Description
概して、本発明は、金属および/または半導体原子を含有し、RNAリガンドを結合しているコアを有することを含むナノ粒子に関する。RNAリガンドとは一般に、低分子干渉RNA(siRNA)およびマイクロRNA配列(miRNA)を模倣するよう設計された短いRNA配列である。ナノ粒子はRNAリガンドを送達するために使用でき、広範な適用において、in vitro系および治療適用または診断適用のための用途がある。一例として、本発明のナノ粒子は(1)ターゲッティングされた転写遺伝子サイレンシングに、(2)ターゲッティングされたmRNA分解に、(3)mRNAのイメージングに、(4)同一または異なるナノ粒子上の複数のRNAリガンドを用いることによる経路の阻害に、(5)例えば、肺へのエアゾール送達に、(6)siRNA耐性mRNAをターゲッティングするためにmRNAサイレンシングと組み合わせて、および(7)機能ゲノミクスにおけるツールを用いるために使用できる。
また、炭水化物の同位体標識を使用して、その検出を容易にすることもできる。
RNAリガンドを含むナノ粒子を用い、低分子干渉RNA(siRNA)によるmRNAのターゲッティングされた分解、転写後遺伝子サイレンシング(PTG)、マイクロRNA(miRNA)による発生的に調節されたmRNAの配列特異的翻訳抑制およびターゲッティングされた転写遺伝子サイレンシングをはじめ、いくつかの方法で遺伝子発現を調節できる。
本発明により、2種以上のRNA分子を送達すること、当技術分野ではこれまで可能でなかったことが可能となることは、特に注目に値する。したがって、いくつかの実施形態では、本発明は、遺伝子経路において発現をダウンレギュレートするのに使用できる、同一のナノ粒子とコンジュゲートしているか、または少なくとも2種の異なる種類のナノ粒子の組成物中に存在している、少なくとも2種の異なるRNA配列を有するナノ粒子組成物を提供する。組成物中に存在するRNAリガンドの数は、経路の複雑性およびダウンレギュレーションのためにターゲッティングされる必要がある遺伝子の数に応じて変わる。研究または治療のいずれかのために、ターゲッティングされ得る経路の例としては、炎症経路、抗ウイルス経路、癌におけるシグナル伝達経路、転移経路または代謝経路が挙げられる。一例として、これとしては、II型糖尿病におけるグルコース産生のためのグルコース新生(neoglucgogenesis)経路の調節が挙げられる。
一適用では、本発明のナノ粒子の組成物を用いて、RNAを標的細胞に送達するためのターゲッティング特性を付与できる。これは、ナノ粒子のコアとコンジュゲートしているさらなる種類のリガンドまたはRNAナノ粒子が標的細胞集団と特異的に相互作用するのを可能にするRNAリガンドと会合しているドメインを含むナノ粒子を提供することによって達成できる。一例として、標的細胞の表面または内側に存在するその結合パートナーと特異的に結合できる特異的結合対の一員であるリガンドを含むナノ粒子を提供することによって、RNA含有ナノ粒子を細胞集団に選択的に向け、例えば、核などの特定の細胞構造をターゲッティングすることができる。ナノ粒子をターゲッティングするために、それらとコンジュゲートしているリガンドとして用いるのに適した特異的結合対の例としては、リガンドおよび受容体が挙げられるが、多数の代替法が当業者には明らかであろう。例えば、グルコース誘導体化したナノ粒子を用いて、GLUTファミリーのタンパク質のメンバーを含有する細胞をターゲッティングできる(18)。ナノ粒子にはその他の糖リガンド、例えば、Glcβ4GlcNAcまたはGlcβ4GlcNH2を使用してもよい。まず前者を用いて、siRNA含有ナノ粒子を細胞表面GLUT輸送タンパク質にターゲッティングし、次いで、細胞に入る際に(グリコシダーゼによる切断後)、GlcNAcがsiRNAナノ粒子を核にさらにターゲッティングする。