【発明の属する技術分野】
本発明は、密閉された造粒室内に原料を投入して目的とする粉・粒状の製品を造粒する流動層、噴流層、及び噴霧造粒等を含む気流式の造粒装置およびこの造粒装置を用いた粉末製造方法に関するものである。

【従来の技術】
一般に、気流式の造粒装置は、造粒室の内部に様々な材料を噴霧して、当該材料からなる粉・粒状あるいは顆粒状の製品を製造するものである。

この造粒装置を、医薬品製造装置や食品製造装置等の洗浄・滅菌の必要な装置に用いた場合、造粒後、造粒室内部を十分に洗浄・滅菌する必要がある。

例えば、特開平5−228353号公報(特許文献1)には、造粒室内で3次元的に回転して該造粒室の内壁面に流体を噴出するノズルを利用して、造粒室内壁面を洗浄する造粒装置が開示されている。

また、造粒装置によって製造される製品は、一部が造粒室に付着し残留するので、製品の回収率(投入した材料から実際に造粒装置から取り出される製品の割合)が低下するという問題が生じる。

そこで、一般に、製品の造粒後、造粒室の洗浄前に、造粒室の外部からエアノッカー等により該造粒室に衝撃を与えて、造粒室内壁面に付着した製品を払い落とし、製品の回収率を上げるようにしている。

【発明が解決しようとする課題】
ところで、上記特許文献1に開示された造粒装置では、造粒室内でノズルが3次元的に回転駆動する構造となっているので、通常、造粒の邪魔にならない位置に格納する必要がある。つまり、上記ノズルは、洗浄位置と格納位置との間を移動することになる。

この場合、ノズルを回転させたり、移動させたりする駆動部の一部あるいは全部が造粒室内に設けられていることになり、この駆動部を構成する部材の摩擦等により異物が発生する虞がある。

したがって、上記特許文献1に開示された造粒装置では、ノズルにより造粒室内が洗浄されるが、洗浄後、格納位置まで移動することになり、この移動によって異物が発生し、洗浄後の造粒室内に異物が混入する虞がある。

また、エアノッカー等により造粒室に外部から衝撃を与えた場合、該造粒室を構成する構成要素、例えば接合フランジ部などが振動により擦れ合って、接合フランジ部を構成するガスケットやフランジなどから異物が発生する虞がある。

したがって、従来の造粒装置では、造粒室の洗浄前後で異物が発生する虞があるので、造粒した製品に異物が混入する虞がある。このため、従来の技術においては、洗浄前後で造粒室に異物の発生がなく、製品への異物の混入が無い造粒装置を提供することは困難であった。つまり、従来の造粒装置では、製品への異物混入を極端に嫌う製剤等の製品を造粒する造粒装置として使用することが困難であった。

従って、従来、このような造粒装置を、異物混入を嫌う食品等の製品(粉末)の製造に使用することは考えられず、特に、無菌・無塵の状態が必要な製剤等の製品(粉末)の製造に使用することは全く考えられなかった。

このため、上記のような異物混入を嫌う粉末、特に、無菌の粉末を製造する方法として、従来では、主に凍結乾燥法、スプレードライ法等が用いられていた。

しかしながら、凍結乾燥法は、冷凍機や真空ポンプ等の付帯設備が多く初期設備投資や保守・運転費用に高額を要するほか、製造に長時間を要するという問題が生じる。

また、スプレードライ法では、凍結乾燥法に比べて短時間で粉末を製造できるが、低溶媒含量の粉末を得るためには、高温での乾燥が必要となるので、熱に弱い物質には適さない上、装置に付着して熱劣化した粉体が粉末内に混入する虞がある。

本発明は、上記の問題点に鑑みなされたものであって、その目的は、洗浄前後で造粒室に異物の発生がなく、製品への異物の混入が無い造粒装置、および本発明の造粒装置を用いて低温で且つ短時間で無菌・無塵の粉末を製造することができる粉末製造方法を提供することにある。

【課題を解決するための手段】
上記の課題を解決するために、本発明の造粒装置は、密閉された造粒室内に原料を投入および/または噴霧して目的とする粉・粒状の製品を造粒する気流式の造粒装置であって、上記造粒室内壁面に、該造粒室内壁面に対して流体を噴出するノズルが複数個設けられ、上記各ノズルの流体噴出方向がそれぞれ所定の方向に固定され、上記ノズルから噴出する流体を、気体、液体、スチームのうち何れかに切り替える切り替え手段が設けられていることを特徴としている。

上記の構成によれば、造粒室内壁面に、該造粒室内壁面に対して流体を噴出するノズルが複数個設けられていることで、該各ノズルによって噴出された流体によって、該造粒室内壁面に付着した粉・粒状の製品を剥離することができる。つまり、造粒室内壁面に付着した製品を、該造粒室に衝撃を与えることなく剥離することが可能となる。

