本明細書中に開示される本発明の実施形態は、免疫原+免疫賦活剤又はBRMを含む免疫原性組成物を被検体に投与し、それにより抗原特異的T細胞応答を誘導するか又は増強する方法に関する。好ましい実施形態では、免疫原性組成物は、指数的漸増方式で被検体に提供される。さらなる実施形態では、免疫原性組成物は、線形漸増方式で被検体に提供される。
本明細書中に開示される免疫原性組成物は、以下の実施例におけるような皮下注射器を用いて、又はワクチン接種のための当該技術分野で既知の他の同様の機能性の用具を用いて、ボーラス注射により送達され得る。送達/投与の他の方法としては、例えばポンプのような免疫原送達手段によるリンパ系への皮下的又は直接的な注入を包含し得る。好ましい実施形態では、送達手段は、動物にとって外部であるが、しかし身体中への、好ましくはリンパ系器官又は高リンパ流の領域へ抗原を送達するための手段(例えば針又はカテーテル)を含有する。免疫原送達手段の一利点は、それが多数回実施される注射を不要にする点である。
概して、本発明の実施形態は、被検体の免疫系が疾患を攻撃するために疾患関連抗原に対する細胞媒介性応答を備える疾患を有する被検体を治療するために有用である。疾患の種類は、例えば悪性腫瘍、又は細胞内に進入し、例えば細胞傷害性Tリンパ球により攻撃される細菌、ウイルス、原生動物、蠕虫、もしくは任意の微生物病原体により引き起こされる感染性疾患であり得る。一治療様式において、本発明の方法は持続性又は慢性症状に良好に適しているが、しかし必ずしもこのようなものに限定されない。さらに、本発明は、感染性疾患又は腫瘍を発症する危険に曝されている被検体を免疫感作するために有用である。
胞、精巣細胞、脳細胞、卵巣細胞、リンパ球、皮膚細胞、脳細胞、骨細胞、柔組織細胞等であり得る。抗原提示細胞は、例えば樹状細胞であり得る。これらの実施形態における投与量は、直前用量のものに比して投与される細胞の数を順次増大することにより、或いは直前用量のものに比して細胞の表面上のエピトープ−MHC複合体の数を順次増大する(この場合、エピトープは標的抗原からのものである)ことにより、或いはその両方により、増大され得る。細胞表面上のエピトープ−MHC複合体の数は、異なる濃度のエピトープでパルス標識することにより、最も容易に操作され得る。
本明細書中に記載される組成物の何れもが、キット中に一緒に集められ得る。非限定な一例では、免疫原性組成物を送達するための1つ又は複数の作用物質又は試薬は、感染性疾患又は癌による疾患又は症状を治療するために、キット中に単独で、又は付加的作用物質と組合せて提供され得る。しかしながらこれらの構成成分は、限定的であるよう意図されない。キットは、作用物質又は試薬を保存及び分散するための適切な容器手段を提供する。
の方法を実行するための材料を含入するための手段、並びに市販の密閉状態の任意の他の試薬容器も包含し得る。このような容器は、所望のバイアルが保持される射出又は吹込み成形プラスチック容器を包含し得る。容器の数又は型とは関係なく、本発明のキット(複数可)は、被検体の身体内への免疫原+免疫賦活剤又はBRMを含む免疫原性組成物の注射/投与を手助けするための器具も含み得るか、或いはそれと共に包装され得る。このような器具は、注射器、ポンプ及び/又は任意のこのような医学的に認可された送達手段であり得る。
材料及び方法
