Description
変異mtDNAを持つ者は、誕生時は正常であるが、成長の過程で、変異mtDNAの比率がある閾値を越えると臨床症状を発現する。例えば、MELAS患者や心筋症患者ではmtDNAの3243位に、MERRF患者は8344位に、CPEOやPearson病ではmtDNAの大きな欠失が報告されている(非特許文献1)。さらに、A3243G置換変異mtDNAは、糖尿病の原因にもなることが明らかにされた。
以上のような変異mtDNAに起因する疾患は、いずれも重篤な症状を引き起こすものであるが、現在までのところ、有効な治療方法がないため、治療法の早期確立が強く望まれている。
一方、ヒト細胞を含む哺乳類細胞のmtDNA複製は、θ型であるとの報告(非特許文献7)がある他、組換えと複製が協働したT4ファージ型の複製を行う可能性を示唆する報告などがあり(非特許文献8)、現在のところ、そのメカニズムの詳細は混沌としており明らかにされていない。しかしながら、ローリングサークル型のmtDNA複製ではないというのが定説となっている(非特許文献9及び非特許文献10)。
そこで、ミトコンドリア病の根治療法を開発するためにも、哺乳類細胞mtDNAのホモプラスミー化のメカニズムについて、さらなる研究の進捗が待たれる状況である。
また、本発明は、上記方法を使用して細胞をホモプラスミー化する方法の提供を目的とする。
さらに、本発明は、細胞内の変異mtDNAの存在比率が変化した細胞、及びホモプラスミー細胞の提供を目的とする。
すなわち、本発明の以下の(1)〜(12)である。
(1)真核細胞に活性酸素種又は活性酸素種を細胞内に発生させる化学種を接触させ、該細胞内の変異mtDNAの存在比率を変化させる方法。
(2)前記活性酸素種が、一重項酸素、スーパーオキシドアニオンラジカル、過酸化水素又はヒドロキシラジカルからなるグループより選択される1又は複数の組合せである上記(1)に記載の方法。
(3)前記活性酸素種を細胞内に発生させる化学種が、過酸化水素、塩化鉄(II)、アロキサン又はアンチマイシンからなるグループより選択される1又は複数の組合せである上記(1)に記載の方法。
(4)前記活性酸素種又は活性酸素種を細胞内に発生させる化学種が、過酸化水素である上記(2)又は(3)に記載の方法。
(5)前記真核細胞に活性酸素種又は活性酸素種を細胞内に発生させる化学種を接触させる時間が、10分〜1時間である上記(1)乃至(4)のいずれかに記載の方法。
(6)細胞内の変異mtDNAの存在比率を5%以下に変化させることを特徴とする上記(1)乃至(5)のいずれかに記載の方法。
(7)上記(1)乃至(6)のいずれかに記載の方法を用いて、細胞内の変異mtDNAの存在比率を変化させた細胞を調製する方法。
(8)上記(1)乃至(6)のいずれかに記載の方法を用いて、ホモプラスミー細胞を調製する方法。
(9)前記ホモプラスミー細胞が、正常なmtDNAからなることを特徴とする上記(8)に記載の方法。
(10)上記(7)乃至(9)のいずれかに記載の方法により調製される細胞。
(11)上記(10)に記載の細胞から調製したiPS細胞。
(12)上記(10)に記載の細胞から、iPS細胞を介さずに調製した、該細胞とは異なる細胞型を有する細胞。
従って、本発明は、変異mtDNAに起因する疾患の治療のための組織材料の提供において極めて有効である。
本発明で使用される真核細胞は、特に限定はされないが、例えば、哺乳類細胞であり、中でも、ヒト及び非ヒト動物由来の細胞である。そのような細胞として、例えば、マウス、ラット、ウシ、ウマ、ブタ、ヒツジ、サル、イヌ、ネコ、トリなどに由来する細胞を例示することができ、特に好ましくは、ヒト由来の細胞である。また、本発明で使用する真核細胞は、mtDNAに変異を有する細胞であるか、又は、mtDNAに変異を有する事が疑われる細胞が特に好ましい。
