Description

The present invention relates to a biological sample transfer device for transferring a biological sample such as a cell such as stem cells or umbilical cord blood or serum to a biological sample storage container, and a biological sample transfer method using the biological sample transfer device.

Conventionally, a biological sample such as a stem cell or umbilical cord blood or a serum prepared from blood is collected from the living body until it is used, and the biological sample storage container has a biological sample storage chamber formed by three-dimensional molding. (See, for example, Patent Document 1). More specifically, a biological sample collected from a living body is first stored in a biological sample storage container such as a blood bag. Next, the biological sample stored in the biological sample storage container is transferred to a biological sample storage container connected to the biological sample storage container via a connection tube.

When transferring a biological sample from a biological sample storage container to a biological sample storage container, the biological sample storage container is positioned above the biological sample storage container so as to drop a gap between the biological sample storage container and the biological sample storage container. Pressure is generated, and the biological sample is transferred to the biological sample storage container by this drop pressure.
However, since air exists in the biological sample storage chamber of the biological sample storage container, when the biological sample is transferred to the biological sample storage container, the work of replacing the air present in the biological sample storage chamber with the biological sample is performed. Necessary.

Here, when storing a biological sample in a biological sample storage container, in order to maintain the aseptic state of the biological sample, it is necessary that external air enters the internal space of the biological sample storage container, the connection tube, and the biological sample storage container. It is required to prevent. Therefore, when transferring a biological sample to the biological sample storage container, the biological sample storage container is crushed to move the air present in the biological sample storage chamber to the biological sample storage container side through the connection tube, and then crushed. The operation of transferring the biological sample to the biological sample storage chamber is repeated a plurality of times.

As described above, when the operation of replacing the biological sample and air is repeated a plurality of times, the biological sample and air are mixed to cause foaming, which adversely affects the biological sample. In addition, foaming of the biological sample causes a part of the biological sample to remain in the biological sample storage container, and the biological sample cannot be sufficiently transferred to the biological sample storage container.

Therefore, an object of the present invention is to provide a biological sample transfer device and a biological sample transfer method that hardly cause adverse effects on a biological sample when the biological sample is transferred from the biological sample storage container to the biological sample storage container.

FIG. 2 is a perspective view of the biological sample storage container 3, and FIG. 3 is a plan view. 2 and 3, the cover portion 314 is omitted.
As illustrated in FIGS. 1 to 3, the biological sample storage container 3 includes a storage container main body 31 and a cylindrical member 32.
The storage container body 31 includes most of the peripheral portions of two sheet-like members made of a thermoplastic resin having flexibility such as EVA resin (ethylene-vinyl acetate copolymer resin) and a predetermined portion (described later). The boundary portion between the first biological sample storage chamber and the second biological sample storage chamber) is joined. In the storage container main body 31, a biological sample storage chamber 311, a biological sample introduction port 312, a biological sample outlet 313, and a cover part 314 are formed.

The biological sample storage chamber 311 is formed by three-dimensionally molding a sheet-like member. More specifically, first, one of the two sheet-like members is subjected to vacuum forming or the like to form a three-dimensional shape. Next, the other planar sheet-like member is joined to one sheet-like member formed with this three-dimensional shape. Thereby, the biological sample storage chamber 311 having a three-dimensional shape is formed.
In the first embodiment, the biological sample storage chamber 311 includes a first biological sample storage chamber 311a that can store a relatively large amount (for example, 10 ml to 30 ml) of a biological sample, as shown in FIGS. The second biological sample storage chamber 311b that can store a smaller amount (for example, 3 ml to 10 ml) of the biological sample than the first biological sample storage chamber 311a, and the first biological sample storage chamber 311a and the second biological sample storage A communication portion 311c for communicating with the chamber 311b.

In the first embodiment, the first biological sample storage chamber 311a and the second biological sample storage chamber 311b are arranged adjacent to each other in the width direction WD of the storage container main body 31, as shown in FIGS. Moreover, the communication part 311c is provided in two places, the upper end side which is the one end side of the height direction HD of the storage container main body 31, and the lower end side which is the other end side.

