本発明の分野は、一般的に、1つ以上のGlycoTag(糖タグ)を含む修飾キャリアタンパク質、及びかかる修飾キャリアタンパク質の、例えばPgILを用いた、効率的なO-結合型グリコシル化における使用に関する。

オリゴサッカリルトランスフェラーゼ(OST又はOTase)は、オリゴ糖を脂質キャリアから新生タンパク質へと転移させる膜埋め込み酵素(membrane-embedded enzyme)である(連続的作用によりオリゴ糖を構築する細胞質中のグリコシルトランスフェラーゼとは異なり、OTaseは、一括してグリカンをタンパク質に転移させる[2])。O-結合型グリコシル化は、糖分子(グリカン)の、タンパク質標的(例えば、ピリン)中のアミノ酸残基(例えば、セリン又はスレオニン)の側鎖ヒドロキシ基への共有結合からなる。例えば、ナイセリア・メニンギティディス(Neisseria meningitidis)由来のピリン-グリコシル化遺伝子L(PglL)タンパク質は、O-結合型グリコシル化に関与するOTaseである。グラム陰性細菌のペリプラズムにおいて、PglLは、グリカンをUnd-PP-グリカンからピリンタンパク質へと転移させる([1])。PglB(N-グリコシル化)とは異なり、PglLは、活性のために、還元末端のC-2位における2-アセトアミド基又は最初の2つの糖間のβ 1,4結合を必要とせず、このため実質的にあらゆるグリカンを転移させることができる(大腸菌細胞及びサルモネラ(Salmonella)細胞の両方において、ナイセリア・メニンギティディス(Neisseria meningitidis)PglLは、例えば、C.ジェジュニ(C. jejuni)七糖、大腸菌O7抗原、大腸菌K30莢膜構造、S.エンテリカ(S. enterica)O抗原、及び大腸菌O16ペプチドグリカンサブユニットを、ピリンに転移させる)([1]、[3]、[14]、[16])。従って、NmPglL及びそのホモログ、例えばナイセリア・ゴノレア(Neisseria gonorrhoeae)由来のPglL(「PglO」と呼ばれる、[6]及び[9])及び緑膿菌(シュードモナス・エルギノーサ(Pseudomonas aeruginosa))由来のPilO([15])は、基質「無差別性(promiscuous)」である(すなわち、これらは緩い基質特異性を有し、このため、多様なオリゴ糖及び多糖を転移させることができる)。[1]及び[14]([3]及び[16]を通して)。ナイセリア・メニンギティディス(Neisseria meningitidis)PglL(NmPglL)ホモログは、本明細書中に記載されており(実施例を参照のこと)、当技術分野で公知である:([17]、[28]、[18])。

本明細書中で用いられる「GlycoTag」は、組換えのO結合型グリコシル化部位であり、ピリンアミノ酸配列のフラグメントからなる。このように、用語「GlycoTag」は、組換えアミノ酸配列(すなわち、野生型ピリンから分離されたアミノ酸配列)を指すために用いられるのに対して、「シークオン」は、野生型ピリン内に位置する(すなわち、野生型ピリンから分離されていない)同じ配列を指すために使用することができる。

コンジュゲーション
本明細書に示されるとおり、修飾キャリアタンパク質は、バイオコンジュゲーションのために使用することができる。特定の実施形態において、修飾キャリアタンパク質は、グラム陰性細菌宿主細胞内のin vivoバイオコンジュゲーションのために使用することができる。特定の実施形態において、修飾キャリアタンパク質は、修飾キャリアタンパク質を、ナイセリア属の(Neisserial)PglL及びPglLグリカン基質と共に、場合により好適なバッファー中でインキュベートすることによるコンジュゲート製造のために使用することができる。
