Abstract
Description
The present invention relates to an external skin preparation containing loxoprofen that has excellent analgesic and antiinflammatory effects. More specifically, the present invention relates to a topical preparation that has significantly improved transdermal absorbability compared to conventional loxoprofen topical preparations and their formulation techniques by incorporating a specific active ingredient into loxoprofen.
Loxoprofen, which is a propionic acid nonsteroidal antipyretic, analgesic, and antiinflammatory drug (NSAIDs), has antipyretic, analgesic, and antiinflammatory effects based on the inhibitory effect on prostaglandin biosynthesis, like other NSAIDs. Furthermore, after oral administration, loxoprofen is absorbed from the gastrointestinal tract as an unchanged form with a weak gastric mucosal stimulating effect, and is a prodrug that becomes active in the body, so it has the characteristic that it causes less gastric mucosal damage than the active form. It is also known (for example, see Non-Patent Document 1).
In recent years, loxoprofen has been commercially available as a topical anti-inflammatory analgesic in the form of poultices, tapes, and gels, and is used clinically (for example, see Non-Patent Document 2). It is known that loxoprofen is converted into a trans-OH form (active form) by ketone reductase even in the skin (see, for example, Patent Document 1).
Chlorpheniramine maleate is used as an antihistamine in topical preparations to treat urticaria and pruritus associated with skin diseases (eczema/dermatitis, pruritus, drug eruption). In addition to being used as a blood circulation promoting ingredient in external preparations, l-menthol is also used as a refreshing agent and fragrance in pharmaceutical additives. Glycyrrhetinic acid is used as an anti-inflammatory ingredient in external preparations. Nonanoic acid vanillylamide is used as a thermal stimulation ingredient and a blood circulation improving ingredient in external preparations. Benzyl nicotinic acid ester is used as a blood circulation improving ingredient in external preparations, and as a solubilizing agent in pharmaceutical excipients. Vitamin E is used as a blood circulation improving ingredient in external preparations, and as a stabilizer and antioxidant in pharmaceutical additives.
So far, in topical preparations containing loxoprofen, there have been topical gel preparations that incorporate carboxyvinyl polymer to increase transdermal absorption of loxoprofen (see Patent Document 2), and formulations that incorporate propylene glycol and propylene glycol fatty acid esters as additives. It has been disclosed that transdermal absorption of loxoprofen was increased by doing so (see Patent Document 3). In addition, polyhydric alcohols such as propylene glycol, butylene glycol, and glycerin are often added to enhance the usability of liquid preparations for external use, but these polyhydric alcohols are used as transdermal absorption enhancers of medicinal ingredients. It is known that this has been done (see Patent Document 4).
Furthermore, it is known that antihistamines enhance the skin permeability of nonsteroidal anti-inflammatory analgesics and enhance the pharmacological effects of the main agent (see Patent Document 5). Further, in a formulation in which loxoprofen and isopropanol are blended to improve storage stability, a liquid formulation is also known in which glycyrrhetinic acid, nonanoic acid vanillylamide, chlorpheniramine maleate, etc. are further added (Patent Document 6).
However, when one or more ingredients selected from glycyrrhetinic acid, nicotinic acid benzyl ester, nonanoic acid vanillylamide, vitamin E, etc. are added to a topical preparation containing loxoprofen and l-menthol, the skin permeability of loxoprofen sodium increases. is not known to decrease. Furthermore, it is not known at all how chlorpheniramine maleate in such external preparations affects the transdermal absorption of loxoprofen.
An object of the present invention is to find an ingredient that further enhances the transdermal absorption of loxoprofen in a skin external preparation (combined preparation) containing loxoprofen and other active ingredients.
The present inventors have developed glycyrrhetinic acid, nicotine, etc. for the purpose of imparting or enhancing anti-inflammatory effects, thermal stimulation effects, and blood circulation promoting effects in skin external preparations (compound preparations) containing loxoprofen, l-menthol, and ethanol. In order to develop an external preparation further containing ingredients such as acid benzyl ester, nonanoic acid vanillylamide, and vitamin E, the percutaneous absorption of external preparations containing these active ingredients was investigated. As a result, by adding one or more selected from glycyrrhetinic acid, nonanoic acid vanillylamide, benzyl nicotinic acid ester, and vitamin E to a skin topical preparation containing loxoprofen, l-menthol, and ethanol, loxoprofen can be administered to the skin easily. It was found that skin absorbability decreased. The inventors have also discovered that the percutaneous absorption of loxoprofen can be significantly improved by further incorporating chlorpheniramine maleate into these external preparations, and have completed the present invention.

Comparison of Samples 10 and 12 in Table 6 shows that transdermal absorption of loxoprofen is reduced by adding glycyrrhetinic acid and nonanoic acid vanillylamide to a topical preparation (combined preparation) containing loxoprofen, l-menthol, and ethanol. I found out what to do. On the other hand, a comparison of Sample 10, Sample 11, and Sample 12 revealed that in the topical preparation containing loxoprofen, l-menthol, ethanol, glycyrrhetinic acid, and nonanoic acid vanillylamide, the reduction was observed when chlorpheniramine maleate was further blended. It was found that the transdermal absorption of loxoprofen was restored.
Furthermore, by comparing Samples 10 and 14, transdermal absorption of loxoprofen was reduced by further blending glycyrrhetinic acid and nicotinic acid benzyl ester in external preparations (combined preparations) containing loxoprofen, l-menthol, and ethanol. I found out what to do. On the other hand, by comparing Samples 13 and 14, it was found that in a topical preparation containing loxoprofen, l-menthol, ethanol, glycyrrhetinic acid, and benzyl nicotinic acid ester, the amount of loxoprofen decreased by further blending chlorpheniramine maleate. It was found that percutaneous absorption was restored.
The topical skin preparation containing loxoprofen of the present invention has an excellent feeling of use, suppresses the decrease in transdermal absorption of loxoprofen due to the ingredients, maintains anti-inflammatory and analgesic effects, has a thermal stimulation effect, a blood circulation promoting effect, and an antihistamine effect. It has enhanced or added effects and is extremely useful.
