Description
少量の親水性化合物(ATP、NAD、NADP、NADH、NADPH、酵素、遊離脂肪酸、保存料、殺生物剤、塩等) は、微生物または例えば粒子と同様に、原油、植物油、石油およびケロセンのような疎水性液体マトリクス中に拡散または分散しがちである。このような化合物または粒子は、夾雑物または不純物となっている場合もあれば、また、特定の濃度であることが要求される添加物や保存料出る場合もある。したがって、このような化合物を検出することおよび/またはその定量を行うことによって、個々の製品が安全であり、かつ個々の用途に対して適切なものであるかを確認することが望ましい。上記したような親水性化合物を、本明細書においては親水性化合物と総称する。
本発明は、疎水性/非極性/非イオン性液体マトリクス中に拡散または分散した親水性化合物の抽出方法に関する。本発明は、捕捉溶液にも関する。
本発明が対象とするのは、疎水性液体マトリクス中に拡散または分散した親水性化合物の抽出および濃縮の方法であって、以下の工程を含むものである。
a)疎水性液体のサンプルの提供
b)水性の捕捉溶液として少なくとも1種の抽出剤を含有するものの、該サンプルへの添加
c)該サンプルと該捕捉溶液の完全な混合
d)水相のサンプル相からの分離
e)前記水相における前記化合物の測定。
図1は、本発明の手順を示している。詳細は例に示した。示されている工程は、(1)1リットルのサンプルを回収する;(2)捕捉溶液を添加する;(3)混合物を10秒間激しく振る;(4)そして混合物を5分間直立させる;(5)捕捉溶液を回収する;(6)捕捉溶液をHY−LiTE(登録商標)ペンチューブに添加する;(7)捕捉溶液をHY−LiTE(登録商標)ペンを用いてテストする;(8)HY−LiTE(登録商標)ペンを読む;HY−LiTE(登録商標)照度計を用いて放出された光を測定する。
本発明において用いられる捕捉溶液は、抽出剤の水性溶液である。この捕捉溶液は、任意に酸、塩基または緩衝材を添加剤として用いて、所定のpHを維持しても、および/または中性塩を添加してイオン強度を維持してもよいし、本発明は保存料を添加して夾雑した微生物が捕捉溶液中において増殖するのを防いでもよい。これらの添加剤の性質は、マトリクス、分析物および分析方法に依存するが、下記を例示することができる;
・次亜塩素酸ナトリウム−使用前の無菌性の維持:
・塩化ナトリウム−溶液の等浸透圧維持:
・リン酸バッファ−溶液のpH維持;
・水酸化ナトリウム−溶液の滴定可能なアルカリ性の維持。
他の例は当業者に自明である。
当技術分野において、界面活性剤および乳化剤は水中油型分散剤または油中水型分散剤の製造および安定化に用いられている。また、これらの物質は、疎水性物質を水性溶液に溶解するためにも用いられている。これらの物質の他の用い方としては、生体膜の破壊がある。
本発明の捕捉溶液を用いることによって、親水性化合物の回収率は50%より高くなり、好ましくは80%より高くなる。
大豆レシチン: 0.50〜10.00g
メチレンブルー: 0.01〜0.20g
次亜塩素酸ナトリウム: 0.01〜0.05g
水: 1000.00mlとする
この捕捉溶液は、燃料、例えばディーゼルオイル、ケロセン中の夾雑物の検出にとくに有用である。
生成した混合物はある時間静置して相分離させる。必要に応じて、相分離を促進するために、さらに少量のイオン性化合物(典型的には塩、酸または塩基)を追加して添加してエマルションを破壊するか、または物理的な処理(変温、振動、遠心分離)を行ってもよい。
本発明の抽出剤の製造は、成分を蒸留水に溶解することによって行う。

以下の例は、本発明の実際の適用を示すものである。
例1:捕捉溶液を用いた抽出効率の改善(ATPの抽出)
本発明の使用法として、遊離化合物の疎水性液体サンプルからの抽出の改善を表1aに示した。選択されたサンプルはディーゼルオイルであり、遊離化合物のマーカーはATPである。10mlのディーゼル燃料を2つの瓶のそれぞれに移す。1μlの1.0x10−4MのATP溶液を各瓶に添加し、ボルテックスミキサーによって60秒間混合する。一方の瓶に1mlの水のみを入れ、他方の瓶に1mlの捕捉溶液(0.1%(w/vのレシチン)を入れる。いずれの瓶も、ボルテックスミキサーによって10秒間混合する。瓶を30分間放置し、安定状態となった水および捕捉液を除去する。回収した水および捕捉溶液に含まれる遊離ATP量の測定をMerck KGaAのバイオ発光試薬を用いてアッセイして行う。100%回収された場合として、1μlの1.0x10−4MのATP溶液を1mlの水および1mlの捕捉溶液に直接添加して遊離ATP量を測定する。
%回収率の計算は、下式によって行う。
本発明の使用法として、遊離化合物の疎水性液体サンプルからの抽出の改善を表2aに示した。選択されたサンプルは航空機燃料であり、バイオマスの決定のマーカーはアデノシン三リン酸(ATP)である。
本発明の使用法として、遊離化合物およびバイオマス成分の疎水性液体サンプルにおける捕捉およびそれに続く測定を表3に示した。
