発現構築物
蛍光タグ付き発現構築物ECFP−PCNA及びhABH21−261−EYFPのクローニングは、以前に説明されている(Aas et al., 2003)。hABH21−261−EYFPを鋳型として利用して、hABH21−10−EYFP及びhABH211−261−EYFPをPCRにより生成した。増幅産物を、それぞれNdeI/AgeI及びAgeI/EcoRIを使用してpEYFP−N1(Clonetech)中にクローニングした。
ESTからのPCR産物(イメージクローン5176979(BC035374)RZPD)を、pEYFP−C1(HindIII/Acc651)中にクローニングして、EYFP−TFIIS−Lを得た。EYFP−XPA構築物は、Wim Vermeulen博士(Department of Cell Biology and Genetics, Rotterdam)から寛大にも提供を受けたHis9
−HA−EGFP−XPA(Rademakers et al., 2003)においてEYFPとEGFP(
NheI/BsrGI断片)とを入れ替えることにより作製した。TFII−I−EYFPは、Robert G. Roeder博士(Laboratory of Biochemistry and Molecular Biology, The Rockefeller University, New York)から寛大にも提供を受けたpI3CX−TFII−I(Roy et al., 1993)からのTFII−IのPCR増幅、及びEYFP−N1(SacI/ApaI)中へのクローニングにより生成した。EYFP−Topo−IIαは、William T. Beck(Division of Molecular Pharmacology, Department of Molecular Genetics, University of Illinois, Chicago)から寛大にも提供を受けたEGFP−Topo−IIα (pT104−1)(Mo and Beck, 1999)からEGFPタグ(EcoRIブラント/NheI)とEYFPタグ(XhoIブラント/NheI)とを入れ替えることにより作製した。F4突然変異体を含むhABH21−7−EYFP構築物は、XhoI/EcoRIオーバーハングでオリゴヌクレオチドをアニーリングし、その後ATGコドンで突然変異したEYFP−N1にクローニングすることにより、作製した。全ての点突然変異を、QuickChange(登録商標)II取扱説明書マニュアルに従い部位特異的突然変異誘発により作製した。制限酵素及び仔牛小腸由来アルカリホスファターゼ(CIP)は、New England Biolabs(登録商標)Inc.から、オリゴヌクレオチドはMedProbe、Eurogentech(Oslo, Norway)から入手した。全ての構築物を、配列決定により確認した。
共焦点画像化及びFRET測定
生きたHeLa細胞を、ECFP及びEYFP融合構築物の一過性のトランスフェクション(製造業者の推奨に従い、Fugene6(Roche Inc.)による)の16時間〜2
4時間後に調べた。蛍光画像を、Plan−Apochromate 63×/1.4油浸対物レンズを備えたZeiss LSM 510 Metaレーザー走査顕微鏡を使用して取得した。連続式スキャンを使用して、増強型シアン蛍光タンパク質(ECFP)はλ=458nmで励起してλ=470nm〜500nmで検出し、増強型黄色蛍光タンパク質(EYFP)はλ=514nmで励起してλ=530nm〜600nmで検出した。切片の厚みは1μmとした。
RET=I2−I1(ID2/ID1)−I3(IA2/IA3)が0より大きい場合)、FRETが起こったものとした。FRETを、式:NFRET=FRET/(I1×I3)1/2を使用して発現レベルに関して正規化した。NFRETは、25ピクセル超を含有する関心領域(ROI)内の平均強度(I)から算出した(全てのピクセルの強度が250未満であり、供与体及び受容体構築物の両方に関する平均強度が100〜200であった)。チャネル1(ECFP)及び3(EYFP)を、上で説明したように画像化のために測定し、チャネル2(FRET)はλ=458nmで励起しλ=530nm〜600nmで検出した。ID1、D2、D3及びIA1、A2、A3は、同時トランスフェクトした細胞(I1及びI3)と同じ設定及び同じ蛍光強度を用いて、ECFP及びEYFP構築物のみでトランスフェクトした細胞に関して確定した。ECFP−PCNA及びEYFP−PCNAは陽性対照として含め、同時発現したタグの二量体形成のために、エンプティベクターから発現したECFP及びEYFPタンパク質を全ての実験において陰性対照として含めた。