特定の実施形態において、O-グリコシル化修飾キャリアタンパク質は、in vivoの方法及び系を用いて製造される。特定の実施形態において、O-グリコシル化修飾キャリアタンパク質(又はバイオコンジュゲート)が作製され、その後宿主細胞のペリプラズムから単離される。本発明のin vivoコンジュゲーション(「バイオコンジュゲーション」)は、グラム陰性細菌細胞中の組換えタンパク質発現及び上記細胞からの単離のための公知の方法論を利用し、この方法論は、配列の選択及び最適化、ベクター設計、プラスミドのクローニング、培養パラメータ、及びペリプラズム精製技術を含む。例えば、[65]、[3]、[5]、[7]、[8]、[9]、[10]、[11]、[1]、[14]、[4]、[63]、及び[31]を参照のこと。宿主細胞を用いたバイオコンジュゲートの製造方法は、例えば、[66]及び[67]に記載されている。バイオコンジュゲーションは、バイオコンジュゲーションが製造のために必要とする化学薬品が少なく、生成される最終製品についてより一貫性があるという点において、in vitroの化学的コンジュゲーションを上回る利点を提供する。
O-グリコシル化修飾キャリアタンパク質をin vitroで製造するため、PglL OTaseを、修飾キャリアタンパク質及びPglLグリカン基質と共に、例えばバッファー中でインキュベートすることができる。特定の実施形態において、上記バッファー(bugger)は、約6~約8のpHを有する。一態様において、バッファーは、リン酸バッファー生理食塩水であり得る。別の態様において、バッファーは、7.5のpHを有するTris-HCl 50mMであり得る。
本発明のO-グリコシル化修飾キャリアタンパク質は、多数の疾患の治療のための治療剤として使用することが可能であり、この場合、有効量のO-グリコシル化修飾キャリアタンパク質が、かかる治療を必要とする対象に投与される。また本発明のO-グリコシル化修飾キャリアタンパク質は、有効量のO-グリコシル化修飾キャリアタンパク質が、かかる治療を必要とする対象に投与される場合、疾患の予防のためのワクチンとして又は免疫原性組成物中で使用することもできる。従って、多数の異なるO-グリコシル化修飾キャリア(crrier)タンパク質を製造するための本明細書中に記載される方法は、ワクチン学において極めて有用であることがわかるであろう。
本発明のグリカン及びコンジュゲートを分析するために様々な方法を使用することが可能であり、例えば、SDS-PAGE又はキャピラリーゲル電気泳動などが挙げられる。O抗原ポリマーの長さは、直線状に構築される反復単位の数によって規定される。これは、典型的な梯子様パターンが、グリカンを構成する様々な反復単位数の結果であることを意味する。従って、SDS PAGE(又はサイズで分離する他の技術)において互いに隣り合う2つのバンドは、単一の反復単位のみで異なる。これらの個別の差は、グリカンのサイズについて糖タンパク質を分析する場合に利用される:異なるポリマー鎖長を有する非グリコシル化キャリアタンパク質とバイオコンジュゲートは、それらの電気泳動移動度に従って分離する。バイオコンジュゲート上に存在する第1の検出可能な反復単位数(n1)及び平均反復単位数(naverage)が測定される。これらのパラメーターを用いて、例えば、バッチ間の一貫性又は多糖安定性を実証することができる。
グリコシル化部位利用は、例えば、糖ペプチドLC-MS/MSにより定量することができる:コンジュゲートをプロテアーゼ(1つ又は複数)で消化して、ペプチドを好適なクロマトグラフィー法(C18、親水性相互作用HPLC HILIC、GlycoSepNカラム、SE HPLC、AE HPLC)で分離し、MS/MSを用いて異なるペプチドを同定する。この方法は、その前の化学的方法(スミス分解(smith degradation))又は酵素的方法による糖鎖短縮を伴って使用することもできるし、又は上記の糖鎖短縮を伴わずに使用することもできる。215~280nmのUV検出を用いた糖ペプチドピークの定量は、グリコシル化部位利用の相対的測定を可能とする。別の実施形態において、サイズ排除HPLCにより:より高いグリコシル化部位利用は、SE HPLCカラムからのより早い溶出時間により反映される。
