1. 本出願は、少なくとも1つの下記性質を有する抗4-1BB結合タンパク質の全ヒト由来抗体を提供する。1)本願発明の抗4-1BB結合タンパク質の全ヒト由来抗体は、全長抗体と、「重鎖」のみを含む全く新しい全ヒト抗体とを含み、ヒト4-1BBおよびカニクイザル4-1BBと結合する活性を有し;その中で、当該4-1BB結合タンパク質の重鎖抗体の大きさは従来のIgG抗体の半分しかなく、軽鎖を含まないため、この抗体は二重特異性抗体に用いることができ、そして軽鎖ミスマッチと異種二量化の問題を解決した。ヒトとサル由来の4-1BBタンパク質を結合することができる;2)結合エピトープは、Urelumabと異なる;3)4-1BB信号経路を活性化することができ、免疫細胞におけるIFNγ、IL2および/またはTNFαの分泌を刺激し、腫瘍の成長及び/又は腫瘍細胞の増殖を抑制する;活性はUtomilumabより顕著に強かった。
実施例1.1.モノクローナル抗体の作製
Harbour H2L2マウス(Harbour Antibodies BV)はヒト免疫グロブリン免疫ライブラリを持つトランスジェニックマウスであり、その産生抗体は完全なヒト抗体可変ドメインとラット定常ドメインを有する。Harbour H2L2マウスに対して、可溶性組換えヒト4-1BB-Fc融合タンパク質を用いて多ラウンド免疫を行った。マウス血清中の4-1BB特異的抗体価が特定のレベルに達したことを検出した後、マウスの脾臓細胞を取り出し、骨髄腫細胞系と融合してハイブリドーマ細胞を得た;ハイブリドーマ細胞を多ラウンドスクリーニング及びクローニングした後、抗4-1BBモノクローナル抗体分子を発現する2つのハイブリドーマ細胞株65D4G5G11及び79B10G8D4を単離した。抗体分子可変ドメインをコードするヌクレオチド配列及び対応するアミノ酸配列を、従来のハイブリドーマ配列決定手段を用いて得た。本実施例では、免疫のHarbour H2L2マウスから得られたモノクローナル抗体分子可変ドメインの配列はヒト由来抗体配列である。抗体可変ドメインのCDR配列は、Kabat、Chothia、またはKabat定義とChothia定義を組み合わせたCombined定義規則によって分析することができる。本発明の実施形態におけるCDR配列は、Chothia定義規則に従って区分される。
抗体分子をコードする軽、重鎖可変ドメイン配列を得た後、従来の組換えDNA技術を用いて、軽、重鎖可変ドメイン配列と対応するヒトの抗体軽、重鎖定常ドメイン配列を融合発現し、組換え抗体分子を得ることができる。
抗体の重鎖可変ドメイン配列は、染色体上の重鎖遺伝子群の胚系遺伝子V、D、J遺伝子フラグメントの遺伝子組み換えと体細胞の高周波突然変異などの事象に由来する;軽鎖可変ドメイン配列は、κまたはλ軽鎖遺伝子群の胚系遺伝子V、J遺伝子フラグメントの遺伝子組み換え、体細胞の高周波突然変異などの事象に由来する。遺伝子組み換えと体細胞の高周波突然変異は、抗体の多様性を増加させる主要な要素である。同じ胚系V遺伝子フラグメントに由来する抗体も異なる配列を産生する可能性があるが、全体的に類似性が高い。IMGT/DomainGapAlign(http://imgt.org/3Dstructure-DB/cgi/DomainGapAlign.cgi)またはNCBI/IgBLAST(https://www.ncbi.nlm.nih.gov/igblast/)などの計算方法により、抗体の可変ドメイン配列から遺伝子組み換えが発生した場合に可能な胚系遺伝子フラグメントを推定することができる。実施例1.1で得られた抗体配列を分析し、その重鎖可変ドメイン(VH)と軽鎖可変ドメイン(VL)の胚系遺伝子V遺伝子断片を表1に示す。
D. Zhangらは、ヒトIgGのFcのE345部位に突然変異E345Rを導入することで、アゴニスト型OX40抗体の活性化作用を効果的に増加させることができることを発見した(The Journal of Biological Chemistry、291:27134-27146、2016)。
ヒト4-1BBを発現するCHO-K1細胞とカニクイザル4-1BBを発現するCHO-K1細胞を増幅培養した後、PBSで3回洗浄し、3×105/ウェルで96ウェル板(Corning,#3799)に添加する。次に100μlの測定抗原結合タンパク質または陽性対照抗体UtomilumabまたはUrelumabを添加し、最高濃度200nM、5倍希釈、4°Cで1時間育成する。FACS緩衝液で2回洗浄し、1:1000で希釈されたヒツジ抗ヒトIgGのAF488コンジュゲーション抗体(Life Technologies,#A11013)を加え、4°Cで1時間インキュベートした。FACS緩衝液で2回洗浄し、100μlのPBSで再懸濁し、FACS(BD Biosciences、Canto II)テストを行い、蛍光信号を読み取る。測定結果から相対結合活性の評価根拠として、半数最大結合効果濃度(EC50)を算出した。
ヒト4-1BB結合タンパク質がインビトロでヒト4-1BBとヒト4-1BBLとの結合を遮断する活性を研究するために、ヒト4-1BBを過発現するCHO-K1細胞株(CHO-K1/hu 4-1BB)を用いて、細胞レベルでのヒト4-1BB/ヒト4-1BBL結合遮断実験を行った。簡単に言えば、CHO-K1/hu 4-1BB細胞を消化し、F-12K完全培地で再懸濁し、細胞密度を1×106細胞/mLに調整する。96ウェルVプレート(Corning,#3894)に100μL細胞/ウェルで接種し、次いで100μL/ウェルで最終濃度の2倍の3倍濃度勾配で希釈した測定対象抗原結合タンパク質を添加し、均一に混合し、そのうち抗原結合タンパク質の最高最終濃度は100nMで、合計8濃度があり、hIgG1を対照とした。細胞を4°Cに置き、遮光して1時間インキュベートした。その後、4°Cで5分間遠心分離し、上清を捨て、その後、50μL/ウェルで1μg/mL濃度のビオチン標識ヒト4-1BBLタンパク質(Acro,#41L-H82F9)を添加し、4°Cで遮光して30分間インキュベートした。100μL/ウェルで予冷のPBSを添加し細胞を2回すすぎ、500g、4°Cで5分間遠心分離し、上清を廃棄した。100μL/ウェルで蛍光二次抗体(PE Streptavidin、BD、#554061、1:200)を加え、4°Cで遮光して30分間インキュベートした。200μL/ウェルで予冷のPBSで細胞を2回洗浄し、500g、4°Cで5分間遠心分離し、上清を廃棄した。最後に、200μL/ウェルで予冷のPBSを用いて細胞を再懸濁し、BD FACS CANTOIIを用いて蛍光発光信号値を読み取り、IC50を計算し、抑制率%=1-MFI(4-1BB Ab)/MFI(iso)。
