本発明の多孔質表面を有する磁性粒子(以下、単に「磁性粒子」ともいう)は、磁性母粒子(A)の表面に、有機溶剤(S)に溶解するポリマー部(B)および有機溶剤(S)に溶解しないポリマー部(C)を形成し、前記有機溶剤(S)に前記ポリマー部(B)を接触させることにより得られる。以下、本発明の磁性粒子の製造に使用される物質およびその製造方法について、詳細に説明する。
磁性母粒子(A)は、磁石で集めることが可能な公知の粒子状物質であり、その粒径dは、好ましくは0.1〜10μm、さらに好ましくは0.2〜7μm、もっとも好ましくは0.5〜5.0μmである。dが0.1μm未満であると、磁力による分離精製に長時間を要することがあり、10μmを超えると生化学物質結合量が少ない場合がある。
非磁性核粒子(A1)は、有機物質および無機物質のいずれも使用可能であるが、好ましくは有機物質である。有機物質の代表例としては、例えばポリマーを挙げることができる。かかるポリマーとしては、特に、ビニル系ポリマーが好ましく、その製造に使用するビニル系モノマーとしては、スチレン、α−メチルスチレン、ハロゲン化スチレン、ジビニルベンゼンなどの芳香族ビニル単量体、酢酸ビニル、プロピオン酸ビニルなどのビニルエステル類、アクリロニトリルなどの不飽和ニトリル、メチルアクリレート、エチルアクリレート、エチルメタクリレート、ブチルアクリレート、ブチルメタクリレート、2−エチルヘキシルアクリレート、2−エチルヘキシルメタクリレート、ラウリルアクリレート、ラウリルメタクリレート、エチレングリコールジアクリレート、エチレングリコールジメタクリレート、シクロヘキシルアクリレート、シクロヘキシルメタクリレートなどのエチレン性不飽和カルボン酸アルキルエステルなどを例示することができる。このビニル系ポリマーは単独重合体であっても、あるいは上記ビニル系モノマーから選ばれた2種以上のモノマーからなる共重合体であってもよい。また、上記ビニル系モノマーとブタジエン、イソプレンなどの共役ジオレフィン、アクリル酸、メタクリル酸、アクリルアミド、メタクリルアミド、グリシジルアクリレート、グリシジルメタクリレート、N−メチロールアクリルアミド、N−メチロールメタクリルアミド、2−ヒドロキシエチルアクリレート、2−ヒドロキシエチルメタクリレート、ジアリルフタレート、アリルアクリレート、アリルメタクリレート、トリメチロールプロパントリアクリレート、トリメチロールプロパントリメタクリレートなどの共重合可能なモノマーとの共重合体も使用することができる。
磁性体微粒子(A2)は、特に制限はないが、酸化鉄系の物質が代表的であり、MFe2O4(M=Co、Ni、Mg、Cu、Li0.5Fe0.5等)で表現されるフェライト、Fe3O4で表現されるマグネタイト、あるいはγFe2O3が挙げられる。特に、飽和磁化が高く、かつ残留磁化が低い磁気材料として、γFe2O3、Fe3O4が好ましい。
本発明の磁性母粒子(A)を製造するにあたり、非磁性核粒子(A1)の表面に磁性体微粒子(A2)を配置させるには、まず、非磁性核粒子(A1)と磁性体微粒子(A2)とを混合し、非磁性核粒子(A2)の表面に磁性体微粒子(A1)を物理的に吸着させる。
有機溶剤(S)は、ポリマー部(B)を溶解し、かつ、ポリマー部(C)を溶解しない。本発明において「ポリマーを溶解する」とは、粉砕したポリマー1gを有機溶剤100gに入れて24時間撹拌した後、目視でポリマーが前記有機溶剤中に確認されないことをいう。また、本発明において「ポリマーを溶解しない」とは、粉砕したポリマー1.0gを有機溶剤100gに入れて24時間攪拌した後、目視で前記有機溶媒中にポリマーが確認されることをいう。
ポリマー部(B)は、有機溶剤(S)に溶解するポリマーである。ポリマー部(B)は、有機溶剤(S)に溶解すれば、いずれのポリマーでもよい。
ポリマー部(C)は有機溶剤(S)に溶解しないポリマーである。ポリマー部(C)は有機溶剤(S)に溶解しない限り、いずれのポリマーでもよい。
タレートなどが挙げられる。エポキシ基を有するモノマーとしては、例えば、グリシジルメタクリレートなどが挙げられる。また、ポリマー部(C)を形成する際に、ポリマー部(C)とともに、その他の単官能性モノマー(b)を併用してもよい。
