【発明の属する技術分野】
本発明は、インジェクション装置に関し、簡易に、しかも確実且つ迅速に試料を分離することが可能な分取液体クロマトグラフに用いられるインジェクション装置に関する。

【従来の技術】
移動相として液体溶媒(溶離液)を用いる液体クロマトグラフにおいては、溶離液を送出するポンプと固定相が充填されたカラムとの間に多方弁であるインジェクションバルブを介してインジェクターが接続され、インジェクターから試料(サンプル)および溶離液がカラムに供給される構成になっている。カラム内に導入されたサンプル内の各成分はそれぞれカラムを通過する時間が異なるため、カラムから排出される時点では各成分が分離されることになる。

この際、分離するサンプルにより、通常2〜4種類の溶離液を混合することにより溶離液の成分比率を変えるのが一般的である。しかも、カラムから排出されるサンプルの各成分が重ならないように、且つ短時間で分離が行われるようにするため、溶離液の混合比を段階状にまたは徐々に変化させることが行われている(グラジエント法)。

係る液体クラマトグラフィにおいて、インジェクターとしては、コイル状に巻回されたチューブ、または、内部に可動栓、遊走栓および固定栓がこの順番で配置されていると共に遊走栓との間隙から液体が流通するようにリング状の凹部が設けられた筒状シリンダ(実公平1−41473号公報参照)が用いられるのが一般的である。

【発明が解決しようとする課題】
しかしながら、インジェクターとしてチューブを用いた場合には、注射器を用いてチューブ内にサンプルを導入する必要が生じてその作業が煩雑となってしまうと共に注射器に残るサンプルが無駄になってしまう。

他方、インジェクターとして筒状シリンダを用いた場合には、以下のような問題が生じる。まず、サンプルと溶離液との界面の面積が比較的大きくなるために、サンプルと溶離液とが混合されサンプルの濃度が大幅に低下してしまうことがある。次に、グラジエント法により液体クロマトグラフを行う際に、異なる比率で混合された溶離液どうしがインジェクター内で混合されて溶離液の成分比率が所定の比率から大きく外れてしまう。これを防止するためには、インジェクター内のサンプルをカラムに入れ終えた段階で、溶離液がインジェクターを通過しないでカラムに供給されるようにインジェクションバルブを切換える必要がある。しかしながら、これを自動的に行おうとするとインジェクションバルブの切り換えタイミングを計るためのセンサを配置しなければならず装置構成が複雑になってしまう。さらに、サンプルが保持される固定栓と遊走栓との間にサンプルと共に不可避的に空気が含まれてしまい、この空気がカラムに送り込まれてカラム内の充填剤が乾燥したり、サンプルの結晶化が起こったりするおそれがある。

そこで、本発明の主な目的は、インジェクター内にサンプルを無駄なく簡易に導入することができ、濃度の大幅な低下を引き起こすことなくサンプルをカラムに供給することができ、溶離液の成分比率が所定の比率から大きく外れるのを防止しつつ装置構成を複雑にすることなく自動化が可能であると共に、カラムに空気が送り込まれることがないインジェクション装置を提供することである。

【課題を解決するための手段】
上記目的を達成するために、請求項1記載のインジェクション装置は、充填剤が充填されたシリンダ、および、これと着脱可能に組み合わされることによって実質的にゼロから所定値までの範囲で容積可変な閉鎖空間を形成するピストンを有していると共に、前記閉鎖空間の容積が実質的にゼロの状態において前記シリンダ内の充填剤と連通する第1および第2の連通孔が設けられたインジェクターと、溶媒を送出するポンプに接続される第1の接続口、サンプルの分離が行われるカラムに接続される第2の接続口、前記インジェクターの第1の連通孔に接続された第3の接続口、および、前記インジェクターの第2の連通孔に接続された第4の接続口を有しており、前記第1の接続口と前記第2の接続口との間が連結される第1の状態と、前記第1の接続口と前記第3の接続口との間および前記第2の接続口と前記第4の接続口との間が連結される第2の状態とを切換え可能なインジェクションバルブとを備えている。

このように、請求項1において、インジェクションバルブは、第1の状態と第2の状態とに切り換えられるため、第1の状態で溶離液を送出するポンプとカラムとを接続しカラムウェッティングを行い、第2の状態で溶離液を送出するポンプとインジェクターとを接続し、インジェクターとカラムとを接続することで、インジェクターに溶離液を送出し、インジェクターからカラムへサンプルを入れることができる。

