Description
【0001】

BACKGROUND OF THE INVENTION
The present invention relates to a microscope image acquisition apparatus.
[0002]
[Prior art]
The image acquisition device includes an image acquisition unit that acquires a plurality of images continuously in time, a display device that displays a sample image acquired by the image acquisition unit over time, and a recording device that records the image. .
[0003]
Conventionally, using an image acquisition device, a phenomenon (change over time) that occurs in a living cell when the living cell (specimen) is irradiated with light is recorded as an image, and the recorded image is reproduced to perform image analysis. Is generally done.
[0004]
At this time, not only an image in which a change has occurred but also images before and after the change (control image) are generally recorded so that the image can be reliably acquired.
[0005]
By the way, when this image acquisition device is used in, for example, an optical scanning microscope, the fading of a living cell or a fluorescent dye previously injected into the living cell is advanced by scanning with a laser beam.
[0006]
Therefore, conventionally, images are recorded by the following two methods.
(1) The timing for recording a control image and the timing for recording an image in which a change has occurred are set as needed while observing the image being recorded. For example, the control image is recorded at a rate of 1 frame / second, the image when the change occurs is set to be recorded at the video rate (30 frames / second), and the control is performed while observing the image being recorded. The timing for recording an image and the timing for recording an image in which a change has occurred are manually switched.
{Circle around (2)} The timing for recording an image in which a change has occurred or the recording time resolution (sheets / second) at that time is programmed in advance. For example, image recording is performed in advance by a program such as 1 frame / second for the first 10 seconds, video rate (30 frames / second) for the next 10 seconds, and 1 frame / second for the next 10 seconds. Set the recording time resolution.
[0007]
[Problems to be solved by the invention]
However, in the method (1), when the change occurs slowly, it can be set at any time while the observer observes the image being recorded. For example, the change is faster than the video rate (30 frames / second). Even if the change is large (for example, a change in a specific calcium in a cell), it is often impossible to detect the change with human dynamic vision, and switching of the recording timing during image acquisition is virtually impossible. It becomes possible.
[0008]
Method (2) is very effective when the next change can be predicted, but predicting when and how the cell will change, as in the case of living cells. If it is difficult, it cannot be adopted.
[0009]
In addition, each method has a problem that it is difficult to prevent fading of living cells and fluorescent dyes and to reliably record an image in which a change occurs.
[0010]
The present invention has been made in view of such circumstances, and the problem is that it is possible to minimize optical damage to the specimen, and to obtain an image for a microscope that can reliably record an image in which a change occurs. Is to provide a device.
[0011]
[Means for Solving the Problems]
In order to solve the above-described problem, the microscope image acquisition apparatus according to the first aspect of the present invention is arranged in the optical path of the illumination light from the illumination means of the microscope, and is configured to irradiate the specimen with the illumination light and not to irradiate the sample. A shutter for setting a state, a shutter control circuit for controlling the shutter, an image acquisition unit for acquiring a plurality of images in time series, and an image of the specimen acquired by the image acquisition unit over time. In a microscope image acquisition device comprising a display means and a recording means for recording the image, a predetermined time period based on a change in luminance between images acquired by the image acquisition means before and after the time. a control means for determining the number of images or image acquisition interval to get to the shutter control circuit, characterized in that it is driven on the basis of the number of images or image acquisition interval determined by said control means
[0012]
Since the number of images to be acquired within a predetermined time or the image acquisition interval is set based on the amount of change in luminance between images acquired by the image acquisition unit in succession to the display unit in time, The number of images acquired in time and the image acquisition interval can be automatically set, and light damage to the specimen can be minimized.
[0013]
The image acquisition device for a microscope according to
[0014]
When there is no change in luminance between images, the image acquisition interval is changed, so that a sample can be recorded with an appropriate number of images and image acquisition interval.
