第1の態様では、本発明は、配列番号1のアミノ酸配列を含む重鎖CDR1、および/または配列番号2のアミノ酸配列を含む重鎖CDR2、および/または配列番号3のアミノ酸配列を含む重鎖CDR3、ならびに/あるいは配列番号4のアミノ酸配列を含む軽鎖CDR1、および/または配列番号5のアミノ酸配列を含む軽鎖CDR2、および/または配列番号6のアミノ酸配列を含む軽鎖CDR3を含む、ヒトOX40と結合するアンタゴニスト抗体またはその断片を提供する。
例えばELISA、BIAcore(登録商標)、ウェスタンブロット、RIAおよびフローサイトメトリー分析を含む、例えばヒトOX40に対する抗体の結合能力を評価するための標準的なアッセイは、当技術分野において知られている。適切なアッセイは、実施例に詳細に記載する。抗体の結合動態(例えばKDのような結合親和性)も、スキャッチャードまたはBIAcore(登録商標)システム分析によるなどの当技術分野において知られる標準的なアッセイにより評価できる。相対的結合親和性Kiは、当技術分野において知られる標準的な競合アッセイにより評価できる。
本開示は、ヒトOX40と結合する抗体およびその断片をコードする単離核酸、ベクターおよび核酸またはベクターを含む宿主細胞も提供する。核酸は、細胞全体、細胞溶解物または部分的に精製もしくは実質的に精製された形で存在することがある。核酸は、アルカリ/SDS処理、CsClバンド分け、カラムクロマトグラフィー、アガロースゲル電気泳動および当技術分野においてよく知られている他のものを含む標準的な技法により、他の細胞構成成分または他の混入物、例えば他の細胞性核酸またはタンパク質から精製分離された場合に、「単離」または「実質的に純粋に」されている。例えばF.Ausubelら編(1987)Current Protocols in Molecular Biology、Greene Publishing and Wiley Interscience、New Yorkを参照されたい。本発明の核酸は、例えば、DNAまたはRNAであり得、イントロン配列を含んでも含まなくてもよい。好ましい実施形態では、核酸は、cDNA分子である。
OX40ポリペプチドに対して作製された抗体は、よく知られており日常的なプロトコールを用いて、動物の免疫化、すなわち動物、好ましくは非ヒト動物へのポリペプチドの投与によりにより得ることができる。例えばHandbook of Experimental Immunology(Weir DM(編)、第4巻、Blackwell Scientific Publishers、Oxford、England、1986)を参照されたい。ウサギ、マウス、ラット、ヒツジ、ウシ、ラクダまたはブタなどの多くの温血動物を免疫できる。しかし、マウス、ウサギ、ブタおよびラット、特にマウスは、一般的に最も適切である。抗体は、当業者に知られる組換えDNA技法によっても生成できる。さらに、抗体は、自然に存在する抗体の酵素または化学的切断により生成できる。本発明のヒト化抗体は、非ヒト動物(例えばマウス)からのVHおよび/またはVL領域からの1もしくは複数のCDRまたはその一部を、ヒトVHおよび/またはVL領域からの1または複数のフレームワーク領域に移行することにより構築できる。場合によって、このようにVHおよび/またはVL領域中に存在するヒトフレームワーク残基は、抗体の免疫原性を減少させ、かつ/または結合親和性を維持するために必要または所望される場合に、対応する非ヒト(例えばマウス)残基で置き換えることができる。場合によって、CDR中に存在する非ヒトアミノ酸残基は、ヒト残基で置き換えてよい。本発明のキメラまたはヒト化抗体は、上記のようにして調製した非ヒトモノクローナル抗体の配列に基づいて調製できる。重鎖および軽鎖免疫グロブリンをコードするDNAは、対象の非ヒトハイブリドーマから得て、非マウス(例えばヒト)免疫グロブリン配列を含有するように、標準的な分子生物学的技法を用いて工学改変できる。例えば、キメラ抗体を創出するために、当技術分野において知られる方法を用いて、マウス可変領域をヒト定常領域に連結できる(例えばCabillyらへの米国特許第4,816,567号を参照されたい)。ヒト化抗体を創出するために、当技術分野において知られる方法を用いて、マウスCDR領域をヒトフレームワークに挿入できる(例えばWinterへの米国特許第5,225,539号、Queenらへの米国特許第5,530,101号;第5,585,089号;第5,693,762号および第6,180,370号を参照されたい)。
抗体についてのスクリーニングを、ヒトOX40との結合を測定するためのアッセイおよび/またはOX40とそのリガンドであるOX40Lとの結合を阻止する能力を測定するためのアッセイを用いて行うことができる。結合アッセイの例は、特に、プレート上に固定化されたヒトOX40とヒトFcとの融合タンパク質を用い、融合タンパク質と結合した抗OX40抗体を検出するためのコンジュゲート2次抗体を採用するELISAである。阻止アッセイの例は、ヒトCD4細胞上のOX40へのOX40リガンド融合タンパク質結合の阻止を測定する、フローサイトメトリーに基づくアッセイである。蛍光標識2次抗体を用いて、細胞と結合するOX40リガンド融合タンパク質の量を検出する。このアッセイは、上清中の抗体が、OX40とリガンド融合タンパク質との結合を阻止するにつれてのシグナルの低減を求める。阻止アッセイのさらなる例は、プレートを被覆するOX40リガンド融合タンパク質が媒介するナイーブヒトT細胞の共刺激の阻止を、チミジン取り込みを測定することにより測定するアッセイである。抗OX40抗体の機能的活性、例えばT細胞活性化の低減を評価するためのアッセイとして、実施例3および8に記載するヒト混合リンパ球反応(MLR)を用いることができる。本発明の抗体は、当業者に知られる様々な方法で単離または精製できる。標準的な精製方法は、大気圧またはFPLCおよびHPLCのようなシステムを用いて高圧で行う、イオン交換、疎水性相互作用、親和性、サイズ分けまたはゲルろ過および逆相を含むクロマトグラフィー技法を含む。精製方法は、電気泳動、免疫学的、沈降、透析および等電点電気泳動の技法も含む。タンパク質濃縮とともに限外ろ過およびダイアフィルトレーション技法も有用である。OX40抗体を精製するために、選択された宿主細胞を、例えばモノクローナル抗体精製のためにスピナーフラスコ中で成長させることができる。上清をろ過し、濃縮した後に、プロテインA−セファロース(Pharmacia、Piscataway、NJ)を用いて親和性クロマトグラフィーを行う。溶出された抗体をゲル電気泳動および高性能液体クロマトグラフィーにより確認して、純度を確認できる。本発明の好ましい抗体は、よって、ヒトOX40と結合する単離および/または精製抗体である。