細胞株の培養、及び細胞抽出物の調製
対象の構築物を安定して発現するHeLa(子宮頸部癌)及びHaCaT(自発的に形質転換したケラチノサイト)細胞を、トランスフェクション(Fugene6による)、その後の細胞選別又は希釈クローニング、並びに10%ウシ胎児血清(FCS)、アンホテリシンB(250μg/ml、Sigma-Aldrich)、ゲンタマイシン(100μg/ml
、Gibco)及びグルタミン(1mM、BioWhittaker(登録商標))を添加した選択(ゲン
チシン(genticine)、G418、400μg/ml(Invitrogen)を使用する)ダルベ
ッコ変法イーグル培地/高グルコース4.5g/l(DMEM)(BioWhittaker(登録商標))での長期培養により調製した。細胞を5%二酸化炭素、加湿雰囲気中において37℃で培養した。HeLa由来の分画した細胞抽出物を、緩衝液I(10mM Tris−HCl、pH8.0、及び50mM KCl)において1×血中血球容積(PCV)で、及び緩衝液II(10mM Tris−HCl、100mM KCl、20%グリセロール、0.5% Nonidet P−40、10mM EGTA、10mM MgCl2、1mM DTT、1×Completeプロテアーゼ阻害剤(Roche)、ホスフェート
阻害剤カクテル(PIC I及びPIC II、Sigma))において1×PCVで細胞ペ
レットを再懸濁することにより調製した。細胞を、一定振とう下で4℃で30分間、インキュベートした。上清(可溶性画分)を回収した。ペレット(核を含有する)を、緩衝液III(10mM Tris−HCl、pH8.0、及び100mM KCl)において1×PCVで、緩衝液IIにおいて1×PCVで再懸濁し、全ての核が破壊されるまで軽く(briefly)超音波処理した。750μgの核小体含有画分を遠心分離し、ペレット(
クロマチン結合画分)を緩衝液II及びIII中で再懸濁した。クロマチン結合画分を、DNAse/RNAseカクテル(2μl Omnicleave(登録商標)エンドヌクレアーゼ(200U/μl、Epicentre(登録商標)Biotecnologies、WI)、2μl
DNAse(10U/μl、Roche Inc.)、2μl Bensonase(250U/
μl、Novagene、Ge)、2μl 小球菌ヌクレアーゼ(100U/μl〜300U/μl、Sigma-Aldrich)、及び2μl RNAse(10mg/ml、Sigma-Aldrich))と共に室温で30分間、及び37℃で1時間インキュベートした。750μgの可溶性画分を、さらなる2μl Omnicleave(登録商標)と共に、IP時に4℃で終夜インキュベートした。
共免疫沈降(co−IP)及びウェスタン分析(WB)
GFPタンパク質に対して産生したインハウスアフィニティ精製ウサギポリクローナル
抗体(EYFP及びECFPタンパク質も認識する)を、New England Biolabs(登録商
標) Incから得た手順に従ってプロテインA常磁性ビーズ(Dynal(登録商標))と共有結合した(以下ではα−GFPビーズと称する)。各画分を、4℃で終夜定速回転時にα−GFPビーズ(10μl)と共にインキュベートした(IP)。IP後、間に氷上でのインキュベーション(5分間)を行いながら、200μl 10mM Tris−HCl、50mM KCl(pH7.5)でビーズを4回洗浄した。ビーズをその後NuPAGE(登録商標)(Invitrogen)ローディング緩衝液及び1mM DTT中で再懸濁し、加熱し、10%又は4%〜12% Bis−Tris−HCl(NuPAGE(登録商標))ゲル上で分離し、PVDF膜(Immobilon(登録商標)、Millipore)に
