Description
a)は、非還元性末端においてタンパク質反応性アルデヒドを形成するための、過ヨウ素酸塩を用いた、コロミン酸(E.coliからの、アルファ−2,8結合ポリシアル酸)の酸化を示し、
b)は、タンパク質アミノ基との安定で不可逆性の共有結合を形成するための、ソディウムシアノボロヒドリドを用いた、シッフ塩基の選択的還元を示す。
i)非対称二量体;
ii)線状ポリマー;
iii)分枝状ポリマー;及び
iv)様々なより複雑な構造
の形成を示す、詳細な概略図である。
a)還元性末端シアル酸ユニットの環を還元的開環し、それによってビシナルジオール基を形成するための還元;及び
b)ステップa)で形成されるビシナルジオール基を酸化してアルデヒド基を形成するための選択的酸化
の連続するステップに供される、シアル酸化合物のアルデヒド誘導体を生産する新規な方法が提供される。
c)非還元性末端のシアル酸ユニットを第7,8ビシナルジオール基において酸化し、第7アルデヒドを形成するための選択的酸化ステップ;及び、

ここで、R2は、二塩基性有機基、例えば、アリーレンオリゴ(アルコキシ)アルカンまたは、好ましくはアルカンジイル基、例えば、C2−12アルカンジイル基である。

Gly1Oはグリコシルであり;そして、
R5は有機基、好ましくは末端還元性サッカライドユニットの還元形態、アルデヒドまたはそのようなアルデヒドの反応生成物(例えば、アミンまたはヒドラジド)であるそれらの酸化された誘導体である、
第2方法の側面による方法の生成物である新規化合物が提供される。
炭酸アンモニウム、エチレングリコール、ポリエチレングリコール(8KDa)、ソディウムシアノボロヒドリド(>98%純度)、メタ過ヨウ素酸ナトリウム及び分子量マーカーは、Sigma Chemical Laboratory(英国)から入手した。使用されるコロミン酸、鎖状α−(2→8)結合E.coli K1ポリシアル酸(平均22.7kDa、高多分散性1.34、39kDa p.d. 1.4;11kDa、p.d. 1.27)はCamida(アイルランド)から、放射性ヨウ化物(Na125I)はAmersham(英国)から入手した。他の材料は、2,4ジニトロフェニルヒドラジン(Aldrich Chemical Company、英国)、透析チューブ(3.5KDa及び10KDaのカットオフ限界;Medicell International Limited、英国)、セファロースSPハイトラップ(HiTrap)、PD−10カラム(Pharmacia、英国)、トリス−グリシンポリアクリルアミドゲル(4〜20%及び16%)、トリス−グリシンドデシル硫酸ナトリウムランニングバッファー及びローディングバッファー(Novex、英国)を含む。脱イオン水は、エルガスタットオプション4水精製ユニット(Elgastat Option 4 water purification unit)(Elga Limited、英国)から得た。使用される全ての試薬は、分析用グレードである。プレートリーダー(Dynex Technologies、英国)を、タンパク質またはCAアッセイにおける分光学的測定のために用いた。CD1非近交系マウス(8〜9週齢;体重29〜35g)は、Charles River(英国)から購入し、これらを使用する前に少なくとも1週間順化させた。
タンパク質及びコロミン酸測定
レゾルシノール試薬を用いたポリシアル酸(シアル酸としての)の量的評価は、別記されるようなレゾルシノール法(Svennerholm 1957)によって行われた(Gregoriadis et.al., 1993; Fernandes and Gregoriadis, 1996, 1997)。Fab(タンパク質)を、BCA比色法によって測定した。
共有結合PSA−タンパク質複合体は、天然の形態のE.coli由来のポリシアル酸(コロミン酸,CA)を用い、その弱反応性還元性末端を介して、ソディウムシアノボロヒドリドを用いて還元アミノ化することによって製造された。CA=コロミン酸; CAO=酸化されたコロミン酸(Fernandes and Gregoriadis, 1996; Jain et al., 2003におけるような)。ソディウムシアノボロヒドリド(NaCNBH3)を、4mg ml−1の濃度で用いた。
