Description
Gタンパク質共役受容体(GPCR)は、ホルモンまたはリガンドの結合を細胞内シグナルに翻訳する細胞表面タンパク質である。GPCRは、すべての動物および昆虫および植物において見出されている。GPCRのシグナル伝達は、光伝達、嗅覚、神経伝達、血管の正常な状態、心拍出量、消化、痛み、ならびに水分平衡および電解質平衡を含む様々な生理学的機能を調節することにおいて重要な役割を果たしている。GPCRは様々な生理学的機能に関与しているが、多数の共通する構造的特徴をともに有する。GPCRは、交互に並ぶ細胞内ループおよび細胞外ループによって架橋された7個の膜ドメインと、様々な長さの細胞内カルボキシル末端テールとを含有する。
本発明は、一般には、1つ以上のGPCRリガンドを含有する不均一な溶液に存在するGPCRリガンドを検出することに関連する。例えば、そのような溶液には、血清、血液、別の生物学的サンプル、または環境サンプルを挙げることができる。
多数の疾患状態がGPCRリガンド濃度の異常な調節と関連している。本発明は、GPCRリガンドの存在、非存在、濃度、または濃度変化を検出する方法を提供する。そのような方法を使用して、本発明は、疾患または疾患原因状態を診断する方法を提供する。
本発明の別の実施形態において、軍隊および市民社会にとって注目される数多くの化学的/生物学的薬剤を、記載されたセンサーによって容易に検知することができる。本発明は、生物学的薬剤、毒素、神経毒および神経ガスなどを検出するために使用することができる。生物学的に関連する分子を非常に高感度で迅速かつ正確に検出および定量することができることは、医療技術、国家安全保障、公衆安全、環境安全、ならびに市民および軍隊の医療診断のために重要な問題である。
本発明は、サンプルにおけるGPCR結合化合物の存在を分析する非常に特異的かつ高感度で、一般化された定量的方法を提供する。
本発明の別の特徴は、本明細書中に開示されるように、バイオセンサーを得るために、GPCRおよび/またはアレスチンをコードするDNA配列を細胞において発現させることである。当分野で周知のように、DNA配列は、DNA配列を適切な発現ベクターにおける発現制御配列に機能的に連結し、そのような発現ベクターを用いて、適切な単細胞宿主を形質転換することによって発現させることができる。
本発明のアッセイ方法において使用される細胞は、アレスチンタンパク質および検出可能な分子のコンジュゲート体、GPCRおよび検出可能な分子のコンジュゲート体、GPCR/アレスチン複合体の任意のメンバーおよび検出可能な分子とコンジュゲート体、検出可能な分子と、GPCR/アレスチン複合体の任意のメンバーと相互作用する分子とのコンジュゲート体などを含むことができる。検出可能な分子は、そのような分子自体だけでなく、検出可能な分子と相互作用する分子の検出を可能にする。
本発明の細胞は、少なくとも1つのGPCRと、アレスチンとを発現する。この場合、これらの分子の少なくとも1つが検出可能に標識される。本発明において有用な細胞には、真核生物細胞および原核生物細胞が含まれ、これらには、細菌細胞、酵母細胞、菌類細胞、昆虫細胞、線虫細胞、植物細胞および動物細胞が含まれるが、これらに限定されない。好適な動物細胞には、HEK細胞、HeLa細胞、COS細胞、U2OS細胞、および様々な霊長類哺乳動物細胞が含まれるが、これらに限定されない。アレスチンおよび検出可能な分子のコンジュゲート体を、その組織全体で、または特定の器官タイプもしくは組織タイプにおいて発現する動物モデルもまた本発明において使用することができる。
検出可能に標識されたアレスチンの細胞内の位置、検出可能に標識されたGPCRの細胞内の位置、あるいは、検出可能に標識されたアレスチン、またはGPCR/アレスチン複合体の他のメンバーとGPCRまたは任意の他の細胞構造体との相互作用(これらには、例えば、アレスチンまたはGPCRの細胞膜における濃縮、エンドソームにおけるGPCRとのアレスチンの共局在化、およびクラスリン被覆小孔におけるアレスチンまたはGPCRの濃縮などが含まれる)を検出する方法は、使用される検出可能な分子(1つまたは複数)に依存して変化する。
