Description
The present invention relates to an application as a light-controlled rotaxane thin film using a novel compound, azoaryl mercaptoalkyl polyethylene glycol.
Polyethylene glycol thin films are widely used as biomaterials that suppress nonspecific adsorption. Among them, mercaptopolyethylene glycol having a mercapto group introduced is a useful thin film material capable of producing a self-assembled film on a substrate coated with a metal thin film or a gold colloid. Furthermore, poly or oligoethylene glycol forms supramolecular polyrotaxane by cyclodextrin (CD) and host-guest interaction, and the polyrotaxane is used in a drug delivery system (DDS).
The present invention provides a poly or oligoethylene glycol capable of forming a monomolecular film with a mercapto group and further controlling the structure of a supramolecular rotaxane with light. A light-controlled supramolecular rotaxane thin film that contributes to the development of light-controlled biomaterials can be provided.
In the present invention, an azoaryl mercaptoalkylpolyethylene glycol, which is a novel compound not described in the literature, forms a self-assembled film, and further, supramolecules selectively interact with α-CD of a polysaccharide to host-guest interaction. It was found that the film has a pseudorotaxane structure. It was found that the azoaryl mercaptoalkylpolyethylene glycol having the supramolecular pseudorotaxane structure was irradiated with light to take a supramolecular rotaxane structure in which α-CD was not dissociated. That is, the present invention provides the following azoaryl mercaptoalkyl polyethylene glycol.
In the compound represented by the general formula (1), m = 2 to 100, preferably m = 6 to 50, most preferably m = 6 to 25, n = 2 to 50, preferably n = 5 to 30, Most preferably, n = 5-20.
The azoaryl mercaptoalkyl polyethylene glycol of the present invention is a novel compound not described in any literature, and its self-assembled film interacts with α-CD and host-guest, and controls the pseudorotaxane-rotaxane structure by light irradiation. Is.
m=6であれば、この化合物は、そのエチレングリコール鎖にα−CDを最大で3個取り込
む。
The azoaryl mercaptoalkyl polyethylene glycol of the present invention can interact with and incorporate one α-CD per 2 units of ethylene glycol. That is, — (C 2 H 4 O) 2 — can interact with one α-CD. Thus, for example, if m = 6 in formula (1) above, this compound incorporates up to three α-CDs into its ethylene glycol chain.
The compound of the present invention forms a self-assembled film on a metal thin film by its mercapto group. In this self-assembled film, the ethylene glycol portion is in a random coil state. Furthermore, this self-assembled film can incorporate α-CD into the ethylene glycol portion, and becomes a thin film having a pseudorotaxane structure. By incorporating α-CD, the ethylene glycol chain in this pseudorotaxane thin film appears to extend. Next, when this pseudorotaxane thin film is continuously irradiated with light, the azoaryl moiety undergoes trans → cis photoisomerization to form a thin film having a rotaxane structure.

This reaction can be performed using a basic compound such as sodium hydroxide, potassium hydroxide or potassium carbonate as a catalyst. The compound of the present invention thus obtained can be easily isolated and purified from the reaction mixture according to conventional separation and purification means.
B) Manufacturing method of light-controlled rotaxane thin film
1) Preparation of self-assembled film An azoaryl mercaptoalkyl polyethylene glycol of the general formula (1) is dissolved in a solvent. Examples of the solvent include common organic solvents such as ethanol, acetone, chloroform, toluene, and water, and ethanol is preferable. The concentration is preferably 1 mM to 10 mM, particularly 1 mM. When the concentration is low, the formation time of the self-assembled film becomes long.
Moreover, you may mix a mercaptoalkyl alcohol with the azoaryl mercaptoalkyl polyethyleneglycol of the compound of General formula (1). As the mixing ratio, the number of moles of the compound of the general formula (1): the number of moles of the mercaptoalkyl alcohol is 1:99 to 100: 0, preferably 1: 4. This mixture is dissolved in a solvent. The solvent is the same as described above. Even when mercaptoalkyl alcohol is mixed with the compound of general formula (1), the concentration is preferably 1 mM to 10 mM, particularly 1 mM, as in the case of not mixing.
The purpose of mixing the mercaptoalkyl alcohol with the compound of the general formula (1) is to reduce the density of the compound of the general formula (1) in the substrate on which the self-assembled film is formed. That is, when the density of the compound of the general formula (1) on the substrate is high, the compound of the general formula (1) hardly takes in α-CD (outer diameter = about 13 cm) which is a large molecule. Therefore, by mixing a mercaptoalkyl alcohol that forms a monomolecular film with a mercapto group in the same manner as the compound of general formula (1), the distance between the compounds of general formula (1) on the substrate can be increased.