CD32bを発現するCHO-K1細胞CHO-K1/CD32b(Genscript,#M00587)またはCHO-K1(ATCC,#CCL-61)を96ウェルプレート(Perkin Elmer,#6005225)に敷き、細胞量を1.5×104/ウェル、100μL/ウェルである。37°Cで一晩5%CO2環境下でインキュベートした。上清を除去し、40μL/ウェルの2倍の測定抗原結合タンパク質希釈液を加え、初期濃度が200nMであり、5倍濃度希釈で、hlgG1は対照群であった。4.5×104/ウェルの4-1BBおよびNF-Kb反応素子を連続発現できるルシフェラーゼレポーター遺伝子のHEK293レポーター細胞(HEK293/4-1BB/NF-kbレポーター細胞、BPS Biosciences,#79289)40μL/ウェルで添加する。37°Cで5%CO2環境下で6時間培養した。ONE-GloTMルシフェラーゼ試薬(Promega,#E6110)を加え、室温で5分間インキュベートし、マイクロプレートリーダー測定器で発光値を測定した。
実施例1.7.1.4-1BB H2L2抗体が4-1BB経路を体外活性化すること
10μg/mlのマイトマイシン(Ruitaibio、10107409001)を用いてCHO-K1-CD32b(ヒトCD32bを過発現するCHO-K1細胞)の細胞を処理し、37°Cで30分間放置した。その後、10%FBS のF-12K培養液で4回洗浄した。処理した細胞を96ウェルプレートに入れ、1ウェルに1.5×104個であり、37°Cの保温箱を一晩培養した。翌日、MACSキット(Miltenyi Biotec,#130-096-535)を用いてヒトPBMCからヒトCD3陽性T細胞を単離した。まず、細胞数を確認し、その後、細胞数に応じてMACS緩衝液とPan-T細胞ビオチン抗体を添加し、混合し、4°Cで5分間静置した。その後、対応する量のマイクロビーズを加え、4°Cで10分間静置した。LSカラムを通過したのはCD3陽性のT細胞であった。前日の96ウェルプレートの培養液を洗浄し、精製のT細胞を加え、ウェルあたり1×105個である。その後、対応する濃度の4-1BB抗原結合タンパク質または対照抗体Utomilumab、PR000196を添加し、OKT3(eBiosciences、#16-0037-85)を添加し、最終濃度を0.3μg/mlにした。37°C保温箱で72時間培養した。72時間後、上清を回収し、ELISAキット(Invitrogen,#88-7316-88)を用いてIFN-γの含有量を検出した。96平底プレートにコーティング抗体を添加し、4°Cで一晩放置した。翌日、ELISA緩衝液を加え、室温で1時間置く。受け取った上清液を加え、室温で2時間培養した。プレートを2回洗浄し、検出抗体を加え、室温で1時間置く。プレートを2回洗浄し、HRP-ストレプトアビジンを加え、室温で1時間インキュベートした。次にTMB基質を加え、後でELISA停止液(BBI,#E661006-0200)を加えた。マイクロプレートリーダー(Perkin ElemerEnspire)で450nmと570nmの吸光度値を読み取り、OD450-OD570を用いてIFN-γ濃度を計算した。
実施例1.7.1の方法に従って、CHO-K1またはCHO-K1/CD32bの細胞を処理し、プレートを敷き、37°Cの保温箱で一晩培養した。翌日、CD8+およびCD4+T細胞を選別するために、選別試薬(Miltenyi Biotec,#130-096-495,#130-096-533)を用いた。その後、対応する濃度の4-1BB抗体または対照抗体を加え、OKT3(eBiosciences,#16-0037-85)を加え、最終濃度を0.3μg/mlにする。37°C保温箱で72時間培養した。72時間後、上清を回収し、ELISAキット(Invitrogen,#88-7316-88)を用いてIFN-γの含有量を検出した。
B6-h4-1BB遺伝子組換えマウス(Biocytogen Pharmaceuticals (Beijing) Co., Ltd、#11004)MC38皮下結腸癌モデルを構築した。まず、マウス結腸癌細胞MC38(Kerafast,ENH204-FP)の蘇生を行った。培養条件:RPMI-1640培地(Gibco、22400089)+100U/mLペニシリン(AMRESCO(登録商標),#0242-100MU)+100μg/mLストレプトマイシン(AMRESCO(登録商標),#0382)+10%FBS(GIBCO(登録商標)Invitrogen(商標),#10099);37°Cで飽和湿度、5%CO2。対数成長期のMC38細胞(継代回数を記録)を収集し、培養液を除去し、PBSで2回洗浄した後に接種(腫瘍前細胞生存率:98.9%、腫瘍後細胞生存率:92%)、接種量:5×105/100μl/匹(マトリックス接着剤を添加しない)、接種位置:右側皮下。グループ分け基準に従ってマウスを5グループに分け、各グループは6匹で、各グループの平均値は近く、平均値の範囲は80-120mm3で、グループ入り基準:各グループ内の単匹マウス腫瘍の体積はできるだけ150mm3を超えず、かつグループ内のSEMはできるだけ平均値の1/10を超えない。グループ分け当日を0日目と定義し、グループ分け当日としての0日目から投与を開始した。
雌性C57BL/6JマウスにおけるPR000448の薬物動態学を評価した。PR000448とUtomilumabを5mg/kgで静脈内に単回投与し(n=5)、注射前と注射後1日、2日、4日、7日、10日、14日に採血した。採取した血液は直ちに4°Cで15000rpmで15分間遠心分離し、血漿を得て、-20°C以下の冷蔵庫に保存した。ELISA法を用いて血漿中の測定抗体の濃度を測定した。PR000448とUtomilumabの血漿中の濃度変化を図7に示す。得られた血漿中の濃度変化データを薬物動力分析ソフトウェアWinNolinにより分析し、薬物半減期(t1/2)を算出した。tl/2は、最終3点またはソフトウェアによって自動的に設定される最終相血漿中の濃度により計算した。得られた末端t1/2を図7及び下表6に示す。
4-1BB経路に対するFc突然変異抗体PR000980の活性化効果を評価するために、実施例1.6.1の方法に従って、ELISAキット(Invitrogen#88-7316-88、Invitrogen#88-7346-88、Invitrogen#88-7025-77)を用いてIFN-γ、TNF-αとIL-2の含有量を検出した。Utomilumab、IgG1、IgG2、IgG4を対照とした。