移した。膜を、5%低脂肪粉乳を含むPBST(0.1% Tween(登録商標)20を含むPBS)中で1時間ブロッキングした。一次抗体であるα−PCNA(PC10、Santa Cruz biotechnology Inc.)及びα−GFPを1%粉乳に含むPBST中で希釈し、1時間インキュベートし、その後それぞれ二次抗体であるポリクローナルウサギ抗マウスIgG/HRP及びポリクローナルブタ抗ウサギIgG/HRP(DakoCytomation, Denmark)と共に1時間インキュベーションした。膜を化学発光試薬(SuperSign
al(登録商標)West Femto Maximum、PIERCE)で処理し、タンパク質をKodak Image Station 2000Rで可視化した。
配列解析
初期配列解析のために、Swiss−Prot及びTrEMBLデータベースを使用して対象の配列領域と類似の部分配列を有するタンパク質を見出した。PROSITE形式のモチーフでデータベースへの問合せを行った。Clustal Wを使用して対象の配列をアラインメントさせた。補足ファイル1に列挙した保存モチーフを、遺伝子オルソログの比較により同定した。Inparanoid5.1版用のデータファイルを、代表的な生物のサブセットに関してInparanoidウェブサーバー<http://inparanoid.sbc.su.se/>からダウンロードした。ヒトの配列を基準として使用し、Inparanoid処理したfastaファイルを、ローカルツールを使用して、APIMモチーフに関する正規表現でサーチした。Ile、Val及びLeuに加えてAlaがモチーフの3位又は4位のいずれかに存在してもよいが同時に両方の位置には存在しないとする、わずかに拡張したモチーフ定義を使用した。計22218種類のタンパク質配列から、APIMモチーフに対して少なくとも1つのヒットを有する636種類の配列が存在した。これらのエントリーを、ウェブサーバー<http://gpcr.biocomp.unibo.it/esldb/>からダウン
ロードしたeSLDBデータベースにおいて実験上の及び予測の細胞内局在化に対してマッチさせ、核に対する標的化の示徴(indication)を有しない349種類のエントリーは除いた。残りの287種類のエントリーに関して対応するInparanoidオルソログを同定し、対応する配列をfastaファイルから抽出し、得られた配列ライブラリをClustal Wを用いてアラインメントさせた。
。正規表現を、ヒット位置が確定する前にコンセンサスに対してさらに試験した。代替手順(個別)では、正規表現を、ヒト配列におけるヒット位置に対応する各オルソログ部分配列に対して試験し、少なくとも50%のオルソログがこの表現とマッチする位置のみを確定した。この評価では、ギャップのみから成る部分配列は、例えば代替的なスプライシング変異体を表し得ると仮定して、除外した。これらの2つの手順はほとんど同一の結果をもたらし、組み合わせたアウトプットを補足ファイル1に示す。計37種類のエントリーをこの手順により除き、残りの226種類のエントリーを列挙して解析した。アウトプ
ットにおいて使用したタンパク質の説明は、Inparanoid未処理ヒトfastaファイル及びEnsembl release 45から取得した。アウトプットファイルはhtml形式であり、標準的なウェブブラウザにより開くことができる。
予測PCNA結合ペプチドのドットブロット分析
28nmolのペプチド(スポットを可視化するためにポンソーで染色した)のドットを含有するアミノ−PEG500−UC540シート(安定性が向上した酸硬化性)を、オスロ大学(Norway)のバイオテクノロジーセンターのペプチド合成実験室で調製した。膜を1μg/ml PCNAで2時間探索し、その後一次抗体(α−PCNA、PC10)で探索し、上で説明したようにWBのために展開した。
細胞生存アッセイ
HeLa及びHaCaT細胞を96ウェルプレート中に播種し(4000細胞/ウェル)、4時間インキュベートした。種々の用量のMMS(メチルメタンスルホネート、Acros)、BCNU(1,3−ビス(2−クロロエチル)−1−ニトロソウレア(nitrosuream)、Sigma)、テモゾロミド(4−メチル−5−オキソ−2,3,4,6,8−ペンタア
