【発明の属する技術分野】
この発明は、照明方法の切替機構を備えた顕微鏡に関し、特に、落射照明と全反射照明の切り替え機構を備えた顕微鏡に関する。

【従来の技術】
従来より顕微鏡、特に蛍光顕微鏡の照明方法としては、一般的な落射照明と、エバネッセント光を用いる全反射照明とが知られている。

落射照明方法は、図1に示したように、光源(水銀ランプまたはレーザー)1からの照明光3を投影管(レンズ群)2を経て、ダイクロイックミラー8により反射させ、対物レンズ4の中央に入射させて試料溶液7中の試料に照射する方法である。試料からの蛍光9は、対物レンズ4を通り、ダイクロイックミラー8を透過し、不図示の結像光学系に入射する。

一方、全反射照明は、図2のように試料溶液7に接したカバーガラスやスライドガラス等のガラス板6で照明光3を全反射させ、ガラス表面から試料溶液側に深さ100nm程度しみ出すエバネッセント光10を使って、ガラス板6近傍の試料を局所的に照明する方法である。ガラス板6に全反射の条件で照明光3を入射させるには、図2のように対物レンズ4を使い、対物レンズ4とガラス板6の間をオイル5で満たす。そして、所定の開口数(NA)の対物レンズ4を用い、対物レンズ4の中心からずれた位置に照明光を入射させることにより全反射条件で照明光3をガラス板6に照射する。

落射照明は、試料全体が観察可能であるという特徴があり、全反射照明は、照明ができる範囲が局所的であるが、バックグラウンド・ノイズ(散乱光など)が極めて低く、例えば、蛍光色素1分子のような観察にも有効であるという特徴がある。そこで、落射照明と全反射照明とを組み合わせ、同一の試料について全体像観察と局所的な観察を行うことが提案されている。例えば、特開平9−159922号公報では、図1のダイクロイックミラー8に相当するミラーを照明光3の光軸対して垂直な方向に平行移動させることにより、照明光が対物レンズの瞳に入射する位置を変え、落射照明と全反射照明とを簡単に切り替える構造が開示されている。

【発明が解決しようとする課題】
上述のように特開平9−159922号公報では、落射照明と全反射照明とを切り替え可能にするために、ミラーを上記方向に平行移動させ、対物レンズの瞳に照明光が入射する位置を変えているが、対物レンズ内での照明光の散乱をおさえ、かつ、全反射を発生させるためには数十μmの単位で照明光の入射位置を調整する必要がある。しかしながら、ミラーの平行移動という直線運動の制御により、数十μm程度単位の微妙な調整は難しい。

本発明は、簡便に落射照明と全反射照明方法とを切り替えることができる顕微鏡を提供することを目的とする。

【課題を解決するための手段】
本願発明は上記目的を達成することにある。

そこで本願発明は、以下のような顕微鏡が提供される。

本願発明は、試料の照明方法を切り替えるために、対物レンズへの照明光束の入射位置を変更する変更手段を有する顕微鏡において、前記変更手段は、照明光束の屈折により光路を変更する透明部材を含み、前記透明部材を前記照明光束の光路中に挿入する挿入手段を有し、前記透明部材は、一部が切り欠かれた回転体であり、前記挿入手段は、前記回転体の前記透明部材を回転軸を中心に回転させる手段であることを特徴とする顕微鏡を提供する。

上記構成によれば、透明部材を回転させることにより、照明光束に透明部材が挿入されている状態と、切り欠きの部分が照明光束にかかり透明部材が照明光束に挿入されていない状態とを交互に切り替えることができる。

【発明の実施の形態】
本発明の実施の形態について説明する。

まず、本実施の形態の落射照明と全反射照明の切り替え方法の基本的原理については、図3(a),(b),(c)を用いて説明する。本実施の形態では、照明光束を透明部材に入射させ、照明光束を透明部材で屈折させることにより照明光束の光路を変更し、対物レンズへ照明光束が入射する位置を変えて照明方法を切り替える。ここでは、透明部材としてガラス製の平行平板を用い、平行平板を光束に対して傾斜させることにより照明光束の光路を切り替える。

