本発明は、皮膚刺激がなく、紫外線による皮膚の炎症抑制効果、メラニン色素の産生抑制効果に優れ、更には皮膚の色素沈着を速やかに淡色化する効果に優れた有用な美白化粧料に関する。

紫外線により皮膚の色調は変化し黒化する。黒化は、メラノサイトにおいて産生され表皮細胞に受け渡されるメラニンの過剰生産が原因であり、このメラニンについては、チロシンが酸化され産生されることが知られている。

従来より、皮膚の黒化、しみやそばかす等の予防や治療を目的とする美白化粧料として、アスコルビン酸およびその誘導体、ハイドロキノン誘導体、コウジ酸等のピロン類、プラセンターエキス等の胎盤抽出物などが配合されたものが提案されている。これらの物質は、メラニン生成の抑制、生成したメラニンの淡色漂白作用の効果などの美白効果を有する物質として開示されている。

しかしながら、これらの物質は、例えばアスコルビン酸およびその誘導体は、保存安定性が不十分であったり、紫外線による炎症防止効果が十分に認められないことが多い。また、ハイドロキノン誘導体は安全性が十分ではないなど問題がある。このようにこれまでの物質ではメラニンの生成抑制効果、メラニンの淡色漂白作用、炎症防止効果、安全性等において総合的に優れる美白化粧料を得ることはなかなか難しいのが実状である。

そこで、本発明の目的は、皮膚刺激がなく、紫外線による皮膚の炎症抑制効果、メラニン色素の産生抑制効果に優れ、更には皮膚の色素沈着を速やかに淡色化する効果に優れた有用な美白化粧料を提供することにある。

本発明者らは、このような実情に鑑み、従来技術の難点を改良せんとして鋭意研究を重ねた結果、下記一般構造式(1)または(2)で示される化合物から選択される少なくとも1種を配合した化粧料は、皮膚刺激がなく、紫外線による皮膚の炎症抑制効果、メラニン色素の産生抑制効果、更には皮膚の色素沈着を速やかに淡色化する効果に優れていることを見出し本発明を完成したものである。

【発明の実施の形態】
以下、本発明の構成を詳細について説明する。

本発明の美白化粧料に用いられるPhPDOとしては、公知の合成方法、例えばアセトンとベンズアルデヒド化合物の縮合によって合成して得られたものが挙げられ、天然物から抽出調製されたもの、市販品なども挙げられる。具体的には1,5−ビス[p−ハイドロキシフェニル]−1,4−ペンタジエン−3−オン、1,5−ビス[o−ハイドロキシフェニル]−1,4−ペンタジエン−3−オン、1,5−ビス[2,4−ジハイドロキシフェニル]−1,4−ペンタジエン−3−オン、1,5−ビス[3−メトキシ−4−ハイドロキシフェニル]−1,4−ペンタジエン−3−オンなどが挙げられる。

そして、本発明に用いられるPhPDOの美白化粧料への配合量としては、総量を基準として0.001〜5.0重量%(以下wt%とする)が好ましい。

以下、実施例及び比較例に基づいて本発明を詳細に説明する。

実施例に記載の(1)紫外線紅斑抑制試験、(2)チロシナーゼ活性阻害試験、(3)皮膚色明度回復試験、(4)美白実用試験、及び(5)光パッチ試験の各試験法は以下の通りである。

(1)紫外線紅斑抑制試験
除毛したハートレー系モルモット10匹の背部皮膚に、UVB領域の紫外線の最小紅斑量の2倍を各2ヶ所ずつ照射する。24時間前と照射直後に試料を塗布し、試料塗布部位とベース塗布部位とを設定して24時間後の紅斑の状態を表1の判定基準に従い評価する。

(2)チロシナーゼ活性阻害試験
マックルベイン緩衝液(pH6.8)1mlに、0.3mg/ml濃度のチロシン溶液と各濃度の試料溶液を加え、37℃にて10分間の予備保温を行う。これに1mg/ml濃度のチロシナーゼ(シグマ社製)0.1mlを加え37℃にて15分間加温した後、分光光度計を用いて、波長475nmにて吸光度(A)を測定した。一方、チロシナーゼの代わりに緩衝液0.1mlを加えたものの吸光度(B)、試料溶液の代わりに緩衝液0.1mlを加えたものの吸光度(C)、更に試料溶液とチロシナーゼの代わりに緩衝液0.2mlを加えたものの吸光度(D)をそれぞれ測定して、下式に従い阻害率(%)を算出した。

(3)皮膚色明度回復試験
被試験者20名の上腕内側部皮膚に、UVA、UVB領域の紫外線の最小紅斑量を3日間連続照射し、照射終了後、試料クリーム塗布部とベースクリーム塗布部皮膚の基準明度(V0 値、V0 ’値)を測定した。引き続いて、1日3回ずつ4週間連続で塗布し、照射開始1、2、4週間後の試料クリーム塗布部とベースクリーム塗布部皮膚の皮膚明度(Vn 値、Vn ’値)を測定して、表2の判定基準により皮膚色の回復評価を行った。
