本明細書で述べるポリヌクレオチドの使用は、特に、主として特定遺伝子又はそれらの
産物のセンス又はアンチセンス配列に対応し、したがってこれらの遺伝子及び/又はそれ
らの産物の発現に直接の影響を及ぼす配列を含むポリヌクレオチドを指す。例えば、遺伝

507; Elbashir et al, 2001, Nature, 411:494-498)。RNAiは、相補的配列を有する
遺伝子の発現を抑制する、植物と哺乳動物に共通する保存された経路である(Hannon and
Rossi, 2004, Nature, 431:371-378; Meister and Tuschl, 2004, Nature, 431, 343-34
9)。RNAiは、下等生物、例えば植物又は線虫において最初に認められた。これらの
系では、長いdsRNAはRNAiの有効なトリガーとして働く。長いdsRNAは実際
のトリガーではないが、エンドリボヌクレアーゼ、ダイサー(Dicer)によってsiRN
Aと呼ばれる小さなエフェクター分子に分解される。哺乳動物では、ダイサーのプロセシ
ングは、RNA結合タンパク質、TRBPとの複合体として起こる。新生siRNAは、

導型サイレンシング複合体(RISC)を形成する(Chendrimada et al, 2005, Nature,
436:740-744)。ひとたびRISCが形成されると、siRNAの一方の鎖(パッセンジ
ャー鎖)は分解されるか又は処分され、他方の鎖(ガイド鎖)は残存してサイレンシング
複合体の配列特異性を指令する。RISCのAgo2成分は、ガイド鎖の指令下で標的R
NAを切断するリボヌクレアーゼである。
ョウバエ(D. melanogaster)においてRNAiを開始させるために一般的に使用される
が、これらの分子は自然免疫系を活性化し、高等生物におけるインターフェロン(IFN
)応答を引き起こす。RNAiは、一般にIFN応答を誘導しない短いRNAを使用して
哺乳動物細胞において実施され得る。多くの研究者らは、今日、長い基質RNAのダイサ
ープロセシングから生じる天然siRNAを模倣する、合成21量体RNA二本鎖をRN
Ai試薬として使用する。選択的なアプローチは、ダイサーのための基質である、長さが
21量体よりも大きい合成RNA二本鎖を使用することである(Tuschl, T. 2002, Natur
e Biotechnology, 20:446)。

鎖に関係すると思われる(Rose et al, 2005, Nucleic Acids Res, 33:4140-4156)。ダ
イサー基質アプローチは、同じ部位で従来の21量体siRNAよりも10倍高い効力を
有する試薬を生じさせることができる。

RNAは、DNAから転写されるがタンパク質には翻訳されない遺伝子によってコードさ
れる(非コードRNA);その代わりに、それらは、プリmiRNAとして知られる一次
転写産物からプレmiRNAと呼ばれる短いステムループ構造へとプロセシングされ、最
終的には機能性miRNAへとプロセシングされる。成熟miRNA分子は、1又はそれ
以上のメッセンジャーRNA(mRNA)と部分的に相補的であり、それらの主たる機能
は遺伝子発現を下方調節することである。miRNAは細胞内で生成され、高度に保存さ
れているが、mRNA配列と完全な相補性を有することはまれである。しかし、miRN
Aは、同じくAgoタンパク質を必要とする(siRNAにおいて認められるように必ず
しもAgo2ではない)、mRNAの3’UTR領域上のその不完全に適合する標的部位
への結合によってタンパク質翻訳及びmRNA分解に影響を及ぼし得る。siRNAをm
iRNAと比較し、対比させると、siRNAが3’UTR上の不完全な相補的標的にヒ
ットした場合、siRNAはマイクロRNAと同様に挙動し、miRNAがmRNA上の
完全に適合する標的にヒットした場合、miRNAはsiRNAのように挙動し得ること
を示す。したがって、構造的に異なるが、siRNAとmiRNAはどちらも宿主動物の
細胞において類似の生物学的機能を有し、作用機構に多少の相違があると考えられる。