ForteBioOctetプラットフォームを用いて、実施例1で得られた4-1BB H2L2抗体(PR000448及びPR000980)及びUtomilumab、Urelumabをエピトープ同定した。ここで、UrelumabはBristol-MyersSquibbの抗4-1BBの抗体BMS-663513(CAS:934823-49-1)であり、実施例1.2の方法に従って発現精製して得た。第一段階に、ヒスチジン標識4-1BB抗原をセンサで捕捉した後、センサを抗体に浸漬し、当該抗体の100%信号を取得する。第二段階に、第一抗体を複数のAHC先端に担持し、次いで緩衝液pH7.5の測定において60秒のベースラインを得た。次いで、抗原結合を可能にするために、先端をヒスチジン標識の4-1BBに180秒間暴露した。先端を測定緩衝液中の第二抗体を含むウェルへに転移し90秒間続け、最終的な信号を当該第二抗体の信号として記録した。抑制率は、下記の式で計算した:抑制率(%)=(A-B)/A*100、A:ある抗体の100%信号(第一段階から得られる)、B:この抗体を第二抗体とする信号(第二段階から得られる)。
実施例1で得られた4-1BB H2L2抗体(PR000448およびPR000980)とUtomilumab、Urelumabを、Biacoreプラットフォームを用いて親和性同定した。試験にはHBS-EP+(10mM HEPES、150mM NaCl、3mM EDTAと0.05%P20、pH7.4、GE Healthcare、BR-1006-69)を運転緩衝液として使用し、シリーズS CM5(GE Healthcare、BR-1005-30)を実験チップとした。流速を10μl/minに設定し、以下の手順によりCM5の4つのチャンネルにProteinAをコンジュゲーションする:1)注入時間を800sに設定し、50mM NHSと200mM EDCを1:1体積比で新鮮に混合した後、4チャンネルに注入する;2)pH4.5の酢酸ナトリウム(GE Healthcare、BR-1003-50)でProteinAを20μg/mlに希釈し、各チャンネルに800s注入する;3)チップ表面の残りの活性カルボキシル基を封止するために、1M pH8.5エタノールアミンを800s注入する。封止後も1×HBS-EP+緩衝液で装置を2時間平衡化させ、ProteinAの最終コンジュゲーション量は約2000RUであった。さらに、多サイクル動力学モードを設定し、抗体とタンパク質の親和性測定を行い、各サイクルには抗体の捕捉、分析物の結合、チップの再生を含む。抗体をすべて1μg/mlに希釈し、10μl/minの流速で2、3、4チャンネルに30s注入し、予めコンジュゲーションされたProteinAによって各抗体を捕捉し、捕捉量は約120RUであった。ヒトの4-1BBまたはカニクイザル4-1BBを順に0nM、0.391nM、0.781nM、1.5625nM、3.125nM、6.25nM、12.5nM、25nMの濃度勾配で4つのチャンネルに注入し、流速が30μl/minであり、PR000448、PR000980に対して解離時間を600sに設定し、Uremulab、Utomilumabに対して解離時間を80sに設定し、注入時間はいずれも120sとする。最後に表面から試験抗体を除去するために、同様の流速で10mMグリシン-塩酸pH1.5(GE Life Sciences、BR-1003-54)を30s注入し、チップを再生した。Biacore T200分析ソフトウェア2.0を用いて実験結果を分析し、1チャネルを参照チャネルとして控除し、分析モデルは1:1動力学フィッティングモデルを選択した。
実施例5の方法に従って、B6-h4-1BBトランスジェニックマウスMC38皮下結腸癌モデルを構築した。平均腫瘍体積が99.14mm3に達した場合、マウスは腫瘍体積に基づいてランダムにグループ化され、各グループが6匹である。グループ分け当日を0日目と定義し、グループ分け当日としての0日目から投与を開始した。測定対象サンプルはPBSで調製し、投与方式は腹腔注射であり、投与量は2mg/kgであった。投与開始後、週に2回体重を計量した。週に2回腫瘍体積を測定し、腫瘍体積(mm3)=0.5×腫瘍長径×腫瘍短径2。
本出願は、1)ヒト及びサル由来の4-1BBタンパク質を結合することができる;2)免疫細胞のIFN-γ、IL2および/またはTNFαの分泌を刺激できる;3)腫瘍の成長及び/又は腫瘍細胞の増殖を抑制することができる;4)4-1BB信号経路を活性化することができる、という特性における一種又は多種を有する抗原結合タンパク質を提供する。本出願はまた、前記抗原結合タンパク質の腫瘍の予防及び治療における使用を提供する。
実施例2.1.全ヒト由来HCAbマウスの免疫と4-1BB抗体の獲得
Harbour HCAbマウス(HarbourAntibodiesBV、WO2002/085945A3)はヒト免疫グロブリン免疫バンクを持つトランスジェニックマウスであり、従来のIgG抗体の半分の大きさしかなく、全く新しい「重鎖」のみからなる抗体を産生することができる。その産生抗体はヒトの抗体「重鎖」可変ドメインとマウスFc定常ドメインのみを有する。軽鎖を含まないという特徴のため、この抗体は軽鎖ミスマッチと異種二量化の問題をほとんど解決し、この技術プラットフォームは従来の抗体プラットフォームでは実現しにくい製品を開発することができる。
6~8週齢のHarbourヒト由来抗体トランスジェニックマウスは、2群の免疫スキームを用いてHarbour HCAbマウスに多ラウンド免疫を行った。免疫スキーム1は、組換えヒト4-1BB-ECD-Fc(ChemPartner、Shanghai)抗原タンパク質で免疫する。各マウスが免疫するたびに、皮下で鼠径部注射または腹腔内注射によって受ける総注射量は100マイクロリットルである。1回目の免疫において、各マウスは50マイクログラムの抗原タンパク質と完全Freundアジュバント(Sigma,#F5881)とを体積比1:1で混合して作製した免疫原試薬によって免疫を受けた。その後の各ラウンドの免疫増強において、各マウスは25マイクログラムの抗原タンパク質とRibiアジュバント(SigmaAdjuvantSystem、Sigma、#S6322)とを混合して作製した免疫原試薬によって免疫を受けた。免疫スキーム2は、ヒト4-1BBを過発現するNIH3T3-h4-1BB(ChemPartner、Shanghai)を用いて細胞系を安定化させて免疫する。各マウスは免疫するたびに2×106細胞懸濁液を腹腔内注射する。1ラウンド当たりの免疫増強間隔は少なくとも2週間であり、通常は5ラウンドを超えない。免疫時間は0、14、28、42、56、70日であり、そして49、77日目にマウス血清抗体価を測定した。HCAbマウス脾臓B細胞分離を行う5日前に、各マウス25マイクログラムの抗原タンパク質の用量で最後の免疫増強を行った。