1a コロミン酸の活性化
新たに調製した0.1Mメタ過ヨウ素酸ナトリウム(NaIO4)溶液を20℃でCAと混合し(100mg CA/ml NaIO4)、該反応混合物を暗所で15分間、磁気的に攪拌した。その後、2倍体積のエチレングリコールを該反応混合物に添加して過剰のNaIO4を消費し、混合物を20℃でさらに30分間攪拌した。酸化されたコロミン酸を、4℃で、0.01%炭酸アンモニウムバッファー(pH7.4)に対して充分に(24時間)透析(カットオフ分子量3.5KDaの透析チューブ)した。限外濾過(カットオフ分子量3.5KDa以上)を、透析チューブからのCAO溶液を濃縮するために用いた。必要な体積に濃縮した後、該濾液を凍結乾燥し、後に使用するまで−40℃で保存した。
酸化されたコロミン酸(CAO;22.7kDa)を、ソディウムボロヒドリドの存在下で還元した。新たに調製した0.15mMソディウムボロヒドリド(NaBH4;希H2SO4溶液で希釈することにより、pH8〜8.5に希釈した0.1M NaOH中の)を20℃でCAOと混合し(100mg CA/ml)、該反応混合物を暗所で2時間に達するまで攪拌した。反応の完了により、pHは7に低下した。該酸化/還元されたコロミン酸(CAOR)を、4℃で、0.01%炭酸アンモニウムバッファー(pH7)に対して透析(カットオフ分子量3.5KDaの透析チューブ)した。限外濾過を、透析チューブからのCAOR溶液を濃縮するために用いた。該濾液を凍結乾燥し、さらに必要とされるまで4℃で保存した。いずれのアルデヒド含有量の測定も、「CAの酸化の測定」に記載されるようにして測定された。
アルデヒドが含まれていないことを確認した後、酸化/還元されたコロミン酸(CAOR)を再度、CAORを過ヨウ素酸塩溶液と共により長時間(1時間まで)インキュベートした以外は、「コロミン酸の活性化」に報告されているのと同様にして酸化した。該CAORO生成物における酸化の程度を、このステージから同様にして得られた凍結乾燥粉末について測定した。
コロミン酸の酸化の程度の定量的評価は、カルボニル化合物との相互作用において、やや溶けにくい2,4ジニトロフェニルヒドラゾンを生じる2,4ジニトロフェニルヒドラジン(2,4−DNPH)を用いて行われた。非酸化(CA)、酸化(CAO)、還元(CAOR)及び再酸化(CAORO)(各5mg)を、2,4−DNPH試薬(1.0ml)に添加し、該溶液を振り混ぜ、その後、結晶性沈殿が観察されるまで37℃で放置した(Shriner et al., 1980)。CAの酸化の程度(量的)は、アルカリ性溶液中のフェリシアニドイオンのフェロシアン化第二鉄(ペルシアンブルー)への還元(その後630nmで測定する)に基づく方法(Park and Johnson, 1949)を用いて測定された。この例では、グルコースを標準液(standard)として用いた。
コロミン酸試料(CA、CAO、CAOR及びCAORO)をNaNO3(0.2M)、CH3CN(10%;5mg/ml)に溶解し、2x以上のGMPWXLカラム上で、屈折率を検出しながら(GPCシステム:VE1121 GPC 溶媒ポンプ、VE3580 RI検出器及びトライセック3ソフトウェア(Viscotek Europe Ltd)による照合)、クロマトグラフした。試料(5mg/ml)を、0.45μmナイロン膜で濾過し、移動相として0.2M NaNO3及びCH3CN(10%)を用いて0.7cm/minで流した(run)。
