本発明は、微粒子、その製造方法及び製造装置、並びに注射剤及びその製造方法に係り、より詳細には、有機化合物の微粒子、その製造方法及び製造装置、並びに注射剤及びその製造方法に関する。

【従来の技術】
有機化合物の微粒子化は、極端な表面積の増大をもたらす。このため、物質固有の性質が出現しやすくなるという利点がある。また、粒子が難溶性・不溶性の物質である場合、その微粒子化により微粒子を溶媒中に擬似的に可溶化した状態(微粒子が溶媒中に懸濁している状態であるが、光散乱が少ないため擬似的に可溶化しているように見える状態)にすることもできる。

このような微粒子化方法として、従来、特開2001−113159号公報に開示されるものがある。同公報には、レーザ光照射により有機化合物の微粒子を生成する方法が開示されており、この方法では、有機化合物として、無機物と有機物の中間の性質を持ち、分子構造が固くて丈夫な有機顔料や芳香族縮合多環化合物が微粒子化の対象とされている。そして、微粒子の生成に際し、有機化合物の吸光帯における波長の光を有機化合物に照射することにより微粒子の生成が図られている。

【発明が解決しようとする課題】
上述した微粒子化の技術を用いれば、物質の新しい調製方法を提供できる可能性があり、幅広い分野での応用が期待される。例えば、創薬においては、合成された新規物質の水などの溶媒に対する溶解度が低い場合、その物質の物理化学的研究やスクリーニングなどの探索ができず、あるいは、ADME試験(吸収・分布・代謝・***試験)など、動物での前臨床試験における一般毒性、一般薬理、薬効薬理、生化学的研究ができないこととなる。これに対して、有機化合物の微粒子化を行うことにより、様々な創薬候補物質の研究ができる可能性がある。

しかしながら、前述した公報に記載の微粒子生成方法は、以下に示す課題を有していた。

すなわち、上記方法では、分子構造の中に比較的弱い化学結合を含む有機化合物の場合、紫外光などの吸光帯波長の光を照射することにより、微粒子を部分的に生成することはできるが、同時に、一部で電子励起状態を経由して有機化合物の光化学反応が生じて不純物が生成されてしまう場合があった。特に、有機化合物が体内に投与される薬物(医薬品)の場合、そのような不純物は副作用の原因となり、生体に悪影響を与えるおそれもあるため、このような事態は極力避けなければならない。すなわち、製薬分野においては、薬物の加工等、製薬プロセスにおける不純物生成の最少化は最優先課題である。

本発明は、以上の問題点を解決するためになされたものであり、有機化合物における光化学反応を充分に防止しながら微粒子を製造することができる微粒子の製造方法及び製造装置、微粒子、並びに注射剤及びその製造方法を提供することを目的とする。

【課題を解決するための手段】
本願発明者は、上記課題を解決するため、薬物などの有機化合物における光化学反応の発生を回避した上で、被処理液における有機化合物の微粒子化を可能にする光照射条件を追求した結果、特定の光照射条件のレーザ光を有機化合物に照射することにより上記課題を解決し得ることを見出し、本発明を完成するに至った。

すなわち、本発明による微粒子の製造方法は、被処理液の溶媒中の有機化合物を微粒子化して、その有機化合物の微粒子を製造する製造方法であって、有機化合物の吸光帯よりも長い波長で、溶媒の吸光帯にあって溶媒が吸収する所定波長のレーザ光を被処理液に照射することによって、有機化合物を微粒子化するとともに、レーザ光の被処理液への照射光強度を、有機化合物において2光子吸収が生じる照射光強度未満とすることを特徴とする。

また、本発明による微粒子の製造装置は、被処理液の溶媒中の有機化合物を微粒子化して、その有機化合物の微粒子を製造する製造装置であって、被処理液を収容するためのチャンバと、チャンバ内に収容される被処理液に、有機化合物の吸光帯とは異なる波長であって溶媒に対して作用する所定波長のレーザ光を照射するレーザ光源とを備えることを特徴とする。

このような製造方法及び装置によれば、被処理液中に含まれる有機化合物の吸光特性にかかわらず、有機化合物の吸光帯とは異なり、溶媒に作用する波長(好ましくは溶媒が吸収する波長)のレーザ光(好ましくは赤外レーザ光)を照射して有機化合物の微粒子化を実現している。これにより、溶媒中の有機化合物における光化学反応の発生を充分に防止しつつ、有機化合物を微粒子化することができる。

上記した製造方法及び装置において、有機化合物がその一部のみ溶媒に溶解するもの、すなわち、溶媒に難溶であるか、もしくは溶媒に不溶なものである場合には、レーザ光照射による有機化合物の微粒子化により、有機化合物を、溶媒に対して擬似的に可溶化させることが可能となる。すなわち、難溶または不溶の有機化合物の微粒子を含む液体を製造することができる。

