Abstract
Description
この出願は、2018年5月25日に提出された中国発明特許出願201810511368.2「マンノースによって修飾された標的ナノ製剤」の優先権を主張し、該優先権発明特許は、引用によって本書に組み込まれる。
本発明は医薬品の分野に関し、具体的にはマンノースによって修飾された標的ナノ製剤、ナノ製剤を調製するための組成物、標的化要素、標的担体、および調製された標的薬剤およびその調製方法と適用に関する。
前記標的物質と前記スペーサー材料を標的化要素に合成する。
ナノ製剤基礎材料をナノ製剤に調製し、ステップAによって得られた標的化要素を前記ナノ薬物送達担体と接続して標的担体を得るか、先に前記スペーサー材料とナノ製剤基礎材料を合成し、そして前記標的物質をさらに接続して標的担体を得る。
既存のナノ製剤の欠点に対し、マンノースおよびその誘導体によって修飾された標的化性を有するナノ製剤を設計する。
マンノース−PEGn−Chol:マンノース−PEG100−Chol合成を例にとると、この合成経路に従って、長さの異なる他のPEGも合成できる。ジエチレングリコール、p−トルエンスルホニルクロリドとトリエチルアミンを取り、DCMに溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して生成物TosOC2H4OC2H4OH(化合物1)を得る。化合物1ペンタアセチル化マンノース和三フッ化ホウ素エーテル(BF3・Et2O)を取り、DCMに溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して生成物Aco−Mannose−OC2H4OC2H4OTos (化合物2)を得る。化合物2とアジ化ナトリウムを取り、DMFに溶解し、60℃で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Aco−Mannose−OC2H4OC2H4N3(化合物3)を得る。化合物3をメトキシドナトリウムのメタノール溶液に溶解させ、室温で3h反応させ、濃縮して生成物Mannose−OC2H4OC2H4N3(化合物4)を得る。コレステロール、ブロモプロピンおよび水素ナトリウムを取ってエーテルとDMFの混合溶液に溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Chol−CH2CCH(化合物5)を得る。化合物4、化合物5およびヨウ化第一銅を取ってDMFに溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Mannose−Chol(化合物6)を得る、合成経路は反応式Iに示す(マンノース−PEG1000−Cholの合成経路は反応式IIに示す)。添付図1の各特徴的水素プロトンの化学シフトδ(ppm)を分析する:5.30−5.42 ppm(1個)それぞれコレステロール中=CH−水素のシフト、7.92ppm(1個)はN−CH=C中の水素のシフトである、これらの特徴的なピークの存在は、化合物4が化合物5のプラグメントに接続されていることを示し、1H−NMR核磁気結果によると、マンノース−PEG100−Cholが正常に結合されていることがわかる。
処方の組成
固体脂質ナノ粒子は、脂質を骨格材料として調製されたナノ構造の担体であり、生理学的適合性、細胞親和性、および標的化性の特徴を有する。この実施例において、マンノースによって修飾された脂質を加えることにより、固体脂質ナノ粒子の標的化性を改善する。
HDLは、組成、密度、粒子径が不均一なリポタンパク質の1種である。 HDLで最も豊富なアポリポタンパク質はApoA−Iであり、その疎水性の内部コアは体内でコレステロールを輸送する。組換え高密度リポタンパク質は、内因的に単離された、またはインビトロで合成されたApoA−Iをリン脂質、コレステロールなどと体外で再結合することによって形成され、生化学的特性および機能において内因性HDLに類似している。高密度リポタンパク質には、高い安全性、高効率の運搬能力(コアの埋め込み、親油性薬物の埋め込み、および組換え高密度リポタンパク質コアの3つの方法で運搬できる。表面嵌入、 リン脂質単層への親油性薬物の挿入、タンパク質結合および)、標的化性などの優れた特性を持つ。この実施例において、調製されたHDL Naine粒子にマンノースリガンドを添加して、ナノ粒子の標的化特性をさらに改善する。
