Description
The present invention relates to a scanning probe microscope used for sample observation in liquid.
また、ダイナミックモードAFMに含まれるNC−AFM(Noncontact Atomic Force Microscope)においても、同様な制御が行われる。 The dynamic mode AFM, one of the scanning probe microscopes (SPM), vibrates the cantilever at the natural frequency of the cantilever or a frequency in the vicinity thereof, so that the amplitude or frequency of the cantilever is kept constant. In addition, the distance between the probe and the sample is controlled to observe the sample surface shape. Similar control is also performed in an NC-AFM (Noncontact Atomic Force Microscope) included in the dynamic mode AFM.
Thus, in the dynamic mode AFM, the cantilever is vibrated at its natural frequency or a frequency in the vicinity thereof. For this reason, in the dynamic mode AFM, it is necessary to accurately find the cantilever natural frequency before observing the sample and set the cantilever excitation frequency based on the found natural frequency. Note that the natural frequency (resonance frequency) of the cantilever is detected by measuring a resonance curve. That is, the detection of the natural frequency of the cantilever is performed by detecting the amplitude of the cantilever while sweeping the cantilever excitation frequency and obtaining the excitation frequency when the amplitude peak (resonance peak) is obtained.
The dynamic mode AFM can observe a sample in a liquid as well as a sample in a vacuum and atmospheric pressure. A dynamic mode AFM capable of performing observation in liquid is disclosed in, for example, Japanese Patent Application Laid-Open No. 2003-329565 (Patent Document 1). In the dynamic mode AFM of
In the liquid, the Q value of the cantilever is very small. That is, even if the cantilever resonates in the liquid, its amplitude is considerably smaller than in the atmosphere or vacuum. For this reason, when observing in liquid in
Further, in
For these reasons, when observing in liquid in
However, when the vibrating piezoelectric element (10) is vibrated with a large amplitude in this way, the cantilever mounting base (8) connected to the vibrating piezoelectric element (10) also vibrates greatly, and further, the cantilever holder and the like. The part of will also vibrate. Then, the vibration excites resonance of a component other than the cantilever (a component of the cantilever holder), and resonance peaks other than the cantilever increase in the above-described resonance curve measurement. Thus, when the resonance peak increases in the resonance curve measurement, the excitation frequency is erroneously set to a frequency that is not the cantilever resonance frequency, and stable SPM observation in liquid becomes impossible.
Further, as described above, when a thing other than the cantilever (a component part of the cantilever holder) vibrates or resonates, the following problems occur due to the influence of the vibration. That is, in the resonance curve measurement described above, the resonance peak of the cantilever appears at a position slightly deviated from the correct frequency position (resonance frequency position of the cantilever). Even in this case, the excitation frequency is erroneously set to a frequency that is not the resonance frequency of the cantilever.
The present invention has been made in view of these points, and an object thereof is to provide a scanning probe microscope capable of observing a sample by vibrating a cantilever at its resonance frequency.
The scanning probe microscope according to
According to the present invention, it is possible to observe a sample by vibrating a cantilever at its resonance frequency.
Hereinafter, embodiments of the present invention will be described with reference to the drawings.
FIG. 1 shows an example of a scanning probe microscope of the present invention, and shows an example of a dynamic mode AFM of the present invention suitable for observation in liquid. 2 is a view of the cantilever holder 6 in FIG. 1 as viewed obliquely from the front.
In FIG. 1,
Reference numeral 5 denotes a glass cover, and the glass cover 5 is attached to the
The outline of the apparatus structure in FIG. 1 has been described above. Hereinafter, the structure of the cantilever holder 6, which is a characteristic part of the present invention, will be described in detail with reference to FIGS.
Further, the
As shown in FIGS. 1 and 2, the
In the figure,
In such a configuration, when the
By applying this voltage, the vibrating
As described above, in the cantilever holder of the present invention, the vibration generated in the
Furthermore, in the cantilever holder of the present invention, as described above, the vibration of the vibrating
FIG. 3 shows a comparison between the resonance curve (solid line) measured in the apparatus of FIG. 1 of the present invention and the resonance curve (broken line) measured in the conventional method.
In the conventional method, the peak A that is not the resonance of the cantilever is the maximum. In addition, a peak B appears at f 1 that is slightly shifted from the resonance frequency position f 0 of the cantilever. That is, the frequency position shift of the resonance peak of the cantilever has occurred. If the cantilever excitation frequency is adjusted to such frequencies of peaks A and B, high-sensitivity SPM observation in liquid cannot be performed.
On the other hand, in the present invention, the resonance peak P of the cantilever is clearly measured at the resonance frequency position f 0 of the cantilever, and the peak P is maximum. As described above, in the present invention, a resonance curve with less spurious can be obtained by excitation from the second surface S2 of the cantilever 7 (surface on the
Heretofore, an example of the present invention has been described.
Next, another example of the present invention will be described with reference to FIG. In FIG. 4, the same components as those in FIG. 1 are denoted by the same reference numerals as those in FIG. 1, and description thereof is omitted.
In FIG. 4, 24 is a cantilever holder, and 25 is the cantilever holder base. The
The shape of the

A vibrating piezoelectric element (vibrating vibrator) 28 is bonded to the surface of the fixing
In such a configuration, when the
By applying this voltage, the vibrating
Also in the present invention, a resonance curve with less spuriousness similar to the example of FIG. 1 can be obtained by excitation from the second surface S2 (surface on the
The present invention is not limited to the above example. For example, the above-mentioned cantilever holder can be used not only for observation in liquid but also for sample observation in air or vacuum.