有機溶剤(S)にポリマー部(B)を接触させる方法としては、例えば以下の方法が挙げられる。まず、上述の工程により、磁性母粒子(A)の表面をポリマー部(B),(C
)で被覆した後、磁気分離により前記粒子および水を分離した後、上澄みの水を除去する。次に、水と任意に混和する溶媒で置換することにより、残留する水を除去する。次いで、前記溶媒を、有機溶剤(S)にさらに置換し、有機溶剤(S)中に前記粒子を保持する。これにより、前記粒子からポリマー部(B)を溶出させることが好ましい。さらに、必要に応じて、前記粒子を有機溶剤(S)で数回洗浄する。次いで、前記粒子を含む有機溶剤(S)を、水と任意に混ざりあう溶媒に置換する。さらに、前記粒子を水で数回洗浄することにより、本発明の多孔質表面を有する磁性粒子の水分散体を得ることができる。なお、有機溶剤(S)にポリマー部(B)を接触させる際に有機溶剤(S)の温度を上げることにより、より多孔質の表面を得ることができる。
本発明の磁性粒子は、生化学分野、塗料、紙、電子写真、化粧品、医薬品、農薬、食品、触媒など広い分野で利用できる。
以下、実施例を挙げて本発明をさらに詳細に説明するが、本発明はこれらによって制限されるものではない。
本実施例および比較例において、得られた磁性粒子の評価は以下の方法によって測定された。また、粒子の粒径は、特に説明がない限り以下の方法により測定された。
本実施例においては、生化学物質結合量として、ビオチン類結合量の評価を行なった。
ビオチン類結合量(pmol/mg)=
{(結合前の蛍光標識化ビオチンの量(pmol))−(未反応蛍光標識化ビオチンの量(pmol))}/(磁性粒子の質量(mg)) ・・・・・(1)
本実施例に用いたビオチン類結合用粒子は、カルボキシル基を表面に有する磁性粒子を、縮合剤である1−エチル−3−ジメチルアミノプロピルカルボジイミド塩酸塩と反応させ、活性化してからストレプトアビジンをさらに投入して、ストレプトアビジンを該磁性粒子に化学結合させることにより作成された。
直径1μm以上の粒子については、レーザ回折式粒度分布測定装置((株)島津製作所製)SALD−200Vにより、直径1μm未満の粒子についてはレーザ散乱回折法粒度分布測定装置LS 13 320((株)ベックマン・コールター)により粒径を測定した。
特開平7−238105号公報記載の重合方法を参考に、スチレン/ジビニルベンゼン=95/5共重合体粒子(平均粒子径1.5μm)を作製し、重合後遠心分離により粒子のみ取り出したものをさらに水洗し、乾燥、粉砕した。これをコア粒子(1)とする。
2.3.1.多孔質表面を有する磁性粒子の作製
ドデシルベンゼンスルホン酸ナトリウム0.5重量%およびノニオン性乳化剤(商品名:「エマルゲン150」,花王(株)製)0.5重量%を含む水溶液750gを、1Lセパラブルフラスコに投入し、次いで、磁性母粒子(A)30gを投入し、ホモジナイザーで分散した後、60℃に加熱した。ドデシルベンゼンスルホン酸ナトリウム0.5重量%およびノニオン性乳化剤(商品名:「エマルゲン150」,花王(株)製)0.5重量%を含む水溶液50gに、メタクリル酸シクロヘキシル15g、tert−ドデカンチオール0.075g、およびターシャリーブチルペルオキシ2−エチルヘキサネート(日本油脂社製;パーブチルO)0.75gを入れて分散させたプレエマルジョンを、60℃にコントロールした前記1Lセパラブルフラスコに2時間かけて滴下した(ポリマー部(B)の形成)。
次に、固形分濃度1%の磁性粒子の水分散体1mLに1−エチル−3−ジメチルアミノプロピルカルボジイミド塩酸塩(同仁化学社製)5mgを溶解した0.1mM HCl溶液0.1mLを添加し、室温で2時間回転攪拌し、さらに、ストレプトアビジン(シグマ社製)1mgを溶解した0.1mM HCl溶液0.1mLを添加し、室温で8時間回転攪拌することより、粒子の表面にストレプトアビジンを固定化させたビオチン結合用粒子を調製した。次いで、このビオチン結合用粒子を含む分散液を磁気分離処理した後、固形物(ビオチン結合用粒子)に、0.1%牛血清アルブミンを含むリン酸塩緩衝液(PBS,0.1%BSA/PBS,pH=7.2)を添加して磁気分離処理する操作を3回繰り返すことにより、未反応のストレプトアビジンを除去した。そして、ビオチン結合用粒子を、その固形分濃度が1%となるように、0.1%牛血清アルブミンを含むリン酸塩緩衝液(PBS,0.1%BSA/PBS,pH=7.2)に分散させることにより、固形分濃度1%のビオチン結合用粒子の分散液を調製し、ビオチン類結合量を評価した。結果を表1に示す。また、実施例1のビオチン類結合用粒子の平均数粒径は2.3μmであった。
