アセトバクター・マローラムNCI1683(Acetobacter malorum NCI1683)株(FERM BP−10595)を、5mlのYPG液体培地(グルコース3重量/容量%、ポリペプトン0.3重量/容量%、酵母エキス0.5重量/容量%、pH6.5)に植菌し、30℃で48時間、120rpmにて振とう培養して前々培養を行った。
実施例1と同様にして、アセトバクター・マローラムNCI1683(Acetobacter malorum NCI1683)株(FERM BP−10595)を、YPG培地に酢酸ナトリウムを最終濃度0.8%になるように加えた培地で48時間培養し、ここで得られた培養液を8,000gで5分間遠心分離して酢酸菌体を回収し、6分の1容量の培養上清で再懸濁して6倍菌体濃縮液を調製した。
なお、上記の静置処理を行わずに、直ちに凍結乾燥した後、実施例1と同様にして、培地1リッター当りの乾燥酢酸菌体量(g/リッター)、乾燥酢酸菌体1g当りの酢酸菌型セラミド含有量(mg/g)、及び、培地1リッター当りの酢酸菌型セラミド生産量(mg/リッター)を求めたものを「処理前」とした。
実施例1と同様にして、アセトバクター・マローラムNCI1683(Acetobacter malorum NCI1683)株(FERM BP−10595)を、YPG培地に酢酸ナトリウムを最終濃度0.8%になるように加えた培地で48時間培養し、ここで得られた培養液を8,000gで5分間遠心分離して酢酸菌体を回収し、6分の1容量の培養上清で再懸濁して6倍菌体濃縮液を調製した。
試験例1と同様にして、アセトバクター・マローラムNCI1683(Acetobacter malorum NCI1683)株(FERM BP−10595)を、YPG培地に酢酸ナトリウムを最終濃度0.8%になるように加えた培地で48時間培養し、ここで得られた培養液を8,000gで5分間遠心分離して酢酸菌体を回収し、6分の1容量の培養上清で再懸濁して6倍菌体濃縮液を調製した。
その後、試験例2と同様にして6N塩酸により上記6倍菌体濃縮液をpH3に調整し、40℃でインキュベーターの中に表4に示す間静置した。
実施例1と同様にして、グルコンアセトバクター・ハンゼニイNCI1468(Gluconacetobacter hansenii NCI1468)株(FERM BP−10596)を、YPG培地に酢酸ナトリウムを最終濃度0.8%になるように加えた培地で48時間培養し、ここで得られた培養液を8,000gで5分間遠心分離して酢酸菌体を回収し、6分の1容量の培養上清で再懸濁して6倍菌体濃縮液を調製した。
Claims (4)
- 培養終了後の酢酸菌を、集菌後、pH2.0〜4.5及び温度30〜70℃で1日〜7日間保持することを特徴とする酢酸菌中の酢酸菌型セラミド含有量の増加方法。
- 酢酸菌として、アセトバクター・マローラムNCI1683(Acetobacter malorum NCI1683)株(FERM BP−10595)を用いることを特徴とする請求項1に記載の酢酸菌中の酢酸菌型セラミド含有量の増加方法。
- 酢酸菌として、グルコンアセトバクター・ハンゼニイNCI1468(Gluconacetobacter hansenii NCI1468)株(FERM BP−10596)を用いることを特徴とする請求項1に記載の酢酸菌中の酢酸菌型セラミドの増加方法。
- セラミド含有量が乾燥菌体1gあたり7〜18.59mgであることを特徴とする請求項2又は請求項3のいずれか1項に記載の酢酸菌中の酢酸菌型セラミド含有量の増加方法。
