Description
−神経系:皮脂機能を悪化させる、情緒的ストレス、神経緊張;
−疲労;
−バランスの悪い食生活;
−一部の薬剤(向精神薬);
−皮膚および/または頭皮を「剥離」し、反応性脂漏症を引き起こす不適合化粧品;
−主要な原因はホルモン型であり、5−αレダクターゼ酵素の過剰な活性化が脂漏症を引き起こす。
−成熟および脂質滴の段階的取り込みの間の皮脂腺細胞による皮脂の生産;
−分化プログラム(15日)後の皮脂腺細胞の溶解およびそれらの内容物である皮脂の毛嚢皮脂溝(漏斗)への放出;
−漏斗内の皮脂が上昇し、漏斗を満たし、角質層の表面に拡がる。
−X=NHである場合、R1、R2、R3、R4はそれぞれ水素原子を表し、
−X=Nである場合、コアは芳香族であり、かつ、R1、R2、R3およびR4は水素原子を表すか、またはその1つはメチル基を表し、かつ、R2=R3=R4=Hである場合、R1はまた、ハロゲン原子またはアリール、ヘテロアリール、アルケニル、アセチレニル基を表してもよい)。

本発明のニコチン誘導体の合成を可能とする一般操作様式を示す。
Tel−E6E7株(Stephen Lyle博士、マサチューセッツ大学、USAの厚意による)は、毛包球の幹細胞に由来するヒト上皮細胞株である。HPV 16ウイルスのE6/E7遺伝子の発現により不死化されたこの株は、特殊な培養条件下で皮脂腺細胞に分化することができる多能性上皮細胞株である(Multi-potentiality of a new immortalized epithelial stem cell line derived from human hair follicles, Roh C., Roche M., Guo Z., Photopoulos C., Tao Q., Lyle S., In Vitro Cell Dev. Biol. Anim., July-August 2008, 44(7): 236-44)。
・一般操作様式:
ビスエステルA/トリエステルB/ラウリン酸グリセリルの反応混合物をエチルエーテル(10V)で可溶化した。
100mlの3つ口フラスコにて、窒素流中、磁気撹拌下、ニペコチン酸(3.7g)を3N水酸化ナトリウム溶液(30ml)中の溶液とした。この溶液を氷浴で0℃に冷却した。あるいは、クロロギ酸ベンジル(5.8ml)を0℃で45分かけて3N水酸化ナトリウム溶液(9.3ml)とともに少量ずつ加えた。添加の終了後、この反応媒体は淡黄色で温度は3℃であった。氷浴を外し、反応媒体を撹拌下、周囲温度で一晩放置した。次に、水相をエチルエーテルで1回抽出した後、3N塩酸溶液を用いてpH1に酸性化した。酸性化された水相をエチルエーテルで3回抽出した。エーテル抽出相を1N塩酸溶液、次いで、飽和塩化ナトリウム溶液で2回順次洗浄した後、硫酸マグネシウムで乾燥させ、濾過し、その後、真空濃縮した。
1Lの3つ口フラスコにて、窒素流下、磁気撹拌下、工程1で得られた酸(3.5g)をジクロロメタン(440ml)中の溶液とした。これにEDCI.HCl(2.81g)およびDMAP(0.81g)を加えた。この無色の溶液を撹拌下、周囲温度で15分間放置した後、ラウリン酸グリセリル(10.95g)を加えた。この反応媒体を撹拌下、周囲温度で一晩放置した後、水(200ml)で加水分解した。有機相を分離し、飽和炭酸水素ナトリウム溶液で2回洗浄した後、硫酸マグネシウムで乾燥させ、濾過し、かつ最後に真空濃縮し、白蝋状固体(15.5g)を得た。
磁気撹拌を備え、三方コックを取り付けた(1つの分岐は窒素バルーンにつなぎ、2つ目の分岐は水素バルーンにつなぎ、3つ目の分岐は真空システムにつないだ)100mlのフラスコにて、工程2で得られたジエステル(2.64g)を酢酸エチル(40ml)中の溶液とした。窒素系で2回パージした後、この黄色溶液にパラジウムカーボン(0.54g)を加えた。この反応媒体を窒素/真空を交互させて3回、その後水素/真空を交互させて3回パージした後、この反応媒体を水素雰囲気下に置いた。1時間20分の反応時間の後、反応媒体をセライトで濾過した。濾液を真空濃縮し、淡黄色油状物を得た(1.88g、全収率=17%)。
Claims (11)
- 下記一般式Iの化合物:
−X=NHである場合、R1、R2、R3、R4はそれぞれ水素原子を表し、
−X=Nである場合、コアは芳香族であり、かつ、
R2、R3およびR4は水素原子を表し、かつ、R1=H、ハロゲン、アリール、ヘテロアリール、アルケニルまたはアセチレニルであるか、
あるいはR1、R2、R3、R4は、R1またはR2またはR3またはR4がメチル基を表すように定義され、かつ、他の3つの基が水素原子を表す)。 - 前記化合物が、ニコチン酸3−(ドデカノイルオキシ)−2−ヒドロキシプロピルである、請求項1に記載の化合物。
- 請求項1および2のいずれか一項に記載の式Iの化合物を含んでなる薬剤。
- 座瘡、脂漏性皮膚炎または男性型脱毛症の治療のための、請求項1および2のいずれか一項に記載の式Iの化合物を含んでなる薬剤。
- 請求項1および2のいずれか一項に記載の式Iの化合物を含んでなる化粧品。
- 脂漏症(脂漏性病態)の治療のための、請求項1および2のいずれか一項に記載の式Iの化合物を含んでなる化粧品。
- 請求項1および2のいずれか一項に記載の式Iの化合物からなる、皮脂腺細胞による脂質生産の阻害剤。
- 請求項1および2のいずれか一項に記載の式Iの化合物を、少なくとも1種類の薬学的または化粧学的に許容可能な賦形剤とともに含んでなる、組成物。
- 薬剤または化粧品としての使用のための、請求項8に記載の組成物。
- 座瘡、脂漏性皮膚炎または男性型脱毛症の治療のための、請求項8および9のいずれか一項に記載の組成物。
- 脂漏症(脂漏性病態)の治療のための、請求項8および9のいずれか一項に記載の組成物。