A homogeneous self-assembled film can be obtained by making the number of alkyls of mercaptoalkyl alcohol and the number of alkyls of azoaryl mercaptoalkyl polyethylene glycol of the compound of general formula (1) substantially the same. Therefore, for example, it is preferable to mix a commercial mercaptoundecanol with azoaryl mercaptohexaethylene glycol, which is a compound of the general formula (1).
When the substrate coated with the metal thin film is immersed in the solution obtained as described above by a known method, a self-assembled film can be obtained.
As the metal thin film, metal thin films such as gold, silver, copper, platinum and the like are generally preferable, and gold or silver thin films are more preferable.
The reaction temperature may be a temperature at which the solvent does not evaporate and is, for example, 0 ° C. to 50 ° C., preferably room temperature. The reaction time varies depending on the solvent and concentration used, but is preferably 5 minutes to 2 hours.
The film thickness of the resulting self-assembled film was observed in situ using surface plasmon spectroscopy (SPR), the SPR spectrum was fitted to the Fresnel equation, and the amount of shift of the dip Can be obtained from The film thickness of the self-assembled film means the length from the mercapto group to the azoaryl group of the compound of the present invention in a state where the self-assembled film is formed. The film thickness of the obtained self-assembled film varies depending on the values of m and n in the general formula (1), and is, for example, 2.0 nm to 40 nm.
2) Preparation of pseudorotaxane thin film (incorporation of α-CD)
Next, when the obtained self-assembled film is immersed in an α-CD aqueous solution, a pseudorotaxane thin film is obtained.
The concentration of the α-CD aqueous solution is preferably large excess, for example, 7.25 mg / ml to 145 mg / ml (saturated concentration).
As reaction temperature, 15-25 degreeC is preferable. Moreover, although reaction time changes with the number of m in the compound of General formula (1), (alpha) -CD aqueous solution density | concentration, etc., it is about 5 minutes-10 days.
The film thickness of the obtained pseudorotaxane thin film varies depending on the values of m and n in the general formula (1), and is, for example, 2.5 nm to 47 nm.
3) Light control When the pseudorotaxane film is continuously irradiated with light of 300 to 400 nm, preferably 350 nm, the azoaryl moiety in the compound of the general formula (1) undergoes trans → cis photoisomerization to obtain a rotaxane structure. (Cis isomer). When irradiation is stopped, it returns to the pseudorotaxane structure (trans isomer) (FIG. 5).
The azoaryl mercaptoalkylpolyethylene glycol of the present invention has the ability to produce a self-assembled film with a mercaptoalkyl group, exhibits molecular recognition ability with oligoethylene glycol, and further forms a rotaxane structure by photoisomerization of the azoaryl moiety. Can be effectively used as a light-controlled rotaxane thin film.
Hereinafter, the present invention will be described with reference to examples.

An SPR spectrum (FIG. 2) was fitted to the Fresnel equation, and it was confirmed that the film thickness was 3.7 nm from the shift amount of the dip.
When α-CD aqueous solution (saturated concentration: 145 mg / ml) was added to this film, the dip shifted more than the refractive index change to the high angle side (Fig. 3). As a result of fitting, it was found that this corresponds to an increase in film thickness of 2.1 nm. This is presumably because pseudorotaxane is formed by the host-guest interaction, the ethylene glycol chain is extended, and further, CD is adsorbed on the membrane surface portion. On the other hand, even when a β-CD solution was added to the same film sample, only a shift corresponding to a change in refractive index was observed, so that it was found that this film selectively incorporated α-CD by molecular recognition.
When the α-CD pseudorotaxane film was continuously irradiated with 350 nm light, a decrease in film thickness was observed (FIG. 4). This is thought to be due to structural changes caused by trans-cis photoisomerization of azobenzene. And when this rotaxane thin film was washed with water, the dip was shifted to the high angle side before the α-CD injection. This shift to the high angle side could be recovered to the position of the dip before CD injection by washing again with water several hours after the light irradiation was stopped. Therefore, during irradiation, the cis isomer of azobenzene acts as a stopper to take a polyrotaxane structure and suppress inclusion CD from escaping from the chain, but by stopping irradiation, azobenzene is reverse isomerized from cis to trans, It is thought to be a pseudorotaxane again. From the above results, it was shown that a mercapto-oligoethylene glycol monomolecular film modified with an azobenzene derivative is useful as a self-assembled film that selectively incorporates α-CD and forms a photo-controllable rotaxane.