請求項1によると、ピストンをシリンダから取り外した状態でシリンダ内に直接サンプルを流し込むことができるので、インジェクター内にサンプルを導入する作業を簡易に行うことができて、しかもサンプルの無駄が生じることもない。また、シリンダ内に流し込まれたサンプルは充填剤に吸着されるので、インジェクター内に溶離液が供給されてもサンプルと溶離液とが混合することがほとんどなく、サンプルの濃度が大幅に低下するのが抑制される。また、グラジエント法を行う際にインジェクター内に成分比率の異なる溶離液が順次導入されたとしても、シリンダ内に充填剤が充填されているために、溶離液どうしが混合してその成分比率が所定の比率から大きく外れることがなくなる。そのため、インジェクター内のサンプルをカラムに入れ終えた段階で、溶離液がインジェクターを通過しないでカラムに供給されるようにインジェクションバルブを切換える必要がなくなり、センサを配置しなくても装置動作の自動化を実現することが可能となる。さらに、充填剤とピストンとの間に形成される閉鎖空間の容積を実質的にゼロとすることができるので、カラムに空気が送り込まれることがなくなる。そのため、カラム内の充填剤が乾燥したり、サンプルの結晶化が起こったりするのが抑制される。

なお、本発明において、特開平2000−275228号公報に記載されているように、複数のインジェクターおよびカラムを並列に並べ、インジェクションバルブでそのいずれかを選択可能とするようにしてもよい。特開平2000−275228号公報のインジェクション装置は、4つのインジェクションバルブ(ロータリーバルブ)に複数のインジェクターおよびカラムを接続し、これらのロータリーバルブを順次切換えて、インジェクター内のサンプルを順次任意のカラムに送出できるようにしたものである。このとき、1つのインジェクターと1つのカラムに対応させる必要は必ずしもなく、複数のインジェクターに1つのカラムを対応させることもできる。

【発明の実施の形態】
以下、図面を参照しつつ本発明の好適な実施の形態を説明する。

図1は、本発明の第1の実施の形態のインジェクション装置を含む液体クロマトグラフ装置の概略的な模式図である。この液体クロマトグラフ装置は、インジェクション装置1と、溶離液供給機構2と、カラム14と、検知器15とを有する。

インジェクション装置1は、インジェクター3と、インジェクションバルブ4とを有する。図2にインジェクター3の断面図を示す。インジェクター3は、充填剤3eが充填されかつ底部に連通孔3dを有するシリンダ3aと、上部に連通孔3eを有するピストン3bと、ピストン3bの連通孔3eに螺合されるボルト3gとを含んでいる。ピストン3bはシリンダ3aに着脱可能であり、ボルト3gを連通孔3eから取り外した状態でピストン3bをシリンダ3aに押し込むことで両者の間の閉鎖空間3fの容積をほぼゼロにまで変化させることができる。

インジェクター3内にサンプルを導入するには、ピストン3bをシリンダ3bから外し、サンプルを充填剤3cの上部に流し込み、サンプルが充填剤3cに吸着されたらボルト3gを取り外して連通孔3eから空気を抜きながらピストン3bを閉鎖空間3fがほぼゼロになるまで押し下げる。このような簡易な作業を行うことで、インジェクター3内に空気が残ることを防止しつつ、しかも注射器などを用いずに無駄なくインジェクター3にサンプルを導入することができる。さらに、ピストン3bをシリンダ3aに近づけていくことによって両者の間に形成された閉鎖空間3fの容積を実質的にゼロとすることができるので、後述するように、カラム14に空気が送り込まれることがなくなる。そのため、カラム14内の充填剤が乾燥したり、サンプルの結晶化が起こったりするのが抑制される。

本実施の形態のインジェクション装置1においては、インジェクションバルブ4として、6つの接続口V1〜V6を有し、接続口V1と接続口V2、接続口V3と接続口V4、接続口V5と接続口V6がそれぞれ連結される溶離液送出状態(第2の状態)と、接続口V2と接続口V3、接続口V4と接続口V5、接続口V6と接続口V1がそれぞれ連結されるカラムウェッティング状態(第1の状態)に切換え可能である六方バルブが用いられている.。図1に示すように、インジェクションバルブ4の接続口V2とインジェクター3の連通孔3d、および、接続口V5とインジェクター3の連通孔3eとがそれぞれ接続される。