修飾キャリアタンパク質を含む組成物が提供される。特定の実施形態において、修飾キャリアタンパク質は、O-グリコシル化されている。特定の実施形態において、修飾キャリアタンパク質に機能的に連結されたグリカンは免疫原性であり、従って上記組成物は免疫原性組成物である。
アジュバントは、抗原に対する免疫応答又は抗体応答を増強及び調節するために、免疫原性組成物及びワクチン組成物において使用される非抗原成分である。アジュバントが抗原の免疫学的効果の誘導、大きさ、及び/又は寿命を増強することは、十分に認識されている。アジュバントは、化合物が単独で投与される場合、抗原に対する免疫応答又は抗体応答を生じない化合物である。

多糖生合成クラスター(O抗原又は莢膜多糖)の染色体コピーを含む、O抗原リポ多糖リガーゼ遺伝子waaLが欠損した大腸菌(大腸菌W3110 ΔwaaL、ΔwecA-wzzE、ΔO16::P.シゲロイデス(P. shigelloides)O17(S.ソンネイ(S. sonnei))のwbgT-wbgZクラスター(本明細書中以降「大腸菌(E. coli)W3110ΔwaaL」))、並びに、PglL及び修飾キャリアタンパク質を発現する2つのプラスミドを使用した。単一コロニーを、50ml TBdev培地(酵母抽出物24g/L、大豆ペプトン12g/L、グリセロール100% 4.6mL/L、K2HPO4 12.5g/L、KH2PO4 2.3g/L、MgCl2x6H2O 2.03g/L)に播種し、30℃でOD 0.8まで増殖させた。この時点で、誘導物質として0.1mM IPTG及び0.1%アラビノースを添加した。この培養液をさらに一晩(o/n)インキュベートし、さらなる分析のために回収した([00119]を参照)。バイオリアクター評価の場合、50mLの(非誘導)一晩(o/n)培養液を使用して、11の培養液を21個のバイオリアクター中に播種した。このバイオリアクターを500~1000rpmで撹拌し、2M KOH又は20%H3PO4のいずれかの自動制御添加によりpHを7.2に維持し、培養温度を30℃に設定した。溶存酸素(pO2)のレベルを10%酸素に維持した。バッチ段階において、細胞を、50g/Lのグリセロールを含有することを除いて上記のとおりのTBdev培地中で増殖させた。供給培養液として、250g/Lグリセロール及び0.1%IPTG(1プラスミド系)又は0.1%IPTG及び2.5%アラビノース(2プラスミド系)を添加したTBdevを使用した。増殖のフェドバッチ段階を開始する前に、0.1 mM IPTGによる誘導(1プラスミド系)、並びに0.1mM IPTG及び0.1%アラビノースによる誘導(2プラスミド系)をOD=35で行った。線形供給速度を24時間維持し、その後16時間の飢餓期間を設けた。バイオリアクター培養液を合計約40時間の培養後に回収し、このとき、この培養液はOD600=±80に達していたはずであった。
緑膿菌外毒素A(EPA)キャリアタンパク質(配列番号1)を、ナイセリア・メニンギティディス(Neisseria meningitidis)ピリンPilE(配列番号137として提供される野生型配列)由来の1つ以上のGlycoTagを組み込むように修飾した(方法については、[29]、[6]、[4]、及び[31]を参照のこと。これらは全て、その全体が参照により本明細書中に組み込まれる)。組換えEPA(rEPA、配列番号1)を修飾して、3つの他の組換えEPAタンパク質を作製した:
- N末端にNmPilE GlycoTag配列番号140(配列番号137の残基45~73に対応する)(29アミノ酸長)を組み込むように修飾された第1の組換えEPAタンパク質 (rEPA1、配列番号51)。