後者の構築物は正の電荷をナノ粒子表面に加え、細胞へ接着し、取り込まれるのがさらに容易になる。自己組織化ナノ粒子を用いて、スペーサーによってジスルフィドと結合している、脂質、ペプチドまたは何らかのその他の化学成分(例えば、リガンドについての議論において上記に記載したような)などの異種リガンドを組み込むことができるので、種々のその他のターゲッティング分子を用いてRNAナノ粒子を特定の細胞種にターゲッティングできる。一例として、これらの技術を用いてRNAを保持するナノ粒子を腫瘍細胞にターゲッティングできる。
さらなる態様では、本発明は、本明細書に記載されるナノ粒子を用いる、mRNAの検出方法および/またはイメージング方法を提供する。特に、本発明以前に、mRNAをイメージングするための当技術分野で公知の方法はなかった。本方法は、ナノ粒子上に存在するRNAリガンドが標的mRNAと相互作用する条件下、in vivoまたはサンプル中のいずれかでRNAをナノ粒子と接触させることと、ナノ粒子-RNA-mRNA複合体を検出することとを含み得る。複合体を検出するステップは、ナノ粒子の固有の特性を用いることであるか、ナノ粒子と会合している標識を検出することによってであり得る。本発明のこの態様における使用に適した標識の好ましい例としては、磁性基、量子ドットまたは放射性核種を含むナノ粒子が挙げられる。例えば、セレン化カドミウムのナノ結晶によって提供されるような量子ドット、またはセレニドに加えその他の陰イオン、例えば、スルフィドを用いることができるが、これは電子装置および光学装置の双方での生物学的イメージング、量子コンピュータおよび候補薬物のスクリーニングにおいて利用される可能性がある。
ゲノムスクリーニングでは、通常、ランダムに作製したRNA配列を利用し、次いで、それを試験細胞にトランスフェクトし、タンパク質発現の変化をモニターする。本発明のナノ粒子には、ゲノムスクリーニングについてのこれらの先行技術の方法を上回る2つの大きな利点がある。第1は、多数のランダムsiRNA配列が実際には効果がないが、目下のところ試験細胞は個々にスクリーニングされる必要があるため、この形のこれらの実験の労力と費用が増大する。本発明によって複数のsiRNA配列がナノ粒子上にリガンドとして含まれることが可能となり、それによってスクリーニング速度を高めることが可能になる。したがって、細胞において効果が観察される場合には、ビーズ上に存在するRNA分子をスクリーニングしてどの配列が効果の原因であったかを調べることができる。さらに、ナノ粒子はRNAの送達系を提供するので、先行技術において必要とされるトランスフェクション剤の使用を避けることができる。トランスフェクション剤はそれ自体、試験される細胞においてタンパク質発現に変化を引き起こし得るので、これは望ましい利点である。したがって、さらなる態様では、本発明のナノ粒子を、例えば、Nature Reviews、第3巻、2004年4月、318〜329頁に記載されるように、機能ゲノミクスにおけるツールとして使用できる。ナノ粒子は、in vivoで遺伝子機能を調べるために、また、ゲノム規模でのスクリーニングのためのツールとして、粒子当たり3種以上、6種以上、11種以上または21種以上または101種以上のRNA配列を有し得る。
さらなる態様では、本発明は、本明細書に記載されるナノ粒子のエアゾール中での使用を提供する。これは小さなサイズのナノ粒子によって可能になる。イメージングおよび/または治療用途のために、例えば、肺に影響を及ぼす状態の治療において、エアゾール組成物を用いて、RNAリガンドを、特に肺に送達できる。