ザビシクロ[4.3.0]ノナ−2,7,9−トリエン−9−カルボキサミド、TZM、Sigma)及びマイトマイシンC(6−アミノ−1,1a,2,8,8a,8b−ヘキサヒ
ドロ−8−(ヒドロキシメチル)−8a−メトキシ−5−メチル−アジリノ[2’,3’:3,4]ピロロ[1,2−a]インドール−4,7−ジオンカルバメート、MMC、Sigma)を、ウェルに添加した。U2OS細胞は、MMS及びTMZのみに曝露した。細胞
を回収まで連続して曝露した。MTTアッセイ(Mosmann, 1983)を使用して細胞を4日
間毎日回収した。ODを570nmで測定し、少なくとも6ウェルの平均を使用して細胞生存性を算出した。示したデータは、1つの代表的な実験で得られた成長であり、少なくとも2回再現された。
実施例1
この実施例で説明する本研究は、複製中心(foci)におけるhABH2の局在化を調べており、hABH2とPCNAとの間の直接の相互作用、及びかかる相互作用に関与するhABH2の領域を特定している。
実施例2
hABH2を阻害するhABH21−10の能力を試験した。
のG2/M停止、及び非常に低いレベルの修復中間体をもたらし、架橋を含有するほとんどの細胞が死滅したことを示している。他方、MMC処理は1日目及び2日目のS期停止をもたらし、これは対照細胞よりもhABH21−10−EYFPを発現する細胞でより顕著であった。細胞は3日目にも依然としてG2/Mで停止していた。MMC処理後のコメットアッセイにおいて細胞株間の有意差は観察することができなかったが、修復中間体の数は2日目に最大となった。
実施例3
複数の異なる種由来のデータベース配列ABH2のアライメントにより、7個のN末端アミノ酸が高度に保存されていることが明らかにされた(図4)。結合配列を同定するために、これらのアミノ酸のペプチド−PCNA相互作用に関する重要性をドットブロットアッセイを使用して調べた。とりわけ、Arg3及びLys7が互いに置換し得ること、並びにPCNAに対する見かけの親和性に影響を及ぼすことなく、Leu5及びVal6が互いに、又は他の脂肪族アミノ酸、例えばIle及びAlaにより置換され得ることが見出された。さらに、完全に保存されている芳香族アミノ酸Phe4を、Tyrにより置き換えることができたが、この位置のAlaはPCNA結合を有意に低減した。
実施例4
Swiss−Prot及びTrEMBLデータベースを使用してhABH2とPCNAとの結合に関与するものとして同定されている配列と類似の部分配列を有するタンパク質を見出した。クエリーとしてコンセンサス[KR]−[FYW]−[LIVA]−[LIVA]−[KR](配列番号30)を使用して、226個のヒットを得たが(概要としては第1表を参照されたい)、この内の幾つかのヒトタンパク質をさらなる解析のために選択した。
α)も調べた。Topo IIαは複製後のDNA脱連環及びDNA分離において機能する。
実施例4で研究したタンパク質におけるコンセンサス配列の機能を実験で調べた。コンセンサス配列における芳香族アミノ酸PheのAlaへの置換がin vitroでのPCNA相互作用及びin vivoでのPCNAとの共局在化を消失したため、対応する突然変異が全長タンパク質に対して同様の効果を有したかどうかを調べた。全長hABH2におけるPhe4のAlaへの突然変異がPCNAとの共局在化を消失した。4つのモチーフを含有するTFII−Iでは、Phe残基を個々に及び集合的にAlaに置換した(F431A、F536A、F641A及びF803A)。単一突然変異はPCNAとの共局在化を低減しなかったが、TFII−Iにおける全てのコンセンサス配列での突然変異は複製中心におけるPCNAとの共局在化を強く低減し、これにより同じタンパク質に存在する幾つかのコンセンサス配列モチーフが最適なPCNA相互作用に寄与し得ることが示された。

実施例6
シグナル配列と共にモチーフ(「APIM」)含有ペプチドを含有する構築物の調製及び試験