照明光束3としては、光束径の小さい平行光束を用いる。照明光束3の径は、使用する対物レンズ4について予め求めておいた、全反射照明を行うために対物レンズ4の瞳に照明光を照射すべき領域31(図3(c)参照)の幅と同等もしくはそれ以下とする。例えば、レーザ光束、もしくは、水銀灯またはLED光源等から出射された光束を絞り等により絞った光束を用いる。

この光束を図3(a),(b),(c)のように平行平板11に入射させる。平行平板11を透過した照明光束3を、ダイクロイックミラー8で反射し、対物レンズ4に入射させ、試料溶液7中の試料に照射する。ダイクロイックミラー8は、図3(a)のように平行平板11に垂直に入射した照明光束3が、対物レンズ4の光軸に沿って対物レンズ4の中央に入射する位置に配置されている。よって、図3(a)のように、平行平板11を照明光束3が垂直に入射するように配置することにより、照明光束3は、平行平板11で屈折されずまっすぐに進むので、落射照明により試料を照明することができる。照明光3は、試料溶液7に接するガラス板6、および、試料溶液7を透過し、透過領域に存在する試料が発した蛍光は、対物レンズ4を通過し、ダイクロイックミラー8を透過し、不図示の結像光学系に入射して結像する。これにより、例えば、蛍光顕微鏡の場合、落射照明された試料全体の蛍光像を観察することができる。

つぎに、平行平板11を図3(b)のように光束に対して傾斜させると、照明光束3は、平行平板11に入射する時と平行平板11から出射する時にそれぞれ屈折するため、平行平板11から出射された照明光束3は、入射前の照明光束3に対してずれ量tだけ平行にずれる。ずれ量tは、図7に示すように平行平板11の傾斜角度θが大きくなるほど大きくなる。よって、平行平板11の傾斜角度θが大きくなるにつれ、対物レンズ4の瞳への照明光束3の入射位置は、対物レンズ4の光軸から遠ざかる。これにより、照明光束3を、対物レンズ4の瞳の全反射照明のための入射領域31まで移動させることができる。全反射照明のための入射領域31に入射した照明光束3は、全反射される入射角度で、試料溶液7に接したガラス板6に入射する。照明光3は、試料溶液7に接したガラス板6面で全反射され、試料溶液7は全反射の際に試料溶液7側ににじみ出すエバネッセント光で照明される。このように、平行平板11の傾斜角度を調整することにより、対物レンズ4への照明光の入射位置を容易に調整できる。

例えば、蛍光顕微鏡の場合、エバネッセント光が到達した領域(深さ数十〜数百nm程度)に存在した試料が発した蛍光は、対物レンズ4を通過し、ダイクロイックミラー8を透過し、不図示の結像光学系に入射して結像する。これにより、従来よりも弱い光強度の照明光であるエバネッセント光が達した極浅い領域に存在した試料のみの蛍光像を観察することができ、局所的な観察ができる。また、本発明の照明光の光強度は小さいので、試料を劣化させてしまうことも防止できる。

なお、対物レンズ4の瞳に対する全反射照明を実行するため照明光3の入射領域31の幅は、対物レンズ4として60倍の開口数1.45のものを用いる場合で383μm(約400μm)となる。また、100倍の開口数1.45の場合では、230μm(約200μm)である。また、対物レンズ4の光軸から入射領域31までの距離は、数mmである。このような幅の狭い領域31に精度のよく照明光3を位置合わせして入射させるためには、数十μmのオーダーで照明光束3を制御して移動させる必要があるが、本実施の形態の平行平板11を傾斜させる方法は、図7に示すように、屈折率1.514、厚さ10mmの平行平板11を用いた場合、50μm移動させるために1°を傾斜させる。これは、平行平板11に直径30mmのノブを取り付けて手動で回転させることを考えると、ノブの円周方向の移動距離で約0.25mmに相当し、人間が手で十分調整可能な量であり、手動で位置合わせすることが可能である。また、平行平板11を45度傾斜させることにより、照明光束を約3mm移動させることができるため、対物レンズ4の瞳の光軸中心の落射照明の入射位置から、全反射照明の入射領域31までノブの回転で移動可能である。よって、対物レンズ4の種類を変更した際に、ノブを回転させて平行平板11を傾斜させ、図3(c)の全反射照明の入射領域31まで照明光束3を移動させて位置合わせを行い、ノブがそれ以上回転しないように不図示の機構によりノブ可動範囲を制限し、ノブを可動範囲いっぱいに回転させることにより図3(a)の落射照明の位置から図3(c)の全反射照明の位置まで照明光束3を移動させることができ、照明方法を容易に切り替えることができる。