溶媒供給機構2は、ポンプ5と、異なる成分を含む溶離液A〜Dがそれぞれ蓄えられた容器6〜9と、容器6〜9からの溶離液A〜Dの供給をそれぞれ制御する電磁弁10、11、12と、混合器13とを有する。電磁弁10〜12は三方電磁弁であり、電磁弁10は溶離液AとBのいずれかを選択して後段に送ることができるようにされており、電磁弁11は溶離液CとDのいずれかを選択して後段に送ることができるようにされている。そして、電磁弁12は電磁弁10の出口と電磁弁11の出口のいずれかを選択して混合器13に送るようにされている。混合器13において溶離液は混合され、混合された溶離液がポンプ5によりインジェクションバルブ4へと送出される。

カラム14には充填剤が充填されており、その一方の端部が検知器15に接続されている。液体クロマトグラフィの実行によりサンプルはカラム14内にて分離され、検知器15により分析され、分析結果が表示される。

図1に示す液体クロマトグラフ装置においては、溶離液供給機構2がインジェクションバルブ4の接続口V1に、カラム14がインジェクションバルブ4の接続口V2にそれぞれ接続されている。

本実施の形態のインジェクション装置1において、インジェクター3は、図3および図4に示すような支持部材40に支持されてよい。図3は、支持部材40の断面図であり、図4はその正面図である。これらの図面に示すように、支持部材40は、水平方向に延在した土台41と、土台41に支持されており上方に延在した2本の支持柱42および43と、支持柱42、43に挿入されかつその上端部近傍に支持されて水平方向に延在したピストン押え46と、支持柱42、43に挿入されかつそのやや上方よりの部分に支持されて水平方向に延在したシリンダ押え47と、ボルト50、51によってシリンダ押え47の下方に固定されたアーム48と、ピストン押え46およびシリンダ押え47と螺合した調節具52とを備えている。

土台41の一端には、インジェクター3と接続される接続口53aおよびチューブを介してインジェクションバルブ4と接続される接続口53bを有する接続部53が取り付けられている。また、支持柱42、43のピストン押え46とシリンダ押え47との間の部分には、コイルバネ44、45が嵌挿されている。また、シリンダ押え47には、これを所望の高さで固定するためのボルト49が螺合されている。シリンダ押え47とピストン押え46との間の距離は、コイルバネ44、45の弾性力に抗して調節具52を回動させることによって調節することができるようになっている。

支持部材40にインジェクター3を取り付けるには、まず、インジェクター3の連通孔3dを接続部53の接続口53aにはめ込み、シリンダ3aを上方から押える位置にシリンダ押え47を移動させた後、これをボルト49で固定する。次に、ピストン押え46が上方からピストン3bを押さえつけるようになるまで調節具52を回動させる。インジェクター3を取り外すには、上述したのと逆の動作を行えばよい。このように、支持部材40を用いることで、予めサンプルが導入されたインジェクター3を比較的簡易な作業で固定配置することができる。また、支持部材40はシリンダ3aに対してピストン3bを押さえ込むことができるような構成となっているので、閉鎖空間3fの容積を確実にゼロ付近にまで減少させることができて、クロマトグラフの準備段階においてインジェクター3内に空気が残ってしまうことを効率よく防止することができる。

次に、上述した装置により液体クロマトグラフを行う工程を説明する。最初、インジェクションバルブ4はカラムウェッティング状態にあり、接続口V1と接続口V6とが連結されることによって溶媒供給機構2とカラム14とがインジェクションバルブ4を介して連結されている。図1において、矢印A1は、カラムウェッティング状態におけるインジェクションバルブ4を通過する溶離液の流れを示す。この矢印A1を参照することから明らかなように、カラムウェッティング状態では、溶媒供給機構2からインジェクションバルブ4を介してカラム14に溶離液が送出され、カラム14内の充填剤14bが溶離液によりウェッティングされる。

カラム14内の充填剤が充分にカラムウェッティングされたら、インジェクションバルブ4を溶離液送出状態に切り換える。すると、接続口V1と接続口V2および接続口V5と接続口V6とが連結されることによって、溶媒供給機構2とインジェクター3の連通孔3dとがインジェクションバルブ4を介して連結されると共にインジェクター3の連通孔3eとカラム14とがインジェクションバルブ4を介して連結される。図1において、矢印A2は、溶離液送出状態におけるインジェクションバルブ4を通過する溶離液(およびサンプル)の流れを示す。この矢印A2を参照することから明らかなように、溶離液供給機構2から送出される溶離液は接続口V1からインジェクションバルブ4に入り接続口V2から出てインジェクター3に送られる。そして、連通孔3dからインジェクター3内に溶離液が注入されることにより、インジェクター3内の充填剤3cに吸着されていたサンプルが成分ごとに分離されて連通孔3eから送出される。