別の態様では、本発明は、細胞毒、薬物(例えば免疫抑制剤)または放射性毒素のような治療剤に連結した、ヒトOX40と結合するアンタゴニストOX40抗体またはその断片を提供する。このようなコンジュゲートは、本明細書において、「イムノコンジュゲート」という。1または複数の細胞毒を含むイムノコンジュゲートは、「免疫毒素」という。細胞毒または細胞傷害性薬剤は、細胞に有害な(例えば死滅させる)任意の薬剤を含む。例は、タキソール、サイトカラシンB、グラミシジンD、臭化エチジウム、エメチン、マイトマイシン、エトポシド、テノポシド、ビンクリスチン、ビンブラスチン、コルヒチン、ドキソルビシン、ダウノルビシン、ジヒドロキシアントラシンジオン、ミトキサントロン、ミトラマイシン、アクチノマイシンD、1−デヒドロテストステロン、グルココルチコイド、プロカイン、テトラカイン、リドカイン、プロプラノロールおよびピューロマイシンならびにそれらの類似体またはホモログを含む。治療剤は、例えば、代謝拮抗薬(例えばメトトレキセート、6−メルカプトプリン、6−チオグアニン、シタラビン、5−フルオロウラシルデカルバジン)、アルキル化剤(例えばメクロレタミン、チオエパ、クロラムブシル、メルファラン、カルムスチン(BSNU)およびロムスチン(CCNU)、シクロトスファミド、ブスルファン、ジブロモマンニトール、ストレプトゾトシン、マイトマイシンCならびにシス−ジクロロジアミン白金(II)(DDP)シスプラチン)、アントラサイクリン(例えばダウノルビシン(以前のダウノマイシン)およびドキソルビシン)、抗生物質(例えばダクチノマイシン(以前のアクチノマイシン)、ブレオマイシン、ミトラマイシンおよびアントラマイシン(AMC))および有糸***阻害剤(例えばビンクリスチンおよびビンブラスチン)も含む。本発明の抗体と連結できる治療用細胞毒の他の例は、デュオカルマイシン、カリチアマイシン、マイタンシンおよびオーリスタチン、ならびにそれらの誘導体を含む。カリチアマイシン抗体コンジュゲートの例は、市販で入手可能である(Mylotarg(登録商標);American Home Products)。細胞毒は、本発明の抗体と、当技術分野において利用可能なリンカー技法を用いて連結できる。細胞毒と抗体とをコンジュゲートするために用いられているリンカーの型の例は、それらに限定されないが、ヒドラゾン、チオエーテル、エステル、ジスルフィドおよびペプチド含有リンカーを含む。例えばリソソーム区画内の低pHによる切断に感受性またはカテプシン(例えばカテプシンB、C、D)のような腫瘍組織において優先的に発現されるプロテアーゼのようなプロテアーゼによる切断に感受性であるリンカーを選択できる。細胞毒の型、リンカーおよび治療剤と抗体をコンジュゲートする方法についてのさらなる議論について、Saito Gら、(2003)Adv.Drug Deliv.Rev.55:199〜215頁;Trail PAら、(2003)Cancer Immunol.Immunother.52:328〜337頁;Payne G(2003)Cancer Cell、3:207〜212頁;Allen TM(2002)Nat.Rev.Cancer,2:750〜763頁;Pastan IおよびKreitman RJ(2002)Curr.Opin.Investig.Drugs、3:1089〜1091頁;Senter PDおよびSpringer CJ、(2001)Adv.Drug Deliv.Rev.53:247〜264頁も参照されたい。本発明の抗体は、放射活性同位体に連結して、ラジオイムノコンジュゲートともよばれる細胞傷害性放射性医薬品を作製することもできる。診断または治療用に用いるために抗体とコンジュゲートできる放射活性同位体の例は、それらに限定されないが、ヨウ素131、インジウム111、イットリウム90およびルテニウム177を含む。ラジオイムノコンジュゲートを調製するための方法は、当技術分野において確立されている。ラジオイムノコンジュゲートの例は、Zevalin(登録商標)(EDEC Pharmaceuticals)およびBexxar(登録商標)(Corixa Pharmaceuticals)を含んで市販で入手可能であり、同様の方法を用いて、本発明の抗体を用いてラジオイムノコンジュゲートを調製できる。本発明の抗体イムノコンジュゲートを用いて所定の生物学的応答を改変でき、薬物部分は、古典的な化学的治療剤に限定されると解釈されない。例えば、薬物部分は、所望の生物学的活性を有するタンパク質またはポリペプチドであってよい。このようなタンパク質は、例えば、アブリン、リシンA、シュードモナス外毒素もしくはジフテリア毒素のような酵素活性毒素またはその活性断片;腫瘍壊死因子もしくはインターフェロン−γのようなタンパク質;あるいは例えばリンホカイン、インターロイキン−1(「IL−1」)、インターロイキン−2(「IL−2」)、インターロイキン−6(「IL−6」)、顆粒球マクロファージコロニー刺激因子(「GM−CSF」)、顆粒球コロニー刺激因子(「G−CSF」)もしくは他の増殖因子のような生物学的応答改変物質を含むことがある。
別の態様では、本発明は、本発明のアンタゴニスト抗体またはその断片と薬学的に許容される担体とを含む組成物、例えば医薬組成物を提供する。このような組成物は、1もしくは組み合わせ(例えば2以上の異なる)抗体、および/または本発明のイムノコンジュゲート、および/または上記のような細胞毒、薬物(例えば免疫抑制剤)もしくは放射性毒素のような治療剤を含むことがある。例えば、本発明の医薬組成物は、標的抗原上の異なるエピトープと結合するか、または相補的な活性を有する抗体の組み合わせ(またはイムノコンジュゲート)を含み得る。本発明の医薬組成物は、併用治療において投与することもでき、すなわち他の薬剤と組み合わせることができる。例えば、併用治療は、少なくとも1つの他の抗炎症剤または免疫抑制剤と組み合わせた本発明のアンタゴニストOX40抗体を含み得る。
本発明のアンタゴニスト抗体は、OX40媒介性障害の診断および処置を含む、多数のin vitroおよびin vivoの診断および治療用の利用性がある。例えば、これらの分子は、in vitroもしくはex vivoで培養中の細胞に、または例えばin vivoでヒト対象に投与して、様々なOX40媒介性障害を処置、防止および診断できる。好ましい対象はヒトであり、OX40活性により媒介される障害(OX40媒介性障害)を有する患者を含む。本発明のアンタゴニスト抗体は、患者のOX40共刺激状態とは独立して、患者を処置するために効果的であり得る。より好ましい対象はヒトであり、低レベルのOX40を発現する患者を含む。
(a)患者血液試料から末梢血単核細胞(PBMC)を精製するステップと、
(b)PBMCをフローサイトメトリー分析に供するステップと、
(c)CD4+および/またはCD8+T細胞中のOX40陽性細胞の数を決定し、前記数を対照レベルと比較するステップと
を含む方法も提供する。
本開示の別の実施形態では、OX40媒介性障害の処置のための、本発明のアンタゴニスト抗体もしくはその断片、組成物またはイムノコンジュゲートを含む製造品が提供される。製造品は、容器と、容器上のまたは容器に付随するラベルまたは添付文書を含むことがある。適切な容器は、例えば瓶、バイアルまたはシリンジを含む。容器は、ガラスまたはプラスチックのような様々な材料から形成してよい。容器は、状態を処置するために効果的であり得る組成物を保持し、滅菌アクセスポートを有することがある(例えば容器は、皮下注射針が突き刺すことができるストッパーを有する静脈内溶液バッグまたはバイアルであってよい)。組成物中の少なくとも1つの活性物質は、本明細書に記載するアンタゴニスト抗体であってよい。ラベルまたは添付文書は、組成物を、がんのような所望の状態を処置するために用いることができることを示してよい。一実施形態では、ラベルまたは添付文書は、アンタゴニスト抗体を含む組成物を用いてOX40媒介性障害を処置できることを示してよい。