The biological sample inlet 312 serves as an inlet for introducing the biological sample into the biological sample storage chamber 311. The biological sample introduction port 312 is configured by non-joining part of the peripheral portions of the two sheet-like members constituting the storage container main body 31. In the first embodiment, the biological sample inlet 312 is provided at the upper end of one end of the storage container body 31 in the width direction WD as shown in FIGS. 1 and 2. Further, the biological sample introduction port 312 is provided in the first biological sample storage chamber 311a.

The biological sample outlet 313 serves as an outlet for leading out the biological sample stored in the biological sample storage chamber 311. Similar to the biological sample introduction port 312, the biological sample outlet 313 is configured by non-joining part of the peripheral portions of the two sheet-like members that constitute the storage container body 31. In the first embodiment, the biological sample outlet 313 is provided at the upper end of each of the first biological sample storage chamber 311a and the second biological sample storage chamber 312b.
The cover part 314 covers the biological sample outlet 313. The cover part 314 is removed when the biological sample is taken out from the biological sample outlet 313.

The cylindrical member 32 is disposed in the biological sample outlet 313 of the biological sample storage container 3. The tubular member 32 includes a tubular body 321, a diaphragm part 322, and a pair of guard parts 323.

The cylinder main body 321 is configured in a cylindrical shape.
The diaphragm part 322 is disposed inside the cylinder body 321 and closes the cylinder body 321. The diaphragm part 322 is formed in a thin disk shape and has a strength enough to be perforated by the action of an external force. The guard part 323 is formed so as to extend in the axial direction of the cylinder main body 321 from the end of the cylinder main body 321 on the biological sample storage chamber 311 side. The guard portion 323 is formed continuously with the cylinder main body 321.
The above cylindrical member 32 is formed of a thermoplastic resin such as EVA resin.

In the biological sample storage container 3 described above, a liquid passing portion capable of flowing liquid and gas is formed only at the biological sample introduction port 312, and the biological sample outlet 313 is formed by the cylindrical member 32 (diaphragm portion 322). It is closed. In addition, the biological sample storage chamber 311 is formed by three-dimensional molding, and maintains a three-dimensional shape when no external force is applied.

The connection tube 4 is made of a thermoplastic resin such as EVA resin, and connects the biological sample storage container 2 and the biological sample storage container 3 in an aseptic and airtight manner. The connection tube 4 includes an upstream tube 41 and a downstream tube 42. One end of the upstream tube 41 is connected to the biological sample outlet 23 of the biological sample storage container 2, and the other end is connected to a port unit 5 described later.
The downstream tube 42 has one end connected to the port unit 5 and the other end connected to the biological sample inlet 312 of the biological sample storage container 3.

The port unit 5 is configured to be able to lead the gas inside the connection tube 4 to the outside while maintaining the airtight state of the biological sample storage container 2, the biological sample storage container 3, and the connection tube 4.
In the first embodiment, as shown in FIGS. 1 and 5, the port portion 5 is provided between the upstream tube 41 and the downstream tube 42, and connects the upstream tube 41 and the downstream tube 42. To do. The port portion 5 includes a septum 51 and a first connection port 52 and a second connection port 53 formed in the septum 51.

As shown in FIGS. 5 and 6, the septum 51 includes a main body portion 511 having a circular opening, an elastic member 512 formed in a film shape or a plate shape disposed in the opening of the main body portion 511, and A slit portion 513 formed in the elastic member 512.
The other end of the upstream tube 41 is connected to the first connection port 52. One end of the downstream tube 42 is connected to the second connection port 53.

According to the port portion 5 described above, when the distal end portion of a suction tool such as a syringe is inserted into the slit portion 513 of the septum 51, the elastic member 512 is elastically deformed to open the slit portion 513. Thereby, the gas inside the connection tube 4 can be sucked by the suction tool and led out to the outside. Further, when the distal end portion of the suction tool is extracted from the slit portion 513, the slit portion 513 is closed by the elastic recovery force of the elastic member 512.

