The present invention relates to a microscope apparatus that can be used by switching between a confocal microscope and a total reflection microscope.
Confocal microscopes and total reflection fluorescence microscopes are widely used for observing living cells and the like. Both of them match in that a laser light source is used, and a microscope apparatus that can be used by switching between a confocal microscope and a total reflection fluorescence microscope has been proposed.
In the conventional microscope, when switching from the confocal microscope to the total reflection microscope, an optical member for condensing the laser light on the total reflection area at the pupil position of the objective lens must be inserted into the illumination optical system. For this switching, a large switching mechanism is required, which leads to an increase in the size of the microscope apparatus.
The present invention has been made in view of such problems, and an object of the present invention is to provide a microscope apparatus that can be used by switching between a confocal microscope and a total reflection microscope, which is miniaturized.

The microscope body 2 includes a

Next, the case where the
When used as a total reflection microscope, the above-described confocal
The condensing
The chief ray of the laser light incident on the
When used as a total reflection microscope, laser light emitted from a laser light source (not shown) of the confocal
The laser light incident on the
The fluorescence excited by the evanescent wave and expressed in the
As described above, the
Note that the tilt angle of each XY mirror constituting the two-
With reference to FIG. 4, the transition to total reflection illumination will be described in detail. FIG. 4A shows an illumination state when used as a scanning microscope or an epifluorescence microscope, and FIG. 4B shows an illumination state when used as a total reflection microscope.
As shown in FIG. 4A, when used as a scanning microscope or an epifluorescence microscope, a laser beam incident on the
On the other hand, when the laser beam is used as a total reflection microscope shown in FIG. 4B, the principal ray of the laser beam is separated from the
As described above, the
In the present embodiment, as shown in FIG. 5, the tilt of the XY mirrors of the two-
For example, as shown in FIG. 4B, when the optical axis I1 of the laser light incident on the
Further, in the present embodiment, the
For example, when the condensing
Further, in the
As described above, in the
As described above, the optical axis I1 of the laser light is shifted upward from the
Therefore, in the
For example, as shown in FIG. 7, the optical axis of the illumination
Further, the
Further, as shown in FIG. 8, the
For example, in FIG. 8, when the reflecting
In the above, the reflecting member is composed of a total of three members, a first reflecting member (mirror 207), a second reflecting member (folding mirror 208), and a third reflecting member (mirror 209). Although the second reflecting
In addition, as shown in FIG. 9, the
For example, in FIG. 9, the laser light that is shifted from the