ナイセリア・メニンギティディス(Neisseria meningitidis)PglL(NmPglL)(配列番号8のポリヌクレオチド配列、配列番号9のアミノ酸配列をコードする)、シゲラ・ソンネイ(Shigella sonnei)O抗原遺伝子クラスター(配列番号6のポリヌクレオチド配列、配列番号208~216のアミノ酸配列をコードする)、並びにキャリアタンパク質rEPA1、rEPA2、及びrEPA3(DsbAペリプラズムシグナル配列(配列番号5、配列番号4をコードする)に機能的に連結されている)のうちの1つを、O抗原リポ多糖リガーゼ遺伝子waaLが欠損した大腸菌W3110中に導入した(大腸菌W3110ΔwaaL)。rEPA1~rEPA3のそれぞれについて1つの、3個の細胞ロットを作製した。クーマシーブルー染色及びウェスタンブロットアッセイで、NmPglLが、脂質キャリア結合S.ソンネイ(S. sonnei)O抗原を、rEPA1、rEPA2、及びrEPA3のそれぞれに効率的に転移させたことを確認した(図2において、それぞれ#1~#4に対応する)。転移は、1プラスミド系(すなわち、PglLとrEPAキャリアが1つのプラスミド中で組み合わされている(図2A及び図2Bの#1~#3))、又は2プラスミド系(すなわち、PglLとEPAが2つの別個のプラスミド上にコードされている、IPTG及びアラビノース誘導性(図2A及び図2Bの#4))のいずれにおいても観察された。質量分析では、S.ソンネイ(S. sonnei)O抗原が、rEPA1に結合される場合に、インタクトであり且つその構造が維持されることを確認した。21~25個の反復単位(以下)がrEPA1に結合していると決定された。
rEPA1-S.ソンネイ(S. sonnei)O抗原バイオコンジュゲートの免疫原性を評価するため、4匹の雌のニュージーランド白ウサギ(3~4月齢)を2群に分け(1群当たり2匹のウサギ)、0日目、7日目、10日目及び18日目に、2μgの糖、40μgのタンパク質、及び非フロイントアジュバント(群1)、又は10μgの糖、200μgのタンパク質、及び非フロイントアジュバント(群2)を含むバイオコンジュゲート組成物を皮下注射した。0日目、21日目、及び28日目に放血させた。28日目に採取した血液サンプルのウェスタンブロットは、全ての対象においてS.ソンネイ(S. sonnei)O抗原及びEPAに対する抗体が生成され(図4)、2μg用量(図4A)が、10μg用量(図4B)よりも優れた抗体応答を誘導することを明らかにした。これらの結果は、NmPglL媒介性rEPA1-S.ソンネイ(S. sonnei)O抗原バイオコンジュゲートが、ウサギにおいて免疫原性であることを示す。
キャリアタンパク質汎用性
キャリアタンパク質上で複数のGlycoTagを使用することの技術的実現可能性を評価するため、EPAを、1コピー又は2コピーのいずれかの配列番号9の配列を有するNmPilE GlycoTagを組み込むように修飾した。1コピーのGlycoTagのみを組み込むEPAについては、このGlycoTagはN末端に位置していた(rEPA1)。2コピーのGlycoTag配列番号140を組み込むEPAについては、第1のGlycoTagはN末端に位置し、第2のGlycoTagはC末端に位置していた(rEPA43、配列番号135)。ナイセリア・メニンギティディス(Neisseria meningitidis)PglL(NmPglL)を、3つの異なる脂質キャリア結合多糖:S.ソンネイ(S. sonnei)O抗原、S.フレックスネリ(S. flexneri)2a CPS、又はストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)12F CPSのうちの1つの存在下で、rEPA1又はrEPA43に適用した。NmPglLは、S.ソンネイ(S. sonnei)O抗原、S.フレックスネリ(S. flexneri)2a CPS、及びストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)12F CPSのそれぞれを、rEPA1及びrEPA43上に転移させた(図5)。