Her-2/neu発癌遺伝子およびそのコードされる産物pl85Her-2/neuは上皮成長因子受容体チロシンキナーゼに属する(Bargmannら、1986)。HER受容体ファミリーは、4種の膜貫通型チロシンキナーゼ:EGFR(Her-1またはerbB-1としても知られる)、erbB-2(Her-2)、erbB-3(Her-3)およびerbB-4(Her-4)からなる。Her-2/neuシグナル伝達経路は、細胞増殖および分化、悪性形質転換および化学療法薬に対する耐性において重要な役割を果たすことがわかっている(Yarden & Sliwkowski、2001)。Her-2/neuは、ヒト乳癌および卵巣癌の約1/3の場合において過剰発現され、その過剰発現が悪い予後と関連している(Berchuckら、1990)。

1.細胞株
ATCCから得たSK-BR-3ヒト乳腺腺癌(カタログ番号HTB-30)。NCI-フレデリック癌DCTD腫瘍/細胞株貯蔵所(NCI-Frederick Cancer DCTD Tumor/cell line repository)(バイアル0502296)から得たOVCAR-3ヒト腹水腺癌。
2.siRNA保存溶液
選択したHer-2/neu DNA標的配列はAAGCCTCACA GAGATCTTGAAであった。
センスsiRNAは配列r(GCCUCACAGAGAUCUUGAA)d(TT)3Thssを有していた。(K-塩の分子量7416.25)また、アンチセンス配列r(UUCAAGAUCUCUGUGAGGC)d(TT)3Thss(K-塩の分子量7409.57)をQiagenから入手した。
対照(非サイレンシング)siRNA二本鎖配列はQiagenから得(カタログ番号1022076)、センスはr(UUC UCC GAA CGU GUC ACG U)d(TT)であり、アンチセンスはr(ACG UGA CAC GUU CGG AGA A)d(TT)であり、アニーリングしたK-塩の分子量は14839.5であった。
1mlの滅菌バッファー(100mM酢酸カリウム、30mM Hepes-KOH、2mM酢酸マグネシウムpH7.4)に、1本のセンスsiRNA試験管の内容物(296.65μg)を溶解して40μMの保存液を作製する。1μl当たり0.297μgのsiRNAを含む。
1mlの滅菌バッファー(100mM酢酸カリウム、30mM Hepes-KOH、2mM酢酸マグネシウムpH7.4)に、1本のアンチセンスsiRNA試験管の内容物(296.38μg)を溶解して40μMの保存液を作製した。1μl当たり0.296μgのsiRNAを含んでいた。
アニーリングするために、各RNAオリゴ溶液30μlを15μlの5×アニーリングバッファーと混合した。最終バッファー濃度は、DEPC処理した水に溶かした、50mM Tris、pH7.5〜8.0、100mM NaClとした。最終容積は75μlとし、siRNA二本鎖の最終濃度は16μMとした。
溶液を90〜95℃の水浴中で1分間インキュベートし、室温(すなわち、30℃より低い)に放冷した。この試験管を短時間遠心分離してすべての液体を試験管の底に集めた。室温に徐々に冷却すると45〜60分かかった。得られた溶液は、使用する準備ができるまで-20℃で保存し、凍結および解凍の繰り返しに対して耐久性があった。
3.siRNA Nanogold保存溶液
一般法
HAuCl4(99.999%)およびNaBH4はAldrich Chemical社から購入した。2-チオエチル-β-D-グルコピラノシドはcurラボラトリー(cur laboratory)において標準的な手順を用いて合成した。すべての実験および溶液には、DEPC(ジエチルピロカーボネート)で処理したNanopure水(18.1mΩ)を用いた。すべてのエッペンドルフ、スパチュラおよびバイアルはRNaseフリーであった。アニーリングしている二本鎖siRNAはQiagen-Xeragon社から購入した。明細は以下の通りであった。