組換えヒトOX40−Fcタンパク質を生成するために、ヒトTNFRSF4についてのcDNAをimaGenes(クローン番号:RZPDB737H0329D;Berlin、Germany)から購入した。このcDNAを鋳型として用いて、ヒトTNFRSF4細胞外ドメインのDNAコード領域(配列番号11)をPCR増幅した。別のPCR反応では、ヒトIgG1のFc領域(EU位223〜451)をPCRにより増幅して、5’GSGGGリンカーおよび3’SA−6×Hisリンカーならびにクローニングのための制限部位を付加した。2つの得られた産物を、次いで、フランキングプライマーを用いるオーバーラップ伸長PCRを用いて融合して、米国特許第5924939号に記載されるIgドナーアクセプター断片(第1イントロン)を有するヒトCMVプロモーター、OriP配列(Koons MDら、(2001)J Virol.75(22):10582〜92頁)、SV40エンハンサーおよびKim Dら、(2003)Biotechnol.Prog.19(5):1620〜2頁に記載されるガストリンターミネーターと融合したSV40ポリAを含有する、InvitrogenからのpcDNA3.1(−)プラスミドに基づく改変哺乳動物発現ベクター(Invitrogen AG、Basel、Switzerland、Cat.No.V795−20)への後続のクローニングのための制限部位を付加した。この組換えプラスミドは、哺乳動物細胞でのヒトTNFRSF4細胞外ドメイン−Fc融合タンパク質の発現と、ヒトTNFRSF4タンパク質の天然のシグナルペプチドにより駆動される細胞培養培地への分泌とを可能にした。組換えタンパク質生成のために、上記の組換えベクターを懸濁馴化HEK293細胞(ATCC番号CRL1573)に、jetPEI(商標)トランスフェクション試薬(Polyplus−transfection S.A.、Strasbourg、France;流通業者:Brunschwig、Basel、Switzerland)を用いてトランスフェクトした。細胞培養上清を5日後に回収し、AKTA FPLCシステム(GE Healthcare Europe GmbH、Glattbrugg、Switzerland)上で操作されるプロテインA親和性精製カラム(HiTrapプロテインAセファロースカラム;GE Healthcare Europe GmbH、Glattbrugg、Switzerland)を用いてさらに精製した。
陽性選択したそれぞれのハイブリドーマについて、トータルRNAを調製し、cDNAに逆転写し、VHおよびVL遺伝子をそれぞれPCRにより増幅した。これらのPCR産物を、レスキューベクター(pDriveベクター;QIAGEN AG、Hombrechtikon、Switzerland;Cat.No.231124)にライゲーションして、個別のPCR産物のDNA配列決定および選択したハイブリドーマの単クローン性または多クローン性の決定を可能にした。このベクターにより、IPTGおよびX−galを含有するLB寒天プレート上での青/白の選択が可能になった(LacZαペプチドによるX−galの分解のために、挿入断片を有さないコロニーは青色であった)。陽性(白)細菌クローンから組換えプラスミドを調製し、ベクター主鎖に特異的な標準的DNA配列決定プライマー(M13rev、M13fwd、T7またはSP6)を用いて配列決定した。DNA配列を、最終的に、哺乳動物細胞での対象の抗体の組換え発現のために発現ベクターにサブクローニングした。
トータルRNAを、2〜10×106細胞から、QIAGENからのRNeasyミニキット(QIAGEN AG、Hombrechtikon、Switzerland;Cat.No.74106)を製造者のプロトコールに従って用いて単離した。試料を、NanoDrop ND−1000分光光度計(WITEC AG、Littau、Switzerland)を用いて定量した。
上記のトータルRNA調製物を、cDNAにさらに逆転写し、VHおよびVL断片をPCRにより、縮合プライマーの2つの異なる混合物(それぞれ1つがマウス免疫グロブリン重鎖可変断片および可変重鎖接合領域の全ての異なるサブファミリーの回収、または全てのマウス免疫グロブリン軽鎖カッパ可変断片および可変軽鎖カッパ接合領域の回収を可能にした)を用いて増幅した。逆転写および増幅のために用いたプライマーは、Microsynth(Balgach、Switzerland)により合成され、HPLC精製された(表1〜4)。逆転写およびPCR増幅はともに、QIAGENワンステップRT−PCRキット(QIAGEN AG、Hombrechtikon、Switzerland;Cat.No.210212)を用いて同時に行った。この技法は特異的プライマーを用いたので、それぞれのmRNA試料を、次いで、2重に処理して、VHまたはVL断片のいずれかの個別の逆転写および増幅を可能にした。RNアーゼフリー水に30μlの最終容量で溶解した2μgのトータルRNAを、10μlのQIAGENワンステップRT−PCR緩衝液の5×ストック溶液、2μlのdNTPミックス(10mMの濃度)、3μlのプライマーミックス(10μMの濃度)および2μlのQIAGENワンステップRT−PCR酵素ミックスと混合した。最終混合物を、次いで、PCRチューブに入れ、以下の設定を用いてPCRサーモサイクラー(BioRad iCyclerバージョン4.006、Bio−Rad Laboratories AG、Reinach、Switzerland)でサイクルにかけた:
50℃にて30分
95℃にて15分
40サイクル:94℃にて30秒

4℃にて保持
PCR産物を、2%アガロースゲルに泳動させた。DNA電気泳動の後に、対象の断片(およそ450bp)をアガロースゲルから切り出し、Macherey−Nagel NucloSpin抽出IIキット250(Macherey−Nagel、Oensingen、Switzerland;Cat.No.740609.250)を用いてさらに抽出した。DNA配列決定のために、抽出したPCR産物を上記のレスキューベクター(pDriveベクター、QIAGEN AG、Hombrechtikon、Switzerland;Cat.No.231124)にクローニングし、E.coli TOP10株(Invitrogen AG、Basel、Switzerland;Cat.No.C404006)を形質転換した。
陽性コロニーを、Macherey−Nagel方形ウェルブロックプレート(Macherey−Nagel、Oensingen、Switzerland;Cat.No.740488.24)中に播種し、100μg/mlアンピシリンを補った1.5mlのLuria Bertani(LB)培地中で37℃にて1晩(振とう250RPM)培養した。次の日に、DNAミニプレップ抽出を、NucleoSpinマルチ8プラスミドキット(Macherey−Nagel、Oensingen、Switzerland;Cat.No.740620.5)を用いて行った。
試料を、DNA配列決定のために、DNA配列決定サービス業者であるFasteris(Plan−les−Ouates、Switzerland)に送った。標準的なプライマーであるM13rev、M13fwd、T7、SP6を用いた(表5)。DNA配列を分析するために、クローンマネージャ9プロフェッショナル版(Scientific&Educational Software、NC、USA)およびBioEdit配列アラインメントエディタ(Hall,TA(1999)Nucl.Acids.Symp.Ser.41:95〜98頁)を用いた。