コロミン酸(CA)(ポリシアル酸)は、鎖状のアルファ−2,8結合した、N−アセチルノイラミン酸(Neu5Ac)残基のホモポリマーである(図1a)。しかしながら、過ヨウ素酸塩は、強力な酸化剤であり、それにもかわらず、隣接した炭素原子においてヒドロキシル基を含む炭水化物に対して選択的(Fleury and Lange, 1932)であり、内部のNeu5Ac残基に時間−依存性の開裂を生じることができる。従って、本研究において、酸化のためのコロミン酸の曝露は、室温で100mMの過ヨウ素酸塩を用いて、15〜60分間に制限された(Lifely et al., 1981)。さらに、過ヨウ素酸塩が光に曝露して分解し、より反応的な種を生じる(Dyer, 1956)ので、反応混合物は暗所で保存された。過ヨウ素酸塩及びボロヒドリド処理後の内部のアルファ−2,8結合Neu5Ac残基の完全性(integrity)を、ゲル浸透クロマトグラフィーによって分析し、酸化(CAO)、酸化還元(CAOR)、2回酸化(CAORO)原料について得られたクロマトグラフを、天然のCAのそれと比較した。酸化(15分間)(CAO)(6b)、還元(CAOR)(6c)、2回酸化(1時間)(CAORO)(6d)及び天然(6a)のCAは、ほとんど同一の溶出プロフィールを示すことが見出され(図6)、連続する酸化及び還元ステップが、ポリマー鎖の顕著なフラグメント化を生じさせる証拠はないことが見出された。小さなピークは、緩衝塩を示す。
実施例2−Fab−コロミン酸複合体の調製
Fabを0.15M PBS(pH7.4)に溶解し、種々のコロミン酸(CA、CAO、CAOR及びCAORO)に、ソディウムシアノボロヒドリド(NaCNBH3)の存在下で還元アミノ化することにより、共有結合させた。CA:Fabモル比(100:1)でのFabとの合成の各ステップから得られたコロミン酸(出発原料及び実施例1a〜cそれぞれの生成物)を、35±2℃のオーブン中で、磁気的に攪拌しながら、封をした容器中で、ソディウムシアノボロヒドリド(4mg/ml)を含む0.15M PBS(pH7.4;2ml)と反応させた。該混合物を、継続的に攪拌しながら、飽和度70% w/vに達するまでゆっくり添加することにより、硫酸アンモニウム((NH4)2SO4)沈殿反応に供した。該試料(4℃で1時間攪拌した)を、15分間遠心分離(5000xg)し、ポリシアリル化されたFabを含むペレットを(NH4)2SO4の飽和溶液に懸濁し、再度15分間遠心分離した(5000xg)。回収した沈殿を、0.9% NaCl(pH7.4;PBS)を追加した1mlの0.15M Naホスフェートバッファーに再溶解し、4℃で、同様のPBSに対して充分に(24時間)透析した。その後、該透析物を、シアル酸及びFab含有量について分析し、複合体収量を、CA:Fabモル比の観点から示した。コントロールは、天然のタンパク質を、記載された条件下で、非酸化CAの存在下またはCAの非存在下、結合の手順に供することを含む。攪拌は、付随するタンパク質の変性を避けるため、最小限に保たれた。ポリシアリル化されたFabを、サイズ排除クロマトグラフィー、イオン交換クロマトグラフィー及びSDS−PAGEにより、さらに特徴付けた。
ゼロ(コントロール)及び反応混合物からの48時間の試料(0.5ml)を、セファロースSPカチオン交換カラム上で、イオン交換クロマトグラフィー(IEC)に供した(1ml;フローレート1ml/min;結合/洗浄バッファー 50mMリン酸ナトリウム、pH4.0;溶出バッファー、1M塩化ナトリウム含有50mMリン酸ナトリウムバッファー、pH4.0)。該カラムを洗浄し、溶出し、溶出画分を、CA及びタンパク質(Fab)含有量について分析した。PD−10カラムを、カラムにアプライする前の試料を脱塩するために用いた。
SDS−PAGE(MiniGel, Vertical Gel Unit, model VGT 1, power supply model Consort E132; VWR, UK)を、ポリシアリル化におけるFabの分子サイズの変化を検出するために使用した。反応混合物からのFab及びその複合体(CA、CAO、CAOR及びCAOROとの)の0(コントロール)及び48時間の試料、並びに方法コントロール(非酸化CA)のSDS−PAGEを、4〜20%ポリアクリルアミドゲルを用いて行った。該試料は、幅広い分子量マーカーに対して較正された。
羊IgGFabフラグメント、あるいはCAOまたはCAOROとの複合体試料を、以下のようにI125で放射性標識した。