TRISバッファー100mM、pH7.7(250μL)中、2-チオエチル-β-D-グルコピラノシド(0.9mg、3.75μmol)およびsiRNA(0.148mg、0.01μmol)の溶液に、HAuCl4水溶液(22μL、0.025M)を加えた。次いで、NaBH4の1N水溶液(30μL)を数回で加え、迅速に振盪した。形成された褐色の懸濁液をさらに1時間、4℃で振盪した。この懸濁液を遠心分離濾過によって精製した(AMICON分子量10000、30分、4℃、14000rpm)。このプロセスを2回反復し、125μLのTRISバッファーで洗浄した。AMICONフィルター中の残渣を250μLのTRISバッファーに溶解し、凍結乾燥すると、4mgのRNA-Au-Glcナノ粒子が得られた(1mLの水に固体を再懸濁することによって、20mM TRISバッファー中のRNAの6±1μM溶液が得られるはずである)。濾液をAMICON(分子量3000、4℃、14000rpm)を用いて脱塩し、凍結乾燥した。残渣の重量は<50μgであった。図1に示される透過型電子顕微鏡写真(TEM)は、粒子の平均サイズが2.8nmであったことを示し、図4に表されるように、平均807個の金原子/粒子、siRNAおよび100分子のグルコース誘導体を有し、およその分子量は>160,000である。
RNA-Au-Glcナノ粒子、Glc-Auナノ粒子およびRNAオリゴヌクレオチドとグルコース誘導体とを含有すると推定されるRNA-Au-Glcナノ粒子の洗浄の残渣を、30μLの水に各々溶解した。これらの溶液のアリコート(1μL)を、エチジウムブロマイド(EtBr)の水溶液(1μL、0.1%v/v)と混合した。UVランプ下で蛍光を観察したところ(図2参照)、このように調製したナノ粒子にはsiRNAが組み込まれているが(図2b、試験管1)、グルコースしか含有していないナノ粒子は蛍光をまったく示さない(図2b、試験管2)ということが実証された。
148μg siRNAから作製した4mgのsiRNA/nanogold複合体を水1mlに溶解し、20mMtris中、6±1μMの保存溶液を得た。溶液1μl当たり0.078μgのsiRNA相当物を含んでいた。
1.トランスフェクションの24時間前に6×104個細胞を24ウェルプレートにピペットで入れ、適当な培養培地で容量を0.5mlとした。
2.この細胞をおよそ24時間かけて50〜80%コンフルエンシーに到達させた。
3.培養培地を除去し、300μlの新鮮培地/ウェルで置き換えた。
1.3.3μl(または12.8μlのnanogold複合体)の適当な二本鎖siRNA保存液を24ウェルプレートに、細胞を含むものに対応して分配した。
2.各ウェルに、96.7μl(またはnanogold複合体については87.2μl)の適当な培養培地を加え、上下に5回ピペッティングすることによって十分に混合した。
3.各ウェル(nanogold複合体のウェル以外)に、6μlのRNAiFectを加え、上下に5回ピペッティングすることによって十分に混合した。
4.この溶液を室温で10〜15分間インキュベートし、複合体を形成させた。
5.300μlの細胞培地中の細胞を100μlの適当なトランスフェクション複合体で覆った。
6.このプレートを穏やかに揺らして回旋をさけながら混合した。
7.このプレートをCO2インキュベーター中、37℃で48〜72時間インキュベートした。
8.培地を除去し、氷冷PBSで細胞を3回洗浄した。
9.細胞を溶解し、溶解物のタンパク質含量を求めた。
10.SDS-PAGEと、続いてCell Signalling Technologyから得たHer2/ErbB2ポリクローナルウサギ抗体(カタログ番号2242)を用いるウェスタンブロッティングによってタンパク質を分離した。
11.ブロットを抗ウサギIgG-HRP複合体で処理し、続いてECLを発現させた。