処方の組成
トランスフェクションの12時間前に、平皿中のRaw264.7細胞を吹き分け、培養液を再懸濁し、カウントし、1mLの細胞懸濁液を10×104個/mLの密度で24ウェルプレートの各ウェルに挿入し、培養箱の中で約12時間培養しておく。培地を廃棄し、最終濃度100ng/mLのリポ多糖(LPS)を各ウェルに加え、37℃で約12時間培養して、マクロファージを刺激する。培地を廃棄し、最終濃度25ng/mLのさまざまな製剤(処方1と処方3)を各ウェルに加え、37℃の暗所で約2時間培養し、マクロファージによるさまざまな製剤の取り込みを観察する。培養時間になると、培地を廃棄し、冷たいPBSで2回洗浄する。PBSで24ウェルプレート中の細胞を収集し、PBSで細胞(1200rpm、5min)を1回洗浄し、最後にフローサイトメーターに入れて結果を分析する。結果を図5に示す。左から順に、Control、処方3、処方1である。これにより、標的リガンドマンノース−PEG1000−Cholを添加した後、細胞のトランスフェクション効率を大幅に向上させることができるとわかる。
実施例7に記載のナノ製剤基礎材料をナノ薬物送達担体に調製し、次に実施例7に記載の標的物質を調製された前記ナノ薬物送達担体の表面に付着させて標的担体を得る。
実施例7の標的物質とスペーサー材料を合成し、ここではマンノースとPEGnとの合成を例に取る。
実施例9の標的化要素とナノ製剤基礎材料をさらに合成し、ここで、マンノース、PEGnとコレステロールの合成を例に取る。
実施例8の標的担体を改善する方法:A.実施例7の標的物質とスペーサー材料を標的化要素に合成する、B.実施例7のナノ製剤基礎材料をナノ製剤に調製し、次にステップAで得られた標的化要素を実施例8のナノ薬物送達担体に接続する。
マンノース−PEGn−Chol:マンノース−PEG100−Cholの合成を例に取る、異なる長さの他のPEGも、この合成経路に従って合成される。ジエチレングリコール、p−トルエンスルホニルクロリドおよびトリエチルアミンを取り、DCMに溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して生成物TosOC2H4OC2H4OH(式I)を得る。式Iの化合物ペンタアセチル化マンノースと三フッ化ホウ素エーテル(BF3・Et2O)を取り、DCMに溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して生成物Aco−Mannose−OC2H4OC2H4OTos (式II)を得る。式IIの化合物とアジ化ナトリウムを取り、DMFに溶解し、60℃で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Aco−Mannose−OC2H4OC2H4N3(式III)を得る。式IIIの化合物をメトキシドナトリウムのメタノール溶液に溶解し、室温で3h反応させ、濃縮して生成物Mannose−OC2H4OC2H4N3(式IV)を得る。コレステロール、ブロモプロピンおよび水素ナトリウムをエーテルとDMFの混合溶液に溶解し、室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Chol−CH2CCH(式V)を得る。式IVの化合物、式Vの化合物とヨウ化第一銅をDMFに溶解し,室温で24h反応させ、シリカゲルクロマトグラフィーによって分離して固体生成物Mannose−Chol(式VI)を得る。図1の各特徴的な水素プロトンの化学シフトδ(ppm)を分析しする:5.30−5.42ppm(1個)はそれぞれコレステロール中=CH−水素のシフトである、7.92ppm(1個)はN−CH=C中の水素のシフトであり、これらの特徴的なピークの存在は、化合物式IVが化合物式Vのフラグメントに接続されていることを示し、1H−NMR核磁気の結果によると、図1に示すように、マンノース−PEG100−Cholが正常に結合されていることがわかりる。合成経路は次のとおりである。
薬物を実施例8で調製された標的担体にカプセル化される。