The upstream closing portion 6 is provided closer to the biological sample storage container 2 than the port portion 5. In the first embodiment, as shown in FIG. 1, the upstream closing portion 6 is configured by a synthetic resin clamp and is attached to the upstream tube 41.
According to the upstream closing portion 6 described above, the flow of the fluid (liquid and gas) in the upstream tube 41 can be blocked by sandwiching the upstream tube 41 with the clamp. Further, the fluid flow in the upstream tube 41 can be allowed by releasing the clamping of the upstream tube 41 by the clamp.
The phrase “provided on the biological sample storage container 2 side with respect to the port unit 5” is not limited to the case of being provided between the port unit 5 and the biological sample storage container 2 as in the present embodiment. The case where it is provided in the biological sample introduction part 22 in the container 2 is included.

The downstream closing part 7 is provided closer to the biological sample storage container 3 than the port part 5. In the first embodiment, the downstream closing portion 7 is configured by a synthetic resin clamp and is attached to the downstream tube 42 as shown in FIG. 1.
According to the upstream closing portion 6 described above, the flow of the fluid (liquid and gas) in the downstream tube 42 can be blocked by sandwiching the downstream tube 42 with the clamp. Further, by releasing the clamping of the downstream side tube 42 by the clamp, the fluid flow in the downstream side tube 42 can be allowed.

Next, an embodiment of a biological sample transfer method using the biological sample transfer apparatus 1 of the first embodiment will be described with reference to FIGS.
The biological sample transfer method of this embodiment includes a first closing step S1, a suction tool connecting step S2, a suction step S3, a second closing step S4, a first opening step S5, a gas supply step S6, and a suction. A tool removal step S7, a biological sample storage step S8, and a biological sample transfer step S9 are provided.

In the first closing step S1, the upstream side closing portion 6 is closed as shown in FIG. Thereby, the flow of fluid in the upstream tube 41 is blocked. In the first closing step S <b> 1, the downstream side closing portion 7 is opened, and fluid flow in the downstream side tube 42 is allowed. In the first closing step S1, the clamp 224 of the biological sample introduction unit 22 is also closed.

In the suction tool connection step S <b> 2, as shown in FIG. 7, a syringe 100 as a suction tool is connected to the port portion 5. More specifically, in the suction tool connection step S <b> 2, the distal end portion of the syringe 100 with the piston 120 pushed into the cylinder portion 110 is pushed into the slit portion 513 of the septum 51, and the syringe 100 is connected to the port portion 5. .

In the suction step S3, the air inside the biological sample storage container 3 is sucked by the syringe 100 as shown in FIG. More specifically, in the suction step S3, the piston 120 that has been pushed into the cylinder part 110 is pulled out. Then, since the upstream side tube 41 is closed by the upstream side closing part 6, the air existing inside the downstream side tube 42 and the biological sample storage chamber 311 of the biological sample storage container 3 are inside the cylinder part 110. The air present in the air is aspirated. As a result, as shown in FIG. 8, the biological sample storage chamber 311 having a three-dimensional shape is deformed so as to be crushed, and negative pressure is generated inside the biological sample storage container 3 and inside the connection tube 4.

In the second closing step S4, as shown in FIG. 9, the downstream side closing portion 7 is closed. In the first opening step S5, the upstream closing portion 6 is opened. Thereby, the flow of fluid in the downstream tube 42 is blocked, and the flow of fluid in the upstream tube 41 is allowed.

In the gas supply step S6, as shown in FIG. 9, the air sucked into the syringe 100 is supplied to the biological sample storage container 2 side. More specifically, in the gas supply step S6, the air that has been sucked into the cylinder portion 110 is supplied to the biological sample storage container 2 side by pushing in the piston 120 that has been pulled out in the suction step S3.

In the suction tool removal step S <b> 7, the syringe 100 is removed from the port unit 5. Thereby, the port part 5 is sealed again.