ここで、有機化合物が「溶媒に難溶」とは、汎用型分光光度計(例えばHITACHI U-3500)を用い、光路長を1cmとして被処理液の吸光度を測定した場合に最大の吸光度が0.01以上となることをいい、最大の吸光度が0.01未満となる場合に有機化合物が被処理液中の溶媒に不溶であるとする。

また、上記した製造方法及び装置において、被処理液に照射するレーザ光の波長は、900nm以上の波長であることが好ましい。あるいは、レーザ光の波長は、溶媒の吸光帯の波長であることが好ましい。これにより、溶媒に対してレーザ光が作用することによる有機化合物の微粒子化を充分に実現しつつ、非処理液中の有機化合物における光化学反応の発生を確実に防止することができる。

また、レーザ光の被処理液への照射光強度を、有機化合物において2光子吸収が生じる照射光強度未満とすることが好ましい。有機化合物で2光子吸収が生じる照射光強度を持つレーザ光を有機化合物に照射した場合、光化学反応を起こさせない波長のレーザ光を用いたにも関わらず、2光子吸収によって有機化合物に光化学反応が生じる場合がある。これに対して、2光子吸収が生じる照射光強度未満の照射光強度を持つレーザ光を有機化合物に照射することで、有機化合物における光化学反応の発生をより確実に防止することができる。

また、被処理液を冷却しつつレーザ光を被処理液に照射することが好ましい。これにより、レーザ光を照射した際の熱分解による有機化合物の劣化等を防止することができる。

また、製造方法は、被処理液へのレーザ光の照射中に、被処理液中の有機化合物の吸光度を測定して有機化合物の微粒子化状態をモニタすることが好ましい。同様に、製造装置は、被処理液中の有機化合物の吸光度を測定して有機化合物の微粒子化状態をモニタするモニタ用吸光帯測定手段を備えることが好ましい。この場合、微粒子化状態がモニタされるため、微粒子化状態に応じてレーザ光照射の停止・継続を決定することができ、有機化合物への必要以上のレーザ光照射を回避することが可能となる。

また、上記製造方法においては、チャンバ内の被処理液を透過したレーザ光の透過光強度を測定しながら、チャンバに照射されるレーザ光の照射光強度を変えることにより、有機化合物で2光子吸収が生じない照射光強度を求めることが好ましい。

被処理液を収容するチャンバに対し、チャンバを透過したレーザ光の透過光強度を測定しながら、チャンバに照射されるレーザ光の照射光強度を変えると、ある照射光強度で有機化合物において2光子吸収が生じるようになる。このとき、チャンバを透過したレーザ光の透過光強度が急激に変化する。このため、2光子吸収が生じない照射光強度を容易に求めることができ、実際には2光子吸収が生じない照射光強度で使用される。

また、被処理液へのレーザ光の照射前または照射中に、被処理液中で製造される微粒子を被処理液中に安定して分散させる安定化剤を被処理液に添加することが好ましい。この場合、安定化剤により、一旦製造された微粒子が被処理液中で安定して分散され、微粒子同士の凝集が充分に防止されるため、微粒子の製造効率を向上させることができる。ここで、安定化剤は界面活性剤であることが好ましい。この場合、微粒子の製造効率を向上させることができることに加えて、有機化合物における光化学反応をより充分に防止しつつ、レーザ光を有機化合物に照射して有機化合物を微粒子化することが可能となる。

また、上記製造装置においては、レーザ光源は、波長可変レーザ光源であることが好ましい。この場合、有機化合物の吸光帯や、溶媒の吸光特性等に基づき、適切な波長のレーザ光を被処理液に照射することが可能となる。

また、製造装置は、チャンバ内の被処理液を透過するレーザ光の透過光強度を測定する透過光強度測定装置と、レーザ光源によりチャンバに照射されるレーザ光の照射光強度を調整する照射光強度調整手段とをさらに備えていることが好ましい。

この場合、有機化合物で2光子吸収が生じる照射光強度になると、レーザ光が有機化合物のみならずチャンバでも大きく吸収されるため、レーザ光の透過光強度がより大きく減少する。このため、有機化合物で2光子吸収が生じない照射光強度を一層容易に求めることができる。

また、被処理液中に含まれる有機化合物は、分子間力が比較的弱い物質、例えば、薬物のようにその融点が250℃以下であることが好ましい。このように融点が低い有機化合物は、レーザ光が溶媒に対して作用することによって微粒子化しやすい。したがって、レーザ光照射による有機化合物の微粒子化を好適に実現することができる。