配列番号98〜配列番号117のペプチドを、標準的な技法を使用して合成し、示される場合は、同様に標準的な技法を使用して蛍光タグを組み込んだ。ペプチドを第3表に示す。この表は各ペプチドに対するアミノ酸配列を示し、また各ペプチド(必要に応じてモチーフ(「APIM」)含有ペプチド、NLS、CPP及びリンカー、並びに含有される場合、使用するタグ)を構成する個々の構成要素を別々に挙げる。配列番号98〜配列番号116のペプチドはL−アミノ酸で構成される。配列番号117のペプチドRI−MDR26−3はD−アミノ酸で構成されるレトロインベルソペプチドである。第3表に示される全てのペプチドは、APIMペプチド、NLS及びCPPを少なくとも1つ有する。
MTTアッセイ
HeLa細胞を96ウェルプレートに播種し(6000細胞/ウェル)、3時間インキュベートした。種々の用量のMMS及びシスプラチンをウェルに加えた。24時間後、ペプチドを血清無含有培地中の細胞に添加し、1時間インキュベートした。細胞増殖抑制薬を有する新たな培地を添加し、さらに24時間、48時間及び72時間後に細胞を採取した。MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド)を細胞に添加し、ODを570nmで測定し[52]、少なくとも6個のウェルの平均を使用して、細胞の生存度を算出した。データは1つの代表的な実験で得られた成長であり、少なくとも2回再現された(reproduced)。
コロニー形成(CFU)アッセイ
750個のHeLa細胞を、細胞増殖抑制薬(MMS)を含有する成長培地11mlの入った10cm径の細胞培養皿に播種した。2日目、細胞を血清無含有条件下で1時間ペプチドで処理した。新たな細胞増殖抑制薬を含有する新たな培地を10日間のさらなるインキュベートのために細胞に添加した。それから細胞を室温で15分間、6%グルタルアルデヒドのPBS溶液中で固定し、PBSで1回洗浄して、クリスタルバイオレットで染
色し、コロニー形成単位を計測した。少なくとも50個の細胞から成るコロニーだけが含まれていた。
細胞傷害性アッセイ
48時間後のペプチドの細胞傷害性を、MTTアッセイを使用して測定した。HeLa細胞を96ウェルプレートに播種し(6000細胞/ウェル)、3時間インキュベートした。種々の用量のペプチドを、血清の存在下又は非存在下でウェルの培地中に添加した。1時間後に1×又は2×の血清を有する等量の培地(血清無含有培地に対して)を添加し、MTTの添加前に細胞を48時間インキュベートした(上記を参照されたい)。
フローサイトメトリによる膜傷害性
細胞を種々の濃度(2μM、4μM及び8μM)のペプチドで1時間処理した。次に、ヨウ化プロピジウム(PI)(PBS中、50μg/ml)を添加し、これは細胞膜が透過性であればDNAを染色する。次の10分以内にFACS Cantoフローサイトメータ(BD-Life Science)で解析を行った。
さらなる細胞傷害性のデータ

であり、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
前記オリゴペプチド化合物が、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含み、かつ
前記オリゴペプチド化合物におけるPCNA相互作用モチーフの少なくとも1つが、前記シグナル配列よりもN末端側に位置する、
PCNA相互作用モチーフを含むオリゴペプチド化合物。 - 異常な又は不要な細胞成長を伴う障害若しくは病態の治療に又は細胞増殖抑制療法を伴う治療に使用される、PCNA相互作用モチーフを含むオリゴペプチド化合物であって、 前記PCNA相互作用モチーフが、
[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)であり、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
前記オリゴペプチド化合物が、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含み、かつ
前記オリゴペプチド化合物におけるPCNA相互作用モチーフの少なくとも1つが、前記シグナル配列よりもN末端側に位置する、
PCNA相互作用モチーフを含むオリゴペプチド化合物。 - (i)PCNA相互作用モチーフを含むオリゴペプチド化合物であって、
前記PCNA相互作用モチーフが、