本発明には、試験サンプルを分析するための試験キットが含まれる。非常に好ましくは、試験キットは、生物学的サンプルまたは環境サンプルにおけるGPCRリガンドの濃度を測定するために有用である。さらにより好ましくは、試験キットは、GPCRおよびアレスチンを発現する宿主細胞、そしてサンプルにおけるリガンド濃度を測定する方法を含む。
本発明は、限定することを意図しない下記の例示的な実施例によってさらに説明される。
ヒト胚腎臓細胞(HEK−239)をアメリカン・タイプ・カルチャー・コレクション(ATCC、Manassas、VA)から入手した。培地、ウシ胎児血清(FBS)および抗生物質をMedia Tech(Hemdon、VA)およびGibco Invitrogen Corp(Carlsbad、California)から購入した。96ウェルガラスプレートをWhatman(Clifton、New Jersey)から、そして結合樹脂をBioRad Laboratories(Hercules、CA)から得た。CCK−A受容体アンタゴニストのデバゼピドおよび特異的なCCK−B受容体アンタゴニストを使用した。ペンタガストリンをSigmaから得た。
hは、輸送された受容体の割合である。比Nb/Naは、輸送後のβアレスチン2−GFPにより占有される容積の変化の大きさを表す。次に、閾値の値を超える典型的な個々の画素に対するシグナル対ノイズ比は下記のように計算することができる。

フローサイトメトリーおよびアレスチン−GFP輸送による細胞におけるアレスチン−GFP発現およびCCK−B受容体発現の均一性の決定
アレスチン−GFP再分配の定量的分析を単純化するために、同一の様式でリガンドに応答する細胞を樹立した。細胞集団全体での受容体発現およびアレスチン発現の大きな均一度を達成するために、CCK−B受容体およびアレスチン−GFPを永続的に発現するHEK−239細胞株を樹立した。本研究で使用されたクローン(クローンA)におけるアレスチン発現の均一性の程度をフローサイトメトリーによって測定し(図4A)、蛍光によって確認した(図4B、左パネル)。アレスチン−GFPを再分配することによって合成ヒトガストリン−17ペプチド(hG17)の存在に応答する細胞の一般的な能力が図4B(右パネル)に示される。
クローンAは、ガストリン受容体アゴニストである[3H]−CCK8およびhG17の結合を測定し、hG17媒介による2次メッセンジャー応答を測定することによってさらに特徴づけが行われた。[3H]−CCK8を用いた飽和結合により、細胞は(14±1)pmolのCCK−B受容体/mg細胞タンパク質を発現し、そして[3H]−CCK8に対する親和性が(9.0±1.6)nMであることが示された(図5A)。イノシトールリン酸(IP3)産生のヒトガストリン−17による刺激はEC50=(3.2±0.7)nMをもたらした(図5B)。図5Bにおける挿入図は、hG17による[3H]−CCK8の競合的置換を示している。EC50は(28±5)nMであり、CCK−B受容体に対するhG17のKdは(17±3.5)nMであった。
hG17の増大する濃度を使用して、細胞質のアレスチン−GFPの時間に依存した減少を、アレスチン再分配の薬理学がIP3産生の薬理学と相関するかどうかを明らかにするために測定した。96ウェルプレートにおいて増殖させたリガンド処理細胞の連続した蛍光像を、共焦点顕微鏡観察によって、5分間、30秒間隔で得て、細胞質の蛍光の減少を測定することによって分析した。増大する濃度のhG17に対するアレスチン再分配の時間および用量の依存性が図6Aにプロットされる。このデータから、用量応答曲線が計算され、輸送に対して4.2±1.5nMのEC50が得られた(図6B)。これはIP3の結果と一致する。
実施例4のこれらの測定のための方法論が図7に示される。蛍光強度を単に測定することによって得られた、バックグラウンドを越えるシグナルは、120倍の増大が観測された図7Bに示されるように、輸送されたアレスチンが小さい容積に濃縮されるために極めて多かった。