マウス 6〜12週齢C57BL/6マウスを、Harlan(Horst, The Netherlands)から購入した。糖タンパク質中に位置するH−2bマウスにおけるリンパ球脈絡髄膜炎ウイルス(LCMV)の免疫優性エピトープを表すペプチドgp33(aa33〜41)に特異的なT細胞受容体を発現するTCR318トランスジェニックマウスは、Cytos Biotechnology AG(Schlieren, Switzerland)から入手した(Pircher, H., et al. 1989. Nature 342: 559-561;Pircher, H. et al., Nature 346, 629-633, 1990)(これらの記載内容は各々、参照によりその全体が本明細書中で援用される)。HLA A2.1を発現するHHDトランスジェニックマウスは、MannKind Corporation(Valencia, CA; Pascolo, S. et al., J Exp Med 185, 2043-2051, 1997)(この記載内容は参照によりその全体が本明細書中で援用される)から最初に得られた。スイスの獣医学機関のガイドラインに従って、チューリッヒ大学病院の特定の無病原体施設で、マウスを繁殖させ、保持した。
)(この記載内容は参照によりその全体が本明細書中で援用される)。LCMV糖タンパク質ペプチドgp33(aa33〜41;KAVYNFATM、配列番号3)及びgp61(aa61〜80;GLNGPDIYKGVYQFKSVEFD、配列番号4)、並びにVSVペプチドnp52(SDLRGYVYQGLKSG、配列番号5)は、EMC Microcollections(Tubingen, Germany)から購入した。インフルエンザ・マトリックスペプチド(GILGFVFTL、配列番号6)は、Neosystems(Strasbourg, France)から入手した。用いられたHPV16 E7(aa49〜57;RAHYNIVTF、配列番号7)ペプチドは、MannKind Corporation(Valencia, CA)で99%超の純度に合成された。ホスホロチオエート修飾高CGオリゴデオキシヌクレオチド1668(5’−TCC ATG ACG TTC CTG AAT AAT−3’、配列番号8)は、Microsynth(Balgach, Switzerland)により合成された。
LCMV−WEに感染させて、脾臓中のウイルス力価をMC57細胞に関して確定した
(Battegay et al., 1991、上記)。
membrane 96-well plate, Millipore)中で37℃で72時間、10μg/ウエルのHPV16 E7ペプチドと共にインキュベートした。コーティング及び検出IFN−γ抗体(U-Cytech biosciences, Utrecht, The Netherlands)を用いて24時間展開後、ELISpot読取器及びソフトウエア(AID, Strassberg, Germany)を用いてELISPOTを定量した。
指数漸増的抗原刺激はCD8+T細胞応答を増強する
T細胞応答が漸増抗原刺激により増強され得るか否かを調べた。第一の実験では、1×106個のトランスジェニックgp33特異的T細胞をC57BL/6野生型レシピエントマウス中に移して、前駆体T細胞頻度を増大し、免疫応答の査定を促した。
意であった(2.23±0.84%対0.19±0.12%、6日目にp=0.02)。
CD8+T細胞応答の増強はT細胞ヘルプとは無関係である
CD8+T細胞初回刺激に関するT−ヘルプの役割は、当該技術分野でよく知られている。Th−エピトープは、機能性CD8+T細胞免疫にきわめて重要であり得る(Johansen et al., Eur J Immunol., 34, 91-97, 2004;Shedlock and Shen, Science, 300, 337-339, 2003;Sun and Bevan, Science, 300, 339-342, 2003)(これらの記載内容は各々、参照によりその全体が本明細書中で援用される)。他方で、前駆体頻度が高い状況では、特に強力な免疫原、例えばLCMV gp33を用いる場合、CD8+T細胞応答は低Th依存性である(Mintern et al., J Immunol., 168, 977-980, 2002)(この記載内容は参照によりその全体が本明細書中で援用される)。さらに、投与経路もT−ヘルプの要求に影響を与え得る(Bour et al., J Immunol., 160, 5522-5529, 1998)(この記載内容は参照によりその全体が本明細書中で援用される)。したがって、CTLのTh依存性は、実験設定によって大きく変わる。この仮説に基づいて、CD8+T細胞応答の増強が指数的漸増ワクチン用量でのワクチン接種によるT細胞ヘルプと無関係であるか否かを、本発明者らは調べた。