[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)であり、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
前記オリゴペプチド化合物が、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含む、PCNA相互作用モチーフを含むオリゴペプチド化合物、及び
(ii)細胞増殖抑制剤、
を備える、キット。 - 前記(i)オリゴペプチド化合物、及び(ii)細胞増殖抑制剤が、異常な又は不要な細胞成長を伴う障害若しくは病態の治療において又は細胞増殖抑制療法を伴う治療において同時に、連続して又は別々に使用される複合調製物として提供される、請求項3に記載のキット。
- 異常な又は不要な細胞成長を伴う障害若しくは病態の治療において又は細胞増殖抑制療法を伴う治療において細胞増殖抑制剤と併用される、PCNA相互作用モチーフを含むオリゴペプチド化合物であって、
前記PCNA相互作用モチーフが、
[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)であり、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
前記オリゴペプチド化合物が、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含む、PCNA相互作用モチーフを含むオリゴペプチド化合物。 - 放射線療法のための増感剤として使用される、PCNA相互作用モチーフを含むオリゴペプチド化合物であって、
前記PCNA相互作用モチーフが、
[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)であり、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
前記オリゴペプチド化合物が、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含む、PCNA相互作用モチーフを含むオリゴペプチド化合物。 - 放射線療法のための増感剤として、異常な又は不要な細胞成長を伴う障害若しくは病態の治療、又は放射線療法を伴う治療に使用される、請求項6に記載のオリゴペプチド化合物。
- 新規な細胞増殖抑制剤をin vitroで同定する方法であって、請求項1に記載のオリゴペプチド化合物を細胞又は細胞培養物に投与することを含む、in vitroでの同定方法。
- 請求項1に記載のオリゴペプチド化合物を含有するリポソーム。
- PCNAと相互作用可能なオリゴペプチド化合物であって、
前記オリゴペプチド化合物が、
[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)を含み、かつ
修復タンパク質とPCNAとの相互作用を阻害し、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含み、
前記オリゴペプチド化合物におけるPCNA相互作用モチーフの少なくとも1つが、前記シグナル配列よりもN末端側に位置する、
オリゴペプチド化合物。 - 配列[K/R]−[F/Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号30)を有し、かつ 修復タンパク質とPCNAとの相互作用を阻害するPCNA相互作用モチーフを有する又はこれを含むペプチドをコードする核酸分子であって、
前記ペプチドが、
18〜70個のアミノ酸を含み、かつ
核局在化シグナル配列及び細胞透過シグナル配列を含み、
前記オリゴペプチド化合物におけるPCNA相互作用モチーフの少なくとも1つが、前記シグナル配列よりもN末端側に位置する、
核酸分子。 - 請求項11に記載の核酸分子を含むベクター。
- 前記PCNA相互作用モチーフが、
[K/R]−[Y/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号31)、
[K/R]−[F/W]−[L/I/V/A]−[L/I/V/A]−[K/R](配列番号33)、
[K/R]−[F/W]−[L/I/V]−[L/I/V]−[K/R](配列番号34)、