血清中ガストリンレベルの上昇が見られない臨床的な高ガストリン血症が数多く報告されている。その結果、血清中ガストリンのRIA検出では、免疫学的に検出できないガストリン受容体アゴニストを有する患者の一団が見逃され得る。従って、CCK−B受容体アゴニストのペンタガストリン(これは従来のRIAによって検出できない)の輸送に対する影響が評価された。図8に例示されるように、増大する量のペンタガストリンに対するバイオセンサーの応答は、輸送に対するEC50が2.4±1.9nMである用量応答をもたらした。これは、CCK−B受容体に結合するペンタガストリンに対する1nMの報告されたKd、およびポリホスホイノシチド回転に対する3.9nMと類似している。
1時間における用量応答
バイオセンサーが様々な合成されたガストリンイソ型に応答し得ることにより、ヒト血清に含有されるガストリンの多数の生物活性な形態を測定するその潜在的能力が示唆される。図9Aの上段パネルには、1時間のインキュベーションの後における患者血清サンプル(RIAにより5000pg/mlの報告された高ガストリン血症、3,400pg/ml〜6,600pg/mlの範囲)に対する細胞応答が示される。アレスチン−GFPが形質膜および小胞内において認められた。輸送されたアレスチン−GFPの量が、図6Cの像により表されるデータから作製されたhG17標準曲線と比較された。hG17用量応答曲線のEC50は(0.80±0.25)nM(図6Bおよび図6C)であり、患者の生物活性なガストリンの血清中濃度が0.63nM±0.16nMであると決定された(図9B、矢を参照のこと)。
〔実施例7〕
バソプレッシンのカルボキシルテールとコンジュゲート化されたヒトムスカリン性受容体タイプ1を発現するHEK−239細胞をマイクロモル濃度のアセチルコリンにさらした。アレスチン−GFPの輸送を測定した。アレスチン−GFPが、図10に示されるように、膜の端部および小胞内において観測された。
HEK−239細胞を、10%ウシ胎児血清(FBS)を含有する最少必須培地においてインキュベーションした。細胞を、アレスチン−GFP、およびバソプレッシンのカルボキシルテールとコンジュゲート化されたヒトムスカリン性受容体タイプ1の発現を誘導するためのcDNAで一過性トランスフェクションした。10%FBSが存在するとき、アレスチン−GFPの輸送は、細胞を10〜100マイクロモル濃度の範囲の濃度のアセチルコリンに30分までさらした後では観測されなかった。しかし、ミリモル濃度の量のアセチルコリンはアレスチン−GFPの輸送をもたらした。細胞が血清の非存在下で10〜20マイクロモル濃度のアセチルコリンにさらされたとき、アレスチン−GFPが細胞膜に容易に輸送された。アセチルコリンエステラーゼは、アセチルコリンを分解することが知られている酵素であり、FBSを含む血清の一般的な成分である。アセチルコリンエステラーゼは、ムスカリン性受容体のリガンドであるアセチルコリンを分解し、それにより、アセチルコリンにより誘導されるムスカリン性受容体およびアレスチンの内在化を妨げた。アセチルコリンエステラーゼの存在下における非常に多量のアセチルコリン(ミリモル濃度)はアレスチン内在化の一時的な量のみをもたらすことができた。対照的に、はるかに小さい濃度のアセチルコリン(10〜20マクロモル濃度)は、血清が培地に存在しないときには頑強な応答をもたらすことができた。このことは、アレスチンの輸送が、アセチルコリンエステラーゼの阻害剤について血清をアッセイするために使用され得ることを示唆している。これは、有機ホスファート化合物などの強力な阻害剤が、アレスチン−GFP、およびアセチルコリンにさらされたムスカリン性受容体を含有する試験細胞の環境から血清およびすべてのその成分を完全に除くことに対して類似する効果をもたらすからである。