また、微粒子化の対象となる有機化合物が薬物である場合には、レーザ光照射による薬物における光化学反応が充分に防止される。このため、薬物の薬効を失うことなくその微粒子を製造することができる。また、薬物の微粒子化により薬物の表面積が増大し、生体組織への吸収性が向上するため、即効性のある微粒子を得ることができる。更に、薬物が溶媒に難溶または不溶なものである場合は、その薬物を溶媒中において擬似的に可溶化することができる。また、このように有機化合物が薬物である場合、溶媒としては水を用いることが好ましい。あるいは、水以外の溶媒を用いても良い。

また、本発明による微粒子は、上述した微粒子の製造方法により製造される微粒子である。このような微粒子によれば、有機化合物が一部しか溶解できなかった溶媒や、全く溶解できなかった溶媒に対しても、擬似的に可溶化させることが可能となる。

さらに、本発明による注射剤の製造方法は、上述した微粒子の製造方法により微粒子を含む液体を製造し、この液体に等張化剤を添加するか、あるいは等張化剤存在下において微粒子を製造する方法により微粒子を含む注射剤を製造することを特徴とする。このような製造方法によれば、水に難溶であるか、あるいは不溶な薬物をその光化学反応を充分に防止しながら水に可溶化できる。このため、水に難溶であるか、あるいは不溶な薬物であっても注射剤として製造することができる。また薬物が微粒子化されるため、生体に対して即効性のある注射剤を製造することができる。

また、本発明による注射剤は、上述した注射剤の製造方法により製造される注射剤である。このような注射剤においては、薬物が微粒子化されてその表面積が増大しており、その微粒子は、生体に対して高い吸収性を有する。このため、この注射剤は、生体に注射した場合に即効性を有する。

【発明の実施の形態】
以下、図面とともに本発明による微粒子、その製造方法、及び製造装置、並びに注射剤及びその製造方法の好適な実施形態について詳細に説明する。なお、図面の説明においては同一要素には同一符号を付し、重複する説明を省略する。また、図面の寸法比率は、説明のものと必ずしも一致していない。

図1は、本発明による微粒子の製造装置に関する一実施形態の構成を概略的に示すブロック図である。図1に示すように、本微粒子製造装置1は、被処理液2を収容するためのチャンバ3を備えている。チャンバ3は、例えば石英で構成されている。被処理液2は、溶媒である水4と、水4中に懸濁される有機化合物である難溶性薬物5とから構成されている。また、難溶性薬物5は、水4中に極僅かに溶解される溶解物質と、水4に溶解されない非溶解物質(固形物)とから構成される。難溶性薬物5としては、例えば、ステロイド外用薬である酪酸クロベタゾンや、抗てんかん薬であるカルバマゼピン、鎮痛薬であるイブプロフェン等が挙げられる。

また、微粒子製造装置1は、チャンバ3内の被処理液2に所定波長のレーザ光を照射するレーザ光源11を備えている。レーザ光源11は、微粒子化対象の有機化合物である薬物5の吸光帯とは異なる波長であって、溶媒である水4に対して作用する波長(好ましくは水4が吸収する波長)のレーザ光を出射することが可能な光源である。このレーザ光源11としては、レーザ光に設定すべき波長があらかじめ分かっている場合には、波長固定レーザ光源を用いることができる。あるいは、レーザ光源11として、レーザ光の波長を変化させることが可能な波長可変レーザ光源を用いても良い。この場合、有機化合物の吸光帯や、溶媒に対して作用する光の波長などに基づき、適切な波長のレーザ光を適宜に設定して照射することができる。

また、レーザ光源11に対し、必要に応じて、レーザ光源11から出射されるレーザ光の照射光強度を調整する照射光強度調整手段が設けられる。照射光強度調整手段としては、例えば高い光耐圧のある減衰フィルタや光干渉・反射を利用した光減衰器などが挙げられる。図1においては、レーザ光源11とチャンバ3との間に、減衰フィルタなどの照射光強度調整器11aを配置した例を示している。またチャンバ3を挟んでレーザ光源11の反対側の所定位置には、レーザ光源11から出射されチャンバ3を透過するレーザ光の透過光強度を測定する透過光強度測定装置12が配置されている。

さらに、微粒子製造装置1は、チャンバ3内の吸光帯を測定できるモニタ用吸光帯測定装置14を備えている。モニタ用吸光帯測定装置14は、チャンバ3を収容するボックスと、ボックス内に設けられる分光用光源及び光検出器とを備えており、チャンバ3内の被処理液2での吸光度を測定して難溶性薬物の微粒子化状態をモニタすることができるようになっている。