1μgのsiRNA/ウェルを用いた予備的観察結果を図3aおよび3bに示す。siRNA-金ナノ粒子をRNAiFectamineを用いずに細胞に加えた。SKBR3細胞は、80%コンフルエンシーに達するのがOVCAR細胞よりも遅かった。SKBR3の結果は、トランスフェクションの48時間後の溶解物から得たものであり、OVCARの結果はトランスフェクションの72時間後の溶解物から得たものであった。ナノ粒子の概略図を図4に示す。
細胞をsiRNA単独を用いて、および金糖ナノ粒子とコンジュゲートしているsiRNAを用いてトランスフェクトした。図5は、ナノ粒子がコンジュゲートしているsiRNAは有効であり、毒性作用がなかったということを示す。細胞数に対する用量依存効果が見られ、このことは、siRNAナノ粒子が細胞増殖を増加させたということを示す。
OVCAR細胞を、siRNA-ナノ粒子を用いて、siRNAを用いる細胞のトランスフェクションには通常必要とされるトランスフェクション試薬を用いて、または用いずにトランスフェクトした。図6は、siRNA-ナノ粒子の細胞への侵入にはトランスフェクション試薬が必要でなかったことを示す。この結果は、siRNAナノ粒子はトランスフェクション試薬の不在下でさえも細胞内に効率的に送達されたことを示し、実際には、送達はトランスフェクション試薬なしでより効率的であると思われた。細胞数に対する効果の用量依存性は、siRNA-ナノ粒子に対する真の応答を示す。

Claims (31)
- in vivoにて標的遺伝子をダウンレギュレートするための医薬の製造におけるナノ粒子の使用であって、該ナノ粒子は金原子を含有するコアを含み、該コアが複数のリガンドと共有結合しており、該リガンドがsiRNAリガンド、miRNAリガンドおよびshRNAリガンドからなる群から選択されるRNAリガンドを含み、該ナノ粒子がチオールリンカー基を介してRNAリガンドと共有結合しており、該RNAリガンドが該標的遺伝子に特異的な配列を有する、上記使用。
- 標的遺伝子をダウンレギュレートするための医薬が、癌、ウイルス感染または眼の黄斑変性症を治療するためのものである、請求項1に記載の使用。
- 癌が乳癌であるか、またはウイルスがHIV、肝炎もしくはインフルエンザである、請求項2に記載の使用。
- ナノ粒子が炭水化物基を含むリガンドをさらに含む、請求項1〜3のいずれか一項に記載の使用。
- RNAリガンドの長さが17リボヌクレオチドと30リボヌクレオチドの間である、請求項1〜4のいずれか一項に記載の使用。
- リガンドがsiRNAリガンドであり、2リボヌクレオチドの3'オーバーハングを含む、請求項1〜5のいずれか一項に記載の使用。
- RNA分子の第1センスまたはアンチセンス鎖が、その5'および/または3'末端を介してナノ粒子コアと共有結合している、請求項1〜6のいずれか一項に記載の使用。
- 第1鎖と相補的であるRNA分子の第2鎖はRNA分子の第1鎖とアニーリングする、請求項7に記載の使用。
- 第2のRNA鎖が、その5'および/または3'末端を介してナノ粒子コアと共有結合している、請求項8に記載の使用。
- RNA分子の第1および第2鎖はナノ粒子コアと個別に結合しているが、後に一緒になってアニーリングする、請求項8または9に記載の使用。
- RNAリガンドがmiRNAリガンドであり、ヘアピンを含む、請求項1〜5または7〜10のいずれか一項に記載の使用。
- RNAリガンドがshRNAリガンドである、請求項1〜5または7〜10のいずれか一項に記載の使用。

- RNAリガンドがHer2遺伝子配列に基づくものである、請求項1〜14のいずれか一項に記載の使用。
- ナノ粒子が標識を含む、請求項1〜15のいずれか一項に記載の使用。