推定される阻害剤を含有するサンプルが適切な溶媒に抽出されるが、場合により、これは水性の緩衝液であり得る。抽出物は、アセチルコリンエステラーゼタンパク質と、ムスカリン性受容体に対するアゴニスト(アセチルコリンエステラーゼ感受性の塩化アセチルコリンなど)とを含有する緩衝液で希釈されるか、またはそのような緩衝液と一緒にされるか、そのいずれかである。アゴニストと、アセチルコリンエステラーゼと、推定される阻害剤とを含有するこの混合物は、0時間〜数時間の間で所定の時間(5分〜60分の間が最も実用的である)にわたってインキュベーションされ、次いで、その天然のテールを有するか、またはバソプレッシン受容体などに由来する高親和性テールで交換されたテールを有するムスカリン性受容体およびアレスチン−GFPを含有する細胞と接触させられる。アセチルコリンエステラーゼの推定される阻害剤が存在する場合、塩化アセチルコリンは分解されず、アレスチン−GFPの形質膜またはエンドソームへの輸送が、アセチルコリンによる受容体の活性化のために生じる。アセチルコリンエステラーゼの阻害剤が存在しない場合、塩化アセチルコリンが分解され、より少ない量のアレスチン−GFPの輸送が生じるか、またはアレスチン−GFPの輸送が全く生じない。このアッセイは、環境中におけるアセチルコリンエステラーゼ酵素の一般に使用されている阻害剤、例えば、有機ホスファート殺虫剤など、例えば、ジアジノン(環境保護庁は危険性評価を完了し、殺虫剤ジアジノンに対する危険性削減協定を発表している;2000年12月5日、環境保護庁はその改訂危険性評価を公開し、有機ホスファート殺虫剤ジアジノンのいくつかの使用の段階的な廃止/排除を行うための登録者との協定を発表した)、そして神経毒サリンの誘導体などの一般には見出されないより強力な阻害剤を評価するために使用することができる。そのようなアッセイシステムの特定の使用が、水の一部を、塩化アセチルコリン様アゴニストおよびアセチルコリンエステラーゼと予備混合した後、アレスチン−GFPおよびムスカリン性受容体を含有する細胞を有するチャンバーに連続的に加え、アレスチン−GFP輸送の阻害の減少について細胞を観測することによって、殺虫剤および同様な化合物に対して都市水道システムを連続的にモニターすることであり得る。これらのアッセイは、この目的のために市販されている装置によって大きい処理能力で行うことができる。このアッセイの別の使用は、アセチルコリンエステラーゼを阻害する化合物に対する人の生理学的暴露を、血清または組織におけるこれらの化合物の存在を測定することによって評価することである。例えば、1滴の血液を、塩化アセチルコリンにさらされた細胞を含有し、アレスチン−GFPおよびムスカリン性アセチルコリン受容体を有するウェルに直接入れることができる。ヒトの血液または血清は、通常、アセチルコリンを迅速に分解するために十分なアセチルコリンエステラーゼを含有する。従って、アレスチン−GFPを輸送する細胞の能力の阻害が減少することにより、アセチルコリンエステラーゼ阻害剤の存在が示される。

- 高ガストリン血症の患者に由来する試験サンプルにおけるガストリン濃度を測定する方法であって、
(a)ガストリン受容体CCK−B及び検出可能に標識されたアレスチンを含む細胞を提供するステップと、ここで、検出可能に標識されたアレスチンの細胞分布が、CCK−Bの活性化に応答して変化し、

- ガストリン濃度が、10nM未満である請求項5に記載の方法。
- 細胞質と比較した、形質膜、クラスリン被覆小孔、エンドサイトーシス小胞またはエンドソームにおける検出可能に標識されたアレスチンの局所的な濃度の増加が、局所的なシグナル強度の増加をもたらす請求項1に記載の方法。
- 局所的なシグナル強度が、試験サンプルにおけるリガンドの濃度の増加により増大する請求項7に記載の方法。
- 形質膜、クラスリン被覆小孔、エンドサイトーシス小胞またはエンドソームにおける標識されたアレスチンのシグナル強度が、細胞質におけるシグナル強度のレベルと比較して増大する請求項1に記載の方法。
- 検出可能な標識が、放射性同位体、エピトープタグ、アフィニティー標識、酵素、蛍光基または化学発光基である請求項1に記載の方法。
- 細胞分布が、フローサイトメトリーにより視覚化される請求項1に記載の方法。
- 細胞分布が、蛍光共焦点顕微鏡観察によって視覚化される請求項1の方法。
- コンピューターが細胞分布のイメージを解析する請求項1に記載の方法。
- 試験サンプルが不均一である請求項1に記載の方法。
- 細胞分布が、試験サンプルにさらしてから15〜30分後に決定される請求項1に記載の方法。