これに対し、従来技術のようにダイクロイックミラー8を平行移動させることにより、照明光束3の入射位置を変化させる場合、照明光束3の移動量に対して、ダイクロイックミラー8を1:1の移動量で移動させる必要がある。例えば、領域31に照明光束3を位置合わせするために、50μm照明光束3を移動させる場合には、ダイクロイックミラー8を50μm移動させることになる。50μmの移動を手動で調整することは困難であるため、ギア等の機構を使用するか、ステップモータ等を電気的に制御して移動させる必要があり、上述の本実施の形態のように簡単な構成にするのは困難である。また、ギアを使用した場合、ギア同士のかみ合わせのずれ等が発生しやすくなるが、本発明によれば、ギアを用いずに微調整可能なので、正確な調整が可能となる。

なお、本実施の形態の平行平板11としては、使用する対物レンズ4の瞳径に合わせて、90°以内の傾斜角度で、図3(a)の落射照明から図3(c)の全反射照明へ切り替えるための照明光束3のずれ量tが得られるような屈折率および厚さであり、望ましくは、傾斜角に対するずれ量tが、領域31への照明光束3の位置合わせを手動で行うことができる量であるものを用いる。平行平板11の傾斜角と照明光束3のずれ量tとの関係の例を、平行平板11の屈折率と厚さごとに図7に示す。

つぎに、本実施の形態の顕微鏡について具体的に説明する。

第1の実施の形態の顕微鏡は、図8のように、照明光源1、試料を搭載するステージ82、ステージ82の下部に配置された対物レンズ4、ベース83、接眼レンズ部84、および、撮像装置18を有する。ベース83の内部には、図9のように光源1の光軸に沿って順に、投影管(レンズ群)61、平行平板91、平行平板11、ダイクロイックミラー8、結像光学系62が配置されている。照明光源1は、予め定めた波長の励起光を照明光束3として発する光源である。よって、これらは、試料の蛍光像を観察する蛍光顕微鏡を構成する。

また、ステージ82の上方には、原子間力顕微鏡81が配置されている。よって、図8の顕微鏡は、試料の下方側から蛍光像を観察しながら、試料の上方側から極微領域の原子間力顕微鏡像を取得できる構造である。

ダイクロイックミラー8,対物レンズ4は、平行平板11に対して、図3(a)の位置関係となるように配置されている。また、平行平板11は、ベース83の外部に配置されたノブ86を回転させることにより、図9のx軸を中心に回転し、これにより、図3(a),(b),(c)を用いてすでに説明した原理により、照明光源1からの照明光束3を平行移動させ、落射照明と全反射照明とを切り替えることができる。

また、平行平板91は、ベース83の外部に配置されたノブ85を回転させることにより、上記x軸と直交するy軸を中心に回転するようにギア等を組み合わせることで構成されている。よって、ノブ85を回転させることにより、平行平板91は、照明光束3に対して傾斜し、その傾斜の方向は、平行平板11が傾斜する方向と直交している。したがって、平行平板91を傾斜させることにより、照明光束3が対物レンズ4の瞳に入射する位置を移動させることができ、移動方向は、平行平板11を傾斜させた場合の移動方向と直交する。よって、対物レンズ4を別の種類のものに交換した場合等において、落射照明の状態(図3(a))で照明光束3が対物レンズ4の瞳に入射する位置が、対物レンズ4の光軸からずれている場合には、平行平板11および平行平板91をそれぞれ傾斜させることにより、入射位置を位置合わせして光軸に一致させる調整を行うことができる。このように落射照明の状態で照明光束3が対物レンズ4の光軸と一致するように位置合わせをすることにより、全反射照明に切り替えた際に、照明光束3を対物レンズ4の瞳の全反射照明のための入射領域31に精度良く入射させることができる。よって、精度良く全反射照明で観察を行うことができる。