マウスの血液を採取し、血液を10倍希釈して6つの濃度(1:100、1:1000、1:10000、1:100000、1:1000000)を採取し、ヒト4-1BB-ECD-Fc(Chempartner、Shanghai)を被覆したELISAプレートでELISA検出を行い、マウスの血液中の抗ヒト4-1BBの抗体価を確定し、そして、フローサイトメトリーにより2つの濃度のマウス血液(1:100、1:1000)の、4-1BB高発現CHO-K1/h4-1BB細胞(Chempartner、Shanghai)とCHO-K1母細胞に対する特異反応性を測定した。空白対照群(PB)は免疫前マウスの血清であった。マウス血清中の4-1BB特異的抗体価が特定のレベルに達したことを検出した後、マウスの脾臓細胞を取り出してB細胞を分離し、BD FACS AriaIICellSorterを用いてCD138陽性の漿細胞とヒト4-1BB抗原陽性のB細胞群を選別した。B細胞のRNAを抽出し、cDNA(SuperScript IV First-Strand synthesis system、Invitrogen、18091200)を逆転写し、次いで特異的プライマーPCRを用いてヒトVH遺伝子を増幅した。PCRプライマー5’-GGTCCCAGTGTSAGAGAGTGTG-3’、5’-AATCCCTGGCACTGAGACTGACC-3’。増幅されたVH遺伝子断片をヒトIgG1抗体重鎖Fcドメイン配列をコードする哺乳動物細胞発現プラスミドpCAGベクターに構築した。
HCAb抗体をコードするプラスミドを哺乳動物宿主細胞(例えばヒト胚腎細胞HEK293)にトランスフェクションし、従来の組換えタンパク質発現と精製技術を用いて、精製された抗4-1BB組換え重鎖抗体を得ることができる。具体的には、HEK293細胞をFreeStyle(商標) F17 Expression Medium培地(Thermo,A1383504)で培養した。瞬時トランスフェクション開始前に細胞濃度を6×105細胞/mlに調整し、37°C、8%CO2ロッカ中で24時間培養し、細胞濃度は1.2×106細胞/mlであった。培養した細胞を30ml準備し、前記HCAb重鎖をコードするプラスミド30μgを1.5ml Opti-MEM血清減少培地(Thermo,#31985088)に溶解し、さらに1.5ml Opti-MEMを1mg/ml PEI(Polysciences,Inc,#23966-2)120μlに溶解し、5分間静置した。PEIをゆっくりとプラスミドに加え、室温で10分間インキュベートし、培養瓶を揺らしながらプラスミドPEI混合溶液をゆっくり滴下し、37°C、8%CO2ロッキングベッドで5日間培養した。5日後に細胞生存率を観測した。培養物を収集し、3300Gの回転速度で10分間遠心分離した後、上清を採取した。その後、上清を高速遠心分離して不純物を除去した。PBS(pH7.4)でMabSelect(商標)(GE Healthcare Life Science,#71-5020-91AE)を含む重力カラム(Bio-Rad,#7311550)を平衡化させ、カラム体積の2-5倍で洗浄した。上清サンプルをカラムに通す。カラム体積の5~10倍のPBSでカラムを洗い流する。目的のタンパク質をpH3.5の0.1Mグリシンで溶出し、pH8.0のTris-HClで中性に調整し、最後に限外ろ過管(Millipore、UFC901024)でPBS緩衝液に濃縮し、精製した抗4-1BB重鎖抗体溶液を得た。抗体濃度はNanoDropで280nm吸光度を測定して得て、抗体の純度はSEC-HPLCとSDS-PAGEで測定して得た。
タンパク質サンプルの純度および多量体の形態を分析するために、分析型分子サイズ排除クロマトグラフィー(SEC)を用いた。分析型カラムTSKgel G3000SWxl(Tosoh Bioscience、08541、5μm、7.8mmx30cm)を高圧液体クロマトグラフィー(HPLC)(型番:Agilent Technologies、Agilent 1260 Infinity II)に接続し、PBS緩衝液で室温で少なくとも1時間平衡させた。適量のタンパク質サンプル(少なくとも10μg、サンプル濃度を1mg/mlに調整)を0.22μm濾過膜で濾過した後、システムに注入し、HPLCプログラムを設定する:PBS(pH7.4)緩衝液でサンプルを1.0ml/minの流速でカラムを流し、最長時間は20分;検出波長が280nmである。採取後、ChemStationソフトウェアを用いてクロマトグラムを積分し、関連データを計算し、分析報告書を生成し、サンプル内の異なる分子サイズ成分の滞留時間を報告した。
分析型疎水相互作用クロマトグラフィー(HIC)を用いて、タンパク質サンプルの純度と疎水性を分析した。分析型カラムTSKge1 Buty1-NPR(Tosoh Bioscience,14947,4.6mm×3.5cm)高圧液体クロマトグラフィー(HPLC)(型番:Agilent Technologies、Agilent 1260 Infinity II)に接続し、PBS緩衝液を用いて室温で少なくとも1時間平衡させる。設定方法は、16分間以内に100%移動相A(20mMヒスチジン、1.8M硫酸アンモニウム、pH6.0)から100%移動相B(20mMヒスチジン、pH6.0)までの線形勾配であった、流速を0.7ml/minに設定し、タンパク質サンプル濃度が1mg/mlで、注入体積が20μlで、測定波長が280nmである。採取後、ChemStationソフトウェアを用いてクロマトグラムを積分し、関連データを計算し、分析報告書を生成し、サンプル内の異なる分子サイズ成分の滞留時間を報告した。
示差走査蛍光法(Differential Scanning Fluorimetry、DSF)は、タンパク質の熱安定性を測定するための一般的な高フラックス法である。リアルタイム蛍光定量PCR装置を用いて、折り畳み解除されたタンパク質分子と結合した染料の蛍光強度の変化を監視することにより、タンパク質の変性の過程を反映し、タンパク質分子の熱安定性を反映する。本実施例はDSF法を用いてタンパク質分子の熱変性温度(Tm)を測定した。10μgのタンパク質を96ウェルPCRプレート(Thermo、AB-0700/W)に添加し、次いで2μl 100X希釈の染料SYPROTM(Invitrogen、2008138)を添加し、次いで最終体積が40μl/ウェルとなるように緩衝液を添加した。PCRプレートを密封し、リアルタイム蛍光定量PCR装置(Bio-Rad CFX96 PCR System)に置き、25°Cで5分間インキュベートした後、0.2°C/0.2分の勾配で25°Cから95°Cに徐々に昇温し、試験終了時に温度を25°Cに下げた。FRETスキャンモードを使用し、Bio-Rad CFX Maestroソフトウェアを使用してデータ分析を行い、サンプルのTmを算出した。