哺乳動物細胞での組換え発現のために、単離マウスVHおよびVL断片を、アセンブリベースのPCR法を用いて、キメラ免疫グロブリンとしてフォーマットした。これらのキメラ抗体は、マウス重鎖可変ドメインがヒトIgG1重鎖定常ドメイン(γ1、ヒンジ、γ2およびγ3領域)と融合した重鎖と、マウス軽鎖可変ドメインがヒトカッパ定常ドメイン(Cκ)と融合した軽鎖とからなる。PCRにより集合させたマウス可変およびヒト定常部分を、その後、実施例1にて言及したが、ヒト免疫グロブリン軽鎖カッパリーダーペプチドを採用してタンパク質分泌を駆動したことが異なるInvitrogenからの改変pcDNA3.1(−)ベクターに基づく改変哺乳動物発現ベクターにクローニングした。免疫グロブリン候補のタンパク質生成のために、等量の重鎖および軽鎖ベクターDNAを、懸濁馴化HEK−293(ATCC番号:CRL−1573)に同時トランスフェクトした。細胞培養上清を5日後に回収し、AKTA FPLCシステム上で操作されるプロテインA親和性精製カラム(HiTrapプロテインAセファロースカラム)(ともにGE Healthcare Europe GmbH、Glattbrugg、Switzerlandから)を用いて精製した。
OX40特異的抗体検出ELISA:
ハイブリドーマおよび組換え抗体候補による抗体力価、特異性および生成を、直接ELISAにより決定した。簡単に述べると、96ウェルマイクロタイタープレート(Costar USA、流通業者VWR AG、Nyon、Switzerland)をPBS中2μg/mlでの100μlの組換えヒトOX40−hisで被覆した(OX40−hisタンパク質の作製について実施例1を参照されたい)。プレートを4℃にて1晩インキュベートし、次いで、PBS2%BSA(ウシ血清アルブミン、PAA Laboratories、Pasching、Austria)を用いて室温(RT)にて1時間ブロッキングした。ブロッキング溶液を除去し、ハイブリドーマ上清または精製抗体を加えた。プレートをRTにて30分間インキュベートし、次いで、PBS0.01%Tween−20(Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)で9回洗浄し、セイヨウワサビペルオキシダーゼ(HRP)標識ヤギ抗マウスH+L検出抗体(Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)を1:1000の希釈で加えた。ヒトFcを有する組換えキメラ抗体(実施例2を参照されたい)を検出するために、HRP標識ウサギ抗ヒトIgG抗体(Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)を1:1000の希釈で検出抗体として用いた。プレートを30分間室温(RT)にてインキュベートし、PBS0.01%Tween−20で9回洗浄し、TMB基質(Bio−rad Laboratories AG、Reinach、Switzerland)をプレートに加え、6分後にH2SO4を加えることにより反応を停止した。吸光度を次いで450nmにてマイクロプレートリーダー(Biotek、USA;流通業者:WITTEC AG、Littau、Switzerland)で読み取った。図1Aは、キメラ1D4抗体およびキメラ2F8抗体がOX40−his被覆タンパク質を認識することを示す。
組換えヒトOX40リガンドタンパク質(OX40L)を以下のようにして作製した:ヒトTNFSF4についてのcDNA(クローン名:IOH46203)をimaGenes(Berlin、Germany)から購入し、ヒトTNFSF4リガンドの細胞外部分(アミノ酸51〜183)(番号付けはUniprot Q6FGS4配列に従う)を、フランキング制限部位とともに増幅した。ASAリンカーおよび8−Hisタグ配列を5’端に包含する得られたPCR産物を、その後、CMVプロモーター、ウシ成長ホルモンポリアデニル化およびマウスVJ2Cリーダーペプチドを有して組換えタンパク質の分泌を駆動する、Invitrogen(Invitrogen AG、Basel、Switzerland)からの改変バージョンのpREP4ベクターにクローニングした。組換えタンパク質生成のために、組換えベクターを懸濁馴化HEK293細胞(ATCC番号CRL1573)に、jetPEI(商標)トランスフェクション試薬(Polyplus−transfection S.A.、Strasbourg、France;流通業者:Brunschwig、Basel、Switzerland)を用いてトランスフェクトした。細胞培養上清を5日後に回収し、AKTA FPLCシステム(GE Healthcare Europe GmbH、Glattbrugg、Switzerland)上で操作されるプロテインA親和性精製カラム(HiTrapプロテインAセファロースカラム;GE Healthcare Europe GmbH、Glattbrugg、Switzerland)を用いて精製した。
2名の異なるドナーからの血液を、クエン酸塩を抗凝固剤として含む3本の10mL S−Monovette(Sarstedt、Numbrecht、Germany)に回収した。ドナー第1号からの細胞をエフェクター細胞として用い、ドナー第2号からの細胞を標的細胞として用いた。2名のドナーからのPBMC(末梢血単核細胞)を、製造者の使用説明に従って50mL Blood−Sep−Filterチューブ(流通業者:Brunschwig、Basel、Switzerland)を用いて精製した。細胞を、FBSを含まないロズウェルパーク記念研究所(RPMI、PAA Laboratories、Pasching、Austria)培地で2回洗浄した。標的細胞を、50μg/mlのマイトマイシンC(Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)と37℃にて30分間インキュベートした。細胞を、次いで、FBSを含まないRPMIで3回洗浄し、RPMI、10%FBS(PAA Laboratories、Pasching、Austria)、2mM L−グルタミン(Lonza、Leuven、Belgium)、100U/mlペニシリン、100μg/mlストレプトマイシン(Biochrom AG、Berlin、Germany)中に1×106細胞/mLにて再懸濁した。96ウェルU底マイクロプレート(TPP、Trasadingen、Switzerland)中に、50’000の標的細胞および80’000のエフェクター細胞を、各ウェルに100μlの最終容量で分配した。100μlの抗体希釈物をウェルに加えた。プレートを37℃にて5%CO2インキュベータ中で7日間インキュベートした。MLRの開始の7日後に、0.5μCiの3Hチミジン(Perkin Elmer)で細胞をパルスした。パルスの18時間後に、細胞を採集し、取り込まれた放射活性を、Wallacベータカウンタで定量した。図2は、キメラ1D4抗体が陽性対照よりも高い程度までMLRを用量依存的な様式で阻止できることを示す。
ヒト細胞