なお、この位置合わせ調整は、試料溶液7として色素溶液を配置し、色素が発する蛍光を観察して、照明光束3の通過軌跡を確認することにより、容易に行うことができる。例えば、平行平板11を図3(a)の落射照明の状態として、色素溶液中の照明光束3の通過軌跡(蛍光を発する線)を確認し、これが対物レンズ4の視野の中心に位置するように、平行平板11および平行平板91の傾斜角度を調整する方法や、平行平板11を傾斜させて図3(b)の状態として、照明光束3の通過軌跡(蛍光を発する線)が、対物レンズ4の視野の中心(光軸)を通過するように、平行平板91を傾斜させる方法を用いることができる。

図8、図9の顕微鏡において、試料溶液7を観察する場合の各部の動作について説明する。

照明光源1から出射された照明光束3は、投影管(レンズ群)61を通過し、平行平板91および平行平板11を通過し、ダイクロイックミラー8で反射されて対物レンズ4に入射する。このとき、落射照明で試料溶液7を照明する場合には、平行平板11を図3(a)の状態とし、照明光束3を対物レンズ4の光軸に入射させる。これにより、対物レンズ4で集光された照明光束3は、オイル5を通過してガラス板6に入射し、ガラス板6および試料溶液7を透過する。透過領域に存在する試料が発した蛍光は、対物レンズ4を通過し、ダイクロイックミラー8を透過し、結像光学系62に入射して結像する。結像した蛍光像は、撮像装置18で撮像されるか、もしくは接眼レンズ部84を介して観察される。これにより、落射照明された試料全体の蛍光像を観察することができる。

一方、全反射照明で試料溶液7を照明する場合には、ノブ86を回転させて平行平板11を図3(c)の状態まで傾斜させて、対物レンズ4の瞳の領域31に照明光束3を入射させる。これにより、照明光束3は、全反射される入射角度で、ガラス板6に入射し、全反射の際にガラス板6から外側ににじみ出すエバネッセント光が生じ、試料溶液7側ににじみ出したエバネセットにより光主平面方向に極狭い領域が照明される。また、エバネッセント光が到達する深さは、試料溶液7とガラス板6の界面から試料溶液側に数十〜数百nm程度と極浅く、ガラス板6からこの深さに存在した試料のみが照明される。試料が発した蛍光は、対物レンズ4を通過し、ダイクロイックミラー8を透過し、不図示の結像光学系に入射して結像する。結像した蛍光像は、撮像装置18で撮像されるか、もしくは接眼レンズ部84を介して観察される。これにより、全反射照明された試料の極狭く、かつ、浅い領域の蛍光像を観察することができる。これにより、蛍光分子を一分子単位で観察することも可能である。

また、このように試料溶液の下側から落射照明または全反射照明を行いながら、ステージ82の上方の原子間力顕微鏡により、試料溶液7中の試料の微小領域の表面像を取得することができるため、同一の試料について、同一時点に微小領域を複数種類の観察方法で容易に観察することができるため、経時変化する試料を多角的に観察するのに有効である。

上述の第1の実施の形態では、図3(a)、(b)、(c)のように、平行平板11が照明光束3の光路中に常に挿入され、平行平板11の傾斜角度を変えることにより、落射照明と全反射照明とを切り替える構成であったが、図3(a)の状態は、平行平板11が挿入されていない状態と同じである。そこで、第2の実施の形態の顕微鏡では、図4(a),(b),(c)および図6に示したように、平行平板を所定の角度で傾斜させてリング状(唐笠状)にしたものを切り欠いた光学部材12を回転させることにより、落射照明と全反射照明とを光束に切り替える構成とした。

光学部材12は、平行平板を所定の角度で傾斜させてリング状(唐笠状)にしたものを180°切り欠いたものである。この光学部材12を図4(a)、図5のように、光学部材12の中心軸15を中心に回転させることにより、照明光束3に光学部材12が挿入されている状態(図4(b))と、切り欠き13の部分が照明光束3にかかり、光学部材12が照明光束3に挿入されていない状態(図4(a))とを交互に切り替えることができる。よって、光学部材12の平行平板の傾斜角度θ(図4(b))を、図3(c)と同様に全反射照明のための領域31に照明光束3を入射させることができる角度に設定しておくことにより、図4(a)の状態では落射照明、図4(b)の状態では全反射照明を行うことができる。よって、光学部材12を回転駆動部19により回転させることにより、落射照明と全反射照明とを交互に切り替えることができる。また、回転駆動部19が回転速度を調整することにより、落射照明と全反射照明とを所望の速度で高速に切り替え可能である。

