【従来の技術及び発明が解決しようとする課題】
動脈硬化症は、高血圧症や血栓症等の種々の循環器系疾患の危険因子である。
そして、その動脈硬化症の原因には高コレステロール血症や血栓形成等がある。
高コレステロール血症は通常、総コレステロール値が高い場合をいうが、血中コレステロールはその比重によりHDL、LDL、VLDLなどに分類され、動脈硬化の危険因子として重要なのはLDLであり、HDLは逆に動脈硬化の防止に役立っているといわれている。従って血中HDLコレステロール量を上昇させることが動脈硬化の予防には重要である。

ところで、動脈硬化症は治療することよりも、日常生活の食事により予防することが重要であるため、日常生活において摂取することのできるものであって、かつHDLを上昇させることのできるものが求められている。

【課題を解決するための手段】
本発明者は、食後の血中中性脂肪の上昇を抑制することが知られているジアシルグリセロールに着目し、その血中HDLコレステロール量に与える影響について検討してきたところ、ジアシルグリセロールが優れたHDLコレステロール上昇作用を有し、日常生活において摂取の容易な健康指向食品や医薬として有用であることを見出した。

すなわち、本発明は、ジアシルグリセロールの構成脂肪酸中、(シス型不飽和脂肪酸量)/(飽和脂肪酸量+トランス型不飽和脂肪酸量)が6以上であるジアシルグリセロールを35重量%以上、及び植物ステロールを0.05重量%以上含有する油脂組成物を有効成分とするHDLコレステロール上昇剤を提供するものである。

【発明の実施の形態】
本発明に用いられるジアシルグリセロールの構成脂肪酸は、炭素数8〜24、特に16〜22であることが好ましい。全構成脂肪酸中不飽和脂肪酸を70重量%(以下単に%で示す)以上とすることが望ましい。更に、全構成脂肪酸中不飽和脂肪酸を70%以上、かつω3系高度不飽和脂肪酸が15%以上であるジアシルグリセロールがより好ましい。特にHDLコレステロール上昇効果をより高めるという点から、(シス型不飽和脂肪酸量)/(飽和脂肪酸量+トランス型不飽和脂肪酸量)〔(シス)/(トランス+飽和)重量比〕≧6の範囲が好ましい。より好ましい重量比〔(シス)/(トランス+飽和)〕は、8以上であり、更に好ましくは9以上である。また、ジアシルグリセロールの構成脂肪酸のうち、トランス型不飽和脂肪酸含量は5%以下が特に好ましく、飽和脂肪酸含量も5%以下が特に好ましい。ここで、シス型不飽和脂肪酸としては、オレイン酸、α−リノール酸、α−リノレン酸、シス−ジホモγ−リノレン酸、シス−アラキドン酸、シス−エイコサペンタエン酸、シス−ドコサヘキサエン酸などが挙げられる。またトランス型不飽和脂肪酸としては上記不飽和脂肪酸のトランス型が挙げられる。飽和脂肪酸としては、パルミチン酸、ステアリン酸、アラキン酸等が挙げられる。

ジアシルグリセロールは、例えば上記目的の構成脂肪酸を有する油脂とグリセリンとをエステル交換反応に付すか、あるいは目的の構成脂肪酸又はそのエステルとグリセリンとの混合物にリパーゼを作用させてエステル化反応を行うことにより製造される。反応中の異性化を防止するうえで、リパーゼを用いたエステル化反応がより好ましい。また、リパーゼを用いたエステル化反応によっても、反応終了後精製手段における異性化を防止するため、精製手段も脂肪酸の異性化が生起しないような穏和な条件で行うのが好ましい。

上記の方法により得られる組成物は通常、ジアシルグリセロールを含有する油脂組成物である。本発明においては、このようなジアシルグリセロールを含有する油脂組成物を用いるのが好ましい。
このような油脂組成物としては、血中中性脂肪の上昇抑制効果及びHDLコレステロール上昇作用の点から、ジアシルグリセロールを35%以上、50%以上、また60%以上、特に80%以上含有するものが好ましい。

植物ステロールはコレステロール低下効果を有する成分であり、通常の植物油に0.05〜1.2%程度含まれている。従って、前記の油脂組成物において、通常の植物油と同等のコレステロール低下効果を得るためには植物ステロール含量は0.05%以上、特に0.3%以上が好ましい。ジアシルグリセロールを含む油脂組成物中の植物ステロール含量は、その製造法によって異なる。例えば、一般に市販されている蒸留して得られた脂肪酸を原料として用いた場合には、組成物中の植物ステロール量は低下してしまう。このような場合には植物ステロールを0.05%以上になるように添加するのが好ましい。また植物ステロール含量の上限は特に限定されない。通常の植物油と同等のコレステロール低下を目的とする場合には0.05〜1.2%の範囲にあればよく、通常の植物油以上のコレステロール低下を目的とする場合には1.2%以上添加してもよい。ここで植物ステロールとしては、例えばα−シトステロール、β−シトステロール、スチグマステロール、カンペステロール、α−シトスタノール、β−シトスタノール、スチグマスタノール、カンペスタノール、シクロアルテノール等のフリー体、及びこれらの脂肪酸エステル、フェルラ酸エステル、桂皮酸エステル等のエステル体が挙げられる。

前記油脂組成物に含まれる他の成分は、トリアシルグリセロール及びモノアシルグリセロールである。モノアシルグリセロール含量は2%以下、特に1.5%以下であるのが好ましい。残部はほとんどがトリアシルグリセロールである。

後記試験例に示す如く、ジアシルグリセロール及びこれを含む油脂組成物は、優れた血中HDLコレステロール上昇作用を有し、かつ日常摂取しやすいという特性を有することから血中HDLコレステロール上昇を目的とした健康指向の食品として、また医薬として有用である。

本発明のHDLコレステロール上昇剤には、通常の油脂組成物に含まれる成分、例えばトコフェロール、アスコルビン酸パルミテート、アスコルビン酸ステアレート、BHT、BHA、リン脂質等の抗酸化剤、ショ糖脂肪酸エステル、ポリグリセリン脂肪酸エステル、有機酸モノグリセリド等の乳化剤等を配合してもよい。

また、通常ジアシルグリセロールでは同じ脂肪酸組成のトリアシルグリセロールと比較して融点上昇が生じるが、上記の組成物では常温で液状となり、食用油として広く使用できるという利点もある。従って、本発明のHDLコレステロール上昇剤は、調理油及び種々の油脂加工食品として好適に使用することができる。加工食品の例としてはドリンク、デザート、アイスクリーム、ドレッシング、トッピング、マヨネーズ等の水中油型油脂加工食品;マーガリン、スプレッド等の油中水型油脂加工食品;ピーナッツバター、フライング、ベーキングショートニング等の加工油脂食品;ポテトチップ、スナック菓子、ケーキ、クッキー、パイ、パン、チョコレート等の加工食品;ベーカリーミックス;加工肉製品;冷凍アントレ;冷凍食品等に利用することができる。また、カプセル、錠剤等の形態でも利用可能である。
本発明のHDLコレステロール上昇剤の1日摂取量は、ジアシルグリセロールとして1〜25g、更に5〜20g、特に6〜15gが好ましい。

カラムクロマトグラフ(和光純薬工業社製、ワコーゲルC−200とヘキサンでトリグリセリド画分を落とした後、ヘキサン/エーテル=70/30にてジアシルグリセロール画分を得た)により油脂中のジアシルグリセロール画分を集め、常法によりメチルエステル化後、キャピラリーカラム(クロムパック社製、CP−SIL88)を用い、ガスクロマトグラフィーにて分析した。

試験例1