このように、モニタ用吸光帯測定装置14で被処理液2の吸光帯変化をモニタすることにより、薬物5の微粒子化状態がモニタされる。このとき、微粒子化状態に応じてレーザ光照射の停止・継続を決定するなど、被処理液2への良好なレーザ光照射時間や照射条件を決定する際に参照することができ、難溶性薬物5への必要以上のレーザ光照射を回避できるという役割を果たす。また、この測定装置14のボックスには、レーザ光源11から出射されたレーザ光がチャンバ3を経て透過光強度測定装置12に到達するようにレーザ光通過口または通過窓が形成されている。なお、図1においては、測定装置14の具体的な構成について図示を省略している。

レーザ光源11、モニタ用吸光帯測定装置14、照射光強度調整器11a、及び透過光強度測定装置12には、コンピュータなどからなる制御装置13が電気的に接続されている。制御装置13は、上記した製造装置1の各部の動作を制御する。

次に、図1に示した微粒子製造装置1を用いた本発明による微粒子の製造方法について、図2のフローチャートを用いて説明する。

まず水4と難溶性薬物5とを混合した後、撹拌して被処理液2を調整する。被処理液2においては、撹拌により、難溶性薬物5の一部が水4に溶解されて溶解物質となり、残りは、水4に溶解されずに非溶解物質となる。続いて、微粒子製造用チャンバ3内に被処理液2を導入する(ステップS201)。そして、有機化合物である薬物5の吸光帯とは異なる波長であって、溶媒である水4に対して作用する波長により、レーザ光源11から被処理液2へと照射するレーザ光の波長λ1を設定する(S202)。このレーザ光の波長としては、好ましくは薬物5の吸光帯よりも長い波長、さらに好ましくは赤外域の波長、が選択される。

溶解物質である薬物5の吸光帯の最長波長λ0がわかっている場合には、その波長λ0を参照して、微粒子製造に用いるレーザ光の波長λ1が決定することが好ましい。例えば、波長λ1は、最長波長λ0よりも長い波長であって、溶媒である水4に対して作用する波長が選択される。

そして、制御装置13によってレーザ光源11が制御され、レーザ光源11において、照射するレーザ光波長が上記のようにして決定したレーザ光波長λ1に設定される。また、波長λ1があらかじめ設定されている場合には、その波長λ1のレーザ光を出射する波長固定レーザ光源をレーザ光源11としても良い。

次に、レーザ光照射波長λ1はそのままにして、微粒子製造時のレーザ光の照射光強度を決定する(S203)。まずレーザ光源11により、微粒子製造用チャンバ3にレーザ光を照射し、微粒子製造用チャンバ3を透過するレーザ光の透過光強度を透過光強度測定装置12で測定する。そして、微粒子製造用チャンバ3を透過したレーザ光の透過光強度を透過光強度測定装置12で測定しながら、照射光強度調整器11aによりチャンバ3に照射されるレーザ光の照射光強度を変える。こうしてレーザ光の照射光強度とレーザ光の透過光強度との関係が得られる。

ここで、難溶性薬物5に2光子吸収が生じる場合には、レーザ光の透過光強度の急激な変化が観測される。よって、上記のように照射光強度と透過光強度との関係を測定することにより、難溶性薬物5で2光子吸収が生じない照射光強度を容易に決定することができる。そして、制御装置13により照射光強度調整器11aが制御され、レーザ光の照射光強度が、上記のようにして決定した2光子吸収が生じる照射光強度より小さい照射光強度となるように調整される。

有機化合物に2光子吸収が生じる照射光強度を持つレーザ光を薬物5などの有機化合物に照射した場合、光化学反応を起こさせない波長のレーザ光を用いたにも関わらず、2光子吸収によって有機化合物に光化学反応が生じる場合がある。これに対して、2光子吸収が生じる照射光強度未満の照射光強度を持つレーザ光を有機化合物に照射することで、有機化合物における光化学反応をより確実に防止することができる。

この状態で、制御装置13によりレーザ光源11を作動させ、レーザ光源11から出射された波長λ1のレーザ光を微粒子製造用チャンバ3に照射させる。これにより、チャンバ3内の被処理液2において、難溶性薬物5が微粒子化され、難溶性薬物5の微粒子が製造される(S204)。

ここで、難溶性薬物5が医薬品の場合は、微粒子の製造時に、必要以上のレーザ光照射を避けるよう処理をすることが求められる。そのため、被処理液2について、レーザ光照射時間に対する被処理液2の吸光度変化をモニタ用吸光帯測定装置14で測定することによって微粒子化状態をモニタし、目的の処理が達成されたか判断する。そして、目的の処理が達成された場合にはレーザ光の照射を止め、目的の処理が達成されていない場合にはレーザ光の照射を継続する(S205、S206)。