が癌ワクチンの効力を高めるために使用された (Kim et al, Cancer Research, 2005, 65
:309-316)。この試験では、ヒトパピローマウイルス16型E7をコードするDNAワク
チンと、鍵となるプロアポトーシスタンパク質Bac及びBaxを標的化するsiRNA
の同時投与が、ワクチン接種マウスにおいてリンパ節中の抗原発現DCの寿命を延長させ
、E7発現腫瘍モデルに対して強力な抗腫瘍作用を有する抗原特異的CD8 T細胞応答
を増強することが示された。したがって、感染症/自己免疫疾患、癌に対する有効なワク
チン接種の間及び移植の間の特定の望ましくない応答を沈黙化させるためのsiRNAの
使用には有望な見通しがある。対象とする細胞の細胞内区画へのsiRNAの効率的な送
達は、そのような方法の成功のために重要であり、強化された送達製剤の使用を必要とす
る。

全な効力を保持するための2’−O−メチルRNA残基の組込みを有し得る。siRNA
はまた、安定性と効力を改善するために2’−フルオロ修飾を有してもよく、これは通常
、ピリミジン塩基において選択的に組み込まれる。

することができるポックスウイルスベクターは、ポリヌクレオチドが哺乳動物細胞におけ
る発現のために適切な条件下でウイルスゲノムに挿入されるように、Kieny et al. 1984,
Nature 312:163に記載されている相同的組換えによって入手し得る。

した。プラスミドを大腸菌のXL1株に形質転換し、LB培地で増殖させて、アンピシリ
ン−LB寒天プレートに画線接種し、37℃で一晩インキュベートした。アンピシリンを
添加したTB培地中で細菌を単一コロニーから増殖させた。LPSの完全な除去を確実に
するためエンドフリーマキシ又はメガプレップキット(Qiagen,Mississa
uga,Ontario,Canada)を使用して大規模細菌培養物からプラスミドD
NAを精製した。
使用して、リン脂質の精製ダイズ由来混合物(Lipoid GmbHより提供されたリ
ン脂質S100)とコレステロールの10:1(W/W)混合物を水和することにより、
多重層リポソームを調製した。次に、手動押出機(Avanti lipids,Ala
baster,Al,USA)を用いて200nmポリカーボネート膜を通してリポソー
ムを押出した。最終リポソーム製剤を、その後、鉱油ベースの油性担体である不完全フロ
イントアジュバント(Sigma,Oakville,Ontario,Canada)
中に、等しい容量のGFPプラスミド含有リポソームと不完全フロイントアジュバントを
混合することによって乳化した。各々のマウスについての最終注射容量は100μlであ
り、この容量は、マウス1匹につき各投与当たりプラスミド40μgを送達した。

