1.発明の分野
一般的に、本願は、光学測定または試験手段を含む、分子生物学および微生物学の化学処理および装置に関する。ある特定の態様は、ケミルミネッセンスまたは他のルミネッセンスを示すアッセイを撮像するためのデバイス、システム、および方法に関する。
ある特定の生体分子を検出および識別することは、植物および動物といった生物システムを研究する際に重要であり得る。そのようなシステムは、いくつかの例ではハイエンドの実験室グレードの機器および処理によってしか精確に測定されることのできない、タンパク質のようなごく少量の生体分子を示し得る。そのような測定値を精確に取得するために、過去10年にわたり、数多くのアッセイ手法が開発されてきた。

第2のパスで前記ルミネッセンスの媒体に対し第2の方向に、前記ルミネッセンスの媒体の端から端まで、前記画像センサアレイを移動させる工程であって、前記第2の方向は、前記第1の方向と反対である、工程と、
前記第1のパスにおける位置からのピクセル読み取り値を、前記第2のパスにおける前記位置からのピクセル読み取り値と、組み合わせる工程と、
前記第1および第2のパスからの前記組み合わせられたピクセル読み取り値から画像を構築する工程と

ターゲット生体分子を空間パターンでサンプル媒体に固定する工程であって、前記パターンは、前記ターゲット生体分子のアイデンティティについての情報を含む、工程と、
前記ターゲット生体分子に対し結合親和性を有するプライマリプローブ生体分子を用いて前記ターゲット生体分子を探索する工程と、
前記プライマリプローブ生体分子に対し結合親和性を有するセカンダリプローブ生体分子を用いて前記プライマリプローブ生体分子を探索する工程であって、前記セカンダリプローブ生体分子は、基質生体分子の変化による検出可能なエネルギーの発生をトリガする酵素活性を有し、前記基質生体分子は、前記セカンダリプローブ生体分子の前記酵素活性のための基質である、工程と、
第1のパスで少なくとも一つの空間次元で前記サンプル媒体の端から端まで画像センサアレイを移動させる工程と、
前記第1のパス中に前記画像センサアレイによってルミネッセンスを読み取る工程と、
前記画像センサアレイによって読み取られた前記ルミネッセンスから画像を構築する工程と

本明細書に提示されるのは、新規な撮像方法と、ハイエンドCCDイメージャーの感度を達成するが、極めてより小さいサイズを有し、冷却の必要のない装置である。したがって、この撮像装置は、従来よりもはるかに低いコストで生産され、維持されることができる。
「検出限界(LOD)」は、システムノイズレベルの標準偏差の3倍大きい信号を生成する、確実に検出され得るサンプルの最低濃度を含み、またはそうでなければ当該技術で周知のとおりである。
従来技術に対する本発明の態様の技術的利点がいくつかある。レンズによって一度にサンプル全体を撮像する代わりに、サンプルにわたり近接してスキャンすることは、より多くの光が薄暗いルミネッセンスから収集されることを可能にする。高いNAの光学部品および大きい有効ピクセルサイズが、収集されるフォトンの量を最大化するために使用され得る。いくらかの深さを有するウェルを有するプレートを撮像するために、サンプルにわたり近接してスキャンすることは、広領域イメージャーと比較して、追加の利点を提供する。領域イメージャーは、撮像光学部品が異なる角度からウェルを見る事実の結果としてもたらされる、パララックス(視差)として知られる、歪みアーチファクトを有することが知られている。近接してスキャンすることは、パララックス(視差)の問題を回避し、最小の歪みとともに、同一の手法で、すべてのウェルの画像を生成する。
図1A〜図1Bは、態様に係るコンパクトな撮像スキャナを示す。ルミネッセンススキャナ装置100は、フラットベッドの紙文書スタイルのスキャナのような形状であり、蓋101、基部102、およびサンプルベッド103を含む。サンプルベッド103は、支持体としての透明なガラスまたはプラスチックを含み、湿った膜を保持することができる。2つの撮像スキャニングバー、検出器バー104および検出器バー105が、スキャニングのために利用可能である。検出器バー104が、サンプル領域の上側に置かれる一方で、検出器バー105は、サンプル領域の中央に置かれる。