また、真核細胞に、活性酸素種又は活性酸素種を細胞内に発生させる化学種を「接触」させるとは、活性酸素種又は活性酸素種を細胞内に発生させる化学種が目的の細胞内へ侵入できるように、該細胞が生育している培地中などに該活性酸素種又は活性酸素種を細胞内に発生させる化学種を存在させ、該細胞と該活性酸素種又は活性酸素種を細胞内に発生させる化学種が接触可能となるように処理することである。細胞と接触させる活性酸素種又は活性酸素種を細胞内に発生させる化学種は、1種でも複数種でもよく、また、活性酸素種と活性酸素種を細胞内に発生させる化学種を組み合わせて培地に添加してもよい。
活性酸素種を細胞内に発生させる化学種を培地中に添加する場合、限定はしないが、例えば、塩化鉄(II)(FeCl2)(培地中に添加する濃度としては、例えば、1〜30μM、好ましくは、5μM〜20μM、より好ましくは、10μM程度)、アロキサン(培地中に添加する濃度としては、例えば、1〜30mM、好ましくは、5mM〜20mM、より好ましくは、10mM程度)、アンチマイシン(培地中に添加する濃度としては、例えば、10〜500μM、好ましくは、50μM〜300μM、より好ましくは、100μM〜250μM、さらに好ましくは、200μM程度)などの化学種を培地中に添加することができる。活性酸素種を細胞内に発生させる化学種を培地中に添加する場合、当該化学種の添加量についても、細胞に対するダメージを予め調べることで容易に確認することができる。
細胞内の変異mtDNAの存在比率の変化は、増加又は減少のいずれであってもよい。例えば、疾患原因となる変異mtDNAの存在比率が通常の方法では検出できない程度に低い場合、本発明を使用して、その存在比率を増加させることで、該疾患の素因の有無を判定することも可能である。従って、本発明には、検出限界以下のmtDNA変異に起因する疾患の罹患可能性を判定する方法も含まれる。
また、細胞内の変異mtDNAの存在比率を可能な限り減少させると、ホモプラスミー細胞(後述する)の調製が可能である。
細胞内の変異mtDNAの存在比率の変化は、例えば、変異の有無によって切断感受性が異なる制限酵素を利用して、簡便に判定することが可能である(例えば、Goto et al., Nature, 348, 651-653, 1990などを参照のこと)。その他、対立遺伝子特異的PCR法、定量PCR法、インベーダー法、次世代シークエンサーを用いた解析法などの方法も利用可能である。
本発明によって変化した「細胞内の変異mtDNAの存在比率」は、その存在比率を減少させる場合、減少した変異mtDNAの存在比率は、例えば30%以下、好ましくは20%以下、より好ましくは10%以下、さらに好ましくは5%以下、最も好ましくは0%である。また、その存在比率を増加させる場合、増加した変異mtDNAの存在比率は、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、好ましくは70%以上、80%以上、より好ましくは90%以上、さらに好ましくは95%以上である。
ホモプラスミーとは、mtDNAの塩基配列が細胞レベル又は個体レベルで均一な状態のことである。これに対して、mtDNAの塩基配列が細胞レベル又は個体レベルで均一な状態でないことをヘテロプラスミーという。例えば、ある細胞(又は個体)に含まれるmtDNAの塩基配列が全てのミトコンドリアゲノムコピーにおいて均一である場合、該細胞(又は個体)は「ホモプラスミーの細胞(又は個体)」である。本明細書においては、正常な遺伝子配列を均一に含む細胞(又は個体)については、特に、「正常なホモプラスミー細胞(又は個体)」、あるいは、「正常なmtDNAからなるホモプラスミー細胞(又は個体)」などと記載する。さらに、本明細書においては、mtDNA上に存在する特定の変異に着目して、該変異箇所の塩基配列が細胞レベル又は個体レベルで均一な場合は、該特定の変異に関しホモプラスミーな状態と称する。