上記実施例1cで合成したCAOROを、5モル当量のN−[β−マレイミドプロピオン酸]ヒドラジドと、0.1M酢酸ナトリウム中、37℃で2時間反応させた。生成物ヒドラゾンを、エタノール中で沈殿させ、酢酸ナトリウムに再懸濁し、エタノール中で再び沈殿させ、水に再溶解し、凍結乾燥した。該生成物は、タンパク質及びペプチド中のシステイン部分のチオール基への部位特異的な結合に有用である。
参考例2.1−大スケール(large scale)における分画
XK50カラム(Amersham Biosciences, UK)を、900mlのセファロースQ FF(Amersham Biosciences)と共に装填し、3カラム体積の洗浄バッファー(20mMトリエタノールアミン;pH7.4)を用いて、50ml/minのフローレートで平衡させた。CA(200mlの洗浄バッファー中に25グラム)をシリンジポートを介して毎分50mlでカラムにロードした。その後、1.5カラム体積(1350ml)の洗浄バッファーでカラムを洗浄した。
小規模の(at smaller scale)分画
以下の試料を、同一の洗浄及びグラジエントシステムを小規模(75mlまでのマトリックス;0.2〜3グラムのコロミン酸)で用い、分画した:
コロミン酸(CA、22.7kDa、pd 1.34;CA、39KDa、pd=1.4)、コロミン酸−アルデヒド(CAO、22.7kDa、pd 1.34)、単官能性コロミン酸(CAORO、22.7kDa;pd 1.34)、コロミン酸−アミン(CA−NH2、22.7kDa、pd 1.34)、コロミン酸マレイミド(CAM、実施例4による)そして、製造されたCAの分子量を終始モニターした。
分別(Differential)エタノール沈殿を、種々の鎖長のコロミン酸を沈殿させるために用いた。
分別エタノール沈殿は、より小さなCAsはより多くのエタノール(EtOH)を必要とすることを示す。ブロードp.d.22.7kDaポリマーを、70%EtOHを用い、製造物ポリマーの収率>80%で沈殿させた。80%EtOHの濃度は、より低い分子量6.5KDa(pd<1.1)を>80%沈殿させるために必要である。このプロセスは、さらに、生成物を汚染するいかなる塩も除去する。
22.7kDaの試料を、種々のカットオフ分子量の膜(5、10、30、50、及び100kDa)上での限外濾過により、精製した。全ての場合において、未透過物をGPC及びネイティブ(native)PAGEにより分析した。
種々のカットオフ分子量の膜上での限外濾過により、精製された22.7kDaの試料は、ポリマーの多分散性の低下及び、膜のカットオフの増加に伴うより高い分子量へのシフトが存在することを示す(図14)。組み合わされた方法及びイオン対クロマトグラフィーも、ポリマーの分画に用いられ得る。
参考例2.2において調製された、酸化されたコロミン酸(CAO;22.7kDa)及び狭く分散した酸化されたコロミン酸(NCAO;27.7kDa pd=1.09;40.9kDa.pd=1.02)を、GH複合体の調製のために用いた。
成長ホルモンを0.15M PBS(pH7.4)に溶解し、種々のコロミン酸(CAO及びNCAO)に共有結合させた。種々のCAs(22.7kDa、CAO;27.7kDa及び40.9kDa、NCA)を、CA:GHモル比(12.5:1)でGH(2mg)にそれぞれ添加し、ソディウムシアノボロヒドリドを最終濃度4mg/mlとなるように添加した。反応混合物に封をし、35±2℃で24時間、磁気的に攪拌した。その後、混合物を、継続的に攪拌しながら、飽和度70% w/vに達するまで硫酸アンモニウムをゆっくり添加することにより、硫酸アンモニウム((NH4)2SO4)沈殿反応に供し、4℃で1時間攪拌した後、15分間遠心(5000xg)し、ペレットを飽和(NH4)2SO4に再懸濁し、再び15分間遠心した(5000xg)。回収した沈殿を1mlのPBS(pH7.4)に再溶解し、4℃で、同様のバッファーに対して、充分に(24時間)透析した。コントロールは、天然のタンパク質を、非酸化CAの存在下またはCAの非存在下、結合の手順に供することを含む。攪拌は、付随するタンパク質の変性を避けるため、最小限に保たれた。ポリシアリル化されたGHを、SDS−PAGEにより、特徴付けた。ポリシアリル化されたGHは、参考例2に記載されているように、アニオン交換クロマトグラフィーを通過され、生成物画分をSDS PAGEに供した。