In the biological sample storage step S <b> 8, as shown in FIG. 10, the clamp 224 is opened, and the biological sample is stored in the biological sample storage container 2. Here, air is supplied to the biological sample storage container 2 in the gas supply step S6. Accordingly, when the biological sample storage container 2 is configured by a container that is not three-dimensionally shaped, such as a blood bag, the biological sample storage container 2 is supplied with air that is maintained in a sterile state. The storage container 2 can be slightly inflated. Therefore, when an additive such as a frost damage protective agent is added to the biological sample stored in the biological sample storage container 2, the biological sample and the additive can be suitably mixed. In this case, since the biological sample is introduced into the biological sample storage container 2 after air is introduced, the biological sample stored in the biological sample storage container 2 does not foam. Further, even when air is introduced after the biological sample is stored in the biological sample storage container 2, the air is supplied from the biological sample lead-out unit 23 provided at the upper part of the biological sample storage container 2 to the biological sample storage unit 21. Therefore, the biological sample does not foam due to the introduction of air into the biological sample reservoir 21.
In this biological sample storing step S8, as shown in FIG. 10, the upstream closing portion 6 opened in the first opening step S5 may be closed again.

In the biological sample transfer step S <b> 9, as shown in FIGS. 11 and 12, the cells stored in the biological sample storage container 2 with the upstream closing portion 6 opened are introduced into the biological sample storage container 3. More specifically, in the biological sample transfer step S9, first, as shown in FIG. 11, the biological sample storage container 2 is moved from the biological sample storage container 3 with the biological sample outlet 23 facing downward. Is also positioned above. Next, as shown in FIG. 12, the downstream closing portion 7 is opened. When the upstream closing unit 6 is closed in the biological sample storage step S8, the upstream closing unit 6 is also opened in the biological sample transfer step S9. Then, the biological sample stored in the biological sample storage container 2 is transferred to the biological sample storage container 3 through the connection tube 4 due to the drop pressure between the biological sample storage container 2 and the biological sample storage container 3.

Here, in this embodiment, the air inside the biological sample storage chamber 311 is sucked in the suction step S3. Accordingly, the biological sample can be transferred from the biological sample storage container 2 to the biological sample storage container 3 without performing an operation of replacing the air present in the biological sample storage chamber 311 with the biological sample. Therefore, when the biological sample is transferred from the biological sample storage container 2 to the biological sample storage container 3, it is possible to prevent foaming from occurring in the biological sample, and thus it is difficult to adversely affect the biological sample. In addition, since bubbling does not occur in the biological sample, it can be transferred to the biological sample storage container without leaving the biological sample in the biological sample storage container.
Further, since a negative pressure is generated in the internal space of the biological sample transfer device 1, the biological sample can be more efficiently transferred from the biological sample storage container 2 to the biological sample storage container 3.

In the above-described embodiment, the biological sample transfer method includes the gas supply step S6. However, the biological sample transfer method may be performed without providing the gas supply step S6. In this case, the suction tool removal step S7, the biological sample storage step S8, and the biological sample transfer step S9 may be performed after the suction step S3.

According to the biological sample transfer device 1 of the first embodiment and the biological sample transfer method using the biological sample transfer device 1 described above, the following effects are obtained.

(1) The biological sample transfer device 1 is provided in the connection tube 4 that connects the biological sample storage container 2 and the biological sample storage container 3, and the gas inside the connection tube 4 can be led out while maintaining an airtight state. The port portion 5 and the upstream closing portion 6 provided on the biological sample storage container 2 side with respect to the port portion 5 are configured. As a result, the air present in the biological sample storage chamber 311 of the biological sample storage container 3 can be aspirated from the port portion 5 with the upstream side closing portion 6 closed, so that the air present in the biological sample storage chamber 311 and the biological sample The biological sample can be introduced from the biological sample storage container 2 to the biological sample storage container 3 without performing the operation of replacing the. Therefore, when the biological sample is introduced from the biological sample storage container 2 to the biological sample storage container 3, it is possible to prevent the biological sample from being bubbled, so that the biological sample is hardly adversely affected.