本実施例は、抗ヒト4-1BBのHCAbモノクローナル抗体がヒトとカニクイザル4-1BBとインビトロで結合する活性を研究するためである。ヒト4-1BBを過発現するCHO-K1細胞株(CHO-K1/hu 4-1BB、Genescript)、カニクイザル4-1BBを過発現するCHO-K1細胞株(CHO-K1/cyno4-1BB、Genescript)を用いて細胞レベルでの抗体結合実験を行った。簡単に言えば、CHO-K1/hu 4-1BB細胞とCHO-K1/cyno4-1BB細胞を消化し、F12K完全培地で再懸濁し、細胞密度をそれぞれ1×106細胞/mlに調整した。96ウェルVプレート(Corning,#3894)に100μL細胞/ウェルで接種し、次いで100μl/ウェルで添加し、最終濃度の2倍の3倍濃度勾配で希釈した測定対象抗体を添加した。細胞を4°Cに置き、遮光して1時間インキュベートした。その後、100μl/ウェルで予冷のPBSを添加し細胞を2回すすぎ、500g、4°Cで5分間遠心分離し、上清を廃棄した。さらに100μl/ウェルで蛍光二次抗体(Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG,Fcγ Fragment Specific、Jackson、#109-545-06、1:500希釈)を添加し、4°Cで遮光し30分間インキュベートした。細胞を100μl/ウェルで予冷のPBSで2回洗浄し、500g、4°Cで5分間遠心分離し、上清を廃棄した。最後に、200μl/ウェルで予冷のPBSを用いて細胞を再懸濁し、BD FACS CANTOIIを用いて蛍光発光信号値を読み取った。
CD32bを発現するCHO-K1細胞(CHO-K1/CD32b)を96ウェルプレート(Perkin Elmer,#6005225)に敷き、細胞量が1.5×104/ウェルで、100μL/ウェルである。37°Cで5%CO2環境下で一晩インキュベートした。上清を除去し、40μL/ウェルで2倍の測定抗原結合タンパク質希釈液を添加し、初期濃度が200nMで、3倍濃度希釈または初期濃度が30nMで、5倍濃度希釈で、hlgG1が対照群である。4.5×104/ウェルで4-1BBおよびNF-Kb反応素子を連続発現可能なルシフェラーゼレポーター遺伝子のHEK293レポーター細胞(HEK293/4-1BB/NF-kbレポーター細胞、BPS Biosciences,#79289)を40μL/ウェルで添加した。37°Cで5%CO2環境下で6時間培養した。ONE-GloTMルシフェラーゼ試薬(Promega,#E6110)を加え、室温で5分間インキュベートし、マイクロプレートリーダー測定器で発光値を測定した。
ヒト4-1BB結合タンパク質がヒト4-1BBとヒト4-1BBLとの結合をインビトロで遮断する活性を研究するために、ヒト4-1BBを過発現するCHO-K1細胞株(CHO-K1/hu 4-1BB)を用いて細胞レベルでのヒト4-1BB/ヒト4-1BBL結合遮断実験を行った。簡単に言えば、CHO-K1/hu 4-1BB細胞を消化し、F-12K完全培地で再懸濁し、細胞密度を1×106細胞/mLに調整した。96ウェルVプレート(Corning,#3894)に100μL細胞/ウェルで接種し、続いて100μL/ウェルで、最終濃度の2倍の3倍濃度勾配で希釈した測定抗原結合タンパク質を添加し、均一に混合し、そのうち抗原結合タンパク質の最高最終濃度は100nMで、8つの濃度があり、hIgG1を対照とした。細胞を4°Cに置き、遮光して1時間インキュベートした。その後、4°Cで5分間遠心分離し、上清を捨て、その後、50μL/ウェルで、1μg/mL濃度のビオチン標識ヒト4-1BBLタンパク質(ACRO、41L-H82F9)を添加し、4°Cで遮光して30分間インキュベートした。100μL/ウェルで予冷のPBSを添加し細胞を2回すすぎ、500g、4°Cで5分間遠心分離し、上清を廃棄した。100μL/ウェルで蛍光二次抗体(PEStreptavidin,BD,#554061,1:200)を添加し、4°Cで遮光して30分間インキュベートした。200μL/ウェルで予冷のPBSで細胞を2回洗浄し、500g、4°Cで5分間遠心分離し、上清を廃棄した。最後に、200μL/ウェルで予冷のPBSを用いて細胞を再懸濁し、BD FACS CANTOIIを用いて蛍光発光信号値を読み取り、IC50を計算し、抑制率%=1-MFI(4-1BBAb)/MFI(iso)。
10μg/mlのマイトマイシン(北京中生瑞泰科技、#10107409001)を用いてCHO-K1(ATCC、#CCL-61)またはCHO-K1/CD32b(ヒトCD32bを過発現するCHO-K1細胞)の細胞を37°Cで30分処理した。その後、10%FBSのF-12K培養液で4回洗浄した。処理した細胞を96ウェルプレートに入れ、1ウェルあたり1.5×104個とし、37°Cの保温箱を一晩培養した。翌日、MACSキット(Miltenyi Biotec,#130-096-535)を用いてヒトPBMCからヒトCD3陽性T細胞を単離した。まず細胞数を確認し、その後、細胞数に応じてMACS緩衝液とPan-T細胞ビオチン抗体を添加し、混合し、4°Cで5分間静置した。その後、対応する量のマイクロビーズを加え、4°Cで10分間静置した。LSカラムを通過したのはCD3陽性のT細胞であった。前日の96ウェルプレートの培養液を洗浄し、精製T細胞をウェルあたり1×105個で添加した。その後、対応する濃度の4-1BB抗体または対照抗体を加え、OKT3(eBiosciences,#16-0037-85)を加え、最終濃度を0.3μg/mlにする。37°C保温箱で72時間培養した。72時間後、上清を回収し、ELISAキット(Invitrogen,#88-7316-88)を用いてIFN-γの含有量を検出した。96平底プレートにコーティング抗体を添加し、4°Cで一晩放置した。翌日、ELISA緩衝液を加え、室温で1時間置く。受け取った上清液を加え、室温で2時間培養した。プレートを2回洗浄し、検出抗体を加え、室温で1時間置く。プレートを2回洗浄し、HRP-ストレプトアビジンを加え、室温で1時間インキュベートした。次にTMB基質を加え、後でELISA停止液(BBI、E661006-0200)を加えた。マイクロプレートリーダー(Perkin ElemerEnspire)は450nm吸光度(OD450)値を読み取り、IFN-γ濃度を算出した。