NmPglLの基質汎用性を評価するため、血清型Sp8、Sp12F、Sp14、Sp22A、Sp23A、及びSp33Fのそれぞれのうちの1つに由来するニューモコッカル(Pneumococcal)莢膜多糖(CPS)をコードする多糖遺伝子クラスター(すなわち、ヌクレオチド配列)を、大腸菌(E. coli)W3110ΔwaaLに染色体導入した(表1)。またNmPglL、及びrEPA1又はrEPA43ヌクレオチド配列(上記の実施例1)も、大腸菌W3110ΔwaaL細胞のそれぞれに機能的に導入した。12個の組換え宿主細胞を作製し、そのうち6個は6つの異なるニューモコッカル(Pneumococcal)CPSのそれぞれのうちの1つとrEPA1を組み込んでおり、別の6個は6つの異なるニューモコッカル(Pneumococcal)CPSのそれぞれのうちの1つとrEPA43を組み込んでいた。このようにして、NmPglLは、rEPA1又はrEPA43の存在下で各脂質キャリア結合ニューモコッカル(Pneumococcal)CPSペプチドグリカンに接触し、NmPglLは、in vivoで、ニューモコッカル(Pneumococcal)CPSグリカンをrEPA1又はrEPA43上に転移させた:
ナイセリア・メニンギティディス(Neisseria meningitidis)PglLホモログの同定及び特性決定
それぞれ異なるナイセリア(Neisseria)種に由来する20個のナイセリア(Neisseria)PglLタンパク質を同定した。確立された方法を用いて、最初に、各PglLを、それらがS.ソンネイ(S. sonnei)O抗原(配列番号208~216のタンパク質をコードする、wbgT、wbgU、wzx、wxy、wbgV、wbgW、wbgX、wbgY、及びwbgZ遺伝子からなるオペロンにより作製され、これらが、N-アセチル-アルトルロン酸-α1,3-4-アミノ-N-アセチル-フコサミンの還元末端構造を有する糖を作製する)7を、NmPglL(対照)の効率と少なくとも同等の(すなわち、これと等しいか又はこれを上回る)効率で、内在性ピリン上に転移させる能力についてスクリーニングした。これにより6個のナイセリア・メニンギティディス(Neisseria meningitidis)PglLホモログを同定した。その後、この6個のナイセリア(Neisseria)PglLタンパク質を、それぞれ、それらがS.ソンネイ(S. sonnei)O抗原を、NmPglL(対照)と少なくとも同等の効率でrEPA1上に転移させる能力についてスクリーニングした。これにより4個のナイセリア・メニンギティディス(Neisseria meningitidis)PglLホモログを同定した。方法については、[4]、[6]、[29]、及び[31](これらは全て、その全体が参照により本明細書中に組み込まれる)を参照のこと。結果は、以下の表2にまとめられるとおりであった:

- N末端に配列番号163のNsPilin GlycoTag(配列番号162の残基53~83に対応;31アミノ酸長)を組み込むように修飾された第6のEPA (rEPA31、配列番号111)。
ナイセリア・ゴノレア(Neisseria gonnorrhoeae)のGlycoTag(1つ又は複数)を含む設計されたキャリアタンパク質
修飾EPAキャリアタンパク質(各タンパク質は、1コピー又は2コピーのナイセリア・ゴノレア(Neisseria gonorrhoeae)のPilin GlycoTag配列を組み込む)を設計し、製造した。内部EPA残基R274、S408、及び/又はA519(配列番号1について番号付けされた)が、配列番号145又は配列番号146(それぞれ、配列番号143のNgPilin配列の残基52~81に対応する30アミノ酸配列、及び配列番号143のNgPilin配列の残基 62~81に対応する20アミノ酸配列)の配列を有するNgPilin GlycoTagに置換された(以下の表4)。