(2) The biological sample transfer apparatus 1 is configured to include the downstream closing portion 7 provided on the biological sample storage container 3 side with respect to the port portion 5. Thus, after the air inside the biological sample storage chamber 311 is aspirated in the aspirating step S3, the downstream closing portion 7 is closed and the upstream closing portion 6 is opened, and then the aspirated air in the gas supply step S6. Can be supplied to the biological sample storage container 2 side.

(3) The port portion 5 is configured to include a septum 51 having an elastic member 512 and a slit portion 513 formed in the elastic member 512. Thereby, since the slit part 513 is open | released by inserting the front-end | tip part of the syringe 100 in the septum 51, the gas inside the connection tube 4 can be attracted | sucked with the syringe 100 and it can guide | induced to the exterior. Moreover, the slit part 513 can be closed and sealed by extracting the tip part of the syringe 100 from the slit part 513. Therefore, it is possible to realize the port portion 5 that can lead out the gas inside the connection tube 4 to the outside while maintaining the airtight state of the biological sample storage container 2, the biological sample storage container 3, and the connection tube 4.

Next, a biological sample transfer apparatus 1A according to a second embodiment of the present invention will be described with reference to FIGS. In the description after the second embodiment, the same constituent elements are denoted by the same reference numerals, and the description thereof is omitted or simplified.

The biological sample transfer apparatus 1A according to the second embodiment is different from the biological sample transfer apparatus 1 according to the first embodiment in that the upstream side closing part and the downstream side closing part are constituted by three-way stopcocks 8.
The three-way stopcock 8 switches the first connection port 81, the second connection port 82, the third connection port 83, and two of these three connection ports to communicate with each other and closes one connection port. Member 84.

In the second embodiment, the first connection port 81 is connected to the upstream tube 41, and the second connection port 82 is connected to the downstream tube 42. And the 3rd connection port 83 is connected to the septum 51A as a port part.
The switching member 84 communicates the second connection port 82 (biological sample storage container 3) and the third connection port 83 (septum 51A) and closes the first connection port 81 (see FIG. 14), first. A second position (see FIG. 15) where the connection port 81 (biological sample storage container 2) and the second connection port 82 (biological sample storage container 3) communicate with each other and the third connection port 83 is closed, and the third connection port 83. A third position (not shown) in which the (septum 51A) and the first connection port 81 (biological sample storage container 2) communicate with each other and the second connection port 82 is closed can be switched.

Transfer of the biological sample from the biological sample storage container 2 to the biological sample storage container 3 using the biological sample transfer apparatus 1A of the second embodiment is performed according to the following procedure.
First, as shown in FIG. 14, the switching member 84 is positioned at the second position to perform the first closing step S1, and then the syringe 100 is inserted into the septum 51A to store the biological sample in the biological sample storage container 3. Air present inside the chamber 311 is sucked (suction tool connection step S2 and suction step S3). Next, the syringe 100 is removed from the septum 51A (suction tool removing step S7). Next, the biological sample is stored in the biological sample storage container 2 (biological sample storage step S8). Next, in a state in which the biological sample storage container 2 is positioned above the biological sample storage container 3, the switching member 84 is positioned at the third position, and the biological sample stored in the biological sample storage container 2 is stored in the biological sample storage container. 3 (biological sample transfer step S9).

In addition, when the three-way cock 8 of the second embodiment is used, when the switching member 84 is switched from the first position to the third position, it is necessary to temporarily go through the second position. Therefore, in the biological sample transfer method using the biological sample transfer device 1A of the second embodiment, when the gas supply step S6 is performed, the upstream tube 41 may be separately provided with an upstream closing portion such as a clamp. preferable. Thus, after the suction step S3, the upstream tube 41 is closed by a clamp or the like, and the second closing step S4 is performed. In this state, the switching member 84 is moved from the first position to the third position via the second position. The first opening step S5 and the gas supply step S6 can be performed by opening the clamp after switching to.