また、ノズルの流体噴出方向が所定方向に固定されているので、ノズルが回転や移動等の駆動せずに、造粒室内壁面に対して流体を噴射することができる。これにより、ノズルの回転や移動等の駆動に伴う造粒室内への異物の発生を無くすことができる。

ここで、流体として気体を使用すれば、造粒中に造粒室内壁面に付着した粉体の払落としを行なうことができる。また、流体として液体を使用すれば、この液体に洗浄成分を含ませることで、造粒室内壁面の洗浄を行なうことができる。さらに、流体として滅菌可能な高温のスチームを使用すれば、造粒室内壁面の滅菌を行なうことができる。

これにより、流体として気体を使用した場合には、造粒室に外部から衝撃を与えることなく、該造粒室内壁面に流体を噴出することにより、該内壁面に付着した製品を払い落とすことができるので、回収する製品には異物が混入する虞がなくなる。したがって、異物混入の無い製品を、高回収率で回収することができる。

また、流体として洗浄成分を含んだ液体を使用した場合には、ノズルの流体噴出方向が所定の方向(内壁面の洗浄可能な方向)に固定されているので、ノズル自体を造粒室内で回転させたり移動させたりする必要がなく、造粒室内壁面の洗浄を行なうことができる。これにより、ノズルが回転、移動等の駆動することによって発生する異物が洗浄後に造粒室に混入するといった問題が生じない。

したがって、洗浄前後で造粒室に異物は発生しなくなるので、造粒室で製造された製品への異物の混入を無くすことができる。この結果、製品への異物混入を極端に嫌う製剤等の製品を造粒する造粒装置として、上記構成の造粒装置を使用することが可能となる。

また、上述のように、流体の種類を順番に切り替えれば、造粒室内壁面に対する製品の払い落としから、洗浄・滅菌までを順に行なうことができる。この流体の切り替えを自動化すれば、製品の払い落としから洗浄・滅菌までを自動的に行なうことが可能となる。

このように、造粒室内の製品の払い落としから洗浄・滅菌までを自動的に行い、しかも、造粒前後において造粒室に異物が混入しない造粒装置では、造粒室を無菌・無塵環境にすることが容易となり、このような無菌・無塵環境で造粒する必要のある製剤、特に用時溶解注射剤の造粒にも上記構成の造粒装置を好適に使用することが可能となる。

また、本発明者等は、種々の検討を行なった結果、本発明の造粒装置を用いて、スプレードライ法での乾燥温度よりも低い温度で、噴霧造粒工程を行い、引き続き流動層乾燥工程を行うことにより、凍結乾燥法で製造した無菌製剤と同程度の化学的品質を持つ粉末を短時間で製造できることを見出した。

また、本発明の造粒装置の内壁面に設けられたノズルから流体(除湿エアー等)を効率的に噴射することで、該内壁面への粉体の付着を軽減すると共に、流動層の形成を促進できるので、製造される粉末の短時間での乾燥を可能とし、且つ回収率の向上が図れることを見出した。

ここで、粉末中の許容溶媒含量については、粉末として製造される薬物の種類によって異なる。そこで、溶媒が水の場合、薬物が加水分解しない程度の溶媒含量と定義し、溶媒が有機溶媒の場合、人体に影響を与えない程度(残留溶媒がない程度)の溶媒含量と定義する。

なお、本願発明に用いられる粉体/製剤となる物質は、医薬品や食品として使用するに際し毒性がなければ、特に限定されず、非常に幅広い物質に適用可能である。但し、噴霧造粒工程および流動層乾燥工程での温度で液状となる物質は、単独で本発明を適用することは原理上困難である。また、噴霧造粒工程および流動乾燥工程での温度で分解する物質も、単独で本発明を適用することは原理上困難である。

【発明の実施の形態】
本発明の一実施の形態について説明すれば、以下の通りである。なお、本実施の形態では、本発明の造粒装置を流動層造粒装置に適用した造粒システムについて説明する。

本実施の形態に係る造粒システムは、図2に示すように、造粒装置1を中心に、原料供給部2(第1流体供給装置)、圧縮エアー供給部3(第2流体供給装置)、流動用エアー供給部4、排気部5、第1洗浄・滅菌部6(第3洗浄装置)、第2洗浄・滅菌部7(第3洗浄装置)、第3洗浄・滅菌部8(第1洗浄装置)、第4洗浄・滅菌部9(第2洗浄装置)が配置された構成である。

上記造粒装置1は、二流体スプレーノズル100を下部に備えた略円筒形状の造粒室200を備えている。

上記二流体スプレーノズル100は、上記原料供給部2からの液状材料20(第1流体)と、上記圧縮エアー供給部3からの圧縮エアー(第2流体)とを混合して先端開口部100aから造粒室200内に噴霧するようになっている。