また、本明細書において、ホモプラスミーとは、同一の塩基配列を持つmtDNAコピーの細胞内比率が、90%以上、特に好ましくは95%以上、最も好ましくは99%以上(変異mtDNAが1%以下)の状態のことである。
「細胞内の変異mtDNAの存在比率が所望の割合に変化した細胞」には、細胞内の変異mtDNAの存在比率が増加した細胞及び減少した細胞のいずれも含まれるものである。例えば、細胞内の変異mtDNAの存在比率が減少した細胞とは、細胞内の変異mtDNAの存在比率が、例えば30%以下、好ましくは20%以下、より好ましくは10%以下、さらに好ましくは5%以下、最も好ましくは0%の細胞である。また、細胞内の変異mtDNAの存在比率が増加した細胞とは、細胞内の変異mtDNAの存在比率が、例えば10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、好ましくは70%以上、80%以上、より好ましくは90%以上、さらに好ましくは95%以上の細胞である。
このような異なる細胞型の細胞を調製する方法としては、本発明で調製した細胞内の変異mtDNAの存在比率が所望の割合に変化した細胞、あるいは、ホモプラスミー細胞からiPS細胞を調製し、該iPS細胞から所望の分化状態を有する所望の細胞型の細胞へ分化誘導する方法の他、iPS細胞を介さずに直接所望の細胞型を有する細胞を調製する方法などがある。
本発明により調製されるホモプラスミー細胞に分化多能性因子をコードする遺伝子を導入し、前掲の文献を参照することで、iPS細胞を樹立することができる。ここで樹立したiPS細胞から分化誘導した各種組織又は細胞は、ホモプラスミー細胞が由来する個体に移植する場合、組織不適合性の問題を解決することが可能で、ミトコンドリア病などの有効な治療材料として利用することができる。
本発明のホモプラスミー細胞から調製される所望の細胞型を有する細胞は、治療等の目的に沿った如何なる細胞型の細胞であってもよいが、例えば、神経細胞、横紋筋細胞、平滑筋細胞、上皮細胞、表皮細胞、角質細胞、造血細胞、メラニン形成細胞、軟骨細胞、網膜上皮細胞、β細胞、心筋細胞、B細胞、T細胞、血液系細胞(赤血球、白血球、血小板など)、線維芽細胞などが好ましい。
ミトコンドリア病の1つであるMELASの発症原因として、mtDNAのA3243G置換変異が確認されている。そこで、MELAS患者の骨格筋及び皮膚に由来する初代培養細胞(国立精神・神経センター、後藤雄一博士により調製された。以下、MELAS細胞と記載する)のmtDNAに存在するA3243G置換変異のヘテロプラスミー状態について検討した。
MELAS細胞をFCS10%含むDMEMの存在下、細胞密度が1×105〜1×106cell/mLとなるようにプレーティングし、37℃、5%CO2の条件下で3日間培養した。3日間の培養後、細胞密度が5×105〜6×106cells/mLに達した後、104〜1.2×105倍に希釈してプレーティングを行い、さらに、15日間培養を行った。15日間の培養後、細胞のシングルコロニーをいくつか選択して、定法によりDNAを抽出した。抽出したDNAサンプルを鋳型として、mtDNAの3243位付近を増幅するプライマーペアーを用いてPCRを行い、PCR産物に対してApaI処理を行った。mtDNAの3243位付近の配列は、正常な場合には、GAGCCC(下線が3243位に対応する)であるが、MELAS患者のmtDNAでは突然変異によって、GGGCCC(下線が3243位に対応する)と変化し、ApaI処理に対して感受性を示すようになる。従って、正常なmtDNAに由来するPCR産物は、ApaI認識配列を含まないため、ApaI処理後も単一のDNAバンド(本実施例においては、294bpのバンド)のみが検出されるが、突然変異を含むmtDNAに由来するPCR産物はApaI感受性となる結果、ApaIによって切断された2本のバンド(本実施例においては、182bpと112bpのバンド)が検出されるようになる(図1の下図を参照)。従って、294bpに対する182bp及び112bpのバンドの比率を算定することによって、細胞のヘテロプラスミー状態を評価することが可能となる(本方法の詳細は、Goto et al., Nature, 348, 651-653, 1990を参照のこと)。
