この発明は粒子分析装置に関し、とくに、粒子の特徴パラメータを用いて2次元頻度分布図(スキャッタグラム)を作成しその分布図に現れる粒子の集団を分画して粒子の種類や数を特定する分析装置に関する。

【従来の技術】
従来、この種の粒子分析装置において、分布図上に予め複数の領域を設定し、分布図に現れる複数の粒子の各々についてその設定領域に対する帰属度を算出し、帰属度に応じて分画領域を決定するようにしたものが知られている(例えば、特開平6−3252号公報参照)。

【発明が解決しようとする課題】
ところで、従来の粒子分析装置を例えば血液中の血球を分析する血液分析装置として使用する場合、測定対象の血液を希釈する試薬の種類や量、血球から電気的、或いは光学的情報を検出する素子の汚れ、検出した情報から特徴パラメータを得るために該情報を電気信号に変換する電気回路の増幅度の変化、などのような何らかの原因により、分布図の作成に用いる特徴パラメータが変動した場合には、2次元頻度分布図に現れる粒子の集団が移動して正しい分画が行われず、誤まった分析結果が得られるという問題点がある。

この発明はこのような事情を考慮してなされたもので、誤まった分画が行われたときにはそれを分画異常と判定する機能を備えた粒子分析装置を提供するものである。

【課題を解決するための手段】
この発明は、複数の粒子の各々から特徴パラメータを検出する検出部と、検出した特徴パラメータを用いて少なくとも2つの2次元頻度分布図を作成する分布図作成部と、各分布図に出現する粒子を粒子集団に分画する分画部と、2つの分布図において共通する種類の粒子を含む粒子集団がそれぞれ分画されるときに、それらの粒子集団の粒子数を比較する演算部と、その比較結果に基づいて上記分布図における分画の異常を判定する判定部とを備える粒子分析装置を提供するものである。

【発明の実施の形態】
この発明の対象粒子は、主に血液や尿のような体液中に含まれる有形物質であるが、工業用の無機又は有機物質からなる粒子であってもよい。
この発明の検出部には、例えばフローサイトメータ、つまり粒子含有液をシース液に包んで流すフローセルと、粒子含有液の各粒子から特徴パラメータを検出する光学素子とからなる装置を用いることができる。
この場合、検出される特徴パラメータとしては、前方散乱光、側方散乱光、蛍光(例えば側方蛍光)などに基づく光学的な情報が挙げられる。
フローサイトメータではこれら各種光学的情報につき前記光学素子によって光電変換が行われ、粒子の特徴に応じたパルス信号が得られるが、そのピークレベルを光強度としたり、パルス信号が所定の閾値を超えている時間をパルス幅としたりして特徴パラメータとすることができる。すなわち、前方散乱光情報としては前方散乱光強度や前方散乱光パルス幅を、側方散乱光情報としては側方散乱光強度や側方散乱光パルス幅を、側方蛍光情報としては側方蛍光強度や側方蛍光パルス幅を、前記特徴パラメータとすることができる。

また、分布図作成部により作成される2次元頻度分布図としては、検出部としてフローサイトメータを用いる場合には、例えば側方散乱光強度と側方蛍光強度をパラメータとする分布図、側方散乱光強度と前方散乱光強度をパラメータとする分布図、側方蛍光強度と前方散乱光強度をパラメータとする分布図、および側方蛍光強度と前方散乱光強度をパラメータとする分布図などが挙げられる。

以下、図面に示す実施例に基づいてこの発明を詳述する。なお、各図面の共通の要素には共通の番号および記号を付記している。

血液分析装置の構成
図1はこの発明の方法を用いた血液分析装置の光学系を示す斜視図である。同図においてレーザダイオード21から出射されたビームはコリメートレンズ22を介してシースフローセル1のオリフィス部13を照射する。オリフィス部を通過する血球から発せられる前方散乱光は、集光レンズ24とピンホール板25とを介してフォトダイオード26に入射する。