2.4.1.多孔質表面を有する磁性粒子の作製
ドデシルベンゼンスルホン酸ナトリウム0.5重量%およびノニオン性乳化剤(商品名:「エマルゲン150」,花王(株)製)0.5重量%を含む水溶液750gを1Lセパラブルフラスコに投入し、次に磁性粒子(A)30gを投入してホモジナイザーで分散した後、60℃に加熱した。次に、ドデシルベンゼンスルホン酸ナトリウム0.5重量%およびノニオン性乳化剤(商品名:「エマルゲン150」,花王(株)製)0.5重量%を含む水溶液75gに、メタクリル酸シクロヘキシル7.5g、エチレングリコールジメタクリレート0.75g、およびターシャリーブチルペルオキシ2−エチルヘキサネート(商品名「パーブチルO」,日本油脂(株)製)0.38gを入れて分散させたプレエマルジョンを、60℃にコントロールした前記1Lセパラブルフラスコに2時間かけて滴下し、80℃に昇温した後、2時間反応させた(磁性母粒子(A)へのポリマー形成)。
得られた実施例2の磁性粒子について、実施例1と同様の方法にてビオチン類結合量を評価した。結果を表1に示す。また、実施例2のビオチン類結合用粒子の平均数粒径は2.1μmであった。
実施例1において、ポリマー部(B)(メタクリル酸シクロヘキシル)の重合を行なわず、ポリマー部(C)の重合のみを行なうことにより、比較例1の磁性粒子を得た。比較例1の磁性粒子をSEM観察したところ、表面が多孔質でないことが確認された。また、比較例1の磁性粒子について、実施例1と同様にビオチン類結合量を評価した。結果を表1に示す。
実施例1において、ポリマー部(B)を有機溶剤(S)(アセトン)に接触させる前の磁性粒子(比較例2の磁性粒子)を得た。比較例2の磁性粒子をSEM観察したところ、表面が多孔質でないことが確認された。また、比較例2の磁性粒子について、実施例1と同様にビオチン類結合量を評価した。結果を表1に示す。
Claims (10)
- 磁性母粒子(A)の表面に、有機溶剤(S)に溶解するポリマー部(B)を形成し、次に、前記有機溶剤(S)に溶解しないポリマー部(C)を形成した後、
前記有機溶剤(S)に前記ポリマー部(B)を接触させることにより得られる、多孔質表面を有する磁性粒子。 - 請求項1において、
前記磁性母粒子(A)の存在下、水系媒体中で単官能性モノマー(b)をラジカル重合することにより、前記ポリマー部(B)を形成する、多孔質表面を有する磁性粒子。 - 請求項1または2において、
前記磁性母粒子(A)の存在下、水系媒体中で多官能性モノマー(c)をラジカル重合することにより、前記ポリマー部(C)を形成する、多孔質表面を有する磁性粒子。 - 請求項1ないし3のいずれかにおいて、
前記磁性母粒子(A)は、
非磁性核粒子(A1)と、
前記非磁性核粒子(A1)の表面上に配置された磁性体微粒子(A2)と、
を含む、多孔質表面を有する磁性粒子。 - 請求項4において、
前記磁性母粒子(A)は、前記非磁性核粒子(A1)および前記磁性体微粒子(A2)を被覆する母粒子コート層(A3)をさらに含む、多孔質表面を有する磁性粒子。 - 請求項1ないし5のいずれかに記載の磁性粒子を用いた、生化学用担体。
- 請求項1ないし5のいずれかに記載の磁性粒子の表面に、ビオチン類結合部位を有する物質が固定化されている、ビオチン類結合用粒子。
- 磁性母粒子(A)の表面に、有機溶剤(S)に溶解するポリマー部(B)を形成し、次に、前記有機溶剤(S)に溶解しないポリマー部(C)を形成する工程と、
前記有機溶剤(S)に前記ポリマー部(B)を接触させる工程と、
を含む、多孔質表面を有する磁性粒子の製造方法。 - 請求項8において、
前記ポリマー部(B)を形成する工程は、前記磁性母粒子(A)の存在下、水系媒体中で単官能性モノマー(b)をラジカル重合する工程を含む、多孔質表面を有する磁性粒子の製造方法。 - 請求項8または9において、
前記ポリマー部(C)を形成する工程は、前記磁性母粒子(A)の存在下、水系媒体中で多官能性モノマー(c)をラジカル重合する工程を含む、多孔質表面を有する磁性粒子の製造方法。