具体的には、目的の処理が達成されたかどうかは、レーザ光源11により被処理液2に対してレーザ光照射を行い、モニタ用吸光帯測定装置14で測定された吸光帯変化を測定することにより判断し、吸光帯の時間変化がほとんど見られなくなった場合に目的の処理が達成できたものとすればよく、処理時間は、レーザ光照射を開始してから、レーザ光照射時間に対して吸光帯がほとんど変化しなくなるまでの時間とすればよい。

本実施形態による微粒子の製造方法及び製造装置の効果について説明する。

上記した微粒子の製造方法及び製造装置によれば、有機化合物の吸光帯とは異なり、水4などの溶媒に作用する波長(好ましくは溶媒が吸収する波長)のレーザ光を照射して、有機化合物の微粒子化を実現している。これにより、溶媒中の有機化合物における光化学反応の発生を充分に防止しつつ、有機化合物を微粒子化することができる。特に、有機化合物がその一部のみ溶媒に溶解するもの、すなわち、溶媒に難溶であるか、もしくは溶媒に不溶なものである場合には、レーザ光照射を用いて有機化合物を微粒子化することにより、有機化合物を、溶媒に対して擬似的に可溶化させることが可能となる。したがって、難溶または不溶の有機化合物の微粒子を含む液体を製造することができる。

すなわち、上記した実施形態では、難溶性薬物5をレーザ光照射によって微粒子化することで、難溶性薬物5が擬似的に水4中に可溶化される。また難溶性薬物5が微粒子化されても、難溶性薬物5の水4中における可溶化状態を長期間にわたって安定に保持することができる。

さらに、レーザ光として、難溶性薬物5の吸光帯とは異なる波長のレーザ光を用い、レーザ光を薬物5に直接に作用させるのではなく、そのレーザ光を溶媒である水4に作用させることによって薬物5を微粒子化している。したがって、水4中の薬物5における光化学反応の発生を充分に防止して、薬物5の持つ薬効を失うことなく微粒子化を達成することができる。

また、被処理液2中に含まれている薬物5などの有機化合物の吸光特性に関係なく、水4などの溶媒に作用(例えば吸収)がある波長のレーザ光だけで微粒子化処理が実現できる。この場合、有機化合物の吸光帯の波長に合わせてレーザ光波長を設定する方法等に比べて、微粒子製造装置1に使用される光源の波長が限定できる。したがって、微粒子製造に適した特定波長のレーザ光源を開発でき、大量処理や処理コストの面で有用である。このような光源としては、例えば半導体レーザ光源が考えられる。例えば、溶媒が水であれば、有機化合物にかかわらず、水の吸収帯の波長、またはそれに基づいて設定された波長のレーザ光を出射するレーザ光源を用いることができる。

具体的なレーザ光波長については、900nm以上の波長とすることにより、有機化合物における光化学反応による不純物の生成が充分に抑制される条件で、有機化合物の微粒子化処理を実現することができる。また、レーザ光波長を溶媒の吸光帯の波長とすることにより、レーザ光を溶媒に対して充分に吸収させて、高効率で微粒子化を達成することができる。

また、上記した製造方法及び装置によって製造される本発明による微粒子によれば、有機化合物が一部しか溶解できなかった溶媒や、全く溶解できなかった溶媒に対しても、擬似的に可溶化させることが可能となる。

薬物などの有機化合物の溶媒としては、上記したように水を用いることが好ましい。あるいは、水以外の溶媒を用いても良い。そのような溶媒としては、1価アルコールであるエチルアルコール、2価アルコールであるグリコール類(プロピレングリコール、ポリエチレングリコール等)、3価アルコールであるグリセロールなどがある。また、植物油であるダイズ油、トウモロコシ油、ゴマ油、ラッカセイ油なども溶媒として用いることができる。これらの溶媒は、注射剤として使用する場合に、非水性注射剤の有機溶媒として好適に用いることができる。

図3は、溶媒の代表的な吸収ピーク波長(nm)及び吸光度(全て、光路長1cm換算の値を吸光度としている)を示す表である。この表では、医薬品として添加が認可されている溶媒である水、ダイズ油、トウモロコシ油、エチルアルコール、ポリエチレングリコール400、及びグリセロールについて吸収ピーク波長及び吸光度を示している。また、図4、図5、図6は、エチルアルコール、ポリエチレングリコール400、及びグリセロールの吸光度の波長依存性を示すグラフである。

これらの吸収ピーク波長は、いずれも900nm以上であり、このようなピーク波長、あるいはその近傍の波長にレーザ光の波長λ1を設定することにより、レーザ光を溶媒に充分に吸収させて、溶媒中にある有機化合物を高効率で微粒子化することができる。例えば、溶媒として水を用いている場合、レーザ光の波長λ1を1450nm、1940nmなどに設定することが好ましい。