結果(図15)は、対照ウェル(GHを有する)において、試料の移動は、新鮮なGHについてのそれと似ていることを示す。複合体のレーンには、ポリシアリル化されたGHを示す質量の増大を典型的に示す、バンドのシフトが存在する。バンドの幅は、広く分散したポリマーとの複合体に比べ、狭く分散したポリマーとの複合体の場合において、有意に狭くなっていた。さらに、GH複合体(広く分散したポリマーとの)を、アニオン交換クロマトグラフィーにより、種々の種に分離した(図16)。
実施例1で調製された、活性化されたポリシアル酸(酸化されたコロミン酸(CAO))及び単官能性ポリシアル酸(酸化−還元−酸化されたコロミン酸(CAORO))を、rh−インシュリン複合体の調製に用いた。
インシュリンを、最小限の体積の15mM HClに溶解し、その後、0.15M PBS(pH7.4)で希釈し、種々のコロミン酸(CA、CAO及び単官能性CAORO)に共有結合させた。コロミン酸(22.7kDa)を、CA:インシュリンモル比25:1で、インシュリン(2mg)と共に、ソディウムシアノボロヒドリド(4mg/ml)を含む0.15M PBS(pH7.4;2ml)中で48時間、封をした容器に入れて磁気的に攪拌しながら、35±2℃のインキュベーター中で反応させた。その後、混合物を、継続的に攪拌しながら、飽和度70% w/vに達するまで硫酸アンモニウムをゆっくり添加することにより、硫酸アンモニウム((NH4)2SO4)沈殿反応に供した。試料を4℃で1時間攪拌した後、15分間遠心(5000xg)し、ペレットを(NH4)2SO4の飽和溶液に懸濁し、再び15分間遠心した(5000xg)。回収した沈殿を、0.9%NaClを追加した1mlの0.15M Naリン酸バッファー(pH7.4;PBS)に再溶解し、4℃で、同じPBSに対して、充分に(24時間)透析した。その後、該透析物を、シアル酸及びタンパク質含有量について分析し、生じた複合体を、CA:インシュリンモル比の観点から示した(実施例1のように)。コントロールは、天然のタンパク質を、記載された条件下で、非酸化CAの存在下またはCAの非存在下、結合の手順に供することを含んだ。攪拌は、付随するタンパク質の変性を避けるため、最小限に保たれた。ポリシアリル化されたインシュリンを、イオン交換クロマトグラフィー及びSDS−PAGEにより、さらに特徴付けた。結果を、得られた複合体における、CA:インシュリンモル比の観点から示す(第7表)。
実施例6のインシュリン及びポリシアリル化されたインシュリン構造物(constructs)を、正常な雌性T/O非近交系マウス(体重22〜24グラム)における、これらの血中グルコースレベル低減能について試験した。動物を5つのグループに分け、インシュリン(0.9%塩化ナトリウム中にマウスあたり0.3単位、または同様のポリシアリル化されたインシュリンのタンパク質等価物を有する)を皮下注射(s.c.)し、血液試料中のグルコースレベルを、グルコースアッセイキット(Accu-Chek Advantage, Roche, UK)を用い、時間間隔をおいて測定した。
ポリシアリル化されたインシュリン構造物の薬裡学的活性は、皮下注射され、時間間隔をおいて採血された(bled)正常なマウスにおけるインタクトなインシュリンのそれに匹敵した。3種類のインシュリンについてのマウスの血中グルコースレベルを、図17に示す。データポイントは、5つの試料の平均を示し、エラーバーは、s.e.m値である。図17における結果は、ポリシアリル化されたインシュリン(元来の方法(CAOを用いた)及び新規な2回−酸化方法(単官能性CAOROを用いた)により調製された)が、血中グルコースレベルのより延長された低減を発揮したことを明白に示す。このように、グルコースレベルが、インタクトなインシュリンを用いた処置の0.75時間後に底値に到達し、2時間後に正常なレベルに戻るのに対し、ポリシアリル化されたペプチドを用いて処置されたマウスにおけるグルコースレベルは、同様に0.75時間において最低であるが、6時間で正常値に戻った。