調製方法:1:上記の処方に従って異なる重量比のリン脂質を秤量し(主薬がPTXの場合、リン脂質と一緒に添加する)、丸底フラスコに入れ、クロロホルム/エタノール=1:1(v/v)を添加して溶解し、減圧下で回転蒸発により有機溶媒を除去し、フラスコの内壁に薄膜を形成し、PBS7.4緩衝液を加えて薄膜を水和させ、100W超音波で3min処理し、リポソームを得る。pGFPをカプセル化する場合、ブランクリポソームとpGFPを室温で培養して得られる。マンノース−PEG1000−Cholから得られたナノ粒子は、MP1000−LPXとして記録できる。得られたリポソームの特性を評価し、リポソームの粒子径が132.93±4.93nm、zeta電位が37.93±2.95mVであることを示す。また、リポソームが明らかな脂質膜構造を有し、形状が球形に近いことが観察された(図2−4)。
固体脂質ナノ粒子は、脂質を骨格材料として調製されたナノ構造の担体であり、生理学的適合性、細胞親和性、および標的化性の特徴を有する。この実施例において、マンノースによって修飾された脂質を加えると、固体脂質ナノ粒子の標的化性を改善する。

DC2.4細胞と調製されたナノ標的製剤を24ウェルプレートで共培養し、ナノ標的製剤のN/P比を5にする。フローサイトメトリーを使用して、GFP陽性DC2.4細胞のトランスフェクション効率と平均蛍光強度(MFI)を検出する。つまり、DC2.4細胞は、前方散乱(FSC)と側方散乱(SSC)によってキャプチャされる。生き残ったDC2.4細胞は領域1(R1)に示され、GFP陽性細胞は領域2(R2)に示される。トランスフェクション効率はR2に自動的に表示される。FlowJoソフトウェアを使用して、GFP陽性細胞でGFPを発現するMFIを取得する。MFIを計算するには、未処理のDC2.4細胞のバックグラウンド値を差し引く必要がある。主薬mRNAのinvitroトランスフェクション動態をさらに理解するために、DC2.4細胞でのマンノース−PEG1000−Cholのトランスフェクション効率も12−72hで確認された。
マンノース−PEG1000−Cholによって修飾された標的製剤の安定性を研究し、その安定性を粒子径、zeta電位、およびトランスフェクション効率によって決定される。その結果、マンノース−PEG1000−Cholによって修飾されたナノ製剤を4℃で保存した場合、マンノース−PEG1000−Cholによって修飾されたナノ製剤の粒子径はわずかに減少したが、zeta電位は低下しなかった(図8)、4℃で3日間保存した場合、トランスフェクション効率は約50%である(図9)、具体的には、下表をご参照ください。また、血清中では、マンノース−PEG1000−Cholで培養したmRNAは解離していなかった(図10)。同様に、マンノース−PEG100−Cholおよびマンノース−PEG2000−Cholによって修飾されたナノ標的製剤も良好な安定性を示している。
指定された処方で24時間培養した後、フローサイトメトリーを使用して細胞毒性分析を行った。その結果を図11−12に示す。対照群、マンノース−PEG1000−Chol群およびLipo 3K群の生細胞率は、それぞれ86.7±3.6%、86.7±1.7%および90.1±1.2%でした。アポトーシスの初期および後期には大きな違いはなく、全体的は良好で、明らかな細胞毒性がないことが分かった。詳細については、次の表を参照してください。

トランスフェクションの12時間前に、平皿中のBMDCs細胞を吹き分け、培養液を再懸濁し、カウントし、1mLの細胞懸濁液を10×104個/mLの密度で24ウェルプレートの各ウェルに挿入し、培養箱の中で約12時間培養しておく。培地を廃棄し、最終濃度100ng/mLのリポ多糖(LPS)を各ウェルに加え、37℃と4℃でそれぞれ約12時間培養して、BMDCsを刺激する。培地を廃棄し、培地を廃棄し、37℃で培養した細胞を対照群、サイトカラシンD群、フィリピン群、ワートマニン群とクロルプロマジン群に分け、各群に最終濃度25ng/mLのMP1000−LPXを加える。MP1000−LPXを4℃で培養した細胞に添加し、上記の各群を37℃および4℃で暗所で約2時間培養し、さまざまな温度およびさまざまな薬剤作用下でのBMDC細胞の取り込み状況を確認する。培養時点に達した後、培地を廃棄し、冷たいPBSで2回洗浄し、PBSで細胞を24ウェルプレートに収集し、次に細胞をPBSで1回洗浄し(1200rpm、5min)、最後にフローサイトメーターに入れて結果を分析する。
