Description
The present invention relates to a cell culture container and a cell culture method using the same.
Cell death is roughly divided into apoptosis and necrosis. Apoptosis is a type of death of the cells that make up the body of a multicellular organism, meaning controlled and regulated cell suicide that is actively triggered to keep an individual in better condition To do. In vivo, most cancerous cells (and other cells with abnormalities in the interior) continue to be removed by apoptosis, which is known to prevent most tumor growth. ing. In addition, in the development process of organisms, cell death occurs at a predetermined time and place (programmed cell death), which works as a driving force for changes in the morphology of the organism. This cell death is also the mechanism of apoptosis. Caused by. For example, the disappearance of the tail when transformed from a tadpole to a frog is due to apoptosis. In nematodes all apoptosis occurring in development is described. It is also known to play an important role in the immune system, such as the removal of cells that react with self-antigens.
On the other hand, cell death caused by deterioration of the environment inside and outside the cell due to poor blood circulation, trauma, etc. is called necrosis or necrosis and is distinguished from this. The origin of Apoptosis comes from the Greek words “apo- (away)” and “ptosis (down)”, meaning “(dead leaves fall from trees)”.
Necrosis is the death of a part of a living tissue. Unlike normal death, only the cells that make up part of the body die. Caused by infection, physical destruction, chemical damage, decreased blood flow, etc. Those caused by decreased blood flow are called infarctions. Even if the cells are dead, normal cells such as blood cells, skin, and mucosal epithelium of the digestive tract, and tissues that are replenished one after another and do not leave functional disorders or histological abnormalities are not called necrosis. The necrotic tissue is finally removed by the living body's immune system, and the defective part is compensated by the regeneration or fibrosis of the original tissue.
In order to examine and test such cells, it is usually necessary to culture cells isolated from tissues. Techniques for testing and examining cells isolated from tissues are indispensable in biotechnology-related fields. It is widely used for diagnosing diseases and pathological conditions, searching for new drugs and determining their efficacy, animal testing, plant testing, and testing for environmental pollutants. The isolated cells may be used immediately for testing, but in many cases, the cells are cultured in a culture dish or a test tube by a cell culture method. Various tests are performed in this culture system.
In these assays, a uniform culture system is usually set up, and the effect is observed by changing the amount and concentration of the drug to be evaluated. Therefore, the culture container used for the culture is a uniform one. As this culture container, a culture dish (cell culture dish) is generally used. Commonly used culture dishes include petri dishes or 6-well plates, 12-well, 48-well, and 96-well plates (Patent Document 1). Due to the recent trend toward micro-volumes, 384-well plates consisting of a large number of culture dishes with a smaller diameter have begun to be used. The bottom of these culture dishes has a flat plate shape.
As described above, in the medical and biotechnology fields, cell culture dishes (petri dishes) and plates that are currently used for culture tests and tissue culture use the flat plate portion on the bottom as the culture surface. .
However, when tissue cells are cultured in a flat plate-shaped culture dish, the cells grow thin and have a non-directional shape, and the shape and function that are originally seen in vivo cannot be exhibited. It was. For example, there has been a problem that even if adipocyte precursor cells are cultured in a plate-shaped culture dish or culture vessel and differentiation is induced, they do not differentiate into the shape of the target adipocyte. In addition, hepatocytes have a problem of not forming spheroid-like cell clusters.
Therefore, since it is difficult to efficiently target and proliferate and differentiate cells in the flat form, methods of immobilizing cytokines and extracellular matrix on the flat surface and methods using special devices have been attempted (patents). Literature 2, Patent Literature 3).
However, the method of immobilizing cell growth factors and extracellular matrix on a flat plate surface increases efficiency only in the initial stage, but it cannot be maintained, the immobilization method is complicated and cannot be stably produced, and the cost is further increased. There is a problem that takes. In addition, the method using a special device has different results depending on the cell type, and there are problems that stable efficiency cannot be expected, that a large device is used, the operability is poor, and the cost is high.