ヒト白血球を含有するフィルタを、La Chaux−de−Fonds、Switzerlandの血液採取センター(Centre de Transfusion Sanguine et Laboratoire de Serologie、rue Sophie−Mairet 29、CH−2300)から回収した。細胞を、10U/mLのリケミン(Drossapharm AG、Lucern、Switzerland)を含有する60mLのPBSを逆に流すことによりフィルタから取り出した。PBMCを、次いで、製造者の使用説明に従って50mL Blood−Sep−Filterチューブ(流通業者:Brunschwig、Basel、Switzerland)を用いて精製した。細胞を、FBS(PAA Laboratories、Pasching、Austria)を含むロズウェルパーク記念研究所(RPMI、PAA Laboratories、Pasching、Austria)培地で3回洗浄した。細胞を、3×106細胞/mlにて、24ウェルプレート(TPP、Trasadingen、Switzerland)中で、RPMI、10%FBS(PAA Laboratories、Pasching、Austria)、2mMウルトラグルタミン(Lonza、Leuven、Belgium)、100U/mlペニシリン、100μg/mlストレプトマイシン(Biochrom AG、Berlin、Germany)、10μg/mlのフィトヘマグルチニン(PHA;Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)+100U/mLのrHu IL−2(プロリュウキン、Novartis、Basel、Switzerland)に再懸濁した。48時間後に、細胞を回収し、以下に記載するようにしてフローサイトメトリーにより分析した。
カニクイザルからの全血(Eric Rouiller教授、Laboratory of Neurophysiology、University of Fribourg、Fribourg、Switzerlandから得た)を、クエン酸塩チューブ(BD Biosciences、Allschwil、Switzerland)に回収した。2mLのPBSを3mLの血液と混合し、混合物を、10mlの85:15のフィコール:PBS混合物(GE Healthcare Europe GmbH、Glattbrugg、Switzerland)の頂部に重層した。試料を室温にて休止せずに20分間遠心分離した。PBMC層を回収し、PBSで3回洗浄した。細胞を3×106細胞/mLにてダルベッコ改変イーグル培地(DMEM、PAA Laboratories、Pasching、Austria)、10%FBS(PAA Laboratories、Pasching、Austria)、非必須アミノ酸(PAA Laboratories、Pasching、Austria)、1mMピルビン酸ナトリウム(PAA Laboratories、Pasching、Austria)、2mMウルトラグルタミン(Lonza、Belgium)、100U/mlペニシリン(Biochrom AG、Germany)、100μg/mlストレプトマイシン(Biochrom AG、Germany)に再懸濁した。1mLの細胞懸濁液を24ウェルプレート(TPP、Trasadingen、Switzerland)に分配し、10ug/mlのPHA(PHA/M、Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)、100U/mLのrHu IL−2(プロリュウキン、Novartis、Basel、Switzerland)を加えた。細胞を50時間、37℃にて、5%CO2インキュベータ中でインキュベートした。活性化PBMCを回収し、PBS/2.5%FBS(FACS緩衝液)に再懸濁した。50μlのFACS緩衝液中の5000細胞を96ウェルV底プレートに分配し、ビオチン化抗ヒトOX40−キメラ1D4抗体またはビオチン化アイソタイプ対照抗体またはビオチン化した、ヒツジで産生させた市販の抗ヒトOX40(BD Biosciences、Allschwil、Switzerland)をウェルに25μg/mlにて加えた。試料を氷上で20分間インキュベートし、次いで、細胞を冷FACS緩衝液で2回洗浄し、1:20希釈でのストレプトアビジン−PE(BD Biosciences、Allschwil、Switzerland)と氷上で15分間インキュベートした。細胞をFACS緩衝液で1回洗浄し、次いで、300μlのFACS緩衝液に再懸濁した。2μlの容量のヨウ化プロピジウム(PI;Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)を各試料に加えて、死滅細胞を除外した。細胞を、フローサイトメトリーにより分析した(Cyan、Beckman Coulter International S.A.、Nyon、Switzerland)。
動的結合親和性定数(KD)を、プロテインA捕捉抗体上で、実施例1に記載するような組換えヒスチジンタグ付加ヒトOX40受容体細胞外ドメインを分析物として用いて測定した。測定は、BIAcore 2000(GE Healthcare−BIAcore、Uppsala、Sweden)で室温にて行い、BiaEvaluationソフトウェア(BIAcore;v4.1)を用いて分析した。
ヒトアクセプターフレームワーク、逆突然変異ならびにヒトCDRグラフト化アクセプターフレームワークの結合特性を実質的に保持および/または改善する変異の選択を含む、抗ヒトOX40マウス抗体1D4のヒト化について本明細書に記載する。
相同性一致を用いて、1D4 CDRにグラフトするヒトアクセプターフレームワークを選択した。データベース、例えばヒトおよびマウスの免疫グロブリン遺伝子座からの生殖系列可変遺伝子のデータベース(IMGTデータベース(the international ImMunoGeneTics information system(登録商標);Lefranc MPら、(1999)Nucleic Acids Res.27(1):209〜12頁;Ruiz Mら、(2000)Nucleic Acids Res.28(1):219〜21頁;Lefranc MP(2001)Nucleic Acids Res.29(1):207〜9頁;Lefranc MP(2003)Nucleic Acids Res.31(1):307〜10頁;Lefranc MPら、(2005)Dev.Comp.Immunol.29(3):185〜203頁;Kaas Qら、(2007)Briefings in Functional Genomics & Proteomics、6(4):253〜64頁)もしくはVBASE2(Retter Iら、(2005)Nucleic Acids Res.33、データベース版D671〜D674)もしくはKabatデータベース(Johnson Gら、(2000)Nucleic Acids Res.28:214〜218頁)または出版物(例えばKabat EAら、既出)を用いて、マウス重鎖および軽鎖V領域が属するヒトサブファミリーを同定し、アクセプター分子として用いるために最もよくフィットするヒト生殖系列フレームワークを決定できる。アクセプターとして用いるためのこれらのサブファミリー内での重鎖および軽鎖可変配列(VHおよびVL)の選択は、配列相同性および/またはグラフト後に6つのCDRの適当な相対的提示を保存する助けとなるCDR1およびCDR2領域の構造の一致に基づくことができる。