上記のようにして得られる難溶性薬物5の微粒子は、水4に擬似的に可溶化されているだけでなく、難溶性薬物5の持つ薬効を失うことなく充分に保持している。このため、難溶性薬物5の微粒子化前の形態では評価できなかった物理化学的研究、スクリーニングなどの候補化合物の探索、決定や、ADME試験(吸収・分布・代謝・***試験)、動物での前臨床試験における一般毒性、一般薬理、薬効薬理、生化学的研究、及び臨床試験などができるようになる。

したがって、入手した化合物ライブラリーや新規に合成された薬物、あるいは天然物が水に対して難溶であったとしても、投資を無駄にすることがない。また難溶性薬物5の微粒子は、微粒子化前の状態に比べて充分に大きな表面積を有している。したがって、生体組織への吸収性が向上し、生体に対する即効性を有するようになる。また上記微粒子製造方法により、極めて多種類の生体に投与可能な薬物を得ることができるため、薬物の投与選択性を飛躍的に拡大することができる。また、このような微粒子化処理は、薬物以外の有機化合物に対しても有効である。

なお、上記した製造方法においては、レーザ光の照射前または照射中に、被処理液2において薬物の微粒子を安定して分散させる安定化剤を添加することが好ましい。このように被処理液2に安定化剤を添加すると、安定化剤により難溶性薬物5が水4中に安定して分散されるため、微粒子の製造効率を向上させることができる。上記安定化剤は界面活性剤であることが好ましい。この場合、微粒子の製造効率を向上させることができる。

なお、上述した製造方法においては、微粒子の製造時に被処理液2の吸光度変化をモニタ用吸光帯測定装置14で測定し、目的の処理が達成された場合にレーザ光の照射を停止するようにしたが、微粒子の製造前に、あらかじめ被処理液2と同一の被処理液についてレーザ光照射による処理時間を決定してもよい。処理時間の決定は、上記したように、モニタ用吸光帯測定装置により有機化合物の吸光帯を測定し、レーザ光照射を開始してから、吸光帯の時間変化がほとんど見られなくなるまでの時間とすればよい。ただし、微粒子の製造前にあらかじめ処理時間を決定している場合は、微粒子の製造時において、その処理時間が経過した時点でレーザ光の照射を止めればよい。したがって、このような場合には、モニタ用吸光帯測定装置14を設置せず、微粒子の製造時に測定装置14で被処理液2中の薬物の微粒子化状態をモニタしなくてもよい。

次に、本発明に係る注射剤の製造方法の実施形態について説明する。

まず、図1に示した微粒子製造装置1を用いて、注射用水4に擬似的に可溶化された難溶性薬物5の微粒子を含む液体を製造する。この液体の製造方法は、上述した微粒子の製造方法と同様である。なお、難溶性薬物5のレーザ光照射前または照射中に、被処理液2に安定化剤を添加しても良いのは、上述した微粒子製造方法と同様である。

続いて、この液体に等張化剤を添加して注射剤を製造する。ここで、液体に添加される等張化剤は、生体の血液と注射液の浸透圧を等しくするように調整する機能を有しており、このような等張化剤としては、例えばショ糖、生理食塩水などが挙げられる。なお、等張化剤の存在下で難溶性薬物の微粒子を製造しても良い。

このような製造方法によれば、難溶性薬物5をその光化学反応を充分に防止しながら注射用水4に可溶化できる。このため、難溶性薬物5であっても、注射剤として製造することができる。また難溶性薬物5が微粒子化されるため、生体に対して即効性のある注射剤を製造することができる。

さらに、上記実施形態では、照射波長決定用吸光帯測定装置10で難溶性薬物5の吸光帯を測定するために被処理液2中の溶媒として水が用いられているが、これには限定されない。このような溶媒としては、上述したように、エチルアルコール等の水溶性の有機溶剤、あるいは植物油を用いることも可能である。

また、ある薬物が水に全く溶解しない、即ち水中でその薬物の吸光帯を測定することができない不溶性薬物である場合には、その薬物の一部を溶解させて吸光帯を測定できるようにするために、水に代えて、例えばエチルアルコール、アセトン、ジメチルスルホキシド等の有機溶媒、又はそれら有機溶媒と水との混合液を用いて、別途、分光光度計によりその吸光帯を測定し、適切な微粒子製造用レーザ光照射波長を決定することができる。

ただし、有機溶媒を用いると、水を用いる場合に比べて吸光帯の最長波長がシフトする傾向がある。このため、薬物の吸光帯を測定する場合には、溶媒として有機溶媒と水との混合液を用いることが好ましい。また薬物にレーザ光を照射してその微粒子を製造する場合は、生体への悪影響を防止する観点から、溶媒として水などの所定の溶媒を用いる必要がある。

