によって担体から非結合のペプチドを除去し、および担体を乾燥させ、
c) 担体に付着したペプチドを染色液で染色し、残存する染色液を洗い流し、および担体を乾燥させ、
d) 担体に付着した染色されたペプチドを、抽出溶液を用いて担体から除去し、および染色されたペプチドを含む上澄み液に対して分光光度測定を行う
ことを特徴とする方法に関する。
2x 10μLのペプチド溶液(2mg/mL、1x PBS中に溶解、pH 7.4、Gibco製)を、HDG鋼鈑(円形物、直径2.3cm)に塗布した。最初の10μLを約15’間乾燥し、次いで第2の10μLを同じ位置上に滴下し、30’間乾燥した。UVP製、HB-1000ハイブリダイゼーションオーブン中、水200mLの存在下に30℃で乾燥を行った。
洗浄のため、1リットルの1x PBS、pH 7.4、を含有する結晶皿(直径19cm)内にプレートを配置し、B.ブラウン製のCertomat U装置中60rpmで、室温で10分間わずかに撹拌した。その後、プレートを30℃で簡単に乾燥させた。
プレートを6ウエル皿中へ移動した。150μLのクマシ染色液を、プレート上にピペットで移した。30秒後、染色液をすすぐためにエッジに6mLの1x PBSバッファー、pH 7.4を注意深く添加した。次いで、残存する染色液を除去するために、2x 6mLの1x PBS、pH 7.4中へプレートを移した。プレートを30℃で簡単に乾燥した。
(クマシ染色液:2gのクマシ・ブリリアントブルーR-250、0.5gのクマシ・ブリリアントブルーG-250、50mLのメタノール、425mLのエタノール、100mLの氷酢酸、および蒸留水約1000mL。)
20μLのDMSO/CH3COOH溶液(v/v 9/1)を、染色されたスポット上にピペットで移した。抽出溶液を室温にて5’間作用させ、次いで、液体を、スポットがプレートから完全に除去されるまで、上下にピペットで移すことにより引き抜いた。反応槽中へ上澄み液を移した。
色強度の測定は、Nanodrop 1000分光光度計(UV VISプログラム)において行なった。この目的で、2-μLのサンプルを塗布し、602nmで測定した。
Claims (7)
- アミノ酸配列RSIVTFSLRQNR(ペプチド8)、QRNLIKSTCRKI(ペプチド9)、SRARLFVVTYHK(ペプチド6)、HMISTMNAASRR(ペプチド7)、RNTIRIRTIKHP(ペプチド13)またはRHSSTLRYRPLP(ペプチド15)を有するドデカペプチド。
- 請求項1に記載のドデカペプチドのためにコードする核酸。
- ドデカペプチドのためにコードする請求項2に記載の核酸を含むベクター。
- ドデカペプチドのためにコードする請求項2に記載の核酸で形質転換された、原核生物又は真核細胞。
- 請求項1に記載のドデカペプチドを含有するコーティング剤、ペイントおよび表面被覆、並びに酸化物表面用接着促進剤又は接着剤。
- 複合材料の少なくとも2つの隣接する層の間、又はコーティングと基材の間の接着促進剤として請求項1に記載のドデカペプチドから完全に又は部分的に構成される化合物を含有する、多層複合材料又は被覆基材。
- コーティング剤、ペイントおよび表面被覆の成分、酸化物表面用接着促進剤又は接着剤の成分、および多層複合材料又は被覆基材の成分としての、請求項1に記載のドデカペプチドの使用。
