本発明は各種成分による皮膚、毛髪および粘膜への刺激を緩和する皮膚細胞毒性緩和剤に関する。

界面活性剤は化粧品などの皮膚外用剤に洗浄成分として、また、クリーム等の乳化剤として広く用いられている。しかし、界面活性剤には皮膚や毛髪、粘膜に対する刺激性を有するという欠点がある。この刺激を緩和するため、アミノ酸系で低刺激性の界面活性剤の開発が進められているが(特公昭50−40125号公報、特開平8−60189号公報)、いまだ十分ではない。また、化粧品等に用いられる抗菌剤、防腐剤は製品の品質保持のために広く用いられるが、反面、皮膚刺激性を有する。

この他にもアルコール類、増粘剤、色素、香料、油脂、植物抽出物等、刺激性を有する成分が化粧品等の皮膚外用剤には多く含まれる。 そのための皮膚粘膜刺激緩和剤として、トレハロースモノ脂肪酸エステル(特開平10−45560号公報)等、アルコール刺激緩和剤として、フェニルエチル−α−グルコシド、フェニルエチル−β−グルコシドのようなグルコース誘導体(特開平8−283121号公報)等が知られている。

近年、環境の変化に伴い、アトピー性皮膚炎などのアレルギー疾患者や、いわゆる敏感肌と感じる人が増加し、これらの人は皮膚が過敏になっているため、化粧品などにより発赤、かゆみといった刺激を感じることが多く、より低刺激な化粧品が求められる。

本発明者は上記のような課題を解決するため研究を行い、その結果、カルダモン、クララ、ボタンボウフウ、ガジュツ、カミツレ、ガラナ、プエラリア、田七人参、ダッタンソバ、ナツメグ、コショウ、ショウガ、桃仁の抽出物に界面活性剤の皮膚細胞毒性緩和作用があることを見出し、本発明を完成した。

カルダモン(Elettaria cardamomum)はショウガ科ショウズク属の植物で、別名しょうづくとも呼ばれる。スパイスの他、媚薬、健胃剤としても用いられる。

クララ(Sophora flavescens)はマメ科エンジュ属の植物で、別名クサエンジュとも呼ばれる。黄疸、下血、健胃薬に用いられる。

ボタンボウフウ(Peucedanum japonicum)はセリ科カワラボウフウ属の植物で、鎮咳、鎮静、利尿、強壮に用いられる。

ガジュツ(Curcuma zedoaria)はショウガ科ウコン属の植物で、健胃、去痰薬として用いられる。

カミツレ(Matricaria chamomilla)はキク科シカギク属の植物で別名カミルレとも呼ばれる。発汗、駆虫に効果がある。

ガラナ(Paullinia cupana)はムクロジ科ガラナ属の植物で、お茶、清涼飲料として用いられている。

プエラリア(Pueraria mirifeda)はマメ科クズ属の植物で、タイ北部に自生する熱帯植物である。イソフラボン誘導体を主成分とする。

田七人参(Panax notoginseng)はウコギ科トチバニンジン属の植物で、別名三七人参とも呼ばれる。止血、鎮痛、消炎などに用いられる。

ダッタンソバ(Fagopyrum tataricum)はタデ科ソバ属の植物で、別名ニガソバとも呼ばれる。種子は食用に用いられる。ルチンを含有し、胃痛、消化不良、下痢などに用いられる。

ナツメグはニクズク科ニクズク属の植物ニクズク(Myristica fragrans)の仮種皮を除いた種子で、スパイスとして広く用いられる。

コショウ(Piper nigrum)はコショウ科コショウ属の植物で、スパイスとして用いられる他、健胃消化、胃痛、腹痛などに用いられる。

ショウガ(Zingiber officinale)はショウガ科ショウガ属の植物で、食用に用いられる他、健胃、新陳代謝機能促進、などに用いられる。

桃仁はバラ科サクラ属の植物モモ(Prunus percica)の種子で、漢方では婦人病、産後腹痛、打身に用いられる。

本発明に用いる植物抽出物は常法に従い、植物の葉、茎、果実または全草を乾燥、必要に応じて焙煎後、例えば水あるいは水性有機溶媒にて、室温あるいは50〜100℃にて抽出して得られた抽出液をろ過後、そのまま、もしくは濃縮、乾燥して粉末として用いることが可能である。ただし、これは抽出例であり、これに限定されることはない。

本発明の植物抽出物の配合量は必ずしも制限されないが、化粧料等の製剤全量に対し、0.001〜30重量%が好ましく、1種類以上を組み合わせて使用することもできる。本発明の皮膚細胞毒性緩和剤は、例えば、皮膚洗浄料、頭髪洗浄料、ローション、スキンクリーム、乳液、リンス、コンディショナー、台所用洗剤、洗濯用洗剤、歯磨等の化粧品、医薬品、医薬部外品等の人に適用される組成物に用いることが可能であり、敏感肌用の組成物に配合することができる。更に必要に応じて本発明の効果を損なわない範囲において、油分、色素、香料、防腐剤、殺菌剤、保湿剤、増粘剤、酸化防止剤、界面活性剤、粉末成分、水、植物抽出物その他の各種添加剤と併用することができる。

本発明の皮膚細胞毒性緩和剤によると、界面活性剤、抗菌剤、防腐剤のような化粧料、医薬、洗剤の原料等による刺激を緩和することができる。また、本発明の植物抽出物を配合することにより、皮膚細胞を各種刺激より防御し、各種刺激に対する皮膚バリアーとなって皮膚を保護することができる。

次に実施例によって本発明を更に詳細に説明するが、本発明はこれら実施例に限定されるものではない。刺激緩和作用の試験では刺激の指標として界面活性剤を用いた。

(植物抽出物の調製)植物の乾燥物をDIONEX社製高速溶媒抽出装置ASE-200を用いて100℃、1500psiで10分間熱水抽出し、凍結乾燥したものを以下の試験に用いた。

(刺激緩和作用評価)植物抽出物について、界面活性剤による刺激緩和作用を、以下の方法により評価した。培養ヒト表皮角化細胞に0.005%ラウリル硫酸ナトリウム水溶液もしくは各濃度の植物抽出物を添加した0.005%ラウリル硫酸ナトリウム水溶液を作用させ、細胞生存量をMTT還元法により測定した。MTT還元法とは、MTT[3-(4,5-dimethylthiazol -2-yl)-2,5-diphenyl tetrazolium bromide]がミトコンドリア内のコハク酸脱水素酵素などにより青色の産物に還元されるが、この生成物を溶解し呈色させたあとでこの吸光度を測定し、生細胞量の指標とする方法である。刺激緩和率を次式より求め評価した。