VH1およびVL1についてのコードDNA配列(cDNA)を、scFvフォーマットにおいてGENEART AG(Regensburg、Germany)により合成して、そのことにより、単一DNA配列が両方の可変ドメイン(配列番号31)を包含することを可能にした。個別の可変ドメインcDNAは、このscFv構築物からPCRにより引き出し、PCRアセンブリ技法を用いて、それらのそれぞれの定常ドメインcDNA配列(複数可)の上流でさらに組み立てた。最後に、完全重鎖および軽鎖cDNAを、CMVプロモーターおよびウシ成長ホルモンポリアデニル化シグナルを有する改変pcDNA3.1ベクター(Invitrogen、CA、USA)に基づく独立ベクターにライゲーションした。軽鎖特異的ベクターは、対象の軽鎖可変ドメインcDNAをカッパ軽鎖定常ドメインcDNAの前にBamHIおよびBsiWI制限酵素部位を用いてライゲーションすることにより、ヒトカッパアイソタイプ軽鎖の発現を可能にしたが、重鎖特異的ベクターは、工学改変して、対象の重鎖可変ドメインcDNAを、ヒトIGHG1 CH1、IGHG1ヒンジ領域、IGHG1 CH2およびIGHG1 CH3定常ドメインをコードするcDNA配列の前に、BamHIおよびSalI制限酵素部位を用いてライゲーションできるようにした。重鎖および軽鎖両方の発現ベクターにおいて、分泌は、BamHI部位を含有するマウスVJ2Cリーダーペプチドにより駆動した。BsiWI制限酵素部位は、カッパ定常ドメイン中にあり、SalI制限酵素部位は、IGHG1 CH1ドメイン中で見出される。
1D4マウス抗体からのCDRの単純なグラフト化によりヒトOX40と結合しない候補が得られたので(表6および図4)、ヒト残基がマウス残基で置換される突然変異誘発を開始した。このプロセスは逆突然変異とよばれ、モノクローナル抗体のヒト化において最も予測不可能な手順である。これは、親和性を保存し、同時にヒト化抗体における免疫原性の可能性を最小限にするために、保持する必要があるマウス抗体からの重要なフレームワーク残基の同定および選択を必要とする。表7、表8および図5は、マウス抗体フレームワークとヒト抗体フレームワークとの間で異なる残基(Kabat番号付け)を示す。CDRの高次構造または可変ドメイン間充填に影響し得る残基は、抗体親和性に最も高い影響を与え得るので、特に興味の対象である。
ヒト化抗体の耐熱性を、示差走査熱量分析(DSC)を用いて測定した。モノクローナル抗体融解プロファイルは、それらのアイソタイプの特徴である(Garber EおよびDemarest SJ(2007)Biochem.Biophys.Res.Commun.355:751〜7頁)が、FAB断片の中間融解温度は、全長IgGの関係においてさえ容易に同定できる。FAB部分のこのような中間融解温度を用いて、ヒト化候補のモノクローナル安定性をモニタリングした。
ヒト化抗OX40抗体のエピトープの特徴を決定するために、VH6/VL9抗体を、様々なヒト−ラットOX40キメラタンパク質を用いて、ヒトOX40細胞外領域の規定されたドメインにマッピングした。
ラットおよびヒト−ラットOX40タンパク質を、実施例1に記載した方法に従ってFc融合タンパク質としてフォーマットした。ELISAのために、PBS中2μg/mLのOX40タンパク質で、4℃にて1晩、高結合96ウェルプレート(Coastar)を被覆した。プレートを、PBS2%ウシ血清アルブミン(BSA)でブロッキングした後に、VH6/VL9抗体またはアイソタイプ対照抗体とインキュベートした。プレートを、次いで、洗浄し、ヤギ−抗ヒトIg F(ab’)2断片特異的−HRP(Jackson ImmunoResearch Europe Ltd、Newmarket、UK)とインキュベートした。洗浄の後に、プレートをTMB基質(Bio−Rad Laboratories AG、Reinach、Switzerland)とインキュベートして、抗体結合を明らかにした。反応を、2M H2SO4を加えることにより停止し、光学密度を450nM(OD 450nM)にて、Synergy HT2分光光度計(Biotek、USA;流通業者:WITTEC AG、Littau、Switzerland)で読み取った。
その起源に関係なく、OX40細胞外領域は、ドメイン1、2、3および4とよばれる4つの構造モジュールに分けられている(Compaan DMおよびHymowitz SG(2006)Structure、14(8):1321〜30頁)。ヒトOX40の細胞外領域(ヒトTNFRSF4のアミノ酸29〜214、番号付けはUniprot P43489配列に従う)に対応するキメラOX40タンパク質を、4つのドメインの1または複数をヒト配列とラット配列との間で交換することにより構築した。例えば、キメラRHRR OX40タンパク質は、第2ドメインが対応するヒトドメイン配列で置き換えられたラットOX40細胞外領域に対応する。
in vitro免疫反応を抑制するVH6/VL9抗体の効力を、1方向同種混合リンパ球反応(MLR)において試験した。MLRは、アロ反応性T細胞活性化および増殖のin vitroモデルである(O’Flaherty Eら、(2000)Immunology、100(3):289〜99頁;DuPont BおよびHansen JA(1976)Adv.Immunol.23:107〜202頁)。2名の無関係のドナーからの末梢血単核細胞(PBMC)を混合した場合に、T細胞は、同種主要組織適合性(MHC)分子の認識により活性化される。この活性化は、Tリンパ球の増殖をもたらす。MLR反応は、T細胞標的化免疫抑制薬の効果を実証するために広く用いられている(Bromelow KVら、(2001)J.Immunol.Methods、247(1〜2):1〜8頁)。シクロスポリンのような免疫抑制薬は、T細胞活性化を阻害することにより主に働く。VH6/VL9抗体による阻止効果を試験することに加えて、MLRの阻害に対する抗体依存性細胞傷害作用(ADCC)のような細胞傷害性機序の寄与も調べた。VH6/VL9抗体の3つの異なる抗体フォーマット:IGHG1フォーマット(本明細書においてVH6/VL9という)、非フコシル化IGHG1(IgG1)フォーマット(本明細書において非フコシル化VH6/VL9という)およびIGHG4(IgG4)フォーマット(本明細書においてVH6/VL9 IGHG4 S228Pという)を、このアッセイにおいて試験した。IGHG1(IgG1)抗体は、ADCCのような細胞傷害作用機序について有能であることが知られている。非フコシル化IGHG1抗体は、ナチュラルキラー細胞(NK細胞)のような細胞傷害性細胞上で発現されるFcγRIIIaに対する親和性がより高いので、ADCC活性の増進を示すことが知られている(Mizushima Tら、(2011)Genes Cells、16(11):1071〜80頁)。対照的に、IGHG4(IgG4)抗体は、ADCCのようなFc媒介性細胞傷害作用機序を有さないことが知られている。
IGHG1 CH1、IGHG1ヒンジ領域、IGHG1 CH2およびIGHG1 CH3定常ドメインをコードするcDNA配列を、実施例6に記載する重鎖特異的ベクター中のIGHG4 CH1、S228P置換を有するIGHG4ヒンジ領域、IGHG4 CH2およびIGHG4 CH3定常ドメインをコードするcDNA配列で置き換えることにより、置換S228Pを有するIGHG4免疫グロブリンフォーマット化を達成した。置換S228Pは、ヒトIGHG4重鎖cDNA鋳型に、標準的なPCR突然変異誘発技法により導入した。得られた重鎖は、配列番号57を有する。非フコシル化VH6/VL9 IGHG1抗体の生成は、WO2010/095031パンフレットの実施例14に記載するプロトコールに従った。