このとき、上述したようにピストン3bをシリンダ3aの間に形成された閉鎖空間3fの容積が実質的にゼロとなっているので、サンプルと共に或いはこれに先だってカラム14に空気が送り込まれることがなく、カラム14内の充填剤が乾燥したり、サンプルの結晶化が起こったりするのが抑制される。さらに、シリンダ3a内に充填剤3cが充填されているために、シリンダ3aの断面積が比較的大きいにもかかわらず、溶離液とサンプルとがほとんど混合することがない。そのため、元々の濃度からほとんど低下していない濃度を有するサンプルが連通孔3eから送出されることになる。

また、溶離液送出状態では、インジェクションバルブ4の接続口V5と接続口V6とが連結されているため、連通孔3eから送出されたサンプルは、接続口V5からインジェクションバルブ4に入り接続口V6から出てカラム14に送出される。カラム14内の充填剤によってサンプルは成分ごとに分離されてカラム14から排出される。カラム14から排出された成分に分けられたサンプルは検知器15に供給される。検知器15は、各成分の定量分析を行ってその結果を表示する。

また、溶離液送出状態では、グラジエント法にしたがい電磁弁10〜12を適宜切り換えることによって、混合器13内の溶離液の成分比率を変更しつつインジェクター3に溶離液を送出する。このとき、シリンダ3a内に充填剤3cが充填されているために、異なる混合比の溶離液どうしが混合することがほとんどなく、各溶離液の成分比率は所定の比率から大きく外れることはない。従って、インジェクター3からカラム14へのサンプルの供給が終了してもインジェクションバルブ4を溶離液送出状態からカラムウェッティング状態に切り換えて溶離液の経路を変更する必要がない。よって、本実施の形態のインジェクション装置1を用いることにより、溶離液供給機構2において、溶離液A〜Dの混合比、混合液それぞれの送出時間を予め設定しておけば、液体クロマトグラフィの開始から終了までの工程を自動化することができる。しかも、上述した筒状シリンダを用いた場合のようにインジェクションバルブの切り換えタイミングを計るためのセンサを配置する必要がなく、装置構成を簡略なものとすることができる。

次に、本発明の第2の実施の形態について説明する。図5は、本実施の形態のインジェクション装置を含む液体クロマトグラフ装置の概略的な模式図である。この液体クロマトグラフ装置は、インジェクション装置31と、溶離液供給機構32と、カラム14と、検知器15とを有している。インジェクション装置31および溶離液供給機構32以外の部材は第1の実施の形態と同様に構成されているので、ここではその詳細な説明を省略する。なお、図5において、矢印B1は、カラムウェッティング状態における溶離液の流れを、矢印B2は溶離液送出状態における溶離液(およびサンプル)の流れをそれぞれ示している。

図5に示すように、インジェクション装置31は、6本のインジェクター21〜26(それぞれが図2に示すインジェクター3と同様に構成されている)と、2つのロータリーバルブ17、18と、インジェクションバルブ4とを備えている。インジェクションバルブ4の接続口V1には溶離液供給機構32が接続されており、インジェクションバルブ4の接続口V2にはカラム14が接続されている。

ロータリーバルブ17および18はそれぞれ7つの接続口RV1〜7を有している。ロータリーバルブ17の接続口RV7は、インジェクションバルブ4の接続口V2に接続されている。ロータリーバルブ18の接続口RV7は、インジェクションバルブ4の接続口V5に接続されている。ロータリーバルブ17には、これを回動させるためのステッピングモータSM1が接続されており、ロータリーバルブ18にはステッピングモータSM2が接続されている。ロータリーバルブ17、18内においては、中心にある接続口RV7と、6つの接続口RV1〜6のいずれか1つとが選択的に連結される。接続口RV7と連結される接続口は、ステッピングモータSM1、SM2によってロータリーバルブ17、18を回動させることで切り換えられる。ロータリーバルブ17および18の接続口RV7はインジェクションバルブ4の接続口V2および接続口V5にそれぞれ接続されている。ロータリーバルブ17の接続口RV1〜6はそれぞれインジェクター21〜26の一方の端部に接続されている。また、ロータリーバルブ18の接続口RV1〜6はそれぞれインジェクター21〜26の他方の端部に接続されている。

ステッピングモータSM1およびSM2は図示しない駆動回路を介して例えばパーソナルコンピュータである制御装置に接続されている。この制御装置によってステッピングモータSM1およびSM2を制御することで、ロータリーバルブ17、18を介してインジェクションバルブ4と連結される任意のインジェクター21〜26を選択することが可能となっている。