[K/R]−[F/Y/W]−[L/I/V]−[L/I/V]−[K/R](配列番号35)、
[K/R]−[Y/W]−[L/I/V]−[L/I/V]−[K/R](配列番号36)、又は
[K/R]−F−[L/I/V]−[L/I/V]−[K/R](配列番号37)である、請求項1、2、5〜7、及び10のいずれか一項に記載のオリゴペプチド化合物。 - 前記PCNA相互作用モチーフが、配列番号2、配列番号3、配列番号4、配列番号5、配列番号6、配列番号7、配列番号8、配列番号9、配列番号10、配列番号11、配列番号12、配列番号13、配列番号14、配列番号16、配列番号17、配列番号18、配列番号19、配列番号20、配列番号21、配列番号22、配列番号23、配列番号24、又は配列番号25に記載の配列である、請求項1、2、5〜7、10及び13のいずれか一項に記載のオリゴペプチド化合物。
- 前記細胞透過シグナル配列がHIV−TAT若しくはPenetratinである、請求項1、2、5〜7、10、13及び14のいずれか一項に記載のオリゴペプチド化合物。
- 前記オリゴペプチド化合物が、細胞増殖抑制剤と共に、同時に、別々に、又は連続して使用される、請求項1、2、5〜7、10、及び13〜15のいずれか一項に記載のオリゴペプチド化合物。
- 前記オリゴペプチド化合物が融合タンパク質又はアプタマーの一部である、請求項1、2、5〜7、10、及び13〜15のいずれか一項に記載のオリゴペプチド化合物。
- 前記細胞増殖抑制剤が、アクチノマイシンD、BCNU(カルマスティン)、カルボプラチン、CCNU、カンプトテシン(CPT)、カンタリジン、シスプラチン、シクロホスファミド、シタラビン、ダカルバジン、ダウノルビシン、ドセタキセル、ドキソルビシン、DTIC、エピルビシン、エトポシド、ゲフィニチブ、ゲムシタビン、イフォサミド、イリノテカン、イオノマイシン、メルファラン、メトトレキサート、メチルメタンスルホネート(MMS)、マイトマイシンC(MMC)、ミトザントロン メルカプトプリン、オキサリプラチン、パクリタキセル(タキソール)、PARP−1阻害剤、タキソテール、テモゾロミド(TZM)、テニポシド、トポテカン、トレオスルファン、ビノレルビン、ビンクリスチン、ビンブラスチン、5−アザシチジン、5,6−ジヒドロ−5−アザシチジン及び5−フルオロウラシルから選択される、請求項5又は16に記載のオリゴペプチド化合物。
- 前記細胞増殖抑制剤がアルキル化剤である、請求項5又は16に記載のオリゴペプチド化合物。
- 前記障害が過剰増殖性障害であり、又は前記治療が骨髄除去である、請求項2、5、及び7のいずれか一項に記載のオリゴペプチド化合物。
- 前記過剰増殖性障害が、悪性、前癌性又は非悪性の腫瘍性障害、炎症、自己免疫障害、血液障害、皮膚障害、ウイルス誘導性の過剰増殖性障害、骨髄異形成(myelodyplastic)障害又は骨髄増殖性障害から選択される、請求項20に記載のオリゴペプチド化合物。
- 前記過剰増殖性障害が、癌、良性腫瘍、乾癬性関節炎、関節リウマチ、炎症性腸疾患、乾癬、ライター症候群、毛孔性紅色粃糠疹、過剰増殖性変異型の角質化障害、再狭窄、糖尿病性腎症、甲状腺過形成、グレイヴズ病、前立腺肥大症、Li−Fraumenti症候群、糖尿病性網膜症、末梢血管疾患、子宮頸部上皮内癌、家族性腸ポリープ症、口腔白板症、組織球増殖症、ケロイド、血管腫、過剰増殖性動脈狭窄、炎症性関節炎、過角化症、関節炎を含む丘疹鱗屑性発疹、疣贅、EBV誘導性疾患、瘢痕形成、多発性硬化症、全身性エリテマトーデス(SLE、ループス)、重症筋無力症、非悪性過形成、肉芽腫、意義不明の単クローン性高ガンマグロブリン血症(MGUS)、新生物性髄膜炎、真性赤血球増加症、硬化性粘液水腫、丘疹性ムチン沈着症、アミロイドーシス及びヴェグナー肉芽腫から選択される、請求項21に記載のオリゴペプチド化合物。
- 炎症の治療において用いる、請求項1、5〜7、10、及び13〜21のいずれか一項に記載のオリゴペプチド化合物。
- 癌の治療において用いる、請求項1、5〜7、10、及び13〜21のいずれか一項に記載のオリゴペプチド化合物。
- 前記癌が、膀胱癌、前立腺癌、及び血液学的癌(例えば、多発性骨髄腫又は白血病)から選択される少なくとも1種である、請求項24に記載のオリゴペプチド化合物。