上記構成の造粒装置1では、上記流動用エアー供給部4から、造粒室200の内部に上向きに流動用エアーを吹き出させると共に、液状材料20を上記二流体スプレーノズル100から流動用エアー中に同じく上向きに噴霧させ、噴霧された液状材料20の微粒子を造粒室200の内部で浮遊させつつ、液状材料20の水分を蒸発させて造粒する。この際、当該微粒子は上記二流体スプレーノズル100から噴霧された液状材料20によって間欠的にレイヤリングされて成長する。このような過程を繰り返して所定の粒径を有する粒子を製造するようになっている。

なお、上記構成の造粒システムでは、図示していないが、造粒室200内に噴霧供給する上記液状材料20の温度を最適化するために、二流体スプレーノズル100の外周部の略全体に流体を流通させるジャケットを設けてある。当該ジャケットに温度調節用の媒体を流通させて二流体スプレーノズル100を加熱または冷却することで、噴霧する液状材料20の温度や粘性を最適化し、噴霧された液状材料20の粒径を均一化する。この結果、噴霧した液状材料20の凝集・成長の程度を揃え、粒径分布の狭い製品粒子を得ることができる。

また、上記構成の造粒装置においては、図2に示すように、造粒途中にある粒子等がエアー排出口となる開口部200bから排出されてしまうのを防止するために、フィルタ206、および、当該フィルタ206に付着した粒子を払い落とすための払落し手段、さらには、上記造粒室200の内部圧力を瞬間的に上昇させるための圧力付加手段とが付加されている。

上記フィルタ206を設けることにより、粒子の排出が防止できるため、造粒室200の容積を比較的小さく構成することができ、上記圧力付加手段を設けていることにより、造粒途中の粒子を締め固めて重質な製品粒子を得ることができる。

以下、上記構成の造粒システムを構成する個々の部材について説明する。

上記原料供給部2は、図2に示すように、液状材料20が収容された容器21と、該容器21に収容された液状材料20を空気圧で押し出すためのポンプ22とで構成されている。この原料供給部2からの液状材料20は、上記ポンプ22のレギュレータバルブ(図示せず)を操作することで、該ポンプ22からフィルター(図示せず)を通して容器21の該液状材料20内に空気が供給され、この空気によって、後述する第3洗浄・滅菌部8と共通の流路を切り替える切替バルブ10と容器21との間に設けられたフィルター(図示せず)を通して該切替バルブ10から二流体スプレーノズル100に供給されるようになっている。

上記圧縮エアー供給部3は、圧縮機31を備え、外部から取り込んだエアーを圧縮して、フィルター(図示せず)を通して切替バルブ12から二流体スプレーノズル100に供給するようになっている。このときの圧縮条件は、0.6MPa、300NL/minとする。

なお、上記二流体スプレーノズル100では、上記原料供給部2から供給される液状材料20と、上記圧縮エアー供給部3から供給される圧縮エアーとは別々の流路に導入され、最終的に混合され先端開口部100aから噴霧されるようになっている。上記二流体スプレーノズル100の詳細については、後述する。

上記流動用エアー供給部4は、所定の温度・湿度に温度設定した流動用エアーを、フィルター(図示せず)を通して開口部200aから造粒室200に上向きに供給するようになっている。当該温度・湿度は、主に造粒室200の内壁面200dに設けた温度計・湿度計(図示せず)を用いて計測する。

上記流動用エアー供給部4は、外部からエアーを取り込むブロワー41と、該ブロワー41によって取り込まれたエアーを貯蔵するエアタンク42と、該エアタンク42に貯蔵されたエアーを乾燥させるドライヤ43と、該ドライヤ43で乾燥されたエアーを所定の温度に温めるヒータ44とで構成されている。すなわち、ドライヤ43によってエアーの湿度を調整し、ヒータ44によってエアーの温度を調整することで、所定の温度・湿度に設定されたエアーを造粒室200に供給するようになっている。

まず、本実施例に係る造粒装置の造粒室200の容量を800L、表面積を10m2、30%固形分のソルビン酸カリ水溶液をモデル原料として、造粒室200の原料の入口温度80℃、出口温度50℃、噴霧液速度120g/min、スプレー空気量300L/min(2本使用)において、33.4kgの液体原料(ソルビン酸カリ水溶液)を吹き込み(液滴径は10μm以下)、約10kgのソルビン酸カリの微細粒子を造粒室200内に生成させた。

ここで、2つの条件によって、造粒室200内壁面に付着した微細粒子の払落としおよび洗浄を行なった結果について以下に述べる。

(a)第1ノズル201〜第4ノズル204を使用しないとき
まず、製品回収管308から製品を排出する際に、流動化空気を流通させつつフィルタ206に逆洗エアーを吹き込むことを行なって、造粒室200内壁面200dに付着した微細粒子を払落とした。この場合、造粒室200内壁面200dに付着した微細粒子を製品として回収することが非常に困難であり、この時の製品回収率は70%(約7kg)であった。残りの約3kgの微細粒子は、造粒室200内壁面200dに残留していることになる。