上記の液体クロマトグラフ装置を用いると、インジェクションバルブ4を切り換えて任意のインジェクター21〜26を選択した後に上述した第1の実施の形態と同様の工程を行うことで、選択したインジェクター21〜26に含まれるサンプルについて液体クロマトゲラフィを行なうことができる。この場合、カラムウェッティングの後インジェクションバルブ4をカラムウェッティング状態から溶離液送出状態に切換えれば、その後インジェクションバルブ4を切換える必要がなく、センサを配置しなくても装置動作を自動化することができる。

また、本実施の形態のような液体クロマトグラフ装置では、ステッピングモータSM1、SM2を自動制御することにより、6つのインジェクター21〜26に含まれるサンプルについて順次自動的に液体クロマトゲラフィを行なうことができるようになる。このようにインジェクター21〜26の選択を自動化することで、無人での動作が可能であり、省力化に著しい効果がある

以上、本発明の好適な実施の形態について説明したが、本発明は上述の実施の形態に限られるものではなく、特許請求の範囲に記載した限りにおいて様々な設計変更が可能なものである。例えば、インジェクターは、必ずしも図示されたような形状のものでなくてもよく、図示された以外の形状を有していてもよい。また、インジェクションバルブとしては、上述した実施の形態で使用したような六方バルブに限らず、様々なタイプのものを使用することができる。

また、支持部材としては、図3および図4で説明した形態に限らず、インジェクターを安定して着脱可能に保持させることができるものであればよい。また、支持部材は、1本とは限らず複数本のインジェクターを安定して保持させるものであってもよい。

【発明の効果】
以上説明したように、請求項1によると、ピストンをシリンダから取り外した状態でシリンダ内に直接サンプルを流し込むことができるので、インジェクター内にサンプルを導入する作業を簡易に行うことができて、しかもサンプルの無駄が生じることもない。また、シリンダ内に流し込まれたサンプルは充填剤に吸着されるので、インジェクター内に溶離液が供給されてもサンプルと溶離液とが混合することがほとんどなく、サンプルの濃度が大幅に低下するのが抑制される。また、グラジエント法を行う際にインジェクター内に成分比率の異なる溶離液が順次導入されたとしても、シリンダ内に充填剤が充填されているために、溶離液どうしが混合してその成分比率が所定の比率から大きく外れることがなくなる。そのため、インジェクター内のサンプルをカラムに入れ終えた段階で、溶離液がインジェクターを通過しないでカラムに供給されるようにインジェクションバルブを切換える必要がなくなり、センサを配置しなくても装置動作の自動化を実現することが可能となる。さらに、充填剤とピストンとの間に形成される閉鎖空間の容積を実質的にゼロとすることができるので、カラムに空気が送り込まれることがなくなる。そのため、カラム内の充填剤が乾燥したり、サンプルの結晶化が起こったりするのが抑制される。このように、請求項1によると、実際に液体クロマトグラフィを行う際に、その利便性が大幅に向上したり、検出精度がよくなるなどの様々な実用的な利益を得ることができる。

BACKGROUND OF THE INVENTION
The present invention relates to an injection apparatus, and more particularly to an injection apparatus used for a preparative liquid chromatograph capable of separating a sample easily, surely and quickly.

In a liquid chromatograph that uses a liquid solvent (eluent) as a mobile phase, an injector is connected via a multi-way injection valve between a pump that delivers the eluent and a column packed with a stationary phase. The sample (sample) and the eluent are supplied to the column. Since each component in the sample introduced into the column has a different time for passing through the column, each component is separated when discharged from the column.

On the other hand, when a cylindrical cylinder is used as an injector, the following problems occur. First, since the area of the interface between the sample and the eluent becomes relatively large, the sample and the eluent may be mixed and the concentration of the sample may be greatly reduced. Next, when liquid chromatography is performed by the gradient method, the eluents mixed at different ratios are mixed in the injector, and the component ratio of the eluent greatly deviates from a predetermined ratio. In order to prevent this, it is necessary to switch the injection valve so that the eluent is supplied to the column without passing through the injector when the sample in the injector has been put into the column. However, if this is to be done automatically, a sensor for measuring the switching timing of the injection valve must be arranged, resulting in a complicated apparatus configuration. In addition, air is unavoidably included with the sample between the fixed stopper that holds the sample and the migratory stopper, and this air is fed into the column to dry the packing material in the column or to crystallize the sample.