また、上記実施形態では、薬物として酪酸クロベタゾンやカルバマゼピン等の難溶性、あるいは不溶性薬物が挙げられているが、これら難溶性、あるいは不溶性薬物に限定されない。さらに、上記実施形態では、薬物として、医薬品物質である酪酸クロベタゾンやカルバマゼピンが用いられているが、本発明の微粒子製造方法及び注射液の製造方法は、上記医薬品物質のみならず医薬品候補物質(天然物、化合物ライブラリー等)、あるいは医薬部外品、化粧品等にも適用可能である。

次に、実施例により、本発明の内容をより具体的に説明するが、本発明は、この実施例に限定されるものではない。

本実施例においては、難溶性薬物として、ステロイド外用薬である酪酸クロベタゾン(Clobetasone Butyrate)の微粒子化を試みた。酪酸クロベタゾン粉末を濃度0.5mg/mlで超音波を用いて水中に10分間懸濁した後、得られた懸濁液(酪酸クロベタゾン懸濁液)3mlを石英製で1cm×1cm×4cmの角セルに入れた。また、角セル中の懸濁液に均等なレーザ光照射が可能なように、液を攪拌するための攪拌マグネットスティックを入れた。懸濁液の温度は、温度依存性に関する実験以外では、すべて室温25℃とした。

また、本実施例では、レーザ光照射による微粒子化及び光化学反応の波長依存性を調査する必要性から、連続的に波長可変なOPOパラメトリック発振器を微粒子化のための光源として用いた。照射レーザ光のパルス幅はFWHM4ns、繰返し周波数は10Hzとした。

図7は、上述した方法を用いた微粒子化処理の前後での酪酸クロベタゾン懸濁液の吸光度の波長依存性を示すグラフである。このグラフにおいて、横軸は光の波長(nm)を、縦軸は懸濁液の吸光度を示している。また、グラフAはレーザ光照射前の吸光特性を示し、グラフBはレーザ光照射後(微粒子化処理後)の吸光特性を示している。

グラフAに示すように、微粒子化処理前の酪酸クロベタゾン懸濁液では、その吸光特性は、ほとんど光散乱損失によるものとなり、波長依存性が小さい平坦な吸光特性となっている。この懸濁液に対し、酪酸クロベタゾンを微粒子化するため、波長1064nm、パルス当たりの照射光強度1700mJ/cm2のYAGパルスレーザ光を1時間照射した。この照射処理後では、グラフBに示すように、酪酸クロベタゾン自体の吸光特性が出現するようになった。この現象は、懸濁液の微粒子化が、サブマイクロメーターオーダーまで進行したことを示している。

次に、レーザ光の照射波長を変えて実験を行うとともに、高速液体クロマトグラフィー(HPLC)を用いて、レーザ光照射による微粒子化処理後の酪酸クロベタゾンの純度(SIGMA製、最低純度98%を使用)を測定し、そのレーザ光波長依存性を調べた。各波長でのレーザ光照射強度は、図7に示したようなサブマイクロメータオーダーの微粒子化が観測できるレベルに選定し、1時間のレーザ光照射処理を行った。

図8は、微粒子化処理後での酪酸クロベタゾン純度のレーザ光波長依存性を示すグラフである。このグラフにおいて、横軸は懸濁液に照射するレーザ光の波長(nm)を、縦軸は酪酸クロベタゾン純度(%)を示している。

このグラフに示すように、照射するレーザ光の波長が500〜800nmの範囲では、非常に高出力の光強度でなければ酪酸クロベタゾンの微粒子化を行うことができない。その結果、酪酸クロベタゾンの一部が光化学反応を引き起こし、大幅な純度の劣化が観測された。また、波長500nm以下では、微粒子化処理に必要な光強度が比較的小さいため劣化も小さいが、1光子あたりのエネルギーが大きく、また酪酸クロベタゾンがレーザ光を直接吸収するため、同様に光化学反応が起きる。その結果、劣化が起こり薬物処理に許容される不純物の生成率とはなっていない。

これに対して、図8に示すように、波長900nm以上の赤外レーザ光を用いて微粒子化処理を行うことにより、薬物などの有機化合物における光化学反応の発生がほとんどない条件で微粒子化処理を実現することが可能である。なお、光強度が高すぎると熱分解による純度の劣化も考えられるので、そのような場合には、被処理液を冷却することによってその温度を低くすることが好ましい。

図9は、赤外波長領域における酪酸クロベタゾンの吸光特性を示すグラフである。このグラフにおいて、横軸は光の波長(nm)を、縦軸は酪酸クロベタゾンの吸光度を示している。また、グラフCは流動パラフィンのみでの吸光特性を示し、グラフDは酪酸クロベタゾン及び流動パラフィンの混合液での吸光特性を示している。