実施例7および実施例9の方法に従って、MP400−LPXおよびメトキシポリエチレングリコール400コレステロール(mP400−LPX)を合成し、これは、主成分E6/E7−mRNA、マンノース標的E6/E7−mRNAを形成するリポソーム複合体を含む。0日目、3日目、10日目、17日目に、マウスに30ugの複合体を注射した。結果を図16〜20に示す。対照群および無関係のmRNA群と比較して、マンノース標的のE6/E7−mRNAリポソーム複合体治療群のマウスの腫瘍増殖データは大幅に減少した。メトキシポリエチレングリコール400コレステロールの対照材料と比較して、mRNAをロードしたマンノースポリエチレングリコール400コレステロールで調製されたワクチンを使用したマウスの生存期間が延長され、1匹のマウスの腫瘍は完全に消失し、標的製剤によって調製されたワクチンは良好な免疫療法効果を示している。
トランスフェクションの12時間前に、平皿中のDC2.4とBMDCs細胞を吹き分け、培養液を再懸濁し、カウントし、1mLの細胞懸濁液を10×104個/mLの密度で24ウェルプレートの各ウェルに挿入し、培養箱の中で約12時間培養しておく。培地を廃棄し、最終濃度100ng/mLのリポ多糖(LPS)を各ウェルに加え、37℃で約12時間培養して、DC2.4とBMDCs細胞を刺激する。培地を廃棄し、各ウェルを6つの群に分け、Cy5−H−mRNAをロードしたさまざまな製剤(対照、LPX、MP100−LPX、MP400−LPX、MP1000−LPXおよびMP2000−LPX)を最終濃度25ng/mLで中央のDC2.4細胞に加える。37℃で暗所で約2時間培養し、さまざまな製剤条件でのDC2.4細胞の取り込み状況を確認する。同時に、GFP−mRNAをロードした異なる製剤(LPX、MP100−LPX、MP400−LPX、MP1000−LPX、MP2000−LPXおよびLipo 3K)を最終濃度25ng/mLでDC2.4およびBMDCs細胞に添加し、暗所で37℃で約2時間培養し、さまざまな製剤でのDC2.4およびBMDC細胞のトランスフェクション効果を確認する。培養時点に達した後、培地を廃棄し、冷たいPBSで2回洗浄し、PBSで細胞を24ウェルプレートに収集し、次に細胞をPBSで1回洗浄し(1200 rpm、5 min)、最後にフローサイトメーターに入れて結果を分析する。
実施例7および実施例9の方法に従って、MP100−LPX、MP400−LPX、MP1000−LPXおよびMP2000−LPXを合成し、主成分E6/E7−mRNA、マンノース標的を形成するE6/E7−mRNAリポソーム複合体を含む。0日目、3日目、10日目、および17日目に、マウスに30ugの複合体を注射し、その結果を図27〜31に示し、MP400−LPXが最も効果的である。他の鎖長の標的比と比較して、ポリエチレングリコール400のマンノースコレステロール標的リガンドは、インビボで最高の抗腫瘍効果を示している。具体的には、マウスの腫瘍治癒率が最も高く、最大60%に達している、また、未治癒のマウスの腫瘍の成長率が低く、担癌マウスの生存期間を延ばすことができる。
Claims (66)
- マンノースとその誘導体によって修飾された標的ナノ製剤であり、標的リガンドであるマンノースおよびその誘導体、ナノ製剤と主薬で構成され、ワクチン送達、腫瘍標的療法、 動脈硬化症および様々な炎症性疾患の治療に使用されることを特徴とする。
- マンノース誘導体は、マンノシド、マンノサミン、マンナンなどの1種または数種であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記ナノ製剤は、リポソーム、エマルジョン、ナノゲル、コアシェルナノ粒子、HDLナノ粒子、固体脂質ナノ粒子、ポリマーミセルの1種または数種であることを特徴とする請求項1に記載のナノ製剤。
- 主薬は小分子薬、タンパク質ポリペプチド薬、または遺伝子薬物の1種または数種であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記標的リガンドであるマンノースおよびその誘導体の含有量は0.05%−40%であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記標的リガンドであるマンノースおよびその誘導体の含有量1%−10%であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記標的リガンドであるマンノースおよびその誘