光学部材12の回転速度と撮像装置18の撮像のタイミングを調節するために、回転制御部19と撮像装置18には、制御部20が接続されている。制御部20は、回転制御部19に光学部材12の180°の回転を指示することにより、落射照明と全反射照明とを切替える。また、制御部20が、撮像装置18の撮像速度に同期させて、光学部材12を高速で連続的に回転させることにより、落射照明による試料の全体の蛍光像と、全反射照明によるガラス板6近傍の蛍光像とを、交互に(例えば1フレームごとに)撮像装置で連続撮像することができる。したがって、撮像後に、落射照明による蛍光像画像と、全反射照明による蛍光像画像とを分離する編集を行うことにより、生きた細胞が試料である場合等に、ほぼ同一瞬間の試料の蛍光像として、落射照明による全体像画像と全反射照明による局所的な画像とをそれぞれ取得できる。よって、試料の変化を2種類の照明方法により観察することができる。

なお、第2の実施の形態の顕微鏡において、図4(a)〜(c)、図5以外の部分の構成は、第1の実施の形態の顕微鏡と同様であるので説明を省略する。

また、第3の実施の形態の顕微鏡として、図6に示したように、第1の実施の形態の平行平板11の代わりに方解石16を用いるものを説明する。方解石16を用いることにより、同一照明光源からの照明光束3の垂直及び水平の偏光成分を分離し、光路を変えることができる。よって、一方の偏光成分の照明光束3a対物レンズ4の光軸に沿って入射させ、他方の偏光成分の照明光束3bを対物レンズ4の全反射照明のための入射領域31に入射させることにより、落射照明と全反射照明の2種類の照明方法で、同時に試料溶液7を照明することができる。

このとき、ダイクロイックミラー8と撮像装置18との間には、2種類の照明方法による蛍光像を偏光成分ごとに分離するために偏光ビームスプリッタ17を挿入する。これにより、全反射照明による蛍光像と、落射照明による蛍光像とを分離することができる。分離された2種類の蛍光像は、それぞれ撮像装置18により撮像する。

第3の実施の形態の顕微鏡は、生きた細胞が試料である場合等に、同一瞬間の試料の蛍光像を、落射照明と全反射照明という2種類の照明方法により同時に観察することができる。2種類の蛍光像は、偏光ビームスプリッタ17を挿入されているため、直接の反射光による背景光が抑制され、高感度な像が得られる。

なお、第3の実施の形態の顕微鏡において、図6以外の部分の構成は、第1の実施の形態の顕微鏡と同様であるので説明を省略する。

また、第3の実施の形態の顕微鏡では、複屈折性を有する他の種類の結晶を、方解石の代わりに用いることが可能である。

上述してきた第1〜第3の実施の形態の顕微鏡は、平行平板11、光学部材12、方解石16という透明部材に照明光束3を透過させ、光路をずらすという簡単な構成で、落射照明と全反射照明とを切り替えることができる。しかも、2種類の照明方法の切替を、第1の実施の形態では手動で精度良く行うことができ、第2の実施の形態では高速切り替えが可能であり、第3の実施の形態では、2種類の照明方法による同時観察が可能である。いずれの場合も、全反射照明のための入射領域31に精度良く照明光束を入射させることができるため、全反射照明時に、高いS/N(像が明るく背景が暗い)の像が得られ、蛍光色素分子の高感度観察が可能となる。

また、上述の第1の実施の形態は、蛍光顕微鏡と原子間力顕微鏡とを搭載した顕微鏡を示したが、原子間力顕微鏡に代えて、トンネル顕微鏡(STM)、フォトンSTM(NSOM)を搭載することも可能である。また、蛍光顕微鏡に限らず、レーザ顕微鏡等にも本発明は有効である。

上述してきたように、本発明によれば、簡便に落射照明と全反射照明方法とを切り替えることができる顕微鏡を提供することができる。

BACKGROUND OF THE INVENTION
The present invention relates to a microscope having an illumination method switching mechanism, and more particularly to a microscope having an epi-illumination and total reflection illumination switching mechanism.