2名の異なるヒトドナーからの血液を、抗凝固剤としてクエン酸塩を含む3本の10mL S−Monovette(Sarstedt、Numbrecht、Germany)に回収した。2名のヒトドナーからの末梢血単核細胞(PBMC)を、製造者の使用説明に従って50ML Blood−Sep−Filterチューブ(流通業者:Brunschwig、Basel、Switzerland)を用いて精製した。細胞を、FBSを含まないロズウェルパーク記念研究所(RPMI、PAA Laboratories、Pasching、Austria)培地で2回洗浄した。2名のドナーからの刺激細胞を、50μg/mlのマイトマイシンC(Sigma−Aldrich Chemie GmbH、Buchs、Switzerland)との37℃にて30分間のインキュベーションにより調製した。細胞を、次いで、FBSを含まないRPMIで3回洗浄し、RPMI、10%FBS(PAA Laboratories、Pasching、Austria)、2mM L−グルタミン(Lonza、Leuven、Belgium)、100U/mlペニシリンおよび100μg/mlストレプトマイシン(Biochrom AG、Berlin、Germany)中に1×106細胞/mLにて再懸濁した。レスポンダー細胞を、96ウェルU底マイクロプレート(TPP、Trasadingen、Switzerland)中でRPMI、10%FBS、L−グルタミン、100U/mlペニシリンおよび100μg/mlストレプトマイシン中に再懸濁した。50’000の刺激細胞および80’000のレスポンダー細胞を、各ウェルに100μlの最終容量で分配した。100μlの抗体希釈物(または培地のみ)をウェルに加えた。プレートを37℃にて5%CO2インキュベータ中で7日間インキュベートした。最後の18時間、0.5μCiの3Hチミジン(Perkin Elmer、Basel Switzerland)で細胞をパルスした。細胞をフィルタマットフィルタ(Perkin Elmer)上に採集し、取り込まれた放射活性を、Wallacベータカウンタ(Perkin Elmer)で定量した。
図8に示す結果は、VH6/VL9抗体が、2名の異なる個体(レスポンダー)についてのMLRを、およそ100ng/mLのEC50値で効率的に阻害できることを実証する。結果は、用いる抗体フォーマットに依存して異なる応答も示し、阻止および細胞傷害性機序の寄与における差が、異なる個体からのMLRにおいて観察される。
異種間移植片対宿主(GVH)反応は、ヒト患者における骨髄移植後に観察される同種間移植片対宿主疾患(GVHD)についてのモデルである。GVH反応は、同種または異種MHC認識の結果として宿主環境を攻撃する、グラフト化免疫細胞により媒介される急性免疫応答である(Murphy WJら、(1996)Semin.Immunol.8(4):233〜41頁)。Tリンパ球は、GVH反応の主なエフェクター細胞である。VH6/VL9抗体の免疫抑制効力を、ヒトPBMCを用いるSCIDマウスの再構築に基づく異種GVHDモデルにおいて試験した。このモデルでは、ヒトPBMCおよび主にTリンパ球が、マウス宿主細胞に対して強い応答を開始する。この反応は、特に、体重減少を伴う重度の皮膚および腸の炎症を導く。このモデルの最も適当な読出しは、動物の生存である。
動物(SCIDマウス)を、致死量以下で照射した後に、30百万のヒトPBMCを用いて腹腔内で再構築した。また、TMベータ1抗体を週2回注射することにより、マウスNK細胞をマウスから枯渇させた。VH6/VL9抗体、Enbrel(登録商標)または媒体を用いる処置を、毎週5回の連続する用量をi.v.で与え、PBMC注射の2日前に開始した。動物は、媒体(PBS)、または10mg/kgもしくは1mg/kgのVH6/VL9抗体、または8mg/kgのEnbrel(登録商標)(ヒトIgG1のFc成分と融合したヒト可溶性TNF受容体2の融合タンパク質、Amgen−Pfizer)のいずれかで処置した。動物を、体重減少、下痢、毛皮の外観および全体的な挙動を含むGVHD症状について毎週3回確認して評点した。症状が重度すぎると考えられるならば、動物を道徳的に屠殺した。
図9は、VH6/VL9抗体が、より低い1mg/kg用量であっても、GVHD反応を非常に有力に抑制したことを示す。驚くべきことに、VH6/VL9抗体は、ヒトにおけるGVHDについての認識された治療であるEnbrel(登録商標)を超える、改善された効力を示した(Xhaard Aら、(2011)Bull.Cancer、98(8):889〜99頁;Simpson D(2001)Expert Opin.Pharmacother.2(7):1109〜17頁)。1または10mg/kgのVH6/VL9抗体で処置した動物の生存時間中央値は、媒体処置群(表10)と比較して4倍長く、Enbrel(登録商標)と比較して2倍長かった。さらに、この結果は、VH6/VL9抗体が、アゴニスト活性を有さないことを強調する。なぜなら、アゴニスト性抗OX40抗体は、同種マウスGVHDモデルにおいてGVHDを悪化させると報告されているが(Valzasina Bら、(2005)Blood、105(7):2845〜51頁;Blazar BRら、(2003)Blood、101(9):3741〜8頁)、この事象は本研究で観察されなかったからである。

- ヒトOX40と結合するアンタゴニスト抗体またはその抗原結合断片であって、下記を含むアンタゴニスト抗体またはその抗原結合断片:
(i)配列番号1のアミノ酸配列を含む重鎖CDR1、および配列番号2のアミノ酸配列を含む重鎖CDR2、および配列番号3のアミノ酸配列を含む重鎖CDR3;
ならびに
(ii)配列番号4のアミノ酸配列を含む軽鎖CDR1、および配列番号5のアミノ酸配列を含む軽鎖CDR2、および配列番号6のアミノ酸配列を含む軽鎖CDR3。 - マウス抗体、キメラ抗体またはヒト化抗体である、請求項1に記載の抗体またはその抗原結合断片。

- 重鎖配列が、配列番号35、36、37または38からなる群より選択される重鎖配列の重鎖可変領域配列の非CDR領域と少なくとも80%同一である非CDR領域を含む、請求項6に記載の抗体またはその抗原結合断片。
- (i)IGHV2−70*10(配列番号19)、IGHV2−70*01(配列番号20)、IGHV2−70*13(配列番号21)、IGHV2−5*09(配列番号22)およびIGHV2−70*11(配列番号23)からなる群より選択されるヒト遺伝子の産物であるかもしくは前記遺伝子に由来する重鎖可変フレームワーク領域を含む;または

- (i)アミノ酸改変が、23位、35b位、48位、50位、60位および62位からなる群より選択されるアミノ酸位置でのアミノ酸置換を含み、各群メンバーのアミノ酸位置がKabat番号付けに従って示される;または
(ii)アミノ酸改変が、23S、35bG、48L、50H、60Nおよび62Aからなる群より選択されるアミノ酸置換を含み、各群メンバーのアミノ酸位置がKabat番号付けに従って示される、
請求項8または9に記載の抗体またはその抗原結合断片。 - (i)配列番号8のアミノ酸配列を含む軽鎖可変領域配列;または
(ii)配列番号60、86、87および89からなる群より選択されるアミノ酸配列を含む軽鎖可変領域配列
を含む、請求項1に記載の抗体またはその抗原結合断片。 - (i)配列番号8の軽鎖可変領域配列の非CDR領域と少なくとも80%同一である軽鎖可変領域配列の非CDR領域;または

鎖配列を含む、請求項1に記載の抗体またはその抗原結合断片。 - 軽鎖配列が、配列番号45、46、47または49からなる群より選択される軽鎖配列の軽鎖可変領域配列の非CDR領域と少なくとも80%同一である非CDR領域を含む、請求項13に記載の抗体またはその抗原結合断片。