Further, in cell culture dishes (petri dishes), culture tests using plates, and tissue culture, changes in pH due to carbon dioxide gas discharged from cultured cells are known to have a significant effect on the activity of cultured cells. For this reason, in long-term culture, researchers maintain the activity of cultured cells by periodically changing the culture solution (for example, every two days). Furthermore, in the latest research example, a bioreactor capable of always circulating a fresh culture solution is produced, and a new artificial organ replacement model is proposed.
However, factors for suppressing cell death of reproducible tissues such as necrosis of tissues that do not regenerate like nerve cells, heart muscle, and skeletal muscle, and mucosal epithelium of skin and digestive tract have been clarified. In addition, the current situation is that cell death cannot be suppressed only by exchanging a fresh culture solution.
In particular, rat cardiomyocytes, nerve cells, corneal cells, etc., which are difficult to cultivate, have a survival rate with respect to the number of cells initially dispersed (arranged) even if the fresh culture medium is replaced. The current situation is 20 to 40%.
As described above, in the conventional cell culture container, the cells have a thin and elongated shape, and the shape and function that are originally expressed in the living body are not exhibited. When tissue cells are cultured on a cell culture dish (petri dish or well plate) that is currently commercially available, the cultured cells are thinly elongated and have a non-directional shape. As a method for confirming the activity of cells, researchers use biological sensors to measure changes in pH due to waste product discharge and carbon dioxide release, and data measured from cells cultured in a culture dish. However, at present, the values indicating the data of living tissue cannot be reproduced in the culture dish. Therefore, it is determined that the cultured cells do not exhibit the functions they have in vivo when cultured on a commercially available cell culture dish.
Therefore, by forming a fine container pattern suitable for tissue cell growth on the culture dish, culturing cells in the fine container pattern, and growing the cells three-dimensionally, the function that had in vivo Research has started to make it appear. A further challenge is to efficiently proliferate and / or differentiate the cells.
According to the culture method using the cell culture container according to the present invention, by providing a fine uneven pattern, the same three-dimensional growth as in a living body is possible, and in addition, by the three-dimensional compartment formed by the fine unevenness. Since physiologically active substances such as cell growth factors and / or differentiation-inducing factors produced by the cells themselves can be cultured at a concentration similar to that in the living body, it is possible to mimic the environment surrounding the cells in the living body. The fine concavo-convex pattern has, for example, a plurality of side walls, a plurality of spaces for arranging cultured cells formed by these side walls, and further, by providing openings on the side walls, the plurality of spaces This can be realized by forming a connected connection structure. By having a plurality of side walls, a plurality of spaces are produced, and the size of the space is set according to the required application.
It is presumed that the present invention can be applied to various culture systems by setting the dimensions of the side wall, space, and opening according to the cell type to be cultured. With an opening part, the structure for the space (concave part) formed by the side wall to communicate is made into an opening part.
The size of the space formed by the side wall needs to be in an optimum range for the purpose of culturing cells. If the space formed by the side walls is too large, the cells will not be elongated and show a three-dimensional structure, as in the case of culturing on a flat plate, and if the space is too small, cells cannot enter the space. Therefore, it is desirable that the size of the space be in a range that can accommodate a single or a plurality of cells depending on the cell type to be cultured.
As a result of diligent research, the present inventors have found that the survival rate of cultured cells can be dramatically improved by preventing the diffusion of substances produced by cells in a specific micropattern shape. It came.
Cells produce substances such as fibronectin, laminin, collagen, and elastin during the culture process. The change in pH due to the discharge of carbon dioxide gas causes a decrease in the activity of cultured cells, but these produced substances are used as a substrate material, and are essential for cell surface adhesion, differentiation and proliferation. It is also a material.
The present inventors have succeeded in preventing the diffusion of cell product and increasing the cell viability by forming a plurality of micropatterns in the cell culture container and culturing the cells under the side wall of the pattern. did.
The purpose is to prevent the diffusion of cell products and increase the cell viability, and in the future, it is expected to increase the productivity of cell cultures. Therefore, the problem with the micropattern in the present invention is how to secure the length of the lower portion of the side wall within the culture area.