メーカーが提供する詳細な操作と方法に従って、OctetRED96機器(Fortiebio)と抗ヒトIgGFc アビジンセンサ(AHCセンサ、PallForteBio、#18-5060)を使用して親和性を測定した。具体的には、0.1%(w/w)BSAと0.02%(v/v)TWEEN20を含有するPBS緩衝液(pH7.4)を用いてヒトの4-1BBタンパク質、Hisタグ(Acrobiosystem,#41B-H5227)またはカニクイザルの4-1BBタンパク質、hisタグ(Acrobiosystem,#41B-C52H4)を400nMに希釈し、AHCセンサとインキュベートした。40nMの4-1BB抗体を、ヒト4-1BBタンパク質またはサル4-1BBタンパク質を持つAHCセンサと30°Cで3分間インキュベートした。反応混合物を0.1%(v/w)BSAと0.02%(v/v)TWEEN20を含むPBS緩衝液(pH7.4)中で30°Cで5分間インキュベートした。OctetRed96は、4-1BB抗体と4-1BBタンパク質の結合及び分離信号をリアルタイムで記録する。親和性、相関、解離定数はOctet使用ソフトウェアによって確認され、結果は表13と表14に示されている。
ForteBio Octetプラットフォームを用いて、実施例1で得られた抗原結合タンパク質とutomilab、urelumabをエピトープ同定した。簡単に言えば、第一抗体を複数のAHC先端に担持し、次いで緩衝液pH7.5の測定において60秒ベースラインを得た。次いで、抗原結合を可能にするために、ヒスチジン標識の4-1BBに先端を180秒間暴露した。測定緩衝液中の第二抗体を含むウェルに先端を90秒間移した。第二抗体が明らかな結合を示す場合、それは非競合剤(すなわち、第一抗体とは異なるエピトープ区間)とみなされる。第二抗体が明らかな結合を示さない場合、それは競合剤(すなわち、第一抗体と同じエピトープ区間)とみなされる。結合アッセイは、第一抗体の存在下での第二抗体の4-1BBとの結合と第一抗体遮断そのものとの比較によって行った。抑制率は式により計算し、抑制率(%)=(A-B)/A*100(注:A:ある抗体の100%信号、B:この抗体を第二抗体の信号とする)。
4-1BBがTNF腫瘍壊死因子受容体スーパーファミリーに属し、このファミリーは、免疫と非免疫細胞機能を媒介する多機能受容体の大きなクラスから構成されている。CD40、OX40、41BB、CD27、GITR、CD30を含む6種類の受容体が重要な役割を果たす免疫共通刺激者であることが同定された。同様に、誘導型T細胞共刺激因子(ICOS)は、活性化されたT細胞または記憶型T細胞の機能および生存に重要な役割を果たす別の受容体である。
本発明の積極的な進歩効果は、下記の通りである。
本発明の4-1BB抗体は、「重鎖」のみを含む全く新しい全ヒト抗体であり、ヒト4-1BB及びカニクイザル4-1BBと特異的に結合する活性を有する。この4-1BB重鎖抗体の大きさは、従来のIgG抗体の半分しかなく、軽鎖を含まないため、この抗体は二重特異性抗体に用いることができ、軽鎖ミスマッチと異種二量化の問題を解決した。
ヒト表皮成長因子受容体2(HER2)は、ERBB2、HER-2、HER-2/neu、NEU、NGL、TKR1およびc-erbB2とも呼ばれ、ラットではErbB2またはneuとも呼ばれ、ErbBタンパク質ファミリーの一員であり、通常は表皮成長因子受容体ファミリーと呼ばれる。乳癌において侵襲性の高いタンパク質である。HER2は、細胞膜表面に結合したチロシンキナーゼであり、通常は細胞の成長と分化をもたらす信号伝達経路に関与する。HER2は孤児受容体とされ、EGFリガンドファミリーによって活性化することはできない。約30%の乳癌は、HER2遺伝子増幅またはそのタンパク質産物の過発現を有し、この受容体の乳癌における過発現は、疾患の再発と予後不良と関連している。HER2は発育、癌、神経筋接続部の通信及び細胞成長と分化の調節に役割を果たす。
本実施例は、対応するアミノ酸配列がIMGTデータベースに由来する抗HER2のIgG抗体trastuzumabを使用した。製造された抗体番号はPR000210であった。

本実施例は、4-1BBの二重特異性抗体がSK-BR-3とインビトロで結合する活性を研究するためである。SK-BR-3はHER2を高発現する乳癌細胞であり、SK-BR-3を用いて細胞レベルでの抗体結合実験を行った。簡単に言えば、SK-BR-3細胞を消化し、完全培地で再懸濁し、細胞密度をそれぞれ1×106細胞/mlに調整する。96ウェルVプレート(Corning,#3894)に100μL細胞/ウェルで接種した後、最終濃度の2倍の3倍濃度勾配で希釈した測定対象抗体を100μl/ウェルで添加した。細胞を4°Cに置き、遮光して1時間インキュベートした。その後、100μl/ウェルで予冷のPBSを添加し細胞を2回すすぎ、500g、4°Cで5分間遠心分離し、上清を廃棄した。さらに100μl/ウェルで蛍光二次抗体(Alexa Fluor(登録商標)647 Affini Pure Goat Anti-Human IgG,F(ab’)2 fragment specific,Jackson Immunoreserach,#109-605-006、1:1000希釈)を添加し、4°Cで遮光し30分間インキュベートした。細胞を100μl/ウェルで予冷のPBSで2回洗浄し、500g、4°Cで5分間遠心分離し、上清を廃棄した。最後に、200μl/ウェルで予冷のPBSを用いて細胞を再懸濁し、BD FACS CANTOIIを用いて蛍光発光信号値を読み取った。

本実施例は、4-1BBの二重特異性抗体が4-1BBとインビトロで結合する活性を研究するためである。カニクイザル4-1BBを発現するCHO-K1細胞株(CHO-K1/cyno4-1BB、Genescript)を用いて細胞レベルでの抗体結合実験を行った。簡単に言えば、細胞CHO-K1/cyno4-1BB細胞を消化し、F12K完全培地で再懸濁し、細胞密度をそれぞれ1×106細胞/mlに調整した。96ウェルVプレート(Corning,#3894)に100μL細胞/ウェルで接種し、次いで100μl/ウェルで添加し、最終濃度の2倍の3倍濃度勾配で希釈した測定対象抗体を添加した。細胞を4°Cに置き、遮光して1時間インキュベートした。その後、100μl/ウェルで予冷のPBSを添加し細胞を2回すすぎ、500g、4°Cで5分間遠心分離し、上清を廃棄した。さらに100μl/ウェルで蛍光二次抗体(Alexa Fluor(登録商標)647 Affini Pure Goat Anti-Human IgG,F(ab’)2 fragment specific,JacksonImmunoreserach,#109-605-006、1:1000希釈)を添加し、4°Cで遮光し30分間インキュベートした。細胞を100μl/ウェルで予冷のPBSで2回洗浄し、500g、4°Cで5分間遠心分離し、上清を廃棄した。最後に、200μl/ウェルで予冷のPBSを用いて細胞を再懸濁し、NovoCyteフローサイトメータ(ACEABiosciences)を用いて蛍光発光信号値を読み出した。