- (i)IGKV3−11*01(配列番号24)、IGKV1−39*01(配列番号25)、IGKV1D−39*01(配列番号26)、IGKV3−11*02(配列番号27)およびIGKV3−20*01(配列番号28)からなる群より選択されるヒト遺伝子の産物であるかもしくは前記遺伝子に由来する軽鎖可変フレームワーク領域を含む;または
(ii)ヒト遺伝子IGKV3−11*01(配列番号24)の産物であるかもしくは前記遺伝子に由来する軽鎖可変フレームワーク領域を含み、軽鎖可変フレームワーク領域が、対応するマウス抗体の軽鎖可変領域の対応するフレームワーク領域からの少なくとも1つのアミノ酸改変を含む、
請求項1に記載の抗体またはその抗原結合断片。 - 抗体またはその抗原結合断片が、配列番号39のアミノ酸配列を含む軽鎖配列を含み、軽鎖可変フレームワーク領域が、対応するマウス抗体の対応する軽鎖可変フレームワーク領域からの少なくとも1つのアミノ酸改変を含む、請求項15に記載の抗体またはその抗原結合断片。
- アミノ酸改変が、
(i)1位、33位、34位、46位、47位、54位、56位および71位からなる群より選択されるアミノ酸位置でのアミノ酸置換を含み、各群メンバーのアミノ酸位置がKabat番号付けに従って示される;
(ii)1Q、33M、34H、46P、47W、54L、56Sおよび71Yからなる群より選択されるアミノ酸置換を含み、各群メンバーのアミノ酸位置がKabat番号付けに従って示される;または
(iii)アミノ酸位置31位でのアミノ酸欠失を含み、アミノ酸位置がKabat番号付けに従って示される;
請求項15または16に記載の抗体またはその抗原結合断片。 - (a)配列番号37または配列番号38のアミノ酸配列を含む重鎖配列と、
(b)配列番号47のアミノ酸配列を含む軽鎖配列と
を含む、請求項1に記載の抗体またはその抗原結合断片。 - (a)配列番号58または配列番号59のアミノ酸配列を含む重鎖可変領域配列と、
(b)配列番号60のアミノ酸配列を含む軽鎖可変領域配列と
を含む、請求項1に記載の抗体またはその抗原結合断片。 - ヒトOX40と結合する抗体またはその抗原結合断片であって、請求項1から19のいずれか一項に記載の抗体またはその抗原結合断片であり、以下の(a)、(b)、(c)、(d)のいずれか少なくとも1つを満たす、抗体またはその抗原結合断片:
(a)重鎖においてカバット番号35bに対応するアミノ酸がSである;
(b)重鎖においてカバット番号60に対応するアミノ酸がSである;
(c)重鎖においてカバット番号62に対応するアミノ酸がSである;ならびに
(d)軽鎖においてカバット番号34に対応するアミノ酸がAである。 - 重鎖および/または軽鎖定常領域をさらに含み、ヒト重鎖定常領域が、IGHG1、非フコシル化IGHG1およびIGHG4からなる、ヒト免疫グロブリンの群から選択される、請求項1から20のいずれか一項に記載の抗体またはその抗原結合断片。

- 抗体が、親抗体のFc領域と比べて少なくとも1つのアミノ酸改変を含むバリアントFc領域を含み、バリアントFc領域を含む抗体が、親抗体と比較して変更されたエフェクター機能を示す、請求項1から21のいずれか一項に記載の抗体またはその抗原結合断片。
- (i)110nM以下の親和性(KD)でヒトOX40と結合する;
(ii)対応するキメラ抗体のOX40結合親和性(KD)の少なくとも75%を保持する;または
(iii)対応するキメラ抗体と比較した場合に、等しいかまたはより高いOX40結合親和性(KD)を有する、
請求項1から24のいずれか一項に記載の抗体またはその抗原結合断片。 - 抗体が、75℃を超えるFAB断片耐熱温度を有する、請求項1から25のいずれか一項に記載の抗体またはその抗原結合断片。
- 請求項1から26のいずれか一項に記載の抗体またはその抗原結合断片をコードする単離核酸。
- (i)配列番号61もしくは62の核酸配列を含む、重鎖可変領域をコードするDNA;および/または
(ii)配列番号63の核酸配列を含む、軽鎖可変領域をコードするDNA
を含む、請求項27に記載の単離核酸。 - 請求項27または28に記載の単離核酸を含むベクター。
- 請求項27もしくは28に記載の単離核酸または請求項29に記載のベクターを含む宿主細胞。
- 核酸が発現され、抗体が生成されるように、請求項30に記載の宿主細胞を培養するステップを含む、ヒトOX40と結合する抗体またはその抗原結合断片を生成する方法。
- 請求項27または28に記載の単離核酸によりコードされる、ヒトOX40と結合する抗体またはその抗原結合断片。
- 請求項1から26のいずれか一項に記載の抗体またはその抗原結合断片と、薬学的に許容される担体とを含む組成物。

- 医薬品として用いるための、請求項1から26のいずれか一項に記載の抗体またはその抗原結合断片。
- OX40媒介性障害が、感染症(ウイルス、細菌、真菌および寄生虫)、感染症に伴う内毒素ショック、関節炎、関節リウマチ、喘息、気管支炎、インフルエンザ、呼吸器多核体ウイルス、肺炎、COPD、特発性肺線維症(IPF)、原因不明の線維化胞隔炎(CFA)、特発性線維化間質性肺炎、肺気腫、骨盤内炎症性疾患、アルツハイマー病、炎症性腸疾患、クローン病、潰瘍性大腸炎、ペロニー病、セリアック病、胆嚢疾患、毛巣病、腹膜炎、乾癬、血管炎、外科的癒合、脳卒中、I型糖尿病、ライム病、関節炎、髄膜脳炎、自己免疫性ぶどう膜炎、中枢および末梢神経系の免疫媒介性炎症性障害、例えば多発性硬化症、ループス(例えば全身性エリテマトーデス)およびギラン−バレー症候群、アトピー性皮膚炎、自己免疫性肝炎、線維化胞隔炎、グレーブス病、IgA腎症、特発性血小板減少性紫斑病、メニエール病、天疱瘡、原発性胆汁性肝硬変、サルコイドーシス、強皮症、ウェゲナー肉芽腫症、他の自己免疫障害、膵炎、外傷(手術)、移植片対宿主疾患(GVHD)、移植片拒絶、虚血性疾患を含む心血管疾患、例えば心筋梗塞およびアテローム動脈硬化症、血管内凝固、骨吸収、骨粗鬆症、変形性関節症、歯周炎、低酸症ならびに視神経脊髄炎からなる群より選択される、OX40媒介性障害を処置するための請求項1から26のいずれかに記載の抗体またはその抗原結合断片。
- 抗体が、1方向同種混合リンパ球反応(MLR)で測定した場合に、ヒト重鎖定常領域IGHG1を有する抗体と比較して、細胞傷害作用が増進している、請求項35または36に記載の抗体またはその抗原結合断片。
- OX40媒介性障害が、GVHDであり、抗体またはその抗原結合断片が、GVHDの抑制においてEnbrel(登録商標)(ヒトIgG1のFc成分と融合したヒト可溶性TNF受容体2の融合タンパク質)より効果的である、OX40媒介性障害を処置するための請求項1から26のいずれかに記載の抗体またはその抗原結合断片。
- 請求項1から20のいずれか一項に記載の抗体もしくはその抗原結合断片、請求項33に記載の組成物または請求項34に記載のイムノコンジュゲートを含む、OX40媒介性障害の処置のためのキットであって、OX40媒介性障害は以下からなる群より選ばれる、キット:
多発性硬化症、ループス、関節リウマチ、自己免疫性ぶどう膜炎、移植片対宿主疾患(GVHD)、ならびに、移植片拒絶。 - 請求項1に記載の抗体またはその抗原結合断片を用いてOX40の発現レベルが低い患者を検出するin vitroスクリーニング方法であって、
(a)患者血液試料から末梢血単核細胞(PBMC)を精製するステップ、
(b)PBMCをフローサイトメトリー分析に供するステップ、および
(c)CD4+および/またはCD8+T細胞中のOX40陽性細胞の数を決定し、前記数を対照レベルと比較するステップ
を含むスクリーニング方法。