LCMV gp61(aa61〜80)Th−エピトープの混合物を用いて、上記プロトコールの場合(表1参照)と同様に、指数的漸増ワクチン用量でマウスを免疫感作した。CD8+T細胞応答に及ぼす用量動態の同一作用が上記プロトコールで免疫感作されたマウスにおいて得られた(データは示されていない)ため、指数的漸増ワクチン用量での
ワクチン接種によるCD8+T細胞応答の増強は、T細胞ヘルプと無関係であることが観察された。
4日以上の抗原投与は、最大CD8+T細胞応答を誘導する
上記実施例で示したように、4日間の期間にわたって指数的に増大した抗原は、ボーラス注射又は均一ワクチン用量の毎日注射より有意に強力なT細胞応答を誘導した。したがって、抗原提示のさらなる延長が応答をさらに増強するか否かを調べるために、実験を実行した。C57BL/6マウスの群を、同一総用量のgp33ペプチド及びCpG(125μgのp33及び12.5nmolのCpG)で、しかし異なる指数的動態に従って、ボーラスとしての用量を注射することにより、或いは4、6又は8日間にわたって(図3A)皮下免疫感作した(ピークは0日目(ボーラス)、3日目、5日目又は7日目)。
指数的漸増抗原刺激は防御的抗ウイルス応答を増強する
上記観察の機能的関連性をさらに解明するために、異なるレジメン(表1に示したようなs1〜s4)に従って固定累積用量のgp33ペプチド及びCpGで雌野生型C57BL/6マウスを免疫感作し、次に、T細胞応答がすでに収縮又は記憶期にある時点でLCMV又はLCMV糖タンパク質(vacc−gp)を発現する組換えワクシニアウイルスで攻撃誘発した(Kaech, S.M., et al., Nat Rev Immunol 2, 251-62, 2002)(この記載内容は参照によりその全体が本明細書中で援用される)。両ウイルスに対する防御は、専らCD8+T細胞によって変わる(Binder, D. and Kundig, T.M., J Immunol 146, 4301-7, 1991;Kundig, T.M. et al., Proc. Natl. Acad. Sci., USA: 93, 9716-23, 1996)(これらの記載内容は各々、参照によりその全体が本明細書中で援用される)。
活性化APCの数は刺激動態の型に依存しない
免疫感作動態が活性化状態及び活性化APCの数に影響を及ぼすか否かを試験するために、図1〜図3に、並びに表1に記載したような免疫感作プロトコールs1(ボーラス注射)及びs4(指数的漸増用量)に従って、gp33ペプチド及びCpGでC57BL/6マウスを免疫感作した。ワクチンを鼠径部に皮下投与した。1、4、6及び8日後、鼠径リンパ節を除去し、そしてその単一細胞懸濁液を、DCマーカーCD11c、並びにCD86及びMHCクラスIIマーカーI−Abの発現に関してフローサイトメトリーによ
り分析した(図5A)。結果は、ナイーブ対照(0日目)に比して平均蛍光として表現される。結果は、異なる動態が流入領域リンパ節中のDCの数にも、MHC−クラスII(I−Ab)及びCD86発現によりモニタリングされるようなそれらの活性化状態にもきわめて重要な影響を及ぼさない、ということを示唆した(図5A)。しかしながら、DC数及び活性化のピークは比較可能であるが、しかしそれらは2〜3日隔てられた。DC活性化は、免疫感作動態の型とは関係なく、最大CpG用量の1日後にその最大に達した。
指数的漸増抗原刺激はT細胞刺激延長に好都合である