ド40μg、第4群のマウスには、前述したように、但し連続疎水性担体を含まずにリポ
ソーム中で製剤したGFPプラスミド40μg、第5群のマウスには、GFPプラスミド
を含まないリポソーム/連続疎水性担体からなる対照製剤を注射し、そして第6群のマウ
スは未処置のままとした。注射はすべて100μlの容量であった。すべてのマウスから
の流入領域リンパ節を、注射後8日目に収集した。リンパ節を解剖し、単細胞懸濁液を調
製した。リンパ節におけるGFP遺伝子発現の効果を、GFP陽性細胞を検出するための
2色免疫蛍光染色によって測定した。チャネルFL1におけるGFP検出と共にチャネル
FL2において抗原提示細胞を同定するため、細胞をフィコエリトリン結合CD11b及
びCD11c抗体で染色した。試料をフローサイトメーター(FACSCalibur,
BD Biosciences,San Jose,CA)にかけた。GFP陽性細胞の
検出精度を高めるため、各々の試料について少なくとも3×105事象を収集した。
、第1群〜第6群から単離したリンパ節細胞の数を図3に示す。PBS中のGFPプラス
ミドを注射したマウス(第1群)から単離したリンパ節細胞は、低レベルのGFPタンパ
ク質発現細胞を示した(<10)。しかし、連続疎水性担体中にリポソームを含む製剤中
でGFPプラスミドを注射したマウス(第2群)から単離したリンパ節細胞は、GFP陽
性細胞のほぼ4倍の増加を示した。さらに、第3群及び第4群からのマウスから単離した
リンパ節細胞は、第2群のマウスと比較して有意に低い数のGFPタンパク質発現細胞を
示した。第5群及び第6群のマウスにおいては、最小限の蛍光が検出され、これらのマウ
スはGFPプラスミド注射を受けなかったので、この最小限の蛍光はバックグラウンド自
己蛍光事象に帰せられた。さらに、GFP陽性細胞の大部分はCD11b/CD11c陽
性リンパ節細胞の集団内で検出された。CD11b/CD11c及びGFP二重陽性であ
る細胞を特異的に標的化するリンパ節細胞の再解析の結果は、図3に提示した所見と相関
し、第2群からのCD11b/CD11c陽性細胞におけるGFP発現は少なくとも4倍
高かった(図4)。この再解析はまた、対照の第5群及び第6群からのリンパ節細胞にお
いては特異的蛍光が検出できないことを確認した。この所見は、この実験におけるGFP
陽性細胞検出の特異性を確認した。従って、本実験では、リポソームと連続疎水性担体の
いずれか又は両方を欠く製剤と比較して、リポソーム/疎水性担体製剤を使用して卓越し
たGFPプラスミドの取込み及びタンパク質発現が達成された。
ヌクレオチド配列に対してsiRNAによってインビボでタンパク質コードヌクレオチド
配列の発現を阻害する能力を明らかにするため、IL−12完全コード配列及びIL−1
2に対するsiRNA配列を用いて作製したモデル発現プラスミドを選択した。IL−1
2プラスミドを最初にリポソーム中で作製し、生じたIL−12プラスミド/リポソーム
製剤を、鉱油からなる連続疎水性担体でモデル油中水型エマルションに乳化した。IL−
12プラスミド注射の1日前に、siRNAを、PBS中又はリポソーム/鉱油からなる
連続疎水性担体を含む油中水型エマルション中で注射した。siRNAのインビボでの機
能的活性を、注射部位の付近のリンパ節から単離した細胞においてIL−12発現能を検
査することによって測定した。

の基部の上方の右側腹部に皮下的に実施した。

免疫検定法(ELISA)によって実施した。簡単に述べると、抗IL−12捕捉抗体を
ELISAプレート上に一晩被覆し、試料と標準IL−12を添加した。十分に洗浄した
後、ビオチニル化抗IL−12検出抗体を添加し、室温で2時間インキュベートして、洗
浄した。アビジン−HRPコンジュゲートと共にインキュベートし、洗浄した後、発色の
ためにABTS液体基質(Sigma,St Louis,MO)を使用した。マイクロ
タイタープレートリーダーを使用して405nmで吸光度を読み取り、キット中で供給さ
れるIL−12標準品を用いて作成した標準曲線を使用してIL−12濃度を外挿した。

少が認められ、ナイーブマウスで認められたのと同程度に低かった。さらに、第3群のマ
ウスからのリンパ節細胞は、PBS中のsiRNAを摂取した第2群のマウスと比較して
有意に低いIL−12を分泌した。これは、IL−12プラスミドによって誘導されるI
L−12タンパク質発現のより良好な阻害を生じさせる、リポソーム/連続疎水性担体中
で注射した場合のインビボでのsiRNAヌクレオチド配列の改善された送達を明らかに
した。

った。さらに、第3群及び第5群のマウスからのリンパ節細胞は、PBS中のsiRNA
を摂取した第2群及び第4群のマウスと比較して有意に低いIL−12を分泌した。これ
は、オボアルブミン抗原によって誘導されるIL−12タンパク質発現の完全な阻害を生
じさせる、リポソーム/連続疎水性担体中で注射した場合のインビボでのsiRNAヌク
レオチド配列の有効で改善された送達を示す。