これらの結果は、CAO−インシュリン及びCAORO−インシュリンが、非誘導体化インシュリンの場合よりも、著しく、有意に長い滞留時間を循環中で生じ、天然のインシュリンと比較し、カーブの上の領域において増大を生じることを証明する。

Claims (25)
- 還元性末端にシアル酸ユニットそしてビシナルジオール基(vicinal diol group)を有する非還元性末端に末端サッカライドを有する出発原料が、以下の連続するステップ:
a)ビシナルジオール基をアルデヒドに酸化するための予備選択的酸化、
b)還元性末端シアル酸ユニットの環を還元的開環し、それによってビシナルジオール基を形成するための還元(そしてここで、ステップa)において形成されたアルデヒドも還元され、ビシナルジオール基の一部ではないヒドロキシ基を形成する);及び
c)ステップb)で形成されたビシナルジオール基を酸化してアルデヒド基を形成するための選択的酸化
に供される、シアル酸のアルデヒド誘導体の製造方法、
ここで、非還元性末端における末端サッカライドがシアル酸であり、出発原料が少なくとも2のシアル酸ユニットを含むポリサッカライドである。 - 還元性末端のシアル酸ユニットが、第8炭素原子を介して隣接するユニットと結合し、それによって、ステップb)において、第6,7ビシナルジオール基が酸化され、第7炭素原子でアルデヒドを形成する、請求項1に記載の方法。
- ポリサッカライドが、実質的にシアル酸ユニットのみからなるポリシアル酸である請求項1または2に記載の方法。
- ポリサッカライドが、少なくとも5のシアル酸ユニットを分子中に有する、請求項1〜3のいずれかに記載の方法。
- ポリサッカライドが、少なくとも10のシアル酸ユニットを分子中に有する、請求項1〜4のいずれかに記載の方法。
- ポリサッカライドが、少なくとも50のシアル酸ユニットを分子中に有する、請求項1〜5のいずれかに記載の方法。
- 予備酸化ステップが、実質的にポリサッカライド鎖の中間鎖(mid-chain)の開裂がないような条件下で行われる、請求項1〜6のいずれかに記載の方法。
- 予備酸化ステップが、水溶液中で、1mM〜1Mの濃度範囲の過ヨウ素酸塩の存在下、3〜10の範囲のpH、0〜60℃の範囲の温度及び1分〜48時間の範囲の時間で行われる、請求項7に記載の方法。
- ステップb)が、出発原料のペンダントカルボキシル基が還元されない条件下で行われる、請求項1〜8のいずれかに記載の方法。
- ステップb)が、水溶液中で、1μM〜0.1Mの濃度範囲のボロヒドリドの存在下、6.5〜10の範囲のpH、0〜60℃の範囲の温度及び1分〜48時間の範囲の時間で行われる、請求項9に記載の方法。
- アルデヒド誘導体が、第1級アミン基またはヒドラジド基を有する基質と反応する、請求項1〜10のいずれかに記載の方法。
- 生成物が還元される、請求項11に記載の方法。
- 基質が、ペプチドまたはタンパク質である、請求項11または12に記載の方法。
- 基質が、ペプチド治療剤(peptide therapeutic)である、請求項13に記載の方法。
- 生成物を、次にチオール基を有する化合物と反応させる、請求項11または12に記載の方法。
- 基質が、ドラッグデリバリーシステム、細胞、ウィルスまたは合成高分子である、請求項11または12に記載の方法。
- 実質的に全てのサッカライドユニットがシアル酸のサッカライドユニットであり、互いに2−8、2−9または2−8/2−9交互で結合しているポリサッカライドである、請求項17に記載の化合物。
- nが5以上の整数である、請求項17または18に記載の化合物。
- nが10以上の整数である、請求項17〜19のいずれかに記載の化合物。
- nが50以上の整数である、請求項17〜20のいずれかに記載の化合物。