免疫感作の動態がCD8+T細胞の増殖にどのように影響を及ぼしたかを試験するために、上記のように、そして表1に記載されたように、gp33ペプチド及びCpGの1回用量(s1)で、均一1日用量(s2)で、又は指数的漸増用量(s4)をマウスに注射した。マウスの一群は、陰性対照として未処置のままであった。増殖をモニタリングするために、全てのマウスに、初回免疫感作の1日前に、トランスジェニックTCR318マウスからの107又は1.5×106個のCFSE標識脾臓細胞を静脈内投与した。異なる時点で、尾からの採血によりリンパ球を単離し、そしてフローサイトメトリーによりCD8発現及びCFSE染色に関して分析した。p値は、***に入ったCFSE標識CD8+T細胞のパーセンテージに関して、s1日程とs4日程との統計学的差を示す。結果は、2つの比較可能な実験のうちの1つを示す。gp33ペプチド及びCpGのボーラス注射は、免疫感作の3日後に***するようCFSE標識CD8+T細胞を誘発した(図6A及び図6B)。増殖は、2日後にすでに検出することができた(図6B)。5日目に、前駆体細胞は未だ***に入ったが、しかし3日目より低度であり、そして7日目までには、CFSE標識細胞は新規の***に入るのを止めていた。これに対して、指数的漸増刺激は、T細胞増殖を顕著に延長した。***に入っている細胞は、全免疫感作レジメンを未だ受けていないにも拘わらず、初回免疫の3日後という早くに確定され、***は5日目及び7日目に延長した(図6B)。9日目でさえ、増殖は依然として観察された(示されていない)。さらに、***指数、即ち受けた***の平均数は、s2よりs4に関して有意に高かった(p<0.05、マン・ホイットニーによる)。
指数的漸増数のペプチドパルス標識DCはCD8+T細胞応答を増強する
異なる数のAPCの寄与を調べるために、同一総数のペプチドパルス標識DCで、しかし異なる動態を用いて、C57BL/6マウスを免疫感作した。骨髄由来DCにHPV E7(aa49〜57、RAHYNIVTF、配列番号2)ペプチドを負荷し、総量1.
11×105細胞を、1日目にボーラスとして鼠径リンパ節中に注射し(s1)、或いは同一総量の細胞を、1日目(103細胞)、3日目(104細胞)及び6日目(105細胞)に漸増(s4)パターンで投与した。さらに、T細胞初回免疫に利用可能なDCの一定総数を保証するために、ワクチンをリンパ内投与した。ナイーブマウスを、陰性対照として用いた。17日目及び22日目に、末梢血中のE7四量体陽性CD8+T細胞の頻度(図7A(黒四角))をフローサイトメトリーにより分析した。値は、平均及びSEMを表わす(n=10)。IFN−γELISPOT(図7A(白四角))を、脾臓から分析した(n=7)。値は、平均及びSEMを表わす(n=7)。21日目に、3匹のワクチン接種マウス及び10匹のナイーブマウスを、HPV形質転換腫瘍細胞株C3.43で攻撃誘発した(図7B)。腫瘍進行を内径測定(mm)によりモニタリングし、これから、腫瘍容積を算定した。VSV np52ペプチドを負荷したDCの皮下注射により免疫感作したC57BL/6マウス(n=4)において、攻撃誘発後の生存を試験した(図7C)。カプラン・マイヤー曲線の対数順位検定:s4≠s2:p=0.0084;s2≠s1:p=0.0082;s1≠ナイーブ:p=0.401。
指数的漸増抗原性刺激はT細胞におけるIL−2産生を増強する:T細胞クローンのレベルでの抗原動態の影響
本発明者らは次に、上記実施例における観察がT細胞クローンのレベルで説明され得るか、或いはそれらが示差的親和性、結合力及び機能性を有するT細胞クローン型を包含するin vivoT細胞選択過程の結果であるか否かを調べた。
et al., Annu. Rev. Immunol., 19, 683-765, 2001)(この記載内容は参照によりその全体が本明細書中で援用される)。したがって、本発明のin vivoデータは、クローンレベルで、T細胞が抗原曝露の動態を復号し得る、ということを示した。