- 標識が蛍光基、放射性核種、磁性標識、色素、NMR活性原子または表面プラズモン共鳴を用いて検出できる原子である、請求項16に記載の使用。
- (a)蛍光基がフルオレセイン、ローダミンまたはテトラメチルローダミン、テキサスレッド、Cy3もしくはCy5であるか;または
(b)放射性核種が99mTc、32P、33P、57Co、59Fe3+、67Cu2+、67Ga3+、68Ge、82Sr、99Mo、103Pd、111In3+、125I、131I、137Cs、153Gd、153Sm、158Au、186Re、201Tl+、39Y3+、71Lu3+もしくは24Cr2+であるか;または
(c)磁性標識がMn+2、Gd+3、Eu+2、Cu+2、V+2、Co+2、Ni+2、Fe+2、Fe+3もしくはランタニド+3を含む常磁性基であるか;または
(d)NMR活性原子がMn+2、Gd+3、Eu+2、Cu+2、V+2、Co+2、Ni+2、Fe+2、Fe+3もしくはランタニド+3である、
請求項17に記載の使用。 - ナノ粒子が水溶性である、請求項1〜18のいずれか一項に記載の使用。
- (a)ナノ粒子のコアの平均直径が0.5と10nmの間であるか;または
(b)ナノ粒子のコアの平均直径が1と5nmの間であるか;または
(c)リガンドを含むナノ粒子の平均直径が10と30nmの間である、
請求項1〜19のいずれか一項に記載の使用。 - ナノ粒子が複数の異なる種類のRNAリガンドを含む、請求項1〜20のいずれか一項に記載の使用。
- (a)リガンドがペプチド、タンパク質ドメイン、核酸セグメントもしくは炭水化物基をさらに含むか;または
(b)リガンドが多糖、オリゴ糖もしくは単糖基をさらに含むか;または
(c)リガンドが糖ナノ複合体をさらに含むか;または
(d)リガンドが糖脂質もしくは糖タンパク質を含む糖ナノ複合体をさらに含む、
請求項1〜21のいずれか一項に記載の使用。 - 標的遺伝子をダウンレギュレートするためのin vitro方法であって、該遺伝子を含む細胞を、請求項1〜22のいずれか一項に定義されるナノ粒子と接触させることを含む、上記方法。
- 標的遺伝子の一時的なノックアウトをもたらす、請求項23に記載の方法。
- 同一のナノ粒子にコンジュゲートしているか、または少なくとも2種の異なる種類のナノ粒子の組成物中に存在している、少なくとも2種の異なるRNAリガンドを有するナノ粒子を用いて、経路中少なくとも2種の遺伝子の発現をダウンレギュレートする、請求項23または24に記載の方法。
- 経路が炎症経路、抗ウイルス経路、癌におけるシグナル伝達経路、転移経路または代謝経路である、請求項25に記載の方法。
- RNAリガンドが遺伝子ファミリーの保存されたドメインに基づいており、遺伝子ファミリーの複数のメンバーの発現をダウンレギュレートする、請求項23〜26のいずれか一項に記載の方法。
- サンプルまたは細胞中のRNAを検出またはイメージングするためのin vitro方法における、請求項1〜22のいずれか一項に定義されるナノ粒子の使用。
- 請求項1〜23のいずれか一項に定義されるナノ粒子を用いる、サンプルまたは細胞中のmRNAを検出および/またはイメージングするin vitro方法であって、ナノ粒子上に存在するRNAリガンドが標的mRNAと相互作用し、ナノ粒子-RNA-mRNA複合体を検出できる条件下で、ナノ粒子を標的mRNAを含有するサンプルまたは細胞と接触させることを含む方法。
- 複合体を検出するステップがナノ粒子の固有の特性を用いるか、またはナノ粒子と関連している標識を検出することによる、請求項29に記載の方法。
- 肺に送達するための医薬を調製するための請求項1〜23のいずれか一項に定義されるナノ粒子を含むエアゾール組成物の使用であって、該ナノ粒子がイメージングのための標識を含むか、または哺乳類の肺に影響を及ぼす状態の治療のためのものである使用。