According to the biological sample transfer apparatus 1A of the second embodiment, in addition to the effects (1) to (3) described above, the following effects can be obtained.

(4) The upstream closing portion and the downstream closing portion are configured by the three-way cock 8 including the switching member 84. Thereby, since the switching of the flow path can be easily performed by switching the switching member 84, the biological sample transfer method of the present invention can be performed more easily.

Next, a biological sample transfer device 1B according to a third embodiment of the present invention will be described with reference to FIG.

The upstream side tube 41 </ b> B is disposed on the biological sample storage container 2 side, and the proximal end portion is connected to the biological sample outlet 23 of the biological sample storage container 2.
The downstream tube 42 </ b> B is disposed on the biological sample storage container 3 side, and the tip is connected to the biological sample inlet 312 of the biological sample storage container 3.
The branch tube 43B has a proximal end connected to a connection portion between the distal end of the upstream tube 41B and the proximal end of the downstream tube 42B. And the port part 5B is attached to the front-end | tip part of this branch tube 43B.
In the third embodiment, the upstream tube 41B, the downstream tube 42B, and the branch tube 43B are connected by a connecting member 44B having three connection ports.

According to the biological sample transfer apparatus 1B of the third embodiment, in addition to the effects (1) to (3) described above, the following effects can be obtained.

(5) The connection tube 4B includes an upstream tube 41B, a downstream tube 42B, and a branch tube 43B. Thereby, since the port part 5B is attached to the front-end | tip part of the branch tube 43B, the operativity of the biological sample transfer apparatus 1B can be improved.

Next, a biological sample transfer apparatus 1C according to a fourth embodiment of the present invention will be described with reference to FIGS.
A biological sample transfer apparatus 1C of the fourth embodiment differs from that of the third embodiment in the configuration of the port portion. In the fourth embodiment, the port portion is configured by a one-way valve 5 </ b> C that allows gas to flow from the inside to the outside of the connection tube 4 and blocks gas from the outside to the inside.

On the other hand, as shown in FIG. 18, the valve 5C includes a first case 54C connected to the distal end portion of the branch tube 43C, a second case 55C connected to the first case 54C, and the first case 54C and the second case. And a valve body 56C disposed inside 55C.

54 C of 1st cases are provided with the tubular insertion part 541 by which one end side is inserted in the front-end | tip part of the branch tube 43C, and the 1st fitting part 542 provided in the other end side of this insertion part 541. The first fitting portion 542 is formed in a cylindrical shape having a larger diameter than the insertion portion 541.
The second case 55C is provided on the other end side of the cylindrical second fitting portion 551 with one end side fitted inside the first fitting portion 542 and the second fitting portion 551, such as a syringe. A suction tool connecting portion 552 to which the suction tool is connected.

The valve body 56C is made of an elastic member such as rubber, and is disposed on the inner surface side of the second fitting portion 551 of the second case 55C. One end side of the valve body 56C is bonded to the inner peripheral surface of the second fitting portion 551, and the other end side protrudes toward the suction tool connecting portion 552. A slit (not shown) is formed at the protruding end of the valve body 56C.

According to the one-way valve 5C described above, when air is sucked from the suction tool connecting portion 552, the slit of the valve body 56C is opened by the suction force acting on the suction tool side inside the suction tool connecting portion 552. Thereby, air distribute | circulates from the connection tube 4C toward the exterior.
On the other hand, when the suction of air from the suction tool connection portion 552 side is stopped, the slit of the valve body 56C is closed, so that no air flows from the suction tool connection portion 552 side to the connection tube 4C side.

According to the biological sample transfer apparatus 1C of the fourth embodiment, in addition to the effects (1), (2), and (5) described above, the following effects can be obtained.

(6) The port portion is constituted by the one-way valve 5C. Accordingly, when the air present inside the biological sample storage chamber 311 of the biological sample storage container 3 is sucked by the suction tool, it is possible to prevent external air from entering the biological sample transfer apparatus 1C. The aseptic condition inside the transfer device 1C can be suitably maintained.