一方、オリフィス部13を通過する血球から発せられる側方散乱光と側方蛍光については、側方散乱光は集光レンズ27とダイクロイックミラー28とを介してフォトマルチプライアチューブ(以下、フォトマルという)29に入射し、側方蛍光は集光レンズ27とダイクロイックミラー28とフィルタ36とピンホール板30を介してフォトマル31に入射する。

図2は図1に示す血液分析装置の流体系を示す系統図である。同図において、先ず洗浄工程においては、バルブ41,50が開かれ、シース液を収容したシース液チャンバー42からシース液が圧力装置43から印加される圧力Pによって送出され、バルブ41と定量シリンジ44とノズル6とを介して廃液チャンバー45へ排出されると共に、バルブ50とシースフローセル1とを介して廃液チャンバー45に排出され、所定時間後にバルブ41,50が閉じられる。これによって、定量シリンジ44,ノズル6,シースフローセル1およびその経路がシース液により洗浄される。

次に、測定工程においては、バルブ46,47が開かれ、血液含有試料液を試薬で反応させて収容する反応チャンバー48から試料液が吸引装置49の負圧により吸引され、バルブ46とノズル6間の経路が試料液で満たされると、バルブ46,47が閉じられる。次に、バルブ50が開かれると、シース液がシース液チャンバー42から圧力装置43の圧力によりシースフローセル1へ送出され、廃液チャンバー45に排出される。

2次元頻度分布図の作成
図8は、図2中に図示しなかった検体,定量部,試薬供給部を示すものであり、試料の調整に関し詳細に説明するための図である。
図8に示すように、検体容器80から血液(検体)が定量部81〜84によってそれぞれ必要量だけ吸引定量され、反応チャンバー48、51、52、53へそれぞれ分配される。つまり、「有核赤血球測定モード」での測定用に定量された血液は反応チャンバー48へ分配される。「白血球、好塩基球測定モード」での測定用に定量された血液は反応チャンバー51へ分配される。「白血球4分類測定モード」での測定用に定量された血液は反応チャンバー52へ分配される。「網赤血球測定モード」での測定用に定量された血液は反応チャンバー53へ分配される。そして、反応チャンバー48、51、52、53のそれぞれへ、対応する試薬供給部85〜88により、所定の試薬が供給され、血液と試薬を反応させる。このようにして各測定モードに応じた複数の試料が、1つの検体から調製され、シースフローセル1にて順次測定される。
つまり、入力部61(図3)において、「有核赤血球測定モード」,「白血球、好塩基球測定モード」、「白血球4分類測定モード」、「網赤血球測定モード」の4種類の測定モードのオーダがそれぞれ設定されると、オーダに応じて各測定モードが次のように実行される。

「有核赤血球測定モード」では、血液18μlがストマトライザーNR溶血剤(シスメックス(株)製)882μlとともに反応チャンバー48に運ばれる。そしてストマトライザーNR染色液(シスメックス(株)製)18μlが添加される。この状態で約7秒間反応させることで、赤血球が溶血させられ、白血球・有核赤血球が染色される。

図9は、この発明の血液分析装置の動作を示すフローチャートである。このフローチャートを用いて上記の試料の調製と測定の工程の一連の流れを整理して説明する。
ステップS1:患者から採取された血液を吸引する。
ステップS2:各測定モードでの測定に必要な量を、定量して分配する。
ステップS3a〜S3d:定量した各血液に、測定モードに応じて希釈液,染色液,溶血剤など所定の試薬を添加して反応処理を施し、各測定モード(「有核赤血球測定モード」,「白血球,好塩基球測定モード」,「白血球4分類測定モード」,「網赤血球測定モード」)ごとの試料を調製する。
ステップS4a〜S4d:測定モードごとに調製した各試料を、順次検出部に送液し、検出部により光学的情報を検出する。
ステップS5a〜S5d:検出した光学液情報に基づき、各測定モードごとに2次元頻度分布図を作成する。
ステップS6a〜S6d:作成した各分布図上で、出現した粒子を分画し、粒子の種類ごとに計数する。
ステップS7:複数の分布図における分画、計数の結果に基づき、分画異常の有無を判定する(このステップは以下に詳述する)。

分画異常の判定