1mg/mlのOKT3(eBiosciences,#16-0037-85)をPBSで希釈し、ウェルあたり100μLの0.08μg/mlのOKT3を取り96ウェルプレート(Corning,#3599)を包み、最終濃度を10μg/mlとした。4°Cで一晩被覆した。翌日、SK-BR-3細胞を消化し、完全培地で再懸濁し、細胞密度をそれぞれ4×105細胞/mlに調整し予備とする。MACSキット(Miltenyi Biotec,#130-096-535)を用いてヒトPBMCからヒトCD3陽性T細胞を単離した。まず細胞数を確認し、その後、細胞数に応じてMACS緩衝液とPan-T細胞ビオチン抗体を添加し、混合し、4°Cで5分間静置した。その後、対応する量のマイクロビーズを加え、4°Cで10分間静置した。LSカラムを通過したのはCD3陽性のT細胞であった。前日の96ウェルのプレートを被覆したOKT3を洗い流し、50μL精製T細胞を加え、ウェル当たり1×105個とした。さらに50μLのSK-BR-3細胞を96ウェルプレートに入れ、ウェルあたり2×104個とした。次いで、対応する濃度のHER2/4-1BBの二重特異性抗体または対照モノクローナル抗体を加え、37°Cで5%CO2培養箱で培養した。72時間培養後に上清を採取し、ELISAキット(Invitrogen,#88-7316)を用いてIFN-γの含有量を検出した。メーカーの説明書に従ってELISA検査を行い、簡単に言えば、96平底プレートにコーティング抗体を加え、4°Cで一晩過ごした。翌日、ELISA緩衝液を加え、室温で1時間置く。受け取った上清液を加え、室温で2時間培養した。プレートを2回洗浄し、検出抗体を加え、室温で1時間置く。プレートを2回洗浄し、HRP-ストレプトアビジンを加え、室温で1時間インキュベートした。次にTMB基質を添加し、10~30分でELISA停止液(BBI life sciences,#E661006-0200)を添加した。ボードリーダー(Molecular Devices,#SpectraMaxPlus)を使用してOD450-570値を読み込みます。Graphad8.0を用いて値を分析し、図を作成した。
本実施例は、抗HER2のIgG抗体の抗原結合ドメインFabと抗4-1BBのHCAb抗体の抗原結合ドメインVHを用いて、IgG-VH四価対称構造のHER2×4-1BB二重抗体分子を構築した。同時に、抗HER2のIgG抗体の抗原結合ドメインFabと抗4-1BBのH2L2抗体を用いて、IgG-scFv四価対称構造の二重特異性抗体を構築した。
プログラム死受容体1(programmed death 1、PD-1)は主にT細胞などの免疫細胞に発現し、それは2つのリガンド、すなわちプログラム死リガンド-1(programmed death ligand 1、PD-L1)とPD-L2を有する。PD-L1は主に抗原提示細胞及び複数の腫瘍細胞に発現される。PD-L1とPD-1の相互作用はT細胞の活性を低下させ、サイトカインの分泌を弱め、免疫抑制作用を果たす。多くのヒト腫瘍組織においてPD-L1タンパク質の発現が検出され、腫瘍部位のミクロ環境は腫瘍細胞上のPD-L1の発現を誘導でき、発現されたPD-L1は腫瘍の発生と成長に有利であり、抗腫瘍T細胞のアポトーシスを誘導し、さらに腫瘍細胞を保護して免疫攻撃から逃れる。
本実施例は、対応するアミノ酸配列がIMGTデータベースに由来する抗PD-L1のIgG抗体PR000151(Atesolizumab類似体)を使用する。
本実施例は、抗PD-L1のIgG抗体PR000265またはPR000151(atezolizumab類似体)の抗原結合ドメインFabと、抗4-1BBのIgG抗体PR000197またはPR000448の抗原結合ドメインFabを用いて、FIT-Ig構造を有する抗PD-L1×4-1BBの二重特異性抗体分子を構築する。FIT-Ig構造の設計は、特許WO2015/103072A1を参照することができ、構造は図23Aに示されている。
本実施例は、抗PD-L1のIgG抗体PR000265の抗原結合ドメインFabと、抗4-1BBのHCAb抗体PR001758、PR001760またはPR001836の抗原結合ドメインVHとを用いて、多種構造の抗PD-L1×4-1BBの二重特異性抗体分子を構築する。

図26A及び図26Bに示すように、本実施例の二重抗体分子はカニクイザル4-1BBと結合することができることに対して、Urelumabはできない。PR004270がカニクイザル4-1BBを結合する能力は、MFI最大値においてUtomilumabよりやや優れている。
本実施例は、標的細胞におけるPD-L1×4-1BB二重抗体分子の存在は4-1BBを結合することによりT細胞の活性を活性化することを研究するためである。標的細胞は、ヒトPD-L1を高発現するCHO-K1/hPDL1(GENSCRIPT、M00543)、またはヒトPD-L1を高発現するMDA-MB-231(ATCC、HTB-26)などの、PD-L1を異なる程度に発現する細胞であってもよい。エフェクター細胞は、単離されたヒトPBMCまたはT細胞であってもよい。
本実施例において、陽性対照分子は抗4-1BBのモノクローナル抗体Urelumabである。
