Description
(1)O2 −、H2O2およびHOClからなる群より選ばれた活性酸素の発生を抑制し、または該活性酸素の量を減少させるための、テアニンからなる抗酸化剤、
(2)食品において活性酸素の発生を抑制し、又は発生した活性酸素を減少させるための、テアニンの使用、ならびに
(3)活性酸素が、O2 −、H2O2およびHOClからなる群より選ばれる、前記(2)記載の使用
に関する。
0.3Mグルタミンおよび1.5M塩酸エチルアミンを0.05Mホウ酸緩衝液(Na2B4O7−NaOH、pH11)中、0.3Uグルタミナーゼ存在下にて30℃で22時間反応させ、225nmolのテアニンを得た。次いで、反応液からのテアニンの精製は、反応液をDowex50×8カラムおよびDowex1×2カラム(共に室町化学工業社製)によるクロマトグラフィーにかけ、次いでエタノール処理することにより行った。
茶(Camellia sinensis)葉10kgを熱水で抽出後、得られた抽出液をカチオン交換樹脂(室町化学工業社製;Dowex HCR W−2)にかけ、1N NaOHにより溶出した。溶出画分を活性炭(二村化学工業社製;太閤活性炭 SG)に通し、15%エタノールによる溶出画分をRO膜(日東電工社製;NTR 729 HF)を用いて濃縮した。次いで、濃縮物をカラムクロマトグラフィーにて精製し、更に再結晶を行い、L−テアニン24.8gを得た。
本発明の組成物の抗酸化作用を、ケミルミネッセンスアナライザー(CLA−SP2、東北電子産業社製)を用いて、活性酸素によって生じる発光物質からの化学発光を測定することにより評価した。検出波長範囲を400〜870nmに設定し、解像度を270と870nm間の帯域波長において4nmに設定した。
テアニンを0、1、10および100mg/mlの最終濃度となるように50mMリン酸緩衝溶液(pH7.4)に溶解し、サンプルを調製した。得られたサンプル200μlをケミルミネッセンスアナライザー(CLA−SP2、東北電子産業社製)のサンプルチャンバに添加し、サンプル添加100秒後に、発光物質として0.1mMルシゲニン500μlをサンプルチャンバに添加した。次いで、サンプル添加150秒後に、0.1mMキサンチン500μlおよび0.004Uキサンチンオキシダーゼ50μlをサンプルチャンバに添加した。この一連の操作中の化学発光を測定した結果を図1に示す。
テアニン0、10、50、100、150、200および300mgを50mMリン酸緩衝生理食塩水(pH7.4)200μlと混合してサンプルを調製した。得られたサンプルをケミルミネッセンスアナライザー(CLA−SP2、東北電子産業社製)のサンプルチャンバに添加し、サンプル添加100秒後に、発光物質として25μMルミノール500μlをサンプルチャンバに添加した。次いで、サンプル添加150秒後に、0.12重量%次亜塩素酸100μlをサンプルチャンバに添加した。次亜塩素酸添加150秒後の化学発光を測定し、その結果を図3に示す。
テアニンを0、200mgおよび300mgを50mMリン酸緩衝生理食塩水(pH7.4)200μlと混合してサンプルを調製した。得られたサンプルをケミルミネッセンスアナライザー(CLA−SP2、東北電子産業社製)のサンプルチャンバに添加し、サンプル添加100秒後に、発光物質として25μMルミノール500μlをサンプルチャンバに添加した。次いで、サンプル添加150秒後に、0.3重量%過酸化水素(H2O2)をサンプルチャンバに添加した。過酸化水素添加150秒後の化学発光を測定し、その結果を図4に示す。
テアニン含有抗酸化組成物の1例として、次に示す原料を混合し、造粒後に1錠0.75gとなるように打錠し、テアニン含有錠剤を製造した。
フロストシュガー 71.67重量%(0.5375g)
トレハロース 10.00重量%(0.0750g)
テアニン 13.33重量%(0.1000g)
ショ糖脂肪酸エステル 1.00重量%(0.0075g)
香料(高砂香料工業社製) 4.00重量%(0.0300g)
合計 100重量%(0.7500g)
テアニン(10kg)を水10kgに溶解してテアニン溶液を調製した。次いでグラニュー糖(64kg)を水20kgに溶解しながら110℃まで加熱し、テアニン溶液および水飴を加えて、145℃まで温度を上げた。次に、火を止め、50重量%酒石酸(1kg)および香料(1kg、高砂香料工業社製)を添加して混合した。75〜80℃まで冷却し、成型ローラーで成型し、テアニン含有抗酸化組成物の1例として、テアニン含有キャンディーを製造した。なお、キャンディー中のテアニンの含有量を測定した結果、含有量は1コ1.2gで89.6mg/gであった。
果糖ブドウ糖(12kg)、ブルーベリー濃縮果汁(1kg)、1/5透明レモン果汁(0.4kg)、クエン酸Na(0.05kg)およびテアニン(0.1kg)を水に加え攪拌溶解した。50重量%クエン酸Na(結晶)を用いpH3.1に調整し95℃まで昇温後香料(0.05kg、高砂香料工業社製)を加えて全量100kgになるように水を加えた。一缶100gとなるように缶充填した後に殺菌し、冷却して、テアニン含有抗酸化組成物の1例として、テアニン含有ブルーベリー飲料を製造した。なお、ブルーベリー飲料中のテアニン含有量を測定した結果、含有量は98.3mg/100gであった。
果糖ブドウ糖液(6kg)、テアニン(0.1kg)、ピロリン酸第二鉄(0.06kg)、プラセンタエキス(0.01kg)およびグレープフルーツ果汁100%(30kg)を水に加え攪拌溶解した。クエン酸Naを用いpH3.1に調製し95℃まで昇温後香料(0.05kg、高砂香料工業社製)を加えて、全量100kgになるように水を加えた。一缶100gとなるように缶充填した後に殺菌し、冷却して、テアニン含有抗酸化組成物の1例として、テアニン含有グレープフルーツ飲料を製造した。なお、グレープフルーツ飲料中のテアニン含有量を測定した結果、含有量は96.4mg/100gであった。100mlづつ充填して冷却し、テアニン含有グレープフルーツ飲料を製造した。

- O2 −、H2O2およびHOClからなる群より選ばれた活性酸素の発生を抑制し、または該活性酸素の量を減少させるための、テアニンからなる抗酸化剤。
- 食品において活性酸素の発生を抑制し、又は発生した活性酸素を減少させるための、テアニンの使用。
- 活性酸素が、O2 −、H2O2およびHOClからなる群より選ばれる、請求項2記載の使用。