導体の含有量は2%−5%であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記標的リガンドであるマンノースおよびその誘導体の修飾方法では、ナノ製剤を調製した後、マンノースとその誘導体をナノ製剤の表面に吸着または結合させ、または、ナノ製剤を調製する前にマンノースとその誘導体をナノ製剤の調製用担体材料と結合させることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記マンノースおよびその誘導体は、溶媒揮発法、高圧均質化法またはマイクロエマルジョン法によってナノ製剤の表面に吸着させることを特徴とする請求項8に記載の標的ナノ製剤。
- ナノ製剤を修飾するマンノースおよびその誘導体は、1個のマンノースおよびその誘導体または2〜10個のマンノースおよびその誘導体であることを特徴とする請求項1に記載の標的ナノ製剤。
- 前記マンノースおよびその誘導体とナノ製剤またはナノ製剤の調製用担体材料は、直接結合またはスペーサーを介して結合することを特徴とする請求項8に記載の標的ナノ製剤。
- 前記スペーサーは、グルタルアルデヒド、エチレンジアミン、マロン酸、PEG、アミノ酸、ジペプチド、オリゴペプチド、ペプチド、ステアロイル、パルミトイルの1種または数種であることを特徴とする請求項11に記載の標的ナノ製剤。
- ナノ製剤の調製に使用される材料は、活性アミノ基またはヒドロキシル基の少なくとも1種であり、薬学的に許容できるミセル、エマルジョン、ナノゲル、コアシェルナノ粒子、HDLナノ粒子、固体脂質ナノ粒子、リポソーム、ポリマーミセルなどの調製用担体材料で薬学的に許容できるナノ製薬の調製方法によって調製されることを特徴とする請求項1−12に記載の標的ナノ製剤。
- 標的機能を備えたナノ製剤の調製に使用される組成物であり、前記組成物は標的物質とナノ製剤基礎材料を備え、前記標的物質はマンノースおよび/またはマンノース誘導体であることを特徴とする。
- 前記マンノース誘導体は、マンノシドおよび/またはマンノサミンおよび/またはマンナンであることを特徴とする請求項14に記載の組成物。
- 前記マンノース誘導体は、メチル−D−マンノシド、1−αホルミルメチル−マンノピラノシド、4−アミノフェニル−α−D−マンノピラノシド、4−ニトロフェニル−α−D−マンノピラノシド、4−メチルウンベリフェリル−α−D−マンノピラノシド、マンノース−6−リン酸、カルバモイル−D−マンノースであることとを特徴とする請求項15に記載の組成物。
- 前記組成物中の標的物質の重量パーセントは、0.05−40%であることを特徴とする請求項14または15または16に記載の組合物。
- 前記組成物中の前記標的物質の重量パーセントは、1−10%であることを特徴とする請求項14または15または16に記載の組成物。
- 前記組成物は、前記標的物質、前記ナノ製剤基礎材料、および標的物質とナノ製剤基礎材料との間に距離を作り出すスペーサー材料から構成され、前記スペーサー材料は、グルタルアルデヒド、エチレンジアミン、マロン酸、短鎖アルキル鎖、PEG、アミノ酸、ジペプチド、オリゴペプチド、ポリペプチド、ステアロイルおよびパルミトイルの1種または数種であることを特徴とする請求項14に記載の組成物。
- 前記ナノ製剤基礎材料は、リポソーム、エマルジョン、ナノゲル、コアシェルナノ粒子、HDLナノ粒子、固体脂質ナノ粒子、およびポリマーミセルを調製するための材料の組成物であることを特徴とする請求項14に記載の組成物。
- 前記スペーサー材料はPEGnであり、n=100−5000であることを特徴とする請求項19に記載の組成物。
- 前記nは100、200、300、400、500、600、800、900、1000または2000であることを特徴とする請求項21に記載の組成物。
- 請求項19に記載の組成物であって、重量部で次の成分からなることを特徴とする:マンノース1部、PEG 0.5〜56部、コレステロール2〜11部。
- 請求項19に記載の組成物であって、重量部で次の成分からなることを特徴とする:マンノース1部、PEG 10〜25部、コレステロール4〜8部。
- 請求項14に記載の組成物で調製された薬物送達のための標的担体。
- 前記薬物は小分子薬および/またはタンパク質ペプチド薬および/または遺伝子薬物であることを特徴とする請求項25に記載の標的担体。