図27A-図27Dに示すように、標的細胞CHO-K1/hPDL1とT細胞を混合するシステムにおいて、架橋に依存しない抗4-1BBモノクローナル抗体Urelumabは、T細胞のIFN-γ放出を活性化することができ、一方、架橋に依存する抗4-1BBモノクローナル抗体(PR000448、PR001758、PR001760、PR001836)はT細胞をほとんど活性化できない。FIT-Ig構造の二重抗体分子(PR003052、PR000701)とIgG-VH四価対称構造の二重抗体分子(PR003549、PR003550、PR003551、PR004268、PR007130、PR007132、PR007133、PR007135、PR007136、PR007137、PR007138、PR007139、PR007141、PR007142、PR007143、PR007145、PR007146、PR007149)とFab-HCAb構造の二重抗体分子(PR004270)はいずれもT細胞を活性化させ、サイトカインを放出することができる。これは、二重抗体分子のT細胞に対する活性化が標的細胞の特異的活性化に依存することを示している。また、IgG-VH四価対称構造の二重抗体分子(PR003549、PR003550、PR007130、PR007132、PR007133、PR007135、PR007136、PR007137、PR007138、PR007139、PR007141、PR007142、PR007143、PR007145、PR007146、PR007149)とFab-HCAb構造の二重抗体分子(PR004270)はFIT-Ig構造の二重抗体分子(PR003052、PR000701)と比べて、より高いサイトカイン放出レベルを引き起こすことができ、より強いT細胞活性化能力を示し、Urelumabより優れている。
図27Eに示すように、標的細胞MDA-MB-231とT細胞を混合するシステムにおいて、IgG-VH四価対称構造の二重抗体分子(PR003549)とFab-HCAb構造の二重抗体分子(PR004270)は類似のT細胞活性化能力を有し、FIT-Ig構造の二重抗体分子(PR003052、PR000701)より優れた。
本実施例は、混合リンパ球反応(MLR)を用いてT細胞に対するPD-L1×4-1BB二重抗体分子の活性化作用を研究する。
本実施例は、マウス体内におけるFab-HCAb対称構造を有するPD-L1×4-1BB二重抗体分子PR004270の薬物動態学特性を研究した。
本実施例は、抗PD-L1のIgG抗体の抗原結合ドメインFabと抗4-1BBのHCAb抗体の抗原結合ドメインVHを用いて、多種類の構造の抗PD-L1×4-1BBの二重特異性抗体分子を構築した。HCAbに基づいて二重特異性抗体分子構造を構築するフレキシビリティを示し、異なる構造タイプ、相対位置、結合価数などのパラメータによってT細胞を活性化する機能活性を調節した。

- 請求項11に記載の単離の核酸を含む、発現ベクター。
- 請求項12に記載の発現ベクターを含み;好ましく、原核細胞または真核細胞である、宿主細胞。
- 請求項13に記載の宿主細胞を培養し、培養物から4-1BB結合タンパク質を獲得することを含む、4-1BB結合タンパク質の作製方法。
- 請求項1-6のいずれか一項に記載の4-1BB結合タンパク質または請求項7-10のいずれか一項に記載の二重特異性抗体を含む、キメラ抗原受容体。
- 細胞毒性剤、及び請求項1-6のいずれか一項に記載の4-1BB結合タンパク質または請求項7-10のいずれか一項に記載の二重特異性抗体を含み;好ましく、前記細胞毒性剤はMMAFまたはMMAEを含む、抗体薬物複合物。
- 請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体および/または請求項16に記載の抗体薬物複合物を含む、薬物組成物。
- 請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体および請求項17に記載の薬物組成物の、癌を治療および/または予防する薬物の作製における用途であって、
前記癌は好ましくHER2、PD-L1及び4-1BBにおける一種または複数種に関連の癌、例えば乳癌、メラノーマ、肺癌、胃癌、肝臓癌、食道癌、子宮頸癌、頭頸部腫瘍または結腸直腸癌である、用途。 - 請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体、請求項15前記キメラ抗原受容体、請求項16に記載の抗体薬物複合物および/または請求項17に記載の薬物組成物を含み;
好ましく、さらに(i)抗体またはその抗原結合フラグメントまたは抗体薬物複合物または薬物組成物を投与するデバイス;および/または(ii)使用説明を含む、試薬キット。 - 薬カセットAおよび薬カセットBを含み、その中で:
前記薬カセットAは請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体、請求項15に記載のキメラ抗原受容体、請求項16に記載の抗体薬物複合物および/または請求項17に記載の薬物組成物を含み;
前記薬カセットBは他の抗腫瘍抗体または前記他の抗腫瘍抗体を含む薬物組成物、および/またはホルモン製剤、標的小分子製剤、プロテアーゼ体阻害剤、イメージング剤、診断剤、化学療法剤、溶腫薬物、細胞毒性剤、サイトカイン、共刺激分子の活性化剤、抑制性分子の阻害剤及びワクチンからなる群における一種または複数種を含む、薬キット。 - 4-1BBに媒介される疾患または障害を診断、治療および/または予防する方法であって、
前記方法は必要な患者に治療有効量の請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体、請求項15に記載のキメラ抗原受容体、請求項16に記載の抗体薬物複合物または請求項17に記載の薬物組成物を投与し、または請求項20に記載の薬キットを用いて必要な患者を治療する、方法。 - 前記疾患または障害は腫瘍であり、好ましく4-1BB陽性腫瘍であり、より好ましく胃癌、食道癌、肺癌、卵巣癌、黒色腫、腎癌、乳癌、結腸直腸癌、肝臓癌、膵臓癌、膀胱癌、頭頸癌、気管支癌、神経膠腫および/または白血病である、請求項21に記載の方法。
- 4-1BBを免疫検出または測定する方法であって、
請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体、請求項15に記載のキメラ抗原受容体、請求項16に記載の抗体薬物複合物または請求項17に記載の薬物組成物を使用することを含み;好ましく、前記検出は診断および/または治療目的のものではない、方法。 - それぞれ必要な患者に請求項1-6のいずれか一項に記載の4-1BB結合タンパク質、請求項7-10のいずれか一項に記載の二重特異性抗体、請求項15に記載のキメラ抗原受容体、請求項16に記載の抗体薬物複合物または請求項17に記載の薬物組成物、および第二治療剤を投与し;前記第二治療剤は、好ましく他の抗腫瘍抗体または前記他の抗腫瘍抗体を含む薬物組成物、および/またはホルモン製剤、標的小分子製剤、プロテアーゼ体阻害剤、イメージング剤、診断剤、化学療法剤、溶腫薬物、細胞毒性剤、サイトカイン、共刺激分子の活性化剤、抑制性分子の阻害剤及びワクチンからなる群における一種または複数種を含む、併用療法。