典型的なリニアCCDは、直線で配列された撮像ダイオードのアレイを有する。光がこれらのダイオード上に当たると、各ダイオードは、そのフォトダイオード上に当たった光の量に比例した光電荷を作り出す。曝露時間と呼ばれる、ある長さの時間の後、ダイオード転送としばしば呼ばれる信号が使用されて、フォトダイオードにおける光電荷をCCD転送アレイへと転送する。CCD転送アレイは、CCD転送アレイの一端に向けて個々の光電荷を移動させる。CCD転送アレイは、段階的に光電荷を移動させる。CCD転送アレイは普通、少なくとも一つのクロック信号、多くのケースで2つ以上のクロック信号によって制御される。必要とされるクロック数は、含まれるCCDの設計によって決定される。典型的には、CCD転送アレイは、CCD転送アレイをドライブするクロック信号の各クロックの立ち上がりのために一段階で光電荷を移動させるだろう。
いくつかの態様の局面は、リニアCCD読み出しクロックの速度を変えることである。これは、画像の信号対ノイズ比を改善することができる。前述したように、リニアCCDアレイの出力増幅器によって提示される電圧を歪めるいくつかのノイズ源が存在する。ノイズ源の2つは、しばしば1/fノイズと呼ばれるフリッカーノイズ、およびホワイトノイズである。
Claims (14)
- 以下を含む、ルミネッセンスのサンプルをスキャンするためのルミネッセンススキャナ装置:
サンプルベッドと、
一つまたは複数の画像センサアレイを有する、前記サンプルベッドに対し移動するように構成された検出器バーであって、各画像センサアレイは、複数のピクセル素子を有する、検出器バーと、
前記検出器バーと動作的に結合されたメモリ装置およびプロセッサであって、前記メモリ装置が前記プロセッサを次の動作を実行するようにプログラムするための命令を有する、メモリ装置およびプロセッサ:
前記検出器バーを前記サンプルベッドに対し第1のパスで移動させるための動作;
前記第1のパスの間に複数の所定の曝露時間にわたってピクセル読み取り値の第1のセットを得るための動作であって、前記第1のセットの各ピクセル読み取り値が、一所定の曝露時間当たり、それぞれのx、y位置で得られる、動作;
前記検出器バーを前記サンプルベッドに対し第2のパスで移動させるための動作であって、前記第2のパスが前記第1のパスと反対方向である、動作;
前記第2のパスの間に複数の所定の曝露時間にわたってピクセル読み取り値の第2のセットを収集するための動作であって、前記第2のセットの各ピクセル読み取り値が、一所定の曝露時間当たり、それぞれのx、y位置で収集される、動作;および、
画像を構築するために、前記プロセッサを用いて、前記ピクセル読み取り値の第1および第2のセットの間でそれぞれのx、y位置のピクセル読み取り値を合計するための動作。 - 経時的に変化するルミネッセンスを有する、前記サンプルベッド上のルミネッセンスのサンプルをさらに含み、前記ピクセル読み取り値の第1および第2のセットの間でそれぞれの位置のピクセル読み取り値を合計するための動作が、前記ルミネッセンスのサンプルの変化するルミネッセンスを補償する、請求項1記載の装置。
- 前記メモリ装置が、前記画像を構築するために、前記合計するための動作を用いて、前記ピクセル読み取り値の第1および第2のセットの間でそれぞれの位置のピクセル読み取り値を平均するための命令をさらに含む、請求項1記載の装置。
- 前記ピクセル読み取り値の第1および第2のセットの前記所定の曝露時間が0.5秒である、請求項1記載の装置。
- 前記サンプルベッドが取り外し可能なウェルプレートを含む、請求項1記載の装置。
- 前記サンプルベッドと前記一つまたは複数の画像センサアレイとの間に設けられた屈折率分布型(GRIN)レンズアレイをさらに含み、前記GRINレンズアレイは、0.10以上の作動開口数(NA)を有し、かつ前記検出器バーとともに移動するように構成されている、請求項1記載の装置。
- 前記一つまたは複数の画像センサアレイが、電荷結合素子(CCD)画像センサアレイを含む、請求項1記載の装置。
- CCD画像センサアレイが、集積回路(IC)内に組み込まれ、
隣接したCCDフォトダイオードから前記CCD画像センサアレイの読み出しコンデンサへの複数の光電荷を合計することによって、前記CCD画像センサアレイの信号対ノイズ比を増大させるように適合された回路をさらに含む、
請求項7記載の装置。 - 検出器バーが、前記検出器バーに沿って互いに対しジグザグ配置された複数のリニア画像センサアレイ集積回路(IC)を含む、請求項1記載の装置。
- 前記検出器バーが第1の検出器バーであり、
一つまたは複数の画像センサアレイを含む第2の検出器バーであって、サンプルベッドに対し移動するように構成された第2の検出器バーをさらに含み、ここで、
前記第1および第2の検出器バーは、前記サンプルベッドの異なる非重複領域にわたりスキャンするように構成され、
前記メモリ装置は、前記プロセッサをさらに次の動作を行うようにプログラムするための命令を有する、請求項1記載の装置:
