Description
(1)プロテインキナーゼA触媒サブユニットをコードする遺伝子が変異された非ヒト動物。
(a)上記(1)記載の非ヒト動物由来の遺伝子又はタンパク質及び野生型非ヒト動物由来の遺伝子又はタンパク質の発現パターンをそれぞれ測定し、
(b)得られた発現パターンを比較することにより、加齢性記憶障害を抑制又は改善す る物質の標的となる物質を同定すること
を含む、前記方法。
本発明者はamn遺伝子産物の神経ペプチドを受け取るキノコ体にAMIの原因があると考え、キノコ体で発現する遺伝子の変異体からAMIの発現に異常のみられる系統の検索を試み、プロテインキナーゼA(PKA)の触媒サブユニットをコードする遺伝子DC0のヘテロ変異体DC0/+に強いAMIの発現抑制があることを見出した。そして、本発明者はPKAの活性を遺伝学的に50〜60%抑制した非ヒト動物でAMIを抑制することが出来ることを示した。本発明は一つにはAMIが抑制される遺伝学的条件を供与すること、また、マウスなどと比べて寿命の短いショウジョウバエを用いることにより、従来考えられていた背景技術に記載した二つの障害を克服するものである。
(1)PKA触媒サブユニット
PKAは、ATPのγ-リン酸基をタンパク質の特定のセリン又はスレオニンのヒドロシル基に転移する酵素であり、真核細胞に広く分布している。この酵素は、cAMPが結合する分子量約4万の調節サブユニットと分子量約5万の触媒サブユニットから構成され、1分子の調節サブユニットに2分子のcAMPが結合すると、触媒サブユニットと調節サブユニットが解離し、活性を示すようになる。本発明は、このような活性型PKAが加齢による記憶障害(AMI)を起こすことを見出したものである。従って、本発明においては、記憶過程に関わるcAMPシグナル経路において重要な役割を担っている触媒サブユニットに変異を導入するとともに、PKA活性を全部抑えることなくマイルドに低下させることにより、若年時の学習や記憶には影響を与えず、寿命に影響を与えることなくAMIを抑制した動物を提供する。
PKAの触媒サブユニットのホモ欠失変異体は胚致死であるため、本発明の動物はヘテロ変異体であることが好ましい。また、本変異体は、染色体上のPKAの触媒サブユニット遺伝子に塩基置換が起こったことによる、PKA触媒サブユニットとしての機能が部分的に失われた低次形態型の変異体(hypomorph変異体という)であり、そのPKA活性はマイルドに抑制され、野生型PKA活性の50〜60%の活性を保持するものである(つまり、野生型と比較して活性が40〜50%低下している)。PKA活性が野生型と比較して50〜60%マイルドに低下していることにより、変異体動物では、若年時の学習や記憶は影響を受けることなくAMIが抑制されうることが可能となっている。しかしPKA活性が過剰に増加又は減少すると学習記憶が障害されてしまう。なお、このようなPKA活性がマイルドに低下する機能は対立遺伝子特異的に獲得されたものである。
本発明のPKA触媒サブユニットをコードする遺伝子のヘテロ変異体動物は、ショウジョウバエ等の非哺乳動物や、ラット、マウス等のヒト以外の哺乳動物が含まれるがこれらに限定されるものではない。特に、ショウジョウバエであるDrosophila melanogasterは、寿命が短く、老齢個体の行動遺伝学的解析に最適であり(Exp Gerontol 37, 1347-1357,2002)、本発明の動物として好ましい。
AMIの予防・改善薬を開発するためにはAMI発現の分子メカニズムを明らかにし、AMIの予防・改善薬のターゲットとなる生体内の分子・遺伝子を見出すことが重要である。
本発明の方法は、以下の工程:
(a)本発明の非ヒト動物由来の遺伝子又はタンパク質及び野生型非ヒト動物由来の遺伝子又はタンパク質の発現パターンをそれぞれ測定し、
(b)得られた発現パターンを比較することにより、加齢性記憶障害を抑制又は改善す る物質の標的となる物質を同定すること
を含むものである。
本発明は、PKA触媒サブユニットをコードする遺伝子が変異された非ヒト動物由来のタンパク質及び野生型非ヒト動物由来のタンパク質を用いてリン酸化タンパク質の有無又はレベルを測定・比較し、得られる測定結果からPKA触媒サブユニットをコードする遺伝子が変異された非ヒト動物でリン酸化が抑制されたタンパク質を同定することを特徴とし、加齢性記憶障害を抑制又は改善する物質の標的となる物質の同定方法を提供する。
野生型のハエとして用いたw(CS10)は、すでに報告のあるようにw1118をCanton-Sに10回交配して得られた白眼のCanton-Sである。この研究に用いたdnc1を除く、すべての変異体はw(CS10)と最低6回、交配して用いた。遺伝子の変異の位置がわかっていないdnc1はw(CS10)と交配して生まれたヘテロ個体を用いた。dnc遺伝子はX染色体上にあるので、dnc遺伝子のヘテロ個体は全てメスである。dnc1/+の実験のコントロールには、メスのw(CS10)、オスのw(CS10)、そしてオスのdnc1とメスのw(CS10)を交配して生まれたオスを用いた。その結果、全てのコントロール間で学習、記憶、そして匂い反応性に差は見られなかった。これより、dnc1/+の結果に遺伝学的バックグラウンドが影響を与えていないことが示された。
典型的な匂い条件付け(参考文献10)では、ハエの嫌いな匂いを2種類(3-オクタノール [OCT]と4-メチルシクロヘキサノール [MCH])を用いる。約100匹のハエに対して最初の匂い(CS+)を60秒間電気ショックと共に与えた(電気ショックは1.5秒間、60Vの直流電気ショックを3秒間間隔で与えた)。最初の匂い呈示後、45秒間の休憩後、次の匂い(CS-)を60秒間、今度は電気ショック無しで与えた。記憶テストではトレーニングを受けたハエにCS+とCS-を選択させた。パフォーマンスインデックス(PI)は、ハエが50:50に分かれたときは0となり、CS+の匂いから離れた方へ0:100に分かれたときは100とした。記憶実験における嗅覚や電気ショックに対する反応性はトレーニングを受けていないハエに対してOCTと空気、MCHと空気の選択、電気ショックありとショックなしを選択させることにより調べた。なお、本発明において使用された変異体及び加齢体では、匂いとショックに対する反応性に顕著な異常は見られなかった。
ショウジョウバエ頭部のPKA活性は、PKAによる基質ペプチドのリン酸化を分析するための標準的なELISA法(MESACUP Protein Kinase Assay Kit、MBL、名古屋、日本)により調べた。
頭部抽出物は以下の手順で得た。まず、ショウジョウバエをドライアイスで凍らせ、頭部を分離した。次に、約100個の頭部を200μLのsample preparation buffer(MESACUP Protein Kinase Assay Kit)を用いて、乳棒ですりつぶした。その後、遠心分離して不純物を取り除き、頭部の抽出物を得た。分析を2μMのcAMP濃度存在下で行い、組換えPKAc(New England Biolabs)を用いて、検量線を作成した。それぞれのデータは最低4回の独立した実験の結果の平均から得た。
DC0転写物の発現レベルの比較はreal time PCR(Applied Biosystems,model 7500)を用いて定量した。ショウジョウバエの頭部を、ドライアイスを用いて分離し、全RNAをTRIzol reagent(Invitrogen)を用いて分離した。cDNAはHigh Capacity cDNA Archive Kit(Applied Biosystems)を用いて合成し、DC0とGAPDH2特異的なプライマーとSYBR-Greenを用いて定量PCRを行なった。用いたプライマーは以下のとおりである。
DC0-fwd: CAAGGAGGAGTTCGAGGACA (配列番号3)
DC0-rev: GTGCTGGACGATCATGACAC (配列番号4)
GAPDH2-fwd: GCGGTAGAATGGGGTGAGAC (配列番号5)
dGAPDH2-rev: TGAAGAGCGAAAACAGTAGC (配列番号6)
PCRは通常の条件で行った。DC0の発現はGAPDH2の発現を標準化して得た。それぞれのデータは最低4回の独立した実験の平均からなる。
口吻を取り除いた3〜5日目のショウジョウバエ成虫の頭部を4%ホルムアルデヒドで4時間固定した。その後、エタノールで30分×2回洗い、さらにトルエンで10分×3回洗った。頭部は100%パラフィンに4回、それぞれ1時間ずつ浸し、その後、パラフィンに包埋した。5ミクロンのスライスを集め、キシレンで5分×3回、濃度を下げたEtOHで5分×3回、再水和させた。DC0の抗体はDaniel Kalderon(コロンビア大学、ニューヨーク)から得た。抗体はPBHT (0.02M NaPO4、0.5M NaCl、0.2%Triton X-100 (pH7.4))と5%ヤギ血清で1:400に希釈して使用した。FITCが結合した抗ウサギ2次抗体は1:2000に希釈して使用した。
AMIに影響を及ぼす1遺伝子変異を同定するために、DC0(参考文献7, 11, 13)、volado (参考文献8)、fasciclin 2 (参考文献9)、dunce (参考文献10)、rutabaga(参考文献10)、など既知の変異体に加えてキノコ体でレポータシグナルを示す GAL4トランスポゾン因子系統などを含む、主にキノコ体で発現している約100個の遺伝子の変異体の供与を受け、それぞれの若齢体および加齢体で1時間記憶をテストし1時間記憶の低下を指標にAMIの抑制変異体を検索した。
AMIは一般に個体老化の結果生じると考えられていることから、DC0/+変異体は個体老化を遅らせることによってAMIを抑制している可能性がある。その可能性を調べるためにDC0B3/+の寿命を調べた。驚いたことにDC0B3/+のハエの寿命は野生型と変わらなかった。オスでは野生型の41.6日(N=238)に対してDC0/+は39.2日(N=239)、メスでは野性型の36.3日(N=236)に対してDC0/+は37.0日(N=227)であった(図2a)。
AMIの抑制モデル系からAMIと個体老化との関連性が示唆されている。しかしながら、たった1つの主要な生体内経路が、もっとも老化に関係する表現型を調節しているのか、あるいは、多くの生体内経路が関係しているのか不明である。心臓機能の老化は個体老化同様、インシュリン/インシュリン様成長因子経路(IIS)によって調節されている(参考文献14, 15)。従って心臓機能の老化は個体老化と同一のメカニズムにより制御されていると考えられる。同様に、哺乳類では代謝活性を下げると、寿命を延ばすのと同じようにAMIが抑制される。一方、過剰なカロリー制限による代謝活性の低下は、寿命は延ばすがAMIは抑制しないことが報告されている(参考文献16)。また、他の動物と比較して人間では更年期の開始時期から寿命による死亡時期までの間が比較的長い。これらのことはこれら2つの過程を分けて考えることができることを示している。また上記の通り、AMIが抑制されたDC0/+では寿命の延長が見られない。つまり、AMIは寿命に影響を与えず抑制できることを示しているが、このことはAMIと個体老化とを分けて考えることができる、というものである。
即ち15日齢での記憶力低下は、mth1と野性型で見分けが付かない。この結果は、mthでも運動活性が通常通り衰えるという以前の結果に類似している(参考文献18)。これらのことは、個体老化とAMIは部分的に一致しているが、個体老化の延長はAMI抑制の必要条件にも十分条件にもならないということを示唆している。
DC0-PKA活性の低下はAMIを強く抑制することから、加齢によりDC0の発現が上昇することで記憶力低下が低下するという可能性が考えられる。DC0をキノコ体に過剰発現させると顕著な記憶障害となる(図4a)。図4aは、DC0-PKAをキノコ体で過剰発現させると加齢体と同程度に1時間記憶が低下することを示す。野性型(+/+)と、キノコ体でDC0遺伝子を強制発現した系統(c747/PKAc+)において、1日齢と20日齢の1時間記憶をそれぞれ調べた。c747はGAL4をキノコ体で特異的に発現するエンハンサートラップ系統(GAL4トランスポゾン因子系統)で、PKAc+はUAS配列下流にDC0遺伝子を持つ系統である。即ち、c747/PKAc+ではGAL4/UASコントロール下、キノコ体で恒常的活性をもった触媒サブユニットを発現する。全てのデータはN=6である。

(a)プロテインキナーゼA触媒サブユニットをコードする遺伝子が変異され、脳における当該プロテインキナーゼAの活性が野生型の活性の50〜60%に低下した非ヒト動物由来の遺伝子又はタンパク質、及び野生型非ヒト動物由来の遺伝子又はタンパク質の発現パターンをそれぞれ測定し、