選択されたサンプルは航空機燃料であり、遊離化合物およびバイオマス成分の決定のマーカーはアデノシン三リン酸(ATP)である。ATPの決定のレベルによって、航空機燃料における微生物の夾雑のレベルが示される。航空機燃料として、航空機の翼部タンクから集めたものをサンプルとして用いてATPのレベルおよびTVC(全生存細胞数)を測定する。燃料のサンプルには下記のとおりの処理を行い、水性捕捉液の試験をATPのレベルおよびTVCについて行えるようにした。
遊離ATP(細胞外ATP)および全ATP(バイオマスATP+細胞外ATP)のレベルの測定は、Merck KGaA, Germany製のHY−LiTE(登録商標)ATP発光アッセイによって行った。遊離ATPの測定は、28μlの捕捉溶液をHY−LiTE(登録商標)ペン(反応キャップを介して)にピペットし、発光量(RLUとして表される−相対発光量)をHY−LiTE(登録商標)発光メータによって測定することによって行う。TVCの決定は、未希釈、10−2および10−4に希釈した100μlの捕捉溶液をトリプトン大豆寒天プレートにプレーティングすることによって行った。プレートを2日間、20℃にてインキュベートし、コロニー数を数えた。コロニー形成単位(cfu/ml)としてmlあたりのものを計算した。
cfu/ml = コロニー数x希釈率x10
r = 0.8872 y = 0.0049293 x + 676,7
このデータは図2に示した。
本発明の使用法として、遊離化合物の疎水性液体サンプルからの抽出の改善を表4に示した。選択されたサンプルは航空機燃料であり、遊離化合物のマーカーは硝酸塩である。100mlの新しい航空機燃料を3つの瓶のそれぞれに移す。9μlの32.4g/リットルの硝酸カリウム溶液を各瓶に添加し、手で振ることによって10秒間混合し、5分間放置する。1つの瓶に1mlの水のみを入れ、他の瓶に1mlの捕捉溶液(0.1%(w/v)のレシチンまたは0.05%(w/v)のSDS)を入れる。全ての瓶を手で振ることによって10秒間混合する。瓶を5分間放置し、安定状態となった水および捕捉液を除去する。回収した水および捕捉溶液に含まれる硝酸塩量の測定をMerck KGaAの硝酸塩アッセイを用いて行う。100%回収された場合を測定するために、9μlの1.0x10−4Mの32.4g/リットルの硝酸カリウム溶液を1mlの水または1mlの捕捉溶液に直接添加して遊離硝酸塩量を測定する。

種々の抽出剤による、遊離化合物の疎水性液体サンプルからの抽出の改善を表5および6に示した。選択されたサンプルはディーゼルオイルであり、抽出対象化合物はATPである。10mlのディーゼル燃料を数多くの瓶のそれぞれに移す。1μlの1.0x10−4MのATP溶液(実験A;表5)または5μlの1.0x10−4MのATP溶液(実験B;表6)を、多くの瓶の各瓶に添加し、ボルテックスミキサーによって60秒間混合する。1つの瓶に1mlの水のみを入れ、他の1つの瓶に1mlの試験化合物溶液または試験対象である化合物の混合物を入れる。全ての瓶をボルテックスミキサーによって10秒間混合する。瓶を5分間放置し、安定状態となった水および捕捉液を除去する。回収した水および捕捉溶液に含まれる遊離ATP量の測定をMerck KGaAのバイオ発光試薬を用いてアッセイして行う。100%回収された場合を測定するために、1μlの1.0x10−4MのATP溶液(実験A)または5μlの1.0x10−4MのATP溶液(実験B)を1mlの水および1mlの捕捉溶液に直接添加して遊離ATP量を測定する。
%回収率の計算は、下式によって行う。
例5の実験を、本発明の抽出剤とは異なる界面活性剤および抽出作業において通常用いられる他の化合物を用いて繰り返す。実験Cにおいては、1μlの1.0x10−4MのATP溶液を瓶および100%コントロールに入れ、実験Dにおいては、5μlの1.0x10−4MのATP溶液を瓶および100%コントロールに入れる。結果を下記表7および8にまとめた。
Claims (8)
- 原油、石油およびケロセンから選択される疎水性液体マトリクス中に拡散または分散したATPの抽出および濃縮の方法であって、以下の工程
a)疎水性液体のサンプルの提供;
b)水性の捕捉溶液として少なくとも1種の抽出剤を含有するものの、該サンプルへの添加;
c)該サンプルと該捕捉溶液の完全な混合;
d)水相のサンプル相からの分離;
e)前記水相におけるATPの測定
を含む、前記方法。 - 原油、石油およびケロセンから選択される疎水性液体マトリクス中に拡散または分散したATPの抽出および濃縮するための、少なくとも1種の抽出剤を含有する水性の捕捉溶液であって、該捕捉溶液中の該抽出剤は、疎水性マトリクスから抽出されたATPの収量を向上せしめる、前記捕捉溶液。
- 抽出剤の選択が、両性リン脂質またはアニオン性リン脂質およびアニオン性界面活性剤からなる群からなされる、請求項2に記載の捕捉溶液。
- 抽出剤がレシチンである、請求項3に記載の捕捉溶液。
- 2種以上の抽出剤を含有する、請求項2〜4のいずれかに記載の捕捉溶液。
- 抽出剤と非イオン性界面活性剤とを含有する、請求項2〜5のいずれかに記載の捕捉溶液。
- 水相の可視性を良好にする量の水溶性染料を含有する、請求項2〜6のいずれかに記載の捕捉溶液。
- 請求項2〜7のいずれかに記載の捕捉溶液を含む、ATPを原油、石油およびケロセンから選択される疎水性マトリクスから抽出し、ATPを検出するための反応キット。
