【従来の技術と発明が解決しようとする課題】
従来の擬似移動層式クロマト分離装置を使用したクロマト分離方法は、内部に充填剤を収容した複数のカラムを直列に連結し、カラムの前端と後端とを流体通路で結合することにより無端状に連結され、内部に液体が一方向に循環している充填床に、分離するべき成分の混合物である原料を含有する原料含有溶液および溶離液を導入し、同時に分離された成分を含有する液と、他の成分を含有する液とを抜き出すことからなり、擬似移動層式クロマト分離装置における前記充填床には、溶離液導入口、吸着されやすい物質を含有する液(エクストラクト;吸着質に富む溶液)の抜き出し口、原料含有溶液導入口、吸着されにくい物質を含有する液(ラフィネート;非吸着質に富む溶液)の抜き出し口が液体の流れ方向に沿ってこの順序で配置され、かつこれらの導入口および抜き出し口は、循環流路内におけるこれらの相対的な位置関係を保持したまま流体の流れ方向に間欠的に逐次移動されるようになっている。

この公報に記載された方法は、得られる分離した成分の濃度を測り、カラムにおける流体の導入口および抜出口の位置を間欠的に移動させる時間を制御することにより純度を維持することを内容とする。

しかしながら、前記公報に記載された方法は、換言すると、濃度パターンの変化を抑制するように流体の導入口および抜出口の位置を間欠的に移動させる時間を制御する方法であるが、時間を制御するだけでは純度を維持することができるものではなく、擬似移動層式クロマト分離装置における吸着工程の非吸着質の吸着、脱着工程における吸着質の脱着を適切に行わなければ、使用する溶剤の低減、得られる成分の純度を制御することはできない。

この発明は前記事情に基づいてなされたものである。すなわち、この発明の目的は、擬似移動層式クロマト分離装置における吸着工程の非吸着質の吸着および脱着工程における吸着質の脱着を適切に行うことのできる擬似移動層式クロマト分離方法および擬似移動層式クロマト分離装置を提供することにある。この発明の目的は、擬似移動層式クロマト分離装置における、溶離液の使用量を低減し、得られる成分の高純度維持を達成することのできる擬似移動層式クロマト分離方法および擬似移動層式クロマト分離装置を提供することにある。

【前記課題を解決するための手段】
前記目的を達成するためにこの発明者が研究したところ、擬似移動層式クロマト分離装置において、ラフィネート抜出口およびエクストラクト抜出口のいずれか一方または両方に、溶質の濃度を検出する検出器を設け、この検出器から出力される電気信号の波形に基づいて、擬似移動層式クロマト分離装置における流体通路への液体の導入口および抜出口を切り替える時間間隔(ステップタイムとも称される。)を調節することによって、溶離液の使用量を低減し、得られる成分の高純度維持を達成することができることを見いだしてこの発明に到達した。

【作用】
この発明における擬似移動層式クロマト分離装置においては、分離用充填剤を収容した複数のカラムが、パイプ等の配管を介して無端状に連結されることにより、循環流体流路が形成されている。この循環流体流路に、流体の流通方向に沿って、光学異性体のラセミ体混合物を含む原料溶液を導入する原料溶液導入口、非吸着質に富む溶液(ラフィネートとも称される。)を抜き出すラフィネート抜き出し口、この循環流体流路に溶離液を導入する溶離液導入口、および吸着質に富む溶液(エクストラクトとも称される。)を抜き出すエクストラクト抜き出し口がこの順に設けられている。

循環流体流路中で一方向に流体を循環させつつ、原料溶液導入口から循環流体流路中に原料を導入すると、原料溶液がカラム中の充填剤と接触し、吸着質成分(吸着の容易な成分であり、強吸着成分とも称される。)が充填剤に吸着され、非吸着質成分(吸着の困難な成分であり、弱吸着成分とも称される。)がラフィネート分として溶離液と共にラフィネート抜き出し口から抜き出される(この工程を吸着工程とも称される)。吸着質成分を吸着した充填剤はエクストラクトの一部と接触し、充填剤上に残存している非吸着質成分が追い出され、吸着質成分が濃縮される(この工程を濃縮工程とも称される。)。濃縮された吸着質成分を含む充填剤は溶離液と接触することにより、吸着質成分が充填剤から追い出され、溶離液を伴ってエクストラクトとしてエクストラクト抜き出し口を介して循環流体流路から抜き出される(この工程を脱着工程とも称される。)。実質的に溶離液のみを吸着した充填剤は、ラフィネートの一部と接触し、充填剤に含まれる溶離液の一部が溶離液回収分として回収される(この工程を溶離液回収工程とも称される。)。

図1に示すように、この発明の実施例装置である擬似移動層式クロマト分離装置1は、第1〜第8単位カラム(単位充填床とも称される。)2a〜2hを有する。第1単位カラム2aの流体出口と第2単位カラム2bの流体入り口、第2単位カラム2bの流体出口と第3単位カラム2cの流体入り口、以後同様にして第7単位カラム2gの流体出口と第8単位カラム2hの流体入口とが流体通路3aにより相互に接続され、第8単位カラム2hの流体出口と第1単位カラム2aの流体入り口とは流体通路3bにより接続されている。また、各単位カラムと次の単位カラムとを結ぶ流体通路3aには逆止弁4がそれぞれ設けられている。この逆止弁4は、単位カラムから次の単位カラムへと流体を導通させるがその逆の流れを阻止する機能を有する。したがって、この実施例においては、逆止弁4は前記機能を有する限り、その構造に特に制限がなく、公知の逆止弁および今後開発される新規の逆止弁を使用することができる。

このラフィネート濃度検出器RFからは、ラフィネートの濃度に対応した電気信号たとえば電圧あるいは電流が出力される。そして、ラフィネート中の溶質の濃度が0であるときに、たとえば図2に示すように、このラフィネート濃度検出器RFからの出力電気信号がベースラインに一致していれば適正な出力信号であると言える。この電気信号は、基本的には、第2ロータリーバルブ9を切り替えた瞬間から次の切り替えまでの時間、すなわち、ステップタイムCの初期においては0であり、抜き出される液中のラフィネートの濃度が上昇するので時間の経過とともに信号強度(たとえば電圧、あるいは電流値)が上昇する。

本発明の方法によると、擬似移動層クロマト分離装置の充填床から抜き出される溶液中の成分の濃度を連続的に監視することにより、吸着工程における非吸着質の吸着および脱着工程における吸着質の脱着を適切に行うことができるようになる。特に、ラフィネート抜き出し口から抜き出されるラフィネート中の溶質を監視することにより吸着工程における非吸着質の吸着を最適状態で効率良く行わせることができるようになる。エクストラクト抜き出し口から抜き出されるエクストラクト中の溶質を監視することにより脱着工程における吸着質の脱着を最適状態で効率良く行わせることができるようになる。