線形的漸増抗原刺激はCD8+T細胞応答を増強する
T細胞応答が漸増抗原刺激により増強され得るか否かを確定するために、調査を実行した。一実験において、1×106個のトランスジェニックgp33特異的T細胞をC57BL/6野生型レシピエントマウス中に移して、前駆体T細胞頻度を増大し、免疫応答の査定を促した。
31 Suppl, 119-22, 2003、上記)。しかしながら、組織中では、CpG ODNは相対的に安定しており、半減期は48時間であった(Mutwiri, G.K., et al., J Control Release 97, 1-17, 2004、上記)。さらに、60分以内に、血清プロテアーゼは検出レベルより低い値に遊離ペプチドを分解させる、ということが文献中で注目される(Falo, L.D., Jr., et al., Proc Natl Acad Sci USA 89, 8347-50, 1992;Widmann, C., et al., J Immunol 147, 3745-51, 1991、上記)。
線形的漸増抗原刺激は防御的抗ウイルス応答を増強する
漸増抗原刺激によって防御的抗ウイルス応答を増強することができるか否かを確定するために調査を実行した。一実験では、異なるレジメン(実施例10に記載したs1〜s4)に従って固定累積用量のgp33ペプチド及びCpG(総量で125μgのgp33及び12.5nmolのCpG)で雌野生型C57BL/6マウスを免疫感作し、次に、T細胞応答がすでに収縮又は記憶期にある時点でLCMV又はLCMV糖タンパク質(vacc−gp)を発現する組換えワクシニアウイルスで攻撃誘発した(Kaech, S.M., et al., Nat Rev Immunol 2, 251-62, 2002、上記)。両ウイルスに対する防御は、専らCD8+T細胞によって変わる(Binder, D. and Kundig, T.M., J Immunol 146, 4301-7, 1991;Kundig, T.M. et al., Proc. 線形的漸増数のペプチドパルス標識DCはCD8+T細胞応答を増強する
異なる数のAPCの寄与を調べるために、同一総数のペプチドパルス標識DCで、しかし異なる動態を用いて、C57BL/6マウスを免疫感作した。骨髄由来DCにHPV E7(aa49〜57、RAHYNIVTF、配列番号2)ペプチドを負荷し、総量1.
2×105細胞を、1日目にボーラスとして鼠径リンパ節中に注射し(s1)、或いは同一総量の細胞を、1日目(2×104細胞)、3日目(4×104細胞)及び6日目(6×104細胞)に線形的漸増(s4)パターンで投与する。さらに、T細胞初回免疫に利用可能なDCの一定総数を保証するために、ワクチンをリンパ内投与する。ナイーブマウスを、陰性対照として用いる。17日目及び22日目に、末梢血中のE7四量体陽性CD8+T細胞の頻度をフローサイトメトリーにより分析し、IFN−γELISPOTを脾臓から分析する。21日目に、3匹のワクチン接種マウス及び10匹のナイーブマウスを、HPV形質転換腫瘍細胞株C3.43で攻撃誘発する。腫瘍進行を内径測定(mm)によりモニタリングし、これから、腫瘍容積を算定する。VSV np52ペプチドを負荷したDCの皮下注射により免疫感作したC57BL/6マウスにおいて、攻撃誘発後の生存を試験する。

- 感染性又は新生物性疾患の治療又は予防におけるクラスI MHC制限T細胞応答を刺激
するための免疫原性組成物であって、該免疫原性組成物はクラスI MHC制限T細胞エ
ピトープを含むかまたはこれをコードする免疫原を含み、該免疫原性組成物は複数回の順次用量として哺乳動物に投与され、初回用量後の各用量が直前用量より多く、前記順次用量が前記初回用量の指数関数として増大する、免疫原性組成物。 - 前記免疫原性組成物が免疫原+免疫賦活剤又は生物学的応答修飾剤を含む、請求項1記載の免疫原性組成物。