- 前記小分子薬は、タキサン類、カンプトテシン類、ヴィンブラスティン類、アドリアマイシン、ファルモルビシン、ダウノルビシン、エピルビシン、アンフォテリシン、メトトレキサート、シタラビン、5−フルオロウラシル、ミトキサントロンまたはその誘導体、ゲフィティニブ、ノスカピン、シスプラチン、カルボプラチン、オキサリプラチン、カルムスティンとケルセチンの1種または数種であることを特徴とする請求項26に記載の標的担体。
- 前記タンパク質ペプチド薬はインターロイキン、成長因子、インターフェロン、インテグリン、モノクローナル抗体、酵素およびインスリンであることを特徴とする請求項26に記載の標的担体。
- 前記インターロイキンには、IL−1、IL−1a、IL−2、IL−3、IL−4、IL−5、IL−6を含むがこれらに限定されない、前記成長因子には、線維芽細胞成長因子、肝臓成長因子、血管内皮成長因子、造血成長因子を含むがこれらに限定されない、前記インターフェロンにはIFNα、IFNβ、IFNγを含むがこれらに限定されない、前記腫瘍壊死因子にはTNFα、TNFβを含むがこれらに限定されないことを特徴とする請求項18に記載の標的担体。
- 前記遺伝子薬物は、DNA、プラスミド、mRNA、siRNA、shRNA、microRNAの1種または数種であることを特徴とする請求項26に記載の標的担体。
- 請求項25に記載の標的担体によってカプセル化された薬物。
- 前記ナノ製剤基礎材料をナノ薬物送達担体に調製し、そして前記標的物質を前記ナノ薬物送達担体の表面に付着させることを特徴とする請求項31に記載の標的担体の調製方法。
- 請求項25に記載の標的担体の標的化性を改善するための方法であって、その特徴は以下のとおりである。
A標的化要素の合成
前記標的物質と前記スペーサー材料を標的化要素に合成する。
B標的担体の調製
ナノ製剤基礎材料をナノ薬物送達担体に調製し、次のステップAで得られた標的化要素と前記ナノ薬物送達担体を接続して標的担体を得る、または前記スペーサー材料とナノ製剤基礎材料を合成し、そして前記標的物質と接続して標的担体を得る。 - 前記標的物質は具体的にはマンノースとその誘導体であり、前記スペーサー材料は具体的にはPEGであることを特徴とする請求項33に記載の方法。
- 前記標的物質と前記ナノ薬物送達担体との距離は0.2−100nmであることを特徴とする請求項33に記載の方法。
- 前記標的化要素と前記ナノ薬物送達担体との距離は0.3−10nmであることを特徴とする請求項35に記載の方法。
- 薬用成分を標的細胞に送達するための方法であって、薬用成分を請求項25に記載の標的担体に入れて送達することを特徴とする。
- 前記標的細胞はマンノース受容体を含む細胞であることを特徴とする請求項37に記載の方法。
- 前記標的細胞には、マクロファージ、樹状細胞、および腫瘍細胞が含まれるが、これらに限定されないことを特徴とする請求項37に記載の方法。
- 標的化要素を調製するための組成物であって、前記組成物は標的物質とスペーサー材料から構成され、前記標的物質はマンノースおよび/またはマンノース誘導体であることを特徴とする。
- 前記マンノース誘導体は、マンノシドおよび/またはマンノサミンおよび/またはマンナンであることを特徴とする請求項40に記載の組成物。
- 前記マンノース誘導体は、メチル−D−マンノシド、1−αホルミルメチル−マンノピラノシド、4−アミノフェニル−α−D−マンノピラノシド、4−ニトロフェニル−α−D−マンノピラノシド グリコシド、4−メチルウンベリフェリル−α−D−マンノピラノシド、マンノース−6−ホスフェート、カルバモイル−D−マンノースのいずれか1種または数種であることを特徴とする請求項40に記載の組成物。
- 前記スペーサー材料は、グルタルアルデヒド、エチレンジアミン、マロン酸、短鎖アルキル鎖、PEG、アミノ酸、ジペプチド、オリゴペプチド、ポリペプチド、ステアロイル、パルミトイルの1種または数種であることを特徴とする請求項40に記載の組成物。
- 前記スペーサー材料と前記標的物質とのモル比は1−10:1であることを特徴とする請求項40に記載の組成物。
- 前記スペーサー材料と前記標的物質とのモル比は5:1であることを特徴とする請求項44に記載の組成物。
- マンノースとPEGとのモル比は1:5であることを特徴とする請求項40に記載の組成物。