図1は、MELAS細胞株から得られるいくつかの細胞クローンについて、A3243G置換変異の存在比率を検討した結果である。MELAS患者由来の細胞から得られた細胞クローンのヘテロプラスミー状態はいずれのクローンについても、30%程度と安定していることが確認された(図1の上図)。
次に、上記MELAS細胞に、活性酸素種として過酸化水素を接触させた場合、ヘテロプラスミー状態にどのような影響が生じるか検討した。
MELAS細胞(細胞密度;1×105〜1×106cell/mL)の培地中に最終濃度100μMとなるように過酸化水素を添加し、37℃、5%CO2の条件下で30分間インキュベートした後、細胞をPBSで3回洗浄し、3日間培養した。3日間の培養後、細胞密度細胞密度が5×105〜6×106cells/mLに達した後、104〜1.2×105倍に希釈してプレーティングを行い、さらに、15日間培養を行った。15日間の培養後、細胞のシングルコロニーをいくつか選択して、定法によりDNAを抽出した。得られたDNAサンプルに対して、mtDNAのA3243G置換変異の細胞内存在比率を上述と同様の方法で調べた。
図2は、過酸化水素処理後に得られた各細胞クローン中に存在するA3243Gの比率を調べた結果である。過酸化水素で処理を行うと、A3243G置換変異がほぼ0%になる細胞クローン(例えば、クローン16、25など。図2の上図を参照のこと)を得ることができた。なお、A3243G置換変異が0%である、正常なホモプラスミー細胞を選択する場合、フッ化ナトリウム(例えば、0.2μg/mL)やヨード酢酸(例えば、0.2μg/mL)などの解糖系を阻害する薬剤を添加すると、正常なホモプラスミー細胞の選択がさらに容易になる(図2の上図、“selected clone”)。
以上のことから、細胞の過酸化水素処理(活性酸素種による処理)は、細胞のホモプラスミー化を促進することが明らかとなった。
Claims (11)
- mtDNAに変異を有する哺乳類細胞に、活性酸素種又は活性酸素種を細胞内に発生させる化学種を接触させ、該細胞内の該変異を有するmtDNAの存在比率を減少させる方法。
- 前記活性酸素種が、一重項酸素、スーパーオキシドアニオンラジカル、過酸化水素又はヒドロキシラジカルからなるグループより選択される1又は複数の組合せである請求項1に記載の方法。
- 前記活性酸素種を細胞内に発生させる化学種が、過酸化水素、塩化鉄(II)、アロキサン又はアンチマイシンからなるグループより選択される1又は複数の組合せである請求項1に記載の方法。
- 前記活性酸素種又は活性酸素種を細胞内に発生させる化学種が、過酸化水素である請求項2又は3に記載の方法。
- 前記真核細胞に活性酸素種又は活性酸素種を細胞内に発生させる化学種を接触させる時間が、10分〜1時間である請求項1乃至4のいずれかに記載の方法。
- 細胞内に存在していた変異mtDNAの存在比率を5%以下に減少させることを特徴とする請求項1乃至5のいずれかに記載の方法。
- 請求項1乃至6のいずれかに記載の方法を用いて、細胞内に存在していた変異mtDNAの存在比率を減少させた細胞を調製する方法。
- 請求項1乃至6のいずれかに記載の方法を用いて、ホモプラスミー細胞を調製する方法。
- 前記ホモプラスミー細胞が、正常なmtDNAからなることを特徴とする請求項8に記載の方法。
- 請求項7乃至9のいずれかに記載の方法により調製される細胞から、iPS細胞を調製する方法。
- 請求項7乃至9のいずれかに記載の方法により調製される細胞から、iPS細胞を介さずに、該細胞とは異なる細胞型を有する細胞を調製する方法。