Since the minimum size of a single cell is 4-5 micrometers, the width | variety of the lower part between side walls is desirable to be 2 micrometers or more, for example.
The width of the side wall is preferably as narrow as possible in order to ensure the length of the lower part of the side wall within the culture area, but is preferably 1 μm or more from the viewpoint of being reproducible from an industrial technical point of view.
The height of the side wall is preferably 1 μm or more, for example, from the viewpoint of suppressing the diffusion of the produced substance.
The ratio of the width and height of the protrusions in each step is preferably as high as possible for the purpose of preventing the diffusion of the product, but from the viewpoint of reproducibility in terms of industrial technology, the ratio of height / width is 1 It is desirable to select from / 1 to 20/1.
Furthermore, the cell culture container according to the present invention is also effective for a bioreactor as an alternative to an artificial organ. When a space for refluxing the culture solution is formed by stacking the substrates, a bridge higher than the side wall for preventing the diffusion of the product of cultured cells is provided so as to be perpendicular to the side wall, and the culture solution is placed on the side wall. By flowing in the direction perpendicular, the PH change due to the release of carbon dioxide from the cultured cells can be prevented, and the product produced by the cultured cells can be diffused.

An uneven pattern is formed in the cell culture container. The concavo-convex pattern is formed in two steps. Therefore, the two-step
The 1st
It is preferable that the uneven pattern has two or more steps and the
The size (width, depth) of the
Moreover, it is preferable that the two-
Moreover, it is preferable that the height of the recessed
Moreover, it is preferable that the height of the 1st
The width of the
The height of the
The ratio between the width and the height of the second
Furthermore, it is preferable to perform a surface treatment on the region where the uneven pattern is provided. That is, it is preferable to perform surface treatment on the upper surface and side surfaces of the
The concavo-convex pattern on the cell culture vessel can be formed by a semiconductor processing technique using silicon as a material or a forming technique using a Ni stamper (master) by a photolithographic method. For example, in the case of a semiconductor processing technique using silicon as a material, a resist is applied on a silicon substrate, and the resist is exposed and developed to pattern the resist. By etching the silicon from above the resist, the silicon substrate is patterned to form an uneven pattern. Then, by removing the resist, a silicon cell culture vessel having a desired uneven pattern is completed. Of course, the substrate for the cell culture container is not limited to a silicon substrate, and may be a glass substrate or a resin substrate. As described above, by using the semiconductor processing technique, the uneven pattern can be formed with high accuracy.
In the case of a molding technique using a Ni stamper, first, a first resist layer is applied and exposed on a substrate, and then a second resist layer is applied and exposed thereon. An uneven pattern can be formed on the substrate by batch original images of the first resist layer and the second resist. Then, a conductive film is attached to the substrate on which the concavo-convex pattern is formed by vapor deposition or sputtering. Then, a metal structure is formed by depositing metal on the conductive thin film by plating. The stamper can be formed by peeling the metal structure from the substrate. By molding using this stamper, a highly accurate cell culture solution can be produced with high productivity. In this case, the cell culture container is formed of, for example, a resin material. Of course, a structure for forming a metal structure may be formed using a silicon substrate, a glass substrate, or the like. Furthermore, the manufacturing method of a cell culture container is not limited to said method. Specifically, the
The
In addition, by culturing cells in the space formed by the fine uneven pattern, the three-dimensional shape originally expressed in the living body can be shown, and the cell growth factor and differentiation produced by the cell itself by the three-dimensional division of the fine unevenness. The pericellular concentration of physiologically active substances such as inducers is improved. Therefore, it is possible to provide a cell culture container that can efficiently proliferate and / or differentiate. Furthermore, it is possible to provide a cell culture container that is inexpensive and easy to observe by being made of a transparent resin material.
In addition, the cell culture container having two concavo-convex patterns is also effective for a bioreactor that substitutes for an artificial organ.

Thus, in the