- マンノースとPEGとのモル比は1:1であることを特徴とする請求項46に記載の組成物。
- 請求項40に記載の組成物で調製された標的化要素である。
- 前記標的化要素は式1に示すようであることを特徴とする請求項48に記載の標的化要素、ここで、R1は−C5H8O2、−C2H7N2、−C3H3O4、−O(CH2CH2O)nH、−C2H3O2NR’または−C2H3ONR’、−C4H5O3N2R1’R2’または−C4H5O2N2R1’R2’、−C3n+1H3n+2On+2Nn+1R1’R2’または−C3n+1H3n+2On+1Nn+1R1’R2’、−C18H35O、C16H31O、−H 、−CHO、−C6H4NH2、− C6H4NO2、−CONH2、−CH2C6CCHCO2CH3であり、前記R1’とR2’は、任意の天然アミノ酸側鎖R’グループであり、R2は− CH3または−PO3H2、R3は−H、R4は−H、R5は−Hまたは−NHCOCH3である。
- 請求項48に記載の標的化要素を含む標的担体。
- 標的化要素とナノ製剤で接続され、前記ナノ製剤は、リポソーム、エマルジョン、ナノゲル、コアシェルナノ粒子、HDLナノ粒子、固体脂質ナノ粒子、ポリマーミセルであることを特徴とする請求項51に記載の標的担体。
- 1個の標的担体に1〜100000個の前記標的化要素を接続していることを特徴とする請求項51に記載の標的担体。
- 前記1個の標的担体に20〜1000個の前記標的化要素を接続していることを特徴とする請求項53に記載の標的担体。
- 前記標的化要素のターゲットヘッドと前記ナノ製剤との距離は0.2−100nmであることを特徴とする請求項52に記載の標的担体。
- 前記標的化要素のターゲットヘッドと前記ナノ製剤との距離は0.3−10nmであることを特徴とする請求項55に記載の標的担体。
- 前記ターゲットヘッドはマンノシドおよび/またはマンノサミンおよび/またはマンナンであることを特徴とする請求項55または56に記載の標的担体。
- 前記標的物質とスペーサー材料を接続した後、ナノ製剤の材料と接続してナノ製剤を形成することを特徴とする請求項48に記載の標的化要素をナノ製剤に接続する方法。
- 請求項48に記載の標的化要素をナノ製剤に接続する方法であって、以下の手順を含むことを特徴とする。
1)ジエチレングリコール、p−トルエンスルホニルクロリドおよびトリエチルアミンをDCMに溶解し、室温で反応させ、カラムクロマトグラフィーによって分離して化合物1を得る。
2)化合物1とペンタアセチル化マンノースおよびその誘導体と三フッ化ホウ素エーテルをDCMに溶解し、室温で反応させ、カラムクロマトグラフィーによって分離して化合物2を得る。
3)化合物2とアジドナトリウムをDMFに溶解し、シリカゲルカラムクロマトグラフィーによって分離して固体化合物3を得る。
4)化合物3をメトキシドナトリウムのメタノール溶液に溶解し、室温で反応させ、濃縮して化合物4を得る。
5)コレステロール、ブロモプロピン、水素ナトリウムをエーテルとDMFの混合溶液に溶解し、室温で反応させ、シリカゲルカラムクロマトグラフィーによって分離して固体化合物5を得る。
6)化合物4、化合物5およびヨウ化第一銅をDMFに溶解し、室温で24時間反応させ、シリカゲルカラムクロマトグラフィーによって標的担体固体化合物6を得る。 - 薬物を標的担体にカプセル化されて送達することを特徴とする請求項51に記載標的担体の薬物送達能力を改善する方法。
- 前記薬物は小分子薬および/またはタンパク質ペプチド薬および/または遺伝子薬物であることを特徴とする請求項60に記載の方法。
- 前記標的担体は薬物をマンノース受容体標的を含む標的細胞に送達できることを特徴とする請求項60に記載の方法。
- 請求項51に記載の標的担体を含む複合体であって、前記複合体は前記標的担体と受容体によって形成された複合体であり、前記標的担体は薬物を運び、前記受容体はマンノース受容体であることを特徴とする。
- トランスフェクション試薬のトランスフェクション効率を改善するための方法であって、高いトランスフェクション効率を有するトランスフェクション試薬を調製するために請求項40に記載の標的化要素の組成物をトランスフェクション試薬を調製するための担体に接続することを特徴とする。
- 請求項48に記載の標的化要素を標的油相として使用される。
- 請求項48に記載の標的化要素を標的水相として使用される。