ここでは、ヌジョール法を用いて赤外波長領域における酪酸クロベタゾンの吸光特性の測定を行った。ヌジョール法は、粒子状のサンプルに流動パラフィンを添加し、乳鉢ですりつぶしてオイル状に加工し、その混合液と、流動パラフィンとの吸光特性の差からサンプル自体の吸光特性を評価する方法である。吸光度の測定においては、光路長100μmの石英セルを用いた。

グラフDに示すように、酪酸クロベタゾン及び流動パラフィンの混合液の吸光特性には、光散乱と若干の吸光帯とが現れている。この吸光特性をグラフCに示す流動パラフィンのみでの吸光特性と比較すると、矢印P、Qで示された1700nm帯及び2300nm帯の吸光帯で両者は合致している。このことから、900〜2500nmの波長域においては、酪酸クロベタゾン自体には大きな吸光帯はなく、したがって、この波長域のレーザ光を微粒子化処理のために照射したとしても、酪酸クロベタゾンにおける光化学反応の発生は充分に小さいと考えられる。

次に、レーザ光波長420〜2150nmの波長範囲において微粒子化効率を求めた。図10は、微粒子化効率のレーザ光波長依存性を示すグラフである。このグラフにおいて、横軸はレーザ光の波長(nm)を、縦軸は波長570nmにおける微粒子化効率を1として規格化した微粒子化効率を示している。

ここで、微粒子化効率の算出手法としては、まず、微粒子化の度合を示す酪酸クロベタゾンの吸光度を求め、照射光強度で割り算し、さらに波長570nmにおける微粒子化効率で規格化して各波長の微粒子化効率を比較した。また、この図10には、水の吸光度の波長依存性のグラフを微粒子化効率のデータと対応させて示している。

このグラフに示すように、照射するレーザ光の波長が570nmのときに微粒子化効率が最も悪く、波長420〜600nmでも大きな差はない。一方、波長900nm以上では効率良く微粒子化が行われている。また、水は960nm、1450nm、1940nmに吸光帯を持っているが、水に吸収がある波長において微粒子化効率が特に高くなることが判明した。このことは、レーザ光照射による微粒子化処理が、近赤外の波長域において酪酸クロベタゾンなどの有機化合物には光の吸収がなくても、水などの溶媒に作用する(吸収がある)波長を選択すれば、微粒子化処理が可能であることを示している。

こうして溶解物質である薬物5の吸光帯の最長波長λ0が決定された後、その波長λ0を参照して、微粒子製造に用いるレーザ光の波長λ1が決定される。例えば、波長λ1は、最長波長λ0よりも長い波長であって、溶媒である水4に対して作用する波長に決定される(S302c)。そして、制御装置13によってレーザ光源11が制御され、レーザ光源11において、照射するレーザ光波長が上記のようにして決定したレーザ光波長λ1に設定される。

本発明による微粒子、その製造方法、及び製造装置、並びに注射剤及びその製造方法は、以上詳細に説明したように、次のような効果を得る。すなわち、被処理液の溶媒中の有機化合物を微粒子化して、その有機化合物の微粒子を製造する際に、有機化合物の吸光帯とは異なる波長であって溶媒に対して作用する所定波長のレーザ光を被処理液に照射する方法及び装置等によれば、被処理液中に含まれる有機化合物の吸光特性にかかわらず、溶媒に作用する所定波長の光を照射することによって有機化合物の微粒子化が実現される。これにより、溶媒中の有機化合物における光化学反応の発生を充分に防止しつつ、有機化合物を微粒子化することができる。

また、本発明の微粒子によれば、有機化合物が一部しか溶解できなかった溶媒や全く溶解できなかった溶媒に対しても、有機化合物を微粒子化して擬似的に可溶化させることが可能となる。また、本発明の注射剤によれば、生体に注射した場合に即効性を有するようになる。さらに、本発明による注射剤の製造方法によれば、水に不溶であるか、水に一部しか溶解しない薬物であっても注射剤として製造することができる。また、生体に対して即効性を有する注射剤を製造することができる。

This publication discloses a method for producing fine particles of an organic compound by laser light irradiation. In this method, the organic compound has a property intermediate between an inorganic substance and an organic substance, and has a solid molecular structure and a strong organic pigment. And aromatic condensed polycyclic compounds are targeted for micronization. In producing fine particles, fine particles are produced by irradiating the organic compound with light having a wavelength in the absorption band of the organic compound.

If the above-described micronization technique is used, a new method for preparing a substance may be provided, and application in a wide range of fields is expected.